Abstract The species-specific character of sounds in the animal kingdom has been extensively documented, yet research on fishes has predominantly focused on a limited number of species, overlooking the potential of acoustic signals to reflect broader taxonomic ranks. In this study, we analyzed acoustic data of hand-held sounds from 388 specimens spanning 5 genera and 33 species within the family Holocentridae, with the objective of evaluating the use of sound characteristics for taxonomic discrimination across various levels (subfamily, genus, species). Sounds could be indicative of grouping. Taxa discriminability depends on taxonomic level; the higher the taxonomic level, the better the discrimination of taxa based on sounds. Analogous to the role of morphological traits in taxonomic delineation, this research corroborates the utility of acoustic features in identifying fish taxa across multiple hierarchical levels. Remarkably, certain holocentrid species have evolved complex sound patterns characterized by unique temporal arrangements where pulses are not continuous but emitted in blocks, facilitating the exploitation of the acoustic space.

Abstract Examples of symbiotic relationships often include cleaning mutualisms, typically involving interactions between cleaner fish and other fish, called the clients. While these cleaners can cooperate by removing ectoparasites from their clients, they can also deceive by feeding on client mucus, a behavior usually referred to as “cheating behavior” that often leads to a discernible jolt from the client fish. Despite extensive studies of these interactions, most research has focused on the visual aspects of the communication. In this study, we aimed to explore the role of acoustic communication in the mutualistic relationship between cleaner fishes and nine holocentrid client species across four regions of the Indo-Pacific Ocean: French Polynesia, Guam, Seychelles, and the Philippines. Video cameras coupled with hydrophones were positioned at various locations on reefs housing Holocentridae fish to observe their acoustic behaviors during interactions. Our results indicate that all nine species of holocentrids can use acoustic signals to communicate to cleaner fish their refusal of the symbiotic interaction or their desire to terminate the cooperation. These sounds were predominantly observed during agonistic behavior and seem to support visual cues from the client. This study provides a novel example of acoustic communication during a symbiotic relationship in teleosts. Interestingly, these vocalizations often lacked a distinct pattern or structure. This contrasts with numerous other interspecific communication systems where clear and distinguishable signals are essential. This absence of a clear acoustic pattern may be because they are used in interspecific interactions to support visual behavior with no selective pressure for developing specific calls required in conspecific recognition. The different sound types produced could also be correlated with the severity of the client response. There is a need for further research into the effects of acoustic behaviors on the quality and dynamics of these mutualistic interactions.

The literature on sound production behaviours in fish in the wild is quite sparse. In several taxa, associations between different sound types and given behaviours have been reported. In the Holocentridae, past nomenclature of the different sound types (knocks, growls, grunts, staccatos and thumps) has been confusing because it relies on the use of several terms that are not always based on fine descriptions. Our study aims to ascertain whether holocentrids can produce a variety of sounds in the wild and if these sounds are associated with specific behaviours. Additionally, we aim to determine whether sounds produced by hand-held specimens, a common methodology to record sounds in standardised conditions in fishes, could correspond to some sounds produced by free-swimming individuals in natural conditions. Our study shows that all holocentrid species are able to produce sounds in 6 behavioural contexts of both agonistic (conspecific and heterospecific chases, competition) and social signalling types (acceleration, broadcasting, body quivering), in addition to previously described mobbing towards moray eels and symbiotic interactions with cleaner wrasses. In holocentrids, acoustic communication is not only based on single calls but can also involve series of sounds of different types that are arranged randomly. The large amount of combinations within acoustical events for each behaviour, resulting from both the quantity of sounds and their diversity, supports the absence of stereotypy. This suggests that sounds are produced to reinforce visual communication during the day in this family. Our results also suggest that sounds recorded by hand-held fishes are produced naturally in the wild. Our study challenges past nomenclatures and demonstrates sound critical function in augmenting visual communication, advancing our comprehension of acoustic ecology in teleost species.

