National audience; INTRODUCTION: The third-generation tyrosine kinase inhibitor (TKI) osimertinib is recommended as a first-line treatment in advanced non-small cell lung cancer harboring an activating mutation of Epidermal Growth Factor Receptor (EGFR). Adverse pulmonary events related to osimertinib exposure have been reported, primarily in Japanese patients. They rarely occur in the Caucasian population. OBSERVATION: Herein we report two clinical cases of osimertinib-induced lung toxicities in patients diagnosed with advanced lung adenocarcinoma harboring an EGFR mutation. In the first case, interstitial pneumonia was asymptomatic and evolved favorably after osimertinib discontinuation. The second patient presented a more extensive form of lung injuries and despite systemic corticosteroid therapy, the evolution was fatal. CONCLUSION: Osimertinib-related lung toxicities remain exceptional. While most forms are mild, consideration of TKI treatment discontinuation may be necessitated. Introduction of another TKI or rechallenge with osimertinib might be considered along with corticosteroid therapy if necessary. Diffuse alveolar damage is a pejorative prognostic factor.

- Abstract
OBJECTIVES: The F386L PSEN1 variant has been reported in 1 Japanese family with limited clinical information. We aimed to prove that F386L is pathogenic by demonstrating that it segregates with early-onset Alzheimer disease (AD). METHODS: Eight individuals in a South Asian family provided DNA for genetic testing and underwent a neurologic examination. RESULTS: The female proband was diagnosed with AD at age 45 years and died at age 49 years. She had a CSF biomarker profile consistent with AD, and her florbetaben PET scan was amyloid positive with high uptake in the striatum. Her MRI showed no prominent white matter disease. Her affected relatives had an age at onset range of 38-57 years and had imaging and biomarker profiles similar to hers. DISCUSSION: The results presented here, in conjunction with the prior report, confirm the pathogenicity of F386L. Furthermore, our study highlights the importance of studying families from underrepresented populations to identify or confirm the pathogenicity of rare variants that may be specific to certain genetic ancestries.

International audience; Imaging genetic studies of large control cohorts such as UK Biobank enable to assess the range of normal variations in brain structures. Previous studies by our group have shown that the width of several cortical sulci is associated with a variant in the upstream region of KCNK2 gene even if this effect is corrected with age. Here we propose to analyze in a multivariate setup the associations between sets of genetic variants and multiple sulci widths. The genetic variants we consider are sets of SNPs of known phase called haplotypes, taken from the upstream region of KCNK2 gene. To the best of our knowledge, multivariate analysis in imaging genetics has never been used in haplotype studies. Our method was able to recover the expected association signal and uncover new associations between imaging data and genetic variants.

The ANTARES neutrino telescope is a large photomultiplier array designed to detect neutrino-induced upward-going muons by their Cherenkov radiation. Understanding the absorption and scattering of light in the deep Mediterranean is fundamental to optimising the design and performance of the detector. This paper presents measurements of blue and UV light transmission at the ANTARES site taken between 1997 and 2000. The derived values for the scattering length and the angular distribution of particulate scattering were found to be highly correlated, and results are therefore presented in terms of an absorption length lambda_abs and an effective scattering length lambda_sct^eff. The values for blue (UV) light are found to be lambda_abs ~ 60(26) m, lambda_sct^eff ~ 265(122) m, with significant (15%) time variability. Finally, the results of ANTARES simulations showing the effect of these water properties on the anticipated performance of the detector are presented., Comment: 40 pages, 23 figures (including 4 available as separate figures), to be published in Astroparticle Physics

International audience; BACKGROUND: There is no standard second-line treatment for small cell lung cancer (SCLC). The prognosis of these patients is poor and special attention should be paid to both quality of life and economic factors. METHODS: The aim of this phase II randomised trial (GFPC0501) is to compare, in patients with progressive SCLC after first-line platinum based chemotherapy, oral multi drug chemotherapy (CCNU, cyclophosphamide, etoposide) and classical intravenous chemotherapy with cyclophosphamide, doxorubicin and vincristine (CAV) in terms of tolerability, efficacy (response rate, median one year survival and overall survival), quality of life and consumption of health care resources. Based on a two-stage Bryant and Day approach, this study will require a total of 138 patients with an interim analysis of the first 38. EXPECTED RESULTS: This trial will provide information on several aspects of second-line chemotherapy for patients with SCLC. Thirty six patients have been enrolled in 16 centres by December 2006 and the results of the interim analysis will be available in June 2007.

