Abstract: Abstract Liquid xenon time projection chambers are promising detectors to search for neutrinoless double beta decay (0 $$\nu \beta \beta $$ ν β β ), due to their response uniformity, monolithic sensitive volume, scalability to large target masses, and suitability for extremely low background operations. The nEXO collaboration has designed a tonne-scale time projection chamber that aims to search for 0 $$\nu \beta \beta $$ ν β β of $$^{136}$$ 136 Xe with projected half-life sensitivity of $$1.35\times 10^{28}$$ 1.35 × 10 28 yr. To reach this sensitivity, the design goal for nEXO is $$\le $$ ≤ 1% energy resolution at the decay Q-value ( $$2458.07\pm 0.31$$ 2458.07 ± 0.31 keV). Reaching this resolution requires the efficient collection of both the ionization and scintillation produced in the detector. The nEXO design employs Silicon Photo-Multipliers (SiPMs) to detect the vacuum ultra-violet, 175 nm scintillation light of liquid xenon. This paper reports on the characterization of the newest vacuum ultra-violet sensitive Fondazione Bruno Kessler VUVHD3 SiPMs specifically designed for nEXO, as well as new measurements on new test samples of previously characterised Hamamatsu VUV4 Multi Pixel Photon Counters (MPPCs). Various SiPM and MPPC parameters, such as dark noise, gain, direct crosstalk, correlated avalanches and photon detection efficiency were measured as a function of the applied over voltage and wavelength at liquid xenon temperature (163 K). The results from this study are used to provide updated estimates of the achievable energy resolution at the decay Q-value for the nEXO design.

Abstract: The Neutron Induced Fission Fragment Tracking Experiment (NIFFTE) is a double-sided Time Projection Chamber (TPC) with micromegas readout designed to measure the energy-dependent neutron-induced fission cross sections of the major and minor actinides with unprecedented accuracy. The NIFFTE project addresses the challenge of minimizing major sources of systematic uncertainties from previous fission chamber measurements such as: target and beam non-uniformities, misidentification of alpha and light charged particles as fission fragments, and uncertainties inherent to the reference standards used. In-beam tests of the NIFFTE TPC at the Los Alamos Neutron Science Center (LANSCE) started in 2010 and have continued in 2011, 2012 and 2013. Abstract: Large detectors employing xenon are a leading technology in existing and planned searches for new physics, including searches for neutrinoless double beta decay ($0\nu\beta\beta$) and dark matter. While upcoming detectors will employ target masses of a ton or more, further extending gas or liquid phase Xe detectors to the kton scale would enable extremely sensitive next-generation searches for rare phenomena. The key challenge to extending this technology to detectors well beyond the ton scale is the acquisition of the Xe itself. We describe the motivation for extending Xe time projection chambers (TPCs) to the kton scale and possible avenues for Xe acquisition that avoid existing supply chains. If acquisition of Xe in the required quantities is successful, kton-scale detectors of this type could enable a new generation of experiments, including searches for $0\nu\beta\beta$ at half-life sensitivities as long as $10^{30}$ yr., Comment: 19 pages, 10 figures; published version

Double beta detectors are large and expensive, so background reduction is essential for extracting the highest sensitivity. The identification, or 'tagging', of the $^{136}$Ba daughter atom from double beta decay of $^{136}$Xe provides a technique for eliminating backgrounds in the nEXO neutrinoless double beta decay experiment. The tagging scheme studied in this work utilizes a cryogenic probe to trap the barium atom in solid xenon, where the barium atom is tagged via fluorescence imaging in the solid xenon matrix. Here we demonstrate imaging and counting of individual atoms of barium in solid xenon by scanning a focused laser across a solid xenon matrix deposited on a sapphire window. When the laser sits on an individual atom, the fluorescence persists for $\sim$30~s before dropping abruptly to the background level, a clear confirmation of one-atom imaging. No barium fluorescence persists following evaporation of a barium deposit to a limit of $\leq$0.16\%. This is the first time that single atoms have been imaged in solid noble element. It establishes the basic principle of a barium tagging technique for nEXO.

Abstract: A high quality genome annotation greatly facilitates successful cell line engineering. Standard draft genome annotation pipelines are based largely onde novogene prediction, homology, and RNA-Seq data. However, draft annotations can suffer from incorrectly predictions of translated sequence, incorrect splice isoforms and missing genes. Here we generated a draft annotation for the newly assembled Chinese hamster genome and used RNA-Seq, proteomics, and Ribo-Seq to experimentally annotate the genome. We identified 4,333 new proteins compared to the hamster RefSeq protein annotation and 2,503 novel translational events (e.g., alternative splices, mutations, novel splices). Finally, we used this pipeline to identify the source of translated retroviruses contaminating recombinant products from Chinese hamster ovary (CHO) cell lines, including 131 type-C retroviruses, thus enabling future efforts to eliminate retroviruses by reducing the costs incurred with retroviral particle clearance. In summary, the improved annotation provides a more accurate platform for guiding CHO cell line engineering, including facilitating the interpretation of omics data, defining of cellular pathways, and engineering of complex phenotypes.

