The spermatogenesis of fish remained unclear about the cytological process, especially on the meiotic process of spermatocytes after meiosis I. In the present study, microstructure and ultrastructure of spermatogenesis in Cyprinus carpio (abbreviated CC) were studied, the meiosis process was traced by chromosomal observation and the corresponding immunohistochemical staining of testis where spermatocytes at different stages were distinguished. Results firstly described the telomere-led bouquet configuration of prophase I-stage cells in fish, which ensure the normal chromosomal behavior just like in other eukaryotes. Compared to the canonical meiotic process, the spermatocytes at leptotene, zygotene, pachytene, diplotene and diakinesis stages in CC were normal and also proceeded metaphase I interacting with the meiotic spindle as a unit, while the subsequently meiosis II in CC demonstrated contradicted findings in most organisms, in which no obvious spindle-dependent sister chromatids aligning and segregating could be observed, but some circular condensed nucleus structures. Furthermore, protein antibody of RAD51, DMC1, RNA-Pol II CTD P–S5, H3K36me3 were used respectively as probes, marked initially between the primary spermatocytes at prophase I and metaphase I stage, and between the spermatids and spermatozoa. The systematical study in cytological characteristics of the whole meiotic process in fish spermatogenesis in this paper, provide a standard reference for fish meiosis, which can be of great significance in the practice of fish genetics and breeding.

Polyploidy occurs naturally in fish; however, the appearance of these species is an occasional and gradual process, which makes it difficult to trace the changes in phenotypes, genotypes, and regulation of gene expression. The allotetraploid hybrids (4nAT) of red crucian carp (RCC; ♀) × common carp (CC; ♂) generated from interspecies crossing are a good model to investigate the initial changes after allopolyploidization. In the present study, we focused on the changes in the active sites of the testicular transcriptome of the allotetraploid by localization of RNA Pol II CTD YSPTSPS (phospho S5) using immunofluorescence and RNA-seq data via bioinformatic analysis. The results showed that there was no significant difference in signal counts of the RNA Pol II CTD (S5) between the different types of fish at the same stages, including RCC, CC, 2nF1, and 4nAT, which means that the number of transcriptionally active sites on germ cell chromosomes was not affected by the increase in chromosome number. Similarly, RNA-seq analysis indicated that in the levels of chromosomes and 10-kb regions in the genome, there were no significant changes in the highly active sites in RCC, 2nF1, and 4nAT. These findings suggest that at the beginning of tetraploid origin, the active transcriptome site of 4nAT in the testis was conserved in the regions of the genome compared to that in RCC and 2nF1. In conclusion, 4nAT shared a similar gene expression model in the regions of the genome with RCC and 2nF1 with significantly different expression levels.

Due to the large errors in simulating lake ice phenology, related key parameters and parameterizations were improved in the coupled model based on observations and physics-based schemes. By improving the key parameters associated with shortwave radiation transfer processes when lake ice exists, both models generally simulated the lake break-up date well. Compared with WRF–CLake without improvements, the coupled model with both revised lake models significantly improved the simulation of lake ice phenology. However, there were still considerable errors in simulating the spatial patterns of freeze-up and break-up dates, implying that significant challenges in simulating the lake ice phenology still exist in representing some important model physics, including lake physics such as grid-scale water circulation and atmospheric processes such as snowfall and surface snow dynamics. Therefore, this work can provide valuable new implications for advancing lake ice phenology simulations in coupled models, and the improved model also has practical application prospects in weather and climate forecasts. A comprehensive assessment of the regional water-saving potential (WSP) for crop production is the foundation for setting targets, formulating strategies, and implementing management measures for agriculture. Water footprint (WF) for crop production is a well-known indicator of blue and green water consumption and an effective tool for evaluating blue and green WSP in crop production. The WSP evaluations based on crop WF primarily rely on crop redistribution (CR) and the crop WF benchmark (WFB). The CR can be completed relatively quickly, but the WSP is limited. In contrast, reaching an area’s maximum WSP is possible with WFB, but it requires long-term and multi-aspect inputs. However, existing evaluations of regional crop WSP are often based on a single methodology, and the results are various, which confuse the policymakers at times. This study estimated both blue and green WSP of wheat and maize based on two approaches—crop redistribution (CR) and crop water footprint benchmark (WFB)—at the county level in the Yellow River basin (YRB) in different hydrological years. The WSP was 7–11% based on CR, whereas 17–24% based on the crop WFB. The green WSPs were larger than blue WSPs. Over half of counties had larger WSP based on the WFB. These values vary with the crop, region, and hydrological year. The analysis emphasises the importance of considering both blue and green water, and quantitatively comparing different water-saving methods when evaluating regional crop WSP.

