These cells can introduce an accessible autologos source of neural precursor cells for treatment of different neurodegenerative diseases. This research was done in order to set up isolation, culture, proliferation and differentiation of human skin derived precursors (hSKPs)."n"nMethods: Human foreskin samples were cut into smaller pieces and cultured in proliferation medium after enzymatic digestion. To induce neural differentiation, cells were cultured in neural differentiation medium after fifth passage. We used immunocytochemistry and RT-PCR for characterization of the cells. Neuron and glial cell differentiation potential was assessed by immunofloresence using specific antibodies. The experiments were carried out in triplicate."n"nResults: After differentiation, βΙΙΙ- tubulin and neurofilament-M positive cells were observed that are specific markers for neurons. Moreover, glial fibrillary acid protein (GFAP) and S100 positive cells were identified that are markers specifically express in glial cells. Detected neurons and glials were also confirmed by their morphologic characterizations."n"nConclusion: Our results demonstrated that skin-derived precursors obtained from human foreskin can exhibit neuronal and glial differentiation potential in vitro, depending on the protocols of induction.

Because of the problems in purification and low amount of collagenases in bacterial or animal sources, it is important to find new sources of the enzymes. So, in the present study actinidin, a plentiful protein in kiwifruit was purified and a mixture of actinidin and trypsin was applied to isolate rat aortic endothelial cells."n"nMethods: Aortic endothelial cells were isolated using digestion solution containing different concentrations of actinidin (from 2 to 16 mg/ml) and trypsin (0.3, 0.6, 1.2 and 2.4 mg/ml) in different times (from 15 to 90 minute). Isolated cells were cultured in DMEM culture medium. Isolated cells were identified by morphological characteristics and immunocytochemical staining; viability of separated cells was estimated by trypan blue exclusion test."n"nResults: Actinidin in concentration of 10 mg/ml with trypsin in concentration of 1.2 mg/ml for one hour could isolate rat aortic endothelial cells. In this condition the viability of cells was estimated 90%. Morphological and immunocytochemical charac-teristics confirmed the isolated cells as endothelial cells."n"nConclusion: The results showed that the mentioned mixture of actinidin and trypsin has not considerable toxic effects on separated cells and is a novel and suitable option for isolation of rat aortic endothelial cells.

To integrate the consideration of environmental issues in the design phase of a product eco-design is an ecological quality improvement and preventive approach of it while verifying that the resulting environmental gain is not at the expense of other environmental aspects and focusing on the preservation of its quality of use of its features or certain stages of its life cycle without losing the own global vision of eco-design. Following this vision, the use of bio-laden animal based polymers derived from a mode of production by implementing optimization practices respectful of natural resources sustainably, has become changing way of thinking largely replace the use of non-renewable resources for human activities, the depletion of fossil resources deposits and to fight against the impact of climate change generated by this practice. Our paper attempts to analyze first the mechanical behavior of a bio-load and then incorporated into a polymer material to test improved mechanical and thermal characteristics made to it after adding and optimizing subsequently, its use. This is a load in the form of powder or chips extracted from an animal sheath. Tensile tests are performed in a first step, in order to mechanically characterize the virgin material and eco-composite obtained after the combination, i.e. the maximum tensile strength, the relative deformation and the modulus of the elasticity, etc

"nBackground: Plasminogen has a central role in fibrinolyrtic system can activate through various activators (PAs) to its active form plasmin and perfoem its vital function that is fibrin clot lysis. Furthermore the fibrinolyrtic system plays a major role in angiogenesis. The fibrinolyrtic system activation control cell migration and invasion. In addition to this, plasmin regulates tumor growth. Monoclonal antibodies, as biological tools, play an important role in basic researches. "nMethods: In the first step the effects of antibodies on the activation of fibrinolyrtic system with PAs were evaluated with several methods including macroscopic observation, quantitative measurement of DD/E fragments by D-dimer assay and activation of plasminogen by S-2251 synthetic substrate (ELISA method), subsequently we studied the effect of antibodies on angiogenesis process in an in- vitro model. "nResults: Results showed that MC2B8 that is an inhibitor of plasminogen activation in presence of plasminogen activators can inhibit angiogenesis process: A1D12 that is against N-terminal domain of Glu-plasminogen, in addition to activation of fibrinolyrtic system in presence of plasminogen activators, can activate in vitro angiogenesis process. "nConclusion: Plasmin formation is a critical step in invasion and migration of endothelial cells to form new vessels. Plasmin directly participates in angiogenesis by direct fibrin and other matrix components degradation, and indirectly by activating matrix degrading metalloproteinase and angiogenic growth factors. According to the in- vitro results, MC2B8 and A1D12 monoclonal antibodies play roles in this process in a dose dependent manner.

