The skin epidermis is continuously exposed to external aggressions, including environmental pollution. The cosmetic industry must be able to offer dedicated products to fight the effects of pollutants on the skin. We set up an experimental model that exposed skin explants maintained in culture to a pollutant mixture. This mixture P representing urban pollution was designed on the basis of the French organization ‘Air Parif’ database. A chamber, called Pollubox®, was built to allow a controlled nebulization of P on the cultured human skin explants. We investigated ultrastructural morphology by transmission electron microscopy of high pressure frozen skin explants. A global transcriptomic analysis indicated that the pollutant mixture was able to induce relevant xenobiotic and antioxidant responses. Modulated detoxifying genes were further investigated by laser micro-dissection coupled to qPCR, and immunochemistry. Both approaches showed that P exposure correlated with overexpression of detoxifying genes and provoked skin physiological alterations down to the stratum basale. The model developed herein might be an efficient tool to study the effects of pollutants on skin as well as a powerful testing method to evaluate the efficacy of cosmetic products against pollution.

International audience; Aphanomyces euteiches is an oomycete pathogen that causes the pea root rot. We investigated the potential role of early belowground defense in pea (susceptible plant) and faba bean (tolerant plant) at three days after inoculation. Pea and faba bean were inoculated with A. euteiches zoospores. Root colonization was examined. Root exudates from pea and faba bean were harvested and their impact on A. euteiches development were assessed by using in vitro assays. A. euteiches root colonization and the influence of the oomycete inoculation on specialized metabolites patterns and arabinogalactan protein (AGP) concentration of root exudates were also determined. In faba bean root, A. euteiches colonization was very low as compared with that of pea. Whereas infected pea root exudates have a positive chemotaxis index (CI) on zoospores, faba bean exudate CI was negative suggesting a repellent effect. While furanoacetylenic compounds were only detected in faba bean exudates, AGP concentration was specifically increased in pea.This work showed that early in the course of infection, host susceptibility to A. euteiches is involved via a plant-species specific root exudation opening new perspectives in pea root rot disease management.

International audience; Drought-induced dehydration of vegetative tissues in lycopods affects growth and survival. Different species of Selaginella have evolved a series of specialized mechanisms to tolerate desiccation in vegetative tissues in response to water stress. In the present study, we report on the structural characterization of the leaf cell wall of the desiccation-tolerant species S. involvens and two desiccation-sensitive species, namely S. kraussiana and S. moellendorffii. Isolated cell walls from hydrated and desiccated leaves of each species were fractionated and the resulting oligosaccharide fragments were analyzed to determine their structural features. Our results demonstrate that desiccation induces substantial modifications in the cell wall composition and structure. Altogether, these data highlight the fact that structural remodeling of cell wall hemicellulosic polysaccharides including XXXG-rich xyloglucan, arabinoxylan and acetylated galactomannan is an important process in order to mitigate desiccation stress in Selaginella.

International audience; exDNA is found in various organisms, including plants. However, plant exDNA has thus far received little attention related to its origin and role in the RET (root extracellular trap). In this study, we performed the first high-throughput genomic sequencing of plant exDNA from a Fabaceae with worldwide interest: soybean (Glycine max (L.) Merr.). The origin of this exDNA was first investigated in control condition, and the results show high-coverage on organelles (mitochondria/plastid) DNA relative to nuclear DNA, as well as a mix of coding and non-coding sequences. In the second part of this study, we investigated if exDNA release was modified during an elicitation with PEP-13 (a peptide elicitor from oomycete genus Phytophthora). Our results show that treatment of roots with PEP-13 does not affect the composition of exDNA.

Background: Agronomical and plant physiological studies employ a large variety of compounds that impact root development. The effect of these compounds is often evaluated by root architecture analyses using dedicated software. During the past decade, a growing number of tools proposed complex and deep analysis of the root system architecture. While these software applications are often complex and require specific set up, here, we propose a simple method based on the most common tools used by biologists: ImageJ and Excel®.Results: First, roots are measured manually with ImageJ following a succession of operations (i. choose a plant; ii. Measure: a. the primary root - b. lateral roots). Secondly, RootIX, an Excel file with dedicated macros that will automatically extract, sort and treat the data, is launched. Tables and graphics describing the data are generated. Root length, elongation rate and additional lateral roots are the main indicators provided among many others. We evaluated the effect of algae extracts on tomato roots growth with RootIX. We highlighted that these compounds negatively affect root growth while promoting generation of lateral roots.Conclusion: Ease of use and settings as well as complete data analyses without any cost are the scaffold of RootIX.

