IntroductionThis article offers an analysis of environmental communication (EC) and citizen science (CS) in the context of Environmental Monitoring and Assessment (EMA) for the implementation of Sustainable Development Goal 15 (Life on Land) in Chile and Sweden.MethodsThe paper is based on fieldwork in two rural study areas of Chile and Sweden where we followed different CS initiatives in relation to EMA and Agenda 2030. We conducted interviews, analyzed documentation and conducted an SDG mapping workshop to understand the implementation of SDG 15 in these two rural forest settings.ResultsOur findings suggest that CS has potential as a democratic innovation for environmental governance in both countries. However, we also found important barriers to the legitimacy of CS as a feature of EMA and local environmental governance in both countries. The paper situates CS in the wider governance and environmental communication processes in regional politics surrounding implementation of national policies for the use of natural resources.DiscussionThe article offers new insights into the barriers and possibilities for public participation in environmental governance and policy at local levels, by addressing the interlinkages between environmental communication and citizen science in rural settings.

Abstract Peatland vegetation takes up mercury (Hg) from the atmosphere, typically contributing to net production and export of neurotoxic methyl-Hg to downstream ecosystems. Chemical reduction processes can slow down methyl-Hg production by releasing Hg from peat back to the atmosphere. The extent of these processes remains, however, unclear. Here we present results from a comprehensive study covering concentrations and isotopic signatures of Hg in an open boreal peatland system to identify post-depositional Hg redox transformation processes. Isotope mass balances suggest photoreduction of HgII is the predominant process by which 30% of annually deposited Hg is emitted back to the atmosphere. Isotopic analyses indicate that above the water table, dark abiotic oxidation decreases peat soil gaseous Hg0 concentrations. Below the water table, supersaturation of gaseous Hg is likely created more by direct photoreduction of rainfall rather than by reduction and release of Hg from the peat soil. Identification and quantification of these light-driven and dark redox processes advance our understanding of the fate of Hg in peatlands, including the potential for mobilization and methylation of HgII.

Erosion, soil loss and consequent nutrient fluxes impair water quality and can degrade arable soils. Erosion rates in Sweden are generally low but episodic losses of suspended solids (SS) can affect water quality. Identifying critical source areas (CSAs) and “hot moments” is essential to reduce erosive losses from arable land. Here we use a distributed, dynamic high-resolution erosion model that simulates the sum of all transported material, i.e., erosion within the soil profile, on the soil surface and transport through drainage systems. We simulate monthly SS transport in six small agricultural catchments with varying soil texture over 8 years. Kling-Gupta Efficiency (KGE) was used as model performance statistics, and calibration (KGE = 0.45–0.78) and validation (KGE = 0.64–0.83) showed acceptable model performance for all catchments, with mean annual SS losses between 2.1 and 31.5 t km-2yr-1. Equifinality could be minimised by using more precise initial parameter values. We suggest that the model can be applied to comparable unmonitored catchments to identify erosion-sensitive periods and CSAs.

The fish biodiversity in the Congo River and its tributaries is extremely rich but the information on fish communities in the headwaters in terms of catch and biomass estimates is rare. Fishes in the running and stagnant waters in this region are of vital importance as a food resource for local residents. This study aimed to describe the fish community, catch, and biomass in the three headwater streams Bambou, Lebomo, and Bongo in the Lake Tumba Landscape (LTL) of the Democratic Republic of the Congo. Such information is of vital importance as a benchmark to understand the sustainability of the fish population for future generations of residents of the LTL. The field data were collected from 2007 through 2011, including dry and wet seasons. Here we present the results of this systematic, multi-annual study which was the first for fishes in streams of this region. In total, 50 species of 15 families were found in the nutrient-poor brown waters of these streams where high concentrations of humic acids cause a low pH. Among abundant species occurring in all three streams were the cyprinid Enteromius holotaenia (Boulenger, 1904), the mormyrid Marcusenius moorii (Günther, 1867), the alestids Clupeocharax schoutedeni Pellegrin, 1926 and Bryconaethiops boulengeri Pellegrin, 1900, and the clariid Clarias angolensis Steindachner, 1866. Bongo Stream was distinguished from the others by a rich abundance of Alestopetersius compressus (Poll et Gosse, 1963). The presence of several species at low pH (between 5.0 and 5.5) is new information that lowers the bottom of the pH interval for these species compared to earlier reports. The maximum total length (TL) of some other species was by 5–20 percentage points higher than those reported earlier. The median weight per unit effort (WPUE) in the streams varied between 30 and 115 g per hour during the dry seasons and between 18 and 86 g per hour during the wet seasons. The fish biomass in the streams varied between 0.05 and 0.7 g ⸱ m–2 with a median 0.14 g ⸱ m–2. This relatively low value compared to other tropical headwaters may be a result of the low pH and dark color of these headwaters. The results of the study serve as a reference point to which future monitoring of fish fauna can be compared for sustainable management of the LTL.