Blue water (irrigation water) and green water (soil moisture), as natural bases in crop production, are vulnerable to natural and socioeconomic disturbances. However, quantitative resilience assessments of blue and green water for crop production are still lacking. Here we constructed a social-ecological based general water resilience assessment framework that distinguishes blue and green water for crop production in four dimensions, including resistance, absorptive capacity, adaptive capacity and transformative capacity. The framework was applied to quantify the spatiotemporal characteristics of water resource resilience for three staple crops (rice, wheat, and maize) at provincial level in mainland China over 2000–2017. Results indicate that water resilience related to crop production of China as a whole was at the lower-middle level over the study period. The green water resilience had higher initial performance, while the blue water resilience had a faster growth rate (>50%). The performance of adaptive and transformative capacity was 8.9∼153.7% better than resistance and absorptive capacity. There were significant heterogeneities in the water resilience score among crops and in time and space. Wheat had the highest resilience of green water, while its blue water resilience was the lowest among the three crops. The water resilience for crop production varied by up to 2.4-fold across provinces. Synergies and trade-offs among the four dimensions of water resilience were further evaluated. The current study provides an effective approach for assessing the state and progress of resilience of regional water resources for crop production.

The gray water footprint (GWF) can quantitatively evaluate the effect of non-point pollution on water quality in the context of water quantity. It is crucial to explore the driving forces behind the GWF to solve water quality problems. This study quantified the unit GWFs of grain crops and oil crops at the municipal scale in six provinces of western China over 2001–2018, then jointly applied the extended STIRPAT model and path analysis methods to analyze the climatic and socioeconomic driving forces of the GWF. Results show that the key driving forces affecting the GWF obtained by the two methods were consistent. Planting structure and population were the main factors increasing the total GWF, while crop yield was the largest factor inhibiting the unit GWF and demonstrates regional differences. However, when the indirect influence of the driving factor through other factors was large, some driving forces obtained by different methods were reversed. For example, the indirect impact of per capita cultivated land area on the total GWF in Inner Mongolia was large, resulting in a significant positive impact in path analysis and a slight negative impact in the STIRPAT model. To draw more comprehensive and referential conclusions, we suggest using multiple methods together to verify the driving forces and account for the regional differences.

ObjectiveTo explore the effect of new antiseizure medication on bone metabolism and bone mineral density in children.MethodsThe Chinese and English databases (PubMed, EMBASE, Cochrane Library, CNKI, Wanfang and VIP) were systematically searched for observational studies evaluating the effects of new antiseizure medication on bone metabolism and bone mineral density in children with epilepsy. The effects of new antiseizure medication on serum calcium, phosphorus, alkaline phosphatase, bone alkaline phosphatase, parathyroid hormone, 25-hydroxyvitamin D and bone mineral density in children were systematically evaluated.ResultsAfter systematic retrieval and screening, 12 studies with high literature quality (including 629 epileptic children and 627 control subjects) were included in the systematic evaluation. Meta-analysis showed that new antiseizure medication decreased bone mineral density (MD: −0.05, 95%CI, −0.09, −0.02; P = 0.004). From different kinds of antiseizure medication, levetiracetam can reduce blood phosphorus concentration in children (MD: −0.04; 95%CI, −0.07, −0.01). A 58-year-old male vegetarian presented with progressive numbness and weakness in the lower extremities. Laboratory examinations showed reduced vitamin B12 level with megaloblastic anaemia. Spinal magnetic resonance imaging (MRI) revealed hyperintensity within the posterior and lateral columns on T2-weighted imaging. The diagnosis of subacute combined degeneration (SCD) of the spinal cord was established. Unexpectedly, the patient developed transitory syncope on the second day after hospitalization. The diagnostic computed tomography pulmonary angiography (CTPA) confirmed multiple small pulmonary emboli. An isolated significantly elevated level of homocysteine (117.1 µmol/l) was documented when screening for hypercoagulable markers. Except for a long-term vegetarian diet, no other risk factors for hyperhomocysteinaemia (such as a family history of homocysteinuria) was found. The severity of the hyperhomocysteinaemia found in this current patient was unusual for patients with an insufficient intake of vitamin B12. In SCD patients, elevated homocysteine may increase the risk of thrombosis, which may exacerbate existing problems. Knowing the risk factors should help physicians choose appropriate diagnostic and therapeutic strategies.