Abstract: Silicon photomultipliers are regarded as a very promising technology for next-generation, cutting-edge detectors for low-background experiments in particle physics. This work presents systematic reflectivity studies of Silicon Photomultipliers (SiPM) and other samples in liquid xenon at vacuum ultraviolet (VUV) wavelengths. A dedicated setup at the University of M\"unster has been used that allows to acquire angle-resolved reflection measurements of various samples immersed in liquid xenon with 0.45{\deg} angular resolution. Four samples are investigated in this work: one Hamamatsu VUV4 SiPM, one FBK VUV-HD SiPM, one FBK wafer sample and one Large-Area Avalanche Photodiode (LA-APD) from EXO-200. The reflectivity is determined to be 25-36% at an angle of incidence of 20{\deg} for the four samples and increases to up to 65% at 70{\deg} for the LA-APD and the FBK samples. The Hamamatsu VUV4 SiPM shows a decline with increasing angle of incidence. The reflectivity results will be incorporated in upcoming light response simulations of the nEXO detector., Comment: 18 pages, 11 figures

Abstract: The nEXO neutrinoless double beta decay experiment is designed to use a time projection chamber and 5000 kg of isotopically enriched liquid xenon to search for the decay in $^{136}$Xe. Progress in the detector design, paired with higher fidelity in its simulation and an advanced data analysis, based on the one used for the final results of EXO-200, produce a sensitivity prediction that exceeds the half-life of $10^{28}$ years. Specifically, improvements have been made in the understanding of production of scintillation photons and charge as well as of their transport and reconstruction in the detector. The more detailed knowledge of the detector construction has been paired with more assays for trace radioactivity in different materials. In particular, the use of custom electroformed copper is now incorporated in the design, leading to a substantial reduction in backgrounds from the intrinsic radioactivity of detector materials. Furthermore, a number of assumptions from previous sensitivity projections have gained further support from interim work validating the nEXO experiment concept. Together these improvements and updates suggest that the nEXO experiment will reach a half-life sensitivity of $1.35\times 10^{28}$ yr at 90% confidence level in 10 years of data taking, covering the parameter space associated with the inverted neutrino mass ordering, along with a significant portion of the parameter space for the normal ordering scenario, for almost all nuclear matrix elements. Abstract: The Panoramic SETI (Search for Extraterrestrial Intelligence) experiment (PANOSETI) aims to detect and quantify optical transients from nanosecond to second precision over a large field-of-view ($\sim$4,450 square-degrees). To meet these challenging timing and wide-field requirements, the PANOSETI experiment will use two assemblies of $\sim$45 telescopes to reject spurious signals by coincidence detection, each one comprising custom-made fast photon-counting hardware combined with ($f/1.32$) focusing optics. Preliminary on-sky results from pairs of PANOSETI prototype telescopes (100 sq.deg.) are presented in terms of instrument performance and false alarm rates. We found that a separation of $>$1 km between telescopes surveying the same field-of-view significantly reduces the number of false positives due to nearby sources (e.g., Cherenkov showers) in comparison to a side-by-side configuration of telescopes. Abstract: We present direct photon-hadron correlations in 200 GeV/A Au+Au, d+Au, and p+p collisions, for direct photon pT from 5–12 GeV/c, collected by the PHENIX Collaboration in the years from 2006 to 2011. We observe no significant modification of jet fragmentation in d+Au collisions, indicating that cold nuclear matter effects are small or absent. Hadrons carrying a large fraction of the quark's momentum are suppressed in Au+Au compared to p+p and d+Au. As the momentum fraction decreases, the yield of hadrons in Au+Au increases to an excess over the yield in p+p collisions. The excess is at large angles and at low hadron pT and is most pronounced for hadrons associated with lower momentum direct photons. Comparison to theoretical calculations suggests that the hadron excess arises from medium response to energy deposited by jets.

Abstract: The Neutron Induced Fission Fragment Tracking Experiment (NIFFTE) collaboration has performed measurements with a fission time projection chamber (fissionTPC) to study the fission process by reconstructing full three-dimensional tracks of fission fragments and other ionizing radiation. The amount of linear momentum imparted to the fissioning nucleus by the incident neutron can be inferred by measuring the opening angle between the fission fragments. Using this measured linear momentum, fission fragment angular distributions can be converted to the center-of-mass frame for anisotropy measurements. Angular anisotropy is an important experimental observable for understanding the quantum mechanical state of the fissioning nucleus and vital to determining detection efficiency for cross section measurements. Neutron linear momentum transfer to fissioning $^{235}$U, $^{238}$U, and $^{239}$Pu and fission fragment angular anisotropy of $^{235}$U and $^{238}$U as a function of neutron energies in the range 130 keV--250 MeV are presented.