Boreal peatlands represent a biogeochemically unique and diverse environment in high-latitude landscape. They represent a long-term globally significant sink for carbon dioxide and a source of methane, hence playing an important role in regulating the global climate. There is an increasing interest in deciphering peatland biogeochemical processes to improve our understanding of how anthropogenic and climate change effects regulate the peatland biogeochemistry and greenhouse gas balances. At present, most studies investigating land-atmosphere exchanges of peatland ecosystems are commonly based on single-tower setups, which require the assumption of homogeneous conditions during upscaling to the landscape. However, the spatial organization of peatland complexes might feature large heterogeneity due to its varying underlying topography and vegetation composition. Little is known about how well single site studies represent the spatial variations of biogeochemical processes across entire peatland complexes. The recently established Kulbäcksliden Research Infrastructure (KRI) includes five peatland study sites located less than 3 km apart, thus providing a unique opportunity to explore the spatial variation in ecosystem-scale processes across a typical boreal peatland complex. All KRI sites are equipped with eddy covariance flux towers combined with installations for detailed monitoring of biotic and abiotic variables, as well as catchment-scale hydrology and hydrochemistry. Here, we review studies that were conducted in the Kulbäcksliden area and provide a description of the site characteristics as well as the instrumentation available at the KRI. We highlight the value of long-term infrastructures with ecosystem-scale and replicated experimental sites to advance our understanding of peatland biogeochemistry, hydrology, ecology, and its feedbacks on the environment and climate system.

Abstract Hermansky-Pudlak syndrome (HPS) is an autosomal recessive disorder characterized by improper biogenesis of lysosome-related organelles (LROs). Lung fibrosis is the leading cause of death among adults with HPS-1 and HPS-4 genetic types, which are associated with defects in the biogenesis of lysosome-related organelles complex-3 (BLOC-3), a guanine exchange factor (GEF) for a small GTPase, Rab32. LROs are not ubiquitously present in all cell types, and specific cells utilize LROs to accomplish dedicated functions. Fibroblasts are not known to contain LROs, and the function of BLOC-3 in fibroblasts is unclear. Here, we report that lung fibroblasts isolated from patients with HPS-1 have increased migration capacity. Silencing HPS-1 in normal lung fibroblasts similarly leads to increased migration. We also show that the increased migration is driven by elevated levels of Myosin IIB. Silencing HPS1 or RAB32 in normal lung fibroblasts leads to increased MYOSIN IIB levels. MYOSIN IIB is downstream of p38-MAPK, which is a known target of angiotensin receptor signaling. Treatment with losartan, an angiotensin receptor inhibitor, decreases MYOSIN IIB levels and impedes HPS lung fibroblast migration in vitro. Furthermore, pharmacologic inhibition of angiotensin receptor with losartan seemed to decrease migration of HPS lung fibroblasts in vivo in a zebrafish xenotransplantation model. Taken together, we demonstrate that BLOC-3 plays an important role in MYOSIN IIB regulation within lung fibroblasts and contributes to fibroblast migration.

