INTRODUCTION: Personalized and tumor-informed circulating tumor DNA (ctDNA) testing is feasible and allows for molecular residual disease (MRD) identification in patients with pancreatic ductal adenocarcinoma (PDAC). METHODS: In this retrospective analysis of commercial cases from multiple US institutions, personalized, tumor-informed, whole-exome sequenced, and germline-controlled ctDNA levels were quantified and analyzed in patients with PDAC. Plasma samples (n = 1329) from 298 clinically validated patients were collected at diagnosis, perioperatively (MRD-window; within 2-12 weeks after surgery, before therapy), and during surveillance (>12 weeks post-surgery if no ACT or starting 4 weeks post-ACT) from November 2019 to March 2023. RESULTS: Of the initially diagnosed patients with stages I-III PDAC who went for resection, the median follow-up time from surgery was 13 months (range 0.1-214). Positive ctDNA detection rates were 29% (29/100) and 29.6% (45/152) during the MRD and surveillance windows, respectively. The aim of this study was to determine changes of reactive oxygen species (ROS), serum antioxidant capacity (SAC), oxidative stress index (OSi), and α-tocopherol (α-T) during the periparturient period in healthy and mastitic cows and to further investigate whether these parameters can be used as a tool for identifying cows at higher risk of developing mastitis.

The social behaviour of twenty-one lambs of three sheep breeds (Chios, Karagouniki, and Synthetic) was observed from the age of 2.5–7 months, divided into three equal periods (A, B, C) of 1.5 months each. The frequency of the performed agonistic behaviours was higher (p < 0.01) before four months of age (period A: 11 × 10−4 ± 2 × 10−4) compared to the other periods (B: 5 × 10−4 ± 1 × 10−4; C: 8 × 10−4 ± 1 × 10−4). The same was noticed for affiliative behaviours performed (A: 21 × 10−4 ± 3 × 10−4; B: 8 × 10−4 ± 1 × 10−4; C: 5 × 10−4 ± 1 × 10−4), and for agonistic and affiliative behaviours received (p < 0.001). The lambs had higher levels of wool cortisol (23.398 ± 5.344 pg/mg; p < 0.01) at period C compared to A (7.899 ± 1.19 pg/mg). Breed affected the affiliative behaviours. Karagouniki showed higher frequencies than Synthetic (16 × 10−4 ± 3 × 10−4 vs. 6 × 10−4 ± 1 × 10−4). Blood cortisol was higher (p < 0.05) in the Synthetic (4.789 ± 0.928 ng/mL) compared to the Chios (1.805 ± 0.417 ng/mL) breed. Overall, lambs' behaviour displayed changes upon time, with four months of age being a step to a next developmental stage with fewer social interactions and higher levels of cortisol. The study assesses the direct effects of climate change by 2060, including extreme events, on the productivity of regional crop farming and livestock in Greece, and the broader socio-economic effects on the agri-food and other sectors. Different approaches (i.e., agronomic models, statistical regression models, and equations linking thermal stress to livestock output) were combined to estimate the effects on productivity from changes in the average values of climatic parameters, and subsequently the direct economic effects from this long-term climate change. Recorded damages from extreme events together with climatic thresholds per event and crop were combined to estimate the direct economic effects of these extremes. The broader socio-economic effects were then estimated through input–output analysis. Under average levels of future extreme events, the total direct economic losses for Greek agriculture due to climate change will be significant, from EUR 437 million/year to EUR 1 billion/year. These losses approximately double when indirect effects on other sectors using agricultural products as inputs (e.g., food and beverage, hotels, and restaurants) are considered, and escalate further under a tenfold impact of extreme events. Pervasive inbreeding is a major genetic threat of population fragmentation and can undermine the efficacy of population connectivity measures. Nevertheless, few studies have evaluated whether wildlife crossings can alleviate the frequency and length of genomic autozygous segments. Here, we provided a genomic inbreeding perspective on the potential effectiveness of mammal population defragmentation measures. We applied a SNP-genotyping case study on the ~2500 wild boar Sus scrofa population of Veluwe, The Netherlands, a 1000-km2 Natura 2000 protected area with many fences and roads but also, increasingly, fence openings and wildlife crossings. We combined a 20K genotyping assessment of genetic status and migration rate with a simulation that examined the potential for alleviation of isolation and inbreeding. We found that Veluwe wild boar subpopulations are significantly differentiated (FST-values of 0.02-0.13) and have low levels of gene flow. One noteworthy exception was the Central and Southeastern subpopulation, which were nearly panmictic and appeared to be effectively connected through a highway wildlife overpass. Estimated effective population sizes were at least 85 for the meta-population and ranged from 31 to 52 for the subpopulations. All subpopulations, including the two connected subpopulations, experienced substantial inbreeding, as evidenced through the