Polysaccharides in brown algae, including the kelp Laminaria japonica, are economically important to both food and pharmaceutical industries. The method used to extract polysaccharides significantly influences their characteristics and functions. In this study, L. japonica polysaccharides (LJP) were extracted using four techniques: hot-water extraction (HWE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and acid-assisted extraction (AAE). The highest yield (9.73 ± 0.29 %) and sulfate content (12.47 ± 0.79 %) were obtained via UAE, whereas EAE yielded the highest total carbohydrate content (53.50 ± 1.38 %), with an extraction yield of 9.12 ± 0.13 %. The minimum molecular weight (Mw) was found after AAE, whereas a higher Mw was obtained after EAE and HWE, and a homogeneous Mw was obtained after UAE. All four LJP extracts contained a helix-coil conformation based on Congo red staining, with five monosaccharides, fucose, galactose, mannose, xylose, and glucuronic acid, in varying proportions. Polysaccharides extracted via EAE and UAE exhibited higher antioxidant activity with morphological and conformation changes. A significant correlation was noted between sulfate content and antioxidant activity, and antioxidant capacity was positively correlated to mannose content rather than uronic acid. The results indicate UAE to be the best of the four extraction methods for polysaccharides with homogeneous Mw and high antioxidant activity. This study sheds light on the effects of extraction processes on the physiochemical characteristics of polysaccharides and the relationship between chemical composition and antioxidant activities.

BACKGROUND The substitution or mislabeling of toothfish is an issue of significant concern for seafood authorities; it also reduces the effectiveness of marine conservation and management programs for its over-exploitation and illegal trafficking, boosting the need for identification methods. RESULTS Two species-specific real-time polymerase chain reaction (PCR) assays for the identification of Patagonian toothfish (Dissostichus eleginoides) and Antarctic toothfish (Dissostichus mawsoni) and a genus-specific real-time PCR assay for Dissostichus spp. identification were developed based on fragments of the 16S rRNA and COI (cytochrome c oxidase subunit I) genes. These methods were confirmed to be rapid, simple, and sensitive (absolute sensitivity of 0.0002 ng μL-1 and relative sensitivity of 0.1 g kg-1 with good specificity). These methods can be applied to processed and commercial fish products. CONCLUSIONS These approaches can be beneficial for protecting both consumers and producers from economic fraud and might also help protect toothfish from over-exploitation as well as combat illegal, unreported, and unregulated (IUU) fisheries. © 2021 Society of Chemical Industry.

