In nature, microbes do not thrive in seclusion but are involved in complex interactions within- and between-microbial kingdoms. Among these, symbiotic associations with mycorrhizal fungi and nitrogen-fixing bacteria are namely known to improve plant health, while providing resources to benefit other microbial members. Yet, it is not clear how these microbial symbionts interact with each other or how they impact the microbiota network architecture. We used an extensive co-occurrence network analysis, including rhizosphere and roots samples from six plant species in a natural desert in AlUla region (Kingdom of Saudi Arabia) and described how these symbionts were structured within the plant microbiota network. We found that the plant species was a significant driver of its microbiota composition and also of the specificity of its interactions in networks at the microbial taxa level. Despite this specificity, a motif was conserved across all networks, i.e., mycorrhizal fungi highly covaried with other mycorrhizal fungi, especially in plant roots—this pattern is known as assortativity. This structural property might reflect their ecological niche preference or their ability to opportunistically colonize roots of plant species considered non symbiotic e.g., H. salicornicum, an Amaranthaceae. Furthermore, these results are consistent with previous findings regarding the architecture of the gut microbiome network, where a high level of assortativity at the level of bacterial and fungal orders was also identified, suggesting the existence of general rules of microbiome assembly. Otherwise, the bacterial symbionts Rhizobiales and Frankiales covaried with other bacterial and fungal members, and were highly structural to the intra- and inter-kingdom networks. Our extensive co-occurrence network analysis of plant microbiota and study of symbiont assortativity, provided further evidence on the importance of bacterial and fungal symbionts in structuring the global plant microbiota network. Objective: In this study, we explored the effect of fruit and vegetable waste addition on the quality of corn stalk silage. Methods: Corn stalks were ensiled 20 days after ear harvesting and mixed with fruit and vegetable waste (FVW) consisting of apple, orange, broccoli, and Chinese cabbage waste as 3% of fresh matter. Fruit waste consisted of solid residue obtained after juicing, and vegetable waste was collected from farms and cut into small pieces (2 to 3 cm). The materials were stored anaerobically in 20-L silo buckets and opened after 60 days of fermentation. Results: There were significant differences in dry matter (DM), acid detergent fiber (ADF), neutral detergent fiber (NDF), total digestible nutrient (TDN), and relative feed value (RFV) levels in FVW derived from all tested raw materials (p<0.05). Corn stalk mixed with orange waste (CSOW) had the highest DM content (28.77%), lowest ADF and NDF content (47.78% and 26.62% of DM, respectively), and highest TDN and RFV content (69.21 and 133, respectively). After 60 days, there were significant differences in all chemical parameters examined (p<0.05). Corn stalk mixed with broccoli waste (CSBW) had the lowest DM loss (2.23%), and the CSOW group had the lowest NDF and ADF content and highest in vitro DM digestibility. CSBW had the lowest pH and ammonia nitrogen content, but the highest lactic acid/acetic acid ratio among the treatment groups. CSOW had the highest lactic acid content (2.27% of DM). The microbial contents of each group differed only in lactic acid bacteria counts before and after ensiling, showing a slight increase (p>0.05) and significant decreases in yeast and mold counts (p<0.05) after ensiling. Conclusion: These findings confirmed that mixing various FVW materials, particularly orange waste, with corn stalks improved the nutritional value of silage. Mining activities have resulted in a substantial accumulation of cadmium (Cd) in agricultural soils, particularly in southern China. Long-term Cd exposure can cause plant growth inhibition and various diseases. Rapid identification of the extent of soil Cd pollution and its driving factors are essential for soil management and risk assessment. However, traditional geostatistical methods are difficult to simulate the complex nonlinear relationships between soil Cd and potential features. In this study, sequential extraction and hotspot analyses indicated that Cd accumulation increased significantly near mining sites and exhibited high mobility. The concentration of Cd was estimated using three machine learning models based on 3169 topsoil samples, seven quantitative variables (soil pH, Fe, Ca, Mn, TOC, Al/Si and ba value) and three quantitative variables (soil parent rock, terrain and soil type). The random forest model achieved marginally better performance than the other models, with an R2 of 0.78. Importance analysis revealed that soil pH and Ca and Mn contents were the most significant factors affecting Cd accumulation and migration. Conversely, due to the essence of controlling Cd migration being soil property, soil type, terrain, and soil parent materials had little impact on the spatial distribution of soil Cd under the influence of mining activities. Our results provide a better understanding of the geochemical behavior of soil Cd in mining areas, which could be helpful for environmental management departments in controlling the diffusion of Cd pollution and capturing key targets for soil remediation. The inclusion of pulse crops in Canadian rotations has the potential to improve cropping system efficiencies, reduce the overall amount of applied nitrogen, provide economic opportunities for producers, and reduce the overall carbon footprint of the cropping system. Although primarily grown in western Canada, many pulse species—field pea in particular, are well suited to temperate growing conditions in the Maritime region of Canada. A study was conducted over 2 years at Harrington, Prince Edward Island, and consisted of four field pea varieties including two yellow varieties (AAC Lacombe and CDC Saffron) and two green varieties (CDC Limerick and CDC Raezer) planted at three plant population densities: 75, 100, and 125 plants m−2. The study also measured the effects of nitrogen fertilizer applied pre-plant (0 kg ha−1 vs. 15 kg ha−1) and applied plant available sulfur (0 kg ha−1 vs. 25 kg ha−1). Overall, yellow pea varieties were higher yielding than green pea varieties, and there was a linear increase in yield with increased seeding rate. There were no significant effects of pre-plant nitrogen fertilizer on yield, although it did slightly increase seed protein. Applied sulfur had a positive effect on yield and a slightly negative effect on thousand seed weight. This