Mushroom cultivation vastly improves the yield of mushrooms under optimized, controlled conditions, but may be susceptible to opportunistic colonization by pest species that can establish themselves, as well as the pathogens and pests they may transmit. Here, we describe our investigation into the bacterial communities of adult Lycoriella ingenua (Diptera: Sciaridae) and Megaselia halterata (Diptera: Phoridae) collected from button mushroom (Agaricus bisporus) production houses in Pennsylvania. We collected adult flies and sequenced the hypervariable v4 region of the bacterial 16S rRNA using the Illumina MiSeq. The most abundant bacterial genus detected in both species was Wolbachia, but phylogenetic analysis revealed that the infections are from different clades. Future studies include the characterization of Wolbachia infections on fly behavior and biology, comparison of microbial diversity of fly species colonizing wild mushrooms, and other microbiota that may contribute to the success of certain pest fly species.

We investigated how the scanning strategy (SS) influences the magnetic properties of ferromagnetic metal specimens additively manufactured using 630 stainless steel powder and a directed energy deposition machine. The magnetization curves were measured by using B and H coils and a magnetic flux meter. The specific saturation magnetization, coercivity, and specific magnetic susceptibility were calculated from the magnetization curves obtained. Moreover, the martensite fraction, magnetization structure, and martensite structure were measured and visualized using X-ray diffraction, the Bitter method, and electron backscatter diffraction, respectively. The magnetic properties and martensite fraction depended on the SS, and magnetic poles were generated in each sub-block structure. Furthermore, a numerical analysis using a magnetic micromechanics model indicated that the variation in the martensite structure due to the SS produced differences in the magnetic properties.

Abstract Background Follicular dendritic cell sarcoma is a rare stromal tumor with no standard treatment. However, some reports have revealed that follicular dendritic cell sarcoma has an inflammatory pseudotumor variant associated with Epstein–Barr virus infection that has a relatively good prognosis. In this report, we present a case of a resected inflammatory pseudotumor variant of follicular dendritic cell sarcoma of the liver, and have reviewed the literature on the clinicopathological, molecular, and genomic features of this tumor. Case presentation The inflammatory pseudotumor variant of follicular dendritic cell sarcoma originates only in the liver or spleen, causes no symptoms, and is more common in middle-aged Asian women. It has no characteristic imaging features, which partially explains why the inflammatory pseudotumor variant of follicular dendritic cell sarcoma is difficult to diagnose. Pathologically, the inflammatory pseudotumor variant of follicular dendritic cell sarcoma has spindle cells mixed with inflammatory cells and is variably positive for follicular dendritic cell markers (CD21, CD23, and CD35) and Epstein–Barr virus-encoded RNA. On genetic analysis, patients with this tumor high levels of latent membrane protein 1 gene expression and extremely low levels of host C–X–C Chemokine Receptor type 7 gene expression, indicating that the inflammatory pseudotumor variant of follicular dendritic cell sarcoma has a latent Epstein–Barr virus type 2 infection. Conclusions The inflammatory pseudotumor variant of follicular dendritic cell sarcoma is an Epstein–Barr virus-associated tumor and a favorable prognosis by surgical resection, similar to Epstein–Barr virus-associated gastric cancer.

Background Type I interferons (IFN-Is), secreted by hematopoietic cells, drive immune surveillance of solid tumors. However, the mechanisms of suppression of IFN-I-driven immune responses in hematopoietic malignancies including B-cell acute lymphoblastic leukemia (B-ALL) are unknown.Methods Using high-dimensional cytometry, we delineate the defects in IFN-I production and IFN-I-driven immune responses in high-grade primary human and mouse B-ALLs. We develop natural killer (NK) cells as therapies to counter the intrinsic suppression of IFN-I production in B-ALL.Results We find that high expression of IFN-I signaling genes predicts favorable clinical outcome in patients with B-ALL, underscoring the importance of the IFN-I pathway in this malignancy. We show that human and mouse B-ALL microenvironments harbor an intrinsic defect in paracrine (plasmacytoid dendritic cell) and/or autocrine (B-cell) IFN-I production and IFN-I-driven immune responses. Reduced IFN-I production is sufficient for suppressing the immune system and promoting leukemia development in mice prone to MYC-driven B-ALL. Among anti-leukemia immune subsets, suppression of IFN-I production most markedly lowers the transcription of IL-15 and reduces NK-cell number and effector maturation in B-ALL microenvironments. Adoptive transfer of healthy NK cells significantly prolongs survival of overt ALL-bearing transgenic mice. Administration of IFN-Is to B-ALL-prone mice reduces leukemia progression and increases the frequencies of total NK and NK-cell effectors in circulation. Ex vivo treatment of malignant and non-malignant immune cells in primary mouse B-ALL microenvironments with IFN-Is fully restores proximal IFN-I signaling and partially restores IL-15 production. In B-ALL patients, the suppression of IL-15 is the most severe in difficult-to-treat subtypes with MYC overexpression. MYC overexpression promotes sensitivity of B-ALL to NK cell-mediated killing. To counter the suppressed IFN-I-induced IL-15 production in MYChigh human B-ALL, we CRISPRa-engineered a novel human NK-cell line that secretes IL-15. CRISPRa IL-15-secreting human NK cells kill high-grade human B-ALL in vitro and block leukemia progression in vivo more effectively than NK cells that do not produce IL-15.Conclusion We find that restoration of the intrinsically suppressed IFN-I production in B-ALL underlies the therapeutic efficacy of IL-15-producing NK cells and that such NK cells represent an attractive therapeutic solution for the problem of drugging MYC in high-grade B-ALL.

