BACKGROUND RESULTS CONCLUSION Encapsulation of bioactive compounds within protein‐based nanoparticles has garnered considerable attention in the food and pharmaceutical industries because of its potential to enhance stability and delivery. Soy protein isolate (SPI) has emerged as a promising candidate, prompting the present study aiming to modify its properties through controlled thermal and trypsin treatments for improved encapsulation efficiency (EE) of lutein and its storage stability.The EE of lutein nanoparticles encapsulated using SPI trypsin hydrolysates (SPIT) with three varying degrees of hydrolysis (4.11%, 6.91% and 10.61% for SPIT1, SPIT2 and SPIT3, respectively) increased by 12.00%, 15.78% and 18.59%, respectively, compared to SPI. Additionally, the photostability of SPIT2 showed a remarkable increase of 38.21% compared to SPI. The superior encapsulation efficiency and photostability of SPIT2 was attributed to increased exposure of hydrophobic groups, excellent antioxidant activity and uniform particle stability, despite exhibiting lower binding affinity to lutein compared to SPI. Furthermore, in SPIT2, the protein structure unfolded, with minimal impact on overall secondary structure upon lutein addition.The precise application of controlled thermal and trypsin treatments to SPI has been shown to effectively produce protein nanoparticles with substantially improved encapsulation efficiency for lutein and enhanced storage stability of the encapsulated lutein. AbstractLutein, zeaxanthin, and mesozeaxanthin, collectively termed as macular pigments, are key carotenoids integral to optimized central vision of the eye. Therefore, nutraceuticals and functional foods have been developed commercially using carotenoid rich flowers, such as marigold and calendula or single celled photosynthetic algae, such as the Dunaliella. Industrial formulation of such products enriched in macular pigments have often suffered from serious bottlenecks in stability, delivery, and bioavailability. The two chief factors largely responsible for decreasing the shelf-life have been solubility and oxidation of these pigments owing to their strong lipophilic nature and presence of conjugated double bonds. In this regard, oil-based formulations have often been found to be more suitable than powder-based formulations in terms of shelf life and targeted delivery. In some cases, addition of phenolic acids in the formulations have also augmented the product value by enhancing micellization. In this regard, a novel proprietary formulation of these pigments has been developed in our laboratory utilizing marigold extracts in a colloidal solution of extra virgin olive oil and canola oil fortified with antioxidants like thyme oil, tocopherol, and ascorbyl palmitate. This review article presents an updated insight into the stability and bioavailability of industrially manufactured macular carotenoids together with their safety and solubility issues. The human body can easily convert β‐carotene into retinol (a precursor of vitamin A), a significant ingredient in dietary supplements. The need for commercial natural pigment production, including β‐carotene, has led to intensive research in this area. Processing after biosynthesis is a vital stage that involves the use of nontoxic solvents, efficient mechanical disruption of cells, and the isolation of the required compounds. With the growing popularity of green extraction methods and the increasing market demand for carotenoids due to their health benefits, the use of clean, nontoxic, flash‐hydrolyzed biomass for carotenoid production presents a significant advantage. Furthermore, the remaining biomass can serve as a protein source for animal feed, thereby aligning with the principles of the biorefinery concept. In this study, our aim was to optimize the disruption of cells in two microalgal species using flash hydrolysis (FH). We applied two distinct levels of pressure and four separate temperature conditions over a brief residence time. Following this, we employed an ethanol extraction method to assess the efficiency of carotenoid pigment extraction. Flash hydrolysis is a chemical‐free subcritical water‐based continuous‐flow process, injected with wet biomass slurry at a high temperature (150–250 °C) for a very short time (8–12 s) to obtain bioproducts. In this study, we discovered that the pigment β‐carotene constituted 0.04 to 0.3% of the dry biomass in the case of Chlorella vulgaris. Bands of lutein and zeaxanthin were also observed in the thin layer chromatography (TLC) analysis of the treated slurry. This was achieved under conditions of an effective temperature of 200 °C and a pressure of 1700 psi. The Arthrospira platensis (1Tex), β‐carotene yield was 0.014 to 0.021% of dry biomass. Some lutein and zeaxanthin bands were also found in the treated slurry with an effective temperature of 150 °C and 1000 psi pressure. This study reports pigment extraction by FH for the first time. It is a viable process to scale up high‐value natural products like carotenoids from microalgae at the industrial level without adverse environmental impacts. Due to low residence time, the temperature does not have a negative effect on pigments; cell rupture was accomplished effectively using water as a solvent. A considerable amount of pigment could be recovered from microalgae at specific combinations of temperature and pressure depending on the type of cell wall that microalgae possess. As a sustainable and ecological dye resource, the biomass materials are receiving more and more attention. In this work, the pigments in Cordyceps militaris were extracted and applied for dyeing wool fabrics, and the various properties were investigated. The extracted pigments and dyed wool fibers were analyzed and characterized by ultraviolet–visible (UV-Vis) spectroscopy, Liquid Chromatography-Mass Spectrometry (LC-MS) and Fourier Transform Infrared (FTIR) spectroscopy. The single-factor experiments were used to screen and optimize the dyeing process. Six metal mordants with different use order were compared with each other, and their possible mordanting mechanisms were proposed. The color fastnesses, UV resistance and antioxidant properties were evaluated after dyeing. The results showed that UV-Vis, LC-MS and FTIR analysis confirmed that the main components of C. militaris extract was lutein. The optimal dyeing process for the extract was as follows: dyestuff mass 200% (o.m.f.), dyeing temperature 90 °C, dyeing time 45 min, bath pH 5.0, Na2SO4 mass 0 g/L. Metal ions are added to form chelate bonds between the dyes and the fibers, further improving the color fastness. The dyed fabric exhibits excellent UV resistance and its UPF (Ultraviolet Protection Factor) value increases with the color yield. Meanwhile, the dyed wool fabric could exhibit up to 94.77% antioxidant activity. These results provide new insight for the application of C. militaris pigments on wool fabric. [ABSTRACT FROM AUTHOR]

Introduction: Excessive glutamate levels induce oxidative stress, resulting in neuronal damage, and cell death. While natural antioxidants show promise for neuroprotection, their effectiveness in the central nervous system (CNS) is limited by the blood-brain barrier. Lutein, a neuroprotective carotenoid, has gained attention for its ability to traverse this barrier and accumulate in various brain regions. This study aimed to elucidate the mechanisms underlying the protective effects of lutein against glutamateinduced cell death in HT22 cells. Methods: HT22 cells were treated with lutein (1.25-20 µM) for 24 hours. Cell viability, ROS levels, apoptosis, and mitochondrial membrane potential were assessed following lutein pretreatment and glutamate exposure. Protein expression of apoptotic markers was analyzed using Western blotting. Results: Lutein effectively attenuated glutamate-induced apoptosis due to its antioxidant properties. Additionally, lutein inhibited glutamate-induced mitochondrial-mediated apoptosis. We observed that lutein modulated the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2) and upregulated the expression of heme oxygenase-1 (HO-1). Inhibition of HO-1 by tin protoporphyrin (SnPP), a synthetic inhibitor, weakened the protective effect of lutein. Furthermore, we demonstrated that lutein prevented the aberrant activation of MAPKs induced by glutamate, including ERK1/2, p38, and JNK, thereby conferring oxidative protection. Discussion: Our study highlights the potent antioxidant properties of lutein, which effectively safeguards against glutamate-induced mitochondrial apoptotic cell death through the Nrf2/HO-1 signaling pathway