The limited available water resources and competition among different water use sectors have become the main constraints of food security and sustainability. Faced with the inability to expand the area of cultivated land due to urbanization and population growth, one of the biggest challenges and risks for developing countries is to ensure the supply of food quantity and quality under extremely limited water resources. To achieve water-saving and improve calorie supply by adjusting crop production allocations, three objectives—of minimum blue water footprint, maximum calorie production, and each crop production no less than the reference level of nine main crops in China—were achieved using a non-dominated sorting genetic algorithm II. The results display that compared with the reference year, model Maize+ (maize production increased) had significant blue water saving (~32%), the blue water footprint of crop production in all provinces reduced, and its calorie production increased by 4%. This solution is not realistic for China because wheat and rice production need to be reduced by 82 and 80%, respectively. However, model Citrus– (citrus production decreased) reduced the blue water footprint of crop production (~16%), and increased calorie production (~12%). Compared with other solutions, it is a sustainable crop production structure that is easier to realize because it is better at meeting the production of each crop. Therefore, China can appropriately increase the planting area of maize and reduce the planting of citrus and other crops that consume more blue water and produce fewer calories to ensure the security and sustainability of food supplies. However, the improvement of water saving-technology, rationalization of agricultural water resources management, crop production allocations mentioned in this study, and other efforts are necessary to achieve this goal.

The water footprint (WF) of crop production has been widely accepted as a comprehensive indicator of agricultural water consumption. Rationality and accuracy in crop WF accounting are thus prerequisites for implementation of WF assessments that yield sustainable regional agricultural water management. However, few studies have focused on the rationality of multiple quantitative approaches and the associated differences in crop WF accounting among different studies. Here we, focusing on maize, wheat and rice, review quantitatively the effects of different quantification approaches and scales on the results of crop WF accounting and simulations in relevant published research during 2002–2018 worldwide. Results show that (i) The number of studies on crop WF accounting has increased by 17 times since 2002 (~the year of creation of WF concepts); the research direction is focused on improvement of quantification and resolution in both time and space. (ii) The current approaches to WF calculation can be divided into five main types: the field crop water requirement (FCWR) approach, field soil water balance (FSWB) approach, regional water balance (RWB) approach, remote sensing (RS) approach and field measured water balance (FMWB) approach. The FCWR and FSWB approaches are more widely adopted than the other three. (iii) There were non-negligible differences in the WF accounting results among approaches and scales. At the global level, the deviations in WF for maize, wheat, and rice were relatively low among different studies, with the world average values of 0.73 m3 kg−1 ± 14.9%, 1.136 m3 kg−1 ± 13.5%, and 1.269 m3 kg−1 ± 27%, respectively. The ranges of uncertainty varied significantly when downscaling to specific countries and provinces. The maximum coefficients of variation (CV) of WF for maize, wheat, and rice in different regions were up to 40%, 49%, and 50%, respectively. (iv) The WF simulations showed very reasonable agreement and lower deviation between the FCWR and FSWB approaches.

Fast-growing food losses and waste (FLW) have caused major environmental issues, yet little is known about the role of trade in the inefficient resource consumption related to FLW. In this study, we estimated the inter-annual water footprints (WFs) and land footprints (LFs) related to FLW in consideration of associated inter-provincial virtual water (VW) and virtual land (VL) flows within China from 2000 to 2017. Additionally, differences between eating at home and out in rural and urban regions were distinguished. The FLW-related WF and LF showed 1.5- and 1.2-fold increasing trends, respectively, over the study period. Eating at home in rural regions caused the most FLW and associated water and land appropriations. Both FLW-related gross inter-provincial VW and VL flows increased by 99% and 22% during the study period, respectively, and inter-provincial VW and VL flows in corresponding FLW-related WF and LF differed significantly. In addition, food losses were found to cause more inefficient resource consumption than food waste. This analysis shows that the substantial role of FLW in shaping the intra-national food-related VW and VL networks, and that the policies for FLW diminution at the food supply chain stage are as important as those at the consumption stage.

