1. Is the threatened land crab Cardisoma guanhumi conquering human‐dominated systems?

Abstract

Abstract Land use changes are heralded as a major driver of biodiversity loss. However, recent findings show that cities, perhaps the most radical habitat transformation, sustain increasing numbers of threatened species. This emerging trend has been mostly chronicled for vertebrates from landlocked cities, although loss of biodiversity and rates or urbanization are higher in coastal marine systems. To advance our understanding on how threatened species may conquer human‐dominated systems, we studied the threatened edible crab Cardisoma guanhumi and assessed how it is proliferating in croplands and urban systems at different spatial scales and whether populations show consequences of long‐term exploitation. We gathered the data on crab populations covering the whole distribution range, including three countries reporting this as a threatened species. The abundance, distribution, and size structure of crab populations among different land uses at local scales were compared and published data for populations thriving in different habitats throughout their distribution range were compiled. We found that at local scale this species is able to thrive in natural and human‐disturbed habitats, where food sources are heavily altered. At larger scales, the species showed no differences in abundance and size structure among natural and anthropogenic habitats. In areas near the southern distribution edge, crab populations were more abundant and composed of larger animals in urban areas and croplands than those in natural habitats, suggesting that human‐disturbed systems are stepping stones to extend the geographic range. However, we found a long‐term reduction in maximum body size, exacerbated by land use changes, that likely reflects exploitation regimes consistently targeting larger crabs. Despite its status as a threatened species, the long history of human exploitation combined with livestock farming practices may explain the proliferation of this crab in human‐dominated systems, which emphasize the need to consider conservation in human‐dominated systems.

We generated global inflammation scores and investigated their associations with cardiometabolic risk factors and adverse outcomes.Patients and Methods: Aggregate Inflammation Scores (AIS) and Principal Component Analysis (PCA) scores were generated for 7287 Framingham Heart Study participants using up to 26 inflammation-related proteins, with higher scores reflecting a pro-inflammatory milieu. Multivariable regression and proportional hazards analyses were conducted to investigate the associations of inflammation with cardiometabolic risk factors and outcomes. The primary outcomes for cross-sectional analyses included age, cigarette smoking, fasting lipid and glucose levels, blood pressure, body mass index (BMI), and hypertension, diabetes, and obesity. For prospective analyses, new-onset hypertension, diabetes, obesity, cardiovascular disease and all-cause mortality were investigated.Results: Higher inflammation scores were associated with smoking and older age, higher BMI, systolic blood pressure, lipids, and glucose levels, and with greater odds of hypertension and diabetes after adjusting for age, sex, cohort, and BMI (all p < 0.001). Higher baseline scores were associated with greater odds of new-onset hypertension after adjusting for traditional risk factors (OR [95% CI] per one standard deviation [1-SD] increase, AIS: 1.33 [1.21– 1.47], PCA score: 1.26 [1.12– 1.42], p < 0.001). The AIS also was associated with new-onset diabetes (1.32 [1.14– 1.52], p < 0.001). Proportional hazards analyses revealed greater risk of new-onset cardiovascular disease events and all-cause mortality (HR [95% CI] per 1-SD, AIS: 1.25 [1.14– 1.37] and 1.32 [1.23– 1.42], PCA score: 1.22 [1.13– 1.33] and 1.40 [1.31– 1.49], p < 0.001).Conclusion: Global inflammation scores encompassing an array of pro- and anti-inflammatory proteins and pathways may enhance risk assessment for cardiometabolic diseases. We developed an efficient method to purify the engineered exosomes from conditioned medium based on sequential centrifugation, ultrafiltration, and precipitation. We characterized decoy exosomes using immune-quantification, nanoparticle tracking analysis, and confocal microscopy to confirm that they retain the correct orientation, size, and shape of naturally produced exosomes. We demonstrated the engineered decoy exosomes specifically antagonize activities of TNFα using an inflammatory reporter cell line.Conclusions: Decoy exosomes produced in human cells serve as a novel biologic reagent for antagonizing inflammatory signaling mediated by TNFα.Keywords: Decoy exosomes, Inflammation, CD63, Receptor, TNFα, Drug delivery

