T cell–mediated immunity is a major component of antitumor immunity. In order to be efficient, effector T cells must leave the circulation and enter into the tumor tissue. Regulatory T cells (Treg) from gastric cancer patients, but not from healthy volunteers, potently inhibit migration of conventional T cells through activated endothelium. In this study, we compared T cells from colon cancer patients and healthy donors to determine the mechanisms used by Tregs from cancer patients to inhibit conventional T-cell migration. Our results showed that circulating Tregs from cancer patients expressed high levels of CD39, an ectoenzyme mediating hydrolysis of ATP to AMP, as a rate-determining first step in the generation of immunosuppressive adenosine. Tumor-associated Tregs expressed even more CD39, and we therefore examined the importance of adenosine in Treg-mediated inhibition of T-cell transendothelial migration in vitro. Exogenous adenosine significantly reduced migration of conventional T cells from healthy volunteers, and blocking either adenosine receptors or CD39 enzymatic activity during transmigration restored the ability of conventional T cells from cancer patients to migrate. Adenosine did not directly affect T cells or endothelial cells, but reduced the ability of monocytes to activate the endothelium. Taken together, our results indicate that Treg-derived adenosine acts on monocytes and contributes to reduced transendothelial migration of effector T cells into tumors. This effect of Tregs is specific for cancer patients, and our results indicate that Tregs may affect not only T-cell effector functions but also their migration into tumors. Background and Aims Uncontrolled activation of intestinal mononuclear phagocytes [MNPs] drives chronic inflammation in inflammatory bowel disease [IBD]. Triggering receptor expressed on myeloid cells 1 [TREM-1] has been implicated in the pathogenesis of IBD. However, the role of TREM-1+ cell subsets in driving IBD pathology and the link with clinical parameters are not understood. We investigated TREM-1 expression in human intestinal MNP subsets and examined blocking TREM-1 as a potential IBD therapy. Methods TREM-1 gene expression was analysed in intestinal mucosa, enriched epithelial and lamina propria [LP] layers, and purified cells from controls and IBD patients. TREM-1 protein on immune cells was assessed by flow cytometry and immunofluorescence microscopy. Blood monocyte activation was examined by large-scale gene expression using a TREM-1 agonist or LP conditioned media [LP-CM] from patients in the presence or absence of TREM-1 and tumour necrosis factor [TNF] antagonist antibodies. Results TREM-1 gene expression increases in intestinal mucosa from IBD patients and correlates with disease score. TREM-1+ cells, which are mainly immature macrophages and CD11b+ granulocytes, increase among LP cells from Crohn’s disease patients and their frequency correlates with inflammatory molecules in LP-CM. LP-CM from Crohn’s disease patients induces an inflammatory transcriptome in blood monocytes, including increased IL-6 expression, which is reduced by simultaneous blocking of TREM-1 and TNF. Conclusions High intestinal TREM-1 expression, reflecting a high frequency of TREM-1+ immature macrophages and TREM-1+CD11b+ granulocytes, is linked to the deleterious inflammatory microenvironment in IBD patients. Therefore, blocking the TREM-1 pathway, especially simultaneously with anti-TNF therapy, has potential as a new IBD therapy.

Perovskite-type oxyhydrides, BaTiO3−xHx, represent a novel class of hydride ion conducting materials of interest for several electrochemical applications, but fundamental questions surrounding the defect chemistry and hydride ion transport mechanism remain unclear. Here we report results from powder X-ray diffraction, thermal gravimetric analysis, nuclear magnetic resonance spectroscopy, inelastic neutron scattering (INS), and density functional theory (DFT) simulations on three metal hydride reduced BaTiO3 samples characterized by the simultaneous presence of hydride ions and oxygen vacancies. The INS spectra are characterized by two predominating bands at around 114 (ω⊥) and 128 (ω∥) meV, assigned as fundamental Ti–H vibrational modes perpendicular and parallel to the Ti–H–Ti bond direction, respectively, and four additional, weaker, bands at around 99 (ω1), 110 (ω2), 137 (ω3) and 145 (ω4) meV that originate from a range of different local structures associated with different configurations of the hydride ions and oxygen vacancies in the materials. Crucially, the combined analyses of INS and DFT data confirm the presence of both nearest and next-nearest neighbouring oxygen vacancies to the hydride ions. This supports previous findings from quasielastic neutron scattering experiments, that the hydride ion transport is governed by jump diffusion dynamics between neighbouring and next-nearest neighbouring hydride ion–oxygen vacancy local structures. Furthermore, the investigation of the momentum transfer dependence of the INS spectrum is used to derive the mean square displacement of the hydride ions, which is shown to be in excellent agreement with the calculations. Analysis of the mean square displacement confirms that the hydrogen vibrational motions are localized in nature and only very weakly affected by the dynamics of the surrounding perovskite structure. This insight motivates efforts to identify alternative host lattices that allow for a less localization of the hydride ions as a route to higher hydride ion conductivities.