International audience; Plants are surrounded by a diverse range of microorganisms that causes serious crop losses and requires the use of pesticides. Flax is a major crop in Normandy used for its fibres and is regularly challenged by the pathogenic fungus Fusarium oxysporum (Fo) f. sp. lini. To protect themselves, plants use "innate immunity" as a first line of defense level against pathogens. Activation of plant defense with elicitors could be an alternative for crop plant protection. A previous work was conducted by screening a chemical library and led to the identification of compounds able to activate defense responses in Arabidopsis thaliana. Four compounds were tested for their abilities to improve resistance of two flax varieties against Fo. Two of them, one natural (holaphyllamine or HPA) and one synthetic (M4), neither affected flax nor Fo growth. HPA and M4 induced oxidative burst and callose deposition. Furthermore, HPA and M4 caused changes in the expression patterns of defense-related genes coding a glucanase and a chitinase-like. Finally, plants pre-treated with HPA or M4 exhibited a significant decrease in the disease symptoms. Together, these findings demonstrate that HPA and M4 are able to activate defense responses in flax and improve its resistance against Fo infection.

International audience; Root border cells (BCs) and their associated secretions form a protective structure termed the root extracellular trap (RET) that plays a major role in root interactions with soil borne microorganisms. In this study, we investigated the release and morphology of BCs of Glycine max using light and cryo-scanning electron microscopy (SEM). We also examined the occurrence of cell-wall glycomolecules in BCs and secreted mucilage using immunofluorescence microscopy in conjunction with anti-glycan antibodies. Our data show that root tips released three populations of BCs defined as spherical, intermediate and elongated cells. The mechanism of shedding seemed to be cell morphotype-specific. The data also show that mucilage contained pectin, cellulose, extracellular DNA, histones and two hemicellulosic polysaccharides, xyloglucan and heteromannan. The latter has never been reported previously in any plant root secretions. Both hemicellulosic polysaccharides formed a dense fibrillary network embedding BCs and holding them together within the mucilage. Finally, we investigated the effect of the RET on the interactions of root with the pathogenic oomycete Phytophthora parasitica early during infection. Our findings reveal that the RET prevented zoospores from colonizing root tips by blocking their entry into root tissues and inducing their lysis.

Plant derived arabinogalactan proteins (AGP) were repeatedly confirmed as immunologically as well as dermatologically active compounds. However, little is currently known regarding their potential activity toward skin innate immunity. Here, we extracted and purified AGP from acacia (Acacia senegal) and baobab (Adansonia digitata) seeds to investigate their biological effects on the HaCaT keratinocyte cell line in an in vitro system. While AGP from both sources did not exhibit any cytotoxic effect, AGP from acacia seeds enhanced cell viability. Moreover, real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analysis showed that AGP extracted from both species induced a substantial overexpression of hBD-2, TLR-5, and IL1-α genes. These data suggest that plant AGP, already known to control plant defensive processes, could also modulate skin innate immune responses. J. Cell. International audience; Pea (Pisum sativum) root cap releases a large number of living border cells that secrete abundant mucilage into the extracellular medium. Mucilage contains a complex mixture of polysaccharides, proteins and secondary metabolites important for its structure and function in defense. Unlike xyloglucan and cellulose, pectin and arabinogalactan proteins have been investigated in pea root and shown to be major components of border cell walls and mucilage. In this study, we investigated the occurrence of xyloglucan and cellulose in pea border cells and mucilage using cytochemical staining, immunocytochemistry and laser scanning confocal microscopy. Our data show that i) unlike cellulose, xyloglucan is highly present in the released mucilage as a dense fibrillary network enclosing border cells and ii) that xyloglucan and cellulose form molecular cross-bridges that tether cells and maintain them attached together. These findings suggest that secreted xyloglucan is essential for mucilage strengthening and border cell attachment and functioning.

International audience; It is well recognized that the world population is ageing rapidly. Therefore, it is important to understand ageing processes at the cellular and molecular levels to predict the onset of age‐related diseases and prevent them. Recent research has focused on the identification of ageing biomarkers, including those associated with the properties of the Golgi apparatus. In this context, Golgi‐mediated glycosylation of proteins has been well characterized. Additionally, other studies show that the secretion of many compounds, including pro‐inflammatory cytokines and extracellular matrix–degrading enzymes, is modified during ageing, resulting in physical and functional skin degradation. Since the Golgi apparatus is a central organelle of the secretory pathway, we investigated its structural organization in senescent primary human dermal fibroblasts using confocal and electron microscopy. In addition, we monitored the expression of Golgi‐related genes in the same cells. Our data showed a marked alteration in the Golgi morphology during replicative senescence. In contrast to its small and compact structure in non‐senescent cells, the Golgi apparatus exhibited a large and expanded morphology in senescent fibroblasts. Our data also demonstrated that the expression of many genes related to Golgi structural integrity and function was significantly modified in senescent cells, suggesting a relationship between Golgi apparatus function and ageing.