- Abstract
In this paper the effect of particle shape and size on the mechanic behavior of glass-particle reinforcing Nylon matrix composite is evaluated. There are many parameters effecting on the composite such as types, size, position and shape of reinforcement material. Particle morphology has received enormous interest in particulate composite; small particles adhere strongly to the polymer, which leads to a strong reinforcing effect. The objective of this research is to investigate the influence of reinforcement shape and size with different volume fractions and for three packing (square, hexagonal and random) using finite element method. The composite is subject to unidirectional tensile loading. Numerical results are presented for a variety of particle shapes and size including circular, triangular, square and two distributions grouping these shapes together. The results show that the variation of the Von Mises and longitudinal stresses consistently increased when the shape of the reinforcement changed from triangle to square, distribution 1 and distribution 2 to circular in this order. 18. Effect of glaucoma medications on 24-hour intraocular pressure-related patterns using a contact lens sensor

Background: The aim of this article was to study the circadian intraocular pressure (IOP)-related effects of ocular hypotensive medications using a contact lens sensor (CLS). Design: This is a university-based prospective, randomized, crossover trial. Participants: A total of 23 patients with primary open-angle glaucoma participated. Methods: Patients underwent ambulatory recording of IOP-related patterns for 24h in one eye during 3 monthly sessions using a CLS. Patients were untreated in session 1 (S1), were randomized to one of four classes of glaucoma drops for S2 and had a prostaglandin analogue add-on for S3. Main Outcome Measures: Changes in IOP-related patterns were defined using (i) slopes from wake/sitting to sleep/supine; (ii) cosinor rhythmometry modelling; and (iii) area under receiver operating curve (AUC) of sleep period. Results: Mean patient age was 63.8±11.8 years. Positive linear slopes were seen from wake/sitting to sleep/supine at S1 (17.1±14.2mVeq/h) and S2 (5.5±23.9mVeq/h) and negative slopes at S3 (-1.9±29.4mVeq/h) (S1-S2, P=0.01; S1-S3, P=0.02). This study explores the intricacies of employing two cooperative robotic manipulators for milling operations. Path planning, tool feed rate regulation, and cutting force management constitute essential elements in the milling process executed by these two collaborative robots. The two cooperative robots' milling process requires the coordination of path planning, cutting force management, and tool feed rate regulation. The path planning for the robotic system has been meticulously designed and seamlessly integrated with milling process expertise, while also adhering to the necessary technical specifications. This has been subsequently implemented on two cooperative robots featuring 3 Degrees of Freedom (3-DOF), utilizing the Staubli TX-90 robot model as a foundation. The milling of a typical workpiece by this robotic system is exemplified as a constrained optimization process. Regarding cooperative robot control, a method based on the Multiple Adaptive Neuro-Fuzzy Inference System (MANFIS) has been embraced. In order to validate the effectiveness of the presented approach, a comprehensive simulation has been conducted by using Matlab/SimMechanics software, and the results from this simulation have unequivocally substantiated the efficacy of the method. Our work entitled improvement of the mechanical properties of a Z80WCV 18-04-01 high-speed steel grooving cutter (three sizes) by heat treatment, the latter can play an essential role in improving the mechanical and structural properties of grooving mills. The results obtained showed that the heat treatments allow to obtain a favorable microstructure, with a martensitic phase formation and a homogeneous distribution of carbides. These structural changes have led to an increase in the hardness, strength and durability of the milling cutters to be grooved in addition, it has been observed that the application of cumulative revenues has a significant impact on the mechanical and structural properties. The successive revenues favored the transformation of quenching martensite into tempering martensite and the elimination of residual austenite, leading to a gradual increase in hardness and wear resistance. This paper proposes an inverse dynamics solution for two cooperative robots in a low depth milling application, by using the Levenberg-Marquardt algorithm. The objective is to achieve precise and coordinated control of the motions of the two robots working cooperatively to perform a milling task on a part. The proposed solution involves modeling the dynamics of the robots and their interactions, and formulating an optimization problem to obtain the forces