Monoterpenes are commonly known for their role in the flavors and fragrances industry and are also gaining attention for other uses like insect repellant and as potential renewable fuels for aviation. Corynebacterium glutamicum, a Generally Recognized as Safe microbe, has been a choice organism in industry for the annual million ton-scale bioproduction of amino acids for more than 50 years; however, efforts to produce monoterpenes in C. glutamicum have remained relatively limited. In this study, we report a further expansion of the C. glutamicum biosynthetic repertoire through the development and optimization of a mevalonate-based monoterpene platform. In the course of our plasmid design iterations, we increased flux through the mevalonate-based bypass pathway, measuring isoprenol production as a proxy for monoterpene precursor abundance and demonstrating the highest reported titers in C. glutamicum to date at 1504.6 mg/L. Our designs also evaluated the effects of backbone, promoter, and GPP synthase homolog origin on monoterpene product titers. Monoterpene production was further improved by disrupting competing pathways for isoprenoid precursor supply and by implementing a biphasic production system to prevent volatilization. With this platform, we achieved 321.1 mg/L of geranoids, 723.6 mg/L of 1,8-cineole, and 227.8 mg/L of linalool. Furthermore, we determined that C. glutamicum first oxidizes geraniol through an aldehyde intermediate before it is asymmetrically reduced to citronellol. Additionally, we demonstrate that the aldehyde reductase, AdhC, possesses additional substrate promiscuity for acyclic monoterpene aldehydes.

Background Objectives Methods Results Conclusions Limonene and linalool are used in cosmetic products for their floral scents, but their oxidation products are strong contact allergens whose mechanisms of action are still not fully understood.The effects of limonene hydroperoxide (Lim‐2‐OOH) and linalool hydroperoxides (Lin‐6/7‐OOH) on the lipid profile of a human keratinocyte cell line (HaCaT) were evaluated. 2,4‐Dinitrofluorobenzene (DNFB) was also included.Lim‐2‐OOH and Lin‐6/7‐OOH were synthesised according to previous methods. HaCaT cells were treated with allergens (10 μM) for 24 h and the cellular lipid extracts were analysed by C18 liquid chromatography with tandem mass spectrometry (LC–MS/MS). Data analysis was performed using Lipostar software. Statistical analysis was carried out using Metaboanalyst and R software.All three sensitisers used caused significant changes in the lipidome of HaCaT cells in a similar trend. There was an upregulation in several plasmanyl/plasmenyl phospholipids (O‐/P‐phosphatidylcholines [PC] and O‐/P‐phosphatidylethanolamines [PE]), sphingolipids (HexCer) and triacylglycerol lipid species, and a decrease in some polyunsaturated fatty acids‐containing phospholipid (PE and PC) species suggesting oxidative stress and inflammation.This study is the first to evaluate the plasticity of the HaCaT cell lipidome in response to allylic hydroperoxide allergens Lim‐2‐OOH and Lin‐6/7‐OOH, together with the experimental contact allergen DNFB. Pichia kudriavzevii (formerly Candida krusei) poses a significant threat to immunocompromised patients due to its inherent resistance to various antifungal drugs. This study explored the anticandidal potential of citral, linalool, and carvacrol in combination with nystatin against P. kudriavzevii strains. Using the microdilution method following CLSI guidelines, Minimum Inhibitory Concentrations (MICs) and fungicidal concentrations (MFCs) were determined. Citral exhibited MIC values ranging from 50 to 100 µg/ml, averaging 70.24 ± 16.99 µg/ml, while carvacrol had MIC values of 50 to 100 µg/ml, averaging 86.90 ± 16.99 µg/ml. Linalool demonstrated weaker antifungal activity, with MIC values between 100 and 200 µg/ml, averaging 150 ± 38.73 µg/ml. The study assessed the synergistic effectsof these phenols with nystatin through fractional inhibitory concentration indices (FICIS). In addition, flow cytometry was employed to assess apoptosis induction in P. kudriavzevii cells. Carvacrol displayed a remarkable synergistic effect in combination with nystatin against all 21 isolates tested. Conversely, linalool showed synergy in 17 isolates, while citral exhibited synergy in only 2 isolates. These findings highlight distinct patterns of synergy between the different compounds and nystatin against P. kudriavzevii. Also, Carvacrol emerged as the most potent inducer of apoptosis across all P. kudriavzevii strains, followed by citral and linalool. This suggests that carvacrol not only possesses a stronger antifungal effect but also has a more pronounced ability to trigger programmed cell death in P. kudriavzevii. In conclusion, the study supports the potential of carvacrol, citral and linalool, as anticandidal agents, suggesting their supplementation with nystatin for treating P. kudriavzevii infections. Simple Summary: Drosophila suzukii (Diptera: Drosophilidae) is an invasive pest that lays eggs in ripe fruits. The short generation time, high fertility, and wide host range allows this species to cause important yield losses. Essential oils, extracted from a number of plant species, have been studied for their attractiveness to and deterrence of D. suzukii. In this study, we found clary sage oil exhibited dose-dependent repellency against both D. suzukii adults and larvae. Also, we identified that linalyl acetate and linalool were two major constituents of clary sage oil by GC-MS. Furthermore, we detected that both linalyl acetate and linalool repelled D. suzukii adults and larvae. In addition, we examined the electroantennography (EAG) responses of D. suzukii to clary sage oil, linalyl acetate, and linalool. These results suggest clary sage oil and its constituents linalyl acetate and linalool could be potential repellents for the management of D. suzukii. Drosophila suzukii (Diptera: Drosophilidae), spotted-wing drosophila, poses a significant threat to soft-skinned fruit crops in the Americas, Europe, Africa, and Oceania, as well as in Asia. The application of chemical insecticides is the primary control strategy for D. suzukii; however, resistance has developed with the indiscriminate use of chemical insecticides. Essential oils, considered potential alternatives to pesticidal strategies, exhibit potent toxic and sublethal behavioral effects against numerous pests, including D. suzukii. Clary sage oil repels a variety of agricultural and household pests; however, whether it has a repellent effect against D. suzukii remains unknown. Here, we found that clary sage oil exhibited dose-dependent repellency against D. suzukii adults in a T-maze assay, a two-choice assay and a two-choice attraction assay. Also, clary sage oil showed a significant repellent effect against D. suzukii larvae. Next, we explored the chemical constituents of clary sage oil by GC-MS and identified two major constituents, linalyl acetate (40.03%) and linalool (23.02%). Furthermore, the behavioral assays of linalyl acetate and linalool showed that both compounds conferred comparable repellency against D. suzukii adults and larvae. Finally, we found clary sage oil, linalyl acetate, and linalool elicited EAG responses in D. suzukii, especially clary sage oil, suggesting the repellency was mediated by the olfactory system. These findings indicate that D. suzukii shows olfactory-based behavioral avoidance of clary sage oil, linalyl acetate, and linalool. Carpesium macrocephalum, a species native to China, Korea, Japan, and Russia, has been used medicinally in the countries of its origin. Though mono- and sesquiterpenoids are known constituents of C. macrocephalum, the complete analysis of essential oils produced by the roots and aerial parts of the plant has not been published until now. The present study discloses considerable differences in the composition and cytotoxic activity of essential oils distilled from roots and shoots of C. macrocephalum. The GC-MS-FID analyses have led to the identification of 131 compounds in all, of which 114 were found in aerial parts and 110 in the roots of the plants. The essential oil distilled from shoots contained a mixture of nerol and thymol methyl ether (c. 26%), neryl isobutyrate (c. 12%) and linalool (c. 9%) as major constituents, whereas alantolactone (c. 29%), thymol methyl ether (c. 7%) and 2,5-dimethoxy-p-cymene (thymohydroquinone dimethyl ether, c. 7%) predominated in the essential oil obtained from the roots. The oils demonstrated weak antibacterial activity against Staphylococcus aureus and, at concentrations up to 2.08 mg/mL (oil from the aerial parts) and up to 3.38 mg/mL (oil from roots), were inactive against Gram-negative bacteria. Anxiety disorder is one of the most common neuropsychiatric disorders, and affects many people's daily activities. Although the pathogenesis and treatments of anxiety disorder have been studied for several decades, the underlying mechanisms remain elusive. Here, we provide evidence that olfactory stimuli with inhaled linalool or 2-phenylethanol decreased mouse anxiety-like behaviors and increased