Acceptor-doped barium zirconate-based proton conductors are currently receiving considerable attention because of their promise as electrolytes in future electrochemical devices, such as solid oxide fuel cells, but the defect chemistry, especially in regard to the local coordination environment and dynamics of protons in these materials, is unclear. Here, we investigate the local coordination environments and vibrational dynamics of protons in samples of the proton conducting material BaZr1-xMxO3Hx with M = In and Sc, and x = 0.1 and 0.5, using inelastic neutron scattering (INS), infrared (IR), and Raman spectroscopy together with ab initio molecular dynamics (AIMD) simulations. The local coordination of protons is shown to exhibit a rather peculiar dependence on the type and concentration of dopant atoms, as they are found to be similar for BaZr1-xScxO3Hx with x = 0.1 and 0.5 and BaZr1-xInxO3Hx with x = 0.1, whereas for BaZr1-xInxO3Hx with x = 0.5 additional proton sites seem to be present. It is argued that these additional proton sites are characterized by local structural arrangements reminiscent of the fully In-substituted material BaInO3H. The presence of these local structural arrangements points toward different local proton mobilities between BaZr1-xInxO3Hx with x = 0.5 and the other three materials and a higher rate of proton transfer events in brownrnillerite-type local structures.

Barium zirconate (BaZrO3), when substituted with trivalent acceptor ions to replace Zr4+, is a proton conducting material of interest for several electrochemical applications. The local coordination environments, and vibrational dynamics, of the protons are known to critically influence the material's proton conducting properties, however, the nature of the static and dynamic structure around the protons and, especially, how it is affected by the dopant atoms for high doping concentrations, remains to be elucidated. Here we report results from X-ray powder diffraction, infrared (IR) spectroscopy, inelastic neutron scattering (INS) and ab initio molecular dynamics (AIMD) simulations on a hydrated sample of BaZrO3 substituted with 50% In3+. The investigation of the momentum-transfer (Q) dependence of the INS spectrum is used to aid the analysis of the spectra and the assignment of the spectral components to fundamental O–H bend and O–H stretch modes and higher-order transitions. The AIMD simulations show that the INS spectrum is constituted of the overlapping spectra of protons in several different local structural environments, whereas the local proton environments for specific protons are found to vary with time as a result of thermally activated vibrations of the perovskite lattice. It is argued that, converse to more weakly doped systems, such as 20% Y-doped BaZrO3, the dopant–proton association effect does not hinder the diffusion of protons due to the presence of percolation paths of dopant atoms throughout the perovskite lattice.

We report results from visible and UV Raman spectroscopy studies of the phonon spectra of a polycrystalline sample of the prototypical perovskite type oxide BaZrO3 and a 500 nm thick film of its Y-doped, proton conducting, counterpart BaZr0.8Y0.2O2.9. Analysis of the Raman spectra measured using different excitation energies (between 3.44 eV and 5.17 eV) reveals the activation of strong resonance Raman effects involving all lattice vibrational modes. Specifically, two characteristic energies were identified for BaZrO3, one around 5 eV and one at higher energy, respectively, and one for BaZr0.8Y0.2O2.9, above 5 eV. Apart from the large difference in spectral intensity between the non-resonant and resonant conditions, the spectra are overall similar to each other, suggesting that the vibrational spectra of the perovskites are stable when investigated using an UV laser as excitation source. These results encourage further use of UV Raman spectroscopy as a novel approach for the study of lattice vibrational dynamics and local structure in proton conducting perovskites, and open up for, e.g., time-resolved experiments on thin films targeted at understanding the role of lattice vibrations in proton transport in these kinds of materials.

The structure and dehydration mechanism of the proton conducting oxide Ba2In2O5(H2O)(x) are investigated by means of variable temperature (20-600 degrees C) Raman spectroscopy together with thermal gravimetric analysis and inelastic neutron scattering. At room temperature, Ba2In2O5(H2O)(x) is found to be fully hydrated (x = 1) and to have a perovskite-like structure, which dehydrates gradually with increasing temperature and at around 600 degrees C the material is essentially dehydrated (x approximate to 0.2). The dehydrated material exhibits a brownmillerite structure, which is featured by alternating layers of InO6 octahedra and InO4 tetrahedra. The transition from a perovskite-like to a brownmillerite-like structure upon increasing temperature occurs through the formation of an intermediate phase at ca. 370 degrees C, corresponding to a hydration degree of approximately 50%. The structure of the intermediate phase is similar to the structure of the dehydrated material, but with the difference that it exhibits a non-centrosymmetric distortion of the InO6 octahedra that is not present in the dehydrated material. The dehydration process upon heating is a two-stage mechanism; for temperatures below the hydrated-to-intermediate phase transition, dehydration is characterized by a homogenous release of protons over the entire oxide lattice, whereas above the transition a preferential desorption of protons originating in the nominally tetrahedral layers is observed. Furthermore, our spectroscopic results point towards the co-existence of two structural phases, which relate to the two lowest-energy proton configurations in the material. The relative contributions of the two proton configurations depend on how the sample is hydrated.

Disruption of the homeostatic balance of intestinal dendritic cells (DCs) and macrophages (MQs) may contribute to inflammatory bowel disease. We characterized DC and MQ populations, including their ability to produce retinoic acid, in clinical material encompassing Crohn’s ileitis, Crohn’s colitis and ulcerative colitis (UC) as well as mesenteric lymph nodes (MLNs) draining these sites. Increased CD14+DRint MQs characterized inflamed intestinal mucosa while total CD141+ or CD1c+ DCs numbers were unchanged. However, CD103+ DCs, including CD141+CD103+ and CD1c+CD103+ DCs, were reduced in inflamed intestine. In MLNs, two CD14− DC populations were identified: CD11cintHLADRhi and CD11chiHLADRint cells. A marked increase of CD11chiHLADRint DC, particularly DRintCD1c+ DCs, characterized MLNs draining inflamed intestine. The fraction of DC and MQ populations expressing aldehyde dehydrogenase (ALDH) activity, reflecting retinoic acid synthesis, in UC colon, both in active disease and remission, were reduced compared to controls and inflamed Crohn’s colon. In contrast, no difference in the frequency of ALDH+ cells among blood precursors was detected between UC patients and non-inflamed controls. This suggests that ALDH activity in myeloid cells in the colon of UC patients, regardless of whether the disease is active or in remission, is influenced by the intestinal environment.

