Antimicrobial resistance is one of the greatest threats to human health. Gram-positive methicillin resistant Staphylococcus aureus (MRSA), in both its planktonic and biofilm form, is of particular concern. Herein we identify the hydrogelation properties for a series of intrinsically fluorescent, structurally related supramolecular self-associating amphiphiles and determine their efficacy against both planktonic and biofilm forms of MRSA. To further explore the potential translation of this hydrogel technology for real-world applications, the toxicity of the amphiphiles was determined against the eukaryotic multicellular model organism, Caenorhabditis elegans. Due to the intrinsic fluorescent nature of these supramolecular amphiphiles, material characterisation of their molecular self-associating properties included; comparative optical density plate reader assays, rheometry and widefield fluorescence microscopy. This enabled determination of amphiphile structure and hydrogel sol dependence on resultant fibre formation.

Species–area models now frequently include habitat heterogeneity. These models often fit real-world data better than those that exclude this factor. However, such models usually link the effects of habitat heterogeneity and study area. Critically, we show that difficulties in quantifying habitat heterogeneity within these models can lead to distortions of the apparent effect of area on species richness. Here, we derive a model that minimises these distortions by partitioning the influence of habitat heterogeneity from that of area, without compromising ease of application. This ‘jigsaw model’ achieves this by assuming that different habitats within an area can support similar numbers of species. We compare the behaviour of this model to that of existing models of similar complexity using both simulated island ecosystems and 40 published empirical datasets. The effects of habitat heterogeneity and area on species richness vary independently in our simulations, and these independent effects are recovered by the jigsaw model. This flexibility, however, is not present when the same data are analysed using other models of similar complexity. When applied to real-world data, the jigsaw model demonstrates that the relative importance of area and habitat heterogeneity varies depending on the study system. The jigsaw model provides the best fit to real-world data (according to AICc) of all tested models in logarithmic form, and the second best fit, after the choros model, in power-law form. Our results demonstrate the importance of partitioning the effects of habitat heterogeneity and area on species richness in biogeographic models. The jigsaw model is a simple but powerful tool for such partitioning. It has the potential to elucidate the underlying drivers of species richness patterns, and to be used as a tool in biological conservation projects, where data are often incomplete.

The Winter Fog Experiment (WiFEX) was an intensive field campaign conducted at Indira Gandhi International Airport (IGIA) Delhi, India, in the Indo-Gangetic Plain (IGP) during the winter of 2017–2018. Here, we report the first comparison in South Asia of high-temporal-resolution simulation of ammonia (NH3) along with ammonium (NH4+) and total NHx (i.e., NH3+ NH4+) using the Weather Research and Forecasting model coupled with chemistry (WRF-Chem) and measurements made using the Monitor for AeRosols and Gases in Ambient Air (MARGA) at the WiFEX research site. In the present study, we incorporated the Model for Simulating Aerosol Interactions and Chemistry (MOSAIC) aerosol scheme into WRF-Chem. Despite simulated total NHx values and variability often agreeing well with the observations, the model frequently simulated higher NH3 and lower NH4+ concentrations than the observations. Under the winter conditions of high relative humidity (RH) in Delhi, hydrogen chloride (HCl) was found to promote the increase in the particle fraction of NH4+ (which accounted for 49.5 % of the resolved aerosol in equivalent units), with chloride (Cl−) (29.7 %) as the primary anion. By contrast, the absence of chloride (HCl / Cl−) chemistry in the standard WRF-Chem model results in the prediction of sulfate (SO42-) as the dominant inorganic aerosol anion. To understand the mismatch associated with the fraction of NHx in the particulate phase (NH4+ / NHx), we added HCl / Cl− to the model and evaluated the influence of its chemistry by conducting three sensitivity experiments using the model: no HCl, base case HCl (using a published waste burning inventory), and 3 × base HCl run. We found that 3 × base HCl increased the simulated average NH4+ by 13.1 µg m−3 and NHx by 9.8 µg m−3 concentration while reducing the average NH3 by 3.2 µg m−3, which is more in accord with the measurements. Thus HCl / Cl− chemistry in the model increases total NHx concentration, which was further demonstrated by reducing NH3 emissions by a factor of 3 (−3 × NH3_EMI) in the 3 × base HCl simulation. Reducing NH3 emissions in the 3 × base HCl simulation successfully addressed the discrepancy between measured and modeled total NHx. We conclude that modeling the fate of NH3 in Delhi requires a correct chemistry mechanism accounting for chloride dynamics with accurate inventories of both NH3 and HCl emissions.

