1. Applying IUCN Red List criteria to birds at different geographical scales: similarities and differences

Abstract: Aplicación de los criterios de la Lista Roja de la Unión Internacional para la Conservación de la Naturaleza en diferentes escalas geográficas a las aves. La Unión Internacional para la Conservación de la Naturaleza (UICN) se encarga de evaluar a escala mundial el riesgo de extinción y el estado de conservación de las especies. Para garantizar su objetividad, repetibilidad y trazabilidad, en las evaluaciones se sigue un proceso estandarizado que hace uso de información fiable y verificable. Asimismo, las evaluaciones se sintetizan de acuerdo con determinadas directrices, que se han ajustado recientemente para su aplicación a escalas inferiores. No obstante, la misma especie puede clasificarse en varios estados de conservación, distintos o superpuestos. Para comparar cuantitativamente las evaluaciones de escala mundial a escala subnacional, analizamos 15 listas de evaluación relativas a 66 especies de aves cinegéticas en Francia; según se había declarado, dichas evaluaciones se realizaron en consonancia con las directrices de la UICN. En general, nuestros resultados ponen de manifiesto que (1) las evaluaciones a gran escala espacial tienden a dar como resultado estados de peligro inferiores que las de pequeña escala; (2) las evaluaciones a gran escala permitieron comprobar de forma oficial la información en que se basan, mientras que las evaluaciones a menor escala no; (3) las evaluaciones a gran escala son más propensas a basarse en pruebas estandarizadas de una reducción significativa del tamaño de la población y a estar menos expuestas a efectos de ‘escala’ o de ‘borde de distribución’; (4) las evaluaciones a gran escala también se basan más frecuentemente en publicaciones científicas en sentido estricto; y (5) las fuentes se sintetizan con mayor exactitud en comparación con las listas rojas a escalas espaciales pequeñas. Por lo tanto, nuestros resultados sugieren que las listas rojas a pequeña escala no coinciden plenamente con las directrices de la UICN y que difieren de forma significativa con respecto a sus procesos de evaluación en comparación con los estándares mundiales. El uso de información subjetiva y poco fiable en las listas rojas en pequeña escala (sobre todo en las listas nacionales y subnacionales) puede poner en peligro el objetivo original de las listas rojas de la UICN de proporcionar información completa y científicamente rigurosa y, por lo tanto, podría comprometer la credibilidad y el prestigio de las listas rojas de la UICN a los ojos de los investigadores, del público en general y de otras partes interesadas.

Abstract: International audience; The Polarized Instrument for Long-wavelength Observation of the Tenuous interstellar medium (PILOT) is a balloon-borne experiment that aims to measure the polarized emission of thermal dust at a wavelength of 240 µm (1.2 THz). A first PILOT flight of the experiment took place from Timmins, Ontario, Canada, in September 2015 and a second flight took place from Alice Springs, Australia in April 2017. In this paper, we present the inflight performance of the instrument. Here we concentrate on the instrument performance as measured during the second flight, but refer to the performance observed during the first flight, if it was significantly different. We present a short description of the instrument and the flights. We measure the time constants of the detectors using the decay of the observed signal during flight following high energy particle impacts (glitches) and switching off the instrument's internal calibration source. We use these time constants to deconvolve the timelines and analyze the optical quality of the instrument as measured on planets. We then analyze the structure and polarization of the instrumental background. We measure the detector response flat field and its time variations using the signal from the residual atmosphere and from the internal calibration source. Finally, we analyze the spectral and temporal properties of the detector noise. The inflight performance is found to be satisfactory and globally in line with expectations from ground calibrations. We conclude by assessing the expected inflight sensitivity of the instrument in light of the measured inflight performance.

Abstract: International audience; PILOT (Polarized Instrument for the Long-wavelength Observations of the Tenuous ISM), is a balloon-borne astronomy experiment dedicated to study the polarization of dust emission from the diffuse ISM in our Galaxy [1]. The observations of PILOT have two major scientific objectives. Firstly, they will allow us to constrain the large-scale geometry of the magnetic field in our Galaxy and to study in details the alignment properties of dust grains with respect to the magnetic field. In this domain, the measurements of PILOT will complement those of the Planck satellite at longer wavelengths. In particular, they will bring information at a better angular resolution, which is critical in crowded regions such as the Galactic plane. They will allow us to better understand how the magnetic field is shaping the ISM material on large scale in molecular clouds, and the role it plays in the gravitational collapse leading to star formation. Secondly, the PILOT observations will allow us to measure for the first time the polarized dust emission towards the most diffuse regions of the sky, where the measurements are the most easily interpreted in terms of the physics of dust. In this particular domain, PILOT will play a role for future CMB missions similar to that played by the Archeops experiment for Planck. The results of PILOT will allow us to gain knowledge about the magnetic properties of dust grains and about the structure of the magnetic field in the diffuse ISM that is necessary to a precise foreground subtraction in future polarized CMB measurements. The PILOT measurements, combined with those of Planck at longer wavelengths, will therefore allow us to further constrain the dust models. The outcome of such studies will likely impact the instrumental and technical choices for the future space missions dedicated to CMB polarization. The PILOT instrument will allow observations in two photometric channels at wavelengths 240 μm and 550 μm, with an angular resolution of a few arcminutes. We will make use of large format bolometer arrays, developed for the PACS instrument on board the Herschel satellite. With 1024 detectors per photometric channel and photometric band optimized for the measurement of dust emission, PILOT is likely to become the most sensitive experiment for this type of measurements. The PILOT experiment will take advantage of the large gain in sensitivity allowed by the use of large format, filled bolometer arrays at frequencies more favorable to the detection of dust emission. This paper presents the optical design, optical characterization and its performance. We begin with a presentation of the instrument and the optical system and then we summarise the main optical tests performed. In section III, we present preliminary end-to-end test results.