AIM: Treatment of Crohn's anal fistula remains challenging and little is known about factors associated with healing. The aim of this study was to assess the rate of healing after surgical treatment and analyse clinical variables related to healing.METHOD: 119 patients (63 female, mean age 36 (±13.7) years with histopathologically verified Crohn's disease underwent a surgical procedure for anal fistula at four main referral centres in Sweden, January 1998 - December 2009. Baseline and treatment-related variables were recorded and analysed for correlation with fistula healing at a final follow-up at a mean 7.2 (median 7.1, 1.0-17.5) years.RESULTS: Of the 119 patients 62 (52%) were healed at final follow-up. Fourteen healed after one procedure and the remaining 48 healed after a further median of 4.0 (2-20) procedures. Ten (8%) patients were subjected to a proctectomy. Final healing was more common in patients operated with a procedure aiming at eradicating the fistula (P=0.0001), without proctitis (P=0.02), and a shorter duration of Crohn' disease (P=.0019).CONCLUSION: Long-term healing of a Crohn's anal fistula can be expected in about half of the patients, usually after repeated surgical treatment. The probability for cure was higher when a curative operation was performed in a patient without proctitis and a shorter duration of Crohn's disease. Mucosa-associated invariant T (MAIT) cells are innate-like T cells with a conserved TCR α-chain recognizing bacterial metabolites presented on the invariant MHC-related 1 molecule. MAIT cells are present in intestinal tissues and liver, and they rapidly secrete IFN-γ and IL-17 in response to bacterial insult. In colon cancer, IL-17–driven inflammation promotes tumor progression, whereas IFN-γ production is essential for antitumor immunity. Thus, tumor-associated MAIT cells may affect antitumor immune responses by their secreted cytokines. However, the knowledge of MAIT cell presence and function in tumors is virtually absent. In this study, we determined the frequency, phenotype, and functional capacity of MAIT cells in colon adenocarcinomas and unaffected colon lamina propria. Flow cytometric analyses showed significant accumulation of MAIT cells in tumor tissue, irrespective of tumor stage or localization. Colonic MAIT cells displayed an activated memory phenotype and expression of chemokine receptors CCR6 and CCR9. Most MAIT cells in unaffected colon tissues produced IFN-γ, whereas only few produced IL-17. Colonic MAIT cells also produced TNF-α, IL-2, and granzyme B. In the tumors, significantly lower frequencies of IFN-γ–producing MAIT cells were seen, whereas there were no differences in the other cytokines analyzed, and in vitro studies showed that secreted factors from tumor tissue reduced IFN-γ production from MAIT cells. In conclusion, MAIT cells infiltrate colon tumors but their ability to produce IFN-γ is substantially reduced. We suggest that MAIT cells have the capacity to promote local immune responses to tumors, but factors in the tumor microenvironment act to reduce MAIT cell IFN-γ production.

The crystal and magnetic structures of multiferroic Bi0.9La0.1FeO3 have been studied using high resolution neutron powder diffraction in the pressure range 0-8 GPa. Two structural phase transitions are observed. The first, at similar to 1 GPa, transforms the polar R3c structure to an antipolar PbZrO3-like root 2a(p) x 2 root 2a(p) x 2a(p) perovskite superstructure; the second, at similar to 5 GPa, results in a smaller, root 2a(p) x root 2a(p) x 2a(p) unit cell and a structure described with Ibmm (nonstandard setting of Imma) symmetry, in which the a(-)a(-)b(0) octahedral tilt system is retained and the antipolar cation displacements lost. Accompanying the changes in the nuclear structure, the antiferromagnetic spin structure evolves from a cycloid, with a modulation length, lambda approximate to 770 angstrom, to collinear arrangements with the moments aligned along the b-axis (Pbam) and the a-axis (Ibmm) of the orthorhombic unit cells. In comparison with BiFeO3 the transition from a rhombohedral to an orthorhombic structure is suppressed by similar to 3 GPa, reflecting the dilution of the stereochemically active bismuth lone pair by lanthanum. The vibrational spectra and short-range structure of the brownmillerite-type oxide Ba2In2O6 and its hydrated form BaInO3H, are investigated by means of Raman, infrared, and inelastic neutron scattering spectroscopies together with density functional theory calculations. For Ba2In2O6, which may be described as an oxygen deficient perovskite structure with alternating layers of InO6 octahedra and InO4 tetrahedra, the results affirm a short-range structure of Icmm symmetry, which is characterized by random orientation of successive layers of InO4 tetrahedra. For the hydrated, proton conducting, form, BaInO3H, the results suggest that the short-range structure is more complicated than the P4/mbm symmetry that has been proposed previously on the basis of neutron diffraction, but rather suggest a proton configuration close to the lowest energy structure predicted by Martinez et al. [J.-R. Martinez, C. E. Moen, S. Stoelen, N. L. Allan, J. Solid State Chem., 180, 3388, (2007)]. An intense Raman active vibration at 150 cm(-1) is identified as a unique fingerprint of this proton configuration.