This paper explores the effect of varying agricultural management practices on different water efficiency indicators: irrigation efficiency (IE), crop water use efficiency (WUE), and green and blue water footprint (WF). We take winter wheat in an experimental field in Northern China as a case study and consider a dry, average and wet year. We conducted 24 modeling experiments with the AquaCrop model, for all possible combinations of four irrigation techniques, two irrigation strategies and three mulching methods. Results show that deficit irrigation most effectively improved blue water use, by increasing IE (by 5%) and reducing blue WF (by 38%), however with an average 9% yield reduction. Organic or synthetic mulching practices improved WUE (by 4% and 10%, respectively) and reduced blue WF (by 8% and 17%, respectively), with the same yield level. Drip and subsurface drip irrigation improved IE and WUE, but drip irrigation had a relatively large blue WF. Improvements in one water efficiency indicator may cause a decline in another. In particular, WUE can be improved by more irrigation at the cost of the blue WF. Furthermore, increasing IE, for instance by installing drip irrigation, does not necessarily reduce the blue WF.

The grey water footprint (GWF) is defined as freshwater requirements for diluting pollutants in receiving water bodies. It is widely used to measure the impact of pollutant loads on water resources. GWF can be transferred from one area to another through trade. Although pollution flow has previously been investigated at the national level, there has been no explicit study on the extent to which crop trade affects GWF across regions and the associated changes in grey water stress (GWS). This study analyzes pollution flow associated with interprovincial crop trade based on nitrogen (N) and phosphorus (P) loss intensity of three major crops, namely, maize, rice and wheat, which is simulated by a grid-based crop model for the period 2008–2012, and evaluates the spatial patterns of GWS across China. The results indicate that the integrated national GWF for N and P was 1271 billion m ^3 yr ^−1 , with maize, rice, and wheat contributing 39%, 37%, and 24%, respectively. Through interprovincial crop trade, southern China outsourced substantial N and P losses to the north, leading to a 30% GWS increase in northern China and 66% GWS mitigation in southern China. Specifically, Jilin, Henan, and Heilongjiang Provinces in the northern China showed increases in GWS by 161%, 114%, and 55%, respectively, while Fujian, Shanghai, and Zhejiang in the south had GWS reductions of 83%, 85%, and 80%, respectively. It was found that the interprovincial crop trade led to reduced national GWF and GWS. Insights into GWF and GWS can form the basis for policy developments on N and P pollution mitigation across regions in China.

Assessing the water footprint (WF) of crops is key to understanding the agricultural water consumption and improving water use efficiency. This study assessed the WF of wheat and maize in the Haihe River Basin (HRB) of Northern China over the period1956–2015, including rain-fed, sufficient, and insufficient irrigation conditions by different irrigation intensity to understand the agricultural water use status. The major findings are as follows: (1) The annual average total WF of wheat and maize production is 20.1 (52% green, 29% blue, and 19% grey) and 15.1 (73% green, 3% blue, and 24% grey) billion m3 year−1, respectively. The proportion of grey WF is much larger than the world average; (2) Wheat has larger unit WF (1580 m3 t−1) than maize (1275 m3 t−1). The unit WF of both wheat and maize shows exponentially decreasing trends, indicating that water use efficiency has been improved. The unit WF is heterogeneous in space, which is larger in Tianjin and Huanghua and smaller in the Southern HRB; (3) Rain-fed crops have the largest unit WF, followed by crops under insufficient and sufficient irrigation conditions for both wheat and maize. To improve the sustainability of water resources, the application of fertilizer must be reduced, and irrigation is an effective way to improve water use efficiency in water-abundant areas.

