This study designs a two-stage algorithm to address the bid generation problem of carriers when adding new vehicle routes in the presence of the existing vehicle routes to provide transportation service. To obtain the best auction combination and bid price of the carrier, a hybrid integer nonlinear programming model is introduced. According to the characteristics of the problem, a set of two-stage hybrid algorithms is proposed, innovatively integrating block coding within a genetic algorithm framework with a depth-first search approach. This integration effectively manages routing constraints, enhancing the algorithm's efficiency. The block coding and each route serve as decision variables in the set partition formula, enabling a comprehensive exploration of potential solutions. After a simulation-based analysis, the algorithm has been comprehensively validated analytically and empirically. The improvement of this research lies in the effectiveness of the proposed algorithm, i.e., the ability to handle a broader range of problem scales with less time in addressing complex operator bid generation in combinatorial auctions. Summary: The present study aimed to comparatively analyse the functional properties of the spray drying particles prepared from the noble scallop (Chlamys nobilis) mantle protein‐pectin hydrated mixtures. And the microstructural characteristics of the particles were assessed by Fourier transform infrared, scanning electronic microscopy, and SDS‐PAGE to detect critical changes. The results showed that the solubility of enzymatically extracted protein was significantly improved by spray drying after pectin hydration (AP‐LMP and AP‐HMP), from 76.77% (AP) to 97.87% (AP‐LMP) and 97.13% (AP‐HMP), respectively. And the spray drying water‐extracted protein (HP) after pectin hydration (HP‐LMP and HP ‐HMP) exhibited a greater emulsifying property, from 103.10 m2/g (HP) to 145.55 ± 0.92 m2/g (HP‐LMP) and 200.29 ± 1.16 m2/g (HP‐HMP). The spray drying after pectin hydration had successfully enhanced emulsifying and rheological properties when compared with AP and HP. The results of SDS‐PAGE showed more protein participated in the formation of macromolecule and FT‐IR spectra demonstrated the formation of protein‐pectin spray drying particles facilitated not only by electrostatic interactions but also by covalent binding via Maillard reaction, made a closely compatible protein structure verified by the improvement of fluorescence intensity. Acoustic tweezing cytometry (ATC) is an ultrasound-based biophysical technique that has shown the capability to promote differentiation of human pluripotent stem cells (hPSCs). This study systematically examined how hPSCs respond to cyclic mechanical strains applied by ATC via displacement of integrin-bound microbubbles (averaged diameter of 4.3 µm) using ultrasound pulses (acoustic pressure 0.034 MPa, center frequency 1.24 MHz and pulse repetition frequency 1 Hz). Our data show downregulation of pluripotency marker Octamer-binding transcription factor 4 (OCT4) by at least 10% and increased nuclear localization of Yes-associated protein (YAP) by almost 100% in hPSCs immediately after ATC application for as short as 1 min and 5 min respectively. Analysis of the movements of integrin-anchored microbubbles under ATC stimulations reveals different stages of viscoelastic characteristic behavior and increasing deformation of the integrin-cytoskeleton (CSK) linkage. The peak displacement of integrin-bound microbubbles increased from 1.45 ± 0.16 to 4.74 ± 0.67 μm as the duty cycle of ultrasound pulses increased from 5% to 50% or the duration of each ultrasound pulse increased from 0.05 to 0.5 s. Real-time tracking of integrin-bound microbubbles during ATC application detects high correlation of microbubble displacements with OCT4 downregulation in hPSCs. Together, our data showing fast downregulation of OCT4 in hPSCs in respond to ATC stimulations highlight the unique mechanosensitivity of hPSCs to integrin-targeted cyclic force/strain dependent on the pulse duration or duty cycle of ultrasound pulses, providing insights into the mechanism of ATC-induced accelerated differentiation of hPSCs. People living in plains tend to decrease in body weight or body fat percentage after entering the plateau. Previous studies have found that plateau animals can burn fat and release calories through white adipose tissues (WATs) browning. However, these studies have focused on the effect of cold stimulation that induced WATs browning while there's hardly study on the effect of hypoxia. In this study, we investigate that whether and how hypoxia contributes to WATs browning in rats from acute to chronic hypoxia. We constructed hypobaric hypoxic rat models by exposing 9-week-old male SD rats to a hypobaric hypoxic chamber for 1, 3, 14 and 28 days (Group H) under simulated environment at altitude of 5000 m. We also established normoxic control groups for each time period (Group C), as well as paired 1-day and 14-day normoxic food-restriction rats that were fed the same amount of food as the hypoxic group ate (Group R). We then observed the growth status of rats and recorded dynamic changes in histologic, cellular and molecular levels of perirenal WATs (PWAT), epididymal WATs (EWAT) and subcutaneous WATs (SWAT) in each group. Results showed that (1) Hypoxic rats had lower food intake, significantly lower body weight than control rats, and showed lower WATs index. (2) In group H14, ASC1 mRNA expressions of PWAT and EWAT in rats were lower than that in group C14, and PAT2 mRNA expression of EWAT was higher than that in both group C14 and R14. In group R14, however, ASC1 mRNA expressions of PWAT and EWAT in rats were higher than both group C14 and H14, and that of SWAT was also significantly higher than group C14. (3) In group H3, both the mRNA and protein levels of uncoupling protein 1 (UCP1) of PWAT in rats were significantly increased than group C3. And in group H14, those of EWAT in rats were significantly increased than group C14. (4) In plasma of rats, norepinephrine (NE) level was significantly increased in group H3 than group C3, and free fatty acids (FFAs) level was significantly increased in group H14 than both group C14 and R14. In group R1, FASN mRNA expressions of PWAT and EWAT in rats were down-regulated than group C1. In group H3, FASN mRNA expressions of PWAT and EWAT in rats were down-regulated while ATGL mRNA expression of EWAT was up-regulated than group C3. Conversely, in group R14, FASN mRNA expressions of PWAT and EWAT in rats were significantly up-regulated than group C14 and H14. These results suggested that hypoxia promoted different WATs browning in rats under simulated environment at