This study aimed to explore the biological influence and mechanism of LINC01272 in GC.Materials and Methods: Using bioinformatic analyses, we investigated the expression of LINC01272 in TCGA database and predicted the biological functions and mechanism of LINC01272 in GC. Then, we detected the expression of LINC01272 in GC cell lines, GC tissues, and corresponding normal tissues using real-time polymerase chain reaction (RT-PCR). Finally, we explored the migration and invasion ability of LINC01272 by wound-healing and Transwell assays and examined the expression of epithelial-mesenchymal transition (EMT)-related proteins through Western blotting.Results: We found that LINC01272 was upregulated in GC and was associated with GC staging and lymph node metastasis. The results ofwound-healing and Transwell assays revealed that the LINC01272 was closely related to GC cell migration and invasion. LINC01272 knockdown inhibited the migration and invasion ability of GC cells by reducing the expression of EMT-related proteins. Overexpression of LINC01272 had the opposite effect.Conclusion: Together, our results showed that LINC01272 promoted GC metastasis ability by regulating the expression of EMT-related proteins and could serve as a potential diagnostic biomarker for GC.Keywords: gastric cancer, long non-coding RNA, migration, invasion, LINC01272

Heat-labile enterotoxin B subunit (LTB) of Escherichia coli is a potential mucosal immune adjuvant for its safety. However, the weaker adjuvant activity fails to meet the clinical requirement. Here, one of LTB mutant numbered LTB26 is constructed with enhanced mucosal immune adjuvanticity than that of LTB wild type (LTB). The transcriptome analysis data suggest that LTB26 enhances mucosal immune adjuvanticity via increased expression of BCR and MHC II + molecular. Furthermore, LTB26 can promote both Th1 and Th2 cell mediated immunity via upregulated expression of IL-4 and IFN-γ. Flow cytometry analysis confirms that LTB26 significantly upregulates the activity of antigen presenting cells (DCs and mature macrophage) compared with LTB and LTB57 mutant. The result demonstrates that LTB26 is a potent mucosal immune adjuvant meeting clinical requirement. The GM1 ganglioside (GM1) binding activity of LTB57 is higher than that of LTB26; instead, the immune adjuvanticity of LTB57 is lower than that of LTB26. The result highlights that the immune adjuvanticity of LTB and its mutant are not positively associated with GM1 affinity, which upends decades understanding of the relationship of LTB adjuvanticity and GM1-binding affinity.

To inspect the effect of main MS analysis conditions and data accuracy through matrix-assisted laser desorption ionization-time of flight-mass spectrometry and to obtain peptides mass fingerprint (PMF) for protein search results. BSA had been separated by SDS-PAGE, hydrolyzed by trypsin, and identified by MS, and PMF data were searched through MS-Fit search engine. The results showed that the sequence coverage and the peptide coverage gradually increase when laser strike numbers under 300 and numbers at 400, which appeared to descend tendency. Target protein score, amino acid sequence, and peptide coverage appeared to increase following increase in laser intensity, and when laser intensity was at 1413, there was a tendency to descend. Peptide coverage showed an increasing tendency following delay-time increase, and when delay time was at 260 ns, there was a tendency to descend. On the basis of optimized analysis conditions and on checking the same site for 3 times, the result showed that reproducibility was better. To inspect the accuracy of MS data, Mix 2 was used as external standard to remedy MS primitive data. It was found to score higher, order ahead, detect fewer numbers, and show better reliability as the MS data approached standard value. In contrast, we almost didn't get target protein. The results showed that MS accuracy and analysis conditions were present, which distinguished the influence on protein database search.

The bioinformatics analysis conditions (databases and major parameters) in the process of Peptide Mass Fingerprint (PMF) identify target proteins were optimized in this study. In this process of experiment design and analysis, the Bovine Carbonic anhydrase-2 (CAH2_BOVIN) and human Hsp70s protein (Heat shock protein 70) were separated by two-dimension electrophoresis(2-DE), proteolysis by Trypsin Gold, and then the lysed peptides were gathered and analyzed using MALDI-TOF-MS, and the peptides mass data and mass spectrogram were obtained. In this study, the Swissprot, MSDB, NCBInr, Random databases, MASCOT, and MS-Fit were selected as search engines, and the standard CAH2_BOVIN was used as model to set up protein search standard conditions. Results show that Swissprot is the better database for protein PMF study, the better missed cleavage is one site, the better peptide tolerance is ±1 Da, and average peptide mass type is better than monoisotopic type to get target proteins. Then human Hsp70s protein mass data was used as sample to check the accuracy of MS search standard conditions. Results show that the optimized databases and parameters are reliable.

Satellite simulator is the simulation of a satellite. It constructs a realistic mission environment to test mission control system before the satellite is launched. And it can be used to train mission operators in the control center to improve their abilities of flight control. Fidelity of a simulator is very important. It can be greatly improved by directly embedding onboard software of the control subsystem into the simulator. At the same time, high-accuracy orbit dynamic simulating the complex space environment between the moon and the earth is also applied in a simulator to improve the fidelity of a simulator.