Cropland is a main source of global nitrogen pollution1,2. Mitigating nitrogen pollution from global croplands is a grand challenge because of the nature of non-point-source pollution from millions of farms and the constraints to implementing pollution-reduction measures, such as lack of financial resources and limited nitrogen-management knowledge of farmers3. Here we synthesize 1,521 field observations worldwide and identify 11 key measures that can reduce nitrogen losses from croplands to air and water by 30–70%, while increasing crop yield and nitrogen use efficiency (NUE) by 10–30% and 10–80%, respectively. Overall, adoption of this package of measures on global croplands would allow the production of 17 ± 3 Tg (1012 g) more crop nitrogen (20% increase) with 22 ± 4 Tg less nitrogen fertilizer used (21% reduction) and 26 ± 5 Tg less nitrogen pollution (32% reduction) to the environment for the considered base year of 2015. These changes could gain a global societal benefit of 476 ± 123 billion US dollars (USD) for food supply, human health, ecosystems and climate, with net mitigation costs of only 19 ± 5 billion USD, of which 15 ± 4 billion USD fertilizer saving offsets 44% of the gross mitigation cost. To mitigate nitrogen pollution from croplands in the future, innovative policies such as a nitrogen credit system (NCS) could be implemented to select, incentivize and, where necessary, subsidize the adoption of these measures.

Fine particulate matter (PM2.5, particles with a mass median aerodynamic diameter of less than 2.5 micrometers) in the atmosphere is associated with severe negative impacts on human health, and the gases sulfur dioxide, nitrogen oxides, and ammonia are the main PM2.5 precursors. However, their contribution to global health impacts has not yet been analyzed. Here, we show that nitrogen accounted for 39% of global PM2.5 exposure in 2013, increasing from 30% in 1990 with rising reactive nitrogen emissions and successful controls on sulfur dioxide. Nitrogen emissions to air caused an estimated 23.3 million years of life lost in 2013, corresponding to an annual welfare loss of 420 billion United States dollars for premature death. The marginal abatement cost of ammonia emission is only 10% that of nitrogen oxides emission globally, highlighting the priority for ammonia reduction.

To assess the status of and trends in agricultural nitrogen (N) flows and their wider consequences for Bangladesh, in this study, we analyzed data from national and international bodies. The increased rates of N fertilizer applied for increased food production leaves behind a huge amount of unutilized reactive N (Nr). N fertilizer use is the largest in the crop sector, an important sector, where current annual consumption is 1190 Gg. The present combined annual Nr production from crop, fishery, and livestock sectors is ~600 Gg, while emissions of nitrous oxide (N2O), a potent greenhouse gas, are ~200 Gg. Poor N management results in Nr leaking into the environment, which has increased approximately 16-fold since 1961. One potential consequence is the disruption of ecosystem functioning. The balanced tradeoff between food production and reducing Nr input needs to be achieved. One solution to reducing Nr may be a holistic approach that optimizes N application rates and incorporates waste of one subsector as an input to another applying the principle of the circular economy.