Abstract Throughout their lifetime, fish maintain a high capacity for regenerating complex tissues after injury. We utilized a larval tail regeneration assay in the zebrafish Danio rerio, which serves as an ideal model of appendage regeneration due to its easy manipulation, relatively simple mixture of cell types, and superior imaging properties. Regeneration of the embryonic zebrafish tail requires development of a blastema, a mass of dedifferentiated cells capable of replacing lost tissue, a crucial step in all known examples of appendage regeneration. Using this model, we show that tail amputation triggers an obligate metabolic shift to promote glucose metabolism during early regeneration similar to the Warburg effect observed in tumor forming cells. Inhibition of glucose metabolism did not affect the overall health of the embryo but completely blocked the tail from regenerating after amputation due to the failure to form a functional blastema. We performed a time series of single-cell RNA sequencing on regenerating tails with and without inhibition of glucose metabolism. We demonstrated that metabolic reprogramming is required for sustained TGF-β signaling and blocking glucose metabolism largely mimicked inhibition of TGF-β receptors, both resulting in an aberrant blastema. Finally, we showed using genetic ablation of three possible metabolic pathways for glucose, that metabolic reprogramming is required to provide glucose specifically to the hexosamine biosynthetic pathway while neither glycolysis nor the pentose phosphate pathway were necessary for regeneration.

Abstract Climate change scenarios predict increases in temperature and organic matter supply from land to water, which affect trophic transfer of nutrients and contaminants in aquatic food webs. How essential nutrients, such as polyunsaturated fatty acids (PUFA), and potentially toxic contaminants, such as methylmercury (MeHg), at the base of aquatic food webs will be affected under climate change scenarios, remains unclear. The objective of this outdoor mesocosm study was to examine how increased water temperature and terrestrially-derived dissolved organic matter supply (tDOM; i.e., lake browning), and the interaction of both, will influence MeHg and PUFA in organisms at the base of food webs (i.e. seston; the most edible plankton size for zooplankton) in subalpine lake ecosystems. We used the distribution of stream-dwelling brown trout (Salmo trutta) in a 67 km2 boreal catchment to explore the importance of environmental organizing factors at a range of spatial scales, including whole-catchment characteristics derived from map data, and stream reach chemical and physical characteristics. Brown trout were not observed at any sites characterized by pH < 5.0 during the spring snowmelt episode, matching published toxicity thresholds. Brown trout distributions were patchy even in less acidic regions of the stream network, positively associated with glaciofluvial substrate and negatively associated with fine sand/silty sediments. A multivariate model including only whole-catchment characteristics explained 43% of the variation in brown trout densities, while models with local site physical habitat characteristics or local stream chemistry explained 33 and 25%, respectively. At the stream reach scale, physical habitat apparently played a primary role in organizing brown trout distributions in this stream network, with acidity placing an additional restriction by excluding brown trout from acidic headwater streams. Much of the strength of the catchment characteristics-fish association could be explained by the correlation of catchment-scale landscape characteristics with local stream chemistry and site physical characteristics. These results, consistent with the concept of multiple hierarchical environmental filters regulating the distribution of this fish species, underline the importance of considering a range of spatial scales and both physical and chemical environments when attempting to manage or restore streams for brown trout.

ZRSR2 (zinc finger CCCH-type, RNA binding motif and serine/arginine rich 2) is an essential splicing factor involved in 3′ splice-site recognition as a component of both the major and minor spliceosomes that mediate the splicing of U2-type (major) and U12-type (minor) introns, respectively. Studies of ZRSR2-depleted cell lines and ZRSR2-mutated patient samples revealed its essential role in the U12-dependent minor spliceosome. However, the role of ZRSR2 during embryonic development is not clear, as its function is compensated for by Zrsr1 in mice. Here, we utilized the zebrafish model to investigate the role of zrsr2 during embryonic development. Using CRISPR/Cas9 technology, we generated a zrsr2-knockout zebrafish line, termed zrsr2hg129/hg129 (p.Trp167Argfs*9) and examined embryo development in the homozygous mutant embryos. zrsr2hg129/hg129 embryos displayed multiple developmental defects starting at 4 days post fertilization (dpf) and died after 8 dpf, suggesting that proper Zrsr2 function is required during embryonic development. The global transcriptome analysis of 3 dpf zrsr2hg129/hg129 embryos revealed that the loss of Zrsr2 results in the downregulation of essential metabolic pathways and the aberrant retention of minor introns in about one-third of all minor intron-containing genes in zebrafish. Overall, our study has demonstrated that the role of Zrsr2 as a component of the minor spliceosome is conserved and critical for proper embryonic development in zebrafish.