and inhibition of MAPK activation. Patients with sickle cell disease (SCD) exhibit high levels of reactive oxygen species and low plasma levels of lipophilic antioxidants, which may contribute to end‐organ damage and disease sequelae. Apolipoprotein A1, the major apolipoprotein of high‐density lipoprotein (HDL), is mainly secreted by the intestine and liver in the form of monomeric ApoA1 (mApoA1) present in plasma. Cholesterol and α‐tocopherol are delivered to ApoA1 via the ATP‐binding cassette transporter, subfamily A, member 1 (ABCA1). We measured cholesterol, mApoA1, ApoA1, and lipophilic antioxidants in the plasma of 17 patients with SCD and 40 healthy volunteers. Mean HDL cholesterol (‐C) levels in SCD patients and healthy subjects were 59.3 and 48.1 mg/dL, respectively, and plasma lutein, zeaxanthin, and α‐tocopherol were 64.0%, 68.7%, and 9.1% lower, respectively. To compare SCD to healthy subjects with similar HDL‐C, we also performed subgroup analyses of healthy subjects with HDL‐C above or below the mean. In SCD, the mApoA1 level was 30.4 μg/mL; 80% lower than 141 μg/mL measured in healthy volunteers with similar HDL‐C (56.7 mg/dL). The mApoA1 level was also 38.4% greater in the higher versus lower HDL‐C subgroups (p = .002). In the higher HDL‐C subgroup, lutein and zeaxanthin transported by HDL were 48.9% (p = .01) and 41.9% (p = .02) higher, respectively, whereas α‐tocopherol was 31.7% higher (p = .003), compared to the lower HDL‐C subgroup. Plasma mApoA1 may be a marker of the capacity of HDL to capture and deliver liposoluble antioxidants, and treatments which raise HDL may benefit patients with high oxidative stress as exemplified by SCD. Summary Co‐digestion of cantaloupe (C) melon fresh cuts (FC) coated with probiotics [Lactiplantibacillus plantarum 75/xanthan gum with Natal plum (NP)] or beetroot leaves (BL) was evaluated to determine their bioaccessibility of individual anthocyanins (cyanidin‐3‐glucoside, cyanidin‐3‐O‐sambubioside) and carotenoids (lutein, trans‐β‐carotene and cis‐β‐carotene). Co‐digestion of probiotic‐coated C‐melon‐FC with NP increased the percentage (%) bioaccessibility of cyanidin‐3‐glucoside (11.18%) and cyanidin‐3‐O‐sambubioside (8.89%) compared to single digestion of Natal plum. Bioaccessibility of lutein (18.2%), trans‐β‐carotene (36.92%) and cis‐β‐carotene (16.74%) increased when co‐digestion of probiotic‐coated C‐melon‐FC with BL compared to the single digestion of uncoated C‐melon. Co‐digestion of C‐melon‐FC‐L75 with BL showed the highest antioxidant activity. L. plantarum had the highest LAB survival at the intestinal phase (6.01–6.26 Log CFU mL−1) during single and co‐digestion showing probiotic potential. Lutein is widely used in medicine, health care, and food processing due to its antioxidant effects; however, it is difficult for the traditional extraction of lutein using marigolds to meet the increasing market demand for lutein. To achieve high-efficiency lutein production, we investigated the effects of different conditions on the biomass accumulation and lutein yield of Chlorella vulgaris. The optimized cultivation conditions include mixotrophic cultivation using sodium nitrate as a nitrogen source, maintaining a total-organic-carbon-to-total-nitrogen ratio of 12:1, a total-nitrogen-to-total-phosphorus ratio of 10:1, and lighting duration of 24 h. The results of the study indicated that under these specific conditions, Chlorella vulgaris attained a final biomass concentration, biomass productivity, and growth yield of 6.08 g·L−1, 1.00 g·L−1·d−1, and 1.67 g biomass/g TOC, respectively. Additionally, the concentrations of total chlorophyll, carotenoid, lutein, and protein reached 139.20 mg·L−1, 31.87 mg·L−1, 15.02 mg·L−1, and 2.17 g·L−1, respectively, and the content of lutein reached 2.47 mg·g−1. To prepare different types of lutein nanostructured lipid carriers (LNLs) using chitosan (CS), chitosan-epigallocatechin gallate covalent (C-CS-EGCG), and chitosan-epigallocatechin gallate ester non-covalent (Non-C-CS-EGCG) modifications combined with high-pressure microfluidization. A simulated colonic digestion model was established to investigate the stability and interaction with gut microbiota of these modified lutein nanostructured lipid carriers. Results showed that after 24 hours of simulated in vitro colonic fermentation, the lutein content in CS-LNLs significantly decreased (P<0.05) compared to other treatment groups. Except for lutein and CS-LNLs, the total sugar content in the fermentation supernatant significantly decreased (P<0.05) in other treatment groups, while the total polyphenol content in C-CS-EGCG-LNLs and Non-C-CS-EGCG-LNLs showed an opposite trend. After 24 hours of fermentation, the production of acetic acid and propionic acid significantly increased (P<0.05) in all treatment groups except the LUT group, with the acetic acid concentration reaching 17.18±0.60 mmol/L in the C-CS-EGCG-LNLs group. The lactic acid concentration reached 2.87±0.05 mmol/L, significantly higher (P<0.05) than the blank control group (BLK) by 7.55 times. The three different types of modified lutein nanostructured lipid carriers had a significant impact on the gut microbiota structure. The relative abundance of Bacteroides, Bifidobacterium, and Faecalibacterium significantly increased (P<0.05) in CS-LNLs, C-CS-EGCG- LNLs, and Non-C-CS-EGCG-LNLs, while C-CS-EGCG-LNLs enriched more Veillonella. Conclusion: Compared to other modification groups, C-CS-EGCG-LNLs had significant intestinal microbial structural differentiation in the colon digestive phase of nanoliposomes. To improve the solubility and stability of lutein (LUT), this study utilized the electrostatic self-assembly method to encapsulate LUT within food-grade protein nanoparticles and composite nanoparticles derived from soybean protein isolates (SPI) and fucoidin (FUD). The impact of the SPI/FUD mass ratio on the formation of ternary composite nanoparticles, known as lutein-soybean protein isolate-fucoidin nanoparticles (LUT-SPI-FUD NPs), was examined. The preparation process underwent optimization through single-factor experiments and the Box-Behnken response surface methodology. Various characterization techniques, including ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, differential scanning calorimetry and X-ray diffraction were employed to delve into the mechanisms governing the formation of these two types of nanoparticles to compare with their solubility, stability, and antioxidant properties. The results showed that LUT-SPI-FUD NPs with SPI/FUD mass ratio of 4:1 had an absolute value of zeta potential greater than 30 mV, a polydispersity index, and a smaller particle size. Under these conditions, the most favorable formulation and process parameters were as follows: A LUT/SPI mass ratio of 1:7.587, SPI mass concentration of 0.83 mg/mL, and a stirring time of 1.493 hours. Characterization analysis verified the ideal particle size and more concentrated distribution of both LUT-SPI NPs and LUT-SPI-FUD NPs, and the binding forces were the interplay of hydrogen bonds, hydrophobic interactions, and electrostatic forces. Solubility, stability, and antioxidant assessments indicated that the conversion of LUT into LUT-SPI NPs and LUT-SPI-FUD NPs substantially increased its solubility in purified water by 350.74 and 432.42 times, respectively. Moreover, both formulations very significant (P<0.01) enhanced the stability and antioxidant capacity of LUT when subjected to ultraviolet light, high temperatures, and oxygen exposure, with LUT-SPI-FUD NPs exhibiting superior performance over LUT-SPI NPs. Consequently, the composite food-grade nanocarrier system, built upon SPI and FUD, represented a more effective technology for augmenting the solubility, stability, and antioxidant properties of LUT, and would offer a promising avenue for future LUT applications. Background Results Conclusions The composition of extra virgin olive oil (EVOO) defines its sensory, nutritional, and human health benefits, and distinguishes it as a key component of the Mediterranean diet. Nevertheless, EVOO constituents are susceptible to degradation during processing and storage, which reduces the olive oil's quality and limits its shelf life. The present study investigated the effect of molecular filtration before storage and the effect of cool storage at 4 °C on the stability of ‘Kolovi’ EVOO, a variety originating from the Greek island of Lesvos, over a 24 month period.Storing EVOO at 4 °C positively affected free acidity, peroxide value, K268, fruity qualities, and concentrations of hydroxytyrosol, tyrosol, ligstroside aglycone, lutein, and