Abstract

We study the broadband emission of Mrk 501 using multiwavelength observations from 2017 to 2020 performed with a multitude of instruments, involving, among others, MAGIC, Fermi's Large Area Telescope (LAT), NuSTAR, Swift, GASP-WEBT, and the Owens Valley Radio Observatory. Mrk 501 showed an extremely low broadband activity, which may help to unravel its baseline emission. Nonetheless, significant flux variations are detected at all wave bands, with the highest occurring at X-rays and very-high-energy (VHE) γ -rays. A significant correlation (>3 σ ) between X-rays and VHE γ -rays is measured, supporting leptonic scenarios to explain the variable parts of the emission, also during low activity. This is further supported when we extend our data from 2008 to 2020, and identify, for the first time, significant correlations between the Swift X-Ray Telescope and Fermi-LAT. We additionally find correlations between high-energy γ -rays and radio, with the radio lagging by more than 100 days, placing the γ -ray emission zone upstream of the radio-bright regions in the jet. Furthermore, Mrk 501 showed a historically low activity in X-rays and VHE γ -rays from mid-2017 to mid-2019 with a stable VHE flux (>0.2 TeV) of 5% the emission of the Crab Nebula. The broadband spectral energy distribution (SED) of this 2 yr long low state, the potential baseline emission of Mrk 501, can be characterized with one-zone leptonic models, and with (lepto)-hadronic models fulfilling neutrino flux constraints from IceCube. We explore the time evolution of the SED toward the low state, revealing that the stable baseline emission may be ascribed to a standing shock, and the variable emission to an additional expanding or traveling shock.

2. A Problem-Based Learning Activity for Enhancing Inquiry Skills and Facilitating Conceptual Change in a Biological Chemistry Course

Abstract

When teaching STEM courses, it is important to introduce state-of-the-art techniques. Students need to learn how to conduct experiments, analyse data and choose the most effective approaches to address meaningful situations. Here we present the assessment of the implementation of a structured inquiry-based activity aimed at teaching students about protein mass and size. This activity emerges as an intervention in our educational module, designed to create a cognitive conflict that effectively drives a conceptual change. To evaluate the efficacy of this module, we collected data on students' perceived and actual knowledge through pre- and post-class surveys (n = 36 and 34, respectively, mean age 26 ± 2). Additionally, we evaluated lab reports using a detailed rubric. Results indicate that the practical innovation we propose is a challenging activity that promotes the accomplishment of our learning objectives. The activity led to improvements both in confidence and in actual mastery of theoretical concepts and techniques. After completing the activity, students were able to choose the most appropriate technique to solve specific problems. Furthermore, we found that the use of a structured questionnaire in lab reports helped students to accurately analyse and process experimental data. It also allows them to demonstrate understanding of technical limitations, while integrating the knowledge and skills acquired during the module. Overall, this activity provides notions that are conceivable and profitable, thus leading to successful conceptual changes.

Abstract

A apoptose é um fator importante em muitos processos biológicos normais, tais como a embriogênese, o desenvolvimento do sistema imune, a maturação e a diferenciação celular. Em situações patológicas, a apoptose parece estar implicada na imunodeficiência, resistência a drogas e carcinogênese. Sabe-se que a carcinogênese envolve alterações genéticas cumulativas em oncogenes e genes supressores de tumor. Dessa forma, o prognóstico de cada tumor humano parece depender do equilíbrio entre os diversos genes, sendo previsível que um conhecimento mais profundo da cooperação e antagonismo entre esses genes possa fornecer num futuro próximo informações clinicamente relevantes. Nós revisamos alguns dos mais recentes progressos a respeito de genes envolvidos na apoptose e nos cânceres humanos (p53, bcl-2, c-myc, fas-APO-l, mdr-1) e suas implicações clínicas.