Recruitment of immune cells to tumors is a complex process crucial for both inflammation-driven tumor progression and specific anti-tumor cytotoxicity. Chemokines control the directed migration of immune cells, and their actions are partly controlled by nonsignaling chemokine decoy receptors. The role of the receptors such as D6, Duffy antigen receptor for chemokines and ChemoCentryx chemokine receptor in immunity to tumors is still unclear. Using real-time PCR, we detected significantly decreased expression of D6 mRNA in colon tumors compared to unaffected mucosa. D6 protein was expressed by lymphatic endothelium and mononuclear cells in the colon lamina propria and detected by immunohistochemistry in two out of six tissue samples containing high D6 mRNA levels, whereas no staining was observed in any tissue samples expressing low mRNA levels. When examining the density of lymphatic vessels in colon tumors, we detected a marked increase in vessels identified by the lymphatic endothelial marker Lyve-1, excluding passive regulation of D6 due to decreased lymphatic vessel density. In parallel, the Treg-recruiting chemokine CCL22, which is sequestered by D6, was threefold increased in tumor tissue. Furthermore, we could show that low D6 expression correlated to more invasive tumors and that tumor location influences D6 expression, which is lower in the more distal parts of the colon. The data support that regulation of D6 by colon tumors results in altered levels of proinflammatory CC chemokines, thereby shaping the local chemokine network to favor tumor survival. This may have implications for the design of future immunotherapy for colon cancer.

Local inflammation is a strong risk factor for the development of gastrointestinal adenocarcinomas. Mucosal regulatory T cells and IgA-secreting cells both contribute to reduce inflammatory responses, and their recruitment to tissues is dependent on local production of chemokines. More specifically, IgA-secreting cells are recruited to mucosal tissues by CCL28 signalling through CCR10. Here, we examined the recruitment of IgA-secreting plasma cells to tumor-associated mucosa in patients suffering from colon adenocarcinoma. Flow cytometric analyses of single cell suspensions from tumor-associated and unaffected colon mucosa showed a marked decrease in CD19(+)CD38(high)IgA(+) plasmablasts in the tumor-associated mucosa, while the total frequencies of B and T cells were similar. This finding was confirmed in ELISPOT assays, demonstrating a 64 % reduction in the frequencies of IgA-secreting cells among cells from the tumor-associated mucosa. The few IgA(+) plasmablasts present in the tumor did not express CCR10, and functional migration assays demonstrated that IgA-secreting cells from tumor-associated mucosa did not migrate in response to CCL28. Taken together, our results show an impaired migration of IgA-secreting cells to colon tumors, presumably caused by a decreased production of CCL28 in the tumor. The lack of local IgA antibodies may lead to impaired barrier function and increased bacterial colonization, driving further inflammatory responses and promoting tumor growth.

Poor pouch function after restorative proctocolectomy for ulcerative colitis is a considerable problem. Pouchitis and functional disorders are the most common reasons. Probiotics seem to have a beneficial effect in pouchitis but have not been assessed in functional pouch disorders. The aim was to analyse the effects of probiotics in patients with poor pouch function.Thirty-three patients were randomized to probiotics (Lactobacillus plantarum 299 and Bifidobacterium infantis Cure 21) or placebo in a double blinded, 1:1 fashion. The treatment effect was assessed by the pouch functional score (PFS; 0-15, 15 worst), pouchitis disease activity index (PDAI; 0-18, 18 worst), and levels of four faecal biomarkers of inflammation (calprotectin, lactoferrin, myeloperoxidase [MPO] and eosinophilic cationic protein [ECP]).Thirty-two patients were included (probiotics = 17, placebo = 16). There was no difference in change in the PFS from before to after treatment between the groups (median difference: -1.00, 95% C.I. -3.00 to 0.00, p = 0.119). Furthermore, probiotics had no effect on PDAI (median difference: 0.00, 95% C.I. 0.00-1.00, p = 0.786), or on faecal biomarkers. Significant correlations were observed between PDAI and each of the faecal biomarkers at study start. There were no correlations between PFS or PDAI symptom subscore and the biomarkers. PDAI endoscopic and histologic subscores correlated significantly to each of the biomarkers.The hypothesis that probiotics improves pouch-related dysfunction was not confirmed. Faecal biomarkers could play a future role in the management of pouch patients.

Background and Aims Failure of ileal pouch-anal anastomosis (IPAA) occurs in around 10% of the patients. Compared to patients with functioning pouches, health related quality of life is deteriorated after failure. Sexual function in patients with pouch failure is however poorly studied. The aim was to study sexual function in patients with pelvic pouch failure; patients with functioning pouches were used as controls. The hypothesis was that patients with pouch failure have worse sexual function. Methods 36 patients with pouch failure were compared with 72 age and sex-matched controls with ulcerative colitis and functioning pouches. The patients answered a set of questionnaires concerning sexual function (Female Sexual Function Index [FSFI] and International Index of Erectile Function [IIEF]), body image (BIS-scale) and health-related quality of life (SF-36). Results Both women and men with pouch failure scored lower than controls in the FSFI and IIEF questionnaires. However, none of the observations were statistically significant. The scores in the failure group (for both sexes) were below the cut-off level for sexual dysfunction. Scores for the BIS instrument were significantly lower for both sexes in the failure group. Women and men in the failure group scored lower than the controls in all domains of the SF-36, however statistically significant only for the social function domain in men. Conclusions The hypothesis, that a failed IPAA is associated with worse sexual function, was not confirmed. Compared to patients with functioning pouches, patients with pouch failure have inferior body image.

