N,N-dimethylglycine (DMG) is a naturally occurring compound being widely used as an oral supplement to improve growth and physical performance. Thus far, its effects on human skin have not been described in the literature. For the first time, we show that N,N-dimethylglycine sodium salt (DMG-Na) promoted the proliferation of cultured human epidermal HaCaT keratinocytes. Even at high doses, DMG-Na did not compromise the cellular viability of these cells. In a scratch wound-closure assay, DMG-Na augmented the rate of wound closure, demonstrating that it promotes keratinocyte migration. Further, DMG-Na treatment of the cells resulted in the upregulation of the synthesis and release of specific growth factors. Intriguingly, DMG-Na also exerted robust anti-inflammatory and antioxidant effects, as assessed in three different models of human keratinocytes, mimicking microbial and allergic contact dermatitis as well as psoriasis and UVB irradiation-induced solar dermatitis. These results identify DMG-Na as a highly promising novel active compound to promote epidermal proliferation, regeneration, and repair, and to exert protective functions. Further preclinical and clinical studies are under investigation to prove the seminal impact of topically applied DMG-Na on relevant conditions of the skin and its appendages.

In situ X-radiography imaging during the isothermal solidification of a 200 µm thin Al-24 at.% Ge sample was performed to determine dendritic growth rates, liquid concentrations and projected solid fractions. The experimental data allowed to follow dendrite tip growth velocities with high accuracy all the way from the initial transient growth related decrease of velocity via the expected increase of velocity during free dendrite growth to its final decrease during neighbor interacted growth. Concentration profiles in front of individual dendrite tips were measured to analyze the increase in far-field solute concentration due to solute accumulation. By time-resolved imaging of almost the entire 12 mm diameter sample, the global melt concentration could be derived. This enabled to estimate the initial nucleation undercooling of this un-refined alloy, which was so far unresolved. The experimental results are compared to a three-dimensional dendrite needle network model that simulates the experimental conditions, such as hexagonal dendrite structures and confined sample geometry. Although the simulation results are very sensitive to variations of the parameters undercooling and dendrite selection constant, one model parameter set is found that reproduces all experimentally measured solidification characteristics. The thorough experiment-model comparison indicates that the dendritic growth rates in this experimental configuration can be lower by a factor of 7 depending on undercooling than in the non-confined case.

Caffeine, particularly after ingestion, is well known to exert various pharmacological effects. A growing body of evidence implicates the ingestion of caffeine with beneficial effects on several diseases. The easy penetration of caffeine across the skin barrier and into human skin makes caffeine an ideal compound for topical application. Hair loss is known to negatively affect the quality of life and predispose to depression and anxiety. Androgenetic alopecia (AGA) is the most common type of hair loss in both men and women. To date, only few approved drug-based treatments for AGA exist, and these are inevitably associated with side effects. Therefore, the development of topical treatments based on well-tolerated natural ingredients such as caffeine to alleviate hair loss may provide a much-needed alternative to drug-based approaches.

International audience; To study dislocation dynamics in a model sample, an intrinsic float zone (FZ) Si wafer is chosen as seed to initiate directional solidification. During the temperature ramp, a Von Laue diffraction spot is recorded by a camera. It provides time-resolved information on the evolution of silicon crystalline quality and on the defect nucleation locations and dynamics. With increasing temperature, dislocations are observed to propagate through the seed starting mainly from the sample edges. The effective thermomechanical local stress is estimated as low as (1.2 ± 0.4) 10^5 Pa in a thermal gradient of (1.8 ± 0.2) 10^2 K m-1. The latter is sufficient to allow dislocation nucleation and motion at temperatures beyond (1523 ± 9) K. Dislocation velocity increases with temperature and reaches a maximum velocity of (3.0 ± 0.5) 10^-4 m s-1 close to the silicon melting point. From the dislocation velocity measurements as a function of temperature, an activation energy of (3.1 ± 0.6) eV is estimated and this value is discussed, along with dynamical interactions between defects and dislocations.