Abstract

Fusarium graminearum is the etiological agent of Fusarium head blight (FHB), a disease that produces a significant decrease in wheat crop yield and it is further aggravated by the presence of mycotoxins in the affected grains that may cause health problems to humans and animals. Plant defensins and defensin-like proteins are antimicrobial peptides (AMPs); they are small basic, cysteine-rich peptides (CRPs) ubiquitously expressed in the plant kingdom and mostly involved in host defence. They present a highly variable sequence but a conserved structure. The γ-core located in the C-terminal region of plant defensins has a conserved β-hairpin structure and is a well-known determinant of the antimicrobial activity among disulphide-containing AMPs. Another conserved motif of plant defensins is the α-core located in the N-terminal region, not conserved among the disulphide-containing AMPs, it has not been yet extensively studied. In this report, we have cloned the putative antimicrobial protein DefSm2, expressed in flowers of the wild plant Silybum marianum. The cDNA encodes a protein with two fused basic domains of an N-terminal defensin domain (DefSm2-D) and a C-terminal Arg-rich and Lys-rich domain. To further characterize the DefSm2-D domain, we built a 3D template-based model that will serve to support the design of novel antifungal peptides. We have designed four potential antifungal peptides: two from the DefSm2-D α-core region (SmAPα1-21 and SmAPα10-21) and two from the γ-core region (SmAPγ27-44 and SmAPγ29-35). We have chemically synthesized and purified the peptides and further characterized them by electrospray ionization mass spectrometry (ESI-MS) and Circular dichroism (CD) spectroscopy. SmAPα1-21, SmAPα10-21, and SmAPγ27-44 inhibited the growth of the phytopathogen F. graminearum at low micromolar concentrations. Conidia exposure to the fungicidal concentration of the peptides caused membrane permeabilization to the fluorescent probe propidium iodide (PI), suggesting that this is one of the main contributing factors in fungal cell killing. Furthermore, conidia treated for 0.5h showed cytoplasmic disorganization as observed by transmission electron microscopy (TEM). Remarkably, the peptides derived from the α-core induced morphological changes on the conidia cell wall, which is a promising target since its distinctive biochemical and structural organization is absent in plant and mammalian cells.

Abstract

We report a characterization of the multiband flux variability and correlations of the nearby (z=0.031) blazar Markarian 421(Mrk 421) using data from Mets ̈ahovi, Swift, Fermi-LAT, MAGIC, FACT, and other collaborations and instruments from 2014November till 2016 June. Mrk 421 did not show any prominent flaring activity, but exhibited periods of historically low activity above 1 TeV (F>1TeV<1.7×10−12ph cm−2s−1) and in the 2–10 keV (X-ray) band (F2−10 keV<3.6×10−11erg cm−2s−1),during which the Swift-BAT data suggest an additional spectral component beyond the regular synchrotron emission. The highest flux variability occurs in X-rays and very high-energy (E>0.1 TeV)γ-rays, which, despite the low activity, show a significant positive correlation with no time lag. The HRkeV and HRTeV show the harder-when-brighter trend observed in many blazars, but the trend flattens at the highest fluxes, which suggests a change in the processes dominating the blazar variability. Enlarging our dataset with data from years 2007 to 2014, we measured a positive correlation between the optical and the GeV emission over a range of about 60 d centred at time lag zero, and a positive correlation between the optical/GeV and the radio emission over a range of about 60 d centred at a time lag of 43+9−6d. This observation is consistent with the radio-bright zone being located about 0.2 parsec downstream from the optical/GeV emission regions of the jet. The flux distributions are better described with a lognormal function in most of the energy bands probed, indicating that the variability in Mrk 421 is likely produced by a multiplicative process.

Abstract

Long-duration γ-ray bursts (GRBs) originate from ultra-relativistic jets launched from the collapsing cores of dying massive stars. They are characterized by an initial phase of bright and highly variable radiation in the kiloelectronvolt-to-megaelectronvolt band, which is probably produced within the jet and lasts from milliseconds to minutes, known as the prompt emission. Subsequently, the interaction of the jet with the surrounding medium generates shock waves that are responsible for the afterglow emission, which lasts from days to months and occurs over a broad energy range from the radio to the gigaelectronvolt bands. The afterglow emission is generally well explained as synchrotron radiation emitted by electrons accelerated by the external shock. Recently, intense long-lasting emission between 0.2 and 1 teraelectronvolts was observed from GRB 190114C. Here we report multifrequency observations of GRB 190114C, and study the evolution in time of the GRB emission across 17 orders of magnitude in energy, from 5 × 10^(−6) to 10^(12) electronvolts. We find that the broadband spectral energy distribution is double-peaked, with the teraelectronvolt emission constituting a distinct spectral component with power comparable to the synchrotron component. This component is associated with the afterglow and is satisfactorily explained by inverse Compton up-scattering of synchrotron photons by high-energy electrons. We find that the conditions required to account for the observed teraelectronvolt component are typical for GRBs, supporting the possibility that inverse Compton emission is commonly produced in GRBs.

Abstract

The HAWC Collaboration released the 2HWC catalogue of TeV sources, in which 19 show no association with any known high-energy (HE; E ≳ 10 GeV) or very-high-energy (VHE; E ≳ 300 GeV) sources. This catalogue motivated follow-up studies by both the Major Atmospheric Gamma-ray Imaging Cherenkov (MAGIC) and Fermi-LAT (Large Area Telescope) observatories with the aim of investigating gamma-ray emission over a broad energy band. In this paper, we report the results from the first joint work between High Altitude Water Cherenkov (HAWC), MAGIC, and Fermi-LAT on three unassociated HAWC sources: 2HWC J2006+341, 2HWC J1907+084*, and 2HWC J1852+013*. Although no significant detection was found in the HE and VHE regimes, this investigation shows that a minimum 1° extension (at 95 per cent confidence level) and harder spectrum in the GeV than the one extrapolated from HAWC results are required in the case of 2HWC J1852+013*, whilst a simply minimum extension of 0.16° (at 95 per cent confidence level) can already explain the scenario proposed by HAWC for the remaining sources. Moreover, the hypothesis that these sources are pulsar wind nebulae is also investigated in detail.