Proton motions in hydrated proton conducting perovskites BaZr(0.90)A(0.10)O(2.95) (A = Y and Sc) have been investigated using quasielastic neutron scattering. The results reveal a localized motion on the ps time scale and with an activation energy of similar to 10-30 meV, in both materials. The temperature dependence of the total mean square displacement of the protons shows an onset of this motion at a temperature of about 300 K. The low activation energy, much lower than the activation energy for the macroscopic proton conductivity, suggests that this motion is not the rate-limiting process for the long-range proton diffusion, i.e. it is not linked to the two materials significantly different proton conductivities. In fact, a comparison of the QENS results with density functional theory calculations indicates that for both materials the observed motion may be ascribed to intra-octahedral proton transfers occurring close to a dopant atom. Using a combination of neutron diffraction, infrared spectroscopy, and first-principles calculations, we have investigated the structure of hydrated and dehydrated proton conducting alkali thio-hydroxogermanates of general formula M2GeS2(OH)2·yH2O (M = K, Rb, and Cs). The results show that the structure of hydrated and dry materials are basically the same, which confirms previous indications that the main effect of heating these materials is just a loss of water. We suggest that in the hydrated state the structure of these materials is built of dimers of thio-hydroxogermanate anions, with the water molecules acting as bridges between such dimers. In the dehydrated structure, the thio-hydroxogermanate anions instead form an extended network through the formation of interdimer hydrogen bonds through the −OH groups in the structure. The alkali ions are suggested to act as “space-fillers” in voids formed by the thio-hydroxogermanate anion dimers, in both the hydrated and the dehydrated state.

Solid-state sintering has been used to prepare the perovskite BaZr0.9Sc0.1O3-delta. Analysis of X-ray powder diffraction data shows that an increase of the unit cell parameter, a, was observed after deuteration. Rietveld analysis of room-temperature neutron powder diffraction data confirmed cubic symmetry (space group Pm-3m). Dynamic thermogravimetric analysis indicates that the hydration process occurs below 335 degrees C and approximately 58% of the theoretical number of protonic defects can be filled. The presence of protons/deutrons is seen from the strong O-H/O-D stretch band in the infrared spectrum of the hydrated/deuterated samples. In a systematic study, we investigate the effect of dopant level and hydration on the short-range structure of the proton-conducting perovskite-type oxide BaInxZr1-xO3-x/2 (x = 0 - 0.75), using infrared and Raman spectroscopy. The results show that the doping leads to significant local distortions of the average cubic structure of these materials. By increasing the In concentration from x = 0 to x = 0.75, new bands appear and grow in intensity in both the IR and the Raman spectra, showing that the local distortions become successively more and more pronounced. The structural distortions are largely uncorrelated to the presence of oxygen vacancies, but instead are mainly driven by the size and charge difference between the In3+ and Zr4+ ions, which leads to displacements of the cations and to tilting of the (In/Zr)O-6 octahedra. On the basis of our results, we suggest that there is a threshold between x = 0.10 and x = 0.25 where the structural distortions propagate throughout the whole perovskite structure. Comparison of our spectroscopic data with the proton conductivity for the same materials indicates that the presence of extended structural distortions is favorable for fast proton transport.

High-resolution neutron powder diffraction data have been collected on deuterated and dried samples of the perovskite BaZr0.5In0.5O2.75 at 5 K and room temperature, respectively. Inspection of Fourier nuclear density maps for the deuterated phase have allowed the deuteron position to be refined on a 12h (1/2, y, 0) crystallographic site, with y = 0.217(4) yielding a chemically reasonable O-D distance of 0.92(2) angstrom. Evidence for anisotropy of the deuteron position is also found consistent with a 24k crystallographic site (0.56, 0.21, 0) indicative of displacements of the ion towards neighbouring oxygen ions. The presence of static oxygen disorder in both the dried and deuterated samples is apparent from the structural analyses. Raman spectra confirm short range deviations from cubic symmetry for both dried and hydrated samples.

Introduction. The aetiology and pathogenesis of ulcerative colitis (UC) are essentially unknown. Galectins are carbohydrate-binding lectins involved in a large number of physiological and pathophysiological processes. Little is known about the role of galectins in human UC. In this immunohistochemical exploratory study, both epithelial and inflammatory cell galectin expression were studied in patients with a thoroughly documented clinical history and were correlated with inflammatory activity.Material and Methods. Surgical whole intestinal wall colon specimens from UC patients (n=22) and controls (n=10) were studied. Clinical history, pharmacological treatment, and modified Mayo-score were recorded. Tissue inflammation was graded, and sections were stained with antibodies recognizing galectin-1, galectin-2, galectin-3, and galectin-4.Results. Galectin-1 was undetectable in normal and UC colonic epithelium, while galectin-2, galectin-3, and galectin-4 were strongly expressed. A tendency towards diminished epithelial expression with increased inflammatory grade for galectin-2, galectin-3, and galectin-4 was also found. In the inflammatory cells, a strong expression of galectin-2 and a weak expression of galectin-3 were seen. No clear-cut correlation between epithelial galectin expression and severity of the disease was found.Conclusion. Galectin expression in patients with UC seems to be more dependent on disease focality and individual variation than on degree of tissue inflammation.