Foodborne pathogens have caused many public health incidents and heavy economic burden. Endolysins have been proven to have efficient bactericidal activity against pathogens with low incidence of resistance. In this study, the recombinant endolysin LysSP1 encoded by Salmonella Typhimurium lytic bacteriophage SLMP1 was obtained by prokaryotic expression, and its characteristics were analyzed. Ethylenediaminetetraacetic acid (EDTA) can be used as the outer membrane permeabilizer to increase the bactericidal activity of LysSP1. Under the synergism of 5 mmol/L EDTA, LysSP1 exhibited a strong bactericidal activity against Salmonella Typhimurium ATCC14028. LysSP1 was stable at 4°C for 7 days and at -20°C for 180 days. LysSP1 remained the optimal activity at 40°C and was efficiently active at alkaline condition (pH 8.0-10.0). Divalent metal ions could not enhance the bactericidal activity of LysSP1 and even caused the significant reduction of bactericidal activity. LysSP1 not only could lyse Salmonella, but also could lyse other Gram-negative strains and Gram-positive strains. These results indicated that LysSP1 is a broad-spectrum endolysin and has potential as an antimicrobial agent against Salmonella and other foodborne pathogens. Coronavirus disease 2019 (COVID-19) is an acute respiratory infectious disease that is often accompanied by diarrhea, patients with symptoms such as diarrhea are more likely to develop severe pneumonia, while diarrhea is the most prominent among atypical symptoms. The incidence of diarrhea in COVID-19 patients is 2.0% to 49.5%. Moxibustion has been proven to have a therapeutic effect on diarrhea; however, there is no high-quality evidence on moxibustion for diarrhea in COVID-19 patients. This study was designed to evaluate the effectiveness and safety of moxibustion for the treatment of diarrhea in patients with COVID-19.Randomized controlled trials from December 2019 to December 2021 will be included without restrictions on language or publication date. PubMed, EMBASE, Cochrane Library, Web of Science, Chinese Biomedical Databases, China National Knowledge Infrastructure, Wanfang database, and VIP database will be searched. Two researchers will independently select studies, extract data and evaluate study quality. Cochrane risk of bias tool for randomized trials will be used to assess the risk of bias of included studies. Statistical analyses will be performed using the Review Manager V.5.3 and stata 14.0.The results of this meta-analysis will be submitted to a peer-reviewed journal for publication.This study will provide evidence for whether moxibustion therapy is beneficial to the treatment of diarrhea in COVID-19.Ethical approval is not required for this study. The systematic review will be published in a peer-reviewed journal, presented at conferences, and shared on social media platforms. Foodborne viruses are a global threat to food safety. Real-time reverse transcription polymerase chain reaction (RT-PCR) is the most commonly used method to detect viral RNA in food. Armored RNA (AR) prepared using the MS2 phage system is a successful positive control for detecting foodborne viruses and is an important quality control process when using real-time RT-PCR. In this study, we report a novel technology for preparing AR using bacteriophage Qβ and compare its stability with AR prepared using the MS2 phage system for packaging norovirus detection target RNA. AR could be successfully and efficiently produced using the developed bacteriophage Qβ system. Two types of AR–AR-QNoV prepared using the Qβ system and AR-MNoV prepared using the MS2 system—were stored at different temperatures for different durations. After incubating at − 20 °C for 360 days, the copy numbers of AR-QNoV and AR-MNoV decreased by 8.9% and 35.9%, respectively. After incubating at 4 °C for 60 days, the copy numbers of AR-QNoV and AR-MNoV decreased by 12.0% and 38.9%, respectively. After incubating at 45 °C, the copy numbers of AR-QNoV decreased by 71.8% after 5 days, whereas those of AR-MNoV decreased by 92.9% after only 4 days. BACKGROUND Oilfish (Ruvettus pretiosus) and escolar (Lepidocybium flavobrunneum) are often used to adulterate high-value aquatic products, causing serious economic losses to consumers, and even keriorrhea in severe cases. It was difficult to identify them by morphological features for these two fish were processed into steak or fillet. The purpose of this study, therefore, is to develop an accurate and efficient method for detecting the oilfish- and escolar-derived components. RESULTS By comparing the mitochondrial 16S ribosomal RNA gene sequences of oilfish, escolar, and 16 varieties susceptible to adulteration, specific primers/probes were designed, and a duplex real-time polymerase chain reaction (PCR) method was established to detect oilfish- and escolar-derived components. The specificity and sensitivity of the method were analyzed, and the method was used to analyze 70 commercial samples to evalutate its applicability to actual samples in the market. The method developed was highly specific, without any cross-reaction on the other 16 species, with a limit of detection (LOD) for DNA of 0.0002 ng μL-1 for R. pretiosus and 0.002 ng μL-1 for L. flavobrunneum. In addition, the LOD for mixed muscle tissues was 0.1 g kg-1 . Oilfish- and escolar-derived components were detected in 12 of the 70 commercial samples, a result that is consistent with the classic DNA barcoding test results. CONCLUSION The duplex real-time PCR method developed can be used to detect oilfish and escolar specifically, sensitively and accurately; it provides a good technical support for the identification of authentic aquatic products. © 2020 Society of Chemical Industry.