Abstract: International audience; The Polarized Instrument for Long‐wavelength Observation of the Tenuous interstellar medium (PILOT) is a balloon‐borne astronomy experiment designed to study the linear polarization of thermal dust emission in two photometric bands centred at wavelengths 240 mu m (1.2 THz) and 550 mu m (545 GHz), with an angular resolution of a few arcminutes. Several end‐to‐end tests of the instrument were performed on the ground between 2012 and 2014, in order to prepare for the first scientific flight of the experiment that took place in September 2015 from Timmins, Ontario, Canada. This paper presents the results of those tests, focussing on an evaluation of the instrument's optical performance. We quantify image quality across the extent of the focal plane, and describe the tests that we conducted to determine the focal plane geometry, the optimal focus position, and sources of internal straylight. We present estimates of the detector response, obtained using an internal calibration source, and estimates of the background intensity and background polarization.

Abstract: PILOT is a stratospheric experiment designed to measure the polarization of dust FIR emission, towards the diffuse interstellar medium. The first PILOT flight was carried out from Timmins in Ontario-Canada on September 20th 2015. The flight has been part of a launch campaign operated by the CNES, which has allowed to launch 4 experiments, including PILOT. The purpose of this paper is to describe the performance of the instrument in flight and to perform a first comparison with those achieved during ground tests. The analysis of the flight data is on-going, in particular the identification of instrumental systematic effects, the minimization of their impact and the quantification of their remaining effect on the polarization data. At the end of this paper, we shortly illustrate the quality of the scientific observations obtained during this first flight, at the current stage of systematic effect removal.

Abstract: Future cosmology space missions will concentrate on measuring the polarization of the Cosmic Microwave Background, which potentially carries invaluable information about the earliest phases of the evolution of our universe. Such ambitious projects will ultimately be limited by the sensitivity of the instrument and by the accuracy at which polarized foreground emission from our own Galaxy can be subtracted out. We present the PILOT balloon project, which aims at characterizing one of these foreground sources, the polarized continuum emission by dust in the diffuse interstellar medium. The PILOT experiment also constitutes a test-bed for using multiplexed bolometer arrays for polarization measurements. This paper presents the instrument and its expected performances. Performance measured during ground calibrations of the instrument and in flight will be described in a forthcoming paper.

Abstract: Euclid-VIS is the large format visible imager for the ESA Euclid space mission in their Cosmic Vision program, scheduled for launch in 2020. Together with the near infrared imaging within the NISP instrument, it forms the basis of the weak lensing measurements of Euclid. VIS will image in a single r+i+z band from 550-900 nm over a field of view of ~0.5 deg2. By combining 4 exposures with a total of 2260 sec, VIS will reach to V=24.5 (10σ) for sources with extent ~0.3 arcsec. The image sampling is 0.1 arcsec. VIS will provide deep imaging with a tightly controlled and stable point spread function (PSF) over a wide survey area of 15000 deg2 to measure the cosmic shear from nearly 1.5 billion galaxies to high levels of accuracy, from which the cosmological parameters will be measured. In addition, VIS will also provide a legacy dataset with an unprecedented combination of spatial resolution, depth and area covering most of the extra-Galactic sky. Abstract: Future cosmology space missions will concentrate on measuring the polarization of the Cosmic Microwave Background, which potentially carries invaluable information about the earliest phases of the evolution of our universe. Such ambitious projects will ultimately be limited by the sensitivity of the instrument and by the accuracy at which polarized foreground emission from our own Galaxy can be subtracted out. We present the PILOT balloon project which will aim at characterizing one of these foreground sources, the polarization of the dust continuum emission in the diffuse interstellar medium. The PILOT experiment will also constitute a test-bed for using multiplexed bolometer arrays for polarization measurements. Abstract: The Planck High Frequency Instrument (HFI) surveyed the sky continuously from August 2009 to January 2012. Its noise and sensitivity performance were excellent, but the rate of cosmic ray impacts on the HFI detectors was unexpectedly high. Furthermore, collisions of cosmic rays with the focal plane produced transient signals in the data (glitches) with a wide range of characteristics. A study of cosmic ray impacts on the HFI detector modules has been undertaken to categorize and characterize the glitches, to correct the HFI time-ordered data, and understand the residual effects on Planck maps and data products. This paper presents an evaluation of the physical origins of glitches observed by the HFI detectors. In order to better understand the glitches observed by HFI in flight, several ground-based experiments were conducted with flight-spare HFI bolometer modules. The experiments were conducted between 2010 and 2013 with HFI test bolometers in different configurations using varying particles and impact energies. The bolometer modules were exposed to 23 MeV protons from the Orsay IPN TANDEM accelerator, and to $^{241}$Am and $^{244}$Cm $\alpha$-particle and $^{55}$Fe radioactive X-ray sources. The calibration data from the HFI ground-based preflight tests were used to further characterize the glitches and compare glitch rates with statistical expectations under laboratory conditions. Test results provide strong evidence that the dominant family of glitches observed in flight are due to cosmic ray absorption by the silicon die substrate on which the HFI detectors reside. Glitch energy is propagated to the thermistor by ballistic phonons, while there is also a thermal diffusion contribution. The implications of these results for future satellite missions, especially those in the far-infrared to sub-millimetre and millimetre regions of the electromagnetic spectrum, are discussed., Comment: 11 pages, 13 figures