Challenges in batch normalization and data integration limit the comparison of existing mass cytometry datasets. Here, the authors report CytofIn that can integrate mass cytometry datasets from the public domain and reveal cellular features associated with immune oncology by analyzing five public cancer datasets.

Introduction: Curettage and dermabrasion are effective in the treatment of giant congenital melanocytic nevi (GCMN); however, local infection and hypertrophic scar formation are major issues. Thus, we applied cultured epithelial autografts (CEA) on skin defects after curettage or abrasion of GCMN and assessed the postoperative outcomes. Methods: Seven nevi lesions of five patients (aged 3 months to 24 years) were treated with CEA after curettage or abrasion with a dermatome or a surgical bar, respectively. We assessed the postoperative outcomes, including CEA take ratio, erosion and/or ulcer formation in the acute phase, hospitalization days, Vancouver scar scale, and color improvement one year after the operation. In addition, a histological evaluation of a skin biopsy was performed over one year after the operation. Results: The CEAs took well on the wound, and the wound surface was mostly epithelized by postoperative day 7 in all cases. While hypertrophic scar formation and slight pigmentation were observed in some lesions, the color was improved in all of the treated lesions. Histopathological examination revealed that the regenerated epidermis had stratified keratinocytes with rete ridges, and the dermal layer without nevus cells regenerated above the remaining dermis layer. Conclusions: In this study, we found that early epithelialization and regeneration of the dermal layer was achieved after the application of CEA, suggesting that CEA could be an effective option after curettage or abrasion of GCMN.

Diamond-Blackfan anemia (DBA) is a ribosomopathy that is characterized by macrocytic anemia, congenital malformations, and early onset during childhood. Genetic studies have demonstrated that most patients carry mutations in one of the 20 related genes, most of which encode ribosomal proteins (RP). Treatment of DBA includes corticosteroid therapy, chronic red blood cell transfusion, and other forms of immunosuppression. Currently, hematopoietic stem cell transplantation is the only cure for DBA. Interestingly, spontaneous remissions occur in 10-20% of transfusion-dependent DBA patients. However, there is no consistent association between specific mutations and clinical manifestations. In the past decades, researchers have made significant progress in understanding the pathogenesis of DBA, but it remains unclear how the ubiquitous RP haploinsufficiency causes the erythroid-specific defect in hematopoiesis in DBA patients, and why there is a difference in penetrance and spontaneous remission among individuals who carry identical mutations. In this paper, we provide a comprehensive review of the development of DBA animal models and discuss the future research directions for these important experimental systems.

Clinical pathways are evidence-based tools that have been integrated into many aspects of pediatric hospital medicine and have proven effective at reducing in-hospital complications from a variety of diseases. Adaptation of similar tools for specific, high-risk patient populations in pediatric oncology has been slower, in part due to patient complexities and variations in management strategies. There are few published studies of clinical pathways for pediatric oncology patients. Pediatric patients with a new diagnosis of leukemia or lymphoma often present with one or more “oncologic emergencies” that require urgent intervention and deliberate multidisciplinary care to prevent significant consequences. Here, we present two clinical pathways that have recently been developed using a multidisciplinary approach at a single institution, intended for the care of patients who present with hyperleukocytosis or an anterior mediastinal mass. These clinical care pathways have provided a critical framework for the immediate care of these patients who are often admitted to the pediatric intensive care unit for initial management. The goal of the pathways is to facilitate multidisciplinary collaborations, expedite diagnosis, and streamline timely treatment initiation. Standardizing the care of high-risk pediatric oncology patients will ultimately decrease morbidity and mortality associated with these diseases to increase the potential for excellent outcomes.