Abstract

This contribution aims to assess the effects on the Italian legal order of the proposed directive of the European Union on minimum wages. The approach adopted in the research was that of the legal-regulatory analysis of the provisions of the proposal and their effects on the Italian statutory and collective sources. The approval of the directive would require the Italian legislator to take transposition action even if it were not decided to introduce legislation on minimum wages. The research has the limit of placing a proposal for a directive at the centre of the investigation which is not known whether and in which version it will be approved but the contribution constitutes a useful tool available to the European and domestic legislators to understand the constraints and potential of regulatory intervention on the matter. 15. Structural coalescence underlies the aggregation propensity of a β-barrel protein motif.

Abstract

A clear understanding of the structural foundations underlying protein aggregation is an elusive goal of central biomedical importance. A step toward this aim is exemplified by the β-barrel motif represented by the intestinal fatty acid binding protein (IFABP) and two abridged all-β sheet forms (Δ98Δ and Δ78Δ). At odds with the established notion that a perturbation of the native fold should necessarily favor a buildup of intermediate forms with an enhanced tendency to aggregate, the intrinsic stability (ΔG°H2O) of these proteins does not bear a straightforward correlation with their trifluoroethanol (TFE)-induced aggregation propensity. In view of this fact, we found it more insightful to delve into the connection between structure and stability under sub-aggregating conditions (10% TFE). In the absence of the co-solvent, the abridged variants display a common native-like region decorated with a disordered C-terminal stretch. Upon TFE addition, an increase in secondary structure content is observed, assimilating them to the parent protein. In this sense, TFE perturbs a common native like region while exerting a global compaction effect. Importantly, in all cases, fatty acid binding function is preserved. Interestingly, energetic as well as structural diversity in aqueous solution evolves into a common conformational ensemble more akin in stability. These facts reconcile apparent paradoxical findings related to stability and rates of aggregation. This scenario likely mimics the accrual of aggregation-prone species in the population, an early critical event for the development of fibrillation.

Abstract

Quick and reliable mass testing of infected people is an effective tool for the contingency of SARS-CoV-2. During the COVID-19 pandemic, Point-of-Care (POC) tests using Loop-Mediated Isothermal Amplification (LAMP) arose as a useful diagnostic tool. LAMP tests are a robust and fast alternative to Polymerase Chain Reaction (PCR), and their isothermal property allows easy incorporation into POC platforms. The main drawback of using colorimetric LAMP is the reported short-term stability of the pre-mixed reagents, as well as the relatively high rate of false-positive results. Also, low-magnitude amplification can produce a subtle color change, making it difficult to discern a positive reaction. This paper presents Hilab Molecular, a portable device that uses the Internet of Things and Artificial Intelligence to pre-analyze colorimetric data. In addition, we established manufacturing procedures to increase the stability of colorimetric RT-LAMP tests. We show that ready-to-use reactions can be stored for up to 120 days at −20 °C. Furthermore, we validated both the Hilab Molecular device and the Hilab RT-LAMP test for SARS-CoV-2 using 581 patient samples without any purification steps. We achieved a sensitivity of 92.93% and specificity of 99.42% (samples with CT ≤ 30) when compared to RT-qPCR.

18. Amyloidogenic propensity of a natural variant of human apolipoprotein A-I: stability and interaction with ligands.

Abstract

A number of naturally occurring mutations of human apolipoprotein A-I (apoA-I) have been associated with hereditary amyloidoses. The molecular mechanisms involved in amyloid-associated pathology remain largely unknown. Here we examined the effects of the Arg173Pro point mutation in apoA-I on the structure, stability, and aggregation propensity, as well as on the ability to bind to putative ligands. Our results indicate that the mutation induces a drastic loss of stability, and a lower efficiency to bind to phospholipid vesicles at physiological pH, which could determine the observed higher tendency to aggregate as pro-amyloidogenic complexes. Incubation under acidic conditions does not seem to induce significant desestabilization or aggregation tendency, neither does it contribute to the binding of the mutant to sodium dodecyl sulfate. While the binding to this detergent is higher for the mutant as compared to wt apoA-I, the interaction of the Arg173Pro variant with heparin depends on pH, being lower at pH 5.0 and higher than wt under physiological pH conditions. We suggest that binding to ligands as heparin or other glycosaminoglycans could be key events tuning the fine details of the interaction of apoA-I variants with the micro-environment, and probably eliciting the toxicity of these variants in hereditary amyloidoses.