on the workpiece. The proposed approach takes into account the kinematic and the dynamic constraints of the robots, as well as their interactions, in order to ensure smooth and coordinated motion. The Levenberg-Marquardt algorithm's effectiveness in achieving precise and synchronized motion and force control for a pair of collaborative robots during low-effort milling is validated through simulations conducted in MATLAB/SimMechanics. The simulation results confirm the robust performance of the proposed solution. This proposed approach shows potential for boosting the efficiency and the precision of collaborative milling tasks within industrial environments, ultimately resulting in improved manufacturing capabilities and increased productivity. This study investigates the response of small-scale length parameters and homogenization models of a simply supported nano-plate composed of functionally graded material. The natural frequency is presented for all cases, and the effect of different modes (Voigt, Reuss, LRVE, and Tamura), thickness ratio, and nonlocal parameter on the natural frequency is analyzed. The results show that the homogenization scheme is more influential in the vibrational response of FGM nanoplate with lower aspect ratios, and an increase in the small scale parameter causes a decrease in the natural frequency. To derive the governing equations and resolve them, the virtual work principle and Navier's model were employed. Some of the material demands in the advanced industries cannot be fulfilled by monolithic materials. Therefore, composite materials have been developed. The combination of desired properties of thermoplastics and glass particles (high strength and high modulus) is the aim of composites production Particles are becoming increasingly popular reinforcing elements in products made by injection molding. Particles reinforcement allows the polymer to be processed employing the same methods as those used for unreinforced polymers. The loads are not directly applied to the reinforcements, but they are applied to the matrix and some of the loads applied are transferred to the particles. The development of micromechanics equations for the particulate composites follows along the same lines as those for the short fiber reinforced composites. Particles are used to increase the strength or other properties of inexpensive materials during reinforcement with other matrix materials. The objective of this study is to analyse the particle breaking effect in composite made of nylon 66 (PA) matrix reinforced with glass particles, in which the particles diameters of 19.61, 26.15, 39.22 and 78.45 μm were used. Функціонально градуйовані матеріали (ФГМ) — це клас композитів, у яких властивості матеріалу поступово змінюються в одному або кількох декартових напрямках, поєднання яких призводить до більш ефективних характеристик, ніж компоненти, взяті окремо. Цей клас композитних матеріалів привернув значну увагу в інженерних галузях, таких як ядерна енергетика, аерокосмічна промисловість та виробництво електроенергії. Мета даної роботи полягала у дослідженні впливу моделей гомогенізації (ідеалізації) та термічних навантажень на статичну поведінку прогину сендвічової функціонально градуйованої плити. Кілька мікромеханічних моделей було використано для отримання ефективних властивостей матеріалу двофазної плівки ФГМ. Плівка ФГМ піддається лінійним і нелінійним тепловим навантаженням. Використана інтегральна теорія містить лише чотири функції змінних на відміну від п’яти у випадку інших моделей. Базове рівняння отримано та розв’язано за допомогою принципу віртуальної роботи та моделі Нав’є. Точність запропонованої аналітичної моделі підтверджується порівнянням результатів з літературними даними. Functionally graded materials (FGM) are a class of composites, in which the properties of the material gradually change over one or more Cartesian directions, the combination of which results in an assembly with higher performance than components taken separately. This class of composite materials has gained considerable attention in the engineering community, especially in high-temperature applications such as nuclear reactors, aerospace, and power generation industries. The aim of the current work is to study the influence of homogenization (idealization) models and thermal loads on static deflection behavior of sandwich functionally graded plate. Several micromechanical models have been employed to obtain the effective material properties of the two-phase FGM plate. The FGM plate is subjected to linear and no linear thermal loads. The integral theory used contains only four variable functions as against five in the case of other HSDTs. The governing equation are derived and resolved via virtual work principle and Navier’s model. The accuracy of the proposed analytical model is confirmed by comparing the results with those given by others model existing in the literature.