During metallurgical processing, gas bubbles nucleate and interact with the microstructure, which can result in severe defects. The research presented in this article focuses on in situ observation of bubble movement in semi-solid aluminum alloys (Al-Ge and Al-Cu) during a melting cycle. X-radiography is used to monitor the migration of H-bubbles in the semi-solid mush. For both alloys, H-bubbles are observed to burrow through the mushy zone towards the colder side of a horizontal gradient furnace leaving behind solute-enriched channels. This stands in contrast to the commonly observed bubble movement towards the hot side of the material caused by Marangoni flow-fields surrounding the gas pocket induced by gradients in the bubble’s surface tension. Different dependencies of the surface tension on temperature and concentration for the two alloys are presented as explanation for the observed atypical phenomenon. These results suggest different dominant effects acting on the bubbles, temperature for Al-Ge and concentration for Al-Cu, and explain the cryophile behavior observed.

A new x-ray isothermal furnace has been developed, suitable for in situ observations of semi-solid processes including the transition from dendritic to globulitic grain morphology and grain coarsening in metallic samples. A homogeneous, isothermal temperature field is achieved using a novel heater concept. The furnace structure is sandwich-like with heating elements positioned in the beam line and parallel to the sample. Planar heat transfer to the sample enables measurements with low cooling rates and a minimized temperature gradient. Cooling rates from 0.1 to 15 K min-1 can be controlled in the temperature range 1170-670 K. The furnace setup is integrable in the existing rotatable laboratory x-ray facility (X-RISE) at the German Aerospace Center (DLR). In this setup, an effective pixel size of 3 mum and a field of view of 8 mm in diameter can be achieved. Preliminary solidification and semi-solid experiments in the hypo-eutectic alloy systems Al-Ge and Al-Cu, inoculated with Al-5Ti-1B grain refiner, are presented. They indicate a very uniform temperature distribution in the sample.

Fuel prices are commonly perceived to be excessively high, which regularly triggers political discussions about fuel price regulations. Consumers demand stricter fuel price regulations to provide transparency about the current price level and to protect them from sudden price fluctuations. Such regulations are already in place in several countries, but whether they indeed help lower the overall fuel price level is unclear. In this paper, we study the effect of Austria's Fuel Price Fixing Act on gasoline and diesel price levels. Using the synthetic control method, we construct a counterfactual and estimate Austria's price trends in absence of the intervention. Our empirical results suggest that, immediately after the Fuel Price Fixing Act came into effect, gasoline prices are 23.4% lower than the synthetic control predicts. The effect on diesel prices appears to be less pronounced and amounts to 6.6% lower prices. Austria's fuel price regulation seems to have been effective in terms of gasoline prices but may need further refinements to effectively tackle diesel prices as well.

Directional solidification of a cast mono silicon seed and of a float-zone (FZ) silicon seed was performed and the grain and defect structures of the seeds as well as of the regrown parts are analyzed. In situ X-ray diffraction imaging enabled the observation of the dislocation arrangements. During the heating process, in the FZ seed, mobile dislocations glide on {111} planes, whereas in the cast mono seed dislocations are arranged in a mainly immobile cellular structure. Ex situ grain orientation mappings reveal the presence of subgrains with misorientations up to 3° in the regrown part of the cast mono-seeded sample, which are not observed in the regrown part of the FZ-seeded sample. Subgrain boundaries characterized by misorientations around the [001] growth axis propagate roughly along the growth axis and increase their misorientation by merging with new subgrain boundaries appearing in their vicinity. Although the first inception of subgrain formation cannot be revealed, the comparison of the dislocation arrangements in the two seeds strongly suggests an influence of the latter on subgrain formation. In the regrown part, interactions between subgrain boundaries and twin boundaries show that they can follow Σ3{111} and Σ9{221} grain boundaries or cross Σ3{111} grain boundaries. Whether Σ3{111} GBs are crossed or not depends among other things on the orientation of the grains on either side of the twin. It demonstrates that the grain orientation relationship and not only the grain boundary character play an important role in the subgrain structure evolution and redistribution in a multicrystalline silicon ingot.