occurrence of many long homozygous segments. Simulation output indicated that whereas one or few migrants per generation could undo genetic differentiation and boost effective population sizes rapidly, genomic inbreeding was only marginally reduced. The implication is that ostensibly successful connectivity restoration projects may fail to alleviate genomic inbreeding of fragmented mammal populations. We put forward that defragmentation projects should allow for (i) monitoring of levels of differentiation, migration and genomic inbreeding, (ii) anticipation of the inbreeding status of the meta-population, and, if inbreeding levels are high and/or haplotypes have become fixed, (iii) consideration of enhancing migration and gene flow among meta-populations, possibly through translocation. In Greece both the black indigenous pig and the wild boar are considered as species of valuable genetic diversity while their products achieve a valuable market price. However, many crop damages are recorded, with farmers to claim that wild boar hybrids are responsible. On the other hand, black pig classification is based on phenotypic characteristics, which does not ensure breed's homogeneity in case of hybridization. Using the PCR-RFLP methodology, pig samples (n=135) from different rearing situations (feral boars, semi-extensive black pigs and extensive wild boars) were examined in order to identify whether or not hybridization exists. In the examined feral population of wild boar a 26% of hybrids was noted, while in the case of the extensive farming population of wild pigs a hybridization of 11.76% was observed. Interestingly, in both cases of the examined black pigs' populations, a mentionable hybridization with wild boar was observed, reflecting probably an implemented breeding practice or uncontrolled mating with wild boars. A pivotal level (5-7%) of inbreeding rate was also noted in the examined populations. The immediate removal of hybrids from all the examined populations should be achieved, in order to prevent and eliminate further introgression, genetic depression and loss of genetic diversity for both populations of wild boar and black pig. Finally, the applied methodology may be used by state authorities or certifying organizations to test, control or inspect farms rearing wild boar or black pig populations in order to record and eliminate hybridization events between them.

Highly endangered local breeds are considered important not only for the maintenance of their genetic diversity for future survival but also because they regarded as part of the cultural heritage of the local and national communities. Using pedigree data and an analysis of 18 microsatellite loci we investigated the genetic diversity of a private (commercial) indigenous Skyros horse population, reared in an insular region of North-Western of Greece. The overall average animal inbreeding value reached 24%. Concerning average inbreeding value over non founding animals, it was estimated to 0.013, while the corresponding value over inbred animals were 0.13.The mean number of alleles per locus amounted to 3.72, ranging between 1 and 7 alleles. The poultry sector is considered to be one of the most industrialized sectors of livestock production. Although the livestock sector contributes the 14.5% of total anthropogenic greenhouse gas (GHG) emissions, less attention has been paid in the respective emissions of the poultry sector compared to other farmed animals such as ruminants. The aim of the study was to estimate the carbon footprint of the poultry sector (layers, broilers, and backyards) in the Greek territory during the last 60 years as a means of exploring further mitigation strategies. Tier 2 methodology was used to estimate GHG emissions. Different mitigation scenarios related to changes in herd population, feeds, and manure management were examined. GHG emissions showed an increased trend over time. The different scenarios explored showed moderate to high mitigating potential depending on the parameters that were changed. Changes in manure management or diet revealed to have a higher potential to eliminate GHG emissions. Changes in population numbers showed a low mitigating potential. However, if mortality could be improved within industrialized farming systems, then it could be an indirect increase in product quantities with a slight increase in emissions. Traceability methods in livestock sector through the tracking of animal species, breed or even individuals, has become of utmost importance as a "vehicle" for ensuring consumers' food safety. The advent of new technology at DNA level has facilitated the convenience and the accuracy of the implementation of traceability methods. The scope of this review is to highlight the most up to date progress on DNA based approaches concerning the traceability procedures for small ruminant species and/or their products, giving emphasis on short tandem repeats (microsatellites) and single nucleotide polymorphisms. The conclusions of this review may be used either from the farmer or the State and other Organisations in order not only to certify traceability throughout the whole food process chain but to ensure also consumers' food safety.