International audience; The root cap releases cells that produce massive amounts of mucilage containing polysaccharides, proteoglycans, extracellular DNA (exDNA) and a variety of antimicrobial compounds. The released cells – known as border cells or border‐like cells – and mucilage secretions form networks that are defined as root extracellular traps (RETs). RETs are important players in root immunity. In animals, phagocytes are some of the most abundant white blood cells in circulation and are very important for immunity. These cells combat pathogens through multiple defence mechanisms, including the release of exDNA‐containing extracellular traps (ETs). Traps of neutrophil origin are abbreviated herein as NETs. Similar to phagocytes, plant root cap‐originating cells actively contribute to frontline defence against pathogens. RETs and NETs are thus components of the plant and animal immune systems, respectively, that exhibit similar compositional and functional properties. Herein, we describe and discuss the formation, molecular composition and functional similarities of these similar but different extracellular traps.

International audience; Extensins are cell wall glycoproteins, belonging to the Hydroxyproline-Rich GlycoProtein (HRGP) family, which are involved in many biological functions, including plant growth and defence. Several reviews have described the involvement of HRGPs in plant immunity but little focus has been given specifically to cell wall extensins. Yet, a large set of recently published data indicates that extensins play an important role in plant protection, especially in root-microbe interactions. Here, we summarize the current knowledge on this topic and discuss the importance of extensins in root defence. We first provide an overview of the distribution of extensin epitopes recognised by different monoclonal antibodies among plants and discuss the relevance of some of these epitopes as markers of the root defence response. We also highlight the implication of extensins in different types of plant interactions elicited by either pathogenic or beneficial microorganisms. We then present and discuss the specific importance of extensins in root secretions as these glycoproteins are not only found in the cell walls but are also released into the root mucilage. Finally, we propose a model to illustrate the impact of cell wall extensin on root secretions.

Glycosylation is the most common posttranslational modification of proteins and consists of the addition of sugar moiety to proteins. The resulting glycosylated proteins are often secreted to the extracellular compartment or integrated into different cell organelles. This modification was identified in plant as well as in mammalian cells. A number of plant and mammal proteins are either N- or O-glycosylated. This review focuses on O-glycosylation which refers to linkage of a glycan to hydroxyl group of serine, threonine or proline residues. O-glycosylation can be altered by the action of chemical inhibitors. For instance, 3,4-dehydro-L-proline, ethyl 3,4-dehydroxy benzoate and a,a-dipyridyl inhibit the activity of prolyl4-hydroxylase, a key enzyme for plant O-glycosylation. In addition, a small molecule inhibitor designated 1-68A inhibits the polypeptide N-acetylgalactosaminyltransferases of mammalian cells. The aim of this review is to summarize the role and mechanism of action of these inhibitors of O-glycosylation and their impact on cell development in plants and mammals.

Summary Xyloglucan is the dominant hemicellulosic polysaccharide of the primary cell wall of dicotyledonous plants that plays a key role in plant development. It is well established that xyloglucan is assembled within Golgi stacks and transported in Golgi-derived vesicles to the cell wall. It is also known that the biosynthesis of xyloglucan requires the action of glycosyltransferases including α-1,6-xylosyltransferase, β-1,2-galactosyltransferase and α-1,2-fucosyltransferase activities responsible for the addition of xylose, galactose and fucose residues to the side chains. There is, however, a lack of knowledge on how these enzymes are distributed within subcompartments of Golgi stacks. We have undertaken a study aiming at mapping these glycosyltransferases within Golgi stacks using immunogold-electron microscopy. To this end, we generated transgenic lines of tobacco (Nicotiana tabacum) BY-2 suspension-cultured cells expressing either the α-1,6-xylosyltransferase, AtXT1, the β-1,2-galactosyltransferase, AtMUR3, or the α-1,2-fucosyltransferase AtFUT1 of Arabidopsis thaliana fused to green-fluorescent protein (GFP). Localization of the fusion proteins within the endomembrane system was assessed using confocal microscopy. Additionally, tobacco cells were high pressure-frozen/freeze-substituted and subjected to quantitative immunogold labelling using anti-GFP antibodies to determine the localization patterns of the enzymes within subtypes of Golgi cisternae. The data demonstrate that: (i) all fusion proteins, AtXT1–GFP, AtMUR3–GFP and AtFUT1–GFP are specifically targeted to the Golgi apparatus; and (ii) AtXT1–GFP is mainly located in the cis and medial cisternae, AtMUR3–GFP is predominantly associated with medial cisternae and AtFUT1–GFP mostly detected over trans cisternae suggesting that initiation of xyloglucan side chains occurs in early Golgi compartments in tobacco cells.

