This study used the bark of the forest species Campomanesia guazumifolia modified with H 2 SO 4 to absorb the anti-inflammatory ketoprofen from aqueous solutions. FTIR spectra confirmed that the main bands remained after the chemical treatment, with the appearance of two new bands related to the elongation of the carbonyl group present in hemicellulose. Micrographs confirmed that the surface started to contain a new textural shape after acid activation, having new pores and cavities. The drug adsorption's optimum conditions were obtained by response surface methodology (RSM). The adsorption was favored at acidic pH (2). The dosage of 1 g L -1 was considered ideal, obtaining good indications of removal and capacity. The Elovich model very well represented the kinetic curves. The isotherm studies indicated that the increase in temperature negatively affected the adsorption of ketoprofen. A maximum adsorption capacity of 158.3 mg g -1 was obtained at the lower temperature of 298 K. Langmuir was the best-fit isotherm. Thermodynamic parameters confirmed the exothermic nature of the system (ΔH 0 = -8.78 kJ mol -1 ). In treating a simulated effluent containing different drugs and salts, the removal values were 35, 50, and 80% at 15, 30, and 180 min, respectively. Therefore, the development of adsorbent from the bark of Campomanesia guazumifolia treated with H 2 SO 4 represents a remarkable alternative for use in effluent treatment containing ketoprofen., (© 2021. Magnetic carbon nanotubes (CNTs) with encapsulated Co nanoparticles (Co@CNTs), was synthesized by exploiting the one-step pyrolysis strategy using ZIF-67 as template. The as-synthesized Co@CNTs is provided with the nanopores, a large specific surface area, and strong magnetic response. The obtained Co@CNTs was used as magnetic solid-phase extraction adsorbents to extract two profens including flurbiprofen and ketoprofen. The parameters of extraction efficiency, involving extraction time, sample solution volume, ionic strength, pH and the conditions of desorption efficiency, were optimized in detail. After determined by high-performance liquid chromatography-ultraviolet (HPLC-UV), the results evinced that Co@CNTs showed a high extraction efficiency with high enrichment factors of 832 and 672. The good linear range of both flurbiprofen and ketoprofen were all 5.0-1000 ng L -1 , with the limit of detection were 0.60 ng L -1 and 0.70 ng L -1 , respectively. Furthermore, a valid method for the extraction of flurbiprofen and ketoprofen from human serum was established. The spiking recoveries of two profens were between 86.74% and 97.22%, and the relative standard deviation was less than 6.55%. Co@CNTs can be repeatedly used at least 10 times, indicating its excellent regeneration and reusability. A (glycidyl methacrylate)-co-(ethylene glycol dimethacrylate) polymer (poly(GMA-co-EDMA)) was functionalized with metal-organic frameworks (MOF) and used as a sorbent for solid-phase extraction (SPE). The polymeric sorbent was prepared in-situ by photopolymerization in a previously wall-modified spin column, and then modified with an amino-modified MOF of type NH 2 -MIL-101(Cr). The sorbents were used for the extraction of nonsteroidal anti-inflammatory drugs (NSAIDs) from human urine samples. The sorbent was compared with the parent monolith and embedded approach, where the MOF particles are admixed in the polymerization mixture before the in-situ polymerization in the modified spin column. SPE is performed by percolating the sample solutions in a centrifuge, which streamlines the SPE steps. The hybrid composites were characterized by scanning electron microscopy and nitrogen intrusion porosimetry. Three NSAIDs (ketoprofen, flurbiprofen, and ibuprofen) were tested. They were eluted from the sorbent with acidified water-acetonitrile mixtures and subsequently analyzed by reversed-phase HPLC with UV detection. The detection limits varied in the range from 0.1 to 7 μg·L -1 , and the precisions (relative standard deviation) were <14% in all the cases. The recoveries were between 71.0 and 78.0% in spiked urine samples. A chitosan membrane composed by 60% (w/w) chitosan and 40% (w/w) Aliquat®336 has been proposed as a new biopolymeric support for electromembrane extraction. The new support has been characterized by Scanning Electron Microscopy, resulting a 30-35 µm thickness. Amoxicillin, nicotinic acid, hippuric acid, salicylic acid, anthranilic acid, ketoprofen, naproxen and ibuprofen have been successfully extracted using the proposed support. Better enrichment factors were obtained for the acidic polar analytes than for the non-steroidal anti-inflammatory compounds (ranging from 118 for hippuric acid and 20 for ibuprofen). Electromembrane extraction was developed applying a DC voltage of 100 V, 1-octanol as supported liquid membrane and 20 min of extraction. The target analytes have also been satisfactorily extracted from human urine samples, providing high extraction efficiencies. The use of polymer inclusion membranes (PIMs) as support of 1-octanol liquid membrane in electromembrane extraction (EME) procedure is proposed. Synthesis of PIMs were optimized to a composition of 29% (w/w) of cellulose triacetate as base polymer and 71% (w/w) of Aliquat®336 as cationic carrier. Flat PIMs of 25µm thickness and 6mm diameter were used. EME protocol was implemented for the simultaneous extraction of four non-steroidal anti-inflammatory drugs (NSAIDs) (salicylic acid, ketoprofen, naproxen and