Abstract

Chagas disease is caused by the protozoan parasite Trypanosoma cruzi and affects 7 million people worldwide. Considering the side effects and drug resistance shown by current treatments, the development of new anti-Chagas therapies is an urgent need. T. cruzi hypoxanthine phosphoribosyltransferase (TcHPRT), the key enzyme of the purine salvage pathway, is essential for the survival of trypanosomatids. Previously, we assessed the inhibitory effect of different bisphosphonates (BPs), HPRT substrate analogues, on the activity of the isolated enzyme. BPs are used as a treatment for bone diseases and growth inhibition studies on T. cruzi have associated BPs action with the farnesyl diphosphate synthase inhibition. Here, we demonstrated significant growth inhibition of epimastigotes in the presence of BPs and a strong correlation with our previous results on the isolated TcHPRT, suggesting this enzyme as a possible and important target for these drugs. We also found that the parasites exhibited a delay at S phase in the presence of zoledronate pointing out enzymes involved in the cell cycle, such as TcHPRT, as intracellular targets. Moreover, we validated that micromolar concentrations of zoledronate are capable to interfere with the progression of cell infection by this parasite. Altogether, our findings allow us to propose the repositioning of zoledronate as a promising candidate against Chagas disease and TcHPRT as a new target for future rational design of antiparasitic drugs. Abstract

Obiettivi: Il contributo si propone di valutare gli effetti sull’ordinamento italiano della proposta di direttiva dell’Unione europea in tema di salario minimo. Metodologia: L'approccio adottato nella ricerca è stato quello dell’analisi giuridico-normativa delle previsioni della proposta e dei loro effetti sulle fonti legali e collettive interne. Risultati: L’approvazione della direttiva imporrebbe un intervento attuativo del legislatore italiano anche nel caso in cui non si decidesse di introdurre una legislazione sui minimi salariali. Limiti e implicazioni: La ricerca presenta il limite di mettere al centro dell’indagine una proposta di direttiva che non è dato sapere se e in quale versione sarà approvata ma costituisce un utile strumento a disposizione del legislatore europeo e di quello interno per comprendere i vincoli e le potenzialità di un intervento normativo in materia. Originalità: I tratti originali della ricerca sono rappresentati dalla delimitazione dei confini della competenza dell’Unione in materia retributiva e dall’interazione fra la dimensione europea e quella italiana.

Abstract

Astronomy and astrophysics 647, A163 (2021). doi:10.1051/0004-6361/202039687, Context. QSO B1420+326 is a blazar classified as a flat-spectrum radio quasar (FSRQ). At the beginning of the year 2020, it was found to be in an enhanced flux state and an extensive multiwavelength campaign allowed us to trace the evolution of the flare.Aims. We search for very high-energy (VHE) gamma-ray emission from QSO B1420+326 during this flaring state. We aim to characterize and model the broadband emission of the source over different phases of the flare.Methods. The source was observed with a number of instruments in radio, near-infrared, optical (including polarimetry and spectroscopy), ultraviolet, X-ray, and gamma-ray bands. We use dedicated optical spectroscopy results to estimate the accretion disk and the dust torus luminosity. We performed spectral energy distribution modeling in the framework of combined synchrotron-self-Compton and external Compton scenario in which the electron energy distribution is partially determined from acceleration and cooling processes.Results. During the enhanced state, the flux of both SED components of QSO B1420+326 drastically increased and the peaks were shifted to higher energies. Follow-up observations with the MAGIC telescopes led to the detection of VHE gamma-ray emission from this source, making it one of only a handful of FSRQs known in this energy range. Modeling allows us to constrain the evolution of the magnetic field and electron energy distribution in the emission region. The gamma-ray flare was accompanied by a rotation of the optical polarization vector during a low -polarization state. Also, a new superluminal radio knot contemporaneously appeared in the radio image of the jet. Abstract

MAGIC Collaboration: et al., [Context]: It has become evident that one-zone synchrotron self-Compton models are not always adequate for very high-energy (VHE) gamma-ray-emitting blazars. While two-component models perform better, they are difficult to constrain due to the large number of free parameters., [Aims]: In this work, we make a first attempt at taking into account the observational constraints from very long baseline interferometry (VLBI) data, long-term light curves (radio, optical, and X-rays), and optical polarisation to limit the parameter space for a two-component model and test whether or not it can still reproduce the observed spectral energy distribution (SED) of the blazars., [Methods]: We selected five TeV BL Lac objects based on the availability of VHE gamma-ray and optical polarisation data. We collected constraints for the jet parameters from VLBI observations. We evaluated the contributions of the two components to the optical flux by means of decomposition of long-term radio and optical light curves as well as modelling of the optical polarisation variability of the objects. We selected eight epochs for these five objects based on the variability observed at VHE gamma rays, for which we constructed the SEDs that we then modelled with a two-component model., [Results]: We found parameter sets which can reproduce the broadband SED of the sources in the framework of two-component models considering all available observational constraints from VLBI observations. Moreover, the constraints obtained from the long-term behaviour of the sources in the lower energy bands could be used to determine the region where the emission in each band originates. Finally, we attempt to use optical polarisation data to shed new light on the behaviour of the two components in the optical band. Our observationally constrained two-component model allows explanation of the entire SED from radio to VHE with two co-located emission regions., VFR and EL were supported by Academy of Finland projects 317636 and 320045. Abstract