Abstract: nEXO is a proposed tonne-scale neutrinoless double beta decay ($0\nu\beta\beta$) experiment using liquid ${}^{136}Xe$ (LXe) in a Time Projection Chamber (TPC) to read out ionization and scintillation signals. Between the field cage and the LXe vessel, a layer of LXe ("skin" LXe) is present, where no ionization signal is collected. Only scintillation photons are detected, owing to the lack of optical barrier around the field cage. In this work, we show that the light originating in the skin LXe region can be used to improve background discrimination by 5% over previous published estimates. This improvement comes from two elements. First, a fraction of the $\gamma$-ray background is removed by identifying light from interactions with an energy deposition in the skin LXe. Second, background from ${}^{222}Rn$ dissolved in the skin LXe can be efficiently rejected by tagging the $\alpha$ decay in the ${}^{214}Bi-{}^{214}Po$ chain in the skin LXe., Comment: 11 pages, 12 figures

Abstract: We present the results of a measurement of isotopic concentrations and atomic number ratio of a double-sided actinide target using α -spectroscopy and mass spectrometry. The double-sided actinide target, with predominantly 239Pu on one side and 235U on the other, was used in the fission Time Projection Chamber (fissionTPC) for a measurement of the neutron-induced fission cross-section ratio between the two isotopes. The measured atomic number ratio is needed to extract an absolute measurement fission cross-section ratio. The 239Pu/235U atom number ratio was measured with a combination of mass spectrometry and α -spectroscopy with a planar silicon detector achieving uncertainties of less than 1%. Different strategies for estimating isotopic concentration from the α -spectrum are presented to demonstrate the potential of these methods for non-destructive target assay. We found that a combination of fitting spectra with constraints from mass spectrometry, and summing counts in a region of the spectrum provided the most consistent results with the lowest uncertainty.

Abstract: Accurate and complete genome sequences are essential in biotechnology to facilitate genome‐based cell engineering efforts. The current genome assemblies for Cricetulus griseus, the Chinese hamster, are fragmented and replete with gap sequences and misassemblies, consistent with most short‐read‐based assemblies. Here, we completely resequenced C. griseus using single molecule real time sequencing and merged this with Illumina‐based assemblies. This generated a more contiguous and complete genome assembly than either technology alone, reducing the number of scaffolds by >28‐fold, with 90% of the sequence in the 122 longest scaffolds. Most genes are now found in single scaffolds, including up‐ and downstream regulatory elements, enabling improved study of noncoding regions. With >95% of the gap sequence filled, important Chinese hamster ovary cell mutations have been detected in draft assembly gaps. This new assembly will be an invaluable resource for continued basic and pharmaceutical research.

Abstract: Coagulation factor II (prothrombin; FII) is the pre-proteolyzed precursor to thrombin in the coagulation cascade. It has 10 sites of gamma-carboxylation, which are required for its bioactivity, and is N-glycosylated at three of four putative sites. Production of recombinant human FII (rhFII) using a platform fed-batch process designed for monoclonal antibody production resulted in low levels of gamma-carboxylation and sialylation. There have not been any prior reports of successful process development and clinical manufacture of rhFII with optimal, consistent gamma-carboxylation and sialylation. In order to develop such a fed-batch process, various process parameters were evaluated to determine their impact on product quality. Process temperature and temperature shift timing were important for both sialic acid level and gamma-carboxyglutamate (Gla) level. In addition, vitamin K concentration and the type of surfactant used for preparation of vitamin K stock solution were also important for gamma carboxylation. A fed-batch study performed with various medium additives known to be involved in the N-glycosylation pathway, such as N-acetyl-d-mannosamine (ManNAc), galactose (Gal), dexamethasone, and manganese sulfate, increased the level of sialylation and enabled the elucidation of some potential bottlenecks in the sialylation pathway. The optimized process based on these studies yielded a reduction in the level of missing Gla by 0.4 moles per mole of rhFII in cell culture and a nearly threefold increase in sialic acid level. The process was successfully implemented at the 2000 L scale where a high Gla level and sialylation levels were achieved in all GMP lots. Biotechnol. Abstract: A combined lipidomics and transcriptomics analysis was performed on mouse myeloma SP2/0, Chinese hamster ovary (CHO), and human embryonic kidney (HEK) cells in order to compare widely used mammalian expression systems. Initial thin layer chromatography (TLC) analysis indicated that phosphatidylethanolamine (PE) and phosphatidylcholine (PC) were the major lipid components in all cell lines with lower amounts of sphingomyelin (SM) in SP2/0 compared to CHO and HEK, which was subsequently confirmed and expanded upon following mass spectrometry (MS) analysis. HEK contained 4-10-fold higher amounts of lyso phosphatidylethanolamine (LPE) and 2-4-fold higher amounts of lyso phosphatidylcholine (LPC) compared to SP2/0 and CHO cell lines. C18:1 followed by C16:1 were the main contributors to the difference in both LPE and LPC levels. Alternatively, the SP2/0 cell line exhibited 30-65-fold lower amounts of SM principally in the amount of 16:0. By mapping the transcriptomics data to KEGG pathways, we found expression levels of secretory phospholipase A2 (sPLA2), lysophospholipid acyltransferase (LPEAT), lysophosphatidylcholine acyltransferase (LPCAT), and lysophospholipase (LYPLA) can contribute to the differences in LPE and LPC. Sphingomyelin synthases (SMS) and sphingomyelin phosphodiesterase (SMase) enzymes may play roles in SM differences across the three cell lines. The results of this study provide insights that will aid the understanding of the physiological and secretory differences across recombinant protein production systems.

