1. Management of Personnel Professional Development as a Condition of Digital Transformation of the Organisation

Abstract

The purpose of the study is to form a methodological approach to managing the professional development of employees in order to achieve the maximum effect of the digital transformation of the organisation. The authors analysed the environment for implementing the processes of digitalisation of organisations in Russia, identified existing limitations and lagging behind developed countries. The article systematises the risks of digital transformation of the organisation. The priority of levelling personnel risks arising from the discrepancy between the level of development of the organisation's personnel and the new digital requirements, as well as due to the low adaptive capacity of the staff, is justified. A methodological approach to the creation of a management system for the professional development of the organisation's personnel is proposed. A model of its implementation is developed based on the formation of a profile of employee competencies, identification and assessment of the personnel risk of digital transformation, and realisation of a differentiated set of measures.

Abstract

The article presents the results of studying the economic and technological foundations for improving the production and operational performance of equipment related to the production of cement mass. It is shown that it is expedient to manufacture the bodies of furnaces made on the basis of a rotating principle with helical surfaces, the centers of curvature of which are located inside the body. The use of the proposed rotary screw equipment in the production of cement mass creates the conditions for increasing the speed of rotation of the kiln body by dozens of times, increasing productivity, reducing the size of kilns and reducing production costs. Abstract

In this work, it is shown that intensive treatment of dispersed systems with ultrasound is an effective method for enhancing the interaction of particles and polymer with the subsequent formation of adsorption layers of high thickness and strength. This makes possible to obtain highly stable dispersed systems with a modified surface of the solid phase. The studies proved the important role of intensive ultrasonic action for controlling the properties of the nanoparticles surface in heterophase systems. It is shown that an increase in the activity of the surface of nanoparticles as a result of ultrasonic treatment is of great importance for the study of their antibacterial activity: after ultrasonic activation, nanoparticles are more active in interaction with cell membranes.

Abstract

Positively charged polyelectrolytes hold significant potential as materials for creating antibacterial coatings. We examined the physicochemical and mechanical properties of the macromolecules in water solutions and in coatings for the series of branched polyethyleneimine (PEI) and linear polydiallyldimethylammonium chloride (PDADMAC) with different molecular weights. The microbiological study was conducted to analyze the biocidal activity of the polycation solutions and coatings towards foodborne bacteria. While the moisture saturation of the polycationic coatings and biocidal activity did not significantly depend on the chemical nature of charged groups or the molecular weight or architecture of macromolecules, the lowering of the molecular weight of polymers resulted in the loss of cohesive forces in the coatings and to a dramatic loss of stability when being washed off with water. The diffusion coefficient (D0) of macromolecules was identified as a key parameter for the wash-off mechanism. Films formed by molecules with a D0 below 1 × 10−7 cm2/s demonstrated a high resistance to wash-off procedures. We demonstrated that PEI and PDADMAC samples with high molecular weights showed high antimicrobial activity towards L. monocytogenes. Abstract

Optimization of fermentation processes requires monitoring the species composition of starter cultures and their growth during fermentation. Most starter cultures contain closely related species. Nowadays, high-resolution melting (HRM) analysis is extensively used for multiplex identification of closely related species. In the present paper, we applied real-time polymerase chain reaction (PCR) with HRM analysis for the detection and differentiation of Lactobacillus sakei and L. curvatus. A primer pair was selected for the site of the rpoA gene of Lactobacillus spp. Eleven starter cultures and fifteen fermented sausages with a known bacterial composition were successfully tested using real-time PCR with HRM analysis with the developed primer pair.

Abstract

The present work investigates the impact of spray dehydration technological parameters upon lactulose yield. It is ascertained that the maximum plant productivity is ensured at dehydrating of solutions with the lactulose mass fraction of 50% at the temperature of 140±1°С and the speed of feeding the solution into the chamber of 6 ml/min. Impact of spray dehydration technological parameters upon the end-product quality characteristics has been studied. It has been ascertained that correspondence of the end-product to the quality characteristics is gained by usage of the dehydration temperature of 140±1°С, the feeding speed of 6 ml/min, the airspeed of 20-25 m 3 /h. Lactulose received by hydrating a solution with the mass fraction of 50% meets the requirements laid on food carbohydrates as the solubility index lies within the range from 0,10±0,02 to 0,30±0,02 cm 3 of wet residue. The particles size does not exceed the standardized value of 10 µm. Microstructure of dry lactulose received at different dehydration temperatures has been studied. It is ascertained that the optimum dehydration temperature is 140°С. At the temperatures of 100-120°С dehydrated lactulose is prone to glueing of the particles. Heated over 140°С, the product has a more dispersed structure, but there are signs of caramelization.