As a microcosm for future challenges, the COVID-19 pandemic exhibits increasingly transboundary dynamics, causing interconnected problems across multiple societal systems. To examine the role of innovations as a social mechanism to reconcile these arising challenges, we view the unfolding of the pandemic through the lens of a content analysis of 707 innovation projects that address the fundamental human needs of consumers and businesses. This study proposes a novel procedure to characterize large-scale innovative activities via text mining and employs a theoretical framework for identifying the pressing societal needs amidst crises. Our typology of rapid-response COVID-19 innovations exhibits a diverse set of domains ranging from technological innovations to what may be described as frugal and social innovations. We provide evidence for the growing prevalence of social needs beyond the basic notion of safety during the early months of the crisis. Our contributions show that a structural model of innovation activities and their latent drivers may help policy makers and innovators to move toward achieving a systemic reaction to such crises.

A compact fully protected microfocus X-radiography facility (XRISE-M) is presented for the study of microstructure evolution during the solidification of thin liquid alloy samples and chemical diffusion in liquid binary alloys in situ and in real-time aboard a sounding rocket. XRISE-M presently enables the simultaneous processing of either two near-isothermal solidification furnaces or a combination of a linear-shear cell diffusion furnace and a near-isothermal solidification furnace. For optimal detector calibration shortly before flight, the furnaces can be rotated around the central beam axis and calibration images can be recorded. The facility allows preheating the samples into the liquid state prior to lift-off without leakage during the ascent phase at accelerations of up to 27 g. Macrosegregation on remelting of thin metal samples for microstructure evolution investigations is prevented by an inclinable furnace metric. The use of ion-getter pumps for vacuum generation enables us to exploit the entire available time of reduced gravity for image recording and data acquisition. With the device and currently available sample environments, microstructure formation upon solidification and chemical diffusion under purely diffusive conditions in alloys can be investigated. The facility can be used equally for other investigations such as granular matter dynamics or metal foaming, provided that suitable experiment inserts are developed in the future.

International audience; To control the final grain structure and the density of structural crystalline defects in silicon (Si) ingots is still a main issue for Si used in photovoltaic solar cells. It concerns both innovative and conventional fabrication processes. Due to the dynamic essence of the phenomena and to the coupling of mechanisms at different scales, the post-mortem study of the solidified ingots gives limited results. In the past years, we developed an original system named GaTSBI for Growth at high Temperature observed by Synchrotron Beam Imaging, to investigate in situ the mechanisms involved during solidification. X-ray radiography and X-ray Bragg diffraction imaging (topography) are combined and implemented together with the running of a high temperature (up to 2073 K) solidification furnace. The experiments are conducted at the European Synchrotron Radiation Facility (ESRF). Both imaging techniques provide in situ and real time information during growth on the morphology and kinetics of the solid/liquid (S/L) interface, as well as on the deformation of the crystal structure and on the dynamics of structural defects including dislocations. Essential features of twinning, grain nucleation, competition, strain building, and dislocations during Si solidification are characterized and allow a deeper understanding of the fundamental mechanisms of its growth.

Crystallographic orientations and growth directions of dendrites in thin Al-15 wt.%Cu (200 µm) and Al-29 wt.%Ge (500 µm) alloys were investigated employing in situ X-radiography and post-mortem X-ray tomography and electron backscatter diffraction. The Al-Cu alloy was solidified under microgravity conditions, while the Al-Ge sample was processed horizontally on ground. It was found that (i) dendrites nucleate on both sides of the surface in microgravity, but accumulate under normal gravity near the top surface due to buoyancy, (ii) dendrites grow along preferred in-plane crystallographic directions, which are 〈100〉 for Al-15 wt.%Cu and 〈100〉, 〈110〉 and 〈210〉 for Al-29 wt.%Ge, indicating that the solid-liquid interfacial anisotropy of this Al-Ge composition is similar for these 〈xy0〉 directions, (iii) slightly inclined dendrite arms in Al-Cu grow along the sample boundary after contact, whereas tips of inclined dendrite arms in Al-Ge never touch the boundary, but develop new primary tips originating from secondary branches.