The processing of N-linked oligosaccharides in the secretory pathway requires the sequential action of a number of glycosidases and glycosyltransferases. We studied the spatial distribution of several type II membrane-bound enzymes from Glycine max, Arabidopsis thaliana, and Nicotiana tabacum. Glucosidase I (GCSI) localized to the endoplasmic reticulum (ER), α-1,2 mannosidase I (ManI) and N-acetylglucosaminyltransferase I (GNTI) both targeted to the ER and Golgi, and β-1,2 xylosyltransferase localized exclusively to Golgi stacks, corresponding to the order of expected function. ManI deletion constructs revealed that the ManI transmembrane domain (TMD) contains all necessary targeting information. Likewise, GNTI truncations showed that this could apply to other type II enzymes. A green fluorescent protein chimera with ManI TMD, lengthened by duplicating its last seven amino acids, localized exclusively to the Golgi and colocalized with a trans-Golgi marker (ST52-mRFP), suggesting roles for protein–lipid interactions in ManI targeting. However, the TMD lengths of other plant glycosylation enzymes indicate that this mechanism cannot apply to all enzymes in the pathway. In fact, removal of the first 11 amino acids of the GCSI cytoplasmic tail resulted in relocalization from the ER to the Golgi, suggesting a targeting mechanism relying on protein–protein interactions. We conclude that the localization of N-glycan processing enzymes corresponds to an assembly line in the early secretory pathway and depends on both TMD length and signals in the cytoplasmic tail.

Plant pathogens including fungi and bacteria cause many of the most serious crop diseases. The plant innate immune response is triggered upon recognition of microbe-associated molecular patterns (MAMPs) such as flagellin22 and peptidoglycan. To date, very little is known of MAMP-mediated responses in roots. Root border cells are cells that originate from root caps and are released individually into the rhizosphere. Root tips of Arabidopsis (Arabidopsis thaliana) and flax (Linum usitatissimum) release cells known as “border-like cells.” Whereas root border cells of pea (Pisum sativum) are clearly involved in defense against fungal pathogens, the function of border-like cells remains to be established. In this study, we have investigated the responses of root border-like cells of Arabidopsis and flax to flagellin22 and peptidoglycan. We found that both MAMPs triggered a rapid oxidative burst in root border-like cells of both species. The production of reactive oxygen species was accompanied by modifications in the cell wall distribution of extensin epitopes. Extensins are hydroxyproline-rich glycoproteins that can be cross linked by hydrogen peroxide to enhance the mechanical strength of the cell wall. In addition, both MAMPs also caused deposition of callose, a well-known marker of MAMP-elicited defense. Furthermore, flagellin22 induced the overexpression of genes involved in the plant immune response in root border-like cells of Arabidopsis. Our findings demonstrate that root border-like cells of flax and Arabidopsis are able to perceive an elicitation and activate defense responses. We also show that cell wall extensin is involved in the innate immunity response of root border-like cells.

The development of pectin structural features during the differentiation of cambial derivatives was investigated in aspen (Populus tremula L. x P. tremuloides Michx.) using biochemical and immunocytochemical methods. Comparisons were also made between active and resting tissues. Active tissues, in particular cambial cells and phloem derivatives, were characterized by a high pectin content. Use of antibodies raised against arabinan side chains of rhamnogalacturonan 1 (LM6), as well as biochemical analysis, revealed an obvious decrease from the cortex to the differentiating xylem. Galactan side chains, detected with LM5 antibodies, were present mainly in the cambial zone and enlarging xylem cells. In contrast, they were totally absent from sieve-tube cell walls. Image analysis of LM5 immunogold labelling in the cambial zone showed a clustered distribution of galactan epitopes in the radial walls, a distribution which might result from the association of two different periodic processes, namely the exocytosis of galactan and wall expansion. Cessation of cambial activity was characterized by cell wall thickening accompanied by a sharp decrease in the relative amount of pectin and a lowering of the degree of methylesterification. The data provide evidence that the walls of phloem and xylem cells differ in their pectin composition even at a very early stage of commitment. These differences offer useful tools for identifying the initial cells among their immediate neighbours.

Using SIMS microscopy an unexpected ionic interaction between calcium and sodium has been characterised within the walls of the xylem cells and of the phloem fibres of hypocotyls of beech seedlings: whereas a decrease of the Na signal in the cell walls was always paralleled by an increase of the calcium signal at the same site, unexpectedly an increase of the normalised Na signal in the cell walls of the Naenriched seedlings was paralleled by an increase of the Ca signal. We have also shown that this interaction was involved in the differentiation of the secondary walls.

The skin is a natural barrier between the body and the environment and is colonised by a large number of microorganisms. Here, we report a complete analysis of the response of human skin explants to microbial stimuli. Using this ex vivo model, we analysed at both the gene and protein level the response of epidermal cells to Staphylococcus epidermidis (S. epidermidis) and Pseudomonas fluorescens (P. fluorescens), which are present in the cutaneous microbiota. We showed that both bacterial species affect the structure of skin explants without penetrating the living epidermis. We showed by real-time quantitative polymerase chain reaction (qPCR) that S. epidermidis and P. fluorescens increased the levels of transcripts that encode antimicrobial peptides (AMPs), including human b defensin (hBD)2 and hBD3, and the pro-inflammatory cytokines interleukin (IL)-1a and (IL)-1-b, as well as IL-6. In addition, we analysed the effects of bacterial stimuli on the expression profiles of genes related to innate immunity and the inflammatory response across the epidermal layers, using laser capture microdissection (LCM) coupled to qPCR. We showed that AMP transcripts were principally upregulated in suprabasal keratinocytes. Conversely, the expression of pro-inflammatory cytokines was upregulated in the lower epidermis. These findings were confirmed by protein localisation using specific antibodies coupled to optical or electron microscopy. This work underscores the potential value of further studies that use LCM on human skin explants model to study the roles and effects of the epidermal microbiota on human skin physiology.