altitude of 5000 m and changed the lipid metabolism in WATs. Hyperactivation of hypothalamic–pituitary–adrenal (HPA) axis and hypothalamic–pituitary–thyroid (HPT) axis were found in acute high altitude challenge, but the role of gut microbiota and metabolites is unknown. We utilized adult male Sprague–Dawley rats at a simulated altitude of 5500 m for 3 days in a hypobaric-hypoxic chamber. ELISA and metabolomic analyses of serum and 16S rRNA and metabolomic analyses of fecal samples were then performed. Compared with the normoxic group, serum corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), corticosterone (CORT), and thyroxine (tT4) were increased in the hypoxia group, whereas thyrotropin-releasing hormone (TRH) was decreased. Bacteroides, Lactobacillus,Parabacteroides,Butyricimonas,SMB53,Akkermansia,Phascolarctobacterium, and Aerococcus were enriched in hypoxia group, whereas [Prevotella], Prevotella,Kaistobacter,Salinibacterium, and Vogesella were enriched in normoxic group. Metabolomic analysis indicated that acute hypoxia significantly affected fecal and serum lipid metabolism. In addition, we found five fecal metabolites may mediate the cross-talk between TRH, tT4, and CORT with [Prevotella], Kaistobacter,Parabacteroides, and Aerococcus, and 6 serum metabolites may mediate the effect of TRH and tT4 on [Prevotella] and Kaistobacter by causal mediation analysis. In conclusion, this study provides new evidence that key metabolites mediate the cross-talk between gut microbiota with HPA and HPT axis under acute hypobaric hypoxia challenge. Film thickness is very important in the preparation of film thermoelectric (TE) devices. To define the effect of the film thickness on the power factor and output power, the ZnO:Al films with thicknesses in the range of 100 nm–900 nm were prepared by thermal evaporation method assisted by radio frequency atomic source. The results show that the film thickness has no effect on the phase composition and the (002) preferred orientation of the wurtzite phase in the films. However, the grain size, surface particle size, and surface roughness increase with increasing film thickness. Transmittance significantly reduced for the 900 nm film. Meanwhile, the film thickness affects the carrier concentration and mobility through the defects and growth modes, so that the TE parameters of the films change with the film thickness. The 700 nm film has the highest power factor, and the maximum power factor is 627 μW m−1 K−2 at 560 K. The output power of the films increases as the increase of the temperature on the hot side and higher power factor results in higher output power. The maximum output power of the 700 nm film is 116.01 pW at temperature difference of 1 K. Background: Minimal change disease (MCD) is the major cause of childhood idiopathic nephrotic syndrome, which is characterized by massive proteinuria and debilitating edema. Proteinuria in MCD is typically rapidly reversible with corticosteroid therapy, but relapses are common, and children often have many adverse events from the repeated courses of immunosuppressive therapy. The pathobiology of MCD remains poorly understood. Prior clinical observations suggest that abnormal T-cell function may play a central role in MCD pathogenesis. Based on these observations, we hypothesized that T-cell responses to specific exposures or antigens lead to a clonal expansion of T-cell subsets, a restriction in the T-cell repertoire, and an elaboration of specific circulating factors that trigger disease onset and relapses. Methods: To test these hypotheses, we sequenced T-cell receptors in fourteen MCD, four focal segmental glomerulosclerosis (FSGS), and four membranous nephropathy (MN) patients with clinical data and blood samples drawn during active disease and during remission collected by the Nephrotic Syndrome Study Network (NEPTUNE). We calculated several T-cell receptor diversity metrics to assess possible differences between active disease and remission states in paired samples. Results: Median productive clonality did not differ between MCD active disease (0.0083; range: 0.0042, 0.0397) and remission (0.0088; range: 0.0038, 0.0369). We did not identify dominant clonotypes in MCD active disease, and few clonotypes were shared with FSGS and MN patients. Conclusions: While these data do not support an obvious role of the adaptive immune system T-cells in MCD pathogenesis, further study is warranted given the limited sample size. Tenebenal, as a new insecticide, with a novel action mode, has shown an excellent insecticidal effect, but there is no relevant report about how it can effectively control termite colonies. In this study, we evaluated the insecticidal effects of tenebenal at different concentrations by laboratory study, including the contact toxicity, stomach toxicity, and other insecticide effects of Reticulitermes flaviceps. The results showed that tenebenal had no repellent effect on R. flaviceps in concentrations ranging from 0 to 32 ppm. LC50 and LC90 of tenebenal on R. flaviceps were 3.752 and 8.278 ppm when treating termites with tenebenal for 24 h, respectively. The survival rate and food consumption of R. flaviceps significantly decreased with the increase of the tenebenal concentration from 1 to 8 ppm, and there was no significant difference in concentration between 8 and 16 ppm. Tenebenal had inhibitive effects on particle transfer, tunneling, carcass-burying, and cannibalism behavior of R. flaviceps with concentrations from 1 to 16 ppm. R. flaviceps are exposed to sand treatment with 16, 32, and 64 ppm of tenebenal for 1 h, and the mortality for both donor and recipient is 36%, 77%, and 100% on day 3, respectively. Background and Aim: Non‐invasive hepatic steatosis algorithms are recommended in detecting metabolic‐associated fatty liver disease (MAFLD) in epidemiological studies. However, the diagnostic accuracy of these models is unclear. This study aimed to evaluate the diagnostic efficiency of five common models in a national survey population. Methods: The Third National Health and Nutrition Examination Survey (NHANES III) datasets were used in this study. The fatty liver index (FLI), hepatic steatosis index (HSI), non‐alcoholic liver disease‐liver fat score (NAFLD‐LFS), the steato text (ST), and visceral adiposity index (VAI) were evaluated. Results: The prevalence of MAFLD in the general population was 31.2%. The proportion of MAFLD estimated using the NAFLD‐LFS (30.8%) was the closest to the real number, whereas the ST model (66.1%) significantly overestimated the prevalence of MAFLD in this cohort. The FLI (36.9%) and HSI models (38.5%) also slightly overestimated the prevalence of MAFLD in the study population. The