Mesophotic coral ecosystems (MCEs) are the deepest part of tropical coral reefs, ranging from depths of 30 to over 170 m. Despite their significance, MCEs remain largely unexplored due to the challenges associated with accessing these depths. However, the application of passive acoustic monitoring methods (PAM) is a suitable approach for studying fish communities within these unique habitats. In French Polynesia, recent PAM studies have unveiled a higher occurrence of frequency-modulated fish sounds in MCEs than in shallower reef environments. This study aims to further enhance our understanding of fish sounds in MCEs by examining their diel patterns, focusing specifically on the two most abundant frequency-modulated fish sounds that were recorded at depths of 60 and 120 m at six Polynesian islands. Both sound types occurred predominantly during the beginning and the end of nocturnal periods. The presence and abundance of these sounds exhibited variation between the islands, highlighting potential regional disparities in vocal activity or the bathymetric distribution of the sound-producing species. By characterizing the diel cycles and bathymetric differences in relation to their geographical distribution, this study offers preliminary insights into identifying the potential sound-producing species.

Summary: Most marine organisms have a biphasic life cycle during which pelagic larvae transform into radically different juveniles. In vertebrates, the role of thyroid hormones (THs) in triggering this transition is well known, but how the morphological and physiological changes are integrated in a coherent way with the ecological transition remains poorly explored. To gain insight into this question, we performed an integrated analysis of metamorphosis of a marine teleost, the false clownfish (Amphiprion ocellaris). We show how THs coordinate a change in color vision as well as a major metabolic shift in energy production, highlighting how it orchestrates this transformation. By manipulating the activity of liver X regulator (LXR), a major regulator of metabolism, we also identify a tight link between metabolic changes and metamorphosis progression. Strikingly, we observed that these regulations are at play in the wild, explaining how hormones coordinate energy needs with available resources during the life cycle.

In the context of the general overexploitation of South Pacific reef fisheries, there is a global push to precisely define fishery parameters such as fishing effort, annual landings, and exploitation levels. In Bora-Bora, several surveys were performed to define the fishing grounds and the fishing effort of the reef fishery for the first time. Maps of the fishing grounds under different fishing pressures were created for the Bora-Bora reef fishery. The average fishing effort throughout the lagoon was 40 trips week−1 km−2. Annual landings were estimated following two different methods: school participatory surveys and landing based on the catch per unit effort. These estimations were used to determine that the maximum sustainable yield of the island's fisheries was 6.5 tons km−2 year−1. the reported annual yield in 2022 was 5.35 tons km−2 year−1, under the estimated maximum sustainable yield. The level of exploitation of the fisheries was also estimated based on the catch size composition of the ten most fished species. This revealed that fishing is responsible for more than 50% of fish mortality in six of the ten studied species, and that catch length could be increased to optimize both catches and biomass. Overall, the Bora-Bora reef fishery may be experiencing growth overfishing (i.e., the catching of fish that are too small to maximise biological sustainability and economical yield), which can endanger stock recruitment in the future if no actions are taken.

IntroductionThe Notch intracellular domain (NICD) and its ligands Jagged-1(Jag1), Delta-like ligand (DLL-3) and DLL4 play an important role in neoangiogenesis. Previous studies suggest a correlation between the tissue levels of NICD and response to therapy with bevacizumab in colorectal cancer (CRC). Another marker that may predict outcome in CRC is radiomics of liver metastases. The aim of this study was to investigate the expression of NICD and its ligands and the role of radiomics in the selection of treatment-naive metastatic CRC patients receiving bevacizumab.MethodsImmunohistochemistry (IHC) for NICD, Jag1 and E-cadherin was performed on the tissue microarrays (TMAs) of 111 patients with metastatic CRC treated with bevacizumab and chemotherapy. Both the intensity and the percentage of stained cells were evaluated. The absolute number of CD4+ and CD8+ lymphocytes was counted in three different high-power fields and the mean values obtained were used to determine the CD4/CD8 ratio. The positivity of tumor cells to DLL3 and DLL4 was studied. The microvascular density (MVD) was assessed in fifteen cases by counting the microvessels at 20x magnification and expressed as MVD score. Abdominal CT scans were retrieved and imported into a dedicated workstation for radiomic analysis. Manually drawn regions of interest (ROI) allowed the extraction of radiomic features (RFs) from the tumor.ResultsA positive association was found between NICD and Jag1 expression (p < 0.001). Median PFS was significantly shorter in patients whose tumors expressed high NICD and Jag1 (6.43 months vs 11.53 months for negative cases; p = 0.001). Those with an MVD score ≥5 (CD31-high, NICD/Jag1 positive) experienced significantly poorer survival. The radiomic model developed to predict short and long-term survival and PFS yielded a ROC-AUC of 0.709; when integrated with clinical and histopathological data, the integrated model improved the predictive score (ROC-AUC of 0.823).DiscussionThese results show that high NICD and Jag1 expression are associated with progressive disease and early disease progression to anti VEGF-based therapy; the preliminary radiomic analyses show that the integration of quantitative information with clinical and histological data display the highest performance in predicting the outcome of CRC patients.