Border cells and border-like cells are released from the root tip as individual cells and small aggregates, or as a group of attached cells. These are viable components of the root system that play a key role in controlling root interaction with living microbes of the rhizosphere. As their separation from root tip proceeds, the cells synthesize and secrete a hydrated mucilage that contains polysaccharides, secondary metabolites, antimicrobial proteins and extracellular DNA (exDNA). This exDNA-based matrix seems to function in root defense in a way similar to that of recently characterized neutrophil extracellular traps (NETs) in mammalian cells. This review discusses the role of the cells and secreted compounds in the protection of root tip against microbial infections.

Root tips of many plant species release a number of border, or border-like, cells that are thought to play a major role in the protection of root meristem. However, little is currently known on the structure and function of the cell wall components of such root cells. Here, we investigate the sugar composition of the cell wall of the root cap in two species: pea (Pisum sativum), which makes border cells, and Brassica napus, which makes border-like cells. We find that the cell walls are highly enriched in arabinose and galactose, two major residues of arabinogalactan proteins. We confirm the presence of arabinogalactan protein epitopes on root cap cell walls using immunofluorescence microscopy. We then focused on these proteoglycans by analyzing their carbohydrate moieties, linkages, and electrophoretic characteristics. The data reveal (1) significant structural differences between B. napus and pea root cap arabinogalactan proteins and (2) a cross-link between these proteoglycans and pectic polysaccharides. Finally, we assessed the impact of root cap arabinogalactan proteins on the behavior of zoospores of Aphanomyces euteiches, an oomycetous pathogen of pea roots. We find that although the arabinogalactan proteins of both species induce encystment and prevent germination, the effects of both species are similar. However, the arabinogalactan protein fraction from pea attracts zoospores far more effectively than that from B. napus. This suggests that root arabinogalactan proteins are involved in the control of early infection of roots and highlights a novel role for these proteoglycans in root-microbe interactions.

We describe, for the first time, an efficient protocol based on laser capture microdissection (LCM) for the isolation of human epidermal layers for gene expression profiling using quantitative real-time PCR. Two areas enriched either in basal or granular layers were isolated by LCM. Skin biopsies were fixed in dry ice-cooled isopentane, cryosectioned and stained before the laser procedure. High-quality total RNA was extracted from each microdissected sample, which allowed the analysis of the spatial distribution of mRNA transcripts from 10 innate immunity-related genes within the epidermal layers. Using integrin alpha-6/integrin beta-4 and corneodesmosin/filaggrin-2 sets as gene markers for the basal and granular layers, respectively, we showed that Toll-like receptor 2, RNase 7, human beta-defensin-2 and -3, psoriasin and nucleotide-binding oligomerization domain 1 are upregulated in the suprabasal layer of normal human epidermis. Our protocol, which is based on the rapid isolation of epidermal layers, can be used to follow transcriptional processes in specific areas of the epidermis and is a very promising tool to use in the study of numerous aspects of dermatology.

International audience; The Golgi apparatus of eukaryotic cells is known for its central role in the processing, sorting , and transport of proteins to intra-and extra-cellular compartments. In plants, it has the additional task of assembling and exporting the non-cellulosic polysaccharides of the cell wall matrix including pectin and hemicelluloses, which are important for plant development and protection. In this review, we focus on the biosynthesis of complex polysaccharides of the primary cell wall of eudicotyledonous plants. We present and discuss the compartmen-tal organization of the Golgi stacks with regards to complex polysaccharide assembly and secretion using immuno-electron microscopy and specific antibodies recognizing various sugar epitopes. We also discuss the significance of the recently identified Golgi-localized glycosyltransferases responsible for the biosynthesis of xyloglucan (XyG) and pectin.

Plant cells are surrounded by a carbohydrate-rich compartment called the cell wall. This compartment plays a central role in growth, development, sexual reproduction and defence (Vicre et al., 2005; Mollet et al., 2007; Hematy et al., 2009; Seifert & Blaukopf, 2010). It is a carbohydrate-based structure composed mostly of polysaccharides (~90%) and glycoproteins (~10%). Polysaccharides include cellulose, pectin domains such as homogalacturonans (HG) with different levels of methylesterification, xylogalacturonan, rhamnogalacturonan-I (RG-I) and rhamnogalacturonanII (RG-II) and hemicelluloses (depending on the species, xyloglucan [XyG] or arabinoxylan) (Liepman et al., 2010), whereas cell wall proteins are either enzymatic such as glucanases (Gilbert, 2010) or nonenzymatic such as arabinogalactan proteins (AGP) (Ellis et al., 2010, Driouich & Baskin, 2008).