FLI had the highest area under the receiver operating characteristic (AUROC) value (0.793) among all models, with a sensitivity of 57.0%, a specificity of 83.8%, a positive predictive value (PPV) of 67.3%, and a negative predictive value (NPV) of 77.0%. The combination of the original algorithm with additional metabolic dysfunction criteria did not improve the diagnostic efficiency. The discriminative ability for MAFLD in all models was lower in participants with a normal body mass index (BMI). Conclusions: Non‐invasive models, especially the FLI, have satisfactory diagnostic performance in detecting MAFLD. However, models in people with normal BMIs require further development. In the past 10 years since its introduction, phenome-wide association studies (PheWAS) have uncovered novel genotype–phenotype relationships. Along the way, PheWAS have evolved in many aspects as a study design with the expanded availability of large data repositories with genome-wide data linked to detailed phenotypic data. Advancement in methods, including algorithms, software, and publicly available integrated resources, makes it feasible to more fully realize the potential of PheWAS, overcoming the previous computational and analytical limitations. We review here the most recent improvements and notable applications of PheWAS since the second half of the decade from its inception. We also note the challenges that remain embedded along the entire PheWAS analytical pipeline that necessitate further development of tools and resources to further advance the understanding of the complex genetic architecture underlying human diseases and traits. Pleiotropy, the concept that a gene or genetic variant affects more than one phenotype or trait, is at least a century old. By contrast, phenome-wide association studies (PheWAS), an approach used to identify cross-phenotype associations, were introduced only in the past decade. Still relatively young, PheWAS has rapidly matured into a widely used study design and analytical approach that can still benefit from improvements in its pipeline as genome-wide datasets expand in their breadth and depth, challenging the computational and statistical limits of today's PheWAS. Bilayer is an effective structure to enhance magnetic properties of films. This study employs high magnetic field (HMF) and amorphous Fe–Sm under-layer to tune the magnetic properties of evaporated Co–Sm/Fe–Sm bilayer films. Various analysis methods are used to explore the relation of microstructure evolution under above conditions with magnetic performance. The results show that both the amorphous Fe–Sm under-layer and the HMF can decrease surface roughness. Further, the HMF exhibits other effects that reduces column size, increases interplanar spacing and affects interface at atomic scale. The structural variation has effect on magnetic properties of bilayer films. The amorphous Fe–Sm under-layer increases magnetization, coercivity and squareness through changing the amount of non-magnetic structure at atomic scale. Meanwhile, the HMF decreases coercivity by changing pinning via the interface variation. Both the HMF and amorphous under-layer have no influence on the anisotropy of bilayer films. These results present an effective method to tune the magnetic properties of films through amorphous under-layer and HMF. • Amorphous FeSm under-layer has effect on microstructure and magnetic performance. • High magnetic field improves magnetization by affecting non-magnetic structure. • High magnetic field decreases coercivity by changing interface pinning. • Effect of high magnetic field on squareness is smaller than that of amorphous layer. In this study, we explored the role of the intrinsic defects on the tuning thermoelectric (TE) performance of Al-doped ZnO (ZnO:Al) films. The intrinsic defects of the Zn interstitial (Zn i) and the oxygen vacancy (V O) were controlled by varying the Zn content by using the radio frequency atomic source vacuum evaporation method. In the Zn-rich case, the excessive Zn formed Zn i to provide most of the carriers. V O became the main origin of the carriers with a decreasing Zn content. When the Zn content was 55.2% and 52.5%, the segregation of Zn i decreased the carrier concentration and subsequently, enhanced the resistivity and the Seebeck coefficient. The ZnO:Al film with a Zn content of 52.5% had the highest power factor of 274.03 μW·m−1·K−2 at 819 K. In addition, the higher Zn i concentration had a more prominent effect on thermal conductivity. The thermal conductivity increased to 1.756 W·m−1·K−1 from 0.608 W·m−1·K−1 with the Zn content decreasing from 55.2% to 50.6%. The ZT value reached the highest of 0.121 when the film had the optimum Zn concentration of 51.5%. It is necessary to develop excellent sensors for volatile organic compounds (VOCs) detection from the perspective of environmental protection and human health. Luminescent lanthanide metal‐organic frameworks (MOFs) have many advantages such as porosity, luminescence, and structural controllability, making them very suitable as luminescent sensing materials. Based on a functional oriented strategy, novel Ln‐MOFs are successfully constructed and their structure and properties are characterized in detail. A series of vapor luminescence sensing tests have shown that Eu/Tb doped L1‐Eu0.1/Tb0.9 not only exhibits high selectivity for alcohols, especially ethanol vapor, but also has high sensitivity and extremely low detection limit (9.76 mg L−1). To the knowledge, there are no reports on ratio luminescence sensors based on Ln‐MOFs for ethanol vapor, especially those with such excellent performance. Crystal structure determination demonstrates that ethanol binds to the framework through multiple hydrogen bonding interactions, altering the energy transfer efficiency between Tb3+ and Eu3+ to achieve ratio luminescence detection. In addition, L1‐Eu0.1/Tb0.9 has good repeatability, the prepared thin film sensing material also demonstrates the advantages of portability and processability. It is difficult to fabricate magnetic films containing small nanoparticles with a narrow size distribution. The nanomagnetic structural evolutions inside the nanoparticles in these types of films play a key role in the development of the magnetic properties. Magnetic composite films with a cubic array distribution of the nanoparticles are obtained using the layered growth of FeNi–SiO and SiO by the vacuum thermal evaporation method. The films contain small nanoparticles with a narrow size distribution (4.59 ± 0.18 nm) and an ordered cubic array. In the films, the magnetic induction lines measured by the differential phase contrast microscopy of the transmission electron