Abstract: The Planck High Frequency Instrument (HFI) has been surveying the sky continuously from the second Lagrangian point (L2) between August 2009 and January 2012. It operates with 52 high impedance bolometers cooled at 100mK in a range of frequency between 100 GHz and 1THz with unprecedented sensivity, but strong coupling with cosmic radiation. At L2, the particle flux is about 5 $cm^{-2} s^{-1}$ and is dominated by protons incident on the spacecraft. Protons with an energy above 40MeV can penetrate the focal plane unit box causing two different effects: glitches in the raw data from direct interaction of cosmic rays with detectors (producing a data loss of about 15% at the end of the mission) and thermal drifts in the bolometer plate at 100mK adding non-gaussian noise at frequencies below 0.1Hz. The HFI consortium has made strong efforts in order to correct for this effect on the time ordered data and final Planck maps. This work intends to give a view of the physical explanation of the glitches observed in the HFI instrument in-flight. To reach this goal, we performed several ground-based experiments using protons and $\alpha$ particles to test the impact of particles on the HFI spare bolometers with a better control of the environmental conditions with respect to the in-flight data. We have shown that the dominant part of glitches observed in the data comes from the impact of cosmic rays in the silicon die frame supporting the micro-machinced bolometric detectors propagating energy mainly by ballistic phonons and by thermal diffusion. 12. Mésusage du sulfate de morphine en Languedoc-Roussillon – comportement déviant : intérêt d’une double approche

Abstract: Introduction Le sulfate de morphine (Skenan®) est indique dans les douleurs severes, rebelles aux autres analgesiques. Depuis plusieurs annees, le mesusage et l’abus du sulfate de morphine font l’objet d’une surveillance nationale par le reseau des centres d’addictovigilance. La region Languedoc-Roussillon (LR) est particulierement impactee par ces mesusages et abus mais peu de donnees sont disponibles. Une etude combinant deux approches epidemiologiques complementaires a permis de faire un premier bilan de ce phenomene de mesusage, en region Languedoc-Roussillon (LR) et sur le territoire francais. Materiel et methodes Premiere approche : une extraction des donnees de la CPAM de l’annee 2011 a permis d’etudier le comportement des usagers de Skenan® en LR. Les sujets ayant recu une premiere delivrance entre le 01/01/2011 et le 28/02/2011 ont ete suivis pendant neuf mois. Une classification non supervisee a partir des donnees de consommation a permis d’identifier plusieurs profils de consommateurs. Les clusters obtenus ont ete caracterises et compares a partir des indicateurs de Docteur Shopping bases essentiellement sur les delivrances obtenues par prescriptions multiples. Deuxieme approche : cette etude a ete completee par les donnees de l’etude pharmaco-epidemiologique transversale, multicentrique nationale, OPPIDUM menee en 2012. Les donnees, recueillies dans le cadre d’entretien aupres de patients suivis dans des centres de soins et de prevention des addictions, ont permis l’estimation de la prevalence de la consommation rapportee de sulfate de morphine, d’heroine et des OMT dans chaque region francaise. Seules les regions de plus de 50 consommateurs ont ete retenues. Resultats Les 1081 sujets selectionnes a partir des donnees de la CPAM ont ete regroupes en trois clusters dont un de 35 sujets au comportement juge deviant : poly-prescriptions (mediane : 15 versus 11 et 8), nombreux medecins (mediane : 5 versus 2 et 1) et pharmacies (mediane : 5 versus 2 et 1), consommation journaliere de Skenan® elevee (mediane : 277 mg versus 52 et 46). Ce sous-groupe apparait significativement plus jeune (mediane : 40 ans versus 53 et 58) et plus masculin (66 % versus 42 % et 45 %). Il est egalement caracterise par une consommation simultanee d’autres traitements (benzodiazepine, OMT) et de delivrances de Skenan® a des dosages eleves (100 et 200 mg). Les indicateurs de docteur shopping (DS) temoignent egalement d’une consommation excessive (DSI : 21,6 % versus 1,97 % et 0,10 %). Les resultats de l’etude OPPIDUM [3], recensant 4366 sujets, montrent de maniere graphique que le LR apparait parmi les regions les plus consommatrices de sulfate de morphine, alors que dans certaines regions, ou cette consommation est moindre, on observe une consommation plus elevee en heroine ou OMT. Conclusion Cette etude montre qu’en LR, le sulfate de morphine, (Skenan®), est largement utilise et parfois detourne de son usage habituel par des sujets au comportement juge deviant. Une des raisons avancee serait une faible disponibilite et une moindre qualite de l’heroine dans la region LR.