Abstract: Measurements of electron drift properties in liquid and gaseous xenon are reported. The electrons are generated by the photoelectric effect in a semi-transparent gold photocathode driven in transmission mode with a pulsed ultraviolet laser. The charges drift and diffuse in a small chamber at various electric fields and a fixed drift distance of 2.0 cm. At an electric field of 0.5 kV/cm, the measured drift velocities and corresponding temperature coefficients respectively are 1 . 97 ± 0 . 04 m m ∕ μ s and ( − 0 . 69 ± 0 . 05 ) %/K for liquid xenon, and 1 . 42 ± 0 . 03 m m ∕ μ s and ( + 0 . 11 ± 0 . 01 ) %/K for gaseous xenon at 1.5 bar. In addition, we measure longitudinal diffusion coefficients of 25 . 7 ± 4 . 6 cm 2 /s and 149 ± 23 cm 2 /s, for liquid and gas, respectively. The quantum efficiency of the gold photocathode is studied at the photon energy of 4.73 eV in liquid and gaseous xenon, and vacuum. These charge transport properties and the behavior of photocathodes in a xenon environment are important in designing and calibrating future large scale noble liquid detectors.

Abstract: The Central Atlantic Magmatic Province (CAMP) is the most aerially extensive magmatic event in Earth’s history, but many questions remain about its origin, volume, and distribution. Despite many observations of CAMP magmatism near Earth’s surface, few constraints exist on CAMP intrusions at depth. Here we present detailed constraints on crustal and upper mantle structure from wide-angle seismic data across the Triassic South Georgia Rift that formed shortly before CAMP. Lower crustal magmatism is concentrated where synrift sedimentary fill is thickest and the crust is thinnest, suggesting that lithospheric thinning influenced the locus and volume of magmatism. The limited distribution of lower crustal intrusions implies modest total CAMP volumes of 85,000 to 169,000 km3 beneath the South Georgia Rift, consistent with moderately elevated mantle potential temperatures (, The Central Atlantic Magmatic Province is the most aerially extensive magmatic event in Earth’s history, yet few constraints exist on the volumes of intrusions at depth. Abstract: The PHENIX collaboration presents first measurements of low-momentum ($0.4$1\,GeV/$c$) direct-photon yield $dN_{\gamma}^{\rm dir}/d\eta$ is a smooth function of $dN_{\rm ch}/d\eta$ and can be well described as proportional to $(dN_{\rm ch}/d\eta)^\alpha$ with $\alpha{\approx}1.25$. This scaling behavior holds for a wide range of beam energies at the Relativistic Heavy Ion Collider and the Large Hadron Collider, for centrality selected samples, as well as for different, $A$$+$$A$ collision systems. At a given beam energy the scaling also holds for high $p_T$ ($>5$\,GeV/$c$) but when results from different collision energies are compared, an additional $\sqrt{s_{_{NN}}}$-dependent multiplicative factor is needed to describe the integrated-direct-photon yield., Comment: 673 authors from 82 institutions, 10 pages, 4 figures. v2 is version accepted for publication in Physical Review Letters. Abstract: Fission fragment angular distributions can provide an important constraint on fission theory, improving predictive fission codes, and are a prerequisite for a precise ratio cross section measurement. Available anisotropy data is sparse, especially at neutron energies above 5 MeV. For the first time, a three-dimensional tracking detector is employed to study fragment emission angles and provide a direct measurement of angular anisotropy. The Neutron Induced Fission Fragment Tracking Experiment (NIFFTE) collaboration has deployed the fission time projection chamber (fissionTPC) to measure nuclear data with unprecedented precision. The fission fragment anisotropy of $^{235}$U has been measured over a wide range of incident neutron energies from 180 keV to 200 MeV; a careful study of the systematic uncertainties complement the data.

Abstract: Chinese hamster ovary (CHO) cells are the predominant production vehicle for biotherapeutics. Quantitative proteomics data were obtained from two CHO cell lines (CHO-S and CHO DG44) and compared with seven Chinese hamster (Cricetulus griseus) tissues (brain, heart, kidney, liver, lung, ovary and spleen) by tandem mass tag (TMT) labeling followed by mass spectrometry, providing a comprehensive hamster tissue and cell line proteomics atlas. Of the 8470 unique proteins identified, high similarity was observed between CHO-S and CHO DG44 and included increases in proteins involved in DNA replication, cell cycle, RNA processing, and chromosome processing. Alternatively, gene ontology and pathway analysis in tissues indicated increased protein intensities related to important tissue functionalities. Proteins enriched in the brain included those involved in acidic amino acid metabolism, Golgi apparatus, and ion and phospholipid transport. The lung showed enrichment in proteins involved in BCAA catabolism, ROS metabolism, vesicle trafficking, and lipid synthesis while the ovary exhibited enrichments in extracellular matrix and adhesion proteins. The heart proteome included vasoconstriction, complement activation, and lipoprotein metabolism enrichments. These detailed comparisons of CHO cell lines and hamster tissues will enhance understanding of the relationship between proteins and tissue function and pinpoint potential pathways of biotechnological relevance for future cell engineering.