The use of zebrafish in functional genomics and disease modeling has become popular due to the ease of targeted mutagenesis with genome editing nucleases, i.e., zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats/Cas9 (CRISPR/Cas9). These nucleases, specifically CRISPR/Cas9, are routinely used to generate gene knockout mutants by causing a double stranded break at the desired site in the target gene and selecting for frameshift insertions or deletions (indels) caused by the errors during the repair process. Thus, a variety of methods have been developed to identify fish with indels during the process of mutant generation and phenotypic analysis. These methods range from PCR and gel-based low-throughput methods to high-throughput methods requiring specific reagents and/or equipment. Here, we provide a comprehensive review of currently used indel detection methods in zebrafish. By discussing the molecular basis for each method as well as their pros and cons, we hope that this review will serve as a comprehensive resource for zebrafish researchers, allowing them to choose the most appropriate method depending upon their budget, access to required equipment and the throughput needs of the projects.

Mutations in the gene AK2 are responsible for reticular dysgenesis (RD), a rare and severe form of primary immunodeficiency in children. RD patients have a severely shortened life expectancy and without treatment die, generally from sepsis soon after birth. The only available therapeutic option for RD is hematopoietic stem cell transplantation (HSCT). To gain insight into the pathophysiology of RD, we previously created zebrafish models for Ak2 deficiencies. One of the clinical features of RD is hearing loss, but its pathophysiology and causes have not been determined. In adult mammals, sensory hair cells of the inner ear do not regenerate; however, their regeneration has been observed in several non-mammalian vertebrates, including zebrafish. Therefore, we used our RD zebrafish models to determine whether Ak2 deficiency affects sensory organ development and/or hair cell regeneration. Our studies indicated that Ak2 is required for the correct development, survival and regeneration of sensory hair cells. Interestingly, Ak2 deficiency induces the expression of several oxidative stress markers and it triggers an increased level of cell death in the hair cells. Finally, we show that glutathione treatment can partially rescue hair cell development in the sensory organs in our RD models, pointing to the potential use of antioxidants as a therapeutic treatment supplementing HSCT to prevent or ameliorate sensorineural hearing deficits in RD patients.

Abstract Low‐order streams are suggested to dominate the atmospheric CO2 source of all inland waters. Yet, many large‐scale stream estimates suffer from methods not designed for gas emission determination and rarely include other greenhouse gases such as CH4. Here, we present a compilation of directly measured CO2 and CH4 concentration data from Swedish low‐order streams (> 1600 observations across > 500 streams) covering large climatological and land‐use gradients. These data were combined with an empirically derived gas transfer model and the characteristics of a ca. 400,000 km stream network covering the entire country. The total stream CO2 and CH4 emission corresponded to 2.7 Tg C yr−1 (95% confidence interval: 2.0–3.7) of which the CH4 accounted for 0.7% (0.02 Tg C yr−1). The study highlights the importance of low‐order streams, as well as the critical need to better represent variability in emissions and stream areal extent to constrain future stream C emission estimates.

Abstract It is well established that stream dissolved inorganic carbon (DIC) fluxes play a central role in the global C cycle, yet the sources of stream DIC remain to a large extent unresolved. Here, we explore large-scale patterns in δ13C-DIC from streams across Sweden to separate and further quantify the sources and sinks of stream DIC. We found that stream DIC is governed by a variety of sources and sinks including biogenic and geogenic sources, CO2 evasion, as well as in-stream processes. Although soil respiration was the main source of DIC across all streams, a geogenic DIC influence was identified in the northernmost region. All streams were affected by various degrees of atmospheric CO2 evasion, but residual variance in δ13C-DIC also indicated a significant influence of in-stream metabolism and anaerobic processes. Due to those multiple sources and sinks, we emphasize that simply quantifying aquatic DIC fluxes will not be sufficient to characterise their role in the global C cycle.