microscope confirm that the disordered domain becomes multi-domain with an ordered cubic array. Furthermore, the saturation magnetization of such a film is higher than that of a pure FeNi film, despite the presence of the nonmagnetic SiO dielectric. R‐spondins (Rspos) are endogenous ligands of leucine‐rich repeat‐containing G‐protein‐coupled receptor 4 (LGR4). Rspos‐LGR4 signaling plays important roles in embryogenesis, gastrointestinal homeostasis, and food intake. Here, we investigated the impacts of Rspos‐LGR4 on hepatic cholesterol synthesis. Rspo1/3 and Lgr4 knockdown mice were used to investigate the impacts of Rspo1/3‐LGR4 on hepatic cholesterol synthesis. AMPKα agonist, antagonist, and shRNA were used to explore the downstream targets of Rspos‐LGR4 signaling. In our study, we reported that LGR4, Rspo1, and Rspo3 were highly expressed in hepatocytes and their expressions were sensitive to energy states. Rspo1 and Rspo3 reversed OA‐induced cholesterol synthesis, accompanying with increased the phosphorylation of AMPKα Thr172, reduced SREBP2 nuclear translocation, and Srebf2 mRNA expression. Conversely, hepatic LGR4 knockdown increased hepatic cholesterol synthesis and decreased the phosphorylation of AMPKα both in vitro and in vivo. Activation or inhibition of AMPKα significantly abolished the effects of LGR4 deficiency or Rspos, respectively, on cholesterol synthesis. Knockdown of AMPKα1 or/and AMPKα2 repressed Rspos‐induced inhibition on cholesterol synthesis. This paper investigates the problem of the distributed adaptive output feedback leader-following consensus control for a class of high-order nonlinear multiagent systems with unknown parameters and nonlinear terms. First, the reduced order dynamic gain ${k}$ -filters are built to estimate the unmeasured state variables. The bounds of the unknown parameters are estimated in order to avoid the over-estimation problem. Then, the dynamic surface control technique is used to design the distributed controller, and the computation load of the system is greatly reduced. High altitude hypobaric hypoxia environment impairs male's reproductive function. Leptin is an adipose tissue-derived hormone which regulates body weight homeostasis. Its receptor (LepR) has been found in all levels of male reproductive axis, indicating that it can affect male reproductive system in a direct or (and) indirect way. However, the role of leptin signaling in hypobaric hypoxia induced male reproductive dysfunction remains to be elucidated. In this study, we investigated the changes of leptin levels in male SD rats in stimulated altitude of 5500 m hypobaric hypoxia environment and their effects on the hypothalamus-pituitary-gonad axis (HPG axis). A hypoxia animal model was established using a hypobaric hypoxia chamber. Rats were divided randomly into 1, 7, 14, 28-day hypoxia group, recovery group (14 days hypoxia+14 days normoxia) and their control groups. Hypoxia groups displayed obvious changes of testicular and epididymis index compared to control groups. The total number of sperm and sperm motility rate decreased dramatically, while sperm deformity rate increased in hypoxia groups. The flow cytometry analysis showed that the percentage of haploid in 1-day, 7-day and 28-day hypoxia groups increased while the proportion of diploid decreased in 14-day and 28-day hypoxia group. TUNEL staining showed that the testis cells apoptosis index (AI) of hypoxia groups increased significantly, and the apoptosis of cells mainly focus on spermatogonia and spermatocytes. The expression of GnRH in hypothalamus decreased dramatically under hypoxia condition, accompanied with the reduction of serum testosterone (T) level in 1-day and 28-day hypoxia groups and free-testosterone level (FT) in 1-day and 14-day hypoxia groups. Importantly, ELISA analysis showed that serum leptin level decreased in 7-day hypoxia groups and acylated-ghrelin, gastrin also changed, accompanying with reduction of LepR in hypothalamus in hypoxia groups. Immunohistochemical staining exhibited increased leptin and LepR in testis under hypobaric hypoxia conditions. Our results suggested that simulated high altitude hypobaric hypoxia environment decreased male reproductive function, depressed HPG axis activity and altered the serum concentration of hormones related to food intake in adult male rats. Unstable nanosheet morphology of polycrystalline SnSe film becomes a limitation for its application in the field of waste heat recovery. This study dopes Co into the evaporated SnSe film to smooth the surface and to improve power factor. The results show that Co dopant makes the rough nanosheet change to surface densely packed structure. The dopant Co forms Co particles distributing around SnSe grain boundary, which suppress SnSe (011) plane growth and subsequent reduce nanosheet formation. In addition, Co dopant increases electric conductivity through increasing the carrier concentration, but the magnetism of Co particles decreases carrier mobility due to spin-charge coupling and also affects the Seebeck coefficient. Electrical and thermal transport controlled by growth mode can be used to optimize thermoelectric properties of ZnO:Al films, which was adjusted by the re‐evaporation of Zn and Al via substrate temperatures. The growth modes include equiaxed crystal, columnar crystal and coexistence of both crystals. In the ZnO:Al film, equiaxed crystals improve the carrier mobility and reduce the lattice thermal conductivity. Thus, the carrier mobility and thermal conductivity are tuned by the ratio of equiaxed crystals to columnar crystals. The carrier mobility is dependent on the growth‐mode‐related defects of oxygen vacancies, zinc interstitials and the substitutional dopant of Al. Improved thermoelectric properties with a power factor of 198.45 µW m−1 K−2 at 510 K were achieved. The application of thermoelectric films is limited to retain the temperature gradient. In this study, the Bi-Te films are deposited on the AAO template with a pore size of 100 nm using thermal evaporation. The results show that the conductive types of the Bi-Te film are tuned by source temperature. The power factor of the p-type porous film decreases 36% by comparing to that of the p-type nonporous film (1020 μW/mK2 at 250 °C). Meanwhile, the temperature difference in the porous device is maintained and is approximately 5.0 °C. Thus, the maximum output power is achieved in the porous device (about 25 pW), which is 5 times higher than that of the nonporous device. Abstract In this study, polycrystalline thin films of 330 nm in thickness with different SnSe and SnSe 2 contents were prepared by co-evaporation of Sn and Se through adjusting source temperatures and substrate