Abstract: GOLF in-flight commissioning and calibration was carried out during the first four months, most of which represented the cruise phase of SOHO towards its final LI orbit. The initial performance of GOLF is shown to be within the design specification, for the entire instrument as well as for the separate sub-systems. Malfunctioning of the polarising mechanisms after 3 to 4 months operation has led to the adoption of an unplanned operating sequence in which these mechanisms are no longer used. This mode, which measures only the blue wing of the solar sodium lines, detracts little from the detection and frequency measurements of global oscillations, but does make more difficult the absolute velocity calibration, which is currently of the order of 20%. Data continuity in the new mode is extremely high and the instrument is producing exceptionally noise-free p-mode spectra. The data set is particularly well suited to the study of effects due to the excitation mechanism of the modes, leading to temporal variations in their amplitudes. The g modes have not yet been detected in this limited data set. In the present mode of operation, there are no indications of any degradation which would limit the use of GOLF for up to 6 years or more.

Abstract: Context. The High Frequency Instrument (HFI) is one of the two focal instruments of the Planck mission. It will observe the whole sky in six bands in the 100 GHz–1 THz range.\ud \ud Aims. The HFI instrument is designed to measure the cosmic microwave background (CMB) with a sensitivity limited only by fundamental sources: the photon noise of the CMB itself and the residuals left after the removal of foregrounds. The two high frequency bands will provide full maps of the submillimetre sky, featuring mainly extended and point source foregrounds. Systematic effects must be kept at negligible levels or accurately monitored so that the signal can be corrected. This paper describes the HFI design and its characteristics deduced from ground tests and calibration.\ud \ud Methods. The HFI instrumental concept and architecture are feasible only by pushing new techniques to their extreme capabilities, mainly: (i) bolometers working at 100 mK and absorbing the radiation in grids; (ii) a dilution cooler providing 100 mK in microgravity conditions; (iii) a new type of AC biased readout electronics and (iv) optical channels using devices inspired from radio and infrared techniques.\ud \ud Results. The Planck-HFI instrument performance exceeds requirements for sensitivity and control of systematic effects. During ground-based calibration and tests, it was measured at instrument and system levels to be close to or better than the goal specification.

Abstract: Context. The Planck satellite was successfully launched on May 14th 2009. We have completed the pre-launch calibration measurements of the High Frequency Instrument (HFI) on board Planck and their processing.\ud \ud Aims. We present the results ot the pre-launch calibration of HFI in which we have multiple objectives. First, we determine instrumental parameters that cannot be measured in-flight and predict parameters that can. Second, we take the opportunity to operate and understand the instrument under a wide range of anticipated operating conditions. Finally, we estimate the performance of the instrument built.\ud \ud Methods. We obtained our pre-launch calibration results by characterising the component and subsystems, then by calibrating the focal plane at IAS (Orsay) in the Saturne simulator, and later from the tests at the satellite level carried out in the CSL (Liège) cryogenic vacuum chamber. We developed models to estimate the instrument pre-launch parameters when no measurement could be performed.\ud \ud Results. We reliably measure the Planck-HFI instrument characteristics and behaviour, and determine the flight nominal setting of all parameters. The expected in-flight performance exceeds the requirements and is close or superior to the goal specifications.

17. Differential remodeling of the HIV-1 nucleosome upon transcription activators and SWI/SNF complex binding

Abstract: Here we have examined HIV-1 nucleosome remodeling upon the binding of transcription factors and the SWI/SNF complex using a novel approach. The approach combines UV laser protein-DNA crosslinking, electrophoretic mobility-shift analysis and DNase I protection analysis with immunochemical techniques. It was found that single activator-bound HIV-1 nucleosomes exhibit very weak perturbation in histone NH(2) tail-DNA interactions. However, the simultaneous binding of the transcription activators Sp1, NF-kB1, LEF-1 and USF synergistically increased the release of histone NH(2) tails from nucleosomal DNA. In contrast, the binding of SWI/SNF complex to HIV-1 nucleosome disrupted structured histone domain-DNA contacts, but not histone NH(2) tail-DNA interactions. Stable remodeled nucleosomes, (obtained after detachment of SWI/SNF), displayed identical structural alterations with those bound to SWI/SNF. These results demonstrate a different in vitro remodeling of the HIV-1 nucleosome upon the binding of multiple transcription activators and of SWI/SNF complex.