Abstract: In this paper we report on the characterization of the Hamamatsu VUV4 (S/N: S13370-6152) Vacuum Ultra-Violet (VUV) sensitive Multi-Pixel Photon Counters (MPPC)s as part of the development of a solution for the detection of liquid xenon scintillation light for the nEXO experiment. Various MPPC features, such as: dark noise, gain, correlated avalanches, direct crosstalk and Photon Detection Efficiency (PDE) were measured in a dedicated setup at TRIUMF. MPPCs were characterized in the range 163 K ≤ T ≤ 233 K . At an over voltage of 3 . 1 ± 0 . 2 V and at T = 163 K we report a number of Correlated Avalanches (CAs) per pulse in the 1 μ s interval following the trigger pulse of 0 . 161 ± 0 . 005 . At the same settings the Dark-Noise (DN) rate is 0 . 137 ± 0 . 002 Hz/mm 2 . Both the number of CAs and the DN rate are within nEXO specifications. The PDE of the Hamamatsu VUV4 was measured for two different devices at T = 233 K for a mean wavelength of 189 ± 7 nm . At 3 . 6 ± 0 . 2 V and 3 . 5 ± 0 . 2 V of over voltage we report a PDE of 13 . 4 ± 2 . 6 % and 11 ± 2 % , corresponding to a saturation PDE of 14 . 8 ± 2 . 8 % and 12 . 2 ± 2 . 3 % , respectively. Both values are well below the 24 % saturation PDE advertised by Hamamatsu. More generally, the second device tested at 3 . 5 ± 0 . 2 V of over voltage is below the nEXO PDE requirement. The first one instead yields a PDE that is marginally close to meeting the nEXO specifications. This suggests that with modest improvements the Hamamatsu VUV4 MPPCs could be considered as an alternative to the FBK-LF Silicon Photo-Multipliers for the final design of the nEXO detector.

Abstract: Characterization of the vacuum ultraviolet (VUV) reflectance of silicon photomultipliers (SiPMs) is important for large-scale SiPM-based photodetector systems. We report the angular dependence of the specular reflectance in a vacuum of SiPMs manufactured by Fondazionc Bruno Kessler (FBK) and Hamamatsu Photonics K.K. (HPK) over wavelengths ranging from 120 nm to 280 nm. Refractive index and extinction coefficient of the thin silicon-dioxide film deposited on the surface of the FBK SiPMs are derived from reflectance data of a FBK silicon wafer with the same deposited oxide film as SiPMs. The diffuse reflectance of SiPMs is also measured at 193 nm. We use the VUV spectral dependence of the optical constants to predict the reflectance of the FBK silicon wafer and FBK SiPMs in liquid xenon.

Abstract: nEXO is a proposed experiment to search for the neutrino-less double beta decay ($0\nu\beta\beta$) of $^{136}$Xe in a tonne-scale liquid xenon time projection chamber (TPC). The nEXO TPC will be equipped with charge collection tiles to form the anode. In this work, the charge reconstruction performance of this anode design is studied with a dedicated simulation package. A multi-variate method and a deep neural network are developed to distinguish simulated $0\nu\beta\beta$ signals from backgrounds arising from trace levels of natural radioactivity in the detector materials. These simulations indicate that the nEXO TPC with charge-collection tiles shows promising capability to discriminate the $0\nu\beta\beta$ signal from backgrounds. The estimated half-life sensitivity for $0\nu\beta\beta$ decay is improved by $\sim$20$~(32)\%$ with the multi-variate~(deep neural network) methods considered here, relative to the sensitivity estimated in the nEXO pre-conceptual design report.