Aims. We measure the Crab Nebula γ-ray spectral energy distribution in the ~100 TeV energy domain and test the validity of existing leptonic emission models at these high energies. Methods. We used the novel very large zenith angle observations with the MAGIC telescope system to increase the collection area above 10 TeV. We also developed an auxiliary procedure of monitoring atmospheric transmission in order to assure proper calibration of the accumulated data. This employs recording optical images of the stellar field next to the source position, which provides a better than 10% accuracy for the transmission measurements. Results. We demonstrate that MAGIC very large zenith angle observations yield a collection area larger than a square kilometer. In only ~ 56 h of observations, we detect the γ-ray emission from the Crab Nebula up to 100 TeV, thus providing the highest energy measurement of this source to date with Imaging Atmospheric Cherenkov Telescopes. Abstract

Metabolic control analysis (MCA) is a promising approach in biochemistry aimed at understanding processes in a quantitative fashion. Here the contribution of enzymes and transporters to the control of a given pathway flux and metabolite concentrations is determined and expressed quantitatively by means of numerical coefficients. Metabolic flux can be influenced by a wide variety of modulators acting on one or more metabolic steps along the pathway. We describe a laboratory exercise to study metabolic regulation of human erythrocytes (RBCs). Within the framework of MCA, students use these cells to determine the sensitivity of the glycolytic flux to two inhibitors (iodoacetic acid: IA, and iodoacetamide: IAA) known to act on the enzyme glyceraldehyde-3-phosphate-dehydrogenase. Glycolytic flux was estimated by determining the concentration of extracellular lactate, the end product of RBC glycolysis. A low-cost colorimetric assay was implemented, that takes advantage of the straightforward quantification of the absorbance signal from the photographic image of the multi-well plate taken with a standard digital camera. Students estimate flux response coefficients for each inhibitor by fitting an empirical function to the experimental data, followed by analytical derivation of this function. IA and IAA exhibit qualitatively different patterns, which are thoroughly analyzed in terms of the physicochemical properties influencing their action on the target enzyme. IA causes highest glycolytic flux inhibition at lower concentration than IAA. This work illustrates the feasibility of using the MCA approach to study key variables of a simple metabolic system, in the context of an upper level biochemistry course. © 2018 International Union of Biochemistry and Molecular Biology, 46(5):502-515, 2018.

Abstract

Natural killer (NK) cells are lymphocytes of the innate immune system that eliminate virally infected or malignantly transformed cells. NK cell function is regulated by diverse surface receptors that are both activating and inhibitory. Among them, the homodimeric Ly49 receptors control NK cell cytotoxicity by sensing major histocompatibility complex class I molecules (MHC-I) on target cells. Although crystal structures have been reported for Ly49/MHC-I complexes, the underlying binding mechanism has not been elucidated. Accordingly, we carried out thermodynamic and kinetic experiments on the interaction of four NK Ly49 receptors (Ly49G, Ly49H, Ly49I and Ly49P) with two MHC-I ligands (H-2Dd and H-2Dk). These Ly49s embrace the structural and functional diversity of the highly polymorphic Ly49 family. Combining surface plasmon resonance, fluorescence anisotropy and far-UV circular dichroism (CD), we determined that the best model to describe both inhibitory and activating Ly49/MHC-I interactions is one in which the two MHC-I binding sites of the Ly49 homodimer present similar binding constants for the two sites (∼106 M–1) with a slightly positive co-operativity in some cases, and without far-UV CD observable conformational changes. Furthermore, Ly49/MHC-I interactions are diffusion-controlled and enthalpy-driven. These features stand in marked contrast with the activation-controlled and entropy-driven interaction of Ly49s with the viral immunoevasin m157, which is characterized by strong positive co-operativity and conformational selection. These differences are explained by the distinct structures of Ly49/MHC-I and Ly49/m157 complexes. Abstract