temperature. The result illustrates that the film with pure SnSe phase is p-type and consists by bulk-like nanoparticles, its resistivity is high up to 139 mΩ·m, which results in a low thermoelectric performance. The films with the coexistence of SnSe and SnSe 2 phases are comprised of nanosheets. The existence of SnSe 2 phase changes the conductivity type of the films to n-type and reduces resistivity. At the same time, the nanosheets perpendicular to the substrate can improve the Seebeck coefficient by filtering the low energy carrier. When the SnSe 2 phase content is 34%, the Seebeck coefficient and the power factor of the film can reach the maximum, 255 μV/K and 155 μW/(m·K2) at 250 °C, respectively. Abstract This study assessed neuropeptide Y (NPY) expression in the hippocampus after long-term survival following traumatic brain injury (TBI) induced by controlled cortical impact (CCI) with or without the development of posttraumatic epilepsy (PTE). We hypothesized that following long-term survival after CCI, the severity of tissue injury and the development of PTE would correlate with the degree of hippocampal neurodegeneration as reflected by NPY + and neuronal nuclear antigen (NeuN)+ cell loss. Adult Sprague–Dawley rats of 2–3 months of age were lesioned in the right parietal cortex and monitored for seizure activity by video and/or video-EEG. Neuropeptide Y and NeuN immunoreactivities (IRs) were quantified by light microscopy and semiautomatic image analysis approaches for unbiased quantification. Severely injured animals, marked by extensive tissue loss in the ipsilateral neocortex and adjacent hippocampus, resulted in significantly lower NeuN + hilar cell density and NPY + cell loss in the contralateral Cornu Ammonis (CA)-3 and dentate hilus (DH). The degree of NPY + cell loss was more severe in CCI-injured animals with PTE than those animals that did not develop PTE. Mildly injured animals demonstrated no significant change of NPY expression compared with control animals. Our findings of long-term alterations of NPY expression in the hippocampus of severely brain-injured animals can provide important insights into the cellular and molecular consequences of severe TBI and posttraumatic epileptogenesis. Joint improvement of the Seebeck coefficient S and conductivity σ is a pressing issue to overcome the limit of the figure of merit ( ZT ) enhancement of thermoelectric materials. This study presents a joint improvement in the ZnO:Al films by tuning the diffusion of Au layer via substrate temperature, using a radio frequency-assisted molecular beam vapor deposition method. The phenomenon occurs in the ZnO:Al films at the room temperature (RT) substrate deposited on a 10 nm Au layer (RT-ZnO:AuAl). The films have a hexagonal wurtzite structure with an in-plane (002) preferred orientation and smooth surface. The resistivity values of the films are at the same magnitude of the transparent conducting oxide (~10 −6  Ω·m). The carrier concentration reaches 2.60 × 10 21  cm −3 for the RT-ZnO:AuAl film. The S is about 29.5 μV/K at 240 °C. Joint improvement of conductivity and Seebeck coefficient makes the PF of the RT-ZnO:AuAl film reaches 3.58 × 10 −5  Wm −1  K −2 , which is two orders of magnitude larger than that of the RT-ZnO:Au film without joint improvement. The results indicate that the higher conductivity originates from the Au layer, in-plane (002) preferred orientation, and the easily excited carriers in the ZnO:Al due to the low binding energy of the Zn 2p electrons. Improving the thermoelectric (TE) properties of typical p-type Bi 0.5 Sb 1.5 Te 3 film is crucial for power generation in film-based thermoelectric devices. The shape of the hexagonal structure in the Bi 0.5 Sb 1.5 Te 3 film can be controlled by adjusting the magnetron sputtering parameters. The results indicate that a high nucleation rate under high sputtering power promotes island growth with a preferred orientation of (015). Thermal energy influences the growth in the in-plane direction along the (00 l) orientation and induces the c-axis orientation in the out-of-plane direction. Consequently, a dense layered hexagonal structure with a high grain boundary density is formed, leading to increased defect density, carrier concentration, and phonon scattering. In this case, the power factor reaches 3149.01 μW·m-1·K-2 at 480 K and the ZT value is 1.14 at 480 K. Transparent ZnO films with high breakdown voltage and resistivity were deposited by the radio frequency-assisted evaporation method. In this paper, we have investigated on the structural, optical, and electrical properties of ZnO:Al2O3. The preferred orientation of the columnar structured in situ-grown film was along (002). The resistivity of the films was five orders of magnitude larger than the current's highest resistivity. The breakdown voltage of the film (8571 V/mm) was five times higher than the highest reported breakdown voltage for a ZnO semiconductor. Furthermore, the ZnO:Al2O3 film was transparent in the visible and infrared regions even though the film had an Al content of about 7% and a thickness of 100 nm. The high-frequency dielectric constant of the ZnO:Al2O3 film was higher than that of Al2O3. The possible reasons for the transparent ZnO:Al2O3 film behavior were second-phase Al2O3, lower carrier concentration, and strong bound electrons. Less defects and strong bonding contribute 4 orders of magnitude improvement to the high resistivity of ZnO films. The obtained results suggest that ZnO:Al2O3 can be used as an insulator layer between the p-n junction in order to improve the efficiency of the solar cell device. [ABSTRACT FROM AUTHOR]

Background & aims Leucine-rich repeat G-protein-coupled receptor 4 (LGR4) and its ligands R-spondin1-4 (Rspos) have been vastly investigated in embryonic development. The biological functions of Rspos-LGR4 system in liver remains largely unknown. Here, we explored whether it protects hepatocytes against hypoxia/reoxygenation (H/R) induced damage. Methods H/R injury was induced by dimethyloxalylglycine (DMOG) in AML12 cells and the effects of Rspo3 on cell proliferation and apoptosis were assessed. Specific shRNAs were used to interfere LGR4 or β-catenin. Results DMOG caused hepatocytes damage evidenced by increase in HIF-1α, cell death and apoptosis genes p27 and Bax , with concurrent decrease of cell proliferation genes PCNA and CyclinD1 . Of all the Rspos, Rspo3 is predominantly expressed in AML12 hepatocytes. Importantly, Rspo3 demonstrated an alteration in a manner similar to proliferation-related genes during H/R injury. Rspo3 pretreatment rendered hepatocytes less vulnerable to DMOG induced H/R injury. Ablation of LGR4 using shRNA attenuated