Epitaxial thin films of the proton conducting perovskite BaZr0.53In0.47O3-dH0.47–2d grown by pulsed laser deposition were investigated in their hydrated and dehydrated conditions through a multitechnique approach with the aim to study the structure and proton concentration depth profile and their relationship to proton conductivity. The techniques used were X ray diffraction X ray and neutron reflectivity nuclear reaction analysis and Rutherford backscattering together with impedance spectroscopy. The obtained proton conductivity and activation energy are comparable to literature values for the bulk conductivity of similar materials thus showing that grain boundary conductivity is negligible due to the high crystallinity of the film. The results reveal an uneven proton concentration depth profile with the presence of a 3–4 nm thick proton rich layer with altered composition likely characterized by cationic deficiency. While this surface layer either retains or reobtains protons after desorption and cooling to room temperature the bulk of the film absorbs and desorbs protons in the expected manner. It is suggested that the protons in the near surface proton rich region are located in proton sites characterized by relatively strong O–H bonds due to weak hydrogen bond interactions to neighboring oxygen atoms and that the mobility of protons in these sites is generally lower than in proton sites associated with stronger hydrogen bonds. It follows that strongly hydrogen bonding configurations are important for high proton mobility.

Failure after ileal pouch-anal anastomosis (IPAA) is reported with a frequency of 10-20%. The failed IPAA can be excised or defunctioned. Indications for excision and further management of an indefinitely diverted pouch are poorly described. The aim of the present investigation was to investigate pouch-related problems and the histopathological pattern of the pouch mucosa in this group of patients.In a cohort of 620 patients having IPAA with a median follow-up of 14 years, 56 patients with failure were identified. The patients with defunctioned pouches were assessed with regard to pouch-related problems and endoscopy with biopsies was performed. Biopsies were stained with haematoxylin-eosin, PAS for neutral mucins and Alcian blue/high iron diamine for sialomucins/sulphomucins. Morphological changes were grouped into three types modified according to Veress and assessed for dysplasia.Twenty-two patients with an indefinitely diverted pouch were found. The follow-up time after surgery for failure was 10 years. Thirteen patients completed the follow-up. Except for two patients with pelvic/perineal pain, there were no clinical problems. The majority of patients displayed mild to moderate macroscopic signs of inflammation. Morphologically, findings ranged from a preserved mucosal pattern to intense inflammatory reaction. No case of dysplasia or carcinoma was found.Most patients with an indefinitely diverted pouch had no complaints regarding the pouch. There was no case of dysplasia. Indefinite diversion may be preferable to pouch excision, especially given the associated morbidity.

Polymer composite proton conducting membranes with increasing amount of tungstophosphoric acid (PWA) have been synthesized starting from poly(vinylidene) fluoride and alumina. Membranes could be prepared with an acid loading up to 16.7 wt.%. The conductivity of the membranes increases with acid loading up to 10- 3 Scm- 1 at the highest loading. The membrane matrices have a good thermal stability up to 240 °C, however a loss of water absorbed during the preparation procedure is observed at a considerably lower temperature. Raman spectroscopy showed that in the membrane the polymer adopts a conformation that could promote a separation into hydrophilic/hydrophobic sites. An interaction between alumina and PWA is also found that might limit the conductivity of the membranes.

The structure of Cs 2 GeS 2 (OH) 2 · y H 2 O in hydrous and anhydrous states has been investigated by means of neutron diffraction and infrared spectroscopy. This material belongs to a new class of intermediate temperature highly proton conducting alkali thio-hydroxogermanates. The results show that this material is amorphous, and that both the short and intermediate range structure of the hydrated and dehydrated states of the materials are largely the same. Moreover, the results point to a porous structure built up of thio-oxoanions and alkali ions. In the hydrated state, the water molecules appear to be confined to cages with a characteristic length of 7 A.

Background Long-term pouch function and physiological characteristics after ileal pouch–anal anastomosis (IPAA) are poorly described. The aim of this study was to undertake a prospective investigation of long-term pouch function and manovolumetric characteristics. Methods Forty-two patients with a median follow-up of 16 years after IPAA were included. Function was assessed using a questionnaire and a score was calculated ranging from 0 to 15 (15 being the worst). Manovolumetry was performed and pouchitis recorded. A paired analysis was conducted, as the results were compared with previous data for each patient. Results The median functional score was 3·5 (range 0–10) at 2 years and 5 (range 1–11) at 16 years (P = 0·013). Resting anal canal pressures were higher (P < 0·001) and squeeze pressures lower (P = 0·008) at long-term follow-up. Ileal pouch volumes at distension pressures of 10, 20 and 40 cmH2O were diminished at 16 years (P < 0·001, P = 0·005 and P = 0·058 respectively). The volume and pressure for first sensation and urge to defaecate were reduced. Increased age correlated positively with a poor functional score. A history of pouchitis did not affect functional or physiological characteristics. Conclusion Ileal pouch function declines in the long term. The reasons are unclear, but the ageing process may have an impact.