Local events that affect specific regions of proteins are of utmost relevance for stability and function. The aim of this study is to quantitatively assess the importance of locally-focused dynamics by means of a simple chemical modification procedure. Taking human Frataxin as a working model, we investigated local fluctuations of the C-terminal region (the last 16 residues of the protein) by means of three L → C replacement mutants: L98C, L200C and L203C. The conformation and thermodynamic stability of each variant was assessed. All the variants exhibited native features and high stabilities: 9.1 (wild type), 8.1 (L198C), 7.0 (L200C) and 10.0 kcal mol−1 (L203C). In addition, kinetic rates of Cys chemical modification by DTNB and DTDPy were measured, conformational dynamics data were extracted and free energy for the local unfolding of the C-terminal region was estimated. The analysis of these results indicates that the conformation of the C-terminal region fluctuates with partial independence from global unfolding events. Additionally, numerical fittings of the kinetic model of the process suggest that the local transition occurs in the seconds to minutes timescale. In fact, standard free energy differences for local unfolding were found to be significantly lower than those of the global unfolding reaction, showing that chemical modification results may not be explained in terms of the global unfolding reaction alone. These results provide unequivocal experimental evidence of local phenomena with global effects and contribute to understanding how global and local stability are linked to protein dynamics.

Abstract

Human pathogenic gram-negative bacteria, such as enteropathogenic Escherichia coli (EPEC), rely on type III secretion systems (T3SS) to translocate virulence factors directly into host cells. The coiled-coil domains present in the structural proteins of T3SS are conformed by amphipathic alpha-helical structures that play an important role in the protein-protein interaction and are essential for the assembly of the translocation complex. To investigate the inhibitory capacity of these domains on the T3SS of EPEC, we synthesized peptides between 7 and 34 amino acids based on the coiled-coil domains of proteins that make up this secretion system. This analysis was performed through in vitro hemolysis assays by assessing the reduction of T3SS-dependent red blood cell lysis in the presence of the synthesized peptides. After confirming its inhibitory capacity, we performed molecular modeling assays using combined techniques, docking-molecular dynamic simulations, and quantum-mechanic calculations of the various peptide-protein complexes, to improve the affinity of the peptides to the target proteins selected from T3SS. These techniques allowed us to demonstrate that the peptides with greater inhibitory activity, directed against the coiled-coil domain of the C-terminal region of EspA, present favorable hydrophobic and hydrogen bond molecular interactions. Particularly, the hydrogen bond component is responsible for the stabilization of the peptide-protein complex. This study demonstrates that compounds targeting T3SS from pathogenic bacteria can indeed inhibit bacterial infection by presenting a higher specificity than broad-spectrum antibiotics. In turn, these peptides could be taken as initial structures to design and synthesize new compounds that mimic their inhibitory pharmacophoric pattern. Fil: Larzabal, Mariano. Abstract

The PAU (Physics of the Accelerating Universe) Survey goal is to obtain photometric redshifts (photo-z) and Spectral Energy Distribution (SED) of astronomical objects with a resolution roughly one order of magnitude better than current broad band photometric surveys. To accomplish this, a new large field of view camera (PAUCam) has been designed, built, commissioned and is now operated at the William Herschel Telescope (WHT). With the current WHT Prime Focus corrector, the camera covers ~1-degree diameter Field of View (FoV), of which, only the inner ~40 arcmin diameter are unvignetted. The focal plane consists of a mosaic of 18 2k$x4k Hamamatsu fully depleted CCDs, with high quantum efficiency up to 1 micrometers in wavelength. To maximize the detector coverage within the FoV, filters are placed in front of the CCDs inside the camera cryostat (made out of carbon fiber) using a challenging movable tray system. The camera uses a set of 40 narrow band filters ranging from ~4500 to ~8500 Angstroms complemented with six standard broad-band filters, ugrizY. The PAU Survey aims to cover roughly 100 square degrees over fields with existing deep photometry and galaxy shapes to obtain accurate photometric redshifts for galaxies down to i_AB~22.5, detecting also galaxies down to i_AB~24 with less precision in redshift. With this data set we will be able to measure intrinsic alignments, galaxy clustering and perform galaxy evolution studies in a new range of densities and redshifts. Abstract

A population of globular clusters (GCs) has been recently established by the Fermi-LAT telescope as a new class of GeV $\gamma$-ray sources. Leptons accelerated to TeV energies, in the inner magnetospheres of MSPs or in their wind regions, should produce $\gamma$-rays through the inverse Compton scattering in the dense radiation field from the huge population of stars. We have conducted deep observations of the globular cluster M15 with the MAGIC telescopes and used 165 hrs in order to search for $\gamma$-ray emission. Abstract