Production and release of root border cells and border-like cells are fundamental processes for plant survival and development. Both types of cells are viable components of the root system that regulate its interactions with living microorganisms of the rhizosphere. Border cells are released as individual cells, whereas border-like cells remain attached to each other into small groups or as sheets after their release from the root tip. So far, border-like cells have been observed only in species belonging to the Brassicaceae family including Arabidopsis. Border cells have been largely studied in the legume species pea; in contrast, relatively little information is available on border-like cells so far due to their recent discovery. In this chapter, we present and discuss the release, organization, and the role of these cells in root protection.

Xyloglucan is the dominant hemicellulosic polysaccharide of the primary cell wall of dicotyledonous plants that plays a key role in plant development. It is well established that xyloglucan is assembled within Golgi stacks and transported in Golgi-derived vesicles to the cell wall. It is also known that the biosynthesis of xyloglucan requires the action of glycosyltransferases including α-1,6-xylosyltransferase, β-1,2-galactosyltransferase and α-1,2-fucosyltransferase activities responsible for the addition of xylose, galactose and fucose residues to the side chains. There is, however, a lack of knowledge on how these enzymes are distributed within subcompartments of Golgi stacks. We have undertaken a study aiming at mapping these glycosyltransferases within Golgi stacks using immunogold-electron microscopy. To this end, we generated transgenic lines of tobacco (Nicotiana tabacum) BY-2 suspension-cultured cells expressing either the α-1,6-xylosyltransferase, AtXT1, the β-1,2-galactosyltransferase, AtMUR3, or the α-1,2-fucosyltransferase AtFUT1 of Arabidopsis thaliana fused to green-fluorescent protein (GFP). Localization of the fusion proteins within the endomembrane system was assessed using confocal microscopy. Additionally, tobacco cells were high pressure-frozen/freeze-substituted and subjected to quantitative immunogold labelling using anti-GFP antibodies to determine the localization patterns of the enzymes within subtypes of Golgi cisternae. The data demonstrate that: (i) all fusion proteins, AtXT1-GFP, AtMUR3-GFP and AtFUT1-GFP are specifically targeted to the Golgi apparatus; and (ii) AtXT1-GFP is mainly located in the cis and medial cisternae, AtMUR3-GFP is predominantly associated with medial cisternae and AtFUT1-GFP mostly detected over trans cisternae suggesting that initiation of xyloglucan side chains occurs in early Golgi compartments in tobacco cells.

International audience; Border-like cells are released by Arabidopsis (Arabidopsis thaliana) root tips as organized layers of several cells that remain attached to each other rather than completely detached from each other, as is usually observed in border cells of many species. Unlike border cells, cell attachment between border-like cells is maintained after their release into the external environment. To investigate the role of cell wall polysaccharides in the attachment and organization of border-like cells, we have examined their release in several well-characterized mutants defective in the biosynthesis of xyloglucan, cellulose, or pectin. Our data show that among all mutants examined, only quasimodo mutants (qua1-1 and qua2-1), which have been characterized as producing less homogalacturonan, had an altered border-like cell phenotype as compared with the wild type. Border-like cells in both lines were released as isolated cells separated from each other, with the phenotype being much more pronounced in qua1-1 than in qua2-1. Further analysis of border-like cells in the qua1-1 mutant using immunocytochemistry and a set of anti-cell wall polysaccharide antibodies showed that the loss of the wild-type phenotype was accompanied by (1) a reduction in homogalacturonan-JIM5 epitope in the cell wall of border-like cells, confirmed by Fourier transform infrared microspectrometry, and (2) the secretion of an abundant mucilage that is enriched in xylogalacturonan and arabinogalactan-protein epitopes, in which the cells are trapped in the vicinity of the root tip.

Characterization of a beta1,2-xylosyltransferase from Arabidopsis thaliana (AtXylT) was carried out by expression in Sf9 insect cells using a baculovirus vector system. Serial deletions at both the N- and C-terminal ends proved that integrity of a large domain located between amino acid 31 and the C-terminal lumenal region is required for AtXylT activity expression. The influence of N-glycosylation on AtXylT activity has been evaluated using either tunicamycin or mutagenesis of potential N-glycosylation sites. AtXylT is glycosylated on two of its three potential N-glycosylation sites (Asn51, Asn301, Asn478) and the occupancy of at least one of these two sites (Asn51 and Asn301) is necessary for AtXylT stability and activity. Contribution of the N-terminal part of AtXylT in targeting and intracellular distribution of this protein was studied by expression of variably truncated, GFP-tagged AtXylT forms in tobacco cells using confocal and electron microscopy. These studies have shown that the transmembrane domain of AtXylT and its short flanking amino acid sequences are sufficient to specifically localize a reporter protein to the medial Golgi cisternae in tobacco cells. This study is the first detailed characterization of a plant glycosyltransferase at the molecular level.