Abstract: The Planck High Frequency Instrument (HFI) is designed to measure the temperature and polarization anisotropies of the cosmic microwave background and Galactic foregrounds in six ~30% bands centered at 100, 143, 217, 353, 545, and 857 GHz at an angular resolution of 10′ (100 GHz), 7′ (143 GHz), and 5′ (217 GHz and higher). HFI has been operating flawlessly since launch on 14 May 2009, with the bolometers reaching 100 mK the first week of July. The settings of the readout electronics, including bolometer bias currents, that optimize HFI's noise performance on orbit are nearly the same as the ones chosen during ground testing. Observations of Mars, Jupiter, and Saturn have confirmed that the optical beams and the time responses of the detection chains are in good agreement with the predictions of physical optics modeling and pre-launch measurements. The Detectors suffer from a high flux of cosmic rays due to historically low levels of solar activity. As a result of the redundancy of Planck's observation strategy, theremoval of a few percent of data contaminated by glitches does not significantly affect the instrumental sensitivity. The cosmic ray flux represents a significant and variable heat load on the sub-Kelvin stage. Temporal variation and the inhomogeneous distribution of the flux results in thermal fluctuations that are a probable source of low frequency noise. The removal of systematic effects in the time ordered data provides a signal with an average noise equivalent power that is 70% of the goal in the 0.6-2.5 Hz range. This is slightly higher than was achieved during the pre-launch characterization but better than predicted in the early phases of the project. The improvement over the goal is a result of the low level of instrumental background loading achieved by the optical and thermal design of the HFI.

Abstract: Ellipticines are intercalating planar polycyclic aromatic molecules that display antitumor activity. The cytotoxicity of these compounds is related to the presence of an hydroxy group at position 9 of the pyridocarbazole ring system and to their interaction with DNA topoisomerase II. The ability of 13 ellipticine derivatives to stabilize the topoisomerase II-DNA covalent complex in vitro is reported. The following observations emerge from our structure-activity relationship study: i) the hydroxy group at position 9 is essential for stabilizing the covalent complex, ii) the replacement of the methyl group at position 5 by an ethyl group (EPC) enhances the complex stabilization. The interaction of EPC and three other ellipticine analogues with DNA shows that the covalent complexes which are most stable have the lowest drug-DNA binding constants. In addition our study suggests that ellipticines induce covalent complex stabilization by a cooperative mechanism. A model is proposed to explain this stabilization by ellipticines. This study supports the idea that topoisomerase II is the primary target involved in the mechanisms of action of ellipticines.

Abstract: Ellipticines are aromatic compounds that intercalate between DNA base pairs and display significant antitumor activity. The cytotoxicity of these compounds is mediated by DNA topoisomerase II, and the presence of a hydroxy group at position 9 of the pyridocarbazole ring system of ellipticines has been found to be essential for high levels of cytotoxicity. The ability of 13 ellipticine derivatives to stabilize the topoisomerase II-DNA covalent complex in vitro was studied, and the data obtained with five pairs of hydroxylated and nonhydroxylated analogues indicate that the hydroxy group at position 9 plays a crucial role in the stabilization of the complex. The influence, upon the complex stabilization, of various substituents at positions 1, 2, 5, and 6 of the pyridocarbazole ring system was investigated. The interaction with DNA of four ellipticine derivatives was studied in the topoisomerase II standard medium. Results suggest that the degree of unwinding might be a determinant of topoisomerase II-DNA-drug complex stability. In addition, the 5-ethyl derivative was observed to induce covalent complex stabilization by a cooperative mechanism.

Abstract: Cytotoxic effects and topoisomerase II-mediated DNA breaks induced in vitro by ellipticine derivatives were examined in connection with 1H NMR and circular dichroism (CD) studies on molecular structures and interactions of drugs with DNA. The compounds included four 9-hydroxyellipticine and two 7-hydroxyisoellipticine derivatives. Structure-activity relationships indicated that a change in nitrogen atom position in the pyridinic ring greatly affected drug effects both on topoisomerase II action and cytotoxicity to L1210 cells. The four 9-hydroxyellipticine derivatives yielded bell-shaped curves in in vitro topoisomerase II-mediated DNA break assays, whereas the two 7-hydroxyisoellipticine derivatives demonstrated an almost linear increase at the same concentration (0-10 microM). In both cases, the intensity of cleavage was modulated by the position and the degree of methylation on the pyridinic ring, and results were correlated with cytotoxic activity expressed as the in vitro ID50 values for L1210 leukemia cells. 1H NMR experiments performed on free drug molecules in solution revealed that the two protons (alpha and beta) contiguous to the biologically important hydroxy group were sensitive to changes in electron distribution produced by the distant chemical modifications and methylations of the pyridinic ring. A linear relationship was observed between the differences in chemical shifts of alpha and beta protons (delta delta alpha-beta) versus ID50 values. CD experiments indicated that, at weak ionic strength I = 0.02 and at pH 7, drugs interact with the poly[d(A-T)] duplex according to a "three-mode binding model" which is governed by the drug structure and the drug to DNA ratio. The intercalation mode was related to the induction of topoisomerase II-mediated DNA cleavage, while the external binding mode consecutive to intercalation was related to cleavage suppression. These two modes concerned the good intercalators 9-hydroxyellipticines. The third was found for the weak intercalators 7-hydroxyisoellipticines and was characterized by self-stacked molecules bound "outside" DNA, presumably in the minor groove. Ligands either could be intercalated partially or linked at the edge of bases with a small number of molecules filling intercalation sites, for the second alternative. In addition to having different binding modes, 9-hydroxyellipticines were better inducers of DNA distortions than 7-hydroxyisoellipticines. The incidence of the drug binding modes on DNA-topoisomerase II recognition was discussed in connection with the in vitro cytotoxic activity exhibited by the drugs.