Background: Many causes of vision impairment can be prevented or treated. With an ageing global population, the demands for eye health services are increasing. We estimated the prevalence and relative contribution of avoidable causes of blindness and vision impairment globally from 1990 to 2020. We aimed to compare the results with the World Health Assembly Global Action Plan (WHA GAP) target of a 25% global reduction from 2010 to 2019 in avoidable vision impairment, defined as cataract and undercorrected refractive error. BackgroundThis study aimed to assess the prevalence and causes of vision loss in sub-Saharan Africa (SSA) in 2015, compared with prior years, and to estimate expected values for 2020.MethodsA systematic review and meta-analysis assessed the prevalence of blindness (presenting distance visual acuity In SSA, age-standardised prevalence of blindness, MSVI and MVI in 2015 were 1.03% (80% uncertainty interval (UI) 0.39–1.81), 3.64% (80% UI 1.71–5.94) and 2.94% (80% UI 1.05–5.34), respectively, for male and 1.08% (80% UI 0.40–1.93), 3.84% (80% UI 1.72–6.37) and 3.06% (80% UI 1.07–5.61) for females, constituting a significant decrease since 2010 for both genders. There were an estimated 4.28 million blind individuals and 17.36 million individuals with MSVI; 101.08 million individuals were estimated to have near vision loss due to presbyopia. Cataract was the most common cause of blindness (40.1%), whereas undercorrected refractive error (URE) (48.5%) was the most common cause of MSVI. Sub-Saharan West Africa had the highest proportion of blindness compared with the other SSA subregions.ConclusionsCataract and URE, two of the major causes of blindness and vision impairment, are reversible with treatment and thus promising targets to alleviate vision impairment in SSA.

However, there are few studies concerning rapamycin effects on the cervical cancer cells. In this study, it was aimed to investigate the possible effect of rapamycin on a cervical cancer cell line and explored the possible mechanism(s) and pathway(s) for this agent.Materials and Methods: To do so, HeLa cells as cervical cancer cell line were used and treated with different concentrations of rapamycin under both normoxic and hypoxic conditions. Then, cell viability assays, Western blot, quantitative real-time polymerase chain reaction (QR-PCR), acridine orange and acridine orange/propidium iodide staining were performed to evaluate rapamycin effect on the mentioned cell line.Results: The results showed that autophagy and apoptosis-related genes increased significantly in rapamycin-treated HeLa cells compared to controls. Moreover, cervical cancer cell death by rapamycin-induced autophagy in hypoxia was greater than normoxia compared with controls. In this study, it was showed that autophagy induction by rapamycin can mediate programmed cell death of cervical cancer cells, especially in hypoxic condition.Conclusion: These findings provide a new evidence that rapamycin may inhibit hypoxic HeLa cell proliferation through the trigger of programmed cell death, facilitating the development of novel anti-cancer therapy.Keywords: rapamycin, cervical cancer, autophagy, apoptosis, programed cell death