the activities of hippocampal dentate granule cells (DGCs). RNA-sequencing analysis identified retrograde endocannabinoid signaling, which is a critical pathway for mood regulation and neuron activation, is altered in the hippocampus of both linalool- and 2-phenylethanol-exposed mice. Further studies found that selective inhibition of endocannabinoid signaling by injecting rimonabant abolished the activation of DGCs and the anxiolytic effect induced by linalool or 2-phenylethanol. Together, these results uncovered a novel mechanism by which linalool or 2-phenylethanol decreases mouse anxiety-like behaviors and increases DG activity likely through activating hippocampal retrograde endocannabinoid signaling. The unimolecular reactions of protonated myrcene and linalool were investigated by collision‐induced dissociation and density functional theory calculations. Experiments on a triple quadrupole mass spectrometer showed that protonated myrcene undergoes two major unimolecular reactions losing propene and isobutene, and two minor reactions of ethene and propane loss. In each case, the product ion consists of a substituted five‐member ring. Protonation of myrcene was found to form four distinct protomers, three of which can be significantly populated in the ion source. The observed fragmentation reactions were calculated and found to depend on the starting protomer. Each pathway consisted of several hydrogen‐migration and ring‐forming/opening steps on the way to the observed products. Likewise, protonation of linalool also produces three distinct protomers, with the global minimum being formed by protonation of a central double bond. The major reaction is water loss to form protonated myrcene, but two minor channels were also observed resulting in loss of acetone and isobutene. The calculated minimum energy reaction pathways were found to be consistent with the relative abundances of the ions in the experimental breakdown diagrams. [ABSTRACT FROM AUTHOR]

Summary: A recent increase in the demand for pineapple necessitates the development of new varieties with superior flavour quality. In this study, we characterised the volatile profiles of three new Okinawan pineapple breeding lines using gas chromatography (GC)–mass spectrometry (MS)‐based electronic nose (e‐nose), glycosylated volatiles, and alcohol‐acyltransferase (AAT) activity measurements. The volatile profiles of the fruits were selected using the GC–MS‐e‐nose and visualised using multivariate statistical plots. The glycosidically bound compounds were collected by solid‐phase extraction and hydrolysed by β‐glucosidase. In total, 12 hydroxyl‐group substances, including chavicol (medicinal‐herbal aroma), geraniol (floral‐sweet), and linalool (floral‐citrus), were released from these glycosides. Crude enzymes containing AAT were extracted, and their in vitro activities in 2‐methylbutyl acetate production were highly related to their contents. These results provide information regarding volatile profiles and bounded‐aroma resources, as well as the esterification potential of new Okinawan pineapple breeding lines for use in the agro‐food industry. Acalabrutinib (ACL) was approved in the United States in the year of 2017 and 2019 for the treatment of chronic lymphocytic leukemia (CLL) and relapsed mantle cell lymphoma (MCL). Low-energy method was employed to curate nanoemulsion (NE) for which various components were screened through solubility study and optimization was done through a pseudo-ternary phase diagram. The NE was evaluated for its droplet size, polydispersity index (PDI), transmittance, morphology, rheology, robustness to dilution, the effect of pH, storage stability, and ex vivo permeation study. The optimized ACL-NE demonstrated an appropriate droplet size (94.35 ± 0.3 nm), PDI (0.27 ± 0.03), and an entrapment efficiency of 67.38 ± 2.69%. It showed non-Newtonian (shear thinning behavior) due to reduced viscosity with an increasing shear rate. The apparent permeability was 3.09-fold higher for ACL-NE than ACL suspension. An in vitro drug release study depicted a higher release of ACL (71.10 ± 10.99%) from the optimized NE in a sustained fashion than the suspension (49.01 ± 1.65%). A dose-dependent cytotoxicity was observed in MOLT-4 and HH-cell lines where the IC50 values of ACL were 5.62- and 16.49-fold reduced when encapsulated in oil globules for the respective cell lines. Blank-NE did cause a reduction in cellular toxicity at higher doses. A 7.31- and 5.1-fold increase in Cmax and bioavailability was noted between ACL suspension and ACL-NE. [ABSTRACT FROM AUTHOR]

The impact of the roasting degree on ultra-high-pressure cold brew (UHP) coffee remains unclear, although it has been found that UHP technology accelerates the extraction of cold brew (CB) coffee. Therefore, this study investigated the effects of three different degrees of roasting (light, medium, and dark) on the physicochemical characteristics, volatile and non-volatile components, and sensory evaluation of UHP coffee. Orthogonal partial least-squares-discriminant analysis (OPLS-DA) and principal component analysis (PCA) were used to assess the effects of different roasting degrees. The results showed that most physicochemical characteristics, including total dissolved solids (TDSs), extraction yield (EY), total titratable acidity (TTA), total sugars (TSs), and total phenolic content (TPC), of UHP coffee were similar to those of conventional CB coffee regardless of the degree of roasting. However, the majority of physicochemical characteristics, non-volatile components, including the antioxidant capacity (measured based on DPPH and ABTS) and melanoidin, caffeine, trigonelline, and CGA contents increased significantly with an increase in roasting degree. The sensory evaluation revealed that as the roasting degree rose, the nutty flavor, astringency, bitterness, body, and aftertaste intensities increased, while floral, fruity, and sourness attributes decreased. The HS-SPME-GC/MS analysis showed that most volatile components increased from light to dark roasting. Moreover, 15 representative differential compounds, including hazelnut pyrazine, linalool, butane-2,3-dione, and 3-methylbutanal, were identified by calculating the odor-active values (OAVs), indicating that these contributed significantly to the odor. The PCA showed that the distance between the three roasting degree samples in UHP coffee was smaller than that in CB coffee. Overall, the effect of roasting degrees on UHP coffee was less than that on CB coffee, which was consistent with the results of physicochemical characteristics, volatile components, and sensory evaluation. Rolling plays an important role in shaping the characteristic quality of black tea. However, the dynamic alterations in volatile compounds during the rolling process remain unclear, which hampers the quality control of black tea. In this study, the dynamic changes of volatile compounds in Congou black tea throughout the rolling process were explored by a combination of GC-E-Nose and GC-MS analysis. The results of PCA and PLS-DA by GC-E-Nose revealed significant alterations in the volatile profiles, particularly at the 15-min rolling stage. A total of 82 volatile compounds including eight categories were identified in Congou black tea during the rolling process. The most abundant volatiles were aldehydes, alcohols, and esters. Through the integration of variable importance in the projection >1 and p < 0.05, 21 volatile compounds were pinpointed as pivotal volatiles responsible for distinguishing diverse rolling processes. According to their pathways, glycoside-derived volatiles (GDVs) (such as linalool and geraniol) and fatty acid-derived volatiles (FADVs) (such as (Z)-2-hexen-1-ol and hexanal) highlighted the pivotal role in the formation of the aroma of black tea during rolling process. GDVs, characterized by floral, honeyed, and fruity aromas, reached the lowest point at 35-min rolling and subsequently rebounded. FADVs exhibited an ascending trajectory during the initial 15-min rolling, followed by a downward trend. The current study is objected to investigate the essential oil yields, chemical compositions, and enantiomeric distribution of wild and cultivated linalool rich Origanum dubium Boiss. essential oils obtained at pre-flowering, initial flowering, and post-flowering growth stages. Wild plant materials are collected from Mediterranean region of Turkey. Samples were also cultivated in Mediterranean and Aegean coastlines of Turkey in the experimental fields of Ministry of Agriculture and Forestry of Turkey. O. dubium samples yielded between 6.7 and 9.7% (mL/100 g) of essential oils. The compositions were dominated by oxygenated monoterpenes (98.2–98.9%). Linalool was detected as the major compound in all essential oils (97.0–97.6%). The dominant enantiomer was discovered as (R)-(-)-linalool with a 99.3–99.4% enantiomeric purity. The essential oil yields, chemical compositions and enantiomeric distributions were found to be constant regardless of cultivation, geographical locations or different growth stages. The result of this manuscript indicates that by culturing linalool chemotype of O. dubium, the industrial need for (R)-(-)-linalool could be met at more affordable costs. [ABSTRACT FROM AUTHOR]