the protective effects of Rspo3. Wnt3a also protected AML12 cells from damages caused by H/R, showing enhanced proliferation activity. Notably, knockdown of β-catenin in hepatocytes completely abolished the effect of Rspo3 pretreatment on the expression levels of PCNA and CyclinD1. The effects of nanowire content on the thermoelectric properties of ZnO films were investigated. The nanowire content of ZnO films was tuned by thermal oxidation of evaporated Zn films. The results showed that hexagonal and polyhedral morphologies on the surface of Zn films can be used to tune the nanowire content of ZnO films. Hexagonal nanoplates with a diameter of 100–350 nm readily grew ZnO nanowires with c-axis preferential orientation. Conversely, it was difficult to grow nanowires on polyhedral nanoparticles with diameters of 500–750 nm because the meeting of ZnO (101) and (001) facets suppressed nanowire growth. Thermoelectric parameters were strongly affected by nanowire content. In particular, carrier concentration increased with nanowire content. Carrier mobility also increased with nanowire content because the nanowires behaved as channels for electronic migration. The band gap of the films narrowed with increasing nanowire content because the binding energy of O 1s electrons with oxygen vacancies decreased. Huge difference of surface energies between dielectric and metal provides an effective method to tune the structure and magnetic properties of films. Due to the difference of surface energies between SiO 2 and Fe, dopant of SiO 2 with 3.6% in atomic ratio in Fe film transforms the strip and column structure in the Fe films to equiaxed crystal formed by nanoparticles of the Fe-SiO 2 films. Zeeman energy of applying high magnetic field (HMF) during film growth increases Fe atomic aggregation and atomic diffusion of SiO 2 into the grain boundary. This leads to the improvement of the particle size with applying HMF during the film growth. These structural evolutions have a significant effect on the performance of the films. After the dopant of SiO 2 , the saturation magnetization M s of the Fe-SiO 2 film (324.6 kA/m) is 73.2% less than that of the Fe films. The coercivity H c decreases 204.0% to 788.04 A/m. Human induced pluripotent stem cells (hiPSCs) hold great hopes for application in regenerative medicine due to their inherent capacity to self-renew and differentiate into cells from the three embryonic germ layers. For clinical applications, a large quantity of hiPSCs produced in standardized and scalable culture processes is required. Several groups, including ours, have developed methodologies for scaled-up hiPSC production in stirred bioreactors in chemically defined medium. In this study, we optimized the critical steps and factors that affect hiPSC expansion and yield in stirred-suspension cultures, including inoculation conditions, seeding density, aggregate size, agitation rate, and cell passaging method. Transdermal delivery of therapeutic biomolecules (including peptides) can avoid enzymatic digestion that occurs in the oral route. (Polyethylene glycol) diacrylate (PEGDA)-based microneedles, with good biocompatibility, are easily fabricated through photo-polymerization with a precisely controlled structure. It has successfully been used for the transdermal delivery of small molecule drugs such as 5-fluorouracil. However, the delivery of peptide-based therapeutics using this platform is seldom reported. This is because of the potential damage to the peptide during the photo-polymerization process of PEGDA. Herein, we introduce a method to load PEGDA microneedles with peptides without compromising peptide potency. Using gap junction inhibitor (Gap 26) as an example, the peptide was loaded into PEGDA microneedles through the swelling effect of PEGDA in the aqueous solution. The peptide-loaded microneedles were applied to a keloid scar model and exhibited inhibition expression of collagen I, a predominant marker of keloid scar, demonstrating its potential therapeutic effects. This paper studies the effects of high magnetic field (HMF) on the structure, optical and thermoelectric properties of the doped ZnO thin films. The results show that both Al dopant and application of HMF can affect the crystal structure, surface morphology, elemental distribution and so on. The particles of the thin films become small and regular by doping Al. The ZnO films oxidized from the Au/Zn bilayer have needle structure. The ZnO films oxidized from the Au/Zn-Al bilayer transform to spherical from hexagonal due to the application of HMF. The transmittance decreases with doping Al because of the opaque of Al element and decreases with the application of HMF due to the dense structure obtained under HMF. Electrical resistivity ( ρ ) of the ZnO films without Al decreases with increasing measurement temperature ( T ) and is about 1.5 × 10 −3 Ω·m at 210 °C. However, the ρ of the Al-doped ZnO films is less than 10 −5 Ω·m. The Seebeck coefficient ( S ) of the films oxidized from the Au/Zn-Al films reduces with increasing T . The S values oxidized under 0 T and 12 T conditions are 2.439 μV/K and −3.415 μV/K at 210 °C, respectively. Power factor reaches the maximum value (3.198 × 10 −4 W/m·K 2 ) at 210 °C for the film oxidized under 12 T condition. Cleft lip with or without palate (CL/P) is a common congenital anomaly in humans and is thought to be caused by genetic and environmental factors. However, the epigenetic mechanisms underlying orofacial clefts are not fully understood. Here, we investigate how the overdose of retinoic acid (RA), which can induce cleft palate in mice and humans, regulates histone methyltransferase, Wolf–Hirschhorn syndrome candidate 1 (WHSC1) during palatal development in mice. We treated mouse embryonic fibroblasts (MEFs) with 1 μM all-trans RA and discovered that the global level of H3K36me3 was downregulated and that expression of the H3K36 methyltransferase gene, Whsc1 , was reduced. The expression level of WHSC1 in embryonic palatal shelves was reduced during palatogenesis, following maternal administration of 100 mg/kg body weight of RA by gastric intubation. Furthermore, the expression of WHSC1 in palatal shelves was observed in epithelial and mesenchymal cells at all stages, suggesting an important role for palatal development. Our results suggest that the pathogenesis of cleft palate observed after excessive RA exposure is likely to be associated with a reduction in the histone methyltransferase, WHSC1. Safety is the foremost issue in all human cell therapies, but human induced pluripotent stem cells (iPSCs) currently lack a useful safety indicator. Studies in chimeric mice have demonstrated that certain lines of iPSCs