Abstract: Understanding reflective properties of materials and photodetection efficiency (PDE) of photodetectors is important for optimizing energy resolution and sensitivity of the next generation neutrinoless double beta decay, direct detection dark matter, and neutrino oscillation experiments that will use noble liquid gases, such as nEXO, DARWIN, DarkSide-20k, and DUNE. Little information is currently available about reflectivity and PDE in liquid noble gases, because such measurements are difficult to conduct in a cryogenic environment and at short enough wavelengths. Here we report a measurement of specular reflectivity and relative PDE of Hamamatsu VUV4 silicon photomultipliers (SiPMs) with 50 micrometer micro-cells conducted with xenon scintillation light (~175 nm) in liquid xenon. The specular reflectivity at 15 deg. incidence of three samples of VUV4 SiPMs is found to be 30.4+/-1.4%, 28.6+/-1.3%, and 28.0+/-1.3%, respectively. The PDE at normal incidence differs by +/-8% (standard deviation) among the three devices. The angular dependence of the reflectivity and PDE was also measured for one of the SiPMs. Both the reflectivity and PDE decrease as the angle of incidence increases. Abstract: The normalized $^{238}$U(n,f)/$^{235}$U(n,f) cross section ratio has been measured using the NIFFTE fission Time Projection Chamber from the reaction threshold to $30$~MeV. The fissionTPC is a two-volume MICROMEGAS time projection chamber that allows for full three-dimensional reconstruction of fission-fragment ionization profiles from neutron-induced fission. The measurement was performed at the Los Alamos Neutron Science Center, where the neutron energy is determined from neutron time-of-flight. The $^{238}$U(n,f)/$^{235}$U(n,f) ratio reported here is the first cross section measurement made with the fissionTPC, and will provide new experimental data for evaluation of the $^{238}$U(n,f) cross section, an important standard used in neutron-flux measurements. Use of a development target in this work prevented the determination of an absolute normalization, to be addressed in future measurements. Instead, the measured cross section ratio has been normalized to ENDF/B-VIII.$\beta$5 at 14.5 MeV.

Abstract: Mammalian expression systems such as Chinese hamster ovary (CHO), mouse myeloma (NS0), and human embryonic kidney (HEK) cells serve a critical role in the biotechnology industry as the production host of choice for recombinant protein therapeutics. Most of the recombinant biologics are glycoproteins that contain complex oligosaccharide or glycan attachments representing a principal component of product quality. Both N-glycans and O-glycans are present in these mammalian cells, but the engineering of N-linked glycosylation is of critical interest in industry and many efforts have been directed to improve this pathway. This is because altering the N-glycan composition can change the product quality of recombinant biotherapeutics in mammalian hosts. In addition, sialylation and fucosylation represent components of the glycosylation pathway that affect circulatory half-life and antibody-dependent cellular cytotoxicity, respectively. In this chapter, we first offer an overview of the glycosylation, sialylation, and fucosylation networks in mammalian cells, specifically CHO cells, which are extensively used in antibody production. Next, genetic engineering technologies used in CHO cells to modulate glycosylation pathways are described. We provide examples of their use in CHO cell engineering approaches to highlight these technologies further. Specifically, we describe efforts to overexpress glycosyltransferases and sialyltransfereases, and efforts to decrease sialidase cleavage and fucosylation. Finally, this chapter covers new strategies and future directions of CHO cell glycoengineering, such as the application of glycoproteomics, glycomics, and the integration of 'omics' approaches to identify, quantify, and characterize the glycosylated proteins in CHO cells. Abstract: Future tonne-scale liquefied noble gas detectors depend on efficient light detection in the VUV range. In the past years Silicon Photomultipliers (SiPMs) have emerged as a valid alternative to standard photomultiplier tubes or large area avalanche photodiodes. The next generation double beta decay experiment, nEXO, with a 5 tonne liquid xenon time projection chamber, will use SiPMs for detecting the $178\,\text{nm}$ xenon scintillation light, in order to achieve an energy resolution of $\sigma / Q_{\beta\beta} = 1\, \%$. This paper presents recent measurements of the VUV-HD generation SiPMs from Fondazione Bruno Kessler in two complementary setups. It includes measurements of the photon detection efficiency with gaseous xenon scintillation light in a vacuum setup and dark measurements in a dry nitrogen gas setup. We report improved photon detection efficiency at $175\,\text{nm}$ compared to previous generation devices, that would meet the criteria of nEXO. Abstract: Chinese hamster ovary (CHO) cells are the preferred host cell line for manufacturing a variety of complex biotherapeutic drugs including monoclonal antibodies. We performed a proteomics and bioinformatics analysis on the spent medium from adherent CHO cells. Supernatant from CHO-K1 culture was collected and subjected to in-solution digestion followed by LC/LC-MS/MS analysis, which allowed the identification of 3281 different host cell proteins (HCPs). To functionally categorize them, we applied multiple bioinformatics tools to the proteins identified in our study including SignalP, TargetP, SecretomeP, TMHMM, WoLF PSORT, and Phobius. This analysis provided information on the presence of signal peptides, transmembrane domains, and cellular localization and showed that both secreted and intracellular proteins were constituents of the supernatant. Identified proteins were shown to be localized to the secretory pathway including ones playing roles in cell growth, proliferation, and folding as well as those involved in protein degradation and removal. After combining proteins predicted to be secreted or having a signal peptide, we identified 1015 proteins, which we termed as CHO supernatant-ome (CHO-SO), or superome. As a part of this effort, we created a publically accessible web-based tool called GO-CHO to functionally categorize proteins found in CHO-SO and identify enriched molecular functions, biological processes, and cellular components. We also used a tool to evaluate the immunogenicity potential of high-abundance HCPs. Among enriched functions were catalytic activity and structural constituents of the cytoskeleton. Various transport related biological processes, such as vesicle mediated transport, were found to be highly enriched. Extracellular space and vesicular exosome associated proteins were found to be the most enriched cellular components. The superome also contained proteins secreted from both classical and nonclassical secretory pathways. The work and database described in our study will enable the CHO community to rapidly identify high-abundance HCPs in their cultures and therefore help assess process and purification methods used in the production of biologic drugs.