- Abstract
The ANTARES Neutrino Telescope was completed in May 2008 and is the first operational Neutrino Telescope in the Mediterranean Sea. The main purpose of the detector is to perform neutrino astronomy and the apparatus also offers facilities for marine and Earth sciences. This paper describes the design, the construction and the installation of the telescope in the deep sea, offshore from Toulon in France. An illustration of the detector performance is given. The ANTARES Collaboration proposes to construct a large area water Cherenkov detector in the deep Mediterranean Sea, optimised for the detection of muons from high-energy astrophysical neutrinos. This paper presents the scientific motivation for building such a device, along with a review of the technical issues involved in its design and construction. The observation of high energy neutrinos will open a new window on the universe. The primary aim is to study particle acceleration mechanisms in energetic astrophysical objects such as AGN's and GRB's, which may also shed light on the origin of ultra-high-energy cosmic rays. At lower energies, non-baryonic dark matter may be detected through the neutrinos produced when gravitationally captured WIMPs annihilate in the cores of the Earth and the Sun. Neutrino oscillations can be measured by studying distortions in the energy spectrum of upward-going atmospheric nu's. The characteristics of the proposed site are an important consideration in detector design. Water properties and detector environment parameters were measured. These tests have shown that the proposed site provides a good-quality environment for the detector, and have also demonstrated the feasibility of the deployment technique. The present proposal concerns the construction and deployment of a detector with surface area 0.1 km^2. The conceptual design for such a detector is discussed, and the physics performance evaluated. An overview of costs and schedules is presented. It is concluded that a 0.1 km^2 detector is technically feasible at realistic cost, and offers an exciting and varied physics and astrophysics programme. Such a detector will also provide practical experience which will be invaluable in the design and operation of future detectors on the astrophysically desirable 1 km^2 scale.

- Abstract
The ANTARES neutrino telescope is being constructed in the Mediterranean Sea. It consists of a large three-dimensional array of photo-multiplier tubes. The data acquisition system of the detector takes care of the digitisation of the photo-multiplier tube signals, data transport, data filtering, and data storage. The detector is operated using a control program interfaced with all elements. The design and the implementation of the data acquisition system are described. (c) 2006 Elsevier B.V. - Abstract
In 2005, the ANTARES Collaboration deployed and operated at a depth of 2500 m a so-called Mini Instrumentation Line equipped with Optical Modules (MILOM) at the ANTARES site. The various data acquired during the continuous operation from April to December 2005 of the MILOM confirm the satisfactory performance of the Optical Modules, their front-end electronics and readout system. as well as the calibration devices of the detector. The in situ measurement of the Optical Module time response yields a resolution better than 0.5 ns. The performance of the acoustic positioning system, which enables the spatial reconstruction of the ANTARES detector with a precision of about 10 cm, is verified. These results demonstrate that with the full ANTARES neutrino telescope the design angular resolution of better than 0.3 degrees can be realistically achieved.

- Abstract
The ANTARES neutrino telescope is a large photomultiplier array designed to detect neutrino-induced upward-going muons by their Cherenkov radiation. Understanding the absorption and scattering of light in the deep Mediterranean is fundamental to optimising the design and performance of the detector. This paper presents measurements of blue and UV light transmission at the ANTARES site taken between 1997 and 2000. The derived values for the scattering length and the angular distribution of particulate scattering were found to be highly correlated, and results are therefore presented in terms of an absorption length;,ab, and an effective scattering length lambda(sct)(eff). The values for blue (UV) light are found to be lambda(abs) similar or equal to 60(26) m, lambda(sct)(eff similar or equal to) 265(122) m, with significant (similar to15%) time variability. Finally, the results of ANTARES simulations showing the effect of these water properties on the anticipated performance of the detector are presented. (C) 2004 Elsevier B.V. All rights reserved.

- Abstract
The ANTARES neutrino telescope, to be immersed depth in the Mediterranean Sea, will consist of a three-dimensional matrix of 900 large area photomultiplier tubes housed in pressure-resistant glass spheres. The selection of the optimal photomultiplier was a critical step for the project and required an intensive phase of tests and developments carried out in close collaboration with the main manufacturers worldwide. This paper provides an overview of the tests performed by the collaboration and describes in detail the features of the photomultiplier tube chosen for ANTARES. (c) 2005 Elsevier B.V. Introduction : L’inhibiteur de tyrosine kinase (TKI) de troisième génération, osimertinib, est recommandé en première intention dans la prise en charge des carcinomes bronchiques métastatiques présentant une mutation Epidermal Growth Factor Receptor (EGFR). La toxicité pulmonaire de l’osimertinib, comme les autres traitements de la même classe thérapeutique est principalement décrite chez les patients japonais. Elle demeure rare dans la population caucasienne.