Abstract Background Targeted anticancer therapies such as BCR-ABL tyrosine kinase inhibitors (TKIs) have improved outcomes for chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph + ALL). However, little is known about long-term risks of TKIs in children. Exposure-based survivorship guidelines do not include TKIs, thus surveillance practices may be variable. Methods We retrospectively examined surveillance for cardiac and endocrine late effects in children receiving TKIs for Ph + leukemias, diagnosed at

Surgical resection of skin tumors leads to large defects in surrounding normal tissues, which should be reconstructed thereafter using the patient's own tissues taken from the other site. Our challenge is to solve this problem in dermal malignant melanoma (MM) by a novel process, named extracorporeal high pressure therapy (EHPT), in which the tissue containing tumor is resected and pressurized, and the treated tissue is re-transplant back to the same position as a tumor-free autologous dermal substitute. The key points are complete tumor death and preservation of native extra cellular matrix (ECM) by the hydrostatic pressure. We found that high hydrostatic pressure at 200 MPa for 10 min at room temperature is completely cytocidal against MM cells in suspension form, in monolayer form, and even in the solid tumor form. MM tumor-bearing nude mice were established by injected human MM cells intradermally and treated by EHTP. The denaturation of the dermal extra cellular matrices was so mild that the pressurized skin was well engrafted as tumor free autologous dermal tissues, resulting in the complete eradication of the MM without any unnecessary skin reconstruction surgery. This very simple and short pressing treatment was proved to make the tumor tissue to the transplantable and tumor-free autologous dermal substitute, which can be applicable to the other temporally resectable tissues.

Oxygen isotope ratios (δ18O) of tree-ring cellulose are a novel proxy for summer hydroclimate in monsoonal Asia. In central Japan, we collected 67 conifer wood samples, mainly Chamaecyparis obtusa, with ages encompassing the past 2600 years. The samples were taken from living trees, archeological wood, architectural wood, and buried logs. We analyzed stable isotope ratios of oxygen (δ18O) and hydrogen (δ2H) in tree-ring cellulose in these samples (more than 15 000 rings in total) without using a pooling method and constructed a statistically reliable tree-ring cellulose δ18O time series for the past 2500 years. However, there were distinct age trends and level offsets in the δ18O record, and cellulose δ18O values showed a gradual decrease as an individual tree matures. This suggested it is difficult to establish a cellulose δ18O chronology for low-frequency signals by simple averaging of all the δ18O time series data. In addition, there were opposite age trends in the cellulose δ2H, and δ2H gradually increased with tree age. There were clear positive correlations in the short-periodicity variations between δ18O and δ2H, probably indicating a common climate signal. A comparison of the δ18O and δ2H time series in individual trees with tree-ring width suggested that the opposite age trends of δ18O and δ2H are caused by temporal changes in the degree of post-photosynthetic isotope exchange with xylem water (physiological effect), accompanied by changes in stem growth rate that are influenced by human activity in the forests of central Japan. Based on the assumptions that cellulose δ18O and δ2H vary positively and negatively with constant proportional coefficients due to climatological and physiological effects, respectively, we solved simultaneous equations for the climatological and physiological components of variations in tree-ring cellulose δ18O and δ2H in order to remove the age trend. This enabled us to evaluate the climatic record from cellulose δ18O variations. The extracted climatological component in the cellulose δ18O for the past 2600 years in central Japan was well correlated with numerous instrumental, historical, and paleoclimatological records of past summer climate at various spatial and temporal scales. This indicates that integration of tree-ring cellulose δ18O and δ2H data is a promising method to reconstruct past summer climate variations on annual to millennial timescales, irrespective of the growth environment. However, analytical and statistical methods need to be improved for further development of this climate proxy.

Label placement is one of the most essential tasks in the fields of cartography and geographic information systems. Numerous studies have been conducted on the automatic label placement for the past few decades. In this study, we focus on automatic label placement of area-feature, which has been relatively less studied than that of point-feature and line-feature. Most of the existing approaches have adopted a rule-based algorithm, and there are limitations in expressing the characteristics of label placement for area-features of various shapes utilizing handcrafted rules, criteria, objective functions, etc. Hence, we propose a novel approach for automatic label placement of area-feature based on deep learning. The aim of the proposed approach is to obtain the complex and implicit characteristics of area-feature label placement by manual operation directly and automatically from training data. First, the area-features with vector format are converted into a binary image. Then a key-point detection model, which simultaneously detect and localize specific key-points from an image, is applied to the binary image to estimate the candidate positions of labels. Finally, the final label placement positions for each area-feature are determined via simple post-process. To evaluate the proposed approach, the experiments with cadastral data were conducted. The experimental results show that the ratios of the estimation errors within 1.2 m (corresponding to one pixel of the input image) were 92.6% and 94.5% in the center and upper-left placement style, respectively. It implies that the proposed approach could place the labels for area-features automatically and accurately.