The discharge of a large amount of flotation reagents wastewater can cause significant environmental pollution. In this study, NiO/La-NaTaO 3 nano-photocatalyst was prepared and applied to degrade synthetic flotation reagent ammonium dibutyl dithiophosphate wastewater. Various characterization results confirmed the successful synthesis of NiO/La-NaTaO 3 , and UV-vis DRS analysis revealed a band gap of 3.96 eV for 4 wt% NiO/2.5% La-NaTaO 3 . Under UV light, the degradation rate of 20 mg 4 wt% NiO/2.5% La-NaTaO 3 photocatalyst reached its optimum within 4.5 h at pH=3, exhibiting a 1.45 times improvement compared to pure NaTaO 3 . Radical trapping experiments and EPR results showed that ·OH and·O 2 - showed major contribution to the degradation. Meiosis is the key process for producing mature gametes. A natural fertile triploid Carassius auratus population (3nDTCC) and an artificially derived sterile triploid crucian carp (3nCC) have been previously observed, providing suitable model organisms for investigating meiosis characteristics in triploid fish. In the present study, the microstructures and ultrastructures of spermatogenesis were studied in these fishes. TdT-mediated dUTP nick end labeling detection was performed to investigate the apoptosis of spermatocytes. Fluorescence in situ hybridization was employed to trace chromatin pairing. In addition, the mRNA expressions of cell cycle-related genes (i.e., cell division control 2 and cell cycle protein B) were determined by quantitative realtime polymerase chain reaction to illustrate the molecular mechanism of abnormal meiosis in the 3nCC. The results showed that the 3nCC undergoes an irregular prophase I, with the chromosomes distributed in a unipolar radial manner and exhibiting partial pairing, hindered metaphase I, and degenerated cells in the subsequent stages. Meanwhile, the 3nDTCC presented a relatively regular meiotic prophase I with complete conjugate chromosome pairs and chromosomes distributed along the karyotheca, which were presented as a ring structure by slicing. Only the spreads with 130-150 irregular chromosomes can be easily detected in the 3nDTCC, suggesting that it may undergo an abnormal metaphase I. Introduction: Cardiotocography (CTG) has its limits in detecting fetal acidosis and intrapartum asphyxia. Our aim was to evaluate a CTG training programme based on fetal physiology in the Mediterranean perinatal network., Methods: Professionals from 41 maternity units of the Mediterranean network were invited to participate in a CTG masterclass based on fetal physiology in March 2019 and October 2019. They were asked to react to three practical cases by a physiological approach before the training course (T0), one month after (T1) and six to seven months after (T2). The mean scores were compared by using a mixed model including lapse of time to evaluation, profession of participants and level of the maternity unit as fixed effects., Results: A total of 248 professionals from 32 maternity units finally participated in the organizational audit. By using a mixed model, we found a significant improvement of the mean score at T1=6.44/10 compared to T0=4.97/10 (p<0.0001), and a significant improvement of the mean score obtained at T2=6.17/10 compared to T0 (p<0.0001). T2 scores were not significantly different from T1 scores (p=0.143)., Discussion: A CTG training programme based on fetal physiology showed a significant improvement in the professionals' interpretation of CTG at short term and stable results at long term. Purpose: The aim of this study was to determine the influence of fluoride-resistant strains of S. mutans on the bonding strength of dental resin composite., Methods: Standardized specimens of resin composite (Z350), and self-etch adhesives (Single bond Universal, 3M) were incubated with S. mutans UA159, fluoride-resistant strains of S. mutans UA159 (UA159-FR) or uninoculated culture medium (Control) or PBS for up to 14 days. Resin-dentin bonded disks were subjected to push-out test. Fractured surface analysis of the specimens was performed by scanning electron microscopy(SEM). SPSS18.0 software package was used for statistical analysis., Results: UA159-FR and UA159 were shown to decrease the bond strengths in levels comparable with those found in the control groups (P<0.05). SEM confirmed the increased degradation of all materials with UA159-FR and UA159, compared with control and PBS., Conclusions: The results suggest that resin-dentin interface can be compromised by oral bacteria that contribute to the progress of secondary caries. Fluoride-resistant strains of S. mutans and its parental strains both decrease bonding strengths in a short time, but no significant difference in the extent of damage.

