2. Colonization and inflammation deficiencies in Mongolian gerbils infected by Helicobacter pylori chemotaxis mutants

Helicobacter pylori causes disease in the human stomach and in mouse and gerbil stomach models. Previous results have shown that motility is critical for H. pylori to colonize mice, gerbils, and other animal models. The role of chemotaxis, however, in colonization and disease is less well understood. Two genes in the H. pylori chemotaxis pathway, cheY and tlpB, which encode the chemotaxis response regulator and a methyl-accepting chemoreceptor, respectively, were disrupted. The cheY mutation was complemented with a wild-type copy of cheY inserted into the chromosomal rdxA gene. The cheY mutant lost chemotaxis but retained motility, while all other strains were motile and chemotactic in vitro. These strains were inoculated into gerbils either alone or in combination with the wild-type strain, and colonization and inflammation were assessed. While the cheY mutant completely failed to colonize gerbil stomachs, the tlpB mutant colonized at levels similar to those of the wild type. With the tlpB mutant, there was a substantial decrease in inflammation in the gerbil stomach compared to that with the wild type. Furthermore, there were differences in the numbers of each immune cell in the tlpB-mutant-infected stomach: the ratio of lymphocytes to neutrophils was about 8 to 1 in the wild type but only about 1 to 1 in the mutant. These results suggest that the TlpB chemoreceptor plays an important role in the inflammatory response while the CheY chemotaxis regulator plays a critical role in initial colonization. Chemotaxis mutants may provide new insights into the steps involved in H. pylori pathogenesis.

6. An investigation of the properties of ions, radicals and molecules by translational-energy-loss spectroscopy

This thesis employs double charge transfer spectroscopy (DCTS) to measure ionisation energies (primarily double ionisation energies) of a number of small molecules. DCTS is described in detail and the previous work in this field is reviewed. A discussion on the stability of molecular dications and the experimental techniques that have been used previously to measure the energies of dications is included. Many characteristics of the target molecule and the projectile ion affect the quality and the type of data obtained using DCTS. This subject is discussed and a number of ions are selected and examined for their potential as prospective projectile ions. The data available for measurement is affected by the cross-section for double electron capture reactions. An extension of the Landau-Zener theory is presented enabling the calculation of absolute cross-sections in double electron capture reactions. Calculated cross-sections are compared with measured cross-sections taken from the literature and with relative cross-sections measured in the present study. A 'reaction window' effect is observed in double electron capture reactions and its effect on double charge transfer spectra is discussed. Using a number of selected projectile ions, double ionisation energies of CO, CO2, COS, CS2 NH3, CH3Br, CCl4, CCl3F, CCl2F2 and C2H4 are measured and assigned, wherever possible, using relevant information from the literature. In addition, the electron-ion recombination energy of both SF5+ and SF3+ are determined as well as the electron affinity of each of the molecules SF4, SF2 and SF.

7. Children’s Economic and Social Rights

- Abstract
Is there such a concept? Do they really exist? To speak of children’s economic and social rights implies an affirmative answer to these questions. But the drafting history of the Convention on the Rights of the Child reveals the fragile nature of the distinction between economic and social rights and civil and political rights for children. Indeed, if not for a late intervention from UNICEF, this distinction may well have been erased from the lexicon of children’s rights. It did however survive but only just. This chapter explores the vulnerable and often clouded relationship between children and economic and social rights. It labours to offer some doctrinal clarity with respect to the nature of this relationship which has been somewhat overlooked within the scholarship on economic and social rights - just what economic and social rights do children actually enjoy? - and what are the attendant obligations on States – are they any different to the obligations owed with respect to generic economic and social rights? But it does not shy away from the constructed and artificial nature of the distinction between the two sets of rights – an artificiality that has only become more acute as the meaning of all rights has expanded and muddied traditional points of distinction. Nor does it shy away from the serious challenges facing the effective implementation of all children’s rights – whether they are classified as economic and social rights or civil and political rights. Particular attention is also drawn to the need to bridge what is termed the disciplinary divide and shift the conception of children from a welfare based approach, which highlights their deficits and vulnerability to a rights-based approach, which recognises their strengths and evolving capacities.

The first evidence for X(3872) production in relativistic heavy ion collisions is reported. The X(3872) production is studied in lead-lead (Pb-Pb) collisions at a center-of-mass energy of root s(NN) = 5.02 TeV per nucleon pair, using the decay chain X(3872) -> J/psi pi(+)pi(-) -> mu(+) mu(-) pi(+)pi(-). The data were recorded with the CMS detector in 2018 and correspond to an integrated luminosity of 1.7 nb(-1). The measurement is performed in the rapidity and transverse momentum ranges vertical bar y vertical bar < 1.6 and 15 < p(T) < 50 GeV/c. The significance of the inclusive X(3872) signal is 4.2 standard deviations. The prompt X(3872) to psi 2S yield ratio is found to be rho(Pb-Pb) = 1.08 +/- 0.49(stat) +/- 0.52(syst), to be compared with typical values of 0.1 for pp collisions. This result provides a unique experimental input to theoretical models of the X(3872) production mechanism, and of the nature of this exotic state.