experiment provides a recommendation for the optimal seeding rate (100 plants m−2) and fertility recommendations to achieve profitable yields growing field pea in the Maritime region of Canada. Zinc solubilizing bacteria inhabiting root endosphere possesses great potential to enhance plant yield by solubilizing the nutrients. The potential of Zn solubilizing bacteria Bacillus sp. SH-10 and Bacillus cereus SH-17 to improve yield and grain zinc content of rice was investigated under different regimes of individual and co-inoculation in the presence and absence of chemical fertilizer zinc sulfate (ZnSO4). The strains were applied to the rice varieties basmati 385 and super basmati under field conditions for two consecutive years. A significantly improved growth of rice plants, such as plant height (102–118 cm), number of tillers per plant (8.5–11.5), chlorophyll content (29.5–35.1), zinc requiring enzymes, i.e., superoxide dismutase (396–570 per gram fresh weight (g−1 FW)), carbonic anhydrase activity (CA) (10–15.06 U g−1 FW) and grain yield (3.0–3.8 tons ha−1), was observed in the plants inoculated with Bacillus sp. in the presence of chemical fertilizer (ZnSO4). Consortium of zinc solubilizing bacteria also caused higher grain Zn content (25.0–30.5 mg kg−1) of the rice varieties basmati 385 and super basmati as compared to that of un-inoculated plants. Hence, it can be concluded that Zn solubilizing bacteria has immense potential to be used as agricultural crop inoculants as they synergize the effect of chemical Zn, increase yield and improve the nutritional value of crops. PGPR and PGPF occupy diverse niches within the rhizosphere, playing a crucial role in soil nutrient cycling and influencing a wide range of plant physiological processes. This review examines the primary mechanisms employed by these microbial agents to promote growth, as well as the strategy of co-inoculation to enhance product efficacy. Furthermore, we provide a comprehensive analysis of the microbial inoculants currently available in Brazil, detailing the microorganisms accessible for major crops, and discuss the market's prospects for the research and development of novel products in light of current challenges faced in the coming years. Introduction: Rhizosphere effects (REs) have recently been identified as important regulators of root and microbial nutrient acquisition and are positively involved in nutrient cycling of belowground carbon (C), nitrogen (N), and phosphorus (P). Nutrient conditions of the fine roots and soil N are likely to influence REs. Still, it is unclear how REs of soil nutrients themselves variably impact the supply of nutrients to plants in terms of the responses to soil N due to succession. Methods: In this study, we applied both fine roots and extracellular enzymes for vector analysis and stoichiometry of N:P to explore the metabolic limitations of roots and rhizospheric soil microbes and their relationships with REs across five levels of soil N (0, 5, 10, 15, and 20 kg N m-2 year-1) along successional age classes of 42, 55, and 65 years in a Pinus tabuliformis forest. Results: Overall, the metabolism of root and rhizospheric soil microbes was mediated by soil N. N limitation of roots initially decreased before increasing, whereas that of microbes demonstrated opposite trends to the N levels owing to competition for inorganic N between them by REs of NO3--N. However, N limitations of both roots and microbes were alleviated in young stands and increased with succession after the application of N. In addition, root N limitations were manipulated by REs of three different soil N-related indicators, i.e., total N, NH4 +-N, and NO3 --N. Rhizospheric soil microbial N limitation was almost unaffected by REs due to their strong homeostasis but was an important driver in the regulation of root N limitation. Discussion: Our results indicated that successional age was the most critical driver that directly and indirectly affected root N metabolism. However, the level of N application had a slight effect on root N limitation. Microbial N limitation and variations in the REs of N indicators regulated root N limitation in the rhizosphere. As a result, roots utilized REs to sequester N to alleviate N limitations. These findings contribute to novel mechanistic perspectives on the sustainability of N nutrition by regulating N cycling in a system of plant-soil-microbes in the rhizosphere to adapt to global N deposition or the heterogeneous distribution of bioavailable soil N with succession. This work aimed to assess microbial inoculants (Lactiplantibacillus plantarum and Lentilactobacillus buchneri), chemical additives (natamycin and hexanoic acid), and their combination on fermentation characteristics and aerobic stability in total mixed ration (TMR) silage. The TMR consisted of 30% water bamboo shell (WBS), 10% alfalfa, 20% rice straw, and 40% concentrate. There were six treatments as follows: (1) deionized water (control, CON). (2) lactic acid bacteria (Lactiplantibacillus plantarum + Lentilactobacillus buchneri; LPB, 1 × 106 cfu/g FW). (3) natamycin (NT, 0.02 g/kg FW). (4) hexanoic acid (HA, 0.02 g/kg FW). (5) lactic acid bacteria + natamycin (SLNT, 0.02 g/kg FW). (6) lactic acid bacteria + hexanoic acid (SLHA, 0.02 g/kg FW). After fermentation, laboratory silos (10 L) were opened to assess fermentation quality, followed by a 6-day aerobic stability test. The results showed that all silages were well fermented with high lactic acid (LA) content, low ammonia nitrogen (NH3-N), and negligible butyric acid (BA) levels. Among all silages, SLNT silage exhibited the greatest LA, acetic acid (AA) levels, LAB counts, and the lowest pH and NH3-N. For aerobic stability, all additives significantly (p < 0.05) enhanced aerobic stability, delayed (p < 0.05) the decrease in LA and water-soluble carbohydrates (WSC) and the increase in pH, and significantly (p < 0.05) minimized yeast proliferation. The SLNT silage showed the best aerobic stability, with SLHA, NT, HA, and LPB following. This is the first study that investigates the effect of kefir with an emphasis on the production of short-chain fatty acids (SCFAs) during the fermentation process in food products. The products developed and characterized were an artisanal cream cheese without cream and one with added cream, and for the analysis of the fatty acid profile, both cream cheeses were compared with commercial cream cheese. The artisanal cream cheese had a high amount of lactic acid bacteria characterizing the product formed by Lactobacilli and a low concentration of lactose due to the fermentation process. Compared to commercial cream cheese, our products without and with added cream had a higher concentration of short-chain fatty acids (SCFAs), especially butyric acid, which is important for the health of the gastrointestinal tract, omega 3, and oleic fatty acid, which has been associated with the prevention and control of some diseases. Overall, the artisanal cream cheese cream with fermented cream with kefir grains is a functional product with an innovative character compared to current products on the market and was well accepted by the younger public. Beneficial microbes colonizing plant tissues are recognized as a value-added soil amendment for enhancing soil fertility. The present study aimed to evaluate the effects of co-inoculation with plant growth-promoting endophytic actinobacteria and nitrogen-fixing rhizobacteria for improved growth attributes, soil properties, microbial activity, yield, and nutrient uptake of wheat under field conditions for two consecutive years. Three endophytic actinobacterial isolates, Streptomyces sp. strain WR27, Streptomyces sp. strain WR15, and Streptomyces sp. strain WS6 were evaluated alone and in combination with nitrogen-fixing bacteria, Azotobacter in field experiment with recommended fertilization rate in north-western Indian plains. Results of the study indicated that all the growth attributes were higher in treatments receiving microbial inoculants than un-inoculated control. It was also observed that inoculation with actinobacterial isolates significantly increased total number of tillers m−2, spike length, number of grains spike−1, and 1000 grain weight as compared to control. Streptomyces sp. strain WR27 + Azotobacter and Streptomyces sp. strain WR15 + Azotobacter exhibited maximum grain yield (5.7 t ha−1, respectively) as compared to control (5.0 t ha−1). The treatment involving inoculation with Streptomyces sp. strain WR27 + Azotobacter recorded higher nutrient contents. Soil properties were significantly increased by application of actinobacterial inoculants in both years over the un-inoculated control. Furthermore, among different inocula, Streptomyces sp. strain WR27 + Azotobacter dual inoculation exhibited highest response efficiency for all the traits studied. Therefore, the results revealed multiple benefits of plant growth-promoting actinobacteria and Azotobacter inoculation for wheat crop in terms of improved soil properties and macronutrient content without hampering the soil fertility and productivity. (1) Background: Plant diversity has long been assumed to predict soil microbial diversity. The mutualistic symbiosis between forest trees and ectomycorrhizal (EM) fungi favors strong correlations of EM fungal diversity with host density in terrestrial ecosystems. Nevertheless, in contrast with host tree effects, neighboring plant effects are less well studied. (2) Methods: In the study presented herein, we examined the α-diversity, community composition, and co-occurrence patterns of EM fungi in Quercus acutissima across different forest types (pure forests, mixed forests with Pinus tabuliformis, and mixed forests with other broadleaved species) to ascertain how the EM fungi of focal trees are related to their neighboring plants and to identify the underlying mechanisms that contribute to this relationship. (3) Results: The EM fungal community exhibited an overall modest but positive correlation with neighboring plant richness, with the associations being more pronounced in mixed forests. This neighboring effect was mediated by altered abiotic (i.e., SOC, TN, LC, and LP) and biotic (i.e., bacterial community) factors in rhizosphere soil. Further analysis revealed that Tomentella_badia, Tomentella_galzinii, and Sebacina_incrustans exhibited the most significant correlations with plant and EM fungal diversity. These keystone taxa featured low relative abundance and clear habitat preferences and shared similar physiological traits that promote nutrient uptake through contact, short-distance and medium-distance smooth contact-based exploration types, thereby enhancing the potential correlations between EM fungi and the neighboring plant community. (4) Conclusions: Our findings contribute to the comprehension of the effect of neighboring plants on the EM fungal community of focal trees of different forest communities and the biodiversity sensitivity to environmental change. Summary: Elevated air temperature (Tair) and vapour pressure deficit (VPDair) significantly influence plant functioning, yet their relative impacts are difficult to disentangle.We examined the effects of elevated Tair (+6°C) and VPDair (+0.7 kPa) on the growth and physiology of six tropical tree species. Saplings were grown under well‐watered conditions in climate‐controlled glasshouses for 6 months under three treatments: (1) low Tair and low VPDair, (2) high Tair and low VPDair, and (3) high Tair and high VPDair. To assess acclimation, physiological parameters were measured at a set temperature.Warm‐grown plants grown under elevated VPDair had significantly reduced stomatal conductance and increased instantaneous water use efficiency compared to plants grown under low VPDair. Photosynthetic biochemistry and thermal tolerance (Tcrit) were unaffected by VPDair, but elevated Tair caused Jmax25 to decrease and Tcrit to increase. Sapling biomass accumulation for all species responded positively to an increase in Tair, but elevated VPDair limited growth.This study shows that stomatal limitation caused by even moderate increases in VPDair can decrease productivity and growth rates in tropical species independently from Tair and has important implications for modelling the impacts of climate change on tropical forests. The objective of this study was to evaluate the effect of microbial inoculants, chitosan, and blend (microbial inoculants and chitosan) on chemical composition, in vitro degradability, fermentative profile, losses, aerobic stability, and microorganism counts of corn, pearl millet and sorghum silages. A randomised block design was used in a factorial arrangement of 3 × 4. Three crops (pearl millet, corn and sorghum) and four additives: Control (without additive), 2 g of microbial inoculants (for ton of forage), 10 g/kg chitosan per kg of ensiled material, and blend (chitosan [5 g/kg] plus microbial inoculants [2 g/ton forage]), with five replicates per treatment. Chitosan increases the CP levels of silages and improves the in vitro degradability of DM in sorghum silage. The microbial inoculants reduced the pH in pearl millet, corn and sorghum silages (P < 0.001). The additives affected (P < 0.05) the aerobic stability of silages throughout the evaluation period. However, when evaluating the chitosan, the average temperature of the pearl millet and corn silages remained stable for a longer period. Chitosan improved the nutritional quality of sorghum silage. Chitosan-treated