In a time of increasing economic constraints, it is crucial that health systems optimize their resource use to ensure that they generate the maximum possible health gain. Therefore, it is necessary for health interventions to be evaluated and compared across therapeutic boundaries. Undertaking such an evaluation a generic utility-based measure is required. But it remains uncertain whether the utility values obtained by direct or indirect methods are comparable and which approach is the most appropriate in Systemic Lupus Erythematosus (SLE) population. In the study, we compared the utility values obtained by an indirect method (EQ-5D) with direct utility instruments, the standard gamble (SG) and visual analog scale (VAS), in SLE patients. The correlations between VAS, EQ-5D and LupusQoL were significant; relative good intraclass correlations or kappa coefficients indicated the reliability of these instruments. A model incorporating the SLEDAI scores and LupusQoL domains of emotional health and pain was a good predictor of VAS. SLEDAI score was a good predictor in the SG regression model. These findings suggested that the VAS and EQ-5D might be valid and reliable measures to assess health related quality of life in SLE patients and represent promising outcome measures for future research in this population.

Extracellular signal regulated kinase½ (ERK1/2) signaling is critical to endothelin-1 (ET-1)-induced cardiomyocyte hypertrophy. This study was to investigate ERK1/2 signaling and hypertrophic response to ET-1 stimulation in cardiomyocytes (CMs) from spontaneous hypertensive rats (SHR) and normotensive Wistar-Kyoto rats (WKY). Primary neonatal SHR and WKY CMs were exposed to ET-1 for up to 24 hrs. Minimal basal ERK1/2 phosphorylation was present in WKY CMs, while a significant baseline ERK1/2 phosphorylation was observed in SHR CMs. ET-1 induced a time- and dose-dependent increase in ERK1/2 phosphorylation in both SHR and WKY CMs. However, ET-1-induced ERK1/2 activation occurred much earlier with significantly higher peak phosphorylation level, and stayed elevated for longer duration in SHR CMs than that in WKY CMs. ET-1-induced hypertrophic response was more prominent in SHR CMs than that in WKY CMs as reflected by increased cell surface area, intracellular actin density, and protein synthesis. Pre-treatment with ERK1/2 phosphorylation inhibitor PD98059 completely prevented ET-1-induced ERK1/2 phosphorylation and increases in cell surface area and protein synthesis in SHR and WKY CMs. The specific PI3 kinase inhibitor LY294002 blocked ET-1-induced Akt and ERK1/2 phosphorylation, and protein synthesis in CMs. These data indicated that ERK1/2 signaling was differentially enhanced in CMs, and was associated with increased cardiac hypertrophic response to ET-1 in SHR. ET-1-induced ERK1/2 activation and cardiac hypertrophy appeared to be mediated via PI3 kinase/Akt signaling in SHR and WKY. The differential ERK1/2 activation in SHR CMs by ET-1 might represent a potential target for combination therapy of hypertension.

Green manuring is a common practice in replenishment of soil organic matter and nutrients in rice paddy field. Owing to the complex interplay of multiple factors, the oxidation--reduction (redox) properties of dissolved organic matter (DOM) from green manure crops are presently not fully understood. In this study, a variety of surrogate parameters were used to evaluate the redox capacity and redox state of DOM derived from Chinese milk vetch (CMV, Astragalus sinicus L.) via microbial decomposition under continuously flooded (CF) and non-flooded (NF) conditions. Additionally, the correlation between the surrogate parameters of CMV-DOM and the kinetic parameters of relevant redox reactions was evaluated in a soil-water system containing CMV-DOM. Results showed that the redox properties of CMV-DOM were substantially different between the fresh and decomposed CMV-DOM treatments. Determination of the surrogate parameters via ultraviolet-visible/Fourier transform infrared absorption spectroscopy and gel permeation chromatography generally provided high-quality data for predicting the redox capacity of CMV-DOM, while the surrogate parameters determined by elemental analysis were suitable for predicting the redox state of CMV-DOM. Depending on the redox capacity and redox state of various moieties/components, NF-decomposed CMV-DOM could easily accelerate soil reduction by shuttling electrons to iron oxides, because it contained more reversible redox-active functional groups (e.g. quinone and hydroquinone pairs) than CF-decomposed CMV-DOM. This work demonstrates that a single index cannot interpret complex changes in multiple factors that jointly determine the redox reactivity of CMV-DOM. Thus, a multi-parametric study is needed for providing comprehensive information on the redox properties of green manure DOM.

