4. How can plant modelling be a leverage for cropping system improvement by integrating plant physiology and smart horticulture?

Abstract

The main objective of the workshop “How can plant modelling be a leverage for cropping system improvement by integrating plant physiology and smart horticulture?”, developed within the frame of IHC2022, was to promote a discussion on the utility of plant modelling in the management of horticultural crops. A better understanding of the physiological response of crops to interaction with environmental factors (temperature, light, CO2, water and nutrients) and crop management (soil, irrigation, phytosanitary treatments) would allow to improve crop growth and production (fruit quality and/or yield), thereby saving resources that are becoming ever scarcer. Modelling is becoming more and more important for horticulture in the broadest sense, both to help advancing innovation and for a better understanding of the functioning of existing systems. This workshop was a great opportunity to create a platform for exchange between researchers working in different areas such as process-based models (PBM), Functional–structural plant models (FSPM) and greenhouse climate models (GCM). Interaction of different models is necessary to analyze the spatial and temporal distribution of crop production. This workshop aimed to establish interactions between the different areas of modeling to obtain decision support tools for plant production in smart horticulture. A panel consisting of four researchers (two invited senior researchers, and the animators of the workshop) promoted an open discussion with the participants, on their views and experiences about the integration of plant physiology and smart horticulture. The workshop allowed an interdisciplinary discussion between scientists to identify the potential roles and new research directions of plant modelling. In this paper some points of common interest for all scientists working with crop models are presented based on the results of the discussion in the workshop and in published papers.

5. Nutrient deficiency symptoms and uptake relations in juvenile hazelnut (Corylus avellana) in response to macronutrient supply

Abstract

The characterization of macronutrient (N, P, K, Ca, Mg, S) deficiencies in Corylus avellana is poorly documented, whereas early detection of nutrient shortages would help to prevent yield and quality reductions in hazelnut production. In a four-month greenhouse trial, nutrient deficiencies of macro-elements were induced by growing one-year-old ‘Tonda di Giffoni’ plants in 5-L containers with perlite with macronutrients individually applied at 0, 50 and 100% reduction in the supply concentration of a standard nutrient solution. Except for P, all nutrient disorders were visually induced in the zero treatments, after four (N and K) to six weeks (Ca, Mg, S). In the 50% treatments, deficiency symptoms appeared only for N, Ca, and S. The zero treatments of N, P, K, and S resulted in a significant reduction in biomass, whereas in the 50% treatments, growth was reduced significantly for N only. The nutrient contents in stem and foliage tissue demonstrated positive relationships with the nutrient concentrations applied at the root zone. Despite the clear response of biomass and tissue content to the P-supply, the lack of visible symptoms of P-deficiency was remarkable. The quantity of P in the initial plant was likely sufficient as a P-source, resulting in slight growth reduction but without initiating symptoms in this short growing cycle. Nevertheless, the average P contents in the final harvested leaves were extremely low (25 mmol kg-1 DM) compared to standard values mentioned in literature (50 mmol kg-1 DM).

Abstract

CONTEXT.—: SARS-CoV-2 can undergo maternal-fetal transmission, heightening interest in the placental pathology findings from this infection. Transplacental SARS-CoV-2 transmission is typically accompanied by chronic histiocytic intervillositis together with necrosis and positivity of syncytiotrophoblast for SARS-CoV-2. Hofbauer cells are placental macrophages that have been involved in viral diseases, including HIV and Zika virus, but their involvement in SARS-CoV-2 is unknown. OBJECTIVE.—: To determine whether SARS-CoV-2 can extend beyond the syncytiotrophoblast to enter Hofbauer cells, endothelium, and other villous stromal cells in infected placentas of liveborn and stillborn infants. DESIGN.—: Case-based retrospective analysis by 29 perinatal and molecular pathology specialists of placental findings from a preselected cohort of 22 SARS-CoV-2-infected placentas delivered to pregnant women testing positive for SARS-CoV-2 from 7 countries. Molecular pathology methods were used to investigate viral involvement of Hofbauer cells, villous capillary endothelium, syncytiotrophoblast, and other fetal-derived cells. RESULTS.—: Chronic histiocytic intervillositis and trophoblast necrosis were present in all 22 placentas (100%). SARS-CoV-2 was identified in Hofbauer cells from 4 of 22 placentas (18.2%). Villous capillary endothelial staining was positive in 2 of 22 cases (9.1%), both of which also had viral positivity in Hofbauer cells. Syncytiotrophoblast staining occurred in 21 of 22 placentas (95.5%). Hofbauer cell hyperplasia was present in 3 of 22 placentas (13.6%). In the 7 cases having documented transplacental infection of the fetus, 2 (28.6%) occurred in placentas with Hofbauer cell staining positive for SARS-CoV-2. CONCLUSIONS.—: SARS-CoV-2 can extend beyond the trophoblast into the villous stroma, involving Hofbauer cells and capillary endothelial cells, in a small number of infected placentas. 15. Consequences of intra-canopy and top LED lighting for uniformity of light distribution in a tomato crop.