The temperature dependence of the mean square displacement, 〈u2〉, calculated by elastic neutron scattering for two phosphate glasses, (AgI)x(AgPO3)1-x with x = 0.3, 0.55, is analysed. The studied samples are probed in a wide range of temperatures going from a few tens of K up to the glass transition temperature and well into the undercooled liquid state. In the low temperature regime a solid like behaviour appears, showing a linear temperature dependence of the Debye-Waller factor, while around Tg the onset of a pronounced increase of 〈u2〉 is observed. The temperature dependence of the normalized elastic intensity and the drawn out Debye-Waller factor are fitted and analyzed by the theoretical previsions of Mode Coupling Theory finding a good agreement. © 2006 Elsevier B.V. The proton conductivity and structural features of In3+ substituted BaZrO3 samples, i.e., BaZr1−xInxO3−δ, were investigated. Rietveld analysis of low temperature (10 K) neutron powder diffraction data collected on as-prepared and deuterated samples confirmed cubic symmetry (space group Pm-3m) for all compositions. The level of oxygen vacancies refined in the as-prepared samples were in good agreement with the values expected to conserve charge neutrality, whilst an increase in oxygen occupancy, reflecting the incorporation of OD− species, was obtained for the deuterated materials. An expansion of the unit cell parameter, a, was observed as a function of In3+ doping as well as after the deuteration reaction. The conductivity of pre-hydrated and dry samples was measured using impedance methods. For 25% In-doped BaZrO3, the low T (300 °C) conductivity of the heating cycle of the dried sample was greater than that of the cooling cycle of the pre-hydrated sample indicating a greater number of protons in the nominally dry sample. In contrast, the conductivity values were similar at higher temperatures e.g. T > 500 °C where proton conduction is not dominant.

A scanning near-field optical microscope probe is used to locally excite carrier and photodoped region is associated with lower T c phase (T c = 75 K) via overdoping. Nanoscale characterizations with optical reflectivity reveal that nanodots (30-nm-diameter) are regularly distributed in 50 nm step. The resultant films with photoinduced nanodots enhance J c (at least 5 times larger than the initial one), a situation being similar to strong pinning effects observed in films modified by either ion irradiation or sputtered nanoparticles. These results suggest that photoinduced nanodots with lower T c act as effective pinning centers.

Conflicting views regarding the use of ileorectal anastomosis (IRA) in ulcerative colitis (UC) exist and this controversy prompted us to review our experience, especially against the background of the current tendency to choose the ileal pouch-anal procedure (IPAA).Thirty-two consecutive patients with IRA were studied. Complications, failure rate, reasons for failure and functional results were assessed. The median follow-up time was 3.5 years.The overall complication rate was 28%. The rectum was excised in 4 patients, indicating a failure rate of 12%. The mean daily evacuation frequency was 5.6. Despite urgency occurring in one-third of the patients, continence function was well preserved.Employed on a selective basis, IRA is a safe procedure with low mortality and morbidity and good prospects for success in many patients with UC. The patients must be prepared to submit to life-long rectoscopy surveillance.

Solid state sintering has been used to prepare the cubic perovskite structured compounds BaZr1−xInxO3−δ (0.0 ≤ x ≤ 0.75). Analysis of X-ray powder diffraction (XRPD) data reveals that the unit cell parameter, a, increases linearly with an increased Indium concentration. XRPD data was also used to demonstrate the completion of sample hydration, which was reached when the materials showed a set of single-phase Bragg-peaks. Dynamic thermogravimetric analysis (TGA) data showed that approx. 89% of the total number of available oxygen vacancies can be filled in BaZr1−xInxO3−δ for x = 0.50, and that the maximum water uptake occurs below 300 °C. Rietveld analysis of the room temperature neutron powder diffraction (NPD) data confirmed the average cubic symmetry (space group Pm-3m), and an expansion of the unit cell parameter after the hydration reaction. The strong O–H stretch band, 2500–3500 cm− 1, in the infrared absorbance spectrum clearly manifests the presence of protons in the hydrated material. Proton conductivity of hydrated BaZr1−xInxO3−δ, x = 0.75 was investigated during heating and cooling cycles under dry argon atmosphere. The total conductivity during the heating cycle was nearly two orders of magnitude greater than that of cooling cycle at 300 °C, whilst these values were similar at higher temperatures i.e. T > 600 °C.

The large scale structure of poly(methyl methacrylate)-based gels and gel electrolytes has been investigated using small angle neutron scattering. The data reveal two different structural length scales: the shorter being typically 4–25 A, while the larger is 200–300 A. Comparison between the gel electrolytes, obtained by doping the gels with lithium salt, and the corresponding salt-free systems shows that the determined length scales are very similar. Thus the dissolved salt has only minor influence on the structure of the polymer network. We also discuss the important relations between structure, dynamics and the performance of the gel electrolytes.

We have investigated the thermal stability and reversibility upon dehydration and re-hydration of a novel class of proton conducting alkali thio-hydroxogermanates. The results indicate that no phase transitions or structural degradation occur in the temperature range 25 to 300 °C, and that repeated dehydration and subsequent re-hydration is a reversible process. The dehydration occurs gradually with increasing temperature, starting at about 80 °C. For temperatures above 180 °C the materials are dry, as all molecular water has been dried off. The dehydration process is shown to be reversible and the material can be rehydrated by exposure to air. The thermal stability and reversibility of the dehydration–rehydration process are attractive properties of functional materials, making the proton conducting alkali thio-hydroxogermanates to potential fuel cell electrolytes.