silages pearl millet and corn silages show great efficiency in aerobic stability due to antifungal activity. The microbial inoculants show reduced gas losses compared to chitosan. Objective: This meta-analysis aimed to evaluate the impact of wilted and unwilted silage on various parameters, such as nutrient content, fermentation quality, bacterial populations, and digestibility. Methods: Thirty-six studies from Scopus were included in the database and analyzed using a random effects model in OpenMEE software. The studies were grouped into two categories: wilting silage (experiment group) and non-wilting silage (control group). Publication bias was assessed using a fail-safe number. Results: The results showed that wilting before ensiling significantly increased the levels of dry matter, water-soluble carbohydrates, neutral detergent fiber, and acid detergent fiber, compared to non-wilting silage (p<0.05). However, wilting significantly decreased dry matter losses, lactic acid, acetic acid, butyric acid, and ammonia levels (p<0.05). The pH, crude protein, and ash contents remained unaffected by the wilting process. Additionally, the meta-analysis revealed no significant differences in bacterial populations, including lactic acid bacteria, yeast, and aerobic bacteria, or in vitro dry matter digestibility between the two groups (p>0.05). Conclusion: Wilting before ensiling significantly improved silage quality by increasing dry matter and water-soluble carbohydrates, as well as reducing dry matter losses, butyric acid, and ammonia. Background and aim: Seed inoculation with effective rhizobia (Rh) strains and phosphorus (P) application are sustainable agricultural practices that enhance soybean grain yield. However, adoption of these practices in northern Ghana is hindered by unpredictable yield responses and poor understanding of the factors that cause yield variation. We compared soybean yield from inputs managed trials such as Rh inoculants and P sources. Methods: Soybean trials were conducted on-station and on-farm with one of three Rh inoculants [Bradyrhizobium japonicum (Bj), B. elkanii (Be), and B. diazoefficiens (Bd)], and with one of three P sources [no-P, Triple Superphosphate (TSP), and a mixture of rock-P (low solubility) and TSP (water-soluble P)]. Yield was compared at harvest. The random forest (RF) model was used to predict yield, and factors of yield variation were determined using forward redundancy analysis (RDA). Results: The soybean yield was consistently higher in the Rh × P treatments compared to the control in on-station and on-farm experiments. The treatments that received P fertilizer (RP and TSP) showed a higher grain yield than the control. The on-station experiment yielded more accurate results than the on-farm dataset, with a trained coefficient of determination (R2) of 0.95 and 0.87, respectively. The yield variation was explained by the Rh × P fertilizer, P sources, Rh strains, and exchangeable soil Mg. Conclusion: The exchangeable magnesium in the soil contributed significantly to the variation in yield. The study aimed to investigate the impact of zinc (Zn) nutrition on enzymatic activity in soil and plant systems of zinc-deficient soils. Additionally, it aimed to understand the impact of solubilizer (such as Zinc Solubilizing Bacteria) and mobilizer (like AM Fungi) on enzyme activity and zinc nutrition in plants across calcareous and non-calcareous soil environments. A pot experiment was carried out with two different soils with three Zn sources (ZnSO4, ZnO and Zn-EDTA), organic manure (FYM) and microbial inoculants - Zinc Solubilizing Bacteria (ZSB) and Arbuscular Mycorrhizal Fungi (AM F). The DTPA-Zn and plant Zn content increased with applied sources of Zn. The microbial inoculants and FYM enhanced the use efficiency of all sources of Zn compared to unapplied treatments. Regarding soil enzyme activity, FYM and AM fungi played a major role which in turn altered the dehydrogenase (SDHA) and alkaline phosphatase (AlP) activities in both soils. The Zn sources and ZSB addition enhanced the plant enzyme activity. Calcareous soil recorded higher CA, SOD and alkaline phosphatase activities compared to non-calcareous soil the latter registered higher dehydrogenase activity and fruit Zn content. Biostimulants can be used in sustainable agriculture as they are regarded as inputs with minimal environmental impact. When applied to the root systems of certain plant species, microbial inoculants confer physiological and morphological benefits to the entire plant. In the present study, we focused on some morphometric parameters of the root system of two Crithmum maritimum L. spontaneous populations (Mediterranean and Atlantic). We compared the effects of two different (for number of strains) biostimulants on the weight, length, diameter, and number of tips on the root system. The experiment was conducted in two controlled environments: i) an incubator in which seeds were placed in Petri dishes on Whatman filter paper and ii) a greenhouse where seeds were placed in pots containing seeding substrate. Under incubator conditions, the biostimulant application showed, on average, a stimulating effect on the analyzed root parameters. Specifically, it increased weight by approximately 85%, diameter by 25%, and length by about 128% compared to the control. Similarly, under greenhouse conditions, the biostimulant application showed favorable effects on the analyzed root variables, exhibiting an increase in root weight by approximately 66%, diameter by 25%, length by about 75%, and number of tips by 56% compared to the control. The present study suggests the potential use of biostimulants during the nursery phases of production to ease the establishment of alternative crops such as Crithmum maritimum in cropping systems for which it is necessary to implement the agronomic technique and encourage rapid colonization in contexts otherwise at risk of desertification. BACKGROUND: The plant growth‐promoting bacteria (PGPB) Azospirillum brasilense is widely used as an inoculant for important grass crops, providing numerous benefits to the plants. However, one limitation to develop viable commercial inoculants is the control of PGPB survival, requiring strategies that guarantee their survival during handling and field application. The application of sublethal stress appears to be a promising strategy to increase bacterial cells tolerance to adverse environmental conditions since previous stress induces the activation of physiological protection in bacterial cell. In this work, we evaluated the effects of thermal and salt stresses on the survival of inoculant containing A. brasilense Ab‐V5 and Ab‐V6 strains and we monitored A. brasilense viability in inoculated