Abstract

In the past decade, the potential of positioning LED lamps in between the canopy (intra-canopy) to enhance crop growth and yield has been explored in greenhouse cultivation. Changes in spatial heterogeneity of light absorption that come with the introduction of intra-canopy lighting have not been thoroughly explored. We calibrated and validated an existing functional structural plant model (FSPM), which combines plant morphology with a ray tracing model to estimate light absorption at leaflet level. This FSPM was used to visualize the light environment in a tomato crop illuminated with intra-canopy lighting, top lighting or a combination of both. Model validation of light absorption of individual leaves showed a good fit (R² = 0.93) between measured and modelled light absorption of the canopy. Canopy light distribution was then quantified and visualized in three voxel directions by means of average absorbed photosynthetic photon flux density (PPFD) and coefficient of variation (CV) within that voxel. Simulations showed that the variation coefficient within horizontal direction was higher for intracanopy lighting than top lighting (CV=48% versus CV= 43%), while the combination of intra-canopy lighting and top lighting yielded the lowest CV (37%). Combined intra-canopy and top lighting (50/50%) had in all directions a more uniform light absorption than intra-canopy or top lighting alone. The variation was minimal when the ratio of PPFD from intra-canopy to top lighting was about 1, and increased when this ratio increased or decreased. Intra-canopy lighting resulted in 8% higher total light absorption than top lighting, while combining 50% intra-canopy lighting with 50% top lighting, increased light absorption by 4%. Variation in light distribution was further reduced when the intra-canopy LEDs were distributed over strings at four instead of two heights. Abstract

Primary central nervous system lymphoma (PCNSL) is a rare type of non-Hodgkin lymphoma with an aggressive clinical course. To investigate the potential of immune-checkpoint therapy, we retrospectively studied the tumor microenvironment (TME) using high-plex immunohistochemistry in 22 PCNSL and compared to 7 secondary CNS lymphomas (SCNSL) and 7 "other" CNSL lymphomas with the presence of the Epstein-Barr virus and/or compromised immunity. The TME in PCNSL was predominantly composed of CD8+ cytotoxic T cells and CD163+ phagocytes. Despite molecular differences between PCNSL and SCNSL, the cellular composition and the functional spectrum of cytotoxic T cells were similar. But cytotoxic T cell activation was significantly influenced by pre-biopsy corticosteroids intake, tumor expression of PD-L1 and the presence of EBV. The presence of low numbers of CD8+ T cells and geographic-type necrosis each predicted inferior outcome in PCNSL. Both M1-like (CD68 + CD163low) and M2-like (CD68 + CD163high) phagocytes were identified, and an increased ratio of M1-like/M2-like phagocytes was associated with a better survival. PD-L1 was expressed in lymphoma cells in 28% of cases, while PD1 was expressed in only 0.4% of all CD8+ T cells. TIM-3, a marker for T cell exhaustion, was significantly more expressed in CD8posPD-1pos T cells compared to CD8posPD-1neg T cells, and a similar increased expression was observed in M2-like pro-tumoral phagocytes. In conclusion, the clinical impact of TME composition supports the use of immune-checkpoint therapies in PCNSL. Based on observed differences in immune-checkpoint expression, combinations that boost cytotoxic T cell activation (by blocking TIM-3 or TGFBR1) prior to the administration of PD-L1 inhibition could be of interest.