Simple Summary Cachexia is a deadly disease that accompanies many different types of cancers. Animal studies on cachexia have so far mostly been conducted using young mice, while cancer in humans is a disease of high age. Mouse models used to date may therefore not be suitable to study cachexia with relevance to patients. By comparing young and old mice of three different strains and two different tumor types, we here show that the age of mice has a substantial effect on cachexia progression (specifically body weight, tissue weight, fiber size, molecular markers) that is dependent on the mouse strain studied. This is independent of glucose tolerance. The cachexia markers IL6 and GDF15 differ between ages in both mice and patients. Future studies on cachexia should consider the age and strain of mice. Abstract Background: Cancer is primarily a disease of high age in humans, yet most mouse studies on cancer cachexia are conducted using young adolescent mice. Given that metabolism and muscle function change with age, we hypothesized that aging may affect cachexia progression in mouse models. Methods: We compare tumor and cachexia development in young and old mice of three different strains (C57BL/6J, C57BL/6N, BALB/c) and with two different tumor cell lines (Lewis Lung Cancer, Colon26). Tumor size, body and organ weights, fiber cross-sectional area, circulating cachexia biomarkers, and molecular markers of muscle atrophy and adipose tissue wasting are shown. We correlate inflammatory markers and body weight dependent on age in patients with cancer. Results: We note fundamental differences between mouse strains. Aging aggravates weight loss in LLC-injected C57BL/6J mice, drives it in C57BL/6N mice, and does not influence weight loss in C26-injected BALB/c mice. Glucose tolerance is unchanged in cachectic young and old mice. The stress marker GDF15 is elevated in cachectic BALB/c mice independent of age and increased in old C57BL/6N and J mice. Inflammatory markers correlate significantly with weight loss only in young mice and patients. Conclusions: Aging affects cachexia development and progression in mice in a strain-dependent manner and influences the inflammatory profile in both mice and patients. Age is an important factor to consider for future cachexia studies.

A key parameter in understanding mantle dynamics beneath continents is the temporal evolution of intraplate volcanism in response to lithospheric thinning and asthenospheric uplift. To contribute to a better understanding of how intraplate volcanic fields evolve through time, we present a high precision 40Ar/39Ar age dataset for volcanic rocks from the Siebengebirge volcanic field (SVF) from central Germany, one of the best studied and compositionally most diverse intraplate volcanic fields of the Cenozoic Central European Volcanic Province (CEVP). Petrological and geochemical investigations suggest that the formation of the different rock types that occur in the SVF can be explained by a combination of assimilation and fractional crystallisation processes, starting from at least two different parental magmas with different levels of silica saturation (alkali basaltic and basanitic), and originating from different mantle sources. These evolved along two differentiation trends to latites and trachytes, and to tephrites and tephriphonolites, respectively. In contrast to their petrogenesis, the temporal evolution of the different SVF suites is poorly constrained. Previous K/Ar ages suggested a time of formation between about 28 and 19 Ma for the mafic rocks, and of about 27 to 24 Ma for the differentiated rocks. Our results confirm at high precision that the differentiated lithologies of both alkaline suites (40Ar/39Ar ages from 25.3 ± 0.2 Ma to 25.9 ± 0.3 Ma) erupted contemporaneously within a very short time period of ~ 0.6 Ma, whereas the eruption of mafic rocks (basanites) lasted at least 8 Ma (40Ar/39Ar ages from 22.2 ± 0.2 Ma to 29.5 ± 0.3 Ma). This implies that felsic magmatism in the central SVF was likely a single event, possibly triggered by an intense phase of rifting, and that ongoing melting and eruption of mostly undifferentiated mafic lavas dominate the > 8 Ma long magmatic history of this region. Among the mafic lavas, most basanites and tephrites predate the alkali basalts and hawaiites, suggesting an overall temporal evolution towards less SiO2-undersaturated primary melts and increasing degrees of melting over time. The peak in alkali basaltic to hawaiitic magmatism slightly post dates the flare-up of genetically related felsic magmatism, by no more than ~ 1 Ma. This is consistent with a model in which the magmatic plumbing system erupted successively from upper to lower levels, i.e. from more evolved to more primitive compositions. One young age for a basanitic sample suggests that silica saturation decreased again towards the end of volcanic activity. This chronology of volcanic events is in good agreement with previous models, suggesting continuous lithospheric thinning beneath the SVF as a response to an extensional regime and asthenospheric uplift in the northern alpine realm.