In plant systems, the green fluorescent protein (GFP) is increasingly used as a marker to study dynamics of the secretory apparatus using fluorescence microscopy. The purpose of this study was to immunogold localize the GFP, at the electron microscopic level, in a line of tobacco BY-2-cultured cells, expressing a GFP-tagged Golgi glycosyltransferase. To this end we have developed a simple, one-step chemical fixation method that allow good structural preservation and specific labeling with anti-GFP antibodies. Using this method, we have been able to show that an N-glycan GFP-tagged xylosyltransferase is specifically associated with Golgi stacks of BY-2 transformed cells and is preferentially located in medial cisternae. As an alternative to cryofixation methods, such as high-pressure freezing, which requires specialized and expensive equipment not available in most laboratories, this method offers researchers the opportunity to investigate GFP-tagged proteins of the endomembrane system in tobacco BY-2 cells.

Using secondary ion mass spectrometry (SIMS), calcium was imaged in cambium cells and in the adjacent secondary phloem and xylem cells during the different phases of cambium functioning in beech (Fagus sylvatica L.). At the end of the period of quiescence, immediately before the resumption of cell divisions (i.e. at the cambium pre-activation phase), a strong temporary increase of calcium concentration was observed to take place in cambium and phloem but not in xylem cells.

The root cap is primordial for seedling growth and supports root apical meristem integrity. The root cap releases root border cells (RBCs) that surround the root tip and ensure seedling protection against numerous stresses. RBCs and their associated mucilage form the root extracellular trap (RET). Here, RET and root exudate characterization of three Sahelian woody seedlings are performed. In contrast to B. aegyptiaca and A. raddiana which thrive in semi-arid areas, T. indica is more sensitive to drought. B. aegyptiaca, A. raddiana and T. indica seedlings were sub-cultured in vitro. RBC morphologies and RET organization were determined using microscopic approaches. The polysaccharide composition and arabinogalactan protein (AGP) content were determined by biochemical approaches in the RET and the root exudates. Moreover, the effect of root exudates on the growth of Azospirillum brasilense a plant benefical bacteria has been performed. While B. aegyptiaca produces only border cell (BC) type, the two Fabaceae seedlings release both BCs and border-like cells (BLCs). BCs are enclosed in a dense mucilage enriched in cell wall polymers. Compared to T. indica, RET and root exudates of B. aegyptiaca and A. raddiana include more abundant AGPs. In this context, AGPs could contribute to woody seedling survival. This work opens new research perspectives regarding involvement of RET in plant survival to aridity., La coiffe racinaire est cruciale à la croissance et survie du méristème subapical de racine. Elle libère des cellules frontières (CFs) quiassurent la protection de l’apex racinaire. Les CFs associées à leur mucilage forment le piège extracellaire de racine (RET). La caractérisation du RET et des exsudats racinaires de trois essences ligneuses sahéliennes à partir de plantules cultivées in vitro a été réalisée. B. aegyptiaca et A. raddiana prospèrent dans les zones semi-arides, à l’opposé de T. indica. La morphologie des CFs et l’organisation du RET ont été déterminées par microscopie. La compostion en glycopolymères et la détection des arabinogalactanes proteines (AGPs) dans le RET et les exsudats racinaires ont été déterminées par des analyses biochimiques. L’effet des exsudats racinaires sur la croissance d’Azospirillum brasilense, une bactérie bénéfique pour la plante a été évalué. B. aegyptiaca produit des CFs de type border cells (BCs) alors que les autres Fabaceae produisent des BCs et des border-like cells. Les BCs sont entourées d’un dense mucilage riche en polymères de paroi. Le RET et les exsudats racinaires issus de B. aegyptiaca et A. raddiana sont plus riches en AGPs que ceux provenant T. indica. Les AGPs pourraient contribuer à la survie des plantules dans un contexte semi-aride. Ce travail ouvre de nouvelles perspectives de recherche concernant l'implication du RET dans la survie des plantes à l'aridité.