Abstract

In melanoma, the lymphocytic infiltrate is a prognostic parameter classified morphologically into ‘brisk’, ‘non-brisk’ and ‘absent’ entailing a functional association that has never been proved. Recently, it has been shown that lymphocytic populations can be very heterogeneous, and that anti-PD-1 immunotherapy supports activated T cells. Here, we characterize the immune landscape in primary melanoma by high-dimensional single-cell multiplex analysis in tissue sections (MILAN technique) followed by image analysis, RT-PCR and shotgun proteomics. We observed that the brisk and non-brisk patterns are heterogeneous functional categories that can be further sub-classified into active, transitional or exhausted. The classification of primary melanomas based on the functional paradigm also shows correlation with spontaneous regression, and an improved prognostic value when compared to that of the brisk classification. Finally, the main inflammatory cell subpopulations that are present in the microenvironment associated with activation and exhaustion and their spatial relationships are described using neighbourhood analysis.

Abstract

The treatment of spinal muscular atrophy (SMA) has considerably changed over the last 3 years. Several approaches that aim to increase the deficient SMN protein have demonstrated an efficacy that is inversely correlated with disease duration. In this context, newborn screening (NBS) is increasingly considered as the next step in several countries or regions. In 2018, we initiated a pilot study for NBS of SMA in French- and German-speaking Belgium. We aim to evaluate the feasibility, the efficacy, and the cost-effectiveness of such a program. Initially covering the region of Liege, the program was recently extended to the whole Southern Belgium and currently covers about 55.000 newborns per year. On June 1st 2019, 35.000 newborns had been screened and 5 affected babies were identified and referred to neuromuscular centers for early treatment. A full evaluation of the program will take place after three years to consider the inclusion of SMA screening in the publically-funded NBS program in Southern Belgium.La prise en charge de l’amyotrophie spinale antérieure (SMA) a considérablement évolué au cours des trois dernières années. Les différents essais visant à augmenter la production de la protéine SMN déficitaire dans la SMA ont systématiquement montré une efficacité inversement proportionnelle à la durée de la maladie. Dès lors, l’implémentation d’un programme de dépistage néonatal s’est rapidement imposée comme une évidence médico-économique dans de nombreux pays. Dans ce contexte, nous avons initié un programme de dépistage néonatal pour la SMA en Belgique francophone et germanophone. En 2018, une étude pilote de trois ans visant à évaluer la faisabilité, l’efficacité et la rentabilité du screening a été initiée au sein du centre de dépistage de Liège. L’étude a récemment été étendue à l’ensemble de la Fédération Wallonie-Bruxelles (FWB) pour couvrir environ 55.000 naissances annuelles. Au 1er juin 2019, 35.000 bébés ont été dépistés et cinq nouveau-nés atteints de SMA ont été identifiés. Tous ont été immédiatement référés pour assurer leur prise en charge dans un centre de référence pour les maladies neuromusculaires. Une évaluation complète du programme aura lieu à l’issue de la phase pilote, afin d’envisager que la SMA soit reconnue comme maladie officielle du programme de dépistage néonatal en FWB.

Abstract

We present a methodology to enhance the electrical capacitance of activated carbon (AC) electrodes based on the introduction of electrically charged defects through argon plasma processing. Extensive characterization using electrochemical techniques incorporating cyclic voltammetry, constant current charge/discharge, and electrical impedance spectroscopy indicated a close to seven-fold increase in capacitance with respect to untreated AC electrodes, not subject to plasma processing.

Abstract

The WHO scheme for prothrombin time (PT) standardization has been limited in application, because of its difficulties in implementation, particularly the need for mandatory manual PT testing and for local provision of thromboplastin international reference preparations (IRP).The value of a new simpler procedure to derive international normalized ratio (INR), the PT/INR Line, based on only five European Concerted Action on Anticoagulation (ECAA) calibrant plasmas certified by experienced centres has been assessed in two independent exercises using a range of commercial thromboplastins and coagulometers. INRs were compared with manual certified values with thromboplastin IRP from expert centres and in the second study also with INRs from local ISI calibrations.In the first study with the PT/INR Line, 8.7% deviation from certified INRs was reduced to 1.1% with human reagents, and from 7.0% to 2.6% with rabbit reagents. In the second study, deviation was reduced from 11.2% to 0.4% with human reagents by both local ISI calibration and the PT/INR Line. With rabbit reagents, 10.4% deviation was reduced to 1.1% with both procedures; 4.9% deviation was reduced to 0.5% with bovine/combined reagents with local ISI calibrations and to 2.9% with the PT/INR Line. Mean INR dispersion was reduced with all thromboplastins and automated systems using the PT/INR Line.The procedure using the PT/INR Line provides reliable INR derivation without the need for WHO ISI calibration across the range of locally used commercial thromboplastins and automated PT systems included in two independent international studies.? 2010 International Society on Thrombosis and Haemostasis.