are tumorigenic; however a similar screen has not been developed for human iPSCs. Here, we show that in vitro cartilage tissue engineering is an excellent tool for screening human iPSC lines for tumorigenic potential. Although all human embryonic stem cells (ESCs) and most iPSC lines tested formed cartilage safely, certain human iPSCs displayed a pro-oncogenic state, as indicated by the presence of secretory tumors during cartilage differentiation in vitro.We observed five abnormal iPSC clones amoungst 21 lines derived from five different reprogramming methods using three cellular origins. Pyroptosis is a newly discovered type of programmed cell death associated with inflammatory and fibrotic diseases. Macrophages play an important role in inducing early immune inflammation in systemic sclerosis (SSc). To investigate the effect of macrophages pyroptosis on fibrosis of SSc. Pyroptosis/inflammatory markers in serum and skin of SSc patients were detected. Bleomycin (BLM) was subcutaneously injected to establish SSc mouse model. The levels of pyroptosis markers, dermal thickness and collagen deposition in skin were assessed before and after the administration of pyroptosis inhibitors, including MCC950, Disulfiram and necrosulfonamide (NSA). Human-derived monocyte-macrophage cell line (THP-1) or mouse bone marrow-derived macrophages (BMDMs) were primed with lipopolysaccharide (LPS) and stimulated by silicon dioxide (SiO 2) to induce cell pyroptosis. Fibroblasts from patients with SSc were co-cultured with pyroptotic THP-1 cells, and the collagen production was assessed. Pyroptotic/inflammatory proteins, including NLRP3, cleaved-Caspase (CASP)1, GSDMD-N terminal and IL-18 were increased in the serum, and ASC aggregation and GSDMD were elevated in macrophages in the skin of SSc patients. SSc mice showed increased pyroptosis markers, dermal thickness and collagen deposition in skins, which were alleviated by MCC950, Disulfiram and NSA. Pyroptosis of THP-1 cells and BMDMs was induced by LPS/SiO 2 , and it was reduced by the inhibitors of Cathepsin B, NLRP3, CASP1 and GSDMD. Co-culture with pyroptotic THP-1 cells increased the fibrotic proteins in fibroblasts, which were alleviated by pyroptosis inhibitors. SSc patients and BLM-induced mouse model presented increased pyroptosis. Thymidylate synthase (TS) is a critical enzyme for DNA biosynthesis and many nonclassical lipophilic antifolates targeting this enzyme are quite eflicient and encouraging as antitumor drugs. In this paper, the binding model of 14 antifolates of substituted benzylamino- and heterocyclylmethylamino-carbodithioate derivatives of 4-(3H)-quinazolinone with TS is examined using molecular simulation methods—FlexiDock and SCORE2.0. The resulting conformation and orientation of these antifolates are directly applied to CoMFA study. The robust QSAR model, its three-dimensional contour map, and binding score of these antifolates derived from SCORE2.0 provide guidelines for structural optimization of current antifolates. Protein kinase catalyzes the transfer of the y-phosphoryl group from ATP to the hydroxyl groups of protein side chains, which plays critical roles in signal transduction pathways by transmitting extracellular signals across the plasma membrane and nuclear membrane to the destination sites in the cytoplasm and the nucleus. Protein kinase C (PKC) is a superfamily of phospholipid-dependent Ser/Thr kinase. There are at least 12 isozymes in PKC family. They are distributed in different tissues and play different roles in physiological processes. On account of their concern with a variety of pathophysiologic states, such as cancer, inflammatory conditions, autoimmune disorder, and cardiac diseases, the inhibitors, which can inhibit the activity of PKC and the interaction of cytokine with receptor, and interfere signal transduction pathway, may be candidates of therapeutic drugs. Therefore, intense efforts have been made to develop specific protein kinase inhibitors as biological tools and therapeutic agents. Mn-based phosphors with the wavelength of 700–750 nm are an important category of far-red phosphors that have promising potential in the application of plant lighting, and the higher ability of the far-red light emitting of the phosphors is beneficial to plant growth. Herein, a series of Mn4+- and Mn4+/Ca2+-doped double perovskite SrGd2Al2O7 red-emitting phosphors with wavelengths centered at about 709 nm were successfully synthesized by means of a traditional high-temperature solid-state method. First-principles calculations were conducted to explore the intrinsic electronic structure of SrGd2Al2O7 for a better understanding of the luminescence behavior in this material. Extensive analysis demonstrates that the introduction of Ca2+ ions into the SrGd2Al2O7:Mn4+ phosphor has significantly boosted the emission intensity, internal quantum efficiency, and thermal stability by 170%, 173.4%, and 113.7%, respectively, which are superior to those of most other Mn4+-based far-red phosphors. The mechanism of the concentration quench effect and the positive effect of co-doping Ca2+ ions in the phosphor were extensively explored. All studies suggest that the SrGd2Al2O7:0.1%Mn4+, 11%Ca2+ phosphor is a novel phosphor that can be used to effectively promote the growth of plants and regulate the flowering cycle. Multi-nanocomposite is an effective method to achieve high thermoelectric (TE) performance by breaking the tradeoff among three TE parameters. This paper presents a comparative study of incorporated carbon nanotube (CNT)/Au in evaporated porous Al-doped ZnO (AZO) films. The effect of the nanocomposite interface on scattering of carriers and phonons is used to independently tune thermal conductivity and power factor in the multi-nanocomposite AZO films. This breaks the tradeoff and improves the figure of merit (ZT) of the film. Moreover, the temperature gradient retains in the out-of-plane of the films. These make the AZO film composited CNT and porosity have higher output power than that of the non-porous films. This provides a simple method for the low-dimension film to increase the TE properties through breaking the tradeoff and makes the TE materials suitable for efficient harvesting in low-grade waste heat. Bi 2 Te 3 is a successful and commercial thermoelectric material. However, Bi 2 Te 3 is easily generates point defects, which limits its thermoelectric (TE) properties. This study presents a novel method to tune point defects by using short-range atomic diffusion via pulsed electric field (PEF) treatment in C-doped Bi 2 Te 3 films. The C atoms migrate from the interstitial position in the Bi layer (C i) and the Van der Waals gap (C vdw) to the Bi vacancy