Ammonia (NH3) in the atmosphere affects both the environment and human health. It is therefore increasingly recognised by policy makers as an important air pollutant that needs to be mitigated, though it still remains unregulated in many countries. In order to understand the effectiveness of abatement strategies, routine NH3 monitoring is required. Current reference protocols, first developed in the 1990s, use daily samplers with offline analysis; however, there have been a number of technologies developed since, which may be applicable for high time resolution routine monitoring of NH3 at ambient concentrations. The following study is a comprehensive field intercomparison held over an intensively managed grassland in southeastern Scotland using currently available methods that are reported to be suitable for routine monitoring of ambient NH3. In total, 13 instruments took part in the field study, including commercially available technologies, research prototype instruments, and legacy instruments. Assessments of the instruments' precision at low concentrations (< 10 ppb) and at elevated concentrations (maximum reported concentration of 282 ppb) were undertaken. At elevated concentrations, all instruments performed well and with precision (r2 > 0.75). At concentrations below 10 ppb, however, precision decreased, and instruments fell into two distinct groups, with duplicate instruments split across the two groups. It was found that duplicate instruments performed differently as a result of differences in instrument setup, inlet design, and operation of the instrument. New metrological standards were used to evaluate the accuracy in determining absolute concentrations in the field. A calibration-free CRDS optical gas standard (OGS, PTB, DE) served as an instrumental reference standard, and instrument operation was assessed against metrological calibration gases from (i) a permeation system (ReGaS1, METAS, CH) and (ii) primary standard gas mixtures (PSMs) prepared by gravimetry (NPL, UK). This study suggests that, although the OGS gives good performance with respect to sensitivity and linearity against the reference gas standards, this in itself is not enough for the OGS to be a field reference standard, because in field applications, a closed path spectrometer has limitations due to losses to surfaces in sampling NH3, which are not currently taken into account by the OGS. Overall, the instruments compared with the metrological standards performed well, but not every instrument could be compared to the reference gas standards due to incompatible inlet designs and limitations in the gas flow rates of the standards. This work provides evidence that, although NH3 instrumentation have greatly progressed in measurement precision, there is still further work required to quantify the accuracy of these systems under field conditions. It is the recommendation of this study that the use of instruments for routine monitoring of NH3 needs to be set out in standard operating protocols for inlet setup, calibration, and routine maintenance in order for datasets to be comparable.

Excess nitrogen is a pollutant and global problem that harms ecosystems and can severely affect human health. Pollutant nitrogen is becoming more widespread and intensifying in the tropics. There is thus a requirement to develop nitrogen biomonitoring for spatial mapping and trend analysis of tropical biodiversity and ecosystems. In temperate and boreal zones, multiple bioindicators for nitrogen pollution have been developed, with lichen epiphytes among the most sensitive and widely applied. However, the state of our current knowledge on bioindicators is geographically biased, with extensive research effort focused on bioindicators in the temperate and boreal zones. The development of lichen bioindicators in the tropics is further weakened by incomplete taxonomic and ecological knowledge. In this study we performed a literature review and meta-analysis, attempting to identify characteristics of lichens that offer transferability of bioindication into tropical regions. This transferability must overcome the different species pools between source information – drawing on extensive research effort in the temperate and boreal zone – and tropical ecosystems. Focussing on ammonia concentration as the nitrogen pollutant, we identify a set of morphological traits and taxonomic relationships that cause lichen epiphytes to be more sensitive, or more resistant to this excess nitrogen. We perform an independent test of our bioindicator scheme and offer recommendations for its application and future research in the tropics.

It has often been suggested that the productivity of an ecosystem affects the number of species that it can support. Despite decades of study, the nature, extent, and underlying mechanisms of this relationship are unclear. One suggested mechanism is the “more individuals” hypothesis (MIH). This proposes that productivity controls the number of individuals in the ecosystem, and that more individuals can be divided into a greater number of species before their population size is sufficiently small for each to be at substantial risk of extinction. Here, we test this hypothesis using REvoSim: an individual‐based eco‐evolutionary system that simulates the evolution and speciation of populations over geological time, allowing phenomena occurring over timescales that cannot be easily observed in the real world to be evaluated. The individual‐based nature of this system allows us to remove assumptions about the nature of speciation and extinction that previous models have had to make. Many of the predictions of the MIH are supported in our simulations: Rare species are more likely to undergo extinction than common species, and species richness scales with productivity. However, we also find support for relationships that contradict the predictions of the strict MIH: species population size scales with productivity, and species extinction risk is better predicted by relative than absolute species population size, apparently due to increased competition when total community abundance is higher. Furthermore, we show that the scaling of species richness with productivity depends upon the ability of species to partition niche space. Consequently, we suggest that the MIH is applicable only to ecosystems in which niche partitioning has not been halted by species saturation. Some hypotheses regarding patterns of biodiversity implicitly or explicitly overlook niche theory in favor of neutral explanations, as has historically been the case with the MIH. Our simulations demonstrate that niche theory exerts a control on the applicability of the MIH and thus needs to be accounted for in macroecology., We use simulation experiments to test the predictions of the more‐individuals hypothesis of biodiversity. We find that many of these predictions are supported, but that interspecific competition and limits to niche diversity can provide additional complexity not predicted by the more‐individuals hypothesis.