Abstract

Organ temperature and variation therein plays a key role in plant functioning and its responses to e.g. climate change. There is a strong feedback between organ, especially leaf, temperature and the climate within the canopy (canopy climate), which in turn interacts with the climate outside the canopy (ambient climate). For greenhouses, the determinants of this interplay and how they drive differences between canopy and ambient climate are poorly understood. Yet, as many experiments on both regular greenhouse crops and field crops are done in greenhouses, this is crucial to know. Therefore, we designed an experiment to quantify the differences between ambient and canopy climate and leaf temperature. A path analysis was performed to quantify the interactions between components of the greenhouse canopy-climate system. We found that with high radiation the canopy climate can be up to 5°C cooler than the ambient climate, while for cloudy days this was only 2°C. Canopy relative humidity (RH) was up to 25% higher compared to ambient RH. We showed that radiation is very important for these climate differences, but that this effect could be partly counteracted by turning off supplementary light (i.e. due to its indirect effects e.g. changing light distribution). Leaf temperature was substantially different, both higher and lower, from the canopy air temperature. This difference was determined by leaf area index (LAI), temperature of the heating pipe and the use of supplementary light, which all strongly influence radiation, either shortwave or thermal radiation. The difference between leaf and ambient air temperature could be decreased by decreasing the LAI or increasing the temperature of the heating pipe. [ABSTRACT FROM AUTHOR]

Abstract

It was aimed to determine whether plasma processing could contribute to enhanced capacitance and energy density of activated carbon electrode based electrochemical capacitors, through the formation of additional surface charges. While an increase of up to 35% of the gravimetric capacitance, along with ~ 20% decrease in resistance, was obtained through optimal plasma processing, increased plasma exposure yielded a drastic reduction (/increase) in the capacitance (/resistance). It was also found that the capacitance and resistance modulation was a sensitive function of sample processing as well as electrochemical testing procedure. Considering the complexity of modeling realistic porous matrices, a metric to parameterize the reach of an electrolyte into the matrix has been posited., 15 pages, 5 figures, 2 tables

Abstract

Little is known about the effects of leaf pigmentation (related to leaf ontogeny), on the spectral dependence of photosynthesis and most observations have been limited so far to single leaves. This study aimed to investigate photosynthesis and the related optical properties of two types of rose leaves, young reddish leaves and middle age green leaves, and to quantify the spectral dependence of photosynthesis at the canopy level. Photosynthesis was measured with CO2/H2O gas analyzer on intact leaves of rose ‘Akito’ at narrow band light of 18 wavelengths. Subsequently, the optical properties (transmittance, absorptance and reflectance) were measured with spectrophotometer. A mechanistic crop model was used for up scaling measurements at the single leaf level to the crop level (crop with LAI = 3). The green and the reddish leaves had similar total PAR absorptance, even though absorptance around 550 nm was slightly lower in the green leaves. The maxima of photosynthesis efficiency were at 640-680 nm for quantum yield (per absorbed light unit) and at 660-680 nm for action spectrum (per incident light unit), regardless the colour of the leaf blade. In the range 500-580 nm, both the quantum yield and the action spectrum were lower in reddish than in green leaves. Differences in optical properties and photosynthetic behaviour were related to the higher content of anthocyanins in red leaves. The spectral dependence of light absorption and photosynthesis at the canopy level differed distinctly from that at leaf level. The spectral differences in absorption at the leaf level almost disappeared at the canopy level. Consequently, while the action spectrum of green light (520-570 nm) was only 67% of that of red light (680 nm) at the leaf level, it increased to 79% at the crop level. Young reddish leaves had higher absorptance but lower action spectrum and quantum yield at green light. Spectral differences in photosynthesis at the canopy level are much smaller than at the leaf level. Our short term measurements suggest that optimizing spectral output of LED lamps may increase photosynthesis up to 12% for a canopy with green leaves and up to 17% for a canopy with reddish leaves when compared to the spectrum of HPS lamps.