Функціонально градуйовані матеріали (FGM) – це новий клас композиційних матеріалів, які мають поступову та безперервну зміну об'ємних часток кожного з компонентів (загалом металу та кераміки) з товщиною, що відповідно спричиняє зміну загальних термомеханічних властивостей структурних елементів. Інтерес даної роботи полягає у використанні теорії високого порядку для дослідження термічного жолоблення FGM пластин, що спираються на пружну основу типу Вінклера-Пастернака. Цей метод веде до системи диференціальних рівнянь, де кількість невідомих дорівнює п'яти. Властивості матеріалу FGM пластин, такі як модуль Юнга та коефіцієнт теплового розширення, вважаються змінними з товщиною відповідно до моделі розподілу Морі-Танака. Теплове навантаження вважається рівномірним, лінійним і нелінійним по товщині пластини. Таким чином, проведено параметричне дослідження, щоб побачити вплив геометричних і механічних характеристик, зокрема, геометричного співвідношення (a/b), співвідношення товщин (a/h) та індексу матеріалу (k), а також впливу параметрів Вінклера і Пастернака на критичне навантаження на вигин. Functionally graded materials (FGM) are a new range of composite materials having a gradual and continuous variation of the volume fractions of each of the constituents (in general, metal and ceramic) in thickness, which accordingly causes changes in the overall thermomechanical properties of the structural elements they constitute. The interest of this work is the use of a high-order plate theory for the study of thermal buckling of FGM plates resting on Winkler-Pasternak type elastic foundation. The present method leads to a system of differential equations, where the number of unknowns is five. The material properties of FGM plate such as Young's modulus and coefficient of thermal expansion are assumed to be variable through the thickness according to the Mori-Tanaka distribution model. The thermal loading is assumed to be uniform, linear and nonlinear through the thickness of the plate. A parametric study is thus developed to see the influence of the geometric and mechanical characteristics, in particular, the geometric ratio (a/b), thickness ratio (a/h) and the material index (k), as well as the impact of the Winkler and Pasternak parameters on the critical buckling load.

У дослідженні запропоновано радіатори з металевої піни (MFHS) для теплового керування електронними пристроями. Металеві піни – це матеріали для покращення ефективності теплопередачі радіаторів завдяки їх унікальним характеристикам, таким як велике співвідношення площі поверхні до об'єму та їх складна форма, яка сприяє змішуванню та конвекції. Проведено чисельні дослідження перехідної теплогідравлічної поведінки та ефективності процесу охолодження електронних пристроїв за допомогою MFHS. Фізична модель складається з конвективного ламінарного потоку повітря всередині каналу, оснащеного кількома потужними електронними пристроями, що охолоджуються MFHS. MFHS складається з трьох пластинчастих радіаторів, які виготовлені з алюмінієвої піни з пористістю 0,95 і проникністю 1,65×10 – 7 м2. Основа радіатора виготовлена з твердого алюмінію. Для розв’язання базових рівнянь використовується програмне забезпечення Comsol. Численні результати показують, що теплова продуктивність MFHS більша, ніж у звичайного радіатора за тих самих умов експлуатації, а теплова поведінка електронних пристроїв, що охолоджуються MFHS, стабільно зберігається при допустимих температурах. Перевірка чисельних результатів вказує на високу узгодженість з експериментальними даними з максимальною відносною похибкою 3 %. In this study, metal foam heat sinks (MFHS) are proposed for thermal management of electronic devices. Metal foams are excellent candidates for improving the heat transfer performance of heat sinks due to their unique characteristics such as the large surface area to volume ratio and their complex form, which favors mixing and convection. Numerical investigations of the transient thermal-hydraulic behavior and performance of the cooling process of electronic devices by MFHS are carried out. The physical model consists of a convective laminar air flow inside a channel equipped with multiple power electronic devices cooled by MFHS. MFHS consist of three plate fin heat sinks which are made of aluminum foam with a porosity of 0.95 and a permeability of 1.65 × 10 – 7 m2, and the heat sink base is made of aluminum solid. Comsol software is used to solve the governing equations. Numerical results reveal that the thermal performance of MFHS is larger than that of a conventional heat sink and a clear channel under the same operating conditions, and the thermal behavior of electronic devices cooled by MFHS is stable and maintained at admissible temperatures. The validation of the numerical results shows perfect agreement with the experimental data with a maximum relative error of 3 %.