Mismatch repair (MMR) is a highly conserved repair pathway essential to safeguard the genome from errors that occur during DNA replication. In Saccharomyces cerevisiae, two Msh complexes initiate MMR, either Msh2-Msh3 or Msh2-Msh6. These heterodimeric complexes recognize a number of DNA structures with varying affinities and play critical roles in DNA metabolism outside of post-replicative MMR; they contribute to genome stability through homologous recombination, double strand break repair (DSBR), and the DNA damage response.In contrast to its role in promoting genome stability, Msh2-Msh3 is associated with genome instability through trinucleotide repeat (TNR) expansions. Msh2-Msh3’s non-canonical activity in TNR expansions appears to be an unfortunate consequence of its intrinsic ability to bind to distinct DNA structures. Msh2-Msh3 binds to a wide range of DNA structures, including branched intermediates with 3’ or 5’ ss/ds DNA. Whereas the binding to 3’ ssDNA tails leads to repair in a specialized form of DSBR called 3’ non-homologous tail removal (3’NHTR), in vitro evidence suggests that Msh2-Msh3 binding to 5’ flap structures interferes with the cleavage activity of the structure-specific endonuclease Rad27 (Fen1 in humans). The endonucleolytic function of Rad27 promotes 5’ ssDNA flap processing during Okazaki fragment maturation and long-patch base excision repair (LP-BER). Here we show that elevated Msh2-Msh3 levels interfere with both DNA replication and BER in vivo in an expression and ATP-dependent manner, consistent with the hypothesis that protein abundance and ATPase activities in Msh3 are key drivers of Msh2-Msh3-mediated genomic instability.

Unsustainable phosphorus use is pushing food security further from reach1,2, leaving a legacy of polluted freshwaters, many now beyond ecological restoration3. Ten years have passed since the global anthropogenic flow of phosphorus was identified to be exceeding its planetary boundary4. In 2013, the opportunity was highlighted for a 20% improvement in nutrient use efficiency by 2020 across the full chain of food and waste systems5. The working group of the Post-2020 Global Biodiversity Framework proposed to reduce pollution from excess nutrients by 50% by 20306. Yet, phosphorus management remains largely ignored in the food and environmental policy agendas of most countries and international conventions7. Progress remains hindered by a lack of policy and public awareness, fragmentation of actions and policies, and the absence of intergovernmental coordination.