maize roots after stress treatment of inoculant. RESULTS: Thermal stress application (> 35 °C) in isolated cultures for both strains, as well as salt stress [sodium chloride (NaCl) concentrations > 0.3 mol L−1], resulted in growth rate decline. The A. brasilense enumeration in maize roots obtained by propidium monoazide quantitative polymerase chain reaction (PMA‐qPCR), for inoculated maize seedlings grown in vitro for 7 days, showed that there is an increased number of viable cells after the salt stress treatment, indicating that A. brasilense Ab‐V5 and Ab‐V6 strains are able to adapt to salt stress (0.3 mol L−1 NaCl) growth conditions. CONCLUSION: Azospirillum brasilense Ab‐V5 and Ab‐V6 strains had potential for osmoadaptation and salt stress, resulting in increased cell survival after inoculation in maize plants. © 2024 Society of Chemical Industry. In a three-year field study, we inoculated two potato varieties with a selection of four beneficial microbial strains (i.e. Rhizophagus irregularis MUCL41833, Trichoderma asperelloides A, Pseudomonas brassicacearum 3Re2-7 and Paraburkholderia phytofirmans PsJN), alone or in combination. Plants were grown under rainfed or irrigated conditions, and potato yield and development of several diseases were evaluated. The microbial inoculants were traced in the root system at different stages of crop development via molecular markers. Whatever the water supply, the inoculants had no effect on yield. Conversely, some of the inoculants were able to lower the incidence and/or severity of several blemish diseases, namely common scab-associated symptoms (CSAS) and silver scurf/black dot-associated symptoms (SSAS). Microbial consortia were more efficient in decreasing symptoms compared to single strain inoculations. The best control was obtained with the combination of R. irregularis and P. brassicacearum, which reduced the incidence of CSAS by 22% and severity of SSAS by 21%. Root tracking revealed that P. brassicacearum and P. phytofirmans PsJN were able to establish in the root system of the potato, while only P. brassicacearum was detected from emergence until flowering of the plants. Purpose: Clean and efficient alternatives for sustainable treatment of pig farming wastes are a priority in suburban areas in Forest Reserves of High Andean ecosystems. Among these technologies are Bokashi Fermentation and composting with added microorganisms (bioaugmentation) or nutrients (biostimulation) as biological activators. Method: In a Colombian rural area located at 3160 m.a.s.l., focused on small-scale swine production, Bokashi fermentation with active dry yeast and in-vessel composting were evaluated using two microbial inoculants and a sterile metabolic stimulant. In addition, these treatments were compared with a non-inoculated composting process used in the study area (control). Results: The biological additives improved the composting process performance and end-product quality by shortening the degradation cycle (19.9−45.9%), higher germination index (> 0.8%), increasing nutrient content (N, P, S, Fe and Cu) and showing suppressive activity against Fusarium oxysporum compared to control. Nevertheless, Bokashi was the only treatment that decreased Enterobacteriaceae concentration below 1E+03 CFU g−1. Conclusion: Bokashi technology complies with Colombia’s environmental regulations for eco-friendly fertilizer production, which allows the sustainable management by-products from swine activities in critical ecological areas such as Forestry Protection Reserves in High Andean ecosystems. Seed inoculation with symbiotic bacteria is a commonly employed practice in soybean cultivation. As a result, nodulation proceeds properly and plants self-supply atmospheric nitrogen, requiring either minimal or no additional nitrogen fertilization. The aim of the study was to investigate the response of soybeans to the application of the recommended or double dose of commercial inoculants (HiStick® Soy or TURBOSOY®) and/or mineral nitrogen fertilization compared to the untreated control. It was demonstrated that a double dose of the tested preparations had the most favorable effect on nodulation. However, the impact of weather conditions modified their effectiveness during the study years, which was especially visible in 2022. Sowing seeds without inoculation (control) resulted in the formation of sparse root nodules and consequently the lowest leaf area index (LAI) and soil plant analysis development (SPAD) measurements. In addition, the values of SPAD and LAI indices varied across the years of the study, indicating that weather conditions modified nitrogen uptake by plants. Overall, seed inoculation and/or nitrogen fertilization positively influenced the chemical composition of seeds compared to the control. The only decrease observed was in the oil content, while the double dose of HiStick® Soy preparation reduced the polyphenol content. The double dose of the tested inoculants had the most favorable impact on yield components and seed yield. However, applying inoculation at the recommended dose or in combination with nitrogen fertilization yielded similar or slightly worse results, depending on the year. Therefore, soybean seed inoculation should be recommended, although the effectiveness of the procedure will depend on various factors, including the type of inoculant, dosage, nitrogen fertilization, and weather conditions. Whole crop corn silage (WCCS) been an important source of roughage for confined ruminants. However, at the silage feed-out phase, the rise in temperature and relative humidity under aerobic conditions breeds the production of undesirable microorganisms, such as yeast and mold. In order to investigate the conservation characteristics and aerobic stability underlying the effects of additives in whole crop corn silage (WCCS), whole crop corn (WCC) at the milk-ripe stage was ensiled with Lentilactobacillus (L.) buchneri (LB) and different proportions of Artemisia argyi (AA) for 90 days (d) at room temperature, respectively, and aerobic exposure after 90 d fermentation was also conducted. The study found that AA as an additive improved the fermentation quality and enhanced aerobic stability of WCCS, for which the addition of 60% AA increased the lactic acid fermentation rate, with the lactic acid concentration at the end of aerobic exposure significantly higher than in all other treatment groups, at 98.21 g/kg DM (p < 0.01), which decreased the relative abundance of none wanted microorganisms and reduced the content of fungal toxins (p < 0.05). After 90 d of fermentation, LB also increased the organic acids and reduced the pH compared with control, thereby improving fermentation quality. Furthermore, we also discovered that the relative