Abstract: An RF ion-funnel technique has been developed to extract ions from a high-pressure (10 bar) noble-gas environment into a vacuum (10(-6) mbar). Detailed simulations have been performed and a prototype has been developed for the purpose of extracting Ba-136 ions from Xe gas with high efficiency. With this prototype, ions have been extracted for the first time from high-pressure xenon gas and argon gas. Systematic studies have been carried out and compared to simulations. This demonstration of extraction of ions, with mass comparable to that of the gas generating the high-pressure, has applications to Ba tagging from a Xe-gas time-projection chamber for double-beta decay, as well as to the general problem of recovering trace amounts of an ionized element in a heavy (m > 40 u) carrier gas. Abstract: Recent advancements in proteomics have enabled the generation of high-quality data sets useful for applications ranging from target and monoclonal antibody (mAB) discovery to bioprocess optimization. Comparative proteomics approaches have recently been used to identify novel disease targets in oncology and other disease conditions. Proteomics has also been applied as a new avenue for mAb discovery. Finally, CHO and Escherichia coli cells represent the dominant production hosts for biopharmaceutical development, yet the physiology of these cells types has yet to be fully established. Proteomics approaches can provide new insights into these cell types, aiding in recombinant protein production, cell growth regulation, and medium formulation. Optimization of sample preparations and protein database developments are enhancing the quantity and accuracy of proteomic results. In these ways, innovations in proteomics are enriching biotechnology and bioprocessing research across a wide spectrum of applications.

Abstract: The next-generation Enriched Xenon Observatory (nEXO) is a proposed experiment to search for neutrinoless double beta ($0\nu\beta\beta$) decay in $^{136}$Xe with a target half-life sensitivity of approximately $10^{28}$ years using $5\times10^3$ kg of isotopically enriched liquid-xenon in a time projection chamber. This improvement of two orders of magnitude in sensitivity over current limits is obtained by a significant increase of the $^{136}$Xe mass, the monolithic and homogeneous configuration of the active medium, and the multi-parameter measurements of the interactions enabled by the time projection chamber. Abstract: Chinese hamster ovary cells represent the dominant host for therapeutic recombinant protein production. However, few large-scale data sets have been generated to characterize this host organism and derived CHO cell lines at the proteomics level. Consequently, an extensive label-free quantitative proteomics analysis of two cell lines (CHO-S and CHO DG44) and two Chinese hamster tissues (liver and ovary) was used to identify a total of 11 801 unique proteins containing at least two unique peptides. 9359 unique proteins were identified specifically in the cell lines, representing a 56% increase over previous work. Additionally, 6663 unique proteins were identified across liver and ovary tissues, providing the first Chinese hamster tissue proteome. Protein expression was more conserved within cell lines during both growth phases than across cell lines, suggesting large genetic differences across cell lines. Overall, both gene ontology and KEGG pathway analysis revealed enrichment of cell-cycle activity in cells. In contrast, upregulated molecular functions in tissue include glycosylation and lipid transporter activity. Furthermore, cellular components including Golgi apparatus are upregulated in both tissues. In conclusion, this large-scale proteomics analysis enables us to delineate specific changes between tissues and cells derived from these tissues, which can help explain specific tissue function and the adaptations cells incur for applications in biopharmaceutical productions.

Abstract: We report a measurement of e+e− pairs from semileptonic heavy-flavor decays in p+p collisions at sNN=200 GeV. The e+e− pair yield from bb¯ and cc¯ is separated by exploiting a double differential fit done simultaneously in dielectron invariant mass and pT. We used three different event generators, pythia, mc@nlo, and powheg, to simulate the e+e− spectra from cc¯ and bb¯ production. The data can be well described by all three generators within the detector acceptance. However, when using the generators to extrapolate to 4π, significant differences are observed for the total cross section. These difference are less pronounced for bb¯ than for cc¯. The same model dependence was observed in already published d+A data. The p+p data are also directly compared with d+A data in mass and pT, and within the statistical accuracy no nuclear modification is seen.

Abstract: Genomic rearrangements are a common phenomenon in rapidly growing cell lines such as Chinese hamster ovary (CHO) cells, a feature that in the context of production of biologics may lead to cell line and product instability. Few methods exist to assess such genome wide instability. Here, we use the population distribution of chromosome numbers per cell as well as chromosome painting to quantify the karyotypic variation in several CHO host cell lines. CHO-S, CHO-K1 8 mM glutamine, and CHO-K1 cells adapted to grow in media containing no glutamine were analyzed over up to 6 months in culture. All three cell lines were clearly distinguishable by their chromosome number distribution and by the specific chromosome rearrangements that were present in each population. Chromosome Painting revealed a predominant karyotype for each cell line at the start of the experiment, completed by a large number of variants present in each population. Over time in culture, the predominant karyotype changed for CHO-S and CHO-K1, with the diversity increasing and new variants appearing, while CHO-K1 0 mM Gln preferred chromosome pattern increased in percent of the population over time. As control, Chinese hamster lung fibroblasts were shown to also contain an increasing number of variants over time in culture.