A comprehensive European dataset on monthly atmospheric NH3, acid gases (HNO3, SO2, HCl), and aerosols (NH4+, NO3-, SO42-, Cl−, Na+, Ca2+, Mg2+) is presented and analysed. Speciated measurements were made with a low-volume denuder and filter pack method (DEnuder for Long-Term Atmospheric sampling, DELTA®) as part of the EU NitroEurope (NEU) integrated project. Altogether, there were 64 sites in 20 countries (2006–2010), coordinated between seven European laboratories. Bulk wet-deposition measurements were carried out at 16 co-located sites (2008–2010). Inter-comparisons of chemical analysis and DELTA® measurements allowed an assessment of comparability between laboratories. The form and concentrations of the different gas and aerosol components measured varied between individual sites and grouped sites according to country, European regions, and four main ecosystem types (crops, grassland, forests, and semi-natural). The smallest concentrations (with the exception of SO42- and Na+) were in northern Europe (Scandinavia), with broad elevations of all components across other regions. SO2 concentrations were highest in central and eastern Europe, with larger SO2 emissions, but particulate SO42- concentrations were more homogeneous between regions. Gas-phase NH3 was the most abundant single measured component at the majority of sites, with the largest variability in concentrations across the network. The largest concentrations of NH3, NH4+, and NO3- were at cropland sites in intensively managed agricultural areas (e.g. Borgo Cioffi in Italy), and the smallest were at remote semi-natural and forest sites (e.g. Lompolojänkkä, Finland), highlighting the potential for NH3 to drive the formation of both NH4+ and NO3- aerosol. In the aerosol phase, NH4+ was highly correlated with both NO3- and SO42-, with a near-1:1 relationship between the equivalent concentrations of NH4+ and sum (NO3-+ SO42-), of which around 60 % was as NH4NO3. Distinct seasonality was also observed in the data, influenced by changes in emissions, chemical interactions, and the influence of meteorology on partitioning between the main inorganic gases and aerosol species. Springtime maxima in NH3 were attributed to the main period of manure spreading, while the peak in summer and trough in winter were linked to the influence of temperature and rainfall on emissions, deposition, and gas–aerosol-phase equilibrium. Seasonality in SO2 was mainly driven by emissions (combustion), with concentrations peaking in winter, except in southern Europe, where the peak occurred in summer. Particulate SO42- showed large peaks in concentrations in summer in southern and eastern Europe, contrasting with much smaller peaks occurring in early spring in other regions. The peaks in particulate SO42- coincided with peaks in NH3 concentrations, attributed to the formation of the stable (NH4)2SO4. HNO3 concentrations were more complex, related to traffic and industrial emissions, photochemistry, and HNO3:NH4NO3 partitioning. While HNO3 concentrations were seen to peak in the summer in eastern and southern Europe (increased photochemistry), the absence of a spring peak in HNO3 in all regions may be explained by the depletion of HNO3 through reaction with surplus NH3 to form the semi-volatile aerosol NH4NO3. Cooler, wetter conditions in early spring favour the formation and persistence of NH4NO3 in the aerosol phase, consistent with the higher springtime concentrations of NH4+ and NO3-. The seasonal profile of NO3- was mirrored by NH4+, illustrating the influence of gas–aerosol partitioning of NH4NO3 in the seasonality of these components. Gas-phase NH3 and aerosol NH4NO3 were the dominant species in the total inorganic gas and aerosol species measured in the NEU network. With the current and projected trends in SO2, NOx, and NH3 emissions, concentrations of NH3 and NH4NO3 can be expected to continue to dominate the inorganic pollution load over the next decades, especially NH3, which is linked to substantial exceedances of ecological thresholds across Europe. The shift from (NH4)2SO4 to an atmosphere more abundant in NH4NO3 is expected to maintain a larger fraction of reactive N in the gas phase by partitioning to NH3 and HNO3 in warm weather, while NH4NO3 continues to contribute to exceedances of air quality limits for PM2.5.

South Asian regional cooperation with the International Nitrogen Initiative and the India-led UN Resolution on Sustainable Nitrogen Management (UNEP/EA.4/L.16) brought South Asia into global focus. Here, we reflect upon its proactive scientific community, growing scientific capacity, and international collaborations, which have enabled the emergence of the UKRI-GCRF South Asian Nitrogen Hub, which has great potential both regionally and globally.