squalene, in comparison with the control sample stored at room temperature, particularly after 1 year. Molecular filtration significantly affected the ratio of unsaturated fatty acids to saturated fatty acids (UFAs/SFAs). Optimal preservation of parameters such as acidity value and lutein content was achieved by combining molecular filtration with refrigeration.The present study recommends storing EVOO in the refrigerator for up to 18 months. Based on the regulatory limits of the quality characteristics of acidity, peroxide value, K232 value and fruity sensory attributes, the shelf‐life of the protected geographical indication (PGI) ‘Kolovi’ EVOO can reach 2 years under cool storage (4 °C) and with molecular filtration before storage. © 2024 The Author(s). BACKGROUND RESULTS CONCLUSION Rice bran oil body is rich in nutritional value, which is a byproduct of rice processing. The aim of this study is to develop a novel emulsion‐filled gel with lutein‐loaded rice bran oil body and investigate its functionality as a fat replacer in cookies. The effects of incorporating structured oil body in the form of emulsion‐filled gel instead of butter in cookies with a ratio of 0, 10, 20 and 50 wt% formulation were determined by measuring appearance, texture, thermodynamic properties, moisture distribution and microstructure.The results demonstrated the relationship between geometry, moisture and structure. The 20 wt% emulsion‐filled gel substitution ratio yielded mobility and distribution abilities of melted fat and sugar in the cookies that were closest to those of butter. The addition of emulsion‐filled gel increased the L* value and decreased the a* value, while the b* value of the cookie increased due to the advanced delivery of lutein by oil body. By controlling the addition ratio, the texture of the cookies can be adjusted. Starch granules were separated due to colloidal particles, reducing saturated fat content and decreasing cookie gelatinization enthalpy. The fat coating on starch particles enhanced the binding capacity of free water, improving air entrapment and forming a constrained gluten network structure.These findings provide a theoretical basis for rice bran oil body as a novel substitute for butter in the development of healthy, high‐quality cookies. © 2024 Society of Chemical Industry. Blue light exposure of the ocular apparatus is currently rising. This has motivated a growing concern about potential deleterious effects on different eye structures. To address this, ARPE-19 cells were used as a model of the retinal pigment epithelium and subjected to cumulative expositions of blue light. The most relevant cellular events previously associated with blue-light-induced damage were assessed, including alterations in cell morphology, viability, cell proliferation, oxidative stress, inflammation, and the induction of DNA repair cellular mechanisms. Consistent with previous reports, our results provide evidence of cellular alterations resulting from repeated exposure to blue light irradiation. In this context, we explored the potential protective properties of the vegetal extract from Polypodium leucotomos, Fernblock® (FB), using the widely known treatment with lutein as a reference for comparison. The only changes observed as a result of the sole treatment with either FB or lutein were a slight but significant increase in γH2AX+ cells and the raise in the nuclear levels of NRF2. Overall, our findings indicate that the treatment with FB (similarly to lutein) prior to blue light irradiation can alleviate blue-light-induced deleterious effects in RPE cells, specifically preventing the drop in both cell viability and percentage of EdU+ cells, as well as the increase in ROS generation, percentage of γH2AX+ nuclei (more efficiently with FB), and TNF-α secretion (the latter restored only by FB to similar levels to those of the control). On the contrary, the induction in the P21 expression upon blue light irradiation was not prevented neither by FB nor by lutein. Notably, the nuclear translocation of NRF2 induced by blue light was similar to that observed in cells pre-treated with FB, while lutein pre-treatment resulted in nuclear NRF2 levels similar to control cells, suggesting key differences in the mechanism of cellular protection exerted by these compounds. These results may represent the foundation ground for the use of FB as a new ingredient in the development of alternative prophylactic strategies for blue-light-associated diseases, a currently rising medical interest. Ocular health has emerged as one of the major issues of global health concern with a decline in quality of life in an aging population, in particular and rise in the number of associated morbidities and mortalities. One of the chief reasons for vision impairment is oxidative damage inflicted to photoreceptors in rods and cone cells by blue light as well as UV radiation. The scenario has been aggravated by unprecedented rise in screen-time during the COVID and post-COVID era. Lutein and Zeaxanthin are oxygenated carotenoids with proven roles in augmentation of ocular health largely by virtue of their antioxidant properties and protective effects against photobleaching of retinal pigments, age-linked macular degeneration, cataract, and retinitis pigmentosa. These molecules are characterized by their characteristic yellow-orange colored pigmentation and are found in significant amounts in vegetables such as corn, spinach, broccoli, carrots as well as fish and eggs. Unique structural signatures including tetraterpenoid skeleton with extensive conjugation and the presence of hydroxyl groups at the end rings have made these molecules evolutionarily adapted to localize in the membrane of the photoreceptor cells and prevent their free radical induced peroxidation. Apart from the benefits imparted to ocular health, lutein and zeaxanthin are also known to improve cognitive function, cardiovascular physiology, and arrest the development of malignancy. Although abundant in many natural sources, bioavailability of these compounds is low owing to their long aliphatic backbones. Under the circumstances, there has been a concerted effort to develop vegetable oil-based carriers such as lipid nano-emulsions for therapeutic administration of carotenoids. This review presents a comprehensive update of the therapeutic potential of the carotenoids along with the challenges in achieving an optimized delivery tool for maximizing their effectiveness inside the body. Lutein and zeaxanthin are the two most abundant natural xanthophylls (oxygenated carotenoids) with a linear C40 tetraterpene/isoprenoid lycopene-based backbone. Presence of extensive conjugation (more than 10 double bonds) enable these molecules to act as accessory light harvesting pigments apart from chlorophyll. More importantly, the xanthophylls prevent photobleaching of the pigments and proteins in the Light Harvesting Complex (LHC) by sequestering the excess unutilized blue light and preventing triplet chlorophyll associated formation of Reactive Oxygen Species. In human eye, lutein, zeaxanthin along with mesozeaxanthin constitute the three macular pigments forming the so called "yellow spot" of the macula and are implicated in maintaining the redox balance, homeostasis and normal physiology of the eyes. However, unlike plants, xanthophylls must be acquired from dietary sources such as colored leafy vegetables and egg yolk. Increase in the number of eye diseases in the aging population coupled with insufficient bioavailability of xanthophylls has mandated the industrial production of supplements enriched in xanthophylls. Carotenoids, with their diverse biological activities and potential pharmaceutical applications, have garnered significant attention as essential nutraceuticals. Microalgae, as natural producers of these bioactive compounds, offer a promising avenue for sustainable and cost-effective carotenoid production. Despite the ability to cultivate microalgae for its high-value carotenoids with health benefits, only astaxanthin and β-carotene are produced on a commercial scale by Haematococcus pluvialis and Dunaliella salina, respectively. This review explores recent advancements in genetic engineering and cultivation strategies to enhance the production of lutein by microalgae. Techniques such as random mutagenesis, genetic engineering, including CRISPR technology and multi-omics approaches, are discussed in detail for their impact on improving lutein production. Innovative cultivation strategies are compared, highlighting their advantages and challenges. This study aimed to determine the optimal pressure and