Scenting is an important process for the formation of aroma quality in floral Longjing tea. There are differences in the aroma quality of osmanthus Longjing teas processed by different scenting processes. The efficient isolated scenting method was employed to process a new product of osmanthus Longjing tea in this study, and this was compared with the traditional scenting method. The volatile compounds of osmanthus Longjing tea were analyzed by a GC-MS instrument. In addition, the effects of scenting time and osmanthus consumption on the aroma quality of Longjing tea were studied. The results indicated that there were 67 kinds of volatile compounds in the osmanthus Longjing tea produced by the isolated scenting process (O-ISP), osmanthus Longjing tea produced by the traditional scenting process (O-TSP), and raw Longjing tea embryo (R), including alcohols, ketones, esters, aldehydes, olefins, acids, furans, and other aroma compounds. The proportions of alcohol compounds, ester compounds, aldehyde compounds, and ketone compounds in O-ISP were higher than in O-TSP and R. When the osmanthus consumption was increased, the relative contents of volatile aroma compounds gradually increased, which included the contents of trans-3,7-linalool oxide II, dehydrolinalool, linalool oxide III (furan type), linalool oxide IV (furan type), 2,6-Dimethyl cyclohexanol, isophytol, geraniol, 1-octene-3-alcohol, cis-2-pentenol, trans-3-hexenol, β-violet alcohol, 1-pentanol, benzyl alcohol, trans-p-2-menthene-1-alcohol, nerol, hexanol, terpineol, 6-epoxy-β-ionone, 4,2-butanone, 2,3-octanedione, methyl stearate, cis-3-hexenyl wasobutyrate, and dihydroanemone lactone. When the scenting time was increased, the relative contents of aroma compounds gradually increased, which included the contents of 2-phenylethanol, trans-3,7-linalool oxide I, trans-3,7-linalool oxide II, dehydrolinalool, isophytol, geraniol, trans-3-hexenol, β-ionol, benzyl alcohol, trans-p-2-menthene-1-ol, nerol, hexanol, terpineol, dihydroβ-ionone, α-ionone, and β-ionone,6,10. The isolated scenting process could achieve better aroma quality in terms of the floral fragrance, refreshing fragrance, and tender fragrance than the traditional scenting process. The isolated scenting process was suitable for processing osmanthus Longjing tea with high aroma quality. Papaya is a tropical fruit crop renowned for its rich nutrition, particularly pro-vitamin A. Aroma substances are a major component of fruit quality. While extensive research has been conducted on papaya aroma, there has been a notable lack of in-depth research into a specific class of substances. To bridge this gap, our study focused on analyzing the aroma components of various papaya varieties and their biosynthesis pathways. We compared the volatile components of three papaya varieties with distinct flavors at various ripeness stages. A continuous accumulation of linalool, a volatile compound, in the 'AU9' fruit was detected as it matured. The linalool content reached 56% of the total volatile components upon full ripening. Notably, this percentage was significantly higher than that observed in the other two varieties, 'ZhongBai' and 'Malaysian 7', indicating that linalool serves as the primary component influencing the papaya's odor. Subsequently, we identified CpTPS18, a gene associated with linalool biosynthesis, and demonstrated its ability to catalyze linalool production from GPP and enhance its accumulation through overexpression in papaya fruits, both in vivo and in vitro. Based on transcriptomic analysis, it was predicted that CpMYB56 and CpNAC56 may transcriptionally activate the expression of CpTPS18. Subsequent yeast one-hybrid assay and dual luciferase analysis revealed that CpNAC56 activates the transcription of CpTPS18. Transient overexpression in vivo demonstrated that this gene could upregulate the expression of CpTPS18 and promote linalool accumulation. These results uncovered the primary volatile molecule responsible for papaya fruit odor and identified two major genes influencing its biosynthesis. Bergamot (Citrus bergamia Risso) is not consumed as fresh fruit as other members of Citrus. Bergamot essential oil has a unique aroma. Bergamot essential oil is a very valuable product used in perfumery, cosmetics, aromatherapy and food industry due to its beautiful scent and functional properties. As with other Citrus species, the composition of bergamot essential oil can vary according to harvest time, climate and extraction technology. Bergamot essential oil could be obtained by cold press extraction or distillation method. Within the scope of the study, the effect of four different hydrodistillation (HD) times (5, 10, 60 and 120 min) on the composition of bergamot essential oil obtained from Turkish fruits was investigated. In the first 60 min, 96.11% of the total essential oil was obtained. Research findings have shown that the distillation time is quite effective in the composition of bergamot essential oil. Limonene, linalool and linalyl acetate, which are the main components of bergamot essential oil, varied in the range, 42.36%–53.25%, 5.59%–12.18% and 20.84%–26.73%, respectively, with respect to distillation time. In cold press method, bergamot essential oil has lower linalool/linalyl acetate ratio (0.15) compared to HD method and a higher quality essence was obtained. [ABSTRACT FROM AUTHOR]

Diabetes mellitus (DM) is a prevalent metabolic disorder often accompanied by oxidative stress, which contributes to various diabetic complications. Investigating the antioxidant activity of linalool (LIN) is crucial as it may offer a natural therapeutic approach to mitigate oxidative damage in DM. The aim of the present study was to investigate the antioxidant activity of LIN in a DM rat model. A total of 40 male Wistar albino rats (age, 8 weeks; weight, 250-300 g) were used. CONTROL and DM groups were administered physiological saline solution by oral gavage for 21 days. In rats in the DM + LIN and LIN groups, 100 mg/kg LIN was administered intragastrically after streptozotocin injection (n=10 per group). In the first (48 h after STZ injection), second (1 week later), third (2 weeks later), and fourth (3 weeks later) blood glucose measurements, a statistically significant increase was found in the blood glucose values of the DM and DM + LIN groups compared with those of the CONTROL group. During the 21-day experimental period, there was no reduction in blood glucose levels of the DM + LIN group. Consequently, no discernible anti-hyperglycemic effect of LIN was observed. Catalase enzyme activity, superoxide dismutase (SOD) enzyme activity, malondialdehyde (MDA) levels and glutathione (GSH) levels were measured spectrophotometrically. All assays were conducted according to the protocols provided in the respective kits. The results were analyzed to assess the oxidative status and antioxidant capacity in the experimental groups. Catalase (CAT) activity was decreased in the DM group compared with that in the CONTROL group in both the serum and liver. However, LIN administration restored CAT activity in the DM + LIN group to the level of the CONTROL group. In the liver, the DM + LIN-treated group showed a notable reduction in malondialdehyde (MDA) levels compared with those in the DM group. In conclusion, the present results suggest that the antioxidant properties of LIN may have a regulatory effect on the oxidative status in diabetes-affected systems, potentially offering therapeutic benefits in managing oxidative stress associated with diabetes. Tribolium castaneum (Herbst) and Tribolium confusum (Jacquelin du Val) are common and damaging pests of stored grain flours in several homes and flour mill industries worldwide. This study examines how food, species, concentration, and exposure time affect the susceptibility and nutritional physiology of T. castaneum and T. confusum when exposed to partially purified methanolic fruit extract of Dennettia tripetala (G. Baker) and two of its major active compounds (linalool and 2‐phenylnitroethane). Gas chromatography–mass spectrometry was used to evaluate the chemical profile of the partially purified plant extract. Thirty‐two compounds were found in the plant, including established insecticidal and insectifuge compounds such as linalool, cis‐vaccenic acid, 2‐phenylnitroethane, and linoleic acid, among others. The interactions among the factors have a differential impact on the susceptibility and nutritional indices of both Tribolium species to D. tripetala, linalool and 2‐phenylnitroethane. According to LC50 values, linalool and 2‐phenylnitroethane were the most and least toxic to both beetles, respectively. With increasing extract concentrations, all nutritional indices except feeding deterrence decreased. Relative to controls, linalool and 2‐phenylnitroethane evoked the highest and least reduction, respectively, in beetles' nutritional indices. Treated corn discs deterred both species over treated wheat discs, regardless of