Here, we report the first comparison in South Asia of the high temporal resolution measurements of NH3 along with water-soluble inorganic ions in PM2.5 (Cl-, NO3-, SO42- and NH4+) and corresponding precursor gases (HCl, SO2, HONO, and HNO3) made at the WiFEX research site, using the Monitor for AeRosols and Gases in Ambient Air (MARGA) and high-resolution simulations with Weather Research and Forecasting model coupled with chemistry (WRF-Chem). The hourly measurements were used to investigate how well the model captures the temporal variation of gaseous and particulate water-soluble species and gas-to-particle partitioning of NH3, using the Model for Simulating Aerosol Interactions and Chemistry (MOSAIC) aerosol scheme. The model frequently simulated higher NH3 and lower NH4+ concentrations than the observations, while total NHx values/variability agreed well with the observations. Under the winter conditions of Delhi, high concentrations of hydrochloric acid (HCl) in the ambient air are found to dominate the gas-to-particle partitioning, as NH3 is usually in excess. The default model set-up of WRF-Chem excludes anthropogenic HCl emissions, so sulfuric acid (H2SO4) dominates the gas-to-particle partitioning with NH3 during the simulation period. The sensitivity experiments, including HCl emissions in the model, showed that the inclusion of HCl emissions improves the simulated gas-to-particle conversion rate of ammonia by 24 % (as indicated by NH4+ concentrations) while reducing the bias in gas phase NH3 by 10 %. Nevertheless, even with waste burning HCl emissions included, we find that WRF-Chem still overestimates sulfur dioxide (SO2) and nitrate (NO3−) formation and underestimates sulfate (SO42−), nitrous acid (HONO), nitric acid (HNO3), and HCl concentration in which it interacts, thus limit the gas-to-particle conversion of NH3 to NH4+ in the model. This indicates that modeling of ammonia requires a correct chemistry mechanism with accurate emission inventories for the industrial HCl emissions.

Translesion synthesis (TLS) by specialized DNA polymerases (Pols) is an evolutionarily conserved mechanism for tolerating replication-blocking DNA lesions. Using the Escherichia coli dinB-encoded Pol IV as a model to understand how TLS is coordinated with the actions of the high-fidelity Pol III replicase, we previously described a novel Pol IV mutant containing a threonine 120-to-proline mutation (Pol IV-T120P) that failed to exchange places with Pol III at the replication fork in vitro as part of a Pol III-Pol IV switch. This in vitro defect correlated with the inability of Pol IV-T120P to support TLS in vivo, suggesting Pol IV gains access to the DNA, at least in part, via a Pol III-Pol IV switch. Interaction of Pol IV with the β sliding clamp and the single-stranded DNA binding protein (SSB) significantly stimulates Pol IV replication and facilitates its access to the DNA. In this work, we demonstrate that Pol IV interacts physically with Pol III. We further show that Pol IV-T120P interacts normally with the β clamp, but is impaired in interactions with the α catalytic and εθ proofreading subunits of Pol III, as well as SSB. Taken together with published work, these results provide strong support for the model in which Pol IV-Pol III and Pol IV-SSB interactions help to regulate the access of Pol IV to the DNA. Finally, we describe several additional E. coli Pol-Pol interactions, suggesting Pol-Pol interactions play fundamental roles in coordinating bacterial DNA replication, DNA repair, and TLS. IMPORTANCE Specialized DNA polymerases (Pols) capable of catalyzing translesion synthesis (TLS) generate mutations that contribute to bacterial virulence, pathoadaptation, and antimicrobial resistance. One mechanism by which the bacterial TLS Pol IV gains access to the DNA to generate mutations is by exchanging places with the bacterial Pol III replicase via a Pol III-Pol IV switch. Here, we describe multiple Pol III-Pol IV interactions and discuss evidence that these interactions are required for the Pol III-Pol IV switch. Furthermore, we describe several additional E. coli Pol-Pol interactions that may play fundamental roles in managing the actions of the different bacterial Pols in DNA replication, DNA repair, and TLS.