Forests and trees are key to solving water availability problems in the face of climate change and to achieving the United Nations Sustainable Development Goals. A recent global assessment of forest and water science posed the question: How do forests matter for water? Here we synthesize science from that assessment, which shows that forests and water are an integrated system. We assert that forests, from the tops of their canopies to the base of the soils in which trees are rooted, must be considered a key component in the complex temporal and spatial dimensions of the hydrologic cycle. While it is clear that forests influence both downstream and downwind water availability, their actual impact depends on where they are located and their processes affected by natural and anthropogenic conditions. A holistic approach is needed to manage the connections between forests, water and people in the face of current governance systems that often ignore these connections. We need policy interventions that will lead to forestation strategies that decrease the dangerous rate of loss in forest cover and that—where appropriate—increase the gain in forest cover. We need collective interventions that will integrate transboundary forest and water management to ensure sustainability of water supplies at local, national and continental scales. The United Nations should continue to show leadership by providing forums in which interventions can be discussed, negotiated and monitored, and national governments must collaborate to sustainably manage forests to ensure secure water supplies and equitable and sustainable outcomes.

Fanconi Anemia (FA) is a genomic instability syndrome resulting in aplastic anemia, developmental abnormalities, and predisposition to hematological and other solid organ malignancies. Mutations in genes that encode proteins of the FA pathway fail to orchestrate the repair of DNA damage caused by DNA interstrand crosslinks. Zebrafish harbor homologs for nearly all known FA genes. We used multiplexed CRISPR/Cas9-mediated mutagenesis to generate loss-of-function mutants for 17 FA genes: fanca, fancb, fancc, fancd1/brca2, fancd2, fance, fancf, fancg, fanci, fancj/brip1, fancl, fancm, fancn/palb2, fanco/rad51c, fancp/slx4, fancq/ercc4, fanct/ube2t, and two genes encoding FA-associated proteins: faap100 and faap24. We selected two indel mutations predicted to cause premature truncations for all but two of the genes, and a total of 36 mutant lines were generated for 19 genes. Generating two independent mutant lines for each gene was important to validate their phenotypic consequences. RT-PCR from homozygous mutant fish confirmed the presence of transcripts with indels in all genes. Interestingly, 4 of the indel mutations led to aberrant splicing, which may produce a different protein than predicted from the genomic sequence. Analysis of RNA is thus critical in proper evaluation of the consequences of the mutations introduced in zebrafish genome. We used fluorescent reporter assay, and western blots to confirm loss-of-function for several mutants. Additionally, we developed a DEB treatment assay by evaluating morphological changes in embryos and confirmed that homozygous mutants from all the FA genes that could be tested (11/17), displayed hypersensitivity and thus were indeed null alleles. Our multiplexing strategy helped us to evaluate 11 multiple gene knockout combinations without additional breeding. Homozygous zebrafish for all 19 single and 11 multi-gene knockouts were adult viable, indicating FA genes in zebrafish are generally not essential for early development. None of the mutant fish displayed gross developmental abnormalities except for fancp-/- fish, which were significantly smaller in length than their wildtype clutch mates. Complete female-to-male sex reversal was observed in knockouts for 12/17 FA genes, while partial sex reversal was seen for the other five gene knockouts. All adult females were fertile, and among the adult males, all were fertile except for the fancd1 mutants and one of the fancj mutants. We report here generation and characterization of zebrafish knockout mutants for 17 FA disease-causing genes, providing an integral resource for understanding the pathophysiology associated with the disrupted FA pathway.

Few environmental issues have resulted in such a heated policy-science controversy in Sweden as the 1990s acidification debate in the north of the country. The belief that exceptionally high stream acidity levels during hydrological events was caused by anthropogenic deposition resulted in a governmentally funded, multi-million dollar surface-water liming program. This program was heavily criticized by a large part of the scientific community arguing that the acidity of northern streams was primarily caused by naturally occurring organic acids. Here, we revisit the acid deposition legacy in northern Sweden two decades after the culmination of the controversy by examining the long-term water chemistry trends in the Svartberget/Krycklan research catchment that became a nexus for the Swedish debate. In this reference stream, trends in acidic episodes do show a modest recovery that matches declines in acid deposition to pre-industrial levels, although stream acidity continues to be overwhelmingly driven by organic acidity. Yet there are legacies of acid deposition related to calcium losses from soils, which are more pronounced than anticipated. Finally, assessment of these trends are becoming increasingly complicated by new changes and threats to water resources that must be recognized to avoid unnecessary, expensive, and potentially counterproductive measures to adapt and mitigate human influences. Here we make the argument that while the acidification era is ending, climate change, land-use transitions, and long-range transport of other contaminants warrant close monitoring in the decades to come.