Abstract: Twenty-four patients with cirrhosis were investigated. A nutritional study and an assessment of cellular immunity were performed prior to portacaval shunt and 2 months, 6 months and 12 months following this procedure. Serum bilirubin concentration and coagulation factors II and V were also measured. Following portacaval shunting, there was a progressive and significant increase of arm muscle circumference (P < 0.001) and serum albumin concentration (P < 0.001) as well as an improvement in delayed hypersensitivity and an increase of peripheral T lymphocytes. Serum bilirubin concentration significantly increased and coagulation factors significantly decreased. Among the 20 patients with alcoholic cirrhosis, 19 stopped drinking: 17 of them markedly improved their nutritional and immunological parameters whereas two remained unchanged. One patient pursued a heavy alcohol intake and his parameters deteriorated. The four patients with non alcoholic cirrhosis had mild malnutrition prior to portal systemic shunt which was not significantly altered by this procedure. These results suggest that: a) portacaval shunt does not aggravate the pre-existing malnutrition in cirrhotic patients; b) there is no relationship between the variations of nutritional status and that of liver function; c) improvement of the nutritional status observed in alcoholics following portal diversion is most likely related to alcohol abstinence.

Abstract: The privileged status of liver allografts compared with other organ allografts is illustrated by the spontaneous tolerance of liver allografts in pigs (Calne 1973) and in several combinations of inbred rats (Houssin et al. 1979; Zimmermann et al. 1979). Another remarkable peculiarity of liver transplantation is the absence of hyperacute rejection of liver allografts in man in case of an ABO-incompatible transplantation or when preformed lymphocytotoxic antibodies directed toward the donor are present in the recipient (Starzl et al. 1982). The reason for the relative resistance of the liver to cytotoxic antibodies is still unclear. In order to study further the effect of cytotoxic antibodies on the liver, the fate of donor-specific liver allografts was compared with that of donor-specific heart allografts in hypersensitized rats.

Abstract: In this paper we discuss the Planck-LFI in-flight calibration campaign. After a brief overview of the ground test campaigns, we describe in detail the calibration and performance verification (CPV) phase, carried out in space during and just after the cool-down of LFI. We discuss in detail the functionality verification, the tuning of the front-end and warm electronics, the preliminary performance assessment and the thermal susceptibility tests. The logic, sequence, goals and results of the in-flight tests are discussed. All the calibration activities were successfully carried out and the instrument response was comparable to the one observed on ground. Abstract: Manure from animal production is commonly spread on agricultural soil as an organic fertiliser to provide macro and trace elements to crops. However, some trace elements can accumulate in the soil and become toxic to plants and microorganisms. These elements include copper (Cu) and zinc (Zn), which can be applied in large quantities when pig manure is spread. The feeding strategy and manure management (e.g. through treatment chains) are two mechanisms identified to better control the use of these elements, but their fate from the feed to the soil in pig production remains poorly documented. Better understanding the fate of Cu and Zn, as well as that of other trace and macro elements, along the feed - excreta - waste chain is required to develop alternative ways to reduce their environmental impacts. This dataset provides insight into the composition (Cu, Zn and other trace and macro elements) of organic products along two contrasting manure management chains: (1) only storage or (2) in-building separation, anaerobic digestion (AD) of solids, and digestate drying. Abstract: Studies about the impact of future climate change on diseases have mostly focused on standard Representative Concentration Pathway climate change scenarios. These scenarios do not account for the non-linear dynamics of the climate system. A rapid ice-sheet melting could occur, impacting climate and consequently societies. Here, we investigate the additional impact of a rapid ice-sheet melting of Greenland on climate and malaria transmission in Africa using several malaria models driven by Institute Pierre Simon Laplace climate simulations. Results reveal that our melting scenario could moderate the simulated increase in malaria risk over East Africa, due to cooling and drying effects, cause a largest decrease in malaria transmission risk over West Africa and drive malaria emergence in southern Africa associated with a significant southward shift of the African rain-belt. We argue that the effect of such ice-sheet melting should be investigated further in future public health and agriculture climate change risk assessments.