During the COVID-19 pandemic, the reduced exports and imports as well as the lack of activity due to the interruption in the international tourism economy seriously impacted food security in many Pacific Islands. People often returned to natural resources to provide for themselves, their families, or to generate income. On Bora-Bora Island, the major tourist destination in French Polynesia, roadside sales are widespread. Our study analyses the impact of the COVID-19 pandemic on roadside sales activities through a census of roadside stalls on the five Bora-Bora districts conducted before (January and February 2020), during (from March 2020 to October 2021) and after (from November to December 2021) health-related activity and travel restrictions. Our results showed that the marketing system for local products (fruits, vegetables, cooked meals, and fish) increased in the form of roadside sales during the COVID-19 in two of the five districts of Bora-Bora. Roadside selling would be an alternative system for providing food to the population at Bora-Bora during a global crisis and that could reveal itself sustainable after this pandemic.

In 2020, the COVID-19 pandemic led to a reduction in human activities and restriction of all but essential movement for much of the world's population. A large, but temporary, increase in air and water quality followed, and there have been several reports of animal populations moving into new areas. Extending on long-term monitoring efforts, we examined how coral reef fish populations were affected by the government-mandated lockdown across a series of Marine Protected Area (MPA) and non-Marine Protected Area (nMPA) sites around Moorea, French Polynesia. During the first six-week lockdown that Moorea experienced between March and May 2020, increases (approx. two-fold) in both harvested and non-harvested fishes were observed across the MPA and nMPA inner barrier reef sites, while no differences were observed across the outer barrier sites. Interviews with local amateur and professional fishers indicated that while rules regarding MPA boundaries were generally followed, some subsistence fishing continued in spite of the lockdown, including within MPAs. As most recreational activities occur along the inner reef, our data suggest that the lockdown-induced reduction in recreational activities resulted in the recolonization of these areas by fishes, highlighting how fish behaviour and space use can rapidly change in our absence.

Abstract Understanding the processes that shape biodiversity is essential for effective environmental management. Across the world’s coral reefs, algal farming damselfish (Stegastes sp.) modify the surrounding benthic community through their creation of algae “farms”. Using a long-term monitoring dataset (2005–2019) from Moorea Island, French Polynesia, we investigated whether the density of dusky damselfish (Stegastes nigricans) is associated with benthic habitat composition, the density of predators and/or competitors, and whether the survey area was inside or outside of a Marine Protected Area (MPA). We found no evidence that benthic cover or number of competitors were associated with dusky damselfish densities, both inside and outside MPAs. In contrast, fluctuations in dusky damselfish densities were negatively associated with the density of predators (e.g. Serranidae, Muraenidae and Scorpaenidae) in the preceding year in non-MPA areas, and both within and outside of MPAs when predator densities were high (2005–2010). These results suggest that healthy predator populations may be important for regulating the abundances of keystone species, such as algal farming damselfish, especially when predator densities are high.

Convergent evolution of small body size occurs across many vertebrate clades and may reflect an evolutionary response to shared selective pressures. However it remains unclear if other aspects of phenotype undergo convergent evolution in miniaturized lineages. Here we present a comparative analysis of body size and shape evolution in marine angelfishes (Pomacanthidae), a reef fish family characterized by repeated transitions to small body size. We ask if lineages that evolve small sizes show convergent evolution in body shape. Our results reveal that angelfish lineages evolved three different stable size optima with one corresponding to the group of pygmy angelfishes (Centropyge). Then, we test if the observed shifts in body size are associated with changes to new adaptive peaks in shape. Our data suggest that independent evolution to small size optima have induced repeated convergence upon deeper body and steeper head profile in Centropyge These traits may favour manoeuvrability and visual awareness in these cryptic species living among corals, illustrating that functional demands on small size may be related to habitat specialization and predator avoidance. The absence of shape convergence in large marine angelfishes also suggests that more severe requirements exist for small than for large size optima.