insecticide type. T. castaneum had a higher overall food consumption than T. confusum. Regardless of insecticide type, the relative growth rate was higher in T. confusum and T. castaneum fed with treated corn and wheat discs, respectively. D. tripetala extract, linalool, and 2‐phenylnitroethane showed higher post‐ingestion toxicity to T. castaneum than T. confusum. This study emphasises the importance of certain intrinsic factors, like beetles' species and cereal food types, when managing Tribolium species with plant‐based insecticides. The various data obtained from this study will help researchers and pesticide manufacturers formulate effective biopesticides from linalool and 2‐phenylnitroethane to control Tribolium species on corn and wheat flours. In this study, we investigated the effects of the smell environment in the work booth on autonomic nervous activity (ANS) and psychomotor vigilance levels (PVLs) using linalool (LNL) and trans-2-nonenal (T2N). The subjects were six healthy males (31 ± 6 years old) and six healthy females (24 ± 5 years old). They sat in the work booth filled with the smells of LNL and T2N for 10 min, and their electrocardiograms (ECGs), skin conductance levels, pulse wave variabilities, skin temperatures, and seat pressure distributions were measured. In addition, the orthostatic load test (OLT) and psychomotor vigilance test (PVT) were performed before and after entering the work booth, and a subjective evaluation of the smell was also performed after the experiment. This paper focused on ECG and PVT data and analyzed changes in heart rate variability indices and PVT scores. Males felt slightly comfortable with the LNL smell and showed promoted sympathetic nerve activity in the OLT after the smell presentation. Females felt slightly uncomfortable with the T2N smell and showed promoted sympathetic nerve activity and a decrease in PVT scores in the OLT after the smell presentation. Gender differences were observed in ANS and PVLs, and it is possible that the comfort of LNL increased sympathetic nervous activity in males, while the uncomfortableness of T2N may have reduced work performance in females. The perfume industry in various countries is increasingly expanding and the appeal of perfume comes from its distinctive scent. One of Indonesia's potential needs in the organic perfume industry is essential oils. Nampu rhizome (Homalomena occulta) has great potential and is widely spread on the island of Java. This is because the Nampu rhizome has the main chemical component in the form of linalool compounds, which are essential oil components with high economic value, so it needs to improve quality by increasing its purity. A technology that was developed to improve the purity of essential oils was vacuum fractionation distillation. The purpose of the study was to analyze the effect of temperature on the quality of distillation in the fractionation results of Nampu rhizome essential oil (linalool compound). The experiment used a material temperature factor between 135-160°C and > 160°C.The analysis of fractionation results was carried out on physical properties including refractive index, specific gravity, optical rotation, colour, and linalool compound with GC-FID and yield. The results showed a refractive index value of 1.4620 nD, specific gravity of 0.8647, optical rotation of -19.80, clear oil colour and yield of 69.80%. Chemical analysis with GC-FID improves the purity level of the final product against the linalool fraction increased by 10.02% or linalool purity by 90.92%.

Abstract Pichia kudriavzevii (formerly Candida krusei) poses a significant threat to immunocompromised patients due to its inherent resistance to various antifungal drugs. This study explored the anticandidal potential of citral, linalool, and carvacrol in combination with nystatin against P. kudriavzevii strains. Using the microdilution method following CLSI guidelines, Minimum Inhibitory Concentrations (MICs) and fungicidal concentrations (MFCs) were determined. Citral exhibited MIC values ranging from 50 to 100 µg/ml, averaging 70.24 ± 16.99 µg/ml, while carvacrol had MIC values of 50 to 100 µg/ml, averaging 86.90 ± 16.99 µg/ml. Linalool demonstrated weaker antifungal activity, with MIC values between 100 and 200 µg/ml, averaging 150 ± 38.73 µg/ml. The study assessed the synergistic effectsof these phenols with nystatin through fractional inhibitory concentration indices (FICIS). In addition, flow cytometry was employed to assess apoptosis induction in P. kudriavzevii cells. Carvacrol displayed a remarkable synergistic effect in combination with nystatin against all 21 isolates tested. Conversely, linalool showed synergy in 17 isolates, while citral exhibited synergy in only 2 isolates. These findings highlight distinct patterns of synergy between the different compounds and nystatin against P. kudriavzevii. Also, Carvacrol emerged as the most potent inducer of apoptosis across all P. kudriavzevii strains, followed by citral and linalool. This suggests that carvacrol not only possesses a stronger antifungal effect but also has a more pronounced ability to trigger programmed cell death in P. kudriavzevii. In conclusion, the study supports the potential of carvacrol, citral and linalool, as anticandidal agents, suggesting their supplementation with nystatin for treating P. kudriavzevii infections.

An imbalance between reactive oxygen species (ROS) production and antioxidant defense driven by oxidative stress and inflammation is a critical factor in the progression of neurodegenerative diseases such as Alzheimer's and Parkinson's. Coriander (Coriandrum sativum L.), a culinary plant in the Apiaceae family, displays various biological activities, including anticancer, antimicrobial, and antioxidant effects. Herein, neuroprotective properties of three major bioactive compounds derived from coriander (i.e., linalool, linalyl acetate, and geranyl acetate) were investigated on hydrogen peroxide-induced SH-SY5Y neuroblastoma cell death by examining cell viability, ROS production, mitochondrial membrane potential, and apoptotic profiles. Moreover, underlying mechanisms of the compounds were determined by measuring intracellular sirtuin 1 (SIRT1) enzyme activity incorporated with molecular docking. The results showed that linalool, linalyl acetate, and geranyl acetate elicited their neuroprotection against oxidative stress via protecting cell death, reducing ROS production, preventing cell apoptosis, and modulating SIRT1 longevity. Additionally, in silico pharmacokinetic predictions indicated that these three compounds are drug-like agents with a high probability of absorption and distribution, as well as minimal potential toxicities. These findings highlighted the potential neuroprotective linalool, linalyl acetate, and geranyl acetate for developing alternative natural compound-based neurodegenerative therapeutics and prevention. In the technology for extracting the required components from plant raw materials, in most cases, the grinding process is used as a preparatory operation before the main process, followed by the separation of the extracted material into target components.In most cases, as a result of such a process, we can talk about the effective extraction of only a single target component, while others inevitably lose their qualitative and quantitative indicators, which is due to the morphology of the plant raw material.One of the promising directions in the processes of destruction of plant material for further operations is selective disintegration. For this purpose optimal process conditions have been selected, such as heating, freezing and others. Raw materials obtained as a result of selective destruction can be effectively fractionated according to morphological and physicochemical characteristics, and only after that can be sent to subsequent operations with the greatest efficiency.Under modern conditions, the loss of essential oil in coriander associated with splitting the fruit reaches 23.4% of the oil weight in whole fruits. At the same time, the essential oil from split fruits is enriched with valuable components – linalool, geraniol, geranyl acetate and, in addition, contains less hydrocarbons and camphor. As shown in [1], oil from split fruits can be used to adjust the composition of batches of coriander essential oil in order to increase the content of linalool while simultaneously reducing the content of undesirable components – hydrocarbons and camphor, and can also be used primarily to isolate valuable components – linalool, geraniol. To reduce losses associated with splitting coriander fruits, it has been proposed to carry out mathematical modeling of the process of cryogenic freezing with subsequent grinding of the frozen mass, which will significantly reduce losses of essential oil. Considering that essential oils extracted from coriander show high antibacterial, antioxidant and antifungal activity [2], increasing the yield of high-quality coriander essential oil will allow its wider use in flavoring and food preservation, as well as for medicinal purposes, which is important and actual task.