Abstract: Slow-release oral morphine sulfate (SM) is one of the most abused prescription opioids in France. However, the regional feature of the abuse of morphine is poorly documented. To investigate the abuse of SM in a French administrative region, a multisource approach was applied. The first approach was based on SM exposition at national and regional level using the OPPIDUM survey. In a second approach, we analyzed a drug reimbursement database to assess the magnitude of SM abuse in Languedoc-Roussillon (LR) region. A clustering method was applied to classify patients and to describe the profile of deviant patients. The third approach was based on a self-administered anonymous questionnaire, proposed to patients seen in addiction care centers in the LR region and consuming oral SM. The OPPIDUM study showed that in most regions, where the prevalence of heroin use is higher than the national average of 9.1%, SM consumers were fewer. With the clustering method, three subgroups were identified. One of them gathered 35 users (3.2%) with a deviant behavior characterized by significantly more dispensations, dispensing pharmacies, and prescribers. These subjects were mainly men, younger, and more consumers of benzodiazepines and opioid maintenance therapy than the others. The third study allowed specifying that SM was mainly injected (93.7%), bought in the street (80%), and used because of unavailability and the poor quality of heroin (33.9%). Abstract: In this study, we describe a new approach for studying protein-DNA interactions in solution. The approach is based on mapping the UV laser-induced protein-DNA cross-links between the amino acids of the protein and the DNA bases that are in direct contact. The approach was applied for studying the solution structure of the human necrosis factor (NF)-kappaB p50 homodimer bound to a 37 base pair DNA. Several points of contact identical to those observed in the NF-kappaB-DNA crystal structure were found between the two biomolecules. Evidence is provided for the occurrence of two new contact points, one for each DNA strand. These new points of contact are located symmetrically a base apart from the extremity of the binding sequence.

Abstract: In eukaryotes cell division is accompanied by phosphorylation of histone H3 at serine 10. In this work we have studied the kinase activity responsible for this histone H3 modification by using cell-free extracts prepared from Xenopus eggs. We have found that the Xenopus aurora-A kinase pEg2, immunoprecipitated from the extract, is able to phosphorylate specifically histone H3 at serine 10. The enzyme is incorporated into chromatin during in vitro chromosome assembly, and the kinetics of this incorporation parallels that of histone H3 phosphorylation. Recombinant pEg2 phosphorylates efficiently histone H3 at serine 10 in reconstituted nucleosomes and in sperm nuclei decondensed in heated extracts. These data identify pEg2 as a good candidate for mitotic histone H3 kinase. However, immunodepletion of pEg2 does not interfere with the chromosome assembly properties of the extract nor with the pattern of H3 phosphorylation, suggesting the existence of multiple kinases involved in this H3 modification in Xenopus eggs. This hypothesis is supported by in gel activity assay experiments using extracts from Saccharomyces cerevisiae.

Abstract: Here we have examined HIV-1 nucleosome remodeling upon the binding of transcription factors and the SWI/SNF complex using a novel approach. The approach combines UV laser protein-DNA crosslinking, electrophoretic mobility-shift analysis and DNase I protection analysis with immunochemical techniques. It was found that single activator-bound HIV-1 nucleosomes exhibit very weak perturbation in histone NH(2) tail-DNA interactions. However, the simultaneous binding of the transcription activators Sp1, NF-kB1, LEF-1 and USF synergistically increased the release of histone NH(2) tails from nucleosomal DNA. In contrast, the binding of SWI/SNF complex to HIV-1 nucleosome disrupted structured histone domain-DNA contacts, but not histone NH(2) tail-DNA interactions. Stable remodeled nucleosomes, (obtained after detachment of SWI/SNF), displayed identical structural alterations with those bound to SWI/SNF. Abstract: Ellipticines are intercalating planar polycyclic aromatic molecules that display antitumor activity. The cytotoxicity of these compounds is related to the presence of an hydroxy group at position 9 of the pyridocarbazole ring system and to their interaction with DNA topoisomerase II. The ability of 13 ellipticine derivatives to stabilize the topoisomerase II-DNA covalent complex in vitro is reported. The following observations emerge from our structure-activity relationship study: i) the hydroxy group at position 9 is essential for stabilizing the covalent complex, ii) the replacement of the methyl group at position 5 by an ethyl group (EPC) enhances the complex stabilization. The interaction of EPC and three other ellipticine analogues with DNA shows that the covalent complexes which are most stable have the lowest drug-DNA binding constants. In addition our study suggests that ellipticines induce covalent complex stabilization by a cooperative mechanism. A model is proposed to explain this stabilization by ellipticines. This study supports the idea that topoisomerase II is the primary target involved in the mechanisms of action of ellipticines.