International audience; Synopsis A setup for simultaneous, time-resolved X-ray radiography and diffraction topography imaging is presented. It is used to study defect generation and growth mechanisms during heating, solidification and cooling of a silicon crystal. Abstract One of the key issues to be resolved to improve the performance of silicon solar cells is to reduce crystalline defect formation and propagation during the growth process fabrication step. For this purpose, the generation of structural defects such as grain boundaries and dislocations in silicon must be understood and characterised. We combine in situ X-ray diffraction imaging, historically named topography, with radiography imaging to analyse the development of crystal defects before, during and after crystallisation. Two individual indirect detector systems are implemented to record simultaneously the crystal structure (topographs) and the solid-liquid morphology evolution (radiographs) at high temperature. This allows for a complete synchronisation of the images and for an increased image acquisition rate compared to previous studies that used X-ray sensitive films to record the topographs. The experiments are performed with X-ray synchrotron radiation at beamline ID19 at the European Synchrotron Radiation Facility (ESRF). We present in situ observations of the heating, melting, solidification and holding stages of silicon samples to demonstrate that with the upgraded setup detailed investigations of time-dependent phenomena are now possible. The motion of dislocations is recorded during the entire experiment, so that their interaction with grain boundaries and their multiplication through the activation of Frank-Read sources can be observed. Moreover, the capability to record with two camera-based detectors allows for the study of the relationship between strain distribution, twinning and nucleation events. In conclusion, the simultaneous recording of topographs and radiographs has great potential for further detailed investigations of the interaction and generation of grains and defects that influence the growth process and the final crystalline structure in silicon and other crystalline materials.

The collection of solid waste and the recovery of recyclable material from waste belong to the many challenges Cuba has been facing over the past years. In this paper, we give a short account of the actual waste situation and analyze the causes of the obvious deficiencies of the waste sector. We conducted a small survey in Havana city in order to assess peoples awareness of the garbage problem, their appraisal of the actual situation and their willingness to contribute personally to an improvement of the waste situation. Based on the insights gained from this survey and from our theoretical analysis of the Cuban waste management we develop suggestions for an improvement of the waste situation in Cuba. These suggestions take into account that the financial means available for a reform of the waste management system in Cuba are severely restricted. Therefore, our suggestions are more focused on organizational and motivational changes than on the introduction of high technology.

We report on a dendrite orientation transition (DOT) discovered in the Al Ge system by conducting isothermal solidification experiments in thin samples under slow cooling conditions. The DOT was revealed using a combination of in situ X-radiography imaging and post-mortem electron backscatter diffraction. In Al-20 wt% Ge the primary arms grow along 〈 100 〉 , whereas in Al-46 wt% Ge the primary arms grow along 〈 110 〉 . At an intermediate composition of Al-29 wt% Ge, we observe both directions growing simultaneously. Phase-field simulations in which the solid-liquid interfacial energy anisotropy was systematically varied to correspond with the composition changes reproduced the experimentally observed microstructures. The remarkable agreement between the experimental observations and the simulations provides strong evidence that the DOT in this alloy system is caused by modification of surface energy anisotropy with increasing Ge content. Furthermore, we show that the confinement due to the thin sample geometry influences the selection of the primary growth directions, so that different growth morphologies develop depending on the orientation of the crystal in the sample.