Abstract Recent years, the rapid advancement of technology has raised concerns. We studied the effects of prenatal exposure to 900 MHz radiofrequency (RF) from mobile phones and the protective effects of linalool on learning and memory, and anxiety in adolescent male and female offspring rats. Pregnant rats were divided into four groups: control, wave, wave + linalool, and linalool. Rats received linalool (25mg/kg) by gavage for 21 days. Irradiation was conducted from day 0 to day 21 of pregnancy. Offsprings underwent behavioral and electrophysiological tests on days 50 and 60 after birth. Exposure to RF during pregnancy caused anxiety-like behavior in the EPM test and impairment of learning and memory in the Morris water maze and shuttle box tests. Electrophysiological properties and synaptic plasticity of the dorsal hippocampal CA3-CA1 synapse showed a decrease in fEPSP amplitude and slope. The trace element levels in both male and female offspring were consistent across all groups compared to their respective controls. In the hippocampus tissue, the levels of Fe, Cu, and Mn, as well as the Cu/Zn ratio, were significantly higher in the exposed groups (wave groups) compared to their controls. Moreover, Zn levels were significantly lower in the hippocampus tissue of the exposed groups. Linalool administration mitigated the excessive increase in Fe, Cu, Mn, and Cu/Zn ratio and normalized the disrupted levels of trace elements, except for Zn levels in both male and female offspring. Sex differences were observed in the EPM and shuttle box tests, females were more sensitive than males. In summary, our study demonstrates that prenatal exposure to mobile phone radiation induces stress-like behaviors, disrupts learning and memory, alters hippocampal electrophysiological properties and trace element balance in offspring. Treatment with linalool mitigates these deleterious effects, highlighting its potential as a therapeutic intervention. These findings contribute to our understanding of the impact of prenatal environmental exposures on neurodevelopment and offer insights into potential strategies for neuroprotection.

Basil (Ocimum basilicum L.) is a vital aromatic crop widely utilized in culinary and fragrance industries. The present study was conducted to implement a two-year field experiment (2019 and 2020) aimed to assess the impact of various fertilizers on the growth, yield, and essential oil composition of basil cultivated in Thanh Hoa Province, Vietnam. The fertilizer treatments included: no fertilizer (control), chemical fertilizer (NPK at 90-120-90 kg/ha), cow manure, chicken manure, and a combination of chemical fertilizer, cow manure, and chicken manure. The application of fertilizers resulted in significant improvements across various parameters compared to the control group. Basil plants treated with fertilizers exhibited increased height, la teral stem count, fresh and dry yields, chlorophyll and carotenoid content, essential oil yield, and essential oil content. The essential oil extracted from basil contained notable volatile compounds, with methyl chavicol, linalool, β-elemene, and epi-α-cadinol identified as the major constituents. The proportions of these compounds varied among the fertilizer treatments. Notably, the combined fertilization approach involving chemical fertilizers, cow manure, and chicken manure emerged as the most effective and significant in promoting basil growth and essential oil production. These findings not only contribute to the optimization of basil cultivation practices but also provide insights for the agricultural community, emphasizing the importance of balanced fertilization for maximizing the quality and yield of basil essential oil in the Vietnamese context.

Background: Zanthoxylum bungeanum Maxim. is widely distributed across China, and the aroma of its peel is primarily determined by its volatile components. In this study, we analyzed the characteristics of volatile components in Z. bungeanum peels from different regions and investigated their correlation with climatic factors. Results: The results identified 126 compounds in Z. bungeanum, with 27 compounds exhibiting distinct odor characteristics. Linalool was the most abundant, with an average relative content of 21.664%. The volatile oil of Z. bungeanum predominantly features spicy, floral, citrus, and mint aromas. The classification results indicated a significant difference in elevation at the ZB10 collection points in Shaanxi Province compared to other groups. Temperature, average annual precipitation, and wind speed were crucial factors influencing the accumulation of volatile components. Conclusions: This study is beneficial for enhancing the quality of Z. bungeanum, expanding the understanding of how climatic factors influence the accumulation of volatile substances, and promoting agricultural practices in regions with similar climatic conditions. Pseudomonas fragi (P. fragi) is usually detected in low-temperature meat products, and seriously threatens food safety and human health. Therefore, the study investigated the antibacterial mechanism of linalool against P. fragi from membrane damage and metabolic disruption. Results from field-emission transmission electron microscopy (FETEM) and atomic force microscopy (AFM) showed that linalool damage membrane integrity increases surface shrinkage and roughness. According to Fourier transform infrared (FTIR) spectra results, the components in the membrane underwent significant changes, including nucleic acid leakage, carbohydrate production, protein denaturation and modification, and fatty acid content reduction. The data obtained from amino acid metabolomics indicated that linalool caused excessive synthesis and metabolism of specific amino acids, particularly tryptophan metabolism and arginine biosynthesis. The reduced activities of glucose 6-phosphate dehydrogenase (G6PDH), malate dehydrogenase (MDH), and phosphofructokinase (PFK) suggested that linalool impair the respiratory chain and energy metabolism. Meanwhile, genes encoding the above enzymes were differentially expressed, with pfkB overexpression and zwf and mqo downregulation. Furthermore, molecular docking revealed that linalool can interact with the amino acid residues of G6DPH, MDH and PFK through hydrogen bonds. Therefore, it is hypothesized that the mechanism of linalool against P. fragi may involve cell membrane damage (structure and morphology), disturbance of energy metabolism (TCA cycle, EMP and HMP pathway) and amino acid metabolism (cysteine, glutamic acid and citrulline). Oral thrush and throat infections can occur in a wide range of patients. Treatments are available; however, resistance to drugs is a major problem for treating oral and throat infections. Three-dimensional printing (3DP) of fast dissolving oral films (FDFs) of linalool oil may provide an alternative solution. Linalool oil FDFs were printed by fused deposition modelling across 1–18 % w/w linalool content range with nozzle diameters of 0.4 or 1 mm at the temperature range of 150°C-185°C. The FDFs were evaluated for physicochemical and mechanical properties. Increasing the printer nozzle diameter to 1 mm allowed reducing the printing temperature from 185°C to 150°C; consequently, more linalool was quantified in the films with improved content uniformity. The higher linalool content in the films increased the film disintegration time and mechanical strength. FDFs containing 10% w/w linalool showed clear antifungal activity against Candida albicans. Raman spectroscopy suggested linalool separation from excipients at higher temperature printing. Viscoelastic measurements indicated that to achieve printing; the elastic modulus of molten filament needed to be higher than the loss modulus at low angular frequencies. This work used headspace solid-phase microextraction with gas chromatography–mass spectrometry (HS-SPME-GC–MS) to analyze the volatile components of hydrosols of Citrus × aurantium 'Daidai' and Citrus × aurantium L. dried buds (CAVAs and CADBs) by immersion and ultrasound–microwave synergistic-assisted steam distillation. The results show that a total of 106 volatiles were detected in hydrosols, mainly alcohols, alkenes, and esters, and the high content components of hydrosols were linalool, α-terpineol, and trans-geraniol. In terms of variety, the total and unique components of CAVA hydrosols were much higher