- Abstract
The understanding and legitimacy of the right to health for persons with a disability remains contested and confused. This chapter seeks to address three issues. First, the extent to which a case can be made to justify the idea of the right to health; second, the meaning of this right and the nature of the obligations imposed on states; and third, the consequences and challenges associated with the use of a human rights-based approach to address the health needs of persons with a disability. It is argued that the right to health has been burdened by a false and misplaced set of assumptions about its meaning and content. Once these concerns are addressed it is possible to offer an understanding of the right to health that is not only pragmatic but also offers a model that recognizes the capacity, agency and dignity of persons with a disability.

The necessity to reprocess spent nuclear fuel has arisen from increasing volumes of highly active and long-lived radionuclides associated with nuclear fission alongside increasing environmental impact concerns. In particular, one major challenge is the separation of the long-lived trivalent minor actinides (here, AmIII and CmIII) from the trivalent lanthanides in order to allow the future transmutation of the actinides into shorter lived radionuclides for the purpose of reducing the long-term radiotoxicity of the waste and the volume build up in storage. There are currently a number of different processes under development world-wide, all of which rely on the coordination chemistry and selective extraction of the minor actinides from the trivalent lanthanides using solvent partitioning. Of these, The TALSPEAK process (Trivalent Actinide Lanthanide Separation by Phosphorus reagent Extraction from Aqueous Komplexations) has showed great promise in separating the trivalent lanthanides from the minor actinides. The process uses DTPA (diethylenetriamine pentaacetic acid) as the chelating holdback reagent, HDEHP (bis-2-ethylhexyl phosphoric acid) as the extracting ligand and lactic acid as the buffer to prevent precipitation of DTPA that occurs at below pH 3.6. However, despite the considerable separation factors obtainable, and the fact the process exhibits promising radiolysis resistance, the process operates at sub optimal pH values. To overcome these limitations, we have developed a family of DTPA-amino acid conjugates that operate under TALSPEAK-like conditions over a much lower pH range of 1.5-2.5. Additionally, these ligands show comparable selectivity to the TALSPEAK process and are also relatively resistant to radiolysis. We will discuss the coordination chemistry of these ligands with the lanthanides and minor actinides using a combination of luminescence and NMR spectroscopies.

The energy levels of triplet states of doubly charge oxygen, obtained using double charge transfer spectroscopy, have been examined in the light of ab initio calculations. Four peaks were found in the double charge transfer spectrum with energies of 41.1±0.2, 43.2±0.2, 48.2±0.3, and 51.3±0.3 eV, respectively. The first and third peaks have been assigned to the 3Σ+u and the 3Πu excited states, respectively. The second peak has been assigned to the population of the 3Πg states. The configuration of the state at 51.3 eV is unknown. The effect of the symmetry rule Σ+\Σ- is discussed and the validity of the spin conservation rule is demonstrated when a proton is used as a projectile in this experimental technique. The rise of globalization and the persistence of global poverty are straining the territorial paradigm of human rights. This book asks if states possess extraterritorial obligations under existing international human rights law to respect and ensure economic, social and cultural rights and how far those duties extend. Taking a departure point in theory and practice, the book is the first of its kind to analyze the principal cross-cutting legal issues at stake: the legal status of obligations, jurisdiction, causation, division of responsibility, and remedies and accountability. The book focuses specifically on the role of states but also addresses their duties to regulate powerful nonstate actors. The authors demonstrate that many key issues have been resolved or clarified in international law while others remain controversial or await the development of further practice, particularly the scope of jurisdiction and the quantitative dimension of extraterritorial obligations to fulfil.

The antiviral activity of methylene blue (MB; a WHO essential medicine) against AHC viruses was investigated using human corneal epithelial cells (HCEC).Methods: Time and concentration-dependent MB accumulation by HCEC was determined colorimetrically and MB inhibition of virus production of 5 E70 and 3 CA24v AHC epidemic isolates in HCEC was determined by micro-plaque assay. AHC virus cytopathy inhibition by MB was detected by reductions in virus-induced caspase-3 activity and polymeric DNA fragments.Results: MB uptake by HCEC was rapid and concentration dependent. MB inhibition of E70 and CA24v production was concentration dependent. AHC virus yields were significantly lower (50 to > 10,000 fold) in HCEC pre-treated with 0.25– 1% MB than in placebo controls (p’s ≤ 0.01). MB pre-treatment significantly inhibited virus-induced caspase-3 activation and DNA fragmentation (p’s< 0.01). Virus-infected cells accumulate oxidized MB and MB application up to 6 h after infection inhibited virus production and virus-induced HCEC cytopathy.Conclusion: The results suggest MB treatment prior to and shortly after infection can inhibit AHC virus production and caspase-mediated HCEC cytopathy.