We determine the tip growth velocities of equiaxed dendrites in an Al-24 at. % Ge alloy using in situ x-ray radiography. The tip growth velocity is a main characteristic of dendritic growth and allows for an accurate comparison against theories and simulations describing the growth of a single dendrite tip or an array of dendrites. The disk-shaped samples are 200 or 350 $\ensuremath{\mu}\mathrm{m}$ thin and are positioned horizontally in a near-isothermal furnace to suppress buoyancy and gravity-driven melt flows. We obtain a large data set of free dendritic tip growth velocities of Al dendrites over one order of magnitude (4--140 $\ensuremath{\mu}\mathrm{m}\phantom{\rule{0.16em}{0ex}}{\mathrm{s}}^{\ensuremath{-}1}$) and over a decade of global undercooling (1--10 K). No indications for a significant deviation from three-dimensional growth kinetics due to the sample confinement are found.

Molecular checkpoints that trigger the onset of islet autoimmunity or progression to human type 1 diabetes (T1D) are incompletely understood. Using T cells from children at an early stage of islet autoimmunity without clinical T1D, we find that a microRNA181a (miRNA181a)-mediated increase in signal strength of stimulation and costimulation links nuclear factor of activated T cells 5 (NFAT5) with impaired tolerance induction and autoimmune activation. We show that enhancing miRNA181a activity increases NFAT5 expression while inhibiting FOXP3+ regulatory T cell (Treg) induction in vitro. Accordingly, Treg induction is improved using T cells from NFAT5 knockout (NFAT5ko) animals, whereas altering miRNA181a activity does not affect Treg induction in NFAT5ko T cells. Moreover, high costimulatory signals result in phosphoinositide 3-kinase (PI3K)-mediated NFAT5, which interferes with FoxP3+ Treg induction. Blocking miRNA181a or NFAT5 increases Treg induction in murine and humanized models and reduces murine islet autoimmunity in vivo. These findings suggest targeting miRNA181a and/or NFAT5 signaling for the development of innovative personalized medicines to limit islet autoimmunity.

Carbonaceous chondrites are an important meteorite group that closely resembles the bulk composition of the solar system. We report the first elemental and isotope dataset for Hf–W in carbonaceous chondrites that includes chondrules, matrix, magnetic fractions as well as bulk compositions. Our study focuses on the three CV3 chondrites, Allende, Vigarano and Bali. Compared to bulk chondrites, matrix splits have low Hf/W ratios and e 182 W compositions, whereas chondrule splits are characterized by high, but more variable, Hf/W ratios and e 182 W compositions. Thus, Hf/W ratios behave complementary between chondrules and matrix in the analysed CV chondrites, supporting the view that both components formed from the same parental reservoir. Strong nucleosynthetic effects were observed in most of the analysed CV3 components, especially in matrices and chondrule splits that were found to have large e 183 W anomalies of several e -units. All separates define a rough correlation between initial 182 W/ 184 W and 183 W/ 184 W ratios, in agreement with theoretical model trends based on calculations for stellar nucleosynthesis. Our results, therefore, indicate a heterogeneous distribution of s- and r-process W isotopes among the different CV3 chondrite components, arguing for selective thermal processing of early solar system matter during chondrule formation. After correcting for nucleosynthetic anomalies, chondrules and matrix splits of reduced (Vigarano) as well as oxidised (Allende) CV3 chondrites define a linear correlation in e 182 W vs. 180 Hf/ 184 W space, which is interpreted as an isochron, covering an age interval within the first ∼2.6 Ma after solar system formation. As peak metamorphic temperatures for CV3 chondrites were well below the 182 Hf– 182 W closure temperature, the resulting isochron within its error most likely defines a common formation interval for all components. The calculated age interval is for the first time based on a combined chondrule-matrix isochron, a marked difference compared to previous studies where only chondrules were analysed. Notably, our formation age interval covers previously reported chondrule formation ages determined using 26 Al and Pb–Pb chronometry, illustrating that chondrule and matrix formation started contemporaneously with CAI formation and lasted over a time interval of about 2–3 Ma. Our results also corroborate previous models from ordinary chondrites, in that chondrite parent bodies were not the first planetesimals to have formed in the early solar system.