SNR G24.7+0.6 is a 9.5 kyrs radio and $\gamma$-ray supernova remnant evolving in a dense medium. In the GeV regime, SNR G24.7+0.6 (3FHL\,J1834.1--0706e/FGES\,J1834.1--0706) shows a hard spectral index ($\Gamma$$\sim$2) up to $200$\,GeV, which makes it a good candidate to be observed with Cherenkov telescopes such as MAGIC. We observed the field of view of \snr\ with the MAGIC telescopes for a total of 31 hours. We detect very high energy $\gamma$-ray emission from an extended source located 0.34\degr\ away from the center of the radio SNR. The new source, named \mgc\ is detected up to 5\,TeV, and its spectrum is well-represented by a power-law function with spectral index of $2.74 \pm 0.08$. The complexity of the region makes the identification of the origin of the very-high energy emission difficult, however the spectral agreement with the LAT source and overlapping position at less than 1.5$\sigma$ point to a common origin. We analysed 8 years of \fermi-LAT data to extend the spectrum of the source down to 60\,MeV. \fermi-LAT and MAGIC spectra overlap within errors and the global broad band spectrum is described by a power-law with exponential cutoff at $1.9\pm0.5$\,TeV. The detected $\gamma$-ray emission can be interpreted as the results of proton-proton interaction between the supernova and the CO-rich surrounding., Comment: accepted for publication by MNRAS

Abstract

Hypoxanthine phosphoribosyl transferase from Trypanosoma cruzi (TcHPRT) is a critical enzyme for the survival of the parasite. This work demonstrates that the full-length form in solution adopts a stable and enzymatically active tetrameric form, exhibiting large inter-subunit surfaces. Although this protein irreversibly aggregates during unfolding, oligomerization is reversible and can be modulated by low concentrations of urea. When the C-terminal region, which is predicted as a disordered stretch, is excised by proteolysis, TcHPRT adopts a dimeric state, suggesting that the C-terminal region acts as a main guide for the quaternary arrangement. These results are in agreement with X-ray crystallographic data presented in this work. On the other hand, the C-terminal region exhibits a modulatory role on the enzyme, as attested by the enhanced activity observed for the dimeric form. Bisphosphonates act as substrate-mimetics, uncovering long-range communications among the active sites. All in all, this work contributes to establish new ways applicable to the design of novel inhibitors that could eventually result in new drugs against parasitic diseases.

Abstract

Metabolic control analysis (MCA) is a promising approach in biochemistry aimed at understanding processes in a quantitative fashion. Here the contribution of enzymes and transporters to the control of a given pathway flux and metabolite concentrations is determined and expressed quantitatively by means of numerical coefficients. Metabolic flux can be influenced by a wide variety of modulators acting on one or more metabolic steps along the pathway. We describe a laboratory exercise to study metabolic regulation of human erythrocytes (RBCs). Within the framework of MCA, students use these cells to determine the sensitivity of the glycolytic flux to two inhibitors (iodoacetic acid: IA, and iodoacetamide: IAA) known to act on the enzyme glyceraldehyde-3-phosphate-dehydrogenase. Glycolytic flux was estimated by determining the concentration of extracellular lactate, the end product of RBC glycolysis. A low-cost colorimetric assay was implemented, that takes advantage of the straightforward quantification of the absorbance signal from the photographic image of the multi-well plate taken with a standard digital camera. Students estimate flux response coefficients for each inhibitor by fitting an empirical function to the experimental data, followed by analytical derivation of this function. IA and IAA exhibit qualitatively different patterns, which are thoroughly analyzed in terms of the physicochemical properties influencing their action on the target enzyme. IA causes highest glycolytic flux inhibition at lower concentration than IAA. This work illustrates the feasibility of using the MCA approach to study key variables of a simple metabolic system, in the context of an upper level biochemistry course. © 2018 International Union of Biochemistry and Molecular Biology, 46(5):502-515, 2018.

Abstract

A neutrino with energy of $\sim$290 TeV, IceCube-170922A, was detected in coincidence with the BL Lac object TXS~0506+056 during enhanced gamma-ray activity, with chance coincidence being rejected at $\sim 3\sigma$ level. We monitored the object in the very-high-energy (VHE) band with the MAGIC telescopes for $\sim$41 hours from 1.3 to 40.4 days after the neutrino detection. Day-timescale variability is clearly resolved. We interpret the quasi-simultaneous neutrino and broadband electromagnetic observations with a novel one-zone lepto-hadronic model, based on interactions of electrons and protons co-accelerated in the jet with external photons originating from a slow-moving plasma sheath surrounding the faster jet spine. We can reproduce the multiwavelength spectra of TXS 0506+056 with neutrino rate and energy compatible with IceCube-170922A, and with plausible values for the jet power of $\sim 10^{45} - 4 \times 10^{46} {\rm erg \ s^{-1}}$. The steep spectrum observed by MAGIC is concordant with internal $\gamma\gamma$ absorption above a few tens of GeV entailed by photohadronic production of a $\sim$290 TeV neutrino, corroborating a genuine connection between the multi-messenger signals. In contrast to previous predictions of predominantly hadronic emission from neutrino sources, the gamma-rays can be mostly ascribed to inverse Compton up-scattering of external photons by accelerated electrons. The X-ray and VHE bands provide crucial constraints on the emission from both accelerated electrons and protons. We infer that the maximum energy of protons in the jet co-moving frame can be in the range $\sim 10^{14}$ to $10^{18}$ eV., Comment: 11 pages, 3 figues, 3 tables, Accepted for publication in ApJL. Abstract