The historical castles (castellated walls), which are cultural heritages in Japan, require regular maintenance, and it is necessary to record the arrangement of individual wall stones in the maintenance work. Recently, image processing techniques are practiced to optimize maintenance and management of the infrastructure assets. In the previous study, we proposed an automatic method for efficiently extracting individual wall stone polygons by improved multiscale image segmentation technique. However, the problem has remained that wall stone polygons could not be extracted properly when there were no clear gaps or boundaries between stones. To address this problem, we improved the multiscale image segmentation technique used in our previous studies. The first improvement is that in the region growing process, selecting the best combination of a plurality of objects instead of two. The second improvement is the modification of the shape criterion to be used. Besides, we proposed three-stage Stacked cGAN for wall stone edge detection that enables us to complement areas with weak or broken boundaries of stone edges. This approach is composed of a coarse-to-fine based image-to-edges translation network. The edge images derived from this method are used as the additional channel in multiscale image segmentation with a higher weight compared to the other RGB channels. It was confirmed that the separation performance of individual wall stone polygons was improved by the proposed method. Furthermore, the proposed method is highly effective to reduce the difficulty in setting of the scale parameter, which is usually sensitive to segmentation results and requires trial and error.

In this paper, we discuss how to improve the quality of classification results when deep learning is applied for the filtering of airborne LiDAR point cloud. We introduce the baseline method which utilizes convolutional neural network (CNN) based on voxelization, and then we propose three methods to improve the quality of classification result. The first method is data pre-processing that aims to exclude data in advance that is clearly not on the ground surface in order to efficiently extract the ground surface data. Data pre-processing can greatly reduce the number of target points and the subsequent processing can be performed efficiently. It also has the effect of preventing noise-like points floating in the air from being misclassified as the ground surface, as compared to the case without pre-processing. The second method is changing the network structure. In recent years, various networks have been proposed for classifying point clouds. In our study, the baseline is using very simple networks. In order to improve the classification result of the baseline method, the layer depth and the range size of convolution are changed, and we investigated about the improvements of the results. The current discussion can be used as a guidance when considering new networks. The third method is the integration of classification results from multiple networks. We integrated individual results from multiple networks with varying layer depths and convolution sizes, starting with the baseline, and investigated whether the results improved. We observed that even if the individual results were similar, the classification results can be improved by integrating the results.

Diamond Blackfan Anemia (DBA) is a congenital bone marrow failure syndrome that is associated with anemia. Here, the authors examine the role of Nemo-like kinase (NLK) in erythroid cells in the pathogenesis of DBA and as a potential target for therapy.

High-yield dairy cows need high energy feed during periods of increased milk production. The transitional feeding to high energy feed increases the risk of developing a variety of metabolic disorders. Here, five Holstein cows were fed a four-stage feeding protocol (3 weeks for each stage) ranging from 54.9 to 73.7% total digestive nutrients (TDN). The purpose of the study was to investigate the effect of lactic acid bacteria on high-energy-fed cows associated with transitional feeding, and to evaluate the effects of probiotics on intestinal bacterial changes and inflammatory responses. Three feed transition periods were established for five cows, and Lactobacillus plantarum RGU-LP1 (LP1) was fed as a probiotic during the high-energy feeding period. The number of lymphocyte subsets such as CD3-, CD4-, and CD8 positive cells decreased in response to the high energy feed. Lipopolysaccharide (LPS)-induced cytokine (IL-1β and IL-2) gene expression in peripheral blood mononuclear cells (PBMCs) was shown to increase in those animals receiving the high energy feed. However, supplementation with LP1 resulted in an increase in the number of lymphocyte subsets and the expression of IL-1β and IL-2 were returned to the level at low energy diet. These results suggest that high energy diets induce inflammatory cytokine responses following LPS stimulation, and that the addition of LP1 mitigates these results by regulating the LPS-induced inflammatory reaction. Therefore, the functional lactic acid bacteria LP1 is expected to regulate inflammation resulting from high energy feeding, and this probiotic could be applied to support inflammatory regulation in high-yield dairy cows.

A divertor simulation experimental module (D-module) in the tandem mirror GAMMA 10/PDX was used for the study of plasma detachment. In previous studies, it was difficult to measure far-upstream plasma parameters in the D-module, and only electrostatic probes on the target plate were used to perform electron temperature and density measurements. To study the detached plasma structure, a Thomson scattering (TS) system and a microwave interferometer system have been installed to measure the inside plasma parameters of the D-module, and a movable electrostatic probe has been placed at the inlet of the D-module to measure the inlet plasma density and temperature. The TS system in the central cell observed the electron temperature and density of the core plasma simultaneously. These measurements revealed the entire density and temperature structure from the core plasma to the divertor plate. The line average electron density measured by the microwave interferometer showed a rollover behavior during detachment. The results indicated that the ionization region was located around the center of the D-module, and it appears to move upstream along the axis.