Because of genetic heterogeneity present in idiopathic scoliosis, we previously defined clinical subsets (a priori) from a sample of families with idiopathic scoliosis to find genes involved with spinal curvature. Previous genome-wide linkage analysis of seven families with at least two individuals with kyphoscoliosis found linkage (P-value = 0.002) in a 3.5-Mb region on 5p13.3 containing only three known genes, IRX1, IRX2, and IRX4. In this study, the exons of IRX1, IRX2, and IRX4, the conserved noncoding elements in the region, and the exons of a nonprotein coding RNA, LOC285577, were sequenced. No functional sequence variants were identified. An intrafamilial test of association found several associated noncoding single nucleotide variants. The strongest association was with rs12517904 (P = 0.00004), located 6.5 kb downstream from IRX1. In one family, the genotypes of nine variants differed from the reference allele in all individuals with kyphoscoliosis, and two of three individuals with scoliosis, but did not differ from the reference allele in all other genotyped individuals. One of these variants, rs117273909, was located in a conserved noncoding region that functions as an enhancer in mice. To test whether the variant allele at rs117273909 had an effect on enhancer activity, zebrafish transgenesis was performed with overlapping fragments of 198 and 687 bp containing either the wild type or the variant allele. Our data suggests that this region acts as a regulatory element; however, its size and target gene(s) need to be identified to determine its role in idiopathic scoliosis.

The Earth has entered the Anthropocene epoch and forest managers are facing unprecedented challenges to meet multiple ecosystem service demands from forests. Understanding the complex forest-water relations under a changing environment must add a human dimension, and this is essential in the move towards sustainable forest management in the 21st century. This Special Issue contains 10 papers presented at a joint international forest and water conference in Chile in 2018. These studies provide global examples on new advancements in sciences in forest ecohydrology, watershed management, and ecosystem service assessment under various geographical and socioeconomic settings.

ATP6V1H is a component of a large protein complex with vacuolar ATPase (V-ATPase) activity. We identified two generations of individuals in which short stature and osteoporosis co-segregated with a mutation in ATP6V1H. Since V-ATPases are highly conserved between human and zebrafish, we generated loss-of-function mutants in atp6v1h in zebrafish through CRISPR/Cas9-mediated gene knockout. Homozygous mutant atp6v1h zebrafish exhibited a severe reduction in the number of mature calcified bone cells and a dramatic increase in the expression of mmp9 and mmp13. Heterozygous adults showed curved vertebra that lack calcified centrum structure and reduced bone mass and density. Treatment of mutant embryos with small molecule inhibitors of MMP9 and MMP13 significantly restored bone mass in the atp6v1h mutants. These studies have uncovered a new, ATP6V1H-mediated pathway that regulates bone formation, and defines a new mechanism of disease that leads to bone loss. We propose that MMP9/MMP13 could be therapeutic targets for patients with this rare genetic disease.

The Water Framework Directive (WFD) is an ambitious piece of legislation designed to protect and improve water quality throughout Europe. However, forests are only mentioned once in the WFD, and forestry is not mentioned at all, despite its potential implications for streams, rivers and lakes. Here we present a transdisciplinary commentary on the WFD and its implications for forests and forestry in Sweden. This commentary has been prepared by forestry stakeholders, biophysical and social scientists. While we were cognizant of a large body of discipline-specific research, there are very few inter- or trans-disciplinary commentaries which link academic and stakeholder perspectives on the WFD. We had originally felt that there would be little commonality in our concerns. However, we found significant areas of agreement. Our key areas of concern about the implications of the WFD for forestry in Sweden included: (i) concerns about what is meant by good ecological status and how it is assessed; (ii) a perceived lack of clarity in the legal framework; (iii) an inadequate environmental impact assessment process; and (iv) uncertainties about appropriate programs of measures for improving water quality. We were also concerned that ecosystem services provided by forests and the positive effects of forestry on water quality are inadequately recognized in the WFD.