The Physics of the Accelerating Universe (PAU) Survey is an international project for the study of cosmological parameters associated with Dark Energy. The PAU 18-CCD camera (PAUCam) is installed at the primary focus of the William Herschel Telescope at Observatorio del Roque de los Muchachos (La Palma, Canary Islands). The camera incorporates a unique set of 40 narrow band filters in the optical range from 450 nm to 850 nm and a set of 6 standard broad band filters. It is used to scan part of the northern sky in order to collect low resolution spectral information from millions of galaxies. The PAU data management (PAUdm) team is in charge of treating the data, including data transfer from the observatory to the PAU Survey data center, hosted at Port d’Informacio Cientifica (PIC). PAUdm is also in charge of the storage, data reduction and, finally, of making the results available to the scientific community. We describe the technical solutions adopted to cover different aspects of the PAU Survey data management, from the computing infrastructure to the software tools and web services for the processing orchestration and data exploration. We give special attention to the PAU database, developed to preserve and guarantee the consistency of data and metadata and also used to coordinate the different PAUdm tasks.

Abstract

The MAGIC telescopes observed S2 0109+22 in 2015 July during its flaring activity in high energy gamma rays observed by Fermi-LAT. We analyse the MAGIC data to characterise the very high energy (VHE) gamma-ray emission of S2 0109+22, which belongs to the subclass of intermediate synchrotron peak (ISP) BL Lac objects. We study the multi-frequency emission in order to investigate the source classification. Finally, we compare the source long-term behaviour to other VHE gamma-ray emitting (TeV) blazars. We performed a temporal and spectral analysis of the data centred around the MAGIC interval of observation (MJD 57225-57231). Long-term radio and optical data have also been investigated using the discrete correlation function. The redshift of the source is estimated through optical host-galaxy imaging and also using the amount of VHE gamma-ray absorption. The quasi-simultaneous multi-frequency spectral energy distribution (SED) is modelled with the conventional one-zone synchrotron self-Compton (SSC) model. MAGIC observations resulted in the detection of the source at a significance level of $5.3\,\sigma$. The VHE gamma-ray emission of S2 0109+22 is variable on a daily time scale. VHE gamma-ray luminosity of the source is lower than the average of TeV BL Lacs. The optical polarization, and long-term optical/radio behaviour of the source are different from the general population of TeV blazars. All these findings agree with the classification of the source as an ISP BL Lac object. We estimate the source redshift as $z = 0.36 \pm 0.07$. Abstract

Fast radio bursts (FRBs) are bright flashes observed typically at GHz frequencies with millisecond duration, whose origin is likely extragalactic. Their nature remains mysterious, motivating searches for counterparts at other wavelengths. FRB 121102 is so far the only source known to repeatedly emit FRBs and is associated with a host galaxy at redshift $z \simeq 0.193$. We conducted simultaneous observations of FRB 121102 with the Arecibo and MAGIC telescopes during several epochs in 2016--2017. This allowed searches for millisecond-timescale burst emission in very-high-energy (VHE) gamma rays as well as the optical band. While a total of five FRBs were detected during these observations, no VHE emission was detected, neither of a persistent nature nor burst-like associated with the FRBs. The average integral flux upper limits above 100 GeV at 95% confidence level are $6.6 \times 10^{-12}~\mathrm{photons\ cm^{-2}\ s^{-1}}$ (corresponding to luminosity $L_{\rm VHE} \lesssim 10^{45}~\mathrm{erg\ s^{-1}}$) over the entire observation period, and $1.2 \times 10^{-7}~ \mathrm{photons\ cm^{-2}\ s^{-1}}$ ($L_{\rm VHE} \lesssim 10^{49}~\mathrm{erg\ s^{-1}}$) over the total duration of the five FRBs. We constrain the optical U-band flux to be below 8.6 mJy at 5-$\sigma$ level for 1-ms intervals around the FRB arrival times. A bright burst with U-band flux $29~\mathrm{mJy}$ and duration $\sim 12$ ms was detected 4.3 s before the arrival of one FRB. However, the probability of spuriously detecting such a signal within the sampled time space is 1.5% (2.2 $\sigma$, post-trial), i.e. consistent with the expected background.