Objective Proctocolectomy and ileal pouch anal anastomosis (IPAA) has become the standard surgery for patients with mucosal ulcerative colitis (MUC). Although there is no absolute age limitation, there are concerns as to its use in elderly patients due to the risks of potential complications and poor function. The aim of this study was to assess the complications and outcome of patients over the age of 70 years with MUC who underwent IPAA. Results in these patients were compared to the results in a group of patients aged less than 70 years who had IPAA. Methods After Institutional Review Board approval, a retrospective review of the medical records of patients with MUC who underwent IPAA was undertaken. Raman and infrared spectroscopy results of protonated and dry samples of the proton conducting double perovskites Sr 3 CaZr 0.5 Ta 1.5 O 8.75 and Ba 3 Ca 1.18 Nb 1.82 O 8.73 (BCN18) are presented. As confirmed by the Raman spectra, the incorporation of protons leads to only small distortions in the host lattice of the perovskites. However, the intensity growth of a band at approximately 650 cm − 1 in the Raman spectrum of Sr 3 CaZr 0.5 Ta 1.5 O 8.75 after protonation may be related to a symmetry reduction towards a monoclinic structure. The O–H stretch region in the infrared spectra of Sr 3 CaZr 0.5 Ta 1.5 O 8.75 and BCN18 are quite different. Stronger bands in the 2000–2500 cm − 1 range in Sr 3 CaZr 0.5 Ta 1.5 O 8.75 may be related to the higher concentration of oxygen vacancies in this material after protonation.

We have investigated new poly-vinylidene fluoride/poly-acrylonitrile (PVdF/PAN) based proton conducting membranes by means of vibrational spectroscopy. We find that a complete phase inversion occurs during the preparation procedure, when the gelling solvents are replaced by an acidic solution, providing the proton conducting property. The uptake of acid is promoted both by the presence of PAN and the ceramic filler, Al 2 O 3 . No particular interaction between the polymer matrix and the acidic solution could be detected, supporting the picture of an inert matrix entrapping a liquid component. However, the dissociation degree of the acid is decreased due to the spatial confinement in the membrane. By comparing the dissociation degree and the actual amount of acid in the membrane to the conductivity, we conclude that the limiting factor for the conductivity is the long-range mobility of the protons, which is governed by the morphology of the membrane.

The relaxation properties of poly(propylene oxide) at temperatures above the glass‐transition temperature are studied by quasi‐elastic neutron scattering and molecular dynamics (MD) simulations. The contributions to the dynamic structure factor from the segmental motion and the methyl group jump rotation are separated using a threefold jump‐rotation model for the methyl side group motion, with a delta function for the distribution of relaxation times, and a stretched exponential function for the segmental motion. This simple model describes momentum transfer and time dependence of the data over a wide temperature range. The results show that with this separation, both neutron and MD data can be well described, and the analysis of both contributions is necessary in order to obtain reliable results for the segmental relaxation and rotational relaxation times at temperature above T g .

The structure of an amorphous polymer electrolyte, poly(propylene oxide) (PPO) complexed with LiClO4, has been studied using reverse Monte Carlo (RMC) simulations. The simulations require no force field but are based on experimental data only, in this case from x-ray and neutron diffraction experiments. Excellent agreement between the experimental data and the structures resulting from the RMC simulation is obtained. Samples with ether-oxygen to lithium concentrations (molar ratios) O:Li=16:1 and 5:1 were studied and compared to results of pure PPO from a previous study. We focus on the effects of the solvated salt on the structure of the polymer matrix, the spatial distribution of ions, and the correlations between the anions and the polymer chains. Analyzing the structures produced in the simulations, we find that for a concentration 16:1, the interchain distance is approximately the same as in pure PPO but more well defined. For a concentration 5:1, we find a larger and less well-defined interchain distance compared to the 16:1 concentration. This signifies that at the 16:1 salt concentration, there is enough free volume in the polymer host to accommodate the ions, and that the solvation of salt induces ordering of the polymer matrix. At the higher salt concentration 5:1, the polymer network must expand and become less ordered to host the ions. We also note, in accordance with previous studies, that the solvation of salt changes the conformation of the polymer chain towards more gauche states. The simulations furthermore reveal marked correlations between the polymer chains and the anions, which we suggest arise predominantly from an interaction mediated via cations, which can simultaneously coordinate both ether oxygens in the polymer chains and anions. Interanionic distances at 5 A, which are consistent with two or more anions being coordinated around the same cation, are also observed. On a larger scale, the RMC structure of PPO-LiClO4 16:1 clearly indicates the presence of salt-rich and salt-depleted domains having a length scale of

The frequency dependent conductivity σ(f) of single alkali glasses LiPO3 and RbPO3 has been compared with the conductivity of the mixed alkali glass Li0.5Rb0.5PO3. The low frequency plateau value σ0 of the conductivity drops by several orders of magnitude in the mixed alkali glass. In this work the effect of mixed alkali on the shape of the conductivity σ(f) in the dispersive region has been analyzed. The slope of the conductivity curve, in the log σ vs. log f plot, increases more rapidly in the single alkali glasses than in the mixed composition. The onset of the dispersive region is more gradual in the mixed alkali glass. The results indicate that the mixed alkali glass behaves as a dilute single alkali glass.

The hydration process of Ca 3 SiO 5 (C 3 S) cement is investigated, and accelerating effects of adding colloidal silica (CS) are established. CS accelerates dissolution of the C 3 S phase, and renders a more rapid formation of the calcium–silicate–hydrate (C–S–H) binding phase. The role of water is demonstrated. Water evaporation correlates with the appearance of a sharp IR band at 3650 cm −1 signifying non-hydrogen bonded OH groups. Incomplete hydration results in Ca(OH) 2 precipitation. Suppression of water evaporation causes the 3650 cm −1 band to vanish. The Ca 2+ ions become incorporated in the C–S–H network, the formation of which is accelerated by CS.