Abstract

We present new experimental measurements of resonance strengths in the astrophysical 23Al(p, {\gamma})24Si reaction, constraining the pathway of nucleosynthesis beyond 22Mg in X-ray burster scenarios. Specifically, we have performed the first measurement of the (d, p) reaction using a radioactive beam of 23Ne to explore levels in 24Ne, the mirror analog of 24Si. Four strong single-particle states were observed and corresponding neutron spectroscopic factors were extracted with a precision of {\sim}20{\%}. Using these spectroscopic factors, together with mirror state identifications, we have reduced uncertainties in the strength of the key {\ell} = 0 resonance at Er= 157 keV, in the astrophysical 23Al(p, {\gamma}) reaction, by a factor of 4. Our results show that the 22Mg(p, {\gamma})23Al(p, {\gamma}) pathway dominates over the competing 22Mg({\alpha}, p) reaction in all but the most energetic X-ray burster events (T>0.85GK), significantly affecting energy production and the preservation of hydrogen fuel., Comment: 5 pages, 3 figures

Abstract

We report evidence for the first generation of XUV spectra from relativistic surface high-harmonic generation (SHHG) on plasma mirrors at a kilohertz repetition rate, emitted simultaneously with energetic electrons. SHHG spectra and electron angular distributions are measured as a function of the experimentally controlled plasma density gradient scale length $L_\mathrm{g}$ for three increasingly short and intense driving pulses: 24~fs and $a_0=1.1$, 8~fs and $a_0=1.6$, and finally 4~fs and $a_0\approx2.1$, where $a_0$ is the peak vector potential normalized by $m_\mathrm{e} c/e$ with the elementary charge $e$, the electron rest mass $m_\mathrm{e}$, and the vacuum light velocity $c$. For all driver pulses, we observe correlated relativistic SHHG and electron emission in the range $L_\mathrm{g}\in[\lambda/25,\lambda/5]$, with an optimum gradient scale length of $L_\mathrm{g}\approx\lambda/10$.

31. Formation and Stability of Ordered and Disordered Ba-substituted Calcite Phases

Abstract

Calcite has the ability to host large amounts of intracrystalline inclusions, a phenomenon that is known to be the case in biominerals and has been demonstrated in bio-inspired synthetic systems. In this study, we focused on barium as the inclusion. Highly substituted Ba-calcite possesses disordered carbonate orientations, characteristic of R-3m symmetry. We show that calcite undergoes an ordered-disordered transition, in which the carbonate groups undergo progressive loss of their rotational order with increasing amounts of incorporated Ba, and reach complete rotational disorder for a critical amount of Ba. We characterize this transition and propose a theoretical model justifying the influence of Ba. Moreover, the disordered R-3m Ba-substituted calcite has been previously identified as a high-temperature phase or as a highly metastable room-temperature phase. We challenge those descriptions by successfully synthesizing it under slow-rate conditions and by studying its thermal behavior, and we conclude that the fully disordered Ba-calcite is stable, whereas the transitional, partially disordered Ba-calcite phase is metastable.

Abstract

X-ray diffraction pressure studies at room temperature demonstrate that the spinel FeAl$_{2}$O$_{4}$ transforms to a tetragonal phase at ~18 GPa. This tetragonal phase has a highly irregular unit-cell volume versus pressure dependence up to ~45 GPa, after which a transformation to a Cmcm post-spinel phase is onset. This is attributable to pressure driven Fe-Al site inversion at room temperature, corroborated by signatures in the 57Fe M\"ossbauer spectroscopy pressure data. At the tetragonal to post-spinel transition, onset in the range 45-50 GPa, there is a concurrent emergence of a non-magnetic spectral component in the M\"ossbauer data at variable cryogenic temperatures. This is interpreted as spin crossover at sixfold coordinated Fe locations emanated from site inversion. Spin crossover commences at the end of the pressure range of the tetragonal phase and progresses in the post-spinel structure. There is also a much steeper volume change dV/V ~ 10% in the range 45-50 GPa compared to the preceding pressure regime, from the combined effects of the structural transition and spin crossover electronic change. At the highest pressure attained, ~106 GPa, the M\"ossbauer data evidences a diamagnetic Fe low-spin abundance of ~50%. The rest of the high-spin Fe in eightfold coordinated sites continue to experience a relatively small internal magnetic field of ~33 T. This is indicative of a magnetic ground state associated with strong covalency, as well as substantive disorder from site inversion and the mixed spin-state configuration. Intriguingly magnetism survives in such a spin-diluted post-spinel lattice at high densities. The R(300 K) data decreases by only two orders of magnitude from ambient pressure to the vicinity of ~100 GPa. X-ray diffraction studies to ~90 GPa at room temperature show that Fe$_{2}$TiO$_{4}$ ferrous inverse spinel undergoes the following sequence of structural transitions : cubic (Fd3m) to tetragonal (I41/amd)to orthorhombic(Cmcm) to orthorhombic(Pmma),at the indicated onset transition pressures. Within the Cmcm phase, site-specific spin crossover is initiated and involves only highly distorted octahedral sites constituting ~25% of all Fe locations. This is manifest as a steeper volume decrease of dV/V0 ~ 3.5% beyond ~40 GPa and an emergent diamagnetic component discerned in 57Fe M\"ossbauer spectroscopy at variable cryogenic temperatures. A subsequent Cmcm to Pmma Fe/Ti disorder-order reconfiguration is facilitated at 6-fold coordinated (octahedral) sites. The rest of the high-spin Fe in 6-fold and 8-fold coordinated sites (~75% abundance) in the Pmma phase exhibit average saturation internal magnetic fields of H$_{hf}$ ~ 42 T to ~90 GPa, typical of spin-only (orbitally quenched) Fermi-contact values. By contrast average H$_{hf}$ ~ 20 T values, signifying unquenched orbital moments, occur below the 40-45 GPa spin-crossover initiation regime in the Cmcm phase. Therefore site-specific spin crossover invokes a cooperative lattice response and polyhedral distortions at the rest of the high-spin Fe sites, translating to 3d level (sub-band) changes and consequential orbital moment quenching. Near ~90 GPa Fe$_{2}$TiO$_{4}$ is a partially spin-converted chemically ordered Pmma post-spinel structure having a persistent charge gap of ~100 meV. Despite structural symmetry changes, partial spin crossover and lattice compressibility resulting in a ~33% total reduction in unit-cell volume and corresponding 3d bandwidth broadening, strong electron correlations persist at high densification., Comment: 35 Pages and 7 Figures