abundance of Candida within the 60% AA was the highest. Candida have the ability to convert WSC into organic acids and lower pH, thus improving the quality of silage. Particularly, 60% AA could improve the fermentation quality and aerobic stability of silage through the biosynthetic pathways of phenylalanine, tyrosine and tryptophan, as well as by participation in the hydrolysis of glycoside hydrolases (GHs). Unexpectedly, the addition of AA was found to reduce the relative abundance of antibiotic resistance genes. WCC, ensiled with 60% AA, exhibited excellent fermentation quality and aerobic stability, providing a theoretical basis for understanding the mechanisms of AA which improve the quality of WCCS during the aeration phase. The combined application of chemical fertilizers and organic material is a transitional approach toward more sustainable and productive farming. This study aimed to investigate the response of low chemical fertilizer rates and supplementing with organic materials on soil health and fruit quality. In plum orchard, combination of reduced chemical fertilizer rates (R80; R70; R60) with organic manures (F; V) at varying levels based the loss in nitrogen requirement and microbial inoculants (Bf; J) were studied over the conventional approach (R100) on soil health and fruit quality in a 2-year experiment. Soil analysis showed soil physico-chemical properties and enzymes activity with application of R70+V10+Bf. Compared to integrated treatments R100 saw highest availability of soil potassium and calcium. Application of R70+V10+J resulted in maximum improvement in fruit weight, TSS and soluble sugar content. Fruit set percentage was higher in treatment R70+V10+Bf. Fruit yield showed significant changes with treatments R70+V10+Bf and R70+V10+J having similar yield to R100. Our results suggest that by reducing the amount of chemical fertilizer by 30% and partially replacing it with organic materials had a positive effect on soil health without affecting the fruit production and quality. Introduction: There is a need to rebuild soil health by implementing regenerative agricultural practices across cropping systems dependent on agrochemicals, such as in United States corn production. One increasingly utilized regenerative practice is the application of biostimulants, or microbial inoculants that can rebuild soil health and productivity. In this study, we describe results from a multi-year corn trial conducted to quantify the impact of Re-Gen, a biostimulant invented to recycle plant biomass and increase nutrient bioavailability in the soil. Methods: Re-Gen was applied across four fields (VA01, BH01, VA04, and LA05) at a dairy cattle farm in Ferrisburgh, VT. Results: Over the two consecutive years (2022-2023) of the trial, Re-Gen application on corn stover and cover crop residues increased ear corn yield by 24% and corn silage yield by 12.5-30%, depending on the field. Analysis of soil nutrients and plant tissues showed increased nutrients, particularly in field LA-05. Multi-year Re-Gen application increased yield and generated additional economic value, indicating that the effects of Re-Gen do not diminish with multiple applications. The increased corn silage yield correlated to increased potential milk yield from cows fed silage grown in Re-Gen-treated fields. Further investigation into the mechanism suggests that increased phosphatase production Re-Gen could contribute to increased phosphorus bioavailability in the soil and uptake in the tissue, potentially increasing yields. Discussion: These results highlight the potential for Re-Gen to foster regenerative agriculture processes on cropland and livestock farms while also increasing corn and milk yield and, therefore, revenue for corn farmers in the United States and for similar cropping systems worldwide. Background: Current RNA-seq analysis software for RNA-seq data tends to use similar parameters across different species without considering species-specific differences. However, the suitability and accuracy of these tools may vary when analyzing data from different species, such as humans, animals, plants, fungi, and bacteria. For most laboratory researchers lacking a background in information science, determining how to construct an analysis workflow that meets their specific needs from the array of complex analytical tools available poses a significant challenge. Results: By utilizing RNA-seq data from plants, animals, and fungi, it was observed that different analytical tools demonstrate some variations in performance when applied to different species. A comprehensive experiment was conducted specifically for analyzing plant pathogenic fungal data, focusing on differential gene analysis as the ultimate goal. In this study, 288 pipelines using different tools were applied to analyze five fungal RNA-seq datasets, and the performance of their results was evaluated based on simulation. This led to the establishment of a relatively universal and superior fungal RNA-seq analysis pipeline that can serve as a reference, and certain standards for selecting analysis tools were derived for reference. Additionally, we compared various tools for alternative splicing analysis. The results based on simulated data indicated that rMATS remained the optimal choice, although consideration could be given to supplementing with tools such as SpliceWiz. Conclusion: The experimental results demonstrate that, in comparison to the default software parameter configurations, the analysis combination results after tuning can provide more accurate biological insights. It is beneficial to carefully select suitable analysis software based on the data, rather than indiscriminately choosing tools, in order to achieve high-quality analysis results more efficiently. An important goal of environmental and comparative physiology research is to identify species or populations that may be susceptible to environmental change such as heat wave events that are predicted to become more frequent and intense in the future. This study tested the hypothesis that fishes inhabiting alkaline lakes face significant physiological challenges, which results in reduced thermal tolerance. Brook stickleback (Culaea inconstans) were collected from an alkaline lake (pH 9.3) in Alberta, Canada and held under neutral conditions in the laboratory. Subsequently, fish were acutely exposed (4 d) to neutral (pH 7) or alkaline (pH 9.5) waters at 10 or 25°C. Exposure to alkaline water reduced critical thermal maximum (CTmax) in stickleback by approximately 1°C, but thermal acclimation capacity ("thermal plasticity") was unaffected by alkaline exposure. Alkaline conditions resulted in physiological disturbances characteristic of exposure to high pH including