temperature for the maximum extraction yield and recovery of lipophilic bioactive compounds (BACs) during the supercritical carbon dioxide extraction (SCO2) of fennel seeds. For this purpose, the SCO2 pressure (78.6–361.4 bar) and temperature (35.9–64.1 °C) were varied and optimized, and all of the extracts obtained were analyzed for the volatiles, fatty acids, sterols, tocochromanols and carotenoids. The results showed that the maximum extract yield and content of all of the compounds analyzed favored a higher pressure (320 bar) and lower temperature (40 °C), except for the volatiles, which were the highest at 120 bar and 42 °C. However, the optimal SCO2 conditions for obtaining the highest overall total lipophilic fraction were 320 bar and 40 °C, respectively. The fennel SCO2 extract obtained under these conditions contained 18 volatiles (trans-anethole as the major component), 12 fatty acids (oleic and petroselinic as the major compounds), 12 sterols (β-sitosterol and stigmasterol as the major compounds), two pentacyclic triterpenoids (α-, β-amyrin), one tocopherol (α-tocopherol), two tocotrienols (γ-, δ-tocotrienol) as well as two carotenoids (lutein and β-carotene). The microalgae Chlorella sorokiniana was used for the treatment of winery wastewater (WWW). Batch experiments were initially conducted to investigate how biomass acclimatization in different media, dilution of wastewater, and addition of ammonium nitrogen (NH4-N) affect the growth of microalgae and the removal of major pollutants. Afterwards, two sequencing batch reactor (SBR) systems were tested applying different configurations and hydraulic retention times. The biomass collected at the end of the experiments was characterized for proteins, lipids, carbohydrates, amino acid profile, and the existence of lutein, β-carotene, chlorophyll a, and tocopherols. Batch experiments showed that Chlorella sorokiniana acclimatization to urban wastewater enhanced the removal of NH4-N and total phosphorus (TP). The operation of a two-stage SBR system achieved COD and NH4-N removal equal to 85 ± 9% and 91 ± 20%, respectively, while the use of a single-stage system feeding with anaerobically pretreated WWW resulted to COD and NH4-N removal of 78 ± 9% and 95 ± 9%, respectively. Analyses of biomass showed higher protein content (up to 58.8%) in batch experiments with NH4-N addition as well as in SBR experiments. The cultivation of microalgae under SBR conditions enhanced the production of pigments and tocopherols. The maximum concentrations of 1075 mg kg−1, 45.5 mg kg−1, and 131.2 mg kg−1 were achieved for lutein, β-carotene, and tocopherols, respectively, in the one-stage system. Our findings suggested that Chlorella sorokiniana cultivation in WWW not only removed nutrients from WWW but also could potentially serve for the production of value-added ingredients used in food industry, cosmetics, and animal feedstock. Novel self-assembled aggregates of stearic acid (SA)-modified burdock polysaccharide (BP) for loading lutein were constructed, and the release and absorption properties of lutein in the aggregates in simulated gastrointestinal fluid were investigated. Three different degrees of substitution (DS) of SA-BPs were used to embed lutein, resulting in the encapsulation efficiency exceeding 90%. The aggregates were uniformly spherical, with a particle size range of 227–341 nm. XRD analysis revealed that lutein was present in a non-crystalline state within the aggregates. FT-IR and FS analysis demonstrated that lutein was located in the hydrophobic domains of SA-BP. The highest bioavailability of lutein in these aggregates reached 4.36 times that in the unmodified samples. These aggregates were able to remain stable in gastric juice and enhance the release rate of lutein in intestinal fluid. The transport of lutein-loaded SA-BP aggregates in Caco-2 cells competed with P-glycoprotein inhibitors, mainly promoting the transmembrane absorption of lutein through caveolae (or lipid raft)-related and clathrin-dependent endocytosis pathways. Opsins are a class of transmembrane proteins encoded by opsin genes, and they play a variety of functional roles. Short wavelength-sensitive opsin 2 (sws2), one of the five classes of visual opsin genes, mainly senses blue light. Previous research has indicated that sws2 is essential for melanocyte formation in fish; however, its specific role in skin color differentiation remains to be elucidated. Here, we identified the sws2 gene in a prized reef-dwelling fish, Plectropomus leopardus. The full-length P. leopardus sws2 gene encodes a protein consisting of 351 amino acids, and exhibits substantial homology with other fish species. The expression of the sws2 gene was widespread across P. leopardus tissues, with high expression in eye and skin tissues. Through immunohistochemistry and in situ hybridization analyses, we discovered that the sws2 gene was primarily localized in the rod and cone cells of the retina, and epidermal cells of the skin. Furthermore, dsRNA interference was used for sws2 gene knockdown in living P. leopardus to elucidate its function in skin color differentiation. Black-color-related genes, melanin contents, and tyrosinase activity in the skin significantly decreased after sws2 knockdown (p < 0.05), but red-color-related genes and carotenoid and lutein contents significantly increased (p < 0.05). Retinoic acid injection produced the opposite results. Our results suggested that the sws2 gene influences P. leopardus skin color regulation by affecting vitamin synthesis and melanin-related gene expression levels. Plant proteins have gained significant attention over animal proteins due to their low carbon footprint, balanced nutrition, and high sustainability. These attributes make plant protein nanocarriers promising for applications in drug delivery, nutraceuticals, functional foods, and other areas. Zein, a major by-product of corn starch processing, is inexpensive and widely available. Its unique self-assembly characteristics have led to its extensive use in various food and drug systems. Zein's functional tunability allows for excellent performance in loading and transporting bioactive substances. Lutein offers numerous bioactive functions, such as antioxidant and vision protection, but suffers from poor chemical stability and low bioavailability. Nano-embedding technology can construct various zein-loaded lutein nanodelivery systems to address these issues. This review provides an overview of recent advances in the construction of zein-loaded lutein nanosystems. It discusses the fundamental properties of these systems; systematically introduces preparation techniques, structural characterization, and functional properties; and analyzes and predicts the target-controlled release and bioaccessibility of zein-loaded lutein nanosystems. The interactions and synergistic effects between Zein and lutein in the nanocomplexes are examined to elucidate the formation mechanism and conformational relationship of zein–lutein nanoparticles. The physical and chemical properties of Zein are closely related to the molecular structure. Zein and its modified products can encapsulate and protect lutein through various methods, creating more stable and efficient zein-loaded lutein nanosystems. The global prevalence of Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) is increasing, now affecting 25%-30% of the population worldwide. MASLD, characterized by hepatic steatosis, results from an imbalance in lipid metabolism, leading to oxidative stress, lipoperoxidation, and inflammation. The activation of autophagy, particularly lipophagy, alleviates hepatic steatosis by regulating intracellular lipid levels. Lutein, a carotenoid with antioxidant and anti-inflammatory properties, protects against liver damage, and individuals who consume high amounts of lutein have a lower risk of developing MASLD. Evidence suggests that lutein could modulate autophagy-related signaling pathways, such as the transcription factor EB (TFEB). TFEB plays a crucial role in regulating lipid homeostasis by linking autophagy to energy metabolism at the transcriptional level, making TFEB a potential target against MASLD. STARD3, a transmembrane protein that binds and transports cholesterol and sphingosine from lysosomes to the endoplasmic reticulum and mitochondria, has been shown to transport and bind lutein with high affinity. This protein may play a crucial role in the uptake and transport of lutein in the liver, contributing to the decrease in hepatic steatosis and the regulation of oxidative stress and inflammation. This review summarizes current knowledge on