Primary cell wall of dicotyledonous plants is composed of cellulose microfibrils, proteins, and complex polysaccharides represented by hemicelluloses and pectins. In Arabidopsis thaliana plant model, xyloglucan (XyG) and rhamnogalacturonan-I (RG-I) represent the main hemicelluloses and pectins polysaccharides respectively. Before cell wall transport, XyG and RG-I are synthesized in the Golgi apparatus, particularly via sugar interconversion enzymes and glycosyltransferases, which will generate and assemble the different sugars to form polysaccharides.The aim of this research project was to localize in Golgi subcompartments different cell wall complex polysaccharides epitopes and enzymes implicated in XyG and RG-I synthesis, as well as enzymatic complexes research, using confocal laser scanning microscopy, transmission electron microscopy and quantitative immunocytochemistry., La paroi primaire des plantes des dicotylédones vraies est constituée de microfibrilles de cellulose, de protéines et de polysaccharides complexes, représentés par les hémicelluloses et les pectines. Chez la plante modèle Arabidopsis thaliana, le xyloglucane (XyG) et le rhamnogalacturonane-I (RG-I) constituent les catégories majeures des hémicelluloses et des pectines respectivement. Avant d’être transportés vers la paroi, le XyG et le RG-I sont synthétisés au sein de l’appareil de Golgi, notamment via l’action d’enzymes d’interconversion des sucres et de glycosyltransférases, qui vont générer et assembler les différents sucres pour former les polysaccharides. L’objectif de ce projet de recherche a été d’effectuer la cartographie golgienne d’épitopes polysaccharidiques et de différentes enzymes impliquées dans la synthèse du XyG et du RG-I, ainsi que de rechercher l’existence de complexes enzymatiques, en utilisant les techniques de microscopie confocale à balayage laser, de microscopie électronique à transmission et d’immunocytochimie quantitative.

Résumé. Interface entre l’organisme et l’environnement, la peau est le plus grand organe qui assure une fonction « barrière » vis-à-vis des stress biotiques ou abiotiques. Elle est constituée de trois couches cellulaires superposées: l’hypoderme, le derme, et l’épiderme. L’épiderme renferme également quatre couches cellulaires distinctes: la couche basale, la couche épineuse, la couche granuleuse et la couche cornée. Les kératinocytes issus de la couche basale, constituent le type cellulaire le plus abondant au sein de l’épiderme. Dans ce projet, nous avons analysé l’expression de nombreux gènes (gènes structuraux et gènes de défense) par PCR quantitative en temps réel, au sein des différentes couches épidermiques isolées d’explant de peau humaine saine par microdissection laser (Laser Capture Microdissection, LCM). Nous avons ensuite évalué les effets de bactéries du microbiome cutané sur l’expression de gènes de défense. La distribution des produits de ces gènes a également été examinée par une approche immunocytochimique. Les résultats majeurs obtenus sont décrits ci-après. 1) Nous avons optimisé le protocole de LCM pour isoler deux fractions épidermiques respectivement enrichies en kératinocytes de la couche basale et de la couche granuleuse (Percoco et al., 2012. Experimental Dermatology 21, 531-534). Cet outil prometteur est maintenant aisément applicable à la recherche en Dermatologie. 2) Nous avons montré, que dans une peau humaine saine, les gènes responsables de l’immunité innée, codant notamment pour les bêta-defensines humaines de type 2 et 3 (human beta-defensin, hBD), sont exprimés de façon différentielle au sein des couches épidermiques. 3) Nous avons observé que les trois bactéries Staphylococcus epidermidis, Staphylococcus aureus et Pseudomonas fluorescens sont capables de moduler positivement l’expression des gènes codant pour des beta-defensines (hBD2 et hBD3) et des cytokines pro-inflammatoires (IL-1 alpha et bêta et IL-6). 4) En utilisant des anticorps spécifiques, nous avons localisé les produits de ces gènes dans les différentes couches épidermiques, à l’échelle cellulaire par microscopie à épifluorescence, et à l’échelle ultrastructurale par microscopie électronique à transmission (Percoco et al., Soumis).; The skin is the largest organ of the body. It is a natural barrier between the environment and the organism that plays a key role in the protection of the body against abiotic or biotic stress. The skin is made up of three different cell layers, namely the hypodermis, the dermis and the epidermis. The epidermis is, in turn, composed of four different layers, the basal, the spinous, the granular and the horny layer. In the epidermis, keratinocytes is the major cell type known to originate from basal cells. In the present work, we have studied the expression of a number of genes (antimicrobial peptide genes and pro-inflammatory cytokine genes) in different layers of healthy human epidermis using quantitative real-time PCR coupled to Laser Capture Microdissection (LCM). Using the same approach, we have also investigated the effect of bacteria belonging the human epidermal microbiota on the expression of the same genes within the epidermal layers. In addition, the spatial distribution of gene products has also been examined using immunocytochemsitry. The major findings are: 1) We have developed an optimized LCM protocol for the isolation of human epidermal layers. We have been able to isolate two fractions highly enriched in keratinocytes from the basal and the granular layers (Percoco et al. 2012. Experimental Dermatology. 21, 531-534). Such a method can now be easily applied in dermatological research. 2) We have found that, in healthy human epidermis, innate immunity-related genes including human beta-defensin genes are differentially expressed within different layers. 3) We have found that the three bacteria Staphylococcus epidermidis, Staphylococcus aureus and Pseudomonas fluorescens induce a significant increase in the expression of beta-defensin (hBD2 and hBD3) and pro-inflammatory cytokines (IL-1 alpha, IL-1 beta and IL-6) genes. 4) We have localized the gene products within different cell layers using specific antibodies in conjunction with immunofluorescence and immunogold-electron microscopy (Percoco et al., Submitted).