Abstract

Inflammation is a pathophysiological response to infection or tissue damage during which high levels of reactive oxygen and nitrogen species are produced by phagocytes to kill microorganisms. Reactive oxygen and nitrogen species serve also in the complex regulation of inflammatory processes. Recently, it has been proposed that peroxiredoxins may play key roles in innate immunity and inflammation. Indeed, peroxiredoxins are evolutionarily conserved peroxidases able to reduce, with high rate constants, hydrogen peroxide, alkyl hydroperoxides and peroxynitrite which are generated during inflammation. In this minireview, we point out different possible roles of peroxiredoxins during inflammatory processes such as cytoprotective enzymes against oxidative stress, modulators of redox signaling, and extracellular pathogen- or damage-associated molecular patterns. A better understanding of peroxiredoxin functions in inflammation could lead to the discovery of new therapeutic targets.

Abstract

Peroxiredoxins (PRDXs or Prxs) are peroxidases able to reduce peroxides with a high efficiency, conferring to these enzymes functions in cytoprotection and redox signaling. PRDXs are abundant and widely expressed in most mammalian tissues. Recently, a modified pattern of PRDXs has been reported in neurodegenerative disorders such as Parkinson’s disease (PD), in term of expression level or post-translational modifications, with consequences for their activity. Emerging evidence point to the potential role of PRDXs in the pathogenesis of these neurodegenerative diseases. No systematic analysis of PRDX expression in the central nervous system (CNS) has been conducted so far. In this work, we first confirmed PRDX cell type expression in the mouse CNS by immunohistochemistry (IHC) through co-localization with specific markers such as NeuN, GFAP, Olig2 and Mac1. Several PRDXs are mainly expressed in glial cells (PRDX4 and PRDX6), but most of them are expressed in neurons (PRDX2 to PRDX5). Moreover, focusing on neuronal PRDXs, we report a complete characterization of PRDX expression levels in the most representative neuronal populations of the murine CNS with IHC procedures. Finally, tyrosine hydroxylase-PRDX double immunofluorescence showed that PRDX2 to PRDX5 are moderately to weakly expressed in the dopaminergic neurons from substantia nigra pars compacta, the neuronal population affected in PD. This faint expression could contribute to the vulnerability of these neurons in the disease. At the opposite, PRDXs are strongly expressed in motor neurons of the spinal cord which degenerate in the Amyotrophic Lateral Sclerosis (ALS). Further investigations will be needed to determine whether PRDXs play a major role in neurodegeneration.

Abstract

Food products may be a source of Salmonella, one of the main causal agents of food poisoning, especially after the emergence of strains resistant to antimicrobial preparations. The present work dealt with investigation of the occurrence of resistance to antimicrobial preparations among S. enterica strains isolated from food. The isolates belonged to 11 serovars, among which Infantis (28%), Enteritidis (19%), and Typhimurium (13.4%) predominated. The isolates were most commonly resistant to trimethoprim/sulfamethoxazole (n = 19, 59.38%), cefazolin (n = 15, 46.86%), tetracycline (n = 13, 40.63%), and amikacin (n = 9, 28.13%). Most of the strains (68.75%) exhibited multiple resistance to commonly used antibiotics. High-throughput sequencing was used to analyse three multidrug-resistant strains (resistant to six or more antibiotics). Two of them (SZL 30 and SZL 31) belonged to S. Infantis, while one strain belonged to S. Typhimurium (SZL 38). Analysis of the genomes of the sequenced strains revealed the genes responsible for antibiotic resistance. In the genomes of strains SZL 30 and SZL 31 the genes of antibiotic resistance were shown to be localized mostly in integrons within plasmids, while most of the antibiotic resistance genes of strain SZL 38 were localized in a chromosomal island (17,949 nt). Genomes of the Salmonella strains SZL 30, SZL 31, and SZL 38 were shown to contain full-size pathogenicity islands: SPI-1, SPI-2, SPI-4, SPI-5, SPI-9, SPI-11, SPI-13, SPI-14, and CS54. Moreover, the genome of strain SZL 38 was also found to contain the full-size pathogenicity islands SPI-3, SPI-6, SPI-12, and SPI-16. The emergence of multidrug-resistant strains of various Salmonella serovars indicates that further research on the transmission pathways for these genetic determinants and monitoring of the distribution of these microorganisms are necessary.