the role of lutein in lipophagy, the pathways it is involved in, its relationship with STARD3, and its potential as a pharmacological strategy to treat hepatic steatosis. Introduction: Lung cancer is one of the most prevalent and deadly forms of cancer worldwide, accounting for a significant number of cancer-related deaths. Despite advances in diagnosis and treatment, the prognosis for lung cancer remains poor, emphasising the need for alternative and complementary therapeutic approaches. Transforming Growth Factor-beta (TGF-β) is a multifunctional cytokine that plays a crucial role in cell growth, differentiation, apoptosis and immune regulation. Suppressor of Mothers against Decapentaplegic (SMAD), a well known transcription factor, plays an essential role in carcinogenesis. Lutein, a naturally occurring carotenoid. Recent studies have suggested that lutein possess anticancer properties and could potentially be used as a therapeutic agent against various types of cancer. Aim: To assess the effect of lutein on TGF-β/SMAD2 gene expression in lung cancer cells. Materials and Methods: The present in-vitro study was carried in the Cancer and Stem Cell Laboratory Facility, Saveetha Dental College and Hospitals, Chennai, Tamil Nadu, India, from April 2023 to May 2023. The lung cancer cell line (A-549) was obtained from the National Centre for Cell Science (NCCS), Pune, India. Cell viability of lutein-treated lung cancer cells was assessed by 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) assay. Cell morphology was studied using a phase contrast microscope. The gene expression of TGF-β/SMAD2 was analysed using real-time Polymerase Chain Reaction (PCR), and the results were presented graphically as fold change. All data obtained were analysed by one-way Analysis of Variance (ANOVA) followed by Student’s t-test using PRISM software version 4. Results: The findings suggest that lutein had good cytotoxic effects on lung cancer cells. The Half-maximal inhibitory concentration (IC50) value was found to be 5.14 μM/mL. Lutein inhibited the TGF-β and SMAD2 gene expression signalling pathway and induced apoptosis in A549. Conclusion: Lutein significantly inhibited the TGF-β and SMAD2 gene expression signalling pathways, making it a potent anticancer drug. The downregulation of key genes involved in the TGF-β/SMAD2 pathway suggests a nuanced and intricate relationship between lutein and the regulatory mechanisms governing cellular signalling in lung cancer. These findings support the notion that lutein may exert a regulatory influence on the intricate balance of signalling cascades, offering a potential avenue for therapeutic intervention in lung cancer. Background: Atopic dermatitis (AD) is a chronic inflammatory skin disease that affects 15%‐30% of children and 10% of adults globally, with its incidence being influenced by genetic, environmental, and various other factors. While the immune plays a crucial role in the development, the composition of gut microbiota and serum metabolites also contribute to its pathogenesis. Subject: Study the characteristics of gut microbiota and serum metabolites in patients with atopic dermatitis Method: In this study, we collected stool and serum samples from 28 AD patients and 23 healthy individuals (NC) for metagenomic sequencing of gut microbiota and non‐targeted metabolomic sequencing of serum. Result: Our results revealed a lower diversity of gut microbiota in the AD group compared to the NC group. The predominant Phylum in AD patients were Bacteroidetes, Pseudomonas, and Verrucomicrobia, with the most dominant bacterial genus being Faecalibacterium. At the species level, Prevotella copri and Faecalibacterium prausnitzii were found to be the most abundant bacteria. Significant differences in serum metabolite profiles were observed between NC and AD patients, with noticeable variations in metabolite expression levels. The majority of metabolites in the serum of AD patients exhibited low expression, while a few showed high expression levels. Notably, metabolites such as Cholesterol glucuronide, Styrene, Lutein, Betaine, Phosphorylcholine, Taurine, and Creatinine displayed the most pronounced alterations. Pathogens have traditionally been studied in isolation within host systems; yet in natural settings they frequently coexist. This raises questions about the dynamics of co-infections and how host lifehistory traits might predict co-infection versus single infection. To address these questions, we investigated the presence of two parasites, a gut parasite (Isospora coccidians) and a blood parasite (Plasmodium spp.), in House Finches (Haemorhous mexicanus), a common passerine bird in North America. We then correlated these parasitic infections with various health and condition metrics, including hematological parameters, plasma carotenoids, lipid-soluble vitamins, blood glucose concentration, body condition, and prior disease history. Our study, based on 48 birds captured in Tempe, Arizona, US, in October 2021, revealed that co-infected birds exhibited elevated circulating lutein levels and a higher heterophil: lymphocyte ratio (H/L ratio) compared to those solely infected with coccidia Isospora spp. This suggests that co-infected birds experience heightened stress and may use lutein to bolster immunity against both pathogens, and that there are potentially toxic effects of lutein in co-infected birds compared to those infected solely with coccidia Isospora sp. Our findings underscore the synergistic impact of coparasitism, emphasizing the need for more co-infection studies to enhance our understanding of disease dynamics in nature, as well as its implications for wildlife health and conservation efforts. Purpose: To report the long-term (23 years) clinical and multimodal imaging features of acquired vitelliform lesions (AVLs) associated with nonneovascular age-related macular degeneration. Methods: Retrospective case report. Color and red-free fundus photographs, highresolution optical coherence tomography, fluorescein and indocyanine green angiography, and optical coherence tomography-angiography were performed. Results: A 58-year-old man presented with bilateral AVLs in the setting of nonneovascular age-related macular degeneration. At baseline, his best-corrected visual acuity was 20/30 in his right eye and 20/20 in his left eye. Red-free fundus photographs showed AVLs with cuticular drusen in both eyes corresponding to a "stars-in-the-sky" pattern on fluorescein. Indocyanine green angiography showed no evidence of macular neovascularization. Throughout the 23-year follow-up, the patient reported consuming 20 mg/day of lutein supplement. At the end of follow-up, his best-corrected visual acuity was 20/20 in both eyes. Color fundus photographs showed resorption of the AVLs in both eyes and High-Res optical coherence tomography showed relative preservation of the outer retinal bands in the fovea. Optical coherence tomography-angiography confirmed the absence of macular neovascularization. Conclusion: In nonneovascular age-related macular degeneration, spontaneous resorption of AVLs may be associated with long-term maintenance of visual acuity and relative preservation of the outer retinal morphology. Lutein is an oxygenated fat-soluble carotenoid and a functional compound with proven health benefits for the human body. Nevertheless, the poor water solubility and low oral bioavailability of lutein greatly limit its application. To address this, we developed an effective approach to enhance the water solubility of lutein through co-amorphous formulation. Specifically, the lutein-sucralose co-amorphous mixture was prepared at a molar ratio of 1:1 using ethanol and water as solvents by employing the solvent evaporation method, followed by solid-state characterization and dissolution testing conducted to assess the properties of the formulation. The X-ray diffraction pattern with an amorphous halo and the differential scanning calorimetry thermogram with no sharp melting peaks confirmed the formation of a binary co-amorphous system. Changes in peak shape, position, and intensity observed in the Fourier transform infrared spectroscopy spectrum revealed intermolecular interactions between lutein and sucralose molecules, while molecular dynamics simulations identified interaction sites between their hydroxyl groups. Additionally, dissolution testing demonstrated better dissolution performance of lutein in the co-amorphous form compared to pure lutein and physical mixture counterparts. Chromochloris zofingiensis, a unicellular green alga, is a potential source of natural carotenoids. In this study, the