Ticks are able to transmit the highest number of pathogen species of any blood-feeding arthropod and represent a growing threat to public health and agricultural systems worldwide. While there are numerous and varied causes and effects of changes to tick-borne disease (re)emergence, three primary challenges to tick control were identified in this review from a U.S. borders perspective. (1) Climate change is implicated in current and future alterations to geographic ranges and population densities of tick species, pathogens they can transmit, and their host and reservoir species, as highlighted by Ixodes scapularis and its expansion across southern Canada. (2) Modern technological advances have created an increasingly interconnected world, contributing to an increase in invasive tick species introductions through the increased speed and frequency of trade and travel. The introduction of the invasive Haemaphysalis longicornis in the eastern U.S. exemplifies the challenges with control in a highly interconnected world. (3) Lastly, while not a new challenge, differences in disease surveillance, control, and management strategies in bordering countries remains a critical challenge in managing ticks and tick-borne diseases. International inter-agency collaborations along the U.S.–Mexico border have been critical in control and mitigation of cattle fever ticks (Rhipicephalus spp.) and highlight the need for continued collaboration and research into integrated tick management strategies. These case studies were used to identify challenges and opportunities for tick control and mitigation efforts through a One Health framework.

The inability to extinguish a traumatic memory is a key aspect of post-traumatic stress disorder (PTSD). While PTSD affects 10–20% of individuals who experience a trauma, women are particularly susceptible to developing the disorder. Despite this notable female vulnerability, few studies have investigated this particular resistance to fear extinction observed in females. Similar to humans, rodent models of Pavlovian fear learning and extinction show a wide range of individual differences in fear learning and extinction, although female rodents are considerably understudied. Therefore, the present study examined individual differences in fear responses, including freezing behavior and ultrasonic vocalizations (USVs), of female Long–Evans rats during acquisition of fear conditioning and cued fear extinction. Similar to prior studies in males, female rats displayed individual variation in freezing during cued fear extinction and were divided into extinction competent (EC) and extinction resistant (ER) phenotypes. Differences in freezing between ER and EC females were accompanied by shifts in rearing during extinction, but no darting was seen in any trial. Freezing behavior during fear learning did not differ between the EC and ER females. Vocalizations emitted in the 22 and 50 kHz ranges during fear learning and extinction were also examined. Unlike vocalizations seen in previous studies in males, very few 22 kHz distress vocalizations were emitted by female rats during fear acquisition and extinction, with no difference between ER and EC groups. Interestingly, all female rats produced significant levels of 50 kHz USVs, and EC females emitted significantly more 50 kHz USVs than ER rats. This difference in 50 kHz USVs was most apparent during initial exposure to the testing environment. These results suggest that like males, female rodents show individual differences in both freezing and USVs during fear extinction, although females appear to vocalize more in the 50 kHz range, especially during initial periods of exposure to the testing environment, and emit very few of the 22 kHz distress calls that are typically observed in males during fear learning or extinction paradigms. Overall, these findings show that female rodents display fear behavior repertoires divergent from males.

The del(5q) myelodysplastic syndrome (MDS) is a distinct subtype of MDS, associated with deletion of the ribosomal protein S14 (RPS14) gene that results in macrocytic anemia. This study sought to identify novel targets for the treatment of patients with del(5q) MDS by performing an in vivo drug screen using an rps14-deficient zebrafish model. From this, we identified the secreted gelatinase matrix metalloproteinase 9 (MMP9). MMP9 inhibitors significantly improved the erythroid defect in rps14-deficient zebrafish. Similarly, treatment with MMP9 inhibitors increased the number of colony forming unit-erythroid colonies and the CD71+ erythroid population from RPS14 knockdown human BMCD34+ cells. Importantly, we found that MMP9 expression is upregulated in RPS14-deficient cells by monocyte chemoattractant protein 1. Double knockdown of MMP9 and RPS14 increased the CD71+ population compared with RPS14 single knockdown, suggesting that increased expression of MMP9 contributes to the erythroid defect observed in RPS14-deficient cells. In addition, transforming growth factor β (TGF-β) signaling is activated in RPS14 knockdown cells, and treatment with SB431542, a TGF-β inhibitor, improved the defective erythroid development of RPS14-deficient models. We found that recombinant MMP9 treatment decreases the CD71+ population through increased SMAD2/3 phosphorylation, suggesting that MMP9 directly activates TGF-β signaling in RPS14-deficient cells. Finally, we confirmed that MMP9 inhibitors reduce SMAD2/3 phosphorylation in RPS14-deficient cells to rescue the erythroid defect. In summary, these study results support a novel role for MMP9 in the pathogenesis of del(5q) MDS and the potential for the clinical use of MMP9 inhibitors in the treatment of patients with del(5q) MDS.