The Thunnini, or tuna, comprise many species with very different commercial values. The principal raw tuna product on the market is sashimi, for which the species used is difficult to identify through conventional morphological analysis. The present study amplified the cytochrome b gene (Cytb) of 4 major tuna species used for preparing sashimi—yellowfin tuna (Thunnus albacares), southern bluefin tuna (Thunnus maccoyii), bigeye tuna (Thunnus obesus), and Atlantic bluefin tuna (Thunnus thynnus)—and 4 species commonly mislabeled as components of tuna sashimi—albacore tuna (Thunnus alalunga), skipjack tuna (Katsuwonus pelamis), striped marlin (Tetrapturus audax), and swordfish (Xiphias gladius). Polymerase chain reaction (PCR) amplicons were digested with 5 restriction enzymes—Eco147 I, Hinf I, Mbo I, Xag I, and Hind II—to obtain characteristic restriction maps of the above‐mentioned raw tuna species and the commonly mislabeled species. An identification method using PCR restriction fragment length polymorphism (PCR‐RFLP) was established and validated using 39 commercial tuna sashimi samples, which verified that this method provides results consistent with those obtained by classical sequencing. PCR‐RFLP has several advantages over classical sequencing, such as simplicity, speed and accuracy. This technique could support species identification for raw tuna and sashimi., Find out the restriction fragment length polymorphism in the Cyt b gene of most common kinds of tuna for sashimi preparation. The aim of this study was to evaluate the effectiveness of bacteriophage (phage) SLMP1 to reduce Salmonella Typhimurium on contaminated raw salmon fillets and scallop adductors as a function of Salmonella inoculum level, phage dose, storage temperature, and storage time. Samples were inoculated with 102 and 104 CFU/g Salmonella and then treated with different concentrations of phage SLMP1, followed by incubation at 4, 15, and 25°C, respectively. The results showed that 108 PFU/g was the optimal concentration of phage for the control of Salmonella, which was applied in the following storage experiments over a 7-day period at 4°C, a 4-day period at 15°C, and a 2-day period at 25°C. For the salmon fillets samples, 102 CFU/g Salmonella could be reduced below the detection limit at all three temperatures, whereas 104 CFU/g Salmonella was first decreased and then increased at 15 and 25°C. Lead, one of the most harmful heavy metals, can cause various hazardous effects on living organisms. This study was undertaken to evaluate the antagonistic and protective effects of two economically important laver species, Pyropia yezoensis and P. haitanensis, against subchronic lead poisoning in rats by a 30-day feeding test. Sixty-four healthy Wistar rats were randomly divided into eight groups with eight rats (4♂ + 4♀) per group, among which, one group was served as the control, the others were respectively treated with lead acetate (5 mg/kg b w), and a combination of lead acetate and P. yezoensis or P. haitanensis at different dosages. Weight gain of rats was observed and recorded. Changes in antioxidant indexes, and liver and renal function markers were determined to evaluate the antagonistic effect. Lead content in rats was determined to investigate lead excretion effect of laver. The results showed that exposure to lead caused lead accumulation in kidney and liver, thus leading to significant oxidative damage and impaired liver and renal function compared to the control group. The co-treatment of laver slightly increased body weight compared to the lead-treated group. The co-administration of laver restored liver and renal function of rats by preventing the increment in the activities of alanine transaminase (ALT), alkaline phosphatase (ALP), and aspartate transaminase (AST), and the levels of blood urea nitrogen (BUN) and creatinine (Cr). The increasing of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities, and lowering of the enhanced malondialdehyde (MDA) contents of rats were observed in the laver co-treated groups, which indicated that laver enhanced the antioxidative capacity of rats. The laver also enhanced lead content in feces and reduced it in liver and kidney. The results indicated that P. yezoensis and P. haitanensis could maintain or promote the normal physiological and biochemical function of lead-induced subchronic poisoning of rats, probably owing to their enhancements of antioxidant capacity and lead excretion.