mutant LUT-4 was acquired from the chemical mutagenesis pool of C. zofingiensis strain. The biomass yield and lutein content of LUT-4 reached 9.23 g·L−1, and 0.209% of dry weight (DW) on Day 3, which was 49.4%, and 33% higher than that of wild-type (WT), respectively. The biomass yields of LUT-4 under 100, 300, and 500 µmol/m2/s reached 8.4 g·L−1, 7.75 g·L−1, and 6.6 g·L−1, which was 10.4%, 21%, and 29.6% lower compared with the control, respectively. Under mixotrophic conditions, the lutein yields were significantly higher than that obtained in the control. The light intensity of 300 µmol/m2/s was optimal for lutein biosynthesis and the content of lutein reached 0.294% of DW on Day 3, which was 40.7% more than that of the control. When LUT-4 was grown under 300 µmol/m2/s, a significant increase in expression of genes implicated in lutein biosynthesis, including phytoene synthase (PSY), phytoene desaturase (PDS), and lycopene epsilon cyclase (LCYe) was observed. The changes in biochemical composition, Ace-CoA, pyruvate, isopentenyl pyrophosphate (IPP), and geranylgeranyl diphosphate (GGPP) contents during lutein biosynthesis were caused by utilization of organic carbon. It was thereby concluded that 300 µmol/m2/s was the optimal culture light intensity for the mutant LUT-4 to synthesize lutein. The results would be helpful for the large-scale production of lutein. In this study, we investigated the factors involved in the flavor discrepancy between Norin61 and Satonosora wheat cultivars by analyzing the isozyme composition of lipoxygenase (LOX) and antioxidant levels. The results revealed that Norin61 exhibited a distinctive LOX isozyme composition, with a significant proportion of LOX-3, which correlates with its previously reported high LOX activity. Moreover, our analysis indicated that the content of lutein, an antioxidant present in flour, influences the formation of volatile flavor compounds. These results not only offer valuable insights for crafting products with specific flavors in the development of various wheat flour-based products, but also have potential genetic implications in the breeding of wheat cultivars boasting diverse flavors in the future. To improve the solubility and stability of lutein (LUT), this study utilized the electrostatic self-assembly method to encapsulate LUT within food-grade protein nanoparticles and composite nanoparticles derived from soybean protein isolates (SPI) and fucoidin (FUD). The impact of the SPI/FUD mass ratio on the formation of ternary composite nanoparticles, known as lutein-soybean protein isolate-fucoidin nanoparticles (LUT-SPI-FUD NPs), was examined. The preparation process underwent optimization through single-factor experiments and the Box-Behnken response surface methodology. Various characterization techniques, including ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, differential scanning calorimetry and X-ray diffraction were employed to delve into the mechanisms governing the formation of these two types of nanoparticles to compare with their solubility, stability, and antioxidant properties. The results showed that LUT-SPI-FUD NPs with SPI/FUD mass ratio of 4:1 had an absolute value of zeta potential greater than 30 mV, a polydispersity index, and a smaller particle size. Under these conditions, the most favorable formulation and process parameters were as follows: A LUT/SPI mass ratio of 1:7.587, SPI mass concentration of 0.83 mg/mL, and a stirring time of 1.493 hours. Characterization analysis verified the ideal particle size and more concentrated distribution of both LUT-SPI NPs and LUT-SPI-FUD NPs, and the binding forces were the interplay of hydrogen bonds, hydrophobic interactions, and electrostatic forces. Drought stress significantly affects crop productivity. Carotenoids are essential photosynthetic pigment for plants, bacteria, and algae, with signaling and antioxidant functions. Lutein is a crucial branch product in the carotenoid synthesis pathway, which effectively improves the stress tolerance of higher plants. lycopene cyclase, a central enzyme for lutein synthesis, holds great significance in regulating lutein production. This research establishes a correlation between lutein content and stress resistance by measuring the drought resistance and lutein content of various cotton materials. To identify which crucial genes are associated with lutein, the lycopene cyclase family (LCYs) was analyzed. The research found that LCYs form a highly conserved family divided into two subfamilies, LCY-ε (lycopene ε-cyclase) and LCY-β (lycopene β-cyclase). Most members of the LCY family contain photoresponsive elements and abscisic acid elements. qRT-PCR demonstrates showed that most genes responded positively to drought stress, and GhLCYε-3 was expressed significantly differently under drought stress. Virus-induced gene silencing (VIGS) assay showed that the content of GhLCYε-3 was significantly increased with MDA and PRO, and the contents of chlorophyll and lutein were significantly decreased in pYL156 plants. The decrease in GhLCYε-3 expression is speculated to lead to reduced lutein content in vivo, resulting in the accumulation of reactive oxygen species (ROS) and decreased drought tolerance. Biochemical parameters, including serum lipids, liver enzymes, nutrient concentrations, serum antioxidant enzyme activity (catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD)), and markers of oxidative stress (thiobarbituric acid reactive substances (TBARS) and ferric reducing ability (FRAP)), were assessed before and after the dietary interventions. Additionally, arterial blood pressure, heart rate, body composition, fluid status, anthropometric measurements, and skin microvascular blood flow responses to various stimuli (postocclusive reactive hyperemia (PORH), acetylcholine- (Ach ID), and sodium nitroprusside- (SNP ID)) were measured using laser Doppler flowmetry (LDF) throughout the study.Results: The intake of Nutri4 eggs led to a significant reduction in LDL cholesterol levels, while the levels of total cholesterol remained within the established reference values. Consuming Nutri4 eggs resulted in a 12.7% increase in serum vitamin E levels, an 8.6% increase in selenium levels, and demonstrated a favorable impact on microvascular reactivity, as evidenced by markedly improved PORH and ACh ID. Carotenoids and chlorophylls are among the most widely distributed pigments in nature that play essential roles in the photosynthetic apparatus and confer diverse colours in plants. Among all vegetables, cauliflower (Brassica oleracea L. ssp. var. botrytis) is rich in phytochemicals and is an important crop grown all over the world. This study investigates carotenoid and chlorophyll concentrations in differently pigmented cultivars and elucidates the role of transcriptional regulation of carotenoid accumulation including lutein and β-carotene. Here, we characterised changes in pigments by UHPLC-DAD-ToF-MS and changes in transcript levels of carotenoid metabolic genes by qRT-PCR in florets and leaves of orange (‘Jaffa’ and ‘Sunset’), purple (‘Di Sicilia Violetto’ and ‘Graffiti’), green (‘Trevi’) and white (‘Clapton’) cultivars. Transcript levels of all carotenoid metabolic genes showed different transcript level patterns in the leaves and florets. Compared to the other cultivars, the orange cultivars had the highest levels of β-carotene in the florets and lutein in the leaves resulting in changes lutein/β-carotene ratios. In the green cultivar, higher transcript levels were also found, especially for phytoene synthase and phytoene desaturase genes of the core biosynthesis pathway. However, no increased carotenoid concentrations were observed, possibly due to a higher carotenoid turnover induced by the carotenoid cleavage dioxygenase 4 in the green cultivar. In the white (‘Clapton’) and purple (‘Di Sicilia Violetto’ and ‘Graffiti’) cultivars the phytoene desaturase transcript levels as well as carotenoid concentrations were low. Chlorophyll concentrations changed in trend comparable to the carotenoid concentrations and were only significantly lower in the leaves of the orange cultivar ‘Jaffa’. Also, the chlorophyll a/b ratio changed in ‘Jaffa’. In florets the highest chlorophylls concentrations were observed for the green cultivar (‘Trevi’) and the purple cultivar (‘Di Sicilia Violetto’). Taken together, the study demonstrates the complex source-sink relationship of carotenoid accumulation in different coloured cauliflower.

