Abstract

Dynein light chain LC8 is the smallest subunit of the dynein motor complex and has been shown to play important roles in both dynein-dependent and dynein-independent physiological functions via its interaction with a number of its binding partners. It has also been linked to pathogenesis including roles in viral infections and tumorigenesis. Structural information for LC8-target proteins is critical to understanding the underlying function of LC8 in these complexes. However, some LC8-target interactions are not amenable to structural characterization by conventional structural biology techniques owing to their large size, low solubility, and crystallization difficulties. Here, we report magic-angle spinning (MAS) NMR studies of the homodimeric apo-LC8 protein as a first effort in addressing more complex, multi-partner, LC8-based protein assemblies. We have established site-specific backbone and side-chain resonance assignments for the majority of the residues of LC8, and show [TALOS.sub.+]-predicted torsion angles φ and ψ in close agreement with most residues in the published LC8 crystal structure. Data obtained through these studies will provide the first step toward using MAS NMR to examine the LC8 structure, which will eventually be used to investigate protein-protein interactions in larger systems that cannot be determined by conventional structural studies. La chaine legere de dyneine, LC8, est la sous unite la plus petite du complexe moteur de la dyneine et il a ete demontre qu'elle joue des roles importants dans les fonctions physiologiques tant dependantes qu'independantes de la dyneine par le biais de son interaction avec un certain nombre de ses partenaires de fixation. Elle a aussi ete reliee a des roles de tumorigenese et de pathogenese dans les infections virales. De l'information sur la structure des proteines cibles de la LC8 est critique pour comprendre la fonction sous-jacente de la LC8 dans ces complexes. Toutefois, en raison de leur grande taille, de leur faible solubilite et de difficultes de cristallisation, certaines interactions entre la LC8 et ses cibles ne peuvent pas etre amenees a une caracterisation structurelle par les techniques conventionnelles de la biologie. Dans ce travail, comme premier effort en vue de comprendre les assemblages de proteines complexes, a base de LC8 avec plusieurs partenaires, on rapporte des etudes de RMN du [sup.13]C a l'etat solide dans le mode de la rotation a l'angle magique (RAM) faites sur la proteine homodimere apo-LC8. On a etabli les attributions des resonances du squelette et de la chaine laterale specifique au site pour la majorite des residus de la LC8 et on montre que les valeurs predites par [TALOS.sub.+] pour les angles de torsion φ et ψ sont en bon accord avec celles publiees pour la plupart des residus dans des structures cristallines de la LC8. Les donnees obtenues par le biais de ces etudes serviront de premier pas dans l' utilisation de la RMN en mode RAM pour examiner la structure de la LC8 et qui pourra eventuellement etre utilisee pour etudier les interactions proteine-proteine dans des systemes plus gros qui ne peuvent pas etre determinees par des etudes structurales conventionnelles. Abstract

Pak1 (p21 activated kinase 1) is a serine/threonine kinase implicated in regulation of cell motility and survival and in malignant transformation of mammary epithelial cells. In addition, the dynein light chain, LC8, has been described to cooperate with Pak1 in malignant transformation of breast cancer cells. Pak1 itself may aid breast cancer development by phosphorylating nuclear proteins, including estrogen receptor alpha. Recently, we showed that the LC8 binding site on Pak1 is adjacent to the nuclear localization sequence (NLS) required for Pak1 nuclear import. Here, we demonstrate that the LC8-Pak1 interaction is necessary for epidermal growth factor (EGF)-induced nuclear import of Pak1 in MCF-7 cells, and that this event is contingent upon LC8-mediated Pak1 dimerization. In contrast, Pak2, which lacks an LC8 binding site but contains a nuclear localization sequence identical to that in Pak1, remains cytoplasmic upon EGF stimulation of MCF-7 cells. Furthermore, we show that severe developmental defects in zebrafish embryos caused by morpholino injections targeting Pak are partially rescued by co-injection of wild-type human Pak1, but not by co-injection of mutant Pak1 mRNA disrupting either the LC8 binding or the NLS site. Collectively, these results suggest that LC8 facilitates nuclear import of Pak1 and that this function is indispensable during vertebrate development.