A method was developed for the determination of trace pentachlorophenol and its sodium salt in animal-origin foods by modified QuEChERS-ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Sodium pentachlorophenolate in samples was converted to pentachlorophenol under acidic condition. The pentachlorophenol was extracted twice with acetonitrile containing 1% (v/v) acetic acid by ultrasonic extraction. The extracts were purified by dispersive solid-phase extraction. The usages of dispersive sorbents were optimized based on the recoveries and matrix effects. Chromatographic analysis was conducted on a Waters Acquity UPLC HSS T3 column with gradient elution. The pentachlorophenol was further analyzed by negative electrospray ionization under the multiple reaction monitoring mode. The recoveries at fortification levels of 1.0, 2.0 and 10.0 μg/kg in six matrices (pork, pork liver, chicken, fish, milk and egg) ranged from 73.2% to 108.4% with the relative standard deviations of 4.0%-14.8%. Vibrio parahaemolyticus is an important foodborne pathogen which commonly inhabits estuarine and marine environments and seafood. In the present study, 90 V. parahaemolyticus isolates from the main seafoods from three coastal provinces surrounding Bohai Sea and Yellow Sea, China were analyzed to elucidate their antimicrobial resistance, virulence and genetic relationship by multilocus sequence typing (MLST). The results showed that the virulence genes tdh and trh were detected in one isolate and five isolates respectively. Most of isolates showed resistance to ampicillin (86/90) and cephazolin (75/90). Some isolates were resistant to amikacin (27/90), cefuroxime sodium (18/90), tetracycline (16/90), sulphamethoxazole/trimethoprim (16/90) and streptomycin (13/90). Forty isolates (44.4%) possessed multiple antimicrobial resistance to at least three antimicrobials. The V. parahaemolyticus population was composed of 68 sequence types, of which 41 were novel to the pubMLST database, displaying a high level of genetic diversity. The phylogenetic relatedness of V. parahaemolyticus isolates was irrelevant to the collection sources. Moreover, there were no associations of antimicrobial resistance and trh positive virulence with genetic population of V. parahaemolyticus isolates. These results indicated that the diversity of antimicrobial-resistant or pathogenic V. parahaemolyticus isolates from coasts of Bohai Sea and Yellow Sea, China could pose a potential risk to human health.

UNLABELLED This study was aimed to evaluate the antimicrobial resistance and molecular resistance mechanisms of 87 Vibrio parahaemolyticus isolates from cultured sea cucumbers (Apostichopus japonicus). The results showed that all isolates were resistant to ampicillin and cephazolin, fewer of them were resistant to streptomycin (43·7%), cefuroxime sodium (18·4%), tetracycline (4·6%), sulphamethoxazole/trimethoprim (2·3%) and four quinolones (2·3%). More than half (56·2%) of the isolates displayed multiple resistance to at least three antimicrobials. The resistance genes were detected in all antimicrobial-resistant isolates except two tetracycline-resistant isolates. Among all these tested resistance genes, blaTEM , sul2, strA and strB were predominant, and none of blaSHV , blaCTX-M , blaOXA , sul1, sul3, tetA, tetM and tetQ genes was detected. Point mutations were found in quinolone resistance-determining regions of gyrA and parC genes in quinolone-resistant isolates. All isolates harboured class 1 integrons but only one carried gene cassette without any resistance genes, and none of them was positive to class 2, 3 integrons and SXT constins. These results indicate that the antimicrobial-resistant V. parahaemolyticus isolates from sea cucumbers and resistance genes could be potential risks to public health or other environments. SIGNIFICANCE AND IMPACT OF THE STUDY This study is the first report on characterization of antimicrobial resistance of Vibrio parahaemolyticus from sea cucumbers (Apostichopus japonicus). Our findings reveal a high level of resistance to some antimicrobials and prevalence of the resistance genes in V. parahaemolyticus isolates from sea cucumbers and underline the need for prudent use of antimicrobials in aquaculture to minimize the spread of antimicrobial-resistant V. parahaemolyticus.

A rapid and efficient method for extracting Mnoiodo-tyrosine (MIT) and Diiodo-tyrosine (DIT) from laminaria was successfully developed. The laminaria samples were first pretreated with microwaves, before the MIT and DIT were extracted with an ultrasonic water bath. A response surface experiment was used to optimize the extraction conditions, and ultimately the best conditions for extraction were successfully identified, which were a microwave power of 250 W, a microwave time of 4.7 min, an addition of 0.31 g of pancreatin, and 4.22 h in the ultrasonic water bathe. A simple and low-cost High-Performance Liquid Chromatography High-Performance Liquid Chromatography Photodiode Array (HPLC-PDA) detection method for MIT and DIT was established. The detection limits ranged from 50.6 to 320.6 ng/g. Scanning Electron Microscopy (SEM) was used to observe the degree of damage to the laminaria cells from the different extraction methods. The molecular weight distribution of the organic iodine of the original laminaria and the laminaria after enzymatic hydrolysis was studied by Size-Exclusion Chromatography Inductively Coupled Plasma Mass Spectrometry (SEC-ICP-MS).