elevated whole-body ammonia and lactate concentrations. Acute warming to CTmax in alkaline-exposed fish resulted in reductions in whole-body sodium and chloride concentrations. In addition, alkaline exposure compromised recovery from exercise at elevated temperatures. Overall, these results suggest that the physiological disturbances observed in response to alkaline exposure may render fish more susceptible to acute warming, reducing thermal tolerance. Background: On tropical regions, phosphorus (P) fixation onto aluminum and iron oxides in soil clays restricts P diffusion from the soil to the root surface, limiting crop yields. While increased root surface area favors P uptake under low-P availability, the relationship between the three-dimensional arrangement of the root system and P efficiency remains elusive. Here, we simultaneously assessed allelic effects of loci associated with a variety of root and P efficiency traits, in addition to grain yield under low-P availability, using multi-trait genome-wide association. We also set out to establish the relationship between root architectural traits assessed in hydroponics and in a low-P soil. Our goal was to better understand the influence of root morphology and architecture in sorghum performance under low-P availability. Result: In general, the same alleles of associated SNPs increased root and P efficiency traits including grain yield in a low-P soil. We found that sorghum P efficiency relies on pleiotropic loci affecting root traits, which enhance grain yield under low-P availability. Root systems with enhanced surface area stemming from lateral root proliferation mostly up to 40 cm soil depth are important for sorghum adaptation to low-P soils, indicating that differences in root morphology leading to enhanced P uptake occur exactly in the soil layer where P is found at the highest concentration. Conclusion: Integrated QTLs detected in different mapping populations now provide a comprehensive molecular genetic framework for P efficiency studies in sorghum. This indicated extensive conservation of P efficiency QTL across populations and emphasized the terminal portion of chromosome 3 as an important region for P efficiency in sorghum. Increases in root surface area via enhancement of lateral root development is a relevant trait for sorghum low-P soil adaptation, impacting the overall architecture of the sorghum root system. In turn, particularly concerning the critical trait for water and nutrient uptake, root surface area, root system development in deeper soil layers does not occur at the expense of shallow rooting, which may be a key reason leading to the distinctive sorghum adaptation to tropical soils with multiple abiotic stresses including low P availability and drought. Distilled grain waste (DGW) is rich in nutrients and can be a potential resource as animal feed. However, DGW contains as much as 14% lignin, dramatically reducing the feeding value. White-rot fungi such as Pleurotus ostreatus could preferentially degrade lignin with high efficiency. However, lignin derivatives generated during alcohol distillation inhibit P. ostreatus growth. Thus, finding a new strategy to adjust the DGW properties to facilitate P. ostreatus growth is critical for animal feed preparation and DGW recycling. In this study, three dominant indigenous bacteria, including Sphingobacterium thermophilum X1, Pseudoxanthomonas byssovorax X3, and Bacillus velezensis 15F were chosen to generate single and compound microbial inoculums for DGW composting to prepare substrates for P. ostreatus growth. Compared with non-inoculated control or single microbial inoculation, all composite inoculations, especially the three-microbial compound, led to faster organic metabolism, shorter composting process, and improved physicochemical properties of DGW. P. ostreatus growth assays showed the fastest mycelial colonization (20.43 μg·g-1 ergosterol) and extension (9 mm/d), the highest ligninolytic enzyme activities (Lac, 152.68 U·g-1; Lip, 15.56 U·g-1; MnP, 0.34 U·g-1; Xylanase, 10.98 U·g-1; FPase, 0.71 U·g-1), and the highest lignin degradation ratio (30.77%) in the DGW sample after 12 h of composting with the three-microbial compound inoculation when compared to other groups. This sample was relatively abundant in bacteria playing critical roles in amino acid, carbohydrate, energy metabolism, and xenobiotic biodegradation, as suggested by metagenomic analysis. The feed value analysis revealed that P. ostreatus mycelia full colonization in composted DGW led to high fiber content retention and decreased lignin content (final ratio of 5% lignin) but elevated protein concentrations (about 130 g·kg-1 DM). An additional daily weight gain of 0.4 kg/d was shown in cattle feeding experiments by replacing 60% of regular feed with it. These findings demonstrate that compound inoculant consisting of three indigenous microorganisms is efficient to compost DGW and facilitate P. ostreatus growth. Background: The need for addition of external electron donors such as ethanol or lactate impairs the economic viability of chain elongation (CE) processes for the production of medium-chain carboxylates (MCC). However, using feedstocks with inherent electron donors such as silages of waste biomass can improve the economics. Moreover, the use of an appropriate inoculum is critical to the overall efficiency of the CE process, as the production of a desired MCC can significantly be influenced by the presence or absence of specific microorganisms and their metabolic interactions. Beyond, it is necessary to generate data that can be used for reactor design, simulation and optimization of a given CE process. Such data can be obtained using appropriate mathematical models to predict the dynamics of the CE process. Results: In batch experiments using silages of sugar beet leaves, cassava leaves, and Elodea/wheat straw as substrates, caproate was the only MCC produced with maximum yields of 1.97, 3.48, and 0.88 g/kgVS, respectively. The MCC concentrations were accurately predicted with the modified Gompertz model. In a semi-continuous fermentation with ensiled sugar beet leaves as substrate and digestate from a biogas reactor as the sole inoculum, a prolonged lag phase of 7 days was observed for the production of MCC (C6–C8). The lag phase was significantly shortened by at least 4 days when an enriched inoculum was added to the system. With the enriched inoculum, an MCC yield of 93.67 g/kgVS and a productivity of 2.05 gMCC/L/d were achieved. Without the enriched inoculum, MCC yield and productivity were 43.30 g/kgVS and 0.95 gMCC/L/d, respectively. The higher MCC production was accompanied by higher relative abundances of Lachnospiraceae and Eubacteriaceae. Conclusions: Ensiled waste biomass is a suitable substrate for MCC production using CE. For an enhanced production of MCC from ensiled sugar beet leaves, the use of an enriched inoculum is recommended for a fast process start and high production performance. [ABSTRACT FROM AUTHOR]