Objective: To investigate the construct and criterion validity of the Euro Qol-5D (EQ-5D), which allows quality-adjusted life-years to be calculated, in patients with systemic lupus erythematosus (SLE)., Methods: Consecutive SLE patients who had been followed at the Renji Hospital, School of Medicine, Shanghai Jiao Tong University were recruited. Cross-sectional correlations of the EQ-5D with equivalent domains in disease-specific health-related quality of life (HRQoL), LupusQol, Systemic Lupus Erythematosus Disease Activity Index (SLEDAI) measures, the Systemic Lupus International Collaborating Clinics Damage Index (SDI), and patient characteristics were tested. Discriminant validity to assess the ability to distinguish between patients of different disease severity was assessed. There also were evaluations of ceiling and floor effects., Results: 240 patients were recruited in total. The EQ-5D correlated moderately to strongly with all domains of the LupusQoL (r: 0.44-0.7) apart from intimate relationships (r = 0.25) and body image (r = 0.18). There was moderate negative correlation between EQ-5D and clinical assessment of disease, SLEDAI (r = -0.589) and SDI (r = -0.509). When compared with equivalent domains on LupusQoL, there was good construct validity in EQ-5D (r: 0.631-0.812). EQ-5D could also discriminate patients with varied disease severity (according SLEDAI and SDI). There was no floor effect in EQ-5D but the ceiling effect remains strong (34%)., Conclusion: Our results provide sufficient evidence that the EQ-5D displays construct and criterion validity for use in SLE patients. Disease-specific measures of HRQoL used alongside may be a better choice.

Objective: To evaluate the psychometric properties and clinical utility of Chinese Multidimensional Health Assessment Questionnaire (MDHAQ-C) in patients with rheumatoid arthritis (RA) in China., Methods: 162 RA patients were recruited in the evaluation process. The reliability of the questionnaire was tested by internal consistency and item analysis. Convergent validity was assessed by correlations of MDHAQ-C with Health Assessment Questionnaire (HAQ), the 36-item Short-Form Health Survey (SF-36) and the Hospital anxiety and depression scales (HAD). Discriminant validity was tested in groups of patients with varied disease activities and functional classes. To evaluate the clinical values, correlations were calculated between MDHAQ-C and indices of clinical relevance and disease activity. Agreement with the Disease Activity Score (DAS28) and Clinical Disease Activity Index (CDAI) was estimated., Results: The Cronbach's alpha was 0.944 in the Function scale (FN) and 0.768 in the scale of psychological status (PS). The item analysis indicated all the items of FN and PS are correlated at an acceptable level. MDHAQ-C correlated with the questionnaires significantly in most scales and scores of scales differed significantly in groups of different disease activity and functional status. MDHAQ-C has moderate to high correlation with most clinical indices and high correlation with a spearman coefficient of 0.701 for DAS 28 and 0.843 for CDAI. The overall agreement of categories was satisfying., Conclusion: MDHAQ-C is a reliable, valid instrument for functional measurement and a feasible, informative quantitative index for busy clinical settings in Chinese RA patients.

Introduction: Glucocorticoid (GC) therapy remains important in improving the prognosis of patients with systemic lupus erythematosus (SLE). However, some patients do not achieve an effective response with GC treatment, creating an obstacle to the remission of SLE. Identification of the underlying mechanisms responsible for steroid resistance can be significant. Macrophage migration inhibitory factor (MIF) arouses our interest because of its reciprocal relationship with GCs. In the present study, we investigated for the first time whether MIF correlated with steroid resistance in SLE and explored potential mechanisms of action., Methods: Sixty-two patients with SLE (40 steroid sensitive and 22 steroid resistant) and 21 normal controls were recruited. Serum levels of MIF were measured by ELISA. Cytosolic MIF and IκB expression in peripheral blood mononuclear cells (PBMCs) were determined by western blotting. The electrophoretic mobility shift assay was assessed by NF-κB in nuclear aliquots. Gene silencing was applied to reduce expression of MIF in PBMCs in steroid-resistant patients. PBMCs obtained from steroid-sensitive patients were treated with recombinant human MIF of different concentrations., Results: MIF levels in serum and PBMCs were higher in steroid-resistant patients compared with steroid-sensitive patients and controls. In contrast to the steroid-sensitive group, NF-κB levels were significantly higher and IκB levels lower in steroid-resistant patients. After MIF gene silencing, IκB levels in cells from steroid-resistant patients were increased. In steroid-sensitive patients, a decrease in IκB levels and an increase in NF-κB expression from baseline were detected in PBMCs treated with a higher concentration of recombinant human MIF. Treatment with recombinant human MIF did not regulate expression of IκB and NF-κB in PBMCs from patients treated with an anti-MIF monoclonal antibody., Conclusions: Our results indicated that MIF may play a role in the formation of steroid resistance in SLE by affecting the NF-κB/IκB signaling cascade. As a regulator of glucocorticoid sensitivity, MIF may be a potential target for steroid sparing.