Abstract

IntroductionThere is conflicting evidence whether high-density lipoprotein cholesterol (HDL-C) is a risk factor for Alzheimer's disease (AD) and dementia. Genetic variation in the cholesteryl ester transfer protein (CETP) locus is associated with altered HDL-C. We aimed to assess AD risk by genetically predicted HDL-C.MethodsTen single nucleotide polymorphisms within the CETP locus predicting HDL-C were applied to the International Genomics of Alzheimer's Project (IGAP) exome chip stage 1 results in up 16,097 late onset AD cases and 18,077 cognitively normal elderly controls. We performed instrumental variables analysis using inverse variance weighting, weighted median, and MR-Egger.ResultsBased on 10 single nucleotide polymorphisms distinctly predicting HDL-C in the CETP locus, we found that HDL-C was not associated with risk of AD (P > .7).DiscussionOur study does not support the role of HDL-C on risk of AD through HDL-C altered by CETP. This study does not rule out other mechanisms by which HDL-C affects risk of AD.

Abstract

This study focused on installing Closed Circuit Television Cameras to prevent the increasing crime rate in Barangay Commonwealth, Quezon City. Two groups from Barangay officials and the Philippine National Police were selected using purposive sampling based on their knowledge and skills regarding the information desired.   The study employed a Quantitative research design. Specifically, the purposive sampling technique, questionnaire with close-ended questions, procedure of data gatherings, collection of material and statistical treatment of the data were used to the effectiveness of the installation of Closed Circuit Television Cameras to prevent an increasing rate of crime in Barangay Commonwealth, Quezon City.   The researcher interviewed the Barangay officials and police officers. The main instrument used to gather information is to answer the specific problem raised in every question included in the questionnaires. In this method, the written respondent is to reply as objectively and honestly as possible. Questions were translated into Tagalog to ensure the validity of the responses, that is, correct interpretation and understanding by the respondents of the specific item in line with the purpose of the study.   The study concluded that based on the demographic profile of the two groups of respondents’ Barangay officials and police officers in Barangay Commonwealth, Quezon City, the majority of the respondents came from the age bracket 18 - 25 years old, male, single and degree holders. In the assessment, the police officers and Barangay officials know the effectiveness of installing CCTV cameras. Abstract

Objective: To derive childhood-onset SLE (cSLE) specific remission definitions for future treat-to-target (T2T) trials, observational studies, and clinical practice. Methods: The cSLE International T2T Task Force conducted Delphi surveys exploring paediatric perspectives on adult-onset SLE remission targets. A modified nominal group technique was used to discuss, refine, and agree on the cSLE remission target criteria.Results: The Task Force proposed two definitions of remission: ‘cSLE clinical remission on steroids (cCR)’ and ‘cSLE clinical remission off steroids (cCR-0)’. The common criteria are: (1) Clinical-SLEDAI-2 K = 0; (2) PGA score < 0.5 (0–3 scale); (4) stable antimalarials, immunosuppressive, and biologic therapy (changes due to side-effects, adherence, weight, or when building up to target dose allowed). Criterion (3) in cCR is the prednisolone dose ≤0.1 mg/kg/day (maximum 5 mg/day), whereas in cCR-0 it is zero. Conclusions: cSLE definitions of remission have been proposed, maintaining sufficient alignment with the adult-SLE definition to facilitate life-course research.