Additions of nutrients, faster growing tree varieties, more intense harvest practices, and a changing climate all have the potential to increase forest production in Sweden, thereby mitigating climate change through carbon sequestration and fossil fuel substitution. However, the effects of management strategies for increased biomass production on soil resources and water quality at landscape scales are inadequately understood. Key knowledge gaps also remain regarding the sustainability of shorter rotation periods and more intensive biomass harvests. This includes effects of fertilization on the long-term weathering and supply of base cations and the consequences of changing mineral availability for future forest production. Furthermore, because soils and surface waters are closely connected, management efforts in the terrestrial landscape will potentially have consequences for water quality and the ecology of streams, rivers, and lakes. Here, we review and discuss some of the most pertinent questions related to how increased forest biomass production in Sweden could affect soils and surface waters, and how contemporary forestry goals can be met while minimizing the loss of other ecosystem services. We suggest that the development of management plans to promote the sustainable use of soil resources and water quality, while maximizing biomass production, will require a holistic ecosystem approach that is placed within a broader landscape perspective.

During the last 100 years the Ethiopian upper Blue Nile Basin (BNB) has undergone major changes in land use, and is now potentially facing changes in climate. Rainfall over BNB supplies over two-thirds of the water to the Nile and supports a large local population living mainly on subsistence agriculture. Regional food security is sensitive to both the amount and timing of rain and is already an important political challenge that will be further complicated if scenarios of climate change are realized. In this study a simple spatial model of the timing and duration of summer rains (Kiremt) and dry season (Bega), and annual rain over the upper BNB was established from observed data between 1952 and 2004. The model was used to explore potential impacts of climate change on these rains, using a down-scaled ECHAM5/MP1-OM scenario between 2050 and 2100. Over the observed period the amount, onset and duration of Kiremt rains and rain-free Bega days have exhibited a consistent spatial pattern. The spatially averaged annual rainfall was 1490 mm of which 93% was Kiremt rain. The average Kiremt rain and number of rainy days was higher in the southwest (322 days) and decreased towards the north (136 days). Under the 2050-2100 scenario, the annual mean rainfall is predicted to increase by 6% and maintain the same spatial pattern as in the past. A larger change in annual rainfall is expected in the southwest (ca. +130 mm) with a gradually smaller change towards the north (ca. +70 mm). Results highlight the need to account for the characteristic spatiotemporal zonation when planning water management and climate adaptation within the upper BNB. The presented simple spatial resolved models of the presence of Kiremt and annual total rainfall could be used as a baseline for such long-term planning.

Recently, it has been shown that targeted mutagenesis using zinc-finger nucleases (ZFNs) and transcription activator-like effector nucleases (TALENs) can be used to generate knockout zebrafish lines for analysis of their function and/or developing disease models. A number of different methods have been developed for the design and assembly of gene-specific ZFNs and TALENs, making them easily available to most zebrafish researchers. Regardless of the choice of targeting nuclease, the process of generating mutant fish is similar. It is a time-consuming and multi-step process that can benefit significantly from development of efficient high throughput methods. In this study, we used ZFNs assembled through either the CompoZr (Sigma-Aldrich) or the CoDA (context-dependent assembly) platforms to generate mutant zebrafish for nine genes. We report our improved high throughput methods for 1) evaluation of ZFNs activity by somatic lesion analysis using colony PCR, eliminating the need for plasmid DNA extractions from a large number of clones, and 2) a sensitive founder screening strategy using fluorescent PCR with PIG-tailed primers that eliminates the stutter bands and accurately identifies even single nucleotide insertions and deletions. Using these protocols, we have generated multiple mutant alleles for seven genes, five of which were targeted with CompoZr ZFNs and two with CoDA ZFNs. Our data also revealed that at least five-fold higher mRNA dose was required to achieve mutagenesis with CoDA ZFNs than with CompoZr ZFNs, and their somatic lesion frequency was lower (

The study presents a hydrology concept developed to include lateral water flow in the biogeochemical model ForSAFE. The hydrology concept was evaluated against data collected at Svartberget in the Vindeln Research Forest in Northern Sweden. The results show that the new concept allows simulation of a saturated and an unsaturated zone in the soil as well as water flow that reaches the stream comparable to measurements. The most relevant differences compared to streamflow measurements are that the model simulates a higher base flow in winter and lower flow peaks after snowmelt. These differences are mainly caused by the assumptions made to regulate the percolation at the bottom of the simulated soil columns. The capability for simulating lateral flows and a saturated zone in ForSAFE can greatly improve the simulation of chemical exchange in the soil and export of elements from the soil to watercourses. Such a model can help improve the understanding of how environmental changes in the forest landscape will influence chemical loads to surface waters.