Shallow coral reefs ensure a wide portfolio of ecosystem services, from fish provisioning to tourism, that support more than 500 million people worldwide. The protection and sustainable management of these pivotal ecosystems require fine-scale but large-extent mapping of their 3D composition. The sub-metre spaceborne imagery can neatly produce such an expected product using multispectral stereo-imagery. We built the first 3D land-sea coral reefscape mapping using the 0.3 m superspectral WorldView-3 stereo-imagery. An array of 13 land use/land cover and sea use/sea cover habitats were classified using sea-, ground- and air-truth data. The satellite-derived topography and bathymetry reached vertical accuracies of 1.11 and 0.89 m, respectively. The value added of the eight mid-infrared (MIR) channels specific to the WorldView-3 was quantified using the classification overall accuracy (OA). With no topobathymetry, the best combination included the eight-band optical (visible + near-infrared) and the MIR8, which boosted the basic blue-green-red OA by 9.58%. The classes that most benefited from this MIR information were the land use “roof” and land cover “soil” classes. The addition of the satellite-derived topobathymetry to the optical+MIR1 produced the best full combination, increasing the basic OA by 9.73%, and reinforcing the “roof” and “soil” distinction.

Anthropogenic stressors affect many aspects of marine organismal health. Here, the authors expose surgeonfish to temperature and pesticide stressors and show that the stressors, separately and in combination, have adverse effects on thyroid signaling, which disrupts several sensory systems and important predation defenses.

Brucella canis has been isolated for the first time in Italy in a commercial breeding kennel. It was diagnosed after a deep investigation related to the onset of reproductive disorders. Animals were tested with direct and indirect techniques. The agent was first detected in two Chihuahua aborted foetuses by direct culture. Further, it was also isolated from blood samples of dogs hosted in the kennel, which also showed reaction to conventional serological tests (microplate serum agglutination test). The isolates were identified as B. canis by standard microbiological methods and a Bruce‑ladder multiplex PCR. To investigate the genomic diversity, whole genome sequencing was used, applying the core genome Multilocus Sequence Typing (cgMLST ). In a first round of serological testing performed on 598 animals, 269 (46.1%) tested positive. In the second round of laboratory testing carried out 4‑5 weeks apart, the number of serologically positive dogs was 241 out of 683 tested (35.3%), while the number of dogs positive to isolation was 68 out of 683 tested (10.0%). The PCR showed a lack of sensitivity when compared to direct isolation. The epidemiological investigation did not identify the source of the infection, given the time elapsed from the onset of abortions to the definitive diagnosis of B. canis infection in the kennel. The genomic analyses featured the strains as ST21 and, according to the cgMLST, revealed the presence of a tight cluster with a maximum diversity of four allelic differences. The observed limited genomic variation, largely within the known outbreak cut‑offs, suggests that the outbreak herein described was likely caused by a single introduction. Moreover, in a broader scale comparison using the public available genomes, we found that the closest genome, isolated in China, differed by more than 50 alleles making not possible to find out the likely origin of the outbreak. The lack of updated data on B. canis genome sequences in the public databases, together with the limited information retrieved from the epidemiological investigations on the outbreak, hampered identification of the source of B. canis infection.