Abstract Background Tick-borne diseases have been increasing at the local, national, and global levels. Researchers studying ticks and tick-borne diseases need a thorough knowledge of the pathogens, vectors, and epidemiology of disease spread. Both active and passive surveillance approaches are typically used to estimate tick population size and risk of tick encounter. Our data consists of a composite of active and long-term passive surveillance, which has provided insight into spatial variability and temporal dynamics of ectoparasite communities and identified rarer tick species. We present a retrospective analysis on compiled data of ticks from Pennsylvania over the last 117 years. Methods We compiled data from ticks collected during tick surveillance research, and from citizen-based submissions. The majority of the specimens were submitted by citizens. However, a subset of the data was collected through active methods (flagging or dragging, or removal of ticks from wildlife). We analyzed all data from 1900–2017 for tick community composition, host associations, and spatio-temporal dynamics. Results In total there were 4491 submission lots consisting of 7132 tick specimens. Twenty-four different species were identified, with the large proportion of submissions represented by five tick species. We observed a shift in tick community composition in which the dominant species of tick (Ixodes cookei) was overtaken in abundance by Dermacentor variabilis in the early 1990s and then replaced in abundance by I. scapularis. We analyzed host data and identified overlaps in host range amongst tick species. Conclusions We highlight the importance of long-term passive tick surveillance in investigating the ecology of both common and rare tick species. Information on the geographical distribution, host-association, and seasonality of the tick community can help researchers and health-officials to identify high-risk areas.

This research numerically investigates the behavior of plasma flow in GAMMA 10/PDX. The impact of impurity gases seeding into the divertor region on the plasma parameters and the energy loss processes has been investigated numerically by using the multi-fluid code “LINDA”. The neutral particles are injected into the bulk hydrogen plasma and the upstream plasma parameters on the z-axis are ni=1.0×1019m−3, Ti=100eV,Te=30eV. A reduction in the plasma temperature has been observed with the increasing H neutral injection. The ionization and Charge-exchange (CX) power loss increase almost linearly with the increasing H neutral density, which induce a reduction in the plasma temperature. During H seeding, Ar and Ne are seeded simultaneously to observe the influence of Ar and Ne on the electron and ion energy loss mechanism. In this study, the fraction of impurity density to upstream plasma density (nz/ni) is used to be 10%. A comparison between Ar and Ne shows that Ar is effective on the electron cooling. The radiation power loss of Ar is found to be higher than that of Ne. The radiation cooling effect and CX loss significantly reduces the electron and ion temperature, respectively. A slight influence of Ar and Ne seeding on the ion energy loss terms has been observed. The electron energy loss terms increase significantly during impurity gas Ar and Ne seeding. These outcomes represent the impact of gas seeding for reducing the plasma energy on the divertor target plates. The transition of a detached to attached plasma experiment has been performed in GAMMA 10/PDX by applying an additional plasma heating pulse of electron cyclotron heating (ECH). In a plasma detachment experiment, a short pulse (25 ms) of ECH was applied at the east plug-cell to examine the effects of electron heating on the plasma parameters in the D-module. It was found that the ion flux increased significantly during ECH injection. In the ECH injection period, the ion flux increased with the increasing impurity injection, which indicates the impurity ion components are enhanced by ionization in the D-module due to the application of ECH. During ECH injection period, the increase of the ion flux near the corner of the target plate shows a clear dependence on the gas species. The 2D visible emission has been captured by the high-speed camera and a bright emission near the corner of the target has been observed. The spectroscopic measurement in the D-module shows that the impurity ion emission increases remarkably during ECH injection. These results indicate the detached plasma change to the attached state. We have performed scanning tunneling microscopy (STM) and angle-resolved photoemission spectroscopy (ARPES) in Pb-deposited bilayer graphene (BLG) on the SiC(0001) substrate to investigate the dependence of the electronic structures on the Pb-deposition amount. We have observed that the Pb atoms form islands by STM and the π bands of the BLG shift toward the Fermi level by ARPES. This hole-doping-like energy shift is enhanced as the amount of Pb is increased, and we were able to tune the Dirac gap to the Fermi level by 4 ML deposition. Considering the band dispersion, we suggest that the hole-doping-like effect is related to the difference between the work functions of Pb islands and BLG/SiC; the work function of BLG/SiC is lower than that of Pb. Our results propose an easy way of band tuning for graphene with an appropriate selection of both the substrate and deposited material.