As the permafrost region warms, its large organic carbon pool will be increasingly vulnerable to decomposition, combustion, and hydrologic export. Models predict that some portion of this release will be offset by increased production of Arctic and boreal biomass; however, the lack of robust estimates of net carbon balance increases the risk of further overshooting international emissions targets. Precise empirical or model-based assessments of the critical factors driving carbon balance are unlikely in the near future, so to address this gap, we present estimates from 98 permafrost-region experts of the response of biomass, wildfire, and hydrologic carbon flux to climate change. Results suggest that contrary to model projections, total permafrost-region biomass could decrease due to water stress and disturbance, factors that are not adequately incorporated in current models. Assessments indicate that end-of-the-century organic carbon release from Arctic rivers and collapsing coastlines could increase by 75% while carbon loss via burning could increase four-fold. Experts identified water balance, shifts in vegetation community, and permafrost degradation as the key sources of uncertainty in predicting future system response. In combination with previous findings, results suggest the permafrost region will become a carbon source to the atmosphere by 2100 regardless of warming scenario but that 65%–85% of permafrost carbon release can still be avoided if human emissions are actively reduced.

Bardet-Biedl Syndrome (BBS, MIM#209900) is a genetically heterogeneous disorder with pleiotropic phenotypes that include retinopathy, mental retardation, obesity and renal abnormalities. Of the 15 genes identified so far, seven encode core proteins that form a stable complex called BBSome, which is implicated in trafficking of proteins to cilia. Though BBS9 (also known as PTHB1) is reportedly a component of BBSome, its direct function has not yet been elucidated. Using zebrafish as a model, we show that knockdown of bbs9 with specific antisense morpholinos leads to developmental abnormalities in retina and brain including hydrocephaly that are consistent with the core phenotypes observed in syndromic ciliopathies. Knockdown of bbs9 also causes reduced number and length of cilia in Kupffer's vesicle. We also demonstrate that an orthologous human BBS9 mRNA, but not one carrying a missense mutation identified in BBS patients, can rescue the bbs9 morphant phenotype. Consistent with these findings, knockdown of Bbs9 in mouse IMCD3 cells results in the absence of cilia. Our studies suggest a key conserved role of BBS9 in biogenesis and/or function of cilia in zebrafish and mammals.

In this paper we investigate the hypothesis that long-term sulphate (SO(4) (2-)) deposition has made peatlands a larger source of methyl mercury (MeHg) to remote boreal lakes. This was done on experimental plots at a boreal, low sedge mire where the effect of long-term addition of SO(4) (2-) on peat pore water MeHg concentrations was observed weekly throughout the snow-free portion of 1999. The additions of SO(4) (2-) started in 1995. The seasonal mean of the pore water MeHg concentrations on the plots with 17 kg ha(-1) yr(-1) of sulphur (S) addition (1.3±0.08 ng L(-1), SE; n = 44) was significantly (p2 ng L(-1) compared to +/-0.5 ng L(-1) in the ambient S deposition plots. The concentrations of pore water MeHg in the S-addition plots were positively correlated (r(2) = 0.21; p = 0.001) to the groundwater level, with the lowest concentrations of MeHg during the period with the lowest groundwater levels. The pore water MeHg concentrations were not correlated to total Hg, DOC concentration or pH. The results from this study indicate that the persistently higher pore water concentrations of MeHg in the S-addition plots are caused by the long-term additions of SO(4) (2-) to the mire surface. Since these waters are an important source of runoff, the results support the hypothesis that SO(4) (2-) deposition has increased the contribution of peatlands to MeHg in downstream aquatic systems. This would mean that the increased deposition of SO(4) (2-) in acid rain has contributed to the modern increase in the MeHg burdens of remote lakes hydrologically connected to peatlands.