Тертя генерує розсіювання тепла, що супроводжується нагріванням системи pin-on-disc, у деяких випадках це нагрівання може впливати на термомеханічні властивості системи. У статті представлено перехідне тривимірне чисельне моделювання за допомогою програмного забезпечення COMSOL multiphysics термомеханічної поведінки під час динамічного контакту системи pin-on-disc. Термомеханічна модель тертя запозичена з літератури. Обговорюється вплив затримки контакту [0..10] секунд, зміни навантаження на pin [1..4] МПа та зміни швидкості диска [10..15] м/с. Результати роботи показують, що збільшення затримки контакту призводить до швидкого підвищення температури контактної зони в перші секунди, а починаючи з четвертої секунди, збільшення дуже незначне і стає постійним через поширення тепла шляхом провідності у вільних ділянках системи pin-on-disc. Вплив поступового збільшення навантаження на pin є більш помітним, ніж вплив швидкості диска на термомеханічну поведінку системи pin-on-disc. Friction generates thermal dissipation accompanied by heating of the pin-on-disc system, in some cases this heating can affect the thermomechanical properties of the system. In this paper, transient 3D numerical simulation by the COMSOL multiphysics software of the thermomechanical behavior during the dynamic contact of the pin-on-disc system is presented. A thermomechanical model of friction is adopted from the literature. The effects of contact delay [0..10] second, pin load variation [1..4] MPa and disc speed variation [10..15] m/s are discussed. The results of the present paper show that an increase in the contact delay generates a rapid increase in the temperature of the contact area in the first seconds, starting from the fourth second, the increase is very small and becomes constant due to the propagation of heat by conduction in the free areas of the pin-on-disc system. The effect of a gradual increase in pin load is more remarkable than that of disc speed on the thermomechanical behavior of the pin-on-disc system.

Functionally graded materials (FGM) are a class of composites, in which the properties of the material gradually change over one or more Cartesian directions, the combination of which results in an assembly with higher performance than components taken separately. This class of composite materials has gained considerable attention in the engineering community, especially in high-temperature applications such as nuclear reactors, aerospace, and power generation industries. The aim of the current work is to study the influence of homogenization (idealization) models and thermal loads on static deflection behavior of sandwich functionally graded plate. Several micromechanical models have been employed to obtain the effective material properties of the two-phase FGM plate. The FGM plate is subjected to linear and no linear thermal loads. The integral theory used contains only four variable functions as against five in the case of other HSDTs. The governing equation are derived and resolved via virtual work principle and Navier's model. The accuracy of the proposed analytical model is confirmed by comparing the results with those given by others model existing in the literature. [ABSTRACT FROM AUTHOR]

The measurement based on the vision of periodic patterns by a remote camera is a more suitable solution for the control and characterization of micro robotic systems. In this way, we can take precise measurements without disturbing the measured objects and without the need for more devices and sensors that cause great confusion and clutter around the workspace. The technique of projection of the coded periodic patterns is perhaps effective and of good performance and especially in the case of a well calibrated system. Indeed, the question arises on the influence of the calibration on the results of measurement by this technique in terms of precision and uncertainty. Thus, the main purpose of this article is to study the influence of calibration on the measurement process, by deducing and calculating the committed error and the uncertainty rate. Based on an algorithm in MATLAB, we first create a coded periodic test pattern to which we apply a certain imposed displacement. Then, we project this pattern using a projection and reduction system on a digital camera which captures a photo and transmits it to a processing algorithm, which, in turn, calculates and deduces a value for the measured displacement. The repetition of this process occurs several times on the same imposed displacement (the same standard) to provide us with several measured values, this redundancy of the data allows us to draw a calibration curve and deduce several metrological parameters from our system of measure. У роботі оцінено вплив форми частинок на механічну поведінку композитів, армованих частинками скла. Дрібні частинки міцно прилипають до полімеру, що призводить до сильного армуючого ефекту. При збільшенні загальної контактної поверхні на армуючі частинки передається більше навантаження. У наших попередніх дослідженнях було показано, що додавання круглих частинок із діаметром, що зменшується, не впливає на композит. Метою даної роботи є дослідження механічної поведінки термопластичних матричних композитів Nylon 66, армованих скляними частинками, при односпрямованому розтягуючому навантаженні за допомогою аналізу кінцевих елементів. Числові результати представлені для різних форм частинок, включаючи круглу, трикутну, квадратну, ромбічну, п'ятикутну та шестикутну. Результати показують, що напруження фон Мізеса послідовно збільшувалося в міру того, як форма армуючих частинок змінювалася з трикутної на квадратну, ромбічну, п'ятикутну, шестикутну та круглу в такому порядку. In this paper, the effect of particle shape on the mechanic behavior of glass particle reinforced composites is evaluated. Small particles adhere strongly to the polymer, which leads to a strong reinforcing effect. When the total contact surface increases, more loads are transferred to the reinforcing particles. In our previous studies, it has been shown that addition of circular particles with decreasing diameter does not affect the composite. The objective of this research is to investigate the mechanical behavior of thermoplastic matrix composite nylon 66, reinforced with glass particles, under unidirectional tensile loading using finite element analysis. Numerical results are presented for a variety of particle shapes, including circular, triangular, square, rhombic, pentagonal and hexagonal. The results show that Von Mises stresses consistently increased as the shape of the reinforcing particles changed from triangular to square, rhombic, pentagonal, hexagonal and circular in this order.