Soybean is the main oilseed cultivated worldwide. Even though Brazil is the world's largest producer and exporter of soybean, its production is severely limited by biotic factors. Soil borne diseases are the most damaging biotic stressors since they significantly reduce yield and are challenging to manage. In this context, the present study aimed to evaluate the potential of a bacterial strain (Ag109) as a biocontrol agent for different soil pathogens (nematodes and fungi) of soybean. In addition, the genome of Ag109 was wholly sequenced and genes related to secondary metabolite production and plant growth promotion were mined. Ag109 showed nematode control in soybean and controlled 69 and 45% of the populations of Meloidogyne javanica and Pratylenchus brachyurus, respectively. Regarding antifungal activity, these strains showed activity against Macrophomia phaseolina, Rhizoctonia solani, and Sclerotinia sclerotiorum. For S. sclerotiorum, this strain increased the number of healthy plants and root dry mass compared to the control (with inoculation). Based on the average nucleotide identity and digital DNA-DNA hybridization, this strain was identified as Bacillus velezensis. Diverse clusters of specific genes related to secondary metabolite biosynthesis and root growth promotion were identified, highlighting the potential of this strain to be used as a multifunctional microbial inoculant that acts as a biological control agent while promoting plant growth in soybean. Context: The ensiling process presents losses that are associated with the fermentative profile, resulting in lower nutritional value, and lower aerobic stability of silages. Aims: This study aimed to evaluate the effects of chitosan and microbial inoculants addition in Mombasa grass (Megathyrsus maximus) silage (MGS) fermentation profile and losses, chemical composition, in situ degradation, and aerobic stability. Methods: Forty experimental silos (PVC tubing with 28-cm inner diameter and 25-cm height) were used in a randomised block (n = 5) design to evaluate the following treatments: (1) MGS without additives (control, CON); (2) MGS treated with 5.0 × 104 colony-forming units (CFU) of Lactobacillus buchneri (NCIM 40788) per gram of fresh matter (LBB); (3) MGS treated with 1.6 × 105 CFU of L. plantarum and 1.6 × 105 CFU of Pediococcus acidilactici per gram of fresh matter (LPP); and (4) MGS treated with 6 g/kg DM of chitosan (CHI). Key results: The treatments did not alter the pH, ammonia-N, butyric, and lactic acid concentrations in the silage. The use of LPP reduced the ethanol content, while CHI increased propionic and branched-chain fatty acids compared with other treatments. Fermentation losses and dry-matter recovery were not affected by treatments. Chitosan reduced the organic matter of the MGS in relation to the other treatments, without having an impact on the other variables of chemical composition. The treatments did not influence the in vitro degradation, nor the pH and temperature after aerobic exposure of the silage. Conclusions: Chitosan increases ethanol compared with homofermentative lactic acid bacteria inoculation and does not affect ammonia-N of Mombasa grass silage. In addition, chitosan and microbial inoculants have limited effects on Mombasa grass silage fermentation losses, nutritional value, and aerobic stability. Implications: Chitosan does not reduce fermentation losses or improve the nutritional value of grass silage. Background: Cowpea (Vigna unguiculata L.) an important and commonly grown kharif pulse crop in India for vegetable, grain, forage and green manuring. Considering the adverse effects of chemical fertilizers, bio-inoculants should be encouraged as natural nutrient source. Methods: Three indigenous bio-inoculants viz., Rhizobium sp., Bacillus megaterium and Glomus mosseae were evaluated in a field experiment which was conducted during kharif season 2020 to study the influence of bioinoculants on growth and yield of cowpea. The experiment was laid out in randomized block design (RCBD) with ten treatments replicated thrice. Result: The application of microbial consortia significantly influenced the number of pods bearing branches, number of nodules, fresh weight of nodules, chlorophyll content, plant nitrogen and phosphorus content and microbial activities in the cowpea rhizosphere. The present study concluded that combined application of bio-inoculants (Rhizobium, PSB, AM fungi) can save 25 per cent of recommended dose of fertilizer by sustaining the crop yield and improving the soil health. This Lilliput article provides a literature overview on ecological effects of the plant microbiome with a focus on practical application in forestry, agriculture and urban greenspace under the spectre of climate change. After an overview of the mostly bacterial microbiome of the model plant Arabidopsis thaliana, worldwide data from forests reveal ecological differentiation with respect to major guilds of predominantly fungal plant root symbionts. The plant‐microbiome association forms a new holobiont, an integrated unit for ecological adaptation and evolutionary selection. Researchers explored the impact of the microbiome on the capacity of plants to adapt to changing climate conditions. They investigated the impact of the microbiome in reforestation programs, after wildfire, drought, salination and pollution events in forestry, grasslands and agriculture. With increasing temperatures plant populations migrate to higher latitudes and higher altitudes. Ecological studies compared the dispersal capacity of plant seeds with that of soil microbes and the response of soil and root microbes to experimental heating of soils. These studies described a succession of microbiome associations and the kinetics of a release of stored soil carbon into the atmosphere enhancing global warming. Scientists explored the impact of synthetic microbial communities (SynComs) on rice productivity or tea quality; of whole soil addition in grassland restoration; or single fungal inoculation in maize fields. Meta‐analyses of fungal inoculation showed overall a positive effect, but also a wide variation in effect sizes. Climate change will be particularly prominent in urban areas ("urban heat islands") where more than half of the world population is living. Urban landscape architecture will thus have an important impact on human health and studies started to explore the contribution of the microbiome from urban greenspace to ecosystem services.