In this paper using the "soft" and "hard" way, two methods of future airspace window are studied and analyzed deeply, one is the firing data distribution method and another is preplacing gun barrel angle method.Study conclusion shows our method is s a kind of effective way to improve the high-speed and complicated maneuvering anti-ship missile attacking.

A fast method based on QuEChERS methodology for the simultaneous determination of 32 sulfonylurea herbicide residues in sweet corns and green soybeans was developed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The clean-up effects of three dispersive sorbents were evaluated in terms of the residue mass for extracts after evaporation and recoveries. The three sorbents were C18, a mixture of two sorbents--silica coated with zirconium dioxide (Z-Sep) and C18, a bonded C18 zirconia-coated silica (Z-Sep+). As a result, the best effects were obtained from using Z-Sep/C18 sorbents. The samples were extracted with acetonitrile, and salted out with anhydrous magnesium sulphate and sodium chloride. The extracts were cleaned up with dispersive solid phase extraction using Z-Sep/C18 sorbents. Chromatographic analysis was carried out using a CSH C18 column with gradient elution. The pesticides were analyzed by negative electrospray ionization tandem mass spectrometry under scheduled multiple reaction monitoring mode. The quantification was achieved using matrix-matched standard calibrations as the external standard. The recoveries at fortification levels of 10, 20, 100 µg/kg in sweet corns and green beans ranged from 80.0% to 108.2% with the relative standard deviations of 1.2%-13.0%. The limits of quantification (S/N ≥ 10) were 0.2-5.0 µg/kg. The method has been proven to be simple, sensitive, environmental, and thus suitable for the determination of the 32 sulfonylurea herbicide residues in sweet corns and green soy- beans.

A method for the simultaneous detection of 21 plant growth regulators in fruits by QuEChERS-high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) was developed. The samples were initially extracted with acetonitrile containing 1% (v/v) acetic acid, followed by clean-up using the powder of magnesium sulfate and C18. The resulting samples were separated on a C18 column, and detected under positive and negative multiple reactions monitoring (MRM) mode through polarity switching between time segments. The matrix-matched external standard calibration curves were used for quantitative analysis. The linearities of chlormequat chloride, mepiquat chloride, choline chloride, cyclanilide, forchlorfenuron, thidiazuron, inabenfide, paclobutrazol, uniconazole and triapenthenol were in the concentration range of 0.1-500 microg/L, daminozide and 6-benzylaminopurine in the concentration range of 1.0-500 microg/L, 2,3,5-triiodobenzoic acid, 2,4-D, cloprop, 4-chlorophenoxyacetic acid (4-CPA) and trinexapac-ethyl in the concentration range of 2.0-1 000 microg/L, abscisic acid (ABA), gibberellic acid (GA3), 1-naphthaleneacetic acid (NAA) and indol-3-ylacetic acid (IAA) in the concentration range of 10-1000 microg/L, with the correlation coefficients higher than 0.990. The limits of detection and the limits of quantification of the method were 0.020-6.0 microg/kg and 0.10-15.0 microg/kg, respectively. For all the samples, the average spiked recoveries ranged from 73.0% to 111.0%, and the relative standard deviations (RSDs, n = 6) were in the range of 3.0% - 17.2%. The method is quick, easy, effective, sensitive and accurate, and can meet the requirements for the determination of the 21 plant growth regulator residues in fruits.