Lutein, a plant-derived xanthophyl-carotenoid, is an exceptional antioxidant and anti-inflammatory constituent found in food. High dietary intake of lutein is beneficial against eye disease, improves cardiometabolic health, protects from neurodegenerative diseases, and is beneficial for liver, kidney, and respiratory health. Lutein protects against oxidative and nitrosative stress, both of which play a major role in post-COVID and mRNA vaccination injury syndromes. Lutein is an important natural agent for therapeutic use against oxidative and nitrosative stress in chronic illnesses such as cardiovascular and neurodegenerative diseases and cancer. It can also potentially inhibit spike protein-induced inflammation. Rich dietary supplementation of lutein, naturally derived in non-biodegradable Extra Virgin Olive Oil (EVOO), can most optimally be used against oxidative and nitrosative stress during post-COVID and mRNA vaccination injury syndromes. Due to its high oleic acid (OA) content, EVOO supports optimal absorption of dietary lutein. The main molecular pathways by which the SARS-CoV-2 spike protein induces pathology, nuclear factor kappa-light-chain-enhancer activated B cells (NF-κB) and activated protein (AP)-1, can be suppressed by lutein. Synergy with other natural compounds for spike protein detoxification is likely.

This work designed and synthesized the first organic fluorescent sensor for lutein in aqueous solutions based on blue to yellow fluorescence change of thiosemicarbazide‐bridged bis‐tetraphenylene (Bis‐TPE). By condensing monohydroxy tetraphenylene derivatives with p‐phenylene di‐isothiocyanate, Bis‐TPE was produced. A fluorescence spectrophotometer was used to scan the fluorescence emission spectra from 380 to 700 nm with 360 nm as the excitation wavelength. The fluorescence spectra showed that Bis‐TPE exhibited high selective sensing ability for lutein in aqueous medium, and the fluorescence showed red‐shift phenomenon with a limit of detection as low as 0.26 μM for lutein. The sensing mechanism was confirmed by mass spectrometry, infrared, and fluorescence Job's plots based on the 1:1 stoichiometric ratio of double hydrogen bonding. The successful application of Bis‐TPE in sensing test strips means that Bis‐TPE can achieve efficient detection of lutein in complex environments. As a result, an aggregation‐induced luminescence‐based lutein fluorescence sensor was constructed, which exhibited highly selective and sensitive sensing of lutein, and provided a new research idea for the object detection of such structures. Microalgae are gaining attention as they are considered green fabrics able to synthesize many bioactive metabolites, with unique biological activities. However, their use at an industrial scale is still a challenge because of the high costs related to upstream and downstream processes. Here, a biorefinery approach was proposed, starting from the biomass of the green microalga Pseudococcomyxa simplex for the extraction of two classes of molecules with a potential use in the cosmetic industry. Carotenoids were extracted first by an ultrasound-assisted extraction, and then, from the residual biomass, lipids were obtained by a conventional extraction. The chemical characterization of the ethanol extract indicated lutein, a biosynthetic derivative of α-carotene, as the most abundant carotenoid. The extract was found to be fully biocompatible on a cell-based model, active as antioxidant and with an in vitro anti-aging property. In particular, the lutein-enriched fraction was able to activate Nrf2 pathway, which plays a key role also in aging process. Finally, lipids were isolated from the residual biomass and the isolated fatty acids fraction was composed by palmitic and stearic acids. These molecules, fully biocompatible, can find application as emulsifiers and softener agents in cosmetic formulations. Thus, an untapped microalgal species can represent a sustainable source for cosmeceutical formulations. The study focuses on the association between serum carotenoids and cancer-related death. Using data from the National Health and Nutrition Examination Survey (2001–2006 and 2017–2018), the study encompasses 10,277 participants older than age 20 years, with recorded baseline characteristics and serum carotenoid concentrations (including α-carotene, trans -β-carotene, cis -β-carotene, β-cryptoxanthin, trans -lycopene, and lutein/zeaxanthin). We hypothesized that serum carotenoid concentrations were negatively associated with cancer-related death. The weighted chi-square analyses indicate significant negative correlations between higher serum concentrations of α-carotene, β-cryptoxanthin, trans -lycopene, and total carotenoids, and the risk of cancer-related deaths. Using weighted Cox regression analysis, this study confirms that α-carotene, β-cryptoxanthin, trans -lycopene, and total carotenoids, as continuous or categorical variables, are inversely related to cancer mortality (P <.0001). Furthermore, considering competitive risk events, lower concentrations of serum β-cryptoxanthin (Fine-Gray P = 1.12e-04), trans -lycopene (P = 5.68e-14), and total carotenoids (P =.03) are associated with an increased risk of cancer-related deaths. The Mediterranean-Dietary Approaches to Stop Hypertension (DASH) Intervention for Neurodegenerative Delay (MIND) dietary pattern is associated with reduced cognitive decline and dementia risk. However, the nutrient features that distinguish the MIND from other patterns are unknown. We investigated the relationship between accordance to the MIND pattern and carotenoid intake (phytonutrients hypothesized to confer neuroprotection) relative to the Mediterranean, DASH, and Healthy Eating Index (HEI-2020). We hypothesized that MIND diet accordance would be a stronger predictor of carotenoid intake relative to other diet indices. A total of 396 adults (aged 19–82 years) completed the Dietary History Questionnaire to assess carotenoid intake and adherence to each diet index. Stepwise regressions with adjustment for covariates followed by the Meng's Z-test were used to compare correlation strength between each diet pattern and carotenoid. All diet patterns were positively associated with lutein and zeaxanthin, β-carotene, α-carotene, and β-cryptoxanthin (all βs ≥0.38, P s <.01). Effect size comparisons revealed that MIND accordance predicted a greater proportion of variance in lutein and zeaxanthin (all Z s ≥ 3.3, P s <.001) and β-carotene (all Z s ≥ 2.6, P s <.01) relative to the Mediterranean, DASH, and HEI-2020. MIND accordance explained a greater proportion of variance in α-carotene (Z = 3.8, P <.001) and β-cryptoxanthin (Z = 3.6, P <.001) relative to the HEI-2020. MIND diet accordance was disproportionately related to carotenoid intake, indicating the MIND index places greater emphasis on carotenoid-rich foods, particularly those containing lutein, zeaxanthin, and β-carotene, relative to other diet indices. Future research is needed to define the role of these carotenoids in nutritional interventions for cognitive health. Greater adherence to the MIND diet pattern was disproportionately associated with higher dietary lutein + zeaxanthin and β-carotene intake relative to the Mediterranean, DASH, and HEI-2020 patterns among 396 healthy adults. The green microalga Scenedesmus almeriensis shows promise for natural carotenoid production, specifically lutein, a pigment inducing health benefits, often lacking in the human diet. In this study, S. almeriensis was successfully cultured heterotrophically for the first time. Heterotrophic biomass accumulation and pigment productivity were compared to a phototrophic culture, and a period of photoinduction followed the heterotrophic culture to stimulate carotenoid pigment synthesis. Under optimal glucose and sodium nitrate levels, the heterotrophic culture achieved a cell density of 22 g L–1 Lutein and violaxanthin contents were 2.2 and 1.5 times higher in the phototrophic culture compared to the heterotrophic culture, while zeaxanthin content was 1.8 times higher in the heterotrophic culture. However, heterotrophic culture exhibited higher productivity for lutein, violaxanthin, and zeaxanthin, with 9.79, 2.08, and 2.46 mg L–1 day–1, respectively. Additionally, the photoinduction stage enhanced carotenogenesis and increased productivity, reaching productivities of 11.52, 4.56, and 5.09 mg L–1 day–1 for lutein, violaxanthin, and zeaxanthin, respectively. To describe the variability in carotenoid content of human milk (HM) in mothers of very to extremely low birth weight preterm infants throughout lactation and to explore the relationship between lutein in HM and the occurrence of retinopathy of prematurity (ROP) in preterm infants. We recruited healthy mothers along with