after the PEF treatment. For this case, it increases the content of the Te–C bonds for the PEF films, resulting in increased electron concentration. And the PEF can eliminate amorphous regions of the C-doped Bi 2 Te 3 films. In addition, the shorter bond length of the Te–C improves the compressive stress of the film and increases the effective mass of carriers. Thus, the conductivity and the Seebeck coefficient are improved simultaneously. In this study, the power factor was 2450.32 μW m−1·K−2 at 513 K after the PEF treatment. In this work, the competitive relationship between the double amorphous phases is systematically investigated to increase the high-temperature wear resistance of the self-lubricating TiAlSiCN coating. These double amorphous phases combine the SiN x phase with high-temperature resistance and the C-related phase with a low friction coefficient. An increase in the Si content facilitates the change in the phase composition. The resultant coating exhibits an adhesion strength of 153 N, a friction coefficient of 0.24 and a hardness of 25.24 GPa upon generation of the TiAlN(C), the SiN x and the C-related phase. In this case, the TiAlSiCN coating achieves long-term self-lubrication at high temperatures with the titanium alloy friction pair. This mechanism stems from the SiN x phase replacing the portion of the easily oxidized amorphous C-related phase, which yields an enhanced high-temperature wear resistance. The cutting distance of this TiAlSiCN is 120% higher than that of the TiAlCN during the high-speed cutting of the titanium alloy. The order-disorder transition (ODT) of RE zirconate (RE 2 Zr 2 O 7) has received much attention due to its potential applications in the design of thermal barrier coatings (TBCs). A series of high entropy RE zirconates (RE = La, Sm, Gd, Er and Lu) were designed and synthesized in order to regulate the fluorite-pyrochlore phases, and to investigate the effect of phase composition on the microstructures and performances. The results showed that the sintering temperature, as well as equivalent ion radius, played an important role in regulating the phase transformation of fluorite-pyrochlore. The coexistence of pyrochlore-fluorite phases not only can result in the decrease in grain size but also increased the lattice strain of each phase, thereby enhancing the phonon scattering and contributing to the reduction in the thermal conductivity. The CMAS corrosion resistance was mainly related to phase compositions, and the thicknesses of reaction layer decreased gradually with the increasing fluorite phase content. Current applications of thermoelectric (TE) materials are limited by their low conversion efficiency. Interfacial engineering is a common way to improve TE properties. This study employed a pulsed electric field to tune the grain interfacial structure of Bi 2 Te 3 films and thereby improve their power factor. The high-angle grain boundaries of the as-deposited films were transformed into low-angle grain boundaries (LAGBs) by the electrostatic force of a high pulse current density. The low interfacial energy of LAGBs maintains high carrier mobility while their reduced defects result in a low carrier concentration. These effects improve the Seebeck coefficient and conductivity simultaneously, with the power factor reaching 5394.77 μW·m−1·K−2 at 513 K. In addition, the compression stress generated by LAGBs enhances phonon scattering, creating a film with the highest power density yet reported (1.31 W·m−2). Background: Previous studies have suggested that Toxoplasma gondii (T. gondii) infection might be associated with fatty liver disease. However, the relationship between non-alcoholic fatty liver disease (NAFLD) and T. gondii infection has not been investigated in a large population. We aimed to study the relationship between those two diseases using a population-based dataset from the United States.Methods: The data were collected from the third National Health and Nutrition Examination Survey (NHANES III) between 1988 and 1994. Statistical analysis was applied to compare the prevalence of NAFLD in anti-T. gondii antibody-positive participants with antibody-negative ones.Results: A total of 9465 persons with a mean age of 44.33 ± 16.21 years, 46.9% of which were males, were included in the final analysis. Their mean BMI was 27.60 ± 5.96 kg/m2. A total of 2520 participants (26.62%) were positive for the T. gondii antibody. There was an increasing trend of seroprevalence of T. gondii with age (P for trend < 0.001). The incidence of NAFLD in the seropositive group was higher than that in the seronegative group (27.10% vs 23.40%, p < 0.001). In addition to this, metabolic biomarkers, including serum lipid, fasting blood-glucose, and uric acid were also significantly higher in the seropositive group. However, multivariate analysis revealed that T. gondii infection was not an independent risk factor for NAFLD. Age was independently correlated with both the prevalence of T. gondii and NAFLD.Conclusions: Patients with T. gondii infection may have a higher prevalence of NAFLD. This study focused on identifying bladder cancer (BC)-associated genes, transcription factors (TFs), and microRNAs (miRNAs). Two microarray data sets GSE37815 and GSE40355 were utilized to screen common differentially expressed genes (DEGs) associated with BC. Then, functional enrichment analysis was performed for elucidating the involved functions of DEGs. Subsequently, the protein–protein interaction (PPI) network and submodule of PPI network were analyzed. Finally, the regulation relationships of TF–DEGs and miRNA–DEGs were obtained to construct miRNA–target–TF regulatory network. DEGs were identified across BC and normal bladder tissues samples. Functional enrichment analysis results showed that most upregulated DEGs were closely associated with the Gene Ontology function of "mitotic spindle assembly checkpoint" and pathway of "Cell cycle," whereas most downregulated DEGs were significantly associated with "Complement and coagulation cascades" pathway (e.g., A2M and F13A1) and "Ras signaling pathway" (e.g., GNG11). DEGs such as F13A1 and A2M were highlighted in the PPI network and Submodule 1. In addition, three centromere-associated CENPK, CENPF, and CENPO were enriched in Submodule 2. Moreover, miR-519d had high degree in the regulatory network and CENPO was predicted to be one target of miR-519d. The upregulated CENPK, CENPF, and CENPO, and downregulated A2M, F13A1, and GNG11 might contribute to the progression of BC. In addition, the downregulated miR-519d might lead to the development of BC by upregulating the expression of CENPO. However, future investigation of those findings should be needed.