Abstract

The objective of this study was to compare the effectiveness of a second tumor necrosis factor inhibitor (TNFi) versus a non‐TNFi biologic following discontinuation of a TNFi for patients with polyarticular‐course juvenile idiopathic arthritis (pJIA). Using the Childhood Arthritis and Rheumatology Research Alliance Registry, patients with pJIA who started receiving a second biologic following a first TNFi were identified. Patients were required to have no active uveitis on the index date and a visit six months after the index date. Outcome measures included Clinical Juvenile Arthritis Disease Activity Score with a maximum of 10 active joints (cJADAS10), cJADAS10 inactive disease (ID; ≤2.5) and cJADAS10 minimal disease activity (MiDA; ≤5). Multiple imputation was used to account for missing data. Adjusted odds ratios (aORs) were calculated using propensity score quintiles to compare outcomes at six months following second biologic initiation. There were 216 patients included, 84% initially received etanercept, and most patients stopped receiving it because of its ineffectiveness (74%). A total of 183 (85%) started receiving a second TNFi, and 33 (15%) started receiving a non‐TNFi. Adalimumab was the most common second biologic received (71% overall, 84% of second TNFi), and tocilizumab was the most common non‐TNFi second biologic received (9% overall, 58% of non‐TNFi). There was no difference between receiving TNFi versus non‐TNFi in cJADAS10 ID (29% vs 25%; aOR 1.23, 95% confidence interval [CI] 0.47–3.20) or at least MiDA (43% vs 39%; aOR 1.11, 95% CI 0.47–2.62) at six months. Most patients with pJIA started receiving TNFi rather than non‐TNFi as their second biologic, and there were no differences in disease activity at six months.

Abstract

A series of amphiphilic LC block copolymers, in which the hydrophobic block is a smectic polymer poly(4-methoxyphenyl 4-(6-acryloyloxy-hexyloxy)-benzoate) (PA6ester1) and the hydrophilic block is polyethyleneglycol (PEG), were synthesized and characterized. The self-assembly of one of them in both the pure state and the dilute aqueous solution was investigated in detail. Nano-structures in the pure state were studied by SAXS and WAXS on samples aligned by a magnetic field. A hexagonal cylindrical micro-segregation phase was observed with a lattice distance of 11.2 nm. The PEG blocks are in the cylinder, while the smectic polymer blocks form a matrix with layer spacing 2.4 nm and layer normal parallel to the long axis of the cylinders. Faceted unilamellar polymer vesicles, polymersomes, were formed in water, as revealed by cryo-TEM. In the lyotropic bilayer membrane of these polymersomes, the thermotropic smectic order in the hydrophobic block is clearly visible with layer normal parallel to the membrane surface.

49. Is the threatened land crab Cardisoma guanhumi conquering human‐dominated systems?

Abstract

Land use changes are heralded as a major driver of biodiversity loss. However, recent findings show that cities, perhaps the most radical habitat transformation, sustain increasing numbers of threatened species. This emerging trend has been mostly chronicled for vertebrates from landlocked cities, although loss of biodiversity and rates or urbanization are higher in coastal marine systems. To advance our understanding on how threatened species may conquer human‐dominated systems, we studied the threatened edible crab Cardisoma guanhumi and assessed how it is proliferating in croplands and urban systems at different spatial scales and whether populations show consequences of long‐term exploitation. We gathered the data on crab populations covering the whole distribution range, including three countries reporting this as a threatened species. The abundance, distribution, and size structure of crab populations among different land uses at local scales were compared and published data for populations thriving in different habitats throughout their distribution range were compiled. We found that at local scale this species is able to thrive in natural and human‐disturbed habitats, where food sources are heavily altered. At larger scales, the species showed no differences in abundance and size structure among natural and anthropogenic habitats. In areas near the southern distribution edge, crab populations were more abundant and composed of larger animals in urban areas and croplands than those in natural habitats, suggesting that human‐disturbed systems are stepping stones to extend the geographic range. However, we found a long‐term reduction in maximum body size, exacerbated by land use changes, that likely reflects exploitation regimes consistently targeting larger crabs. Despite its status as a threatened species, the long history of human exploitation combined with livestock farming practices may explain the proliferation of this crab in human‐dominated systems, which emphasize the need to consider conservation in human‐dominated systems.