than those of CADB hydrosols; the relative contents of 13 components of CAVA hydrosols were greater than those of CADB hydrosols, with geranyl acetate up to 15-fold; all hydrosols had a citrus, floral, and woody aroma. From the pretreatment, more volatile components were retained in the immersion; the relative contents of linalool and α-terpineol were increased by the ultrasound–microwave procedure; and the ultrasound–microwave procedure was favorable for the stimulation of the aroma of CAVA hydrosols, but it diminished the aroma of the CADB hydrosols. This study provides theoretical support for in-depth exploration based on the medicine food homology properties of CAVA and for improving the utilization rate of waste resources. Citrus species have undergone immense diversification ever since their ancestral origin. Ponkan and Murcott are two mandarin varieties widely consumed in Brazil and their aroma producing active compounds have not yet been extensively investigated. The present study analyzed the volatile constituents of the Ponkan and Murcott varieties employing the stir bar sorptive extraction (SBSE) technique and gas chromatography‐mass spectrometry (GC‐MS) analysis for the first time. Extraction was performed using the two phases of Twister bar, polydimethyl siloxane (PDMS), and ethylene glycol (EG) silicone in immersion and headspace modes. Among the detected 62 compounds comprising alcohols, aldehydes, esters, terpenes, and others identified, 55 and 37 compounds in the Ponkan and Murcott variety were determined, respectively, from both immersion and headspace modes using the two phases of Twister bar. From the odor active values, the Ponkan was characterized with the domination of compounds such as hexanal, decanal, nonanal, (E)‐2‐hexanal, ethyl hexanoate, d‐limonene, linalool, and geraniol. Similarly, the Murcott variety was characterized with predominant compounds, namely, nonanal, octanal, hexanal, (E)‐2‐hexanal, ethyl hexanoate, d‐limonene, and linalool. The profile of volatile compounds was found to be unique in both the varieties studied, and SBSE technique with GC‐MS analysis favored the characterization of their respective profile due to the high amount of phase in the Twister bars, when compared to similar techniques, like solid phase microextraction. The PDMS Twister bar showed better capacity of adsorption of volatile compounds, since it is a relatively low‐polarity polymer, which discriminates less analytes with different structures and polarities sampled from complex matrices, such as fruits juice. The clinical use of gentamicin (GM) is restricted by its nephrotoxic effects. This study aimed for the first time to elucidate the ameliorative effects of the monoterpene linalool (Lin) against GM-mediated acute kidney injury in rats. A total of thirty-two rats were subdivided into four equal groups: control (saline), Lin (100 mg/kg/day), GM (100 mg/kg/day), and GM + Lin (100 and 100 mg/kg/day). Lin and GM were intraperitoneally administered for 12 days. Our results illustrated that Lin ameliorated GM-mediated renal histopathological abnormalities and reduced serum urea and creatinine levels in rats exposed to GM. Lin treatment mitigated oxidative stress in nephrotoxic animals as manifested by reducing serum and renal levels of malondialdehyde and increasing the activities of serum and renal glutathione peroxidase and renal catalase. Moreover, Lin markedly inhibited GM-triggered inflammation by downregulating NF-κB, iNOS, TNF-α, and IL-1β and reducing renal myeloperoxidase activity and nitric oxide levels. Interestingly, Lin repressed GM-induced apoptosis, as reflected by a marked downregulation of Bax and caspase-3 expression, concurrent with the upregulation of Bcl2 expression. Finally, Lin administration led to a significant downregulation of TGF-β expression in nephrotoxic animals. Leaf spreading is a key processing step that affects the aroma formation of green tea. The effects of a single-light wavelength on the aroma and taste of tea have been extensively studied. Less attention has been paid to the effect of different complex light intensities on the formation of green tea's volatile aroma during leaf spreading. The current study was designed to evaluate how leaf spreading under different complex light intensities relates to the quality of green tea. Using headspace solid-phase micro-extraction and gas chromatography-mass spectrometry (HS-SPME/GC-MS), volatile flavor compounds in green tea were analyzed during leaf spreading in five different light conditions. Multivariate statistical analysis and odor activity values (OAVs) were used to classify these samples and identify key odors. Eight distinct groups, including ninety volatile compounds, were detected. The most prevalent volatile compounds found in green tea samples were hydrocarbons and alcohols, which accounted for 29% and 22% of the total volatile compounds, respectively. Fourteen volatile compounds (OAV > 1) were identified as key active differential odorants. The chestnut-like aroma in green tea was mostly derived from 3-methyl-butanal and linalool, which were significantly accumulated in medium-intensity light (ML). [ABSTRACT FROM AUTHOR]

Parkinson's disease is one of the most prevalent neurological disorders affecting millions of people worldwide. There is a growing demand for novel and natural substances as complementary therapies. Essential oils and their various compounds are highly investigated natural plant-based products as potential treatment options for common human diseases, such as microbial infections, chronic diseases, and neurodegenerative disorders. The present study focuses on the beneficial effects of linalool and geraniol, the major compounds of lavender (Lavandula angustifolia L.) and geranium (Pelargonium graveolens L'Hér. in Aiton) essential oils, on oxidative stress, inflammation, and iron metabolism of the rotenone and 6-hydroxydopamine-induced in vitro Parkinson's models. The experiments were carried out on all-trans retinoic acid differentiated SH-SY5Y cells. The effects of linalool and geraniol were compared to rasagiline, an MAO-B inhibitor. The results revealed that both essential oil compounds reduce the level of reactive oxygen species and alter the antioxidant capacity of the cells. They lower the secretion of IL-6, IL-8, and IL-1β pro-inflammatory cytokines. Moreover, linalool and geraniol change the expression of iron-related genes, such as the iron importer transferrin receptor 1, heme-oxygenase-1, and ferroportin iron exporter, and influence the intracellular iron contents. In addition, it has been unveiled that iron availability is concatenated with the actions of the essential oil compounds. Sodium chloride (NaCl) can cause oxidative stress in plants, which represents a potential obstacle to the development of monocultures worldwide. Onion (Allium cepa L.) is a famous vegetable consumed and used in world cuisine. In the present study, we analyzed the influence of soil physicochemical profile and the remedial capacity of linalool on seed emergence, roots, and leaf growth in onions subjected to salt stress, as well as its in vivo and in vitro antioxidant potential, Fe2+chelating activity, and reducing power of Fe3+. The outcome of the soil analysis established the following order of abundance: sulfur (S) > calcium (Ca) > potassium (K) > magnesium (Mg) > sodium (Na). NaCl (150 mM) significantly reduced the emergence speed index (ESI), leaf and root length, while increasing the peroxidation content. The length of leaves and roots significantly increased after treatment with linalool (300 and 500 μg/mL). Our data showed negative correlations between seed emergence and K+ concentration, which was reversed after treatments. Linalool (500 μg/mL) significantly reduced oxidative stress, but increased Fe2+ concentration and did not show potential to reduce Fe3+. The in vivo antioxidant effect of linalool is thought to primarily result from an enzymatic activation process. This mechanism underscores its potential as a therapeutic agent for oxidative stress–related conditions. Further investigation into this process could unveil new avenues for antioxidant therapy. The essential oil obtained from the dry leaves of Gynoxys szyszylowiczii Hieron. was described in this study for the first time. The chemical analysis, conducted on two stationary phases of different polarity, permitted to