- Abstract
BACKGROUND: Many causes of vision impairment can be prevented or treated. With an ageing global population, the demands for eye health services are increasing. We estimated the prevalence and relative contribution of avoidable causes of blindness and vision impairment globally from 1990 to 2020. We aimed to compare the results with the World Health Assembly Global Action Plan (WHA GAP) target of a 25% global reduction from 2010 to 2019 in avoidable vision impairment, defined as cataract and undercorrected refractive error. METHODS: We did a systematic review and meta-analysis of population-based surveys of eye disease from January, 1980, to October, 2018. We fitted hierarchical models to estimate prevalence (with 95% uncertainty intervals [UIs]) of moderate and severe vision impairment (MSVI; presenting visual acuity from <6/18 to 3/60) and blindness (<3/60 or less than 10° visual field around central fixation) by cause, age, region, and year. Because of data sparsity at younger ages, our analysis focused on adults aged 50 years and older. FINDINGS: Global crude prevalence of avoidable vision impairment and blindness in adults aged 50 years and older did not change between 2010 and 2019 (percentage change -0·2% [95% UI -1·5 to 1·0]; 2019 prevalence 9·58 cases per 1000 people [95% IU 8·51 to 10·8], 2010 prevalence 96·0 cases per 1000 people [86·0 to 107·0]). Age-standardised prevalence of avoidable blindness decreased by -15·4% [-16·8 to -14·3], while avoidable MSVI showed no change (0·5% [-0·8 to 1·6]). However, the number of cases increased for both avoidable blindness (10·8% [8·9 to 12·4]) and MSVI (31·5% [30·0 to 33·1]). BACKGROUND: To contribute to the WHO initiative, VISION 2020: The Right to Sight, an assessment of global vision impairment in 2020 and temporal change is needed. We aimed to extensively update estimates of global vision loss burden, presenting estimates for 2020, temporal change over three decades between 1990-2020, and forecasts for 2050. - Abstract
Background: Humans are exposed to tens of thousands of chemical substances that need to be assessed for their potential toxicity. Acute systemic toxicity testing serves as the basis for regulatory hazard classification, labeling, and risk management. However, it is cost- and time-prohibitive to evaluate all new and existing chemicals using traditional rodent acute toxicity tests. In silica models built using existing data facilitate rapid acute tox- icity predictions without using animals. Objkctivks: The U.S. Interagency Coordinating Committee on the Validation of Alternative Methods (ICCVAM) Acute Toxicity Workgroup organ- ized an international collaboration to develop in silica models for predicting acute oral toxicity based on five different end points: Lethal Dose 50 (LD50 value, U.S. Environmental Protection Agency hazard (four) categories, Globally Harmonized System for Classification and Labeling hazard (five) categories, very toxic chemicals [LD50 (LD50 ≤ 50 mg/kg)], and nontoxic chemicals (LD50 > 2,000 mg/kg). Mkthods: An acute oral toxicity data inventory for 11,992 chemicals was compiled, split into training and evaluation sets, and made available to 35 participating international research groups that submitted a total of 139 predictive models. Predictions that fell within the applicability domains of the submitted models were evaluated using external validation sets. These were then combined into consensus models to leverage strengths of individual approaches. Results: The resulting consensus predictions, which leverage the collective strengths of each individual model, form the Collaborative Acute Toxicity Modeling Suite (CATMoS). CATMoS demonstrated high performance in terms of accuracy and robustness when compared with in viva results. Discussion: CATMoS is being evaluated by regulatory agencies for its utility and applicability as a potential replacement for in viva rat acute oral toxicity studies.