Abstract: Ellipticines are aromatic compounds that intercalate between DNA base pairs and display significant antitumor activity. The cytotoxicity of these compounds is mediated by DNA topoisomerase II, and the presence of a hydroxy group at position 9 of the pyridocarbazole ring system of ellipticines has been found to be essential for high levels of cytotoxicity. The ability of 13 ellipticine derivatives to stabilize the topoisomerase II-DNA covalent complex in vitro was studied, and the data obtained with five pairs of hydroxylated and nonhydroxylated analogues indicate that the hydroxy group at position 9 plays a crucial role in the stabilization of the complex. The influence, upon the complex stabilization, of various substituents at positions 1, 2, 5, and 6 of the pyridocarbazole ring system was investigated. The interaction with DNA of four ellipticine derivatives was studied in the topoisomerase II standard medium. Results suggest that the degree of unwinding might be a determinant of topoisomerase II-DNA-drug complex stability. In addition, the 5-ethyl derivative was observed to induce covalent complex stabilization by a cooperative mechanism.

Abstract: Cytotoxic effects and topoisomerase II-mediated DNA breaks induced in vitro by ellipticine derivatives were examined in connection with 1H NMR and circular dichroism (CD) studies on molecular structures and interactions of drugs with DNA. The compounds included four 9-hydroxyellipticine and two 7-hydroxyisoellipticine derivatives. Structure-activity relationships indicated that a change in nitrogen atom position in the pyridinic ring greatly affected drug effects both on topoisomerase II action and cytotoxicity to L1210 cells. The four 9-hydroxyellipticine derivatives yielded bell-shaped curves in in vitro topoisomerase II-mediated DNA break assays, whereas the two 7-hydroxyisoellipticine derivatives demonstrated an almost linear increase at the same concentration (0-10 microM). In both cases, the intensity of cleavage was modulated by the position and the degree of methylation on the pyridinic ring, and results were correlated with cytotoxic activity expressed as the in vitro ID50 values for L1210 leukemia cells. 1H NMR experiments performed on free drug molecules in solution revealed that the two protons (alpha and beta) contiguous to the biologically important hydroxy group were sensitive to changes in electron distribution produced by the distant chemical modifications and methylations of the pyridinic ring. A linear relationship was observed between the differences in chemical shifts of alpha and beta protons (delta delta alpha-beta) versus ID50 values. CD experiments indicated that, at weak ionic strength I = 0.02 and at pH 7, drugs interact with the poly[d(A-T)] duplex according to a "three-mode binding model" which is governed by the drug structure and the drug to DNA ratio. The intercalation mode was related to the induction of topoisomerase II-mediated DNA cleavage, while the external binding mode consecutive to intercalation was related to cleavage suppression. These two modes concerned the good intercalators 9-hydroxyellipticines. The third was found for the weak intercalators 7-hydroxyisoellipticines and was characterized by self-stacked molecules bound "outside" DNA, presumably in the minor groove. Ligands either could be intercalated partially or linked at the edge of bases with a small number of molecules filling intercalation sites, for the second alternative. In addition to having different binding modes, 9-hydroxyellipticines were better inducers of DNA distortions than 7-hydroxyisoellipticines. The incidence of the drug binding modes on DNA-topoisomerase II recognition was discussed in connection with the in vitro cytotoxic activity exhibited by the drugs.

Abstract: Natural killer (NK) cell activity against two types of target cells was found to be low in patients with inactive alcoholic cirrhosis (AC). This defect was significantly more pronounced in AC patients with severe malnutrition than in those with mild or moderate malnutrition. This was not due to modifications of the kinetics of NK activity. The sera from AC patients had no inhibitory effect on the NK activity of normal subjects. Lymphocytes and macrophages from AC patients did not exert major suppressive effect on the NK activity of normal subjects. Interferon boosted the NK activity of cells from AC patients, but to a lesser degree than cells from normal controls. The findings show that a deficit of NK activity is clearly associated with inactive AC. This seems to be another consequence of AC on cellular immunity, and might be related to the protein calorie malnutrition often present in AC.

Abstract: Severe malnutrition and related impaired cell-mediated immunity are commonly found in cirrhotic patients with intractable ascites and may be responsible for the poor prognosis of these patients. The effects of LeVeen peritoneovenous shunting of ascites on protein reserves and cell-mediated immunity were studied in 31 cirrhotic patients with intractable ascites over a period of 1 year following operation. Arm-muscle circumference, serum albumin, and transferrin levels markedly increased and became close to the normal values. In the same period, delayed hypersensitivity improved. Although the lymphocyte count and the absolute T cell concentration in peripheral blood remained low after peritoneovenous shunting, the in vitro lymphoblastic transformation in response to phytohemagglutinin was augmented. An increased capacity to eat normal meals and the resumption of a good appetite due to the discontinuation of a salt-restricted diet seemed to be the most important factors in the dramatic improvement in the nutritional status of these patients. Restriction from heavy alcohol use may have also contributed, although the patients had already stopped drinking for several months before operation. Improvement in cell-mediated immunity might have been secondary to the increase in protein reserves.