identify sixty-four compounds, that were quantified with at least one column. The main components, on a non-polar and polar stationary phase respectively, were germacrene D (21.6–19.2%), α-pinene (4.4–4.9%), n-tricosane (4.3% on both columns), (E)-β-caryophyllene (3.3–4.3%), 1-docosene (3.2–2.8%), α-cadinol (2.8–3.1%), and cis-β-guaiene (2.6–3.0%). This investigation was complemented by the enantioselective analysis of some major chiral compounds, carried out on two chiral selectors based on β-cyclodextrines. As a result, (S)-(+)-α-phellandrene, (S)-(+)-β-phellandrene, and (1S,2R,6R,7R,8R)-(+)-α-copaene appeared enantiomerically pure, whereas α-pinene, β-pinene, terpinen-4-ol, and germacrene D were detected as scalemic mixtures. Developing effective and targeted drug delivery systems is crucial in searching for improved pain management strategies. Here, it is shown the preparation of a nanocarrier using chitosan hydrogel, and polymeric nanocapsules loaded with linalool for enhancing transdermal permeation and targeted delivery of this antinociceptive monoterpene for the pain treatment. Extensive characterization was conducted to evaluate the properties of the nanocarrier, and several in vitro studies were performed to investigate the release kinetics of linalool from the nanocarrier and its permeation through the layers of the skin. As results, the prepared polymeric nanocarrier exhibited an average size of 160 ± 9 nm, with a polydispersion index of 0.12 ± 0.01. The kinetic study revealed that the nanocarrier effectively controlled and prolonged the release of linalool, following a pseudo‐second order model (R2 = 0.98). To evaluate the permeation of the nanocarrier through the transdermal barrier, swine ear skin was employed. The nanocarrier efficiently penetrated the transdermal barrier and successfully delivered linalool to the skin layers. Additionally, an in vivo toxicity study indicated no toxicity for the nanocarrier at the tested concentrations (<0.950 μg mL−1). This investigation undertakes a thorough examination of the thermophysical properties of linalool (C10H18O) blended with a series of 2-alkanols (ranging from 2-propanol to 2-hexanol) across a temperature spectrum of 293.15 K to 323.15 K. The primary objective of this study is to elucidate the density and viscosity behavior of these binary systems. The experimental results reveal negative deviations from ideality in excess molar volume for linalool with 2-propanol up to 2-pentanol, and positive values for linalool with 2-hexanol, accompanied by negative viscosity deviations across all examined mixtures. The observed negative excess volume suggests the presence of strong intermolecular interactions between linalool and the 2-alkanols, which is indicative of the formation of hydrogen bonds in the mixtures. The positive values of excess molar volumes in linalool and 2-hexanol are indicative of the governing of steric hindrance effects on attractive intermolecular forces. Moreover, the Friction theory (f-theory) was utilized to model the viscosity of the binary mixtures, yielding an excellent concordance with the experimental data, with a maximum discrepancy of merely 2.25 % observed in the linalool + 2-pentanol system. This negligible deviation underscores the efficacy of the f-theory in accurately capturing the viscosity measurements, thereby validating its applicability in predicting the rheological behavior of such binary systems. Several Uvariodendron species are known for their scented leaves and stem bark, hence this study. The leaves, twigs, stem and root barks of the newly described Uvariodendron species, U. mbagoi Dagallier and Couvreur from Kwedijela forest (KF) and Kimboza Forest Reserve (KFR) collected in dry and wet seasons, were steam distilled to yield essential oils that was more abundant in the root bark (2.07-3.85%) than in the leaves, twigs and stem bark (0.09-0.71%). The essential oils from the root bark collected from KF in the wet season (2.07%) was slightly lower than that obtained from the plant part collected from the same locality during the dry season (3.85%). GC-MS analysis indicated that linalool was the most prominent constituent of the leaf (82.18%, KF) and (98.86%, KFR), twig (89.66%, KFR), and stem bark (97.97%, KFR) oil while methyl eugenol (41.08%, KFR) and elemicin (39.28%, KFR) were the major constituents of the root bark oil. The essential oil from the leaves, stem bark, twigs and root bark of U. mbagoi exhibited mild to moderate potency when screened for antioxidant, antibacterial, and protease (trypsin) inhibitory activities. Employing the Molecular Electron Density Theory, [3 + 2] cycloaddition processes between 4-chlorobenzenenitrileoxide and linalool, have been applied using the DFT/B3LYP/6–311(d,p) method, activation, reaction energies and the reactivity indices are calculated. In an investigation of conceptual DFT indices, LIL-1 will contribute to this reaction as a nucleophile, whilst NOX-2 will participate as an electrophile. This cyclization is regio, chemo and stereospecific, as demonstrated by the reaction and activation energies, in clear agreement with the experiment's results, in addition, ELF analysis revealed that the mechanism for this cycloaddition occurs in two steps. Furthermore, a docking study was conducted on the products studied, and the interaction with the protein protease COVID-19 (PDB ID: 6LU7), our results indicate that the presence of the –OH group increases the affinity of these products, moreover, adsorption study by chromatography was made on silica gel as support; our outcome reveals that the –OH group creates an intramolecular hydrogen bond in the product P2, while in the product P3 will create a hydrogen bond with the silica gel which makes the two products P2 and P3 are very easy to separate by chromatography, this result is in excellent agreement with the Rf retention value. The current study examines the effects of linalool in preventing postoperative abdominal adhesions. Twenty male Wistar rats were randomly divided into four groups. (1) Sham: in this group, the abdomen was approached, and without any manipulations, it was sutured. (2) Control: rats in this group underwent a surgical procedure to induce adhesions. This involved making three incisions on the right abdominal side and removing a 1×1-cm piece of the peritoneum on the left abdominal side. (3) Treatment groups: these groups underwent the same surgical procedure as the control group to induce adhesions. Animals in these groups received linalool orally with doses of 50 and 100 mg/kg, respectively, for a period of 14 days. Moreover, rats in the sham and control groups received normal saline via gavage for 14 days. The evaluation of TNF-α, TGF-β, VEGF, and caspase 3 was performed using western blot and IHC methods. Furthermore, oxidative stress biomarkers such as MDA, TAC, GSH, and NO were assessed in the peritoneal adhesion tissue. The findings revealed that linalool significantly reduced peritoneal adhesions by reducing TNF-α, TGF-β, VEGF, and caspase 3 levels. Moreover, MDA concentration was significantly decreased, while NO, TAC, and GSH levels were notably increased. Overall, linalool was effective in preventing adhesion formation and reduced inflammation, angiogenesis, apoptosis, and oxidative stress. Therefore, linalool as a potent antioxidant is suggested for reducing postoperative adhesions in rats. [ABSTRACT FROM AUTHOR]