Abstract Marine natural products isolated from the sponge Fascaplysinopsis cf reticulata, in French Polynesia, were investigated as an alternative to antibiotics to control pathogens in aquaculture. The overuse of antibiotics in aquaculture is largely considered to be an environmental pollution, because it supports the transfer of antibiotic resistance genes within the aquatic environment. One environmentally friendly alternative to antibiotics is the use of quorum sensing inhibitors (QSIs). Quorum sensing (QS) is a regulatory mechanism in bacteria which control virulence factors through the secretion of autoinducers (AIs), such as acyl-homoserine lactone (AHL) in gram-negative bacteria. Vibrio harveyi QS is controlled through three parallel pathways: HAI-1, AI-2, and CAI-1. Bioassay-guided purification of F. cf reticulata extract was conducted on two bacterial species, i.e., Tenacibaculum maritimum and V. harveyi for antibiotic and QS inhibition bioactivities. Toxicity bioassay of fractions was also evaluated on the freshwater fish Poecilia reticulata and the marine fish Acanthurus triostegus. Cyclohexanic and dichloromethane fractions of F. cf reticulata exhibited QS inhibition on V. harveyi and antibiotic bioactivities on V. harveyi and T. maritimum, respectively. Palauolide (1) and fascaplysin (2) were purified as major molecules from the cyclohexanic and dichloromethane fractions, respectively. Palauolide inhibited QS of V. harveyi through HAI-1 QS pathway at 50 μg ml–1 (26 μM), while fascaplysin affected the bacterial growth of V. harveyi (50 μg ml–1) and T. maritimum (0.25 μg). The toxicity of fascaplysin-enriched fraction (FEF) was evaluated and exhibited a toxic effect against fish at 50 μg ml–1. This study demonstrated for the first time the QSI potential of palauolide (1). Future research may assess the toxicity of both the cyclohexanic fraction of the sponge and palauolide (1) on fish, to confirm their potential as alternative to antibiotics in fish farming.

Understanding the natural state of coral reefs is paramount to evaluate the response of these ecosystems to local and global human impacts as well as management and conservation strategies. In French Polynesia, some islands are still pristine or uninhabited, such as Tupai atoll. Tupai has been uninhabited, with access to the lagoon prohibited since 2010. However, fishers from nearby islands often take from the outer reef slope at Tupai. Our marine biodiversity survey (coral, macro-invertebrates, and fish) conducted in 2019 highlighted a low density of commercial fish species and top-predators on the outer slope in comparison to the lagoon, where the top-predators represented 16% (of the density) of functional trophic groups. Our surveys also showed a high living coral cover (46%) on the outer slope of Tupai, perhaps due to the absence of both touristic sub-aquatic activities and local pollution from private and commercial activities. Overall, this initial scientific assessment of Tupai has granted an understanding of the spatial patterns of coral, macro-invertebrates, and fish assemblages in the absence of human impacts (i.e., in the lagoon), representing an ecological baseline that could inform conservation management strategies to ensure the preservation of coral reef ecosystem.

Advancement and diversification of the aquaculture industry is reliant on the development of captive breeding and rearing protocols for novel fish species. Orbicular batfish (Platax orbicularis), a major emerging species in Polynesian aquaculture, live in brackish and marine waters around coral reefs, and are highly prized by Pacific Island communities for their high-quality meat. The present study describes the larval growth of P. orbicularis from hatching until 16 days after hatching (DAH) using meristics and thyroid hormone (T3) quantification. Our study highlighted that metamorphosis of P. orbicularis is critical to their production in aquaculture, as found for other species. Levels of the thyroid hormone T3 in P. orbicularis reached a peak at 12 DAH (i.e. hormonal metamorphosis). This peak occurred in concert with important morphological changes and increased mortality and growth between 9 and 12 DAH, clearly illustrating the sensitivity of this fish during metamorphosis. Overall, our study sheds light on the metamorphosis and larval development of a novel aquaculture species, and the interplay between hormonal and morphological changes throughout a critical period of its rearing. These findings may promote the development of protocols for mass-scale production of orbicular batfish in captivity, which may be particularly beneficial to the aquaculture industries of many Pacific Island countries.