The raw extracts of six vegetables (tomato, green bean, shallot, broccoli, ginger and carrot) were analyzed using gas chromatography-mass spectrometry (GC-MS) in full scan mode combined with NIST library search to confirm main matrix compounds. The effects of cleanup and adsorption mechanisms of primary secondary amine (PSA) , octadecylsilane (C18) and PSA + C18 on co-extractives were studied by the weight of evaporation residue for extracts before and after cleanup. The suitability of the two versions of QuEChERS method for sample preparation was evaluated for the extraction of 51 carbamate pesticides in the six vegetables. One of the QuEChERS methods was the original un-buffered method published in 2003, and the other was AOAC Official Method 2007.01 using acetate buffer. As a result, the best effects were obtained from using the combination of C18 and PSA for extract cleanup in vegetables. The acetate-buffered version was suitable for the determination of all pesticides except dioxacarb. Un-buffered QuEChERS method gave satisfactory results for determining dioxacarb. Based on these results, the suitable QuEChERS sample preparation method and liquid chromatography-positive electrospray ionization-tandem mass spectrometry under the optimized conditions were applied to determine the 51 carbamate pesticide residues in six vegetables. The analytes were quantified by matrix-matched standard solution. The recoveries at three levels of 10, 20 and 100 microg/kg spiked in six vegetables ranged from 58.4% to 126% with the relative standard deviations of 3.3%-26%. The limits of quantification (LOQ, S/N > or = 10) were 0.2-10 microg/kg except that the LOQs of cartap and thiofanox were 50 microg/kg. The method is highly efficient, sensitive and suitable for monitoring the 51 carbamate pesticide residues in vegetables.

Arsenic, a potent environmental toxic agent, causes various hazardous effects on human health. This study was performed to evaluate the protective effects of edible marine algae, Laminaria japonica and Porphyra haitanensis, on subchronic stress of rats induced by arsenic trioxide (As2O3). The co-treatment of marine algae could slightly increase the growth rates of body weights compared to the As2O3-treated group. The marine algae application restored liver and renal function by preventing the increment in the activities of alanine transaminase and alkaline phosphatase, and the levels of total protein, blood urea nitrogen, and creatinine. The increase in the contents of total cholesterol, triglyceride, and low density lipoprotein cholesterol, and decrease in the contents of high density lipoprotein cholesterol were observed in algae co-treated groups which indicated that marine algae could reverse the abnormal lipid metabolisms induced by arsenic. Moreover, these algae could protect the rats from lipid peroxidation by restoring the depletion of superoxide dismutase and glutathione peroxidase activities and sulfhydryl group contents, and lowering the enhanced malondialdehyde contents. Therefore, evidences indicate that L. japonica and P. haitanensis can serve as an effective regimen for treating arsenic poisoning.

A comparison was made between the two versions of QuEChERS sample preparation method for the extraction of 66 organophosphorus pesticides (OPPS) in vegetables. The two QuEChERS methods were the original method without buffer published in 2003, and the AOAC Official Method 2007. 01 with acetate buffer. The adsorption behaviors of primary secondary amine (PSA) sorbent and C18 sorbent on the OPPS were studied. The method of after-extraction addition was used to evaluate matrix effects for OPPS in matrix of broccoli, tomato, green soybean, radish and shallot during liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. As a result, the QuEChERS method with acetate buffer gave higher and more consistent recoveries for some of OPPS. The PSA sorbent and C18 sorbent can adsorb dibrom, so the QuEChERS method was not suitable for the analysis of dibrom. The maximum matrix effects were in the extracts of broccoli during LC-MS/MS analysis. In this article, OPPS were extracted using the QuEChERS method with acetate buffer, and analysed by LC-MS/MS under the optimized conditions with monitoring 132 MS/MS transitions of precursor ions (two for each pesticide) in one single run. Recoveries for all but dibrom at fortification levels of 10, 40, 80 microg/kg in broccoli, tomato, green soybean, radish and shallot ranged from 55% to 122% with relative standard deviations of 1.6% - 18%. The limits of quantification (S/Nor = 10) were 0.1 - 8 microg/kg. Based on these results, the analytical method was proven to be highly efficient, robust and sensitive, and suitable for the monitoring of the maximum residue limits (MRLs) of 66 OPPS in vegetables.