Castellated walls are positioned as tangible cultural heritage, which require regular maintenance to preserve their original state. For the demolition and repair work of the castellated wall, it is necessary to identify the individual stones constituting the wall. However, conventional approaches using laser scanning or integrated circuits (IC) tags were very time-consuming and cumbersome. Therefore, we herein propose an efficient approach for castellated wall management based on an extended multiscale image segmentation technique. In this approach, individual stone polygons are extracted from the castellated wall image and are associated with a stone management database. First, to improve the performance of the extraction of individual stone polygons having a convex shape, we developed a new shape criterion named convex hull fitness in the image segmentation process and confirmed its effectiveness. Next, we discussed the stone management database and its beneficial utilization in the repair work of castellated walls. Subsequently, we proposed irregular-shape indexes that are helpful for evaluating the stone shape and the stability of the stone arrangement state in castellated walls. Finally, we demonstrated an application of the proposed method for a typical castellated wall in Japan. Consequently, we confirmed that the stone polygons can be extracted with an acceptable level. Further, the condition of the shapes and the layout of the stones could be visually judged with the proposed irregular-shape indexes.

In recent years, extensive research has been conducted to automatically generate high-accuracy and high-precision road orthophotos using images and laser point cloud data acquired from a mobile mapping system (MMS). However, it is necessary to mask out non-road objects such as vehicles, bicycles, pedestrians and their shadows in MMS images in order to eliminate erroneous textures from the road orthophoto. Hence, we proposed a novel vehicle and its shadow detection model based on Faster R-CNN for automatically and accurately detecting the regions of vehicles and their shadows from MMS images. The experimental results show that the maximum recall of the proposed model was high – 0.963 (intersection-over-union > 0.7) – and the model could identify the regions of vehicles and their shadows accurately and robustly from MMS images, even when they contain varied vehicles, different shadow directions, and partial occlusions. Furthermore, it was confirmed that the quality of road orthophoto generated using vehicle and its shadow masks was significantly improved as compared to those generated using no masks or using vehicle masks only.

Ribosomal S6 Kinases (RSKs) are a group of serine/threonine kinases that function downstream of the Ras/Raf/MEK/ERK signaling pathway. Four RSK isoforms are directly activated by ERK1/2 in response to extracellular stimuli including growth factors, hormones, and chemokines. RSKs phosphorylate many cytosolic and nuclear targets resulting in the regulation of diverse cellular processes such as cell proliferation, survival, and motility. In hematological malignancies such as acute myeloid leukemia (AML), RSK isoforms are highly expressed and aberrantly activated resulting in poor outcomes and resistance to chemotherapy. Therefore, understanding RSK function in leukemia could lead to promising therapeutic strategies. This review summarizes the current information on human RSK isoforms and discusses their potential roles in the pathogenesis of AML and mechanism of pharmacological inhibitors.

In the tandem mirror GAMMA 10/PDX, molecular activated recombination (MAR) leading to plasma detachment has been observed by additional hydrogen gas injection to the divertor simulation plasma (i.e. end loss plasma) which is exposed to the V-shaped target in the divertor simulation experimental module (D-module). The temperature near the corner of the V-shaped target decreased from ∼23eV to ∼2eV as the neutral pressure in the D-module increased. A clear density rollover was observed at ∼2Pa. A position of the density maximum moves to upstream of the plasma with increase in the neutral pressure and the density near the corner of the target decreases to detach the plasma from the target. After the occurrence of the density rollover, the Balmer β intensity decreases as with the density but the Balmer α intensity continues to increase, indicating the dissociative attachment process in MAR is more dominant than the ion conversion process although the rate coefficient of the former process is lower than that of the latter one, which is calculated by using a collisional radiative model. This would be caused by the MAR process related to triatomic hydrogen molecules which significantly contributed to the detachment process.