Obesity and type-2 diabetes (T2D) are associated with metabolic defects and inflammatory processes in fat depots. FoxP3 + regulatory T cells (Tregs) control immune tolerance, and have an important role in controlling tissue-specific inflammation. In this mini-review we will discuss current insights into how cross-talk between T cells and adipose tissue shapes the inflammatory environment in obesity-associated metabolic diseases, focusing on the role of CD4 + T cells and Tregs. We will also highlight potential opportunities for how the immunoregulatory properties of Tregs could be harnessed to control inflammation in obesity and T2D and emphasize the critical need for more research on humans to establish mechanisms that are conserved in both mice and humans.

An important tool to study dendritic growth in metallic alloys is the in-situ polychromatic X-radiography. Since solutal interaction is a dominant factor influencing dendrite growth, the determination of liquid concentrations is crucial to interpret the image data. Laboratory experiments with polychromatic X-ray sources require reference measurements of samples of known composition and thickness to convert intensities into concentrations because of the wavelength dependency of the absorption coefficient. Calibration measurements and fitted intensity functions are used to determine solutal concentrations in an Al-Ge sample without using in-situ reference samples.

In this paper, we present a newly developed near-isothermal X-ray transparent furnace for in situ imaging of solidification processes in thin metallic samples. We show that the furnace is ideally suited to study equiaxed microstructure evolution and grain interaction. To observe the growth dynamics of equiaxed dendritic structures, a minimal temperature gradient across the sample is required. A uniform thermal profile inside a circular sample is achieved by positioning the sample in the center of a cylindrical furnace body surrounded by a circular heater arrangement. Performance tests with the hypo-eutectic Al-15wt.%Cu and the near-eutectic Al-33wt.%Cu alloys validate the near-isothermal character of the sample environment. Controlled cooling rates of less than 0.5 K min(-1) up to 10 K min(-1) can be achieved in a temperature range of 720 K-1220 K. Integrated in our rotatable laboratory X-ray facility, X-RISE, the furnace provides a large field of view of 10.5 mm in diameter and a high spatial resolution of ∼4 μm. With the here presented furnace, equiaxed dendrite growth models can be rigorously tested against experiments on metal alloys by, e.g., enabling dendrite growth velocities to be determined as a function of undercooling or solutal fields in front of the growing dendrite to be measured.

The combination of micro synthetic structures with bacterial flagella motors represents an actual trend for the construction of self-propelled micro-robots. The development of methods for fabrication of these bacteria-based robots is a first crucial step towards the realization of functional miniature and autonomous moving robots. We present a novel scheme based on optical trapping to fabricate living micro-robots. By using holographic optical tweezers that allow three-dimensional manipulation in real time, we are able to arrange the building blocks that constitute the micro-robot in a defined way. We demonstrate exemplarily that our method enables the controlled assembly of living micro-robots consisting of a rod-shaped prokaryotic bacterium and a single elongated zeolite L crystal, which are used as model of the biological and abiotic components, respectively. We present different proof-of-principle approaches for the site-selective attachment of the bacteria on the particle surface. The propulsion of the optically assembled micro-robot demonstrates the potential of the proposed method as a powerful strategy for the fabrication of bio-hybrid micro-robots.