Main conclusion: One of seven Solanum taxa studied displayed associations between pollen presence and floral scent composition and volume, suggesting buzz-pollinated plants rarely use scent as an honest cue for foraging pollinators. Floral scent influences the recruitment, learning, and behaviour of floral visitors. Variation in floral scent can provide information on the amount of reward available or whether a flower has been visited recently and may be particularly important in species with visually concealed rewards. In many buzz-pollinated flowers, tubular anthers opening via small apical pores (poricidal anthers) visually conceal pollen and appear similar regardless of pollen quantity within the anther. We investigated whether pollen removal changes floral scent composition and emission rate in seven taxa of buzz-pollinated Solanum (Solanaceae). We found that pollen removal reduced both the overall emission of floral scent and the emission of specific compounds (linalool and farnesol) in S. lumholtzianum. Our findings suggest that in six out of seven buzz-pollinated taxa studied here, floral scent could not be used as a signal by visitors as it does not contain information on pollen availability. Cotton, a crucial economic crop, is also the preferred host plant of the mirid bug Apolygus lucorum. In our previous field experiments, we found that cotton cultivars Kelin 08–15 and BR-S-10 (healthy and herbivore-damaged plants) exhibit distinct attraction and repellence to A. lucorum, respectively. However, the key plant volatiles determining attraction or repulsion effects remain unknown. Here, we investigated the volatiles emitted by these two cotton cultivars before and after herbivore infestation. We found that susceptible Kelin 08–15 emitted a greater diversity and quantity of volatiles than those of BR-S-10, with herbivore-damaged cottons releasing more volatile substances. Electroantennogram (EAG) recordings further revealed that 15 representative volatiles identified above could elicited electrophysiological responses in female and male A. lucorum antennae. Among them, behavioral assays showed that two compounds, 1,3-Diethylbenzene and 4-Ethylbenzaldehyde, exhibited attractive properties, whereas six volatiles including Hexyl Acrylate, Cumene, 2,4-Dimethylstyrene, Eucalyptol, Linalool and Butyl Acrylate demonstrated repellent effects on A. lucorum. Taken together, our findings suggest the critical role of volatile compounds in mediating bug-plant interactions and provide a foundation for the development of strategies to prevent and control of A. lucorum in cotton fields. This study evaluated the pharmacokinetic profile of basil (Ocimum basilicum) essential oil (EO) in juvenile tambaqui (Colossoma macropomum). In trial I, juvenile tambaqui (119.9 ± 9.2 g) were exposed to 800 mg L−1 of EO for 10 min for anesthetic induction. After the blood samples, muscle and brain tissue samples were collected at 2, 4, 6, 8, and 10 min to determine the absorption time and distribution of methyl chavicol and linalool, the major compounds in the EO, the concentration of lactate and ammonia in the muscles, and the plasma levels of lactate, chloride, ammonia, and total calcium. In trial II, anesthetic induction occurred at the same time and concentration and blood, muscle, and brain tissue samples were collected at 0.0, 0.5, 1.0, 3.0, and 6.0 h to quantify the elimination of methyl chavicol and linalool and for a histological evaluation of the gills, liver, and spleen. In assay I, both compounds were identified in the plasma, muscle, and brain as early as 2 min, with maximum concentrations observed within 6 min. In assay II, the half-life of methyl chavicol and linalool in plasma was 0.30 h and 0.46 h, respectively. The half-life of methyl chavicol and linalool in muscle was 1.59 h and 0.32 h, respectively, and the half-life of methyl chavicol and linalool in the brain was 0.69 h and 0.33 h, respectively. The histological lesions observed in the gills between 0 and 3 h were reversible, with organ recovery 6 h after anesthesia induction. There were no changes in the liver and spleen during the times evaluated. The biochemical and pharmacokinetic results in this study confirm that the EO of O. basilicum at 800 mg L−1 is a safe anesthetic for tambaqui, inhibits the stress response, and is eliminated rapidly from the fish. Breast cancer (BC) is a common cancer for women. This study aims to construct a prognostic risk model of BC and identify prognostic biomarkers through machine learning approaches, and clarify the mechanism by which linalool exerts tumor‐suppressive function. Three mRNA microarray/RNA sequencing data sets (GSE25055, GSE103091, and TCGA‐BRCA) were obtained from Gene Expression Omnibus database and The Cancer Genome Atlas database, and prognostic genes were obtained by univariate COX analysis. Multiple machine learning methods were used to screen core genes and construct prognostic risk models. The enrichment analysis of crucial genes was analyzed using the DAVID database. UALCAN, human protein atlas, geneMANIA, and LinkedOmics databases were used to analyze gene expression and co‐expressed genes. Molecular docking and molecular dynamics simulation was applied to verify the binding affinity between linalool and phosphoglycerate kinase 1 (PGK1). Cell counting kit 8 (CCK‐8, Edu, transwell, flow cytometry, and Western blot assay were used to analyze cell activity, apoptosis, cell cycle and protein expression. Eight prognostic genes were obtained by bioinformatics analysis and machine learning, and prognostic risk models were constructed. This model could well predict the prognosis of patients, and the risk score could be used as an independent risk factor for BC. Overall survival (OS) and immune cell infiltration characteristics were distinct between high and low risk groups. PGK1 was highly expressed in BC and the OS of patients with high PGK1 expression was shorter. PGK1 was related to cell cycle and PPAR signaling pathway. Linalool and PGK1 had good binding activity, and linalool could inhibit the viability, proliferation, migration, and invasion of BC cells, promote cell apoptosis, and induce G0/G1 arrest. In addition, linalool can promote PPARγ protein expression and inhibit PGK1 expression. Machine learning and molecular docking were promising for exploration of new drug targets for BC, and linalool exerts tumor‐suppressive effects in BC by inhibiting PGK1 expression and activating PPAR signaling pathway. Variations in seed yield and essential oil composition of 48 coriander (Coriandrum sativum L.) germplasms collected from various geographical locations in India were evaluated. The variation in seed yield plant−1 (2.52–10.97 g plant−1), essential oil content (0.08–0.56%) and linalool content (49.04–82.14%). Linalool, γ-terpinene, β-pinene, p-cymene, β-myrcene, limonene, geraniol, caryophyllene, geranyl acetate, α-terpineol, citronellol and caryophyllene were detected in one or other germplasm lines. Cluster analysis was done using Ward's method based on the value of all the traits, resulting in 6 distinct groups of germplasms. Among six clusters, the maximum number of accessions was grouped into cluster II (16). The seed yield plant−1 is positively correlated with the number of umbels plant−1, number of umbellets plant−1, number of seeds plant−1, number of primary and secondary branches. The selection of these traits may result in the identification of germplasm with high seed yield. Rhanterium epapposum Oliv. (Family: Asteraceae) is a common plant in Saudi Arabia, possessing a variety of traditional uses. The current targets were studying the comparative chemical profile and antioxidant of the essential oil (EO) of R. epapposum collected from a sandy habitat at Qareenah, Riyadh, Saudi Arabia. The EO profiling was constructed via gas chromatography-mass spectrometry (GC-MS) and GC-flame ionization detection (GC-FID). Forty-eight compounds were found, including monoterpenes (hydrocarbons 3.25% and oxygenated 48.64%), sesquiterpenes (hydrocarbons 12.64 % and oxygenated 32.38%), which are the backbone of the oil along with a minor of other non-terpenoid compounds (1.31%). Linalool (18.02%), α-eudesmol (11.98%), trans-sesquisabinene hydrate (11.63%), aromadendrene oxide-(1) (7.68%), nerol (6.88%), and α-terpineol (5.01%) were identified as the dominant components. The EO was evaluated for its antioxidant activity using in vitro DPPH, ABTS, and FRAP methods. The results demonstrated that R. epapposum EO possesses a dosage-dependent ability for antioxidant scavenging activity as per all three techniques. This EO exhibited IC50 values of 517.59, 350.24, and 713.72 μg/mL for the DPPH, ABTS, and FRAP, respectively, through the comparison with the ascorbic acid with respective values of 36.10, 26.12, and 44.58 μg/mL. The main compounds were subjected to docking analysis on cytochrome P450 1A1 and xanthine oxidase. All evaluated compounds exhibited good binding affinities especially α-eudesmol and linalool for both enzymes, respectively. The current research confirmed the potent free radical scavenging properties of R. epapposum EO. More research on the antioxidant content of this oil and/or its primary constituents in pure form on the two enzymes, cytochrome P450 1A1 and xanthine oxidase, is proposed to develop a better understanding of the oxidative action routes in vivo and in vitro.