Objectives: It was found that alpha-enolase was dramatically up-regulated in the hypertrophic hearts of SHR in our previous study. The purposes of this study were to examine the expression pattern of alpha-enolase in pre- and postnatal myocardium of spontaneously hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats, and to explore the relationship between the overexpression of alpha-enolase and left ventricular hypertrophy., Methods: HE staining was used for the measurement of cardiac hypertrophy. Immunohistochemical technique was used to evaluate the location of alpha-enolase. The expressions of alpha-enolase in the left cardiac ventricles at different development times were examined by Real-time RT-PCR and Western blot., Results: Cardiac hypertrophy was found in SHR rats at 4 weeks of age and remained up to 24 weeks of age. The signals of alpha-enolase protein were strong and existed extensively in hypertrophic myocardium in SHR, while in the normal myocardium of WKY, the signals were scarcely found and weak. The levels of alpha-enolase mRNA and protein in SHR and WKY hearts during fetal stage and newborn stage were similar, while from 4 weeks of age to 24 weeks of age, accompanied by the cardiac hypertrophy, the levels of alpha-enolase mRNA and protein in left ventricle of SHR were significantly higher than that in WKY., Conclusions: The expressions of alpha-enolase in the left ventricle of the rats during normal and pathological cardiac development were different. This phenomenon provides the potential clues to understanding pathophysiological mechanisms in cardiac hypertrophy of SHR.

Total of 260 soil profiles were reported to investigate the arsenic spatial distribution and vertical variation in Guangdong province. The arsenic concentration followed an approximately lognormal distribution. The arsenic geometric mean concentration of 10.4 mg/kg is higher than that of China. An upper baseline concentration of 23.4 mg/kg was estimated for surface soils. The influence of soil properties on arsenic concentration was not important. Arsenic spatial distributions presented similar patterns that high arsenic concentration mainly located in limestone, and sandshale areas, indicating that soil arsenic distribution was dependent on bedrock properties than anthropogenic inputs. Moreover, from A- to C-horizon arsenic geometric mean concentrations had an increasing tendency of 10.4, 10.7 to 11.3 mg/kg. This vertical variation may be related to the lower soil organic matter and soil degradation and erosion. Consequently, the soil arsenic export into surface and groundwaters would reach 1040 t year-1 in the study area.

A label-free semiquantitative peptide feature profiling method was developed in response to challenges associated with analysis of two-dimensional liquid chromatography-tandem mass spectrometry data. One hundred twenty human sera (49 from invasive breast carcinoma patients, 26 from non-invasive breast carcinoma patients, 35 from benign breast disease patients, and 10 from normal controls) were repeatedly analyzed using a standardized two-dimensional liquid chromatography-mass spectrometry method. Data were extracted using the novel semiquantitative peptide feature profiling method, which is based on comparisons of normalized relative ion intensities. Hierarchical cluster analyses and principle component analyses were used to evaluate the predicative capability of the extracted data, and results were promising. Extracted data were also randomly assigned to either a training group (65%) or to a test group (35%) for artificial neural network modeling. Models best identified invasive breast carcinomas (212 predictions, 94% accurate) and benign non-neoplastic breast disease (96 predictions, 81.3% accurate). These results suggest that, after further development, the novel method may be useful for large scale clinical proteomic profiling.