Obesity and type 2 diabetes are associated with metabolic defects and adipose tissue inflammation. Foxp3(+) regulatory Tcells (Tregs) control tissue homeostasis by counteracting local inflammation. However, if and how Tcells interlink environmental influences with adipocyte function remains unknown. Here, we report that enhancing sympathetic tone by cold exposure, beta3-adrenergic receptor (ADRB3) stimulation or a short-term high-calorie diet enhances Treg induction invitro and invivo. CD4(+) Tcell proteomes revealed higher expression of Foxp3 regulatory networks in response to cold or ADRB3 stimulation invivo reflecting Treg induction. Specifically, Ragulator-interacting protein C17orf59, which limits mTORC1 activity, was upregulated in CD4(+) Tcells by either ADRB3 stimulation or cold exposure, suggesting contribution to Treg induction. By loss- and gain-of-function studies, including Treg depletion and transfers invivo, we demonstrated that a Tcell-specific Stat6/Pten axis links cold exposure or ADRB3 stimulation with Foxp3(+) Treg induction and adipose tissue function. Our findings offer a new mechanistic model in which tissue-specific Tregs maintain adipose tissue function.

Exposure of these crystals to aqueous protein solutions leads to site-selective immobilization of proteins onto the crystals.

Systematic studies are presented demonstrating the complementarity of directed ortho metalation (DoM) and Ir-catalyzed strategies for the provision of borylated aromatics and their subsequent Suzuki-Miyaura coupling reactions. A new concept, the use of the TMS group, readily introduced by DoM, as a latent regiodirective moiety to overcome the otherwise problematic production of isomeric borylated product mixtures is presented. Additional electrophile-induced ipso-deborylation and DoM reactions of the Bpin products are described.

We sought to determine the performance of the new high sensitivity cardiac troponin T assay (TnThs) for early diagnosis of myocardial infarction in patients with suspected acute coronary syndrome (ACS) and compare it with the fourth generation cTnT assay, myoglobin and heart-type fatty acid binding protein (h-FABP). Objective Obesity and type-2 diabetes (T2D) are metabolic diseases that represent a critical health problem worldwide. Metabolic disease is differentially associated with fat distribution, while visceral white adipose tissue (VAT) is particularly prone to obesity-associated inflammation. Next to their canonical function of immune suppression, regulatory T cells (Tregs) are key in controlling adipose tissue homeostasis. Towards understanding the molecular underpinnings of metabolic disease, we focus on how environmental-metabolic stimuli impinge on the functional interplay between Tregs and adipose tissue. Here, cold exposure or beta3-adrenergic signaling are a promising tool to increase energy expenditure by activating brown adipose tissue, as well as by reducing local inflammation within fat depots by supporting immunosuppressive Tregs. However, in humans, the underlying mechanisms that enable the environmental-immune crosstalk in the periphery and in the respective tissue remain currently unknown. Methods We used combinatorial approaches of next generation humanized mouse models and in vitro and in vivo experiments together with beta3-adrenergic stimulation to dissect the underlying mechanisms of human Treg induction exposed to environmental stimuli such as cold. To test the translational relevance of our findings, we analyzed samples from the FREECE study in which human subjects were exposed to individualized cooling protocols. Samples were analyzed ex vivo and after in vitro Treg induction using qRT-PCR, immunofluorescence, as well as with multicolor flow cytometry and cell sorting. Results In vivo application of the beta3-adrenergic receptor agonist mirabegron in humanized mice induced thermogenesis and improved the Treg induction capacity of naïve T cells isolated from these animals. Using samples from the human FREECE study, we demonstrate that a short-term cold stimulus supports human Treg induction in vitro and in vivo. Mechanistically, we identify BORCS6 encoding the Ragulator-interacting protein C17orf59 to be significantly induced in human CD4+ T cells upon short-term cold exposure. Strong mTOR signaling is known to limit successful Treg induction and thus likely by interfering with mTOR activation at lysosomal surfaces, C17orf59 improves the Treg induction capacity of human naïve T cells upon cold exposure. Conclusions These novel insights into the molecular underpinnings of human Treg induction suggest an important role of Tregs in linking environmental stimuli with adipose tissue function and metabolic diseases.