Abstract Background Biologists have long been fascinated by the striking diversity of complex color patterns in tropical reef fishes. However, the origins and evolution of this diversity are still poorly understood. Disentangling the evolution of simple color patterns offers the opportunity to dissect both ultimate and proximate causes underlying color diversity. Results Here, we study clownfishes, a tribe of 30 species within the Pomacentridae that displays a relatively simple color pattern made of zero to three vertical white stripes on a dark body background. Mapping the number of white stripes on the evolutionary tree of clownfishes reveals that their color pattern diversification results from successive caudal to rostral losses of stripes. Moreover, we demonstrate that stripes always appear with a rostral to caudal stereotyped sequence during larval to juvenile transition. Drug treatments (TAE 684) during this period leads to a dose-dependent loss of stripes, demonstrating that white stripes are made of iridophores and that these cells initiate the stripe formation. Surprisingly, juveniles of several species (e.g., Amphiprion frenatus) have supplementary stripes when compared to their respective adults. These stripes disappear caudo-rostrally during the juvenile phase leading to the definitive color pattern. Remarkably, the reduction of stripe number over ontogeny matches the sequences of stripe losses during evolution, showing that color pattern diversification among clownfish lineages results from changes in developmental processes. Finally, we reveal that the diversity of striped patterns plays a key role for species recognition. Conclusions Overall, our findings illustrate how developmental, ecological, and social processes have shaped the diversification of color patterns during the radiation of an emblematic coral reef fish lineage.

At Makemo Atoll (French Polynesia), the carapid fish Onuxodon fowleri lives in symbiosis with the black-lip pearl oyster Pinctada margaritifera. Although the symbiont seems to live inside its host bivalve by using it as a shelter, additional data are still needed to better understand the exact nature of this association. For this purpose, we implemented an approach using stable isotope ratios of carbon (13C/12C) and nitrogen (15N/14N). The δ13C and δ15N values were measured in tissues of the pearl oyster (gonads, gills, mantle and muscles), white muscle tissue from the fish and other food sources. This stable isotope approach was also complemented by the analysis of stomach contents in the carapid fish. Overall, the isotopic compositions measured in the present study support a commensal relationship between O. fowleri and P. margaritifera. In addition, our isotopic data bring new information about another guest living inside P. margaritifera, namely the palaemonid shrimp Conchodytes meleagrinae. Based on the δ13C and δ15N values, it appears that the shrimp might feed on the bivalve gonads.

Coastal urbanisation is a widespread phenomenon throughout the world and is often linked to increased erosion. Small Pacific islands are not spared from this issue, which is of great importance in the context of climate change. The French Polynesian island of Bora Bora was used as a case study to investigate the historical evolution of its coastline classification and position from 1955 to 2019. A time series of very high-resolution aerial imagery was processed to highlight the changes of the island’s coastline. The overall length of natural shores, including beaches, decreased by 46% from 1955 to 2019 while human-made shores such as seawalls increased by 476%, and as of 2019 represented 61% of the coastline. This evolution alters sedimentary processes: the time series of aerial images highlights increased erosion in the vicinity of seawalls and embankments, leading to the incremental need to construct additional walls. In addition, the gradual removal of natural shoreline types modifies landscapes and may negatively impact marine biodiversity. Through documenting coastal changes to Bora Bora over time, this study highlights the impacts of human-made structures on erosional processes and underscores the need for sustainable coastal management plans in French Polynesia.

Abstract Lateralization, i.e. the preferential use of one side of the body, may convey fitness benefits for organisms within rapidly-changing environments, by optimizing separate and parallel processing of different information between the two brain hemispheres. In coral reef-fishes, the movement of larvae from planktonic to reef environments (recruitment) represents a major life-history transition. This transition requires larvae to rapidly identify and respond to sensory cues to select a suitable habitat that facilitates survival and growth. This ‘recruitment’ is critical for population persistence and resilience. In aquarium experiments, larval Acanthurus triostegus preferentially used their right-eye to investigate a variety of visual stimuli. Despite this, when held in in situ cages with predators, those larvae that previously favored their left-eye exhibited higher survival. These results support the “brain’s right-hemisphere” theory, which predicts that the right-eye (i.e. left-hemisphere) is used to categorize stimuli while the left-eye (i.e. right-hemisphere) is used to inspect novel items and initiate rapid behavioral-responses. While these experiments confirm that being highly lateralized is ecologically advantageous, exposure to chlorpyrifos, a pesticide often inadvertently added to coral-reef waters, impaired visual-lateralization. This suggests that chemical pollutants could impair the brain function of larval fishes during a critical life-history transition, potentially impacting recruitment success.