Abstract: A new design for the anode of a time projection chamber, consisting of a charge-detecting "tile", is investigated for use in large scale liquid xenon detectors. The tile is produced by depositing 60 orthogonal metal charge-collecting strips, 3~mm wide, on a 10~\si{\cm} $\times$ 10~\si{\cm} fused-silica wafer. These charge tiles may be employed by large detectors, such as the proposed tonne-scale nEXO experiment to search for neutrinoless double-beta decay. Modular by design, an array of tiles can cover a sizable area. The width of each strip is small compared to the size of the tile, so a Frisch grid is not required. A grid-less, tiled anode design is beneficial for an experiment such as nEXO, where a wire tensioning support structure and Frisch grid might contribute radioactive backgrounds and would have to be designed to accommodate cycling to cryogenic temperatures. The segmented anode also reduces some degeneracies in signal reconstruction that arise in large-area crossed-wire time projection chambers. A prototype tile was tested in a cell containing liquid xenon. Very good agreement is achieved between the measured ionization spectrum of a $^{207}$Bi source and simulations that include the microphysics of recombination in xenon and a detailed modeling of the electrostatic field of the detector. An energy resolution $\sigma/E$=5.5\% is observed at 570~\si{keV}, comparable to the best intrinsic ionization-only resolution reported in literature for liquid xenon at 936~V/\si{cm}., Comment: 18 pages, 13 figures, as published

Abstract: Chinese hamster ovary (CHO) cell lines are the preferred host for manufacturing therapeutic proteins. The use of proteomics for recombinant protein production in CHO cells has expanded recently because of the ease in identifying protein levels and its potential to elucidate targets for improved growth and productivity. Coupling advanced analytical methods, labeling strategies and bioinformatics with mass spectrometry-based proteomics facilitates identification and quantification of large numbers of proteins. CHO proteomics studies have increased the knowledge of proteins that affect cell culture events, thereby facilitating media development and cell line engineering to increase growth and production, delay apoptosis and utilize nutrients more effectively. The applications of proteomics offers numerous ways for creating superior hosts and improving biotherapeutics production in CHO cells.

Abstract: The Neutron Induced Fission Fragment Tracking Experiment (NIFFTE) collaboration is developing a Time Projection Chamber (TPC) to measure neutron-induced fission cross sections with unprecedented accuracy. Modifying TPC technology for nuclear physics applications is a challenging endeavor, and frequent testing and evaluation is required to ensure that all components are behaving as expected. In-beam tests of the NIFFTE TPC at the Los Alamos Neutron Science Center (LAN-SCE) commenced in 2010. An overview of the NIFFTE TPC experiments performed at LANSCE will be presented, along with preliminary performance results.

Abstract: Medium to large channel count detectors are usually faced with a few unattractive options for data acquisition (DAQ). Small to medium-sized TPC experiments, for example, are too small to justify the expense and development time of application specific integrated circuits (ASIC). Commercial rack mounted electronics are too bulky and expensive for large channel counts. The combination of commercial high-speed high-density FPGAs, ADCs, and small discrete components provides another option that scales to tens of thousands of channels and is only slightly larger than ASICs using off-the-shelf components. A working example of this alternative solution is presented.

50. New constraints on buried Triassic basins and regional implications for subsurface CO2 storage from the SeisData6 seismic profile across the Southeast Georgia coastal plain

Abstract: Reprocessing of the SeisData6 coastal plain profile was motivated by the need to provide enhanced subsurface imaging critical to site characterization studies for CO2 storage within the South Georgia Rift (SGR) basin. The objectives were to identify and interpret subsurface reflectors for evidence of the buried Triassic basin and its underlying characteristics. Our new interpretation, supported by analysis of well data, has helped substantiate the presence of a Triassic basin beneath the coastal plain sediments in Southeast Georgia. This basin is approximately 2.2 km (1.7 mi) deep and 170 km (106 mi) wide and appears to coincide with the subsurface convergence of the southwest and northeast extensions of the Riddleville and Dunbarton basins that are subsidiaries of the main SGR. It is characterized by distinctively higher seismic velocities relative to the overlying coastal plain sediments and manifests a series of subhorizontal reflectors below the topmost reflector. We reinterpreted the topmost reflector to originate from a change in velocity and density between the Cretaceous coastal plain sediments and the underlying Triassic rocks. This does not always originate from the Pre-Cretaceous basalt contrary to previous interpretations. The interpreted absence of basalt from this study is consistent with Heffner et al. (2012) showing that basalt is not prevalent throughout the SGR basin. Seismic discontinuities in the southeast of the basin suggest Triassic normal faults. Our data show no clear evidence for the Augusta fault that was identified in other studies in the vicinity of the Piedmont–coastal plain boundary in Georgia and South Carolina.