An increasing concentration of nitrous oxide (N2O) in the global atmosphere can perturb the ecological balance, affecting the climate and human life. South Asia, one of the world’s most populous regions, is a hotspot for N2O emission. Although agriculture traditionally dominated the region, economic activities are rapidly shifting towards industry and energy services. These activites may become the largest emitters of N2O in future. Yet, few attempts have been made to estimate long-term direct N2O emission from fuel combustion for the different energy-consuming sectors in the South Asian region. Therefore, the present study developed a comprehensive sectoral N2O emission inventory for South Asian countries for the time period of 1990–2017, with projections till 2041. It revealed that the average N2O emission from fuel combustion in the South Asia region is about 40.96 Gg yr−1 with a possible uncertainty of ±12 Gg yr−1, showing an increase of more than 100% from 1990 to 2017. Although India is the major contributor, with an average of 34 Gg yr−1 of N2O emissions, in terms of growth, small countries like Bhutan and Maldives are dominating other South Asian countries. Sector-wise, the residential sector contributed a maximum emission of 14.52 Gg yr−1 of N2O but this is projected to reduce by more than 50% by 2041. This is because of the successful promotion of cleaner fuels like liquefied petroleum gas over more polluting fuelwood. Power generation contributed 9.43 Gg yr−1of N2O emissions, exhibiting a maximum growth of 395%, followed by road transport (289%) and industry (231%). Future N2O emissions from transport, power and industry are projected to rise by 2.8, 3.3, and 23.9 times their 2017 estimates, respectively, due to the incapability of current policies to combat rising fossil fuel consumption. Mitigation options, such as replacing diesel and compressed natural gas vehicles with electricity-driven vehicles, can decelerate N2O emissions to 45% by 2041 for road transport. A 41% reduction is possible by displacing coal with renewables in the power and industry sectors. Overall, the South Asian contribution to global N2O emissions has enlarged from 2.7% in 1990 to 5.7% in 2007–2016, meaning there is an urgent need for N2O emission mitigation in the region.

Ammonia (NH3) in the atmosphere affects both the environment and human health. It is therefore increasingly recognised by policy makers as an important air pollutant that needs to be mitigated. In order to understand the effectiveness of abatement strategies, routine NH3 monitoring is required. Current reference protocols, developed in the 1990s, use daily samplers with offline analysis but there have been a number of technologies developed since, which may be applicable for high time resolution routine monitoring of NH3 at ambient concentrations. The following study is a comprehensive field intercomparison held over an intensively managed grassland in South East Scotland using currently available methods that are reported to be suitable for routine monitoring of ambient NH3. In total 13 instruments took part in the field study. The instruments include: an online ion chromatography system (MARGA, Metrohm-Applikon,NL), two wet chemistry continuous flow analysis systems (AiRRmonia, Mechatronics, NL), a photoacoustic spectrometer (NH3 monitor, LSE, NL), two mini Differential Optical Absorption Spectrometers (miniDOAS; NTB Interstate University of Applied Sciences Buchs, now part of "Eastern Switzerland University of Applied Sciences, CH and RIVM, NL), as well as seven spectrometers using cavity enhanced techniques: a Quantum Cascade Laser Absorption Spectrometer (QCLAS, Aerodyne, Inc. US), Picarro G2103 Analyzer (Picarro US), Economical NH3 Analyser (Los Gatos Research, US), Tiger-i 2000 (Tiger Optics, US) and LaserCEM® gas analyser (AP2E, FR). Assessments of the instruments’ precision at low concentrations (< 10 ppb) and at elevated concentrations (maximum reported concentration of 282 ppb) were undertaken. At elevated concentrations all instruments performed well on precision (r2 > 0.75). At concentrations below 10 ppb however, instruments fell into two distinct groups and the duplicate instruments, miniDOAS, AiRRmonia, LGR and Picarro were split across the two groups. It was found that identical instruments performed differently at low concentrations, highlighting the impact of the setup, inlet design and operation of the instrument used. Accuracy in determining absolute concentrations in the field was assessed using a calibration-free CRDS Optical Gas Standard (OGS, PTB, DE), serving as an instrumental reference standard. Accuracy was also assessed using well established metrological standards for calibration gases, i) a permeation system (ReGaS1, METAS, CH) and ii) Primary Standard gas Mixtures (PSMs) prepared by gravimetry (NPL, UK). This study showed that though the OGS good performance with respect to sensitivity and linearity with reference gas standards, this in itself is not enough for the OGS to be a field reference standard because a closed path spectrometer has limitations due to losses to surfaces in sampling NH3, which need to be taken into account. Overall, the instruments studied performed well against the standard gases but we note that not every instrument could be calibrated using gas standards due to incompatible inlet designs and limitations in the gas flow rates of the standards. This work provides evidence that though NH3 instrumentation have greatly progressed in measurement precision, there is still further work required to quantify the accuracy of these systems under field conditions. It is the recommendation of this study that the use of instruments for routine monitoring of NH3 needs to be set out in standard operating protocols for inlet set-up, calibration and routine maintenance, in order for datasets to be comparable.