A simple method to evaluate the thickness distribution of the deposition layer formed on the first wall is proposed using an innovative measurement concept of the optical reflection coefficient, which is measured as the RGB (red, green, blue) value using a compact color analyzer. Analysis of the samples exposed to plasmas during an experimental campaign shows the relationship between the thickness of the deposition layer and the reflection coefficient, which is followed by the single layer model. The reflection coefficient clearly indicates the thickness of the deposition layer between 10 and 100nm. The reflection coefficients of stainless steel plates on the helically twisted coil in one of the 10 toroidal sections of the vacuum vessel in the Large Helical Device (LHD) are measured. There is almost no deposition layer on the inner side of the torus, however, the deposition layer reaches a thickness of over 100nm on the first wall near the divertor region. On the outer side of the torus, almost the entire area is covered by the deposition layer. Reflection coefficient measurements indicate that approximately 60% of the area on the measured coil can is coated with a deposition layer over 10nm thick, which suggests that this area plays a role in the wall retention. Abstract The bed bug Cimex lectularius is a blood-feeding re-emerging annoyance pest insect that has the ability to transmit Trypanosoma cruzi under experimental laboratory conditions. Aquaporins (AQPs) are water channel proteins that are essential in biological organisms. C. lectularius are constantly exposed to water-related stress, suggesting that AQPs may offer novel control avenues. We identified and cloned four AQPs from C. lectularius, assessed tissue and lifestage-specific expression, and characterized biochemical functions in vitro and in vivo. We identified an efficient water-specific AQP (ClAQP1), two aquaglyceroporins (ClGlp1 and ClGlp2) and a homolog of Drosophila melanogaster big brain (ClBib). ClGlp1 was only functional when co-expressed with the water-specific AQP. Simultaneous RNAi gene silencing of ClAQP1 and ClGlp1 significantly reduced water and urea excretion post blood feeding. The Bib homologue was enriched in embryos, exclusively expressed in ovaries, and when silenced, dramatically increased bug fecundity. Our data demonstrate that AQPs have critical roles in excretion, water homeostasis and reproduction in C. lectularius, and could be potential targets for control in this notorious pest.

A frequency multiplied microwave interferometer, a Hα line emission measurement system, and a high speed camera system were installed on the Pilot-PSI device for low frequency fluctuation study in the detached plasma condition. The two dimensional Hα line emission and its fluctuation were monitored with a fast visible camera with Hα filter. The coherent low frequency fluctuations of frequency of approximately 13 kHz were measured by all measurement systems. The stronger fluctuation intensities were observed in the downstream of the ionization front region in the detached plasma condition. Moreover, we show the clear difference between the strong fluctuation regions of the electron line density and Hα line emission for the first time. This means that the fluctuations of Hα line emissions was caused by not only electrons but also by hydrogen ions.

Reaction processes of molecular activated recombination (MAR) have been investigated using end loss plasma in GAMMA 10/PDX. A tungsten V-shaped target in a divertor simulation experimental module (D-module) is exposed to the end loss plasma. The additional hydrogen gas is supplied into the D-module, leading to the plasma detachment by MAR. The vibrational temperature (Tvib) of H2(X1Σg+) increased up to ∼10,000 K with increasing the hydrogen gas pressure, and the Ha line intensity (IHa) continued to increase even though the electron density decreased after the electron density rollover. The increase in Tvib should be caused mainly by the increase in the number of vibrationally excited molecules which are produced by the molecular ion conversion process in MAR. The continuous increase in IHa is attributed to the dissociative attachment in MAR. When the hydrogen gas pressure was more than 12 Pa, Balmer emissions from highly excited atom suddenly increased and a population inversion with nH(n = 5) larger than nH (n = 4) was observed although Tvib became rather low (∼2,000 K). The formation of population inversion and increase in highly excited Balmer line intensity are considered to be caused by the reaction of mutual neutralization between molecular ion and negative ion, since the cross section of H(n = 5) production is the largest in the reaction. The negative ion should be produced by the resonant ionization process between H(2s), since the negative ion production by the dissociative attachment reaction cannot be expected due to low Tvib. From the last decade, the multi-scale image segmentation is getting a particular interest and practically being used for object-based image analysis. In this study, we have addressed the issues on multi-scale image segmentation, especially, in improving the performances for validity of merging and variety of derived region’s shape. Firstly, we have introduced constraints on the application of spectral criterion which could suppress excessive merging between dissimilar regions. Secondly, we have extended the evaluation for smoothness criterion by modifying the definition on the extent of the object, which was brought for controlling the shape’s diversity. Thirdly, we have developed new shape criterion called aspect ratio. This criterion helps to improve the reproducibility on the shape of object to be matched to the actual objectives of interest. This criterion provides constraint on the aspect ratio in the bounding box of object by keeping properties controlled with conventional shape criteria. These improvements and extensions lead to more accurate, flexible, and diverse segmentation results according to the shape characteristics of the target of interest. Furthermore, we also investigated a technique for quantitative and automatic parameterization in multi-scale image segmentation. This approach is achieved by comparing segmentation result with training area specified in advance by considering the maximization of the average area in derived objects or satisfying the evaluation index called F-measure. Thus, it has been possible to automate the parameterization that suited the objectives especially in the view point of shape’s reproducibility.