their preterm infants that were born at gestational age 24 + 2 to 29 + 6 weeks or with a birth weight under 1500 g and were exclusively breastfed HM. Each participant provided up to 7 HM samples (2–10 ml) on day 0–3 and once a week until 6 weeks. Additionally, when possible, a blood sample was collected from the infant at week 6. Concentrations of the major carotenoids (lutein, zeaxanthin, beta-carotene, and lycopene) in all HM and blood samples were assessed and compared. Thirty-nine mother-infant dyads were included and 184 HM samples and 21 plasma samples were provided. Mean lutein, zeaxanthin, beta-carotene, and lycopene concentration decreased as lactation progressed, being at their highest in colostrum samples (156.9 vs. 66.9 vs. 363.9 vs. 426.8 ng/ml, respectively). Lycopene (41%) and beta-carotene (36%) were the predominant carotenoids in colostrum and up to 2 weeks post-delivery. Inversely, the proportion of lutein and zeaxanthin increased with lactation duration to account for 45% of the carotenoids in mature HM. Lutein accounted for 58% of the carotenoids in infant plasma and only 28% in HM. Lutein content of transition and mature HM did not differ between mothers of ROP and non-ROP infants. Conclusion Carotenoid content of HM was dynamic and varied between mothers and as lactation progressed. Infant plasma displayed a distinct distribution of carotenoids from HM. Simple Summary: The abdominal fat of broiler chickens was used as one of the important parts for color evaluation, which directly affects the market price and demand of broilers. This study aimed to identify the contributors and the regulatory network involved in the formation of yellow and white color in abdominal fat. Four xanthophyll compounds were generally significantly correlated with eight metabolites. The high expression of 3-hydroxyanthranilate 3,4-dioxygenase (HAAO) in the yellow abdominal fat group leads to the degradation of tryptophan and its intermediate 5-hydroxyindole compounds, and subsequently to the formation of the four xanthophyll compounds. This study provides a genetic theoretical basis for improving the external appearance and internal quality of chicken abdominal fat. Abdominal fat, which in the past was often regarded as waste and discarded, has in recent years been used as a fat source to produce meat by-products. Yellow abdominal fat has higher economic value. Therefore, improving the color of abdominal fat plays an important role in improving the appearance of meat products. This study aimed to identify the contributors and the regulatory network involved in the formation of yellow and white color in abdominal fat. We found that four xanthophyll compounds were significantly different in yellow and white abdominal fat chicken, including zeaxanthin, lutein, canthaxanthin, and β-cryptoxanthin. There were 551 different and 8 common metabolites significantly correlated with these 4 xanthophyll compounds. Similarly, a total of 54 common genes were identified in 4 common related pathways (Complement and coagulation cascades, Metabolic pathways, PPAR signaling pathway, Carbon metabolism) of the 8 common metabolites. The high expression of HAAO in the yellow abdominal fat group leads to the degradation of tryptophan and its intermediate 5-hydroxyindole, and subsequently to the formation of the four xanthophyll compounds. This process is also regulated by tyrosine, kynurenine 3-monooxygenase (KMO), homogentisate 1, 2-dioxygenase (HGD), etc. Together, these findings show the effect of tryptophan on abdominal fat color, as well as a negative regulatory effect of HAAO and 5-hydroxyindole on the production of xanthophyll compounds involved in abdominal fat coloration. Maize (Zea mays L.) stands as a vital staple food globally, holding significant nutritional and economic value. However, its susceptibility to mycotoxin contamination under stressful environmental conditions poses a considerable concern. This study aimed to assess the quality and pasting characteristics of maize varieties across two distinct regions and examine the occurrence of mycotoxins influenced by climatic factors. Five maize varieties were cultivated in triplicate in the Golegã and Coruche regions. The nutritional composition (protein, fat, fiber, ash, starch, and lutein), pasting properties, and mycotoxin levels were evaluated. A statistical analysis revealed notable differences in the nutritional profiles of the maize varieties between the two regions, particularly in the protein and lutein content. The peak viscosity ranged from 6430 to 8599 cP and from 4548 to 8178 cP in the maize varieties from the Coruche and Golegã regions, respectively. Additionally, a significant correlation was observed between the climatic conditions and the grain nutritional quality components (p < 0.05). The M variety showed the highest ash content, protein content, final viscosity, and setback viscosity and the lowest peak viscosity. The Y variety revealed the lowest fat, fiber, and lutein content and the maximum peak viscosity. The incidence of mycotoxins was notably higher in the varieties from Coruche, which was potentially attributable to higher temperatures and lower precipitation levels leading to more frequent drought conditions. Fumonisin B1 was detected in 58% of the varieties from Coruche and 33% of the samples from Golegã, while deoxynivalenol was found in 87% and 80% of the varieties from Coruche and Golegã, respectively. The H variety, which was harvested in Coruche, exhibited the highest number of fumonisins and higher amounts of protein, lutein, and fat, while fumonisins were not detected in the Golegã region, which was potentially influenced by the precipitation levels. The K variety revealed higher protein and lutein contents, a lower amount of fat, excellent pasting properties (a higher peak viscosity and holding strength and a lower peak time), and no fumonisins B1 or B2. This variety may be considered well adapted to higher temperatures and drier conditions, as verified in the Coruche region. Physiological and environmental cues prompt microbes to synthesize diverse carotenoids, including dihydroxy xanthophylls, facilitating their adaptation and survival. Lutein and its isomeric counterpart, zeaxanthin, are notable dihydroxy xanthophylls with bioactive properties such as antioxidative, anti-inflammatory, anticancer, and neuroprotective effects, particularly beneficial for human ocular health. However, global natural resources for co-producing lutein and zeaxanthin are scarce, with zeaxanthin lacking commercial sources, unlike lutein sourced from marigold plants and microalgae. Traditionally, dihydroxy xanthophyll production primarily relies on petrochemical synthetic routes, with limited biological sourcing reported. Nonetheless, microbiological synthesis presents promising avenues as a commercial source, albeit challenged by low dihydroxy xanthophyll yield at high cell density. Strategies involving optimization of physical and chemical parameters are essential to achieve high-quality dihydroxy xanthophyll products. Lutein is a naturally occurring carotenoid synthesized by plants and algae that has a beneficial effect on several biological processes and associated ailments. Its immediate application is in ophthalmology, where it significantly lowers the incidences of age‐related macular degeneration (AMD). It also has anti‐inflammatory action, treatment of diabetic retinopathy, and cataracts, and enhancement of visual contrast. To critically assess lutein biosynthesis, therapeutic applicability, and market research literature. We have discussed its theoretical frameworks, experimental evidence, limitations, as well as clinical trial results, and future research prospects. The literature for this review article was mined and compiled by collecting and analyzing articles from several databases, including ScienceDirect, Google Scholar, PubMed, Wiley Online Library, Patentscope, and ClinicalTrials.gov published until March 30, 2022. Patent publications were identified using the search terms like IC:(C07C67/56) AND EN_AB:(lutein) OR EN_TI:(lutein) OR EN_AB:(extraction) OR EN_TI:(process). According to the literature, lutein is an essential nutrient given that it cannot be synthesized in the human body and acts as an antioxidant, affecting AMD, diabetic retinopathy, Rheumatic diseases, inflammation, and cancer. Due to inadequate production and laborious extraction, lutein is expensive despite its high demand and applicability. Market research predicts a 6.3% compound annual growth rate for lutein by 2032. Optimizing lutein extraction for high yield and purity is necessary.