ibuprofen) and four highly polar acidic drugs (anthranilic acid, nicotinic acid, amoxicillin and hippuric acid). Posterior HPLC separation of the extracted analytes was developed with diode array detection. Recoveries in the 81-34% range were obtained. Gradient operation of a simulated moving bed (SMB) can improve the separation and purification performance by adjusting adsorption strength in each individual zone according to its functional role. The feasibility of an internal temperature gradient (ITG) established by a difference between feed and desorbent temperatures for binary separation of ketoprofen enantiomers was investigated based on simultaneous optimization of purity and productivity of S-ketoprofen, the preferentially adsorbed species and desired product. ITG operation with a temperature difference of 20K has a unit productivity higher than isothermal mode by about 20%. Due to the combined effects of temperature transition and downstream dilution, concentration profile may exhibit a remarkable peak and a pattern of two-step drop in the temperature descending and ascending areas, respectively. Both areas, if properly located under optimal conditions, are favorable for unit productivity, which cannot be predicted by the direct use of triangle theory and average Henry's constants. The present work represents a simple and effective preparation of a novel mixed-mode anion-exchange (MAX) sorbent based on porous poly[2-(diethylamino)ethyl methacrylate-divinylbenzene] (poly(DEAEMA-DVB)) spherical particles synthesized by one-step Pickering emulsion polymerization. The poly(DEAEMA-DVB) particles were quaternized with 1,4-butanediol diglycidyl ether (BDDE) followed by triethylamine (TEA) via epoxy-amine reaction to offer strong anion exchange properties. The synthesized MAX sorbent was characterized by scanning electron microscopy, Fourier-transform infrared spectroscopy, nitrogen adsorption-desorption measurements and elemental analysis. The MAX sorbent possessed regular spherical shape and narrow diameter distribution (15-35μm), a high IEC of 0.54meq/g, with carbon and nitrogen contents of 80.3% and 1.62%, respectively. Compared to poly(DEAEMA-DVB), the MAX sorbent exhibited decreased S BET (390.5 vs. 515.3m 2 g -1 ), pore volume (0.74 vs. 0.85cm 3 g -1 ) and pore size (16.8 vs. 17.3nm). Moreover, changes of N content for producing the MAX sorbent reveal a successful two-step quaternization, which can be highly related to such a high IEC. Finally, the MAX sorbent was successfully evaluated for selective isolation and purification of some selected acidic pharmaceuticals (ketoprofen, KEP; naproxen, NAP; and ibuprofen, IBP) from neutral (hydrocortisone, HYC), basic (carbamazepine, CAZ; amitriptyline, AMT) pharmaceuticals and other interferences in water samples using solid phase extraction (SPE). An efficient analytical method based on the MAX-based mixed-mode SPE coupled with HPLC-UV was developed for highly selective extraction and cleanup of acidic KEP, NAP and IBP in spiked wastewater samples. A new class of efficient stationary phase has been investigated for use in the liquid chromatographic separation of low molecular weight analytes and high molecular weight biomolecules, based on the application of immobilised stimuli-responsive polymers (SRPs). To this end, two polymeric units, namely poly(2-dimethylaminoethyl methacrylate) (PDMAEMA) and poly(acrylic acid) (PAA) were tethered to a triazine core. The derived poly(2-dimethyl-aminoethyl methacrylate)-block-poly(acrylic acid) (PDMAEMA-b-PAA), as a diblock co-polymer, was then immobilised onto the surface of porous silica particles. The performance of this microparticulate adsorbent was evaluated under various temperature, ionic strength and/or pH conditions in packed columns in a high-performance liquid chromatography (HPLC) format. Baseline separations of a variety of low molecular weight analytes were achieved at different temperatures with this SRP-based adsorbent using 10 mM sodium phosphate buffer, pH 6.0, as the mobile phase. Moreover, when the ionic strength of the mobile phase was increased to 40 mM sodium phosphate buffer, pH 6.0, similar temperature changes resulted in further increases in resolution for the hydrophobic analytes. In addition, changes in the pH of the mobile phase from pH 6.0 to pH 8.0 led to significant changes in selectivity of the analytes, including reversal in their elution orders. Upon increasing the temperature, the retention times of all analytes decreased but without loss of resolution. These findings can be attributed to the consequence of the immobilised copolymer undergoing a phase transition at its lower critical solution temperature (LCST), which leads to changes in its solvated structure, including how the electrostatic, hydrophilic and hydrophobic regions/domains of the copolymer are exposed to the bulk mobile phase. Thermodynamic data were indicative of a temperature-related re-organisation of the structure of the immobilised PDMAEMA-b-PAA stationary phase with exothermic binding of the analytes occurring at temperatures below the lower critical solution temperature (LCST). In this manner; changes in the system temperature could directly be used to manipulate the adsorption and desorption behaviour of these analytes with this stimuli-responsive, polymer-modified porous silica stationary phase. Additional studies with several proteins further documented the versatility of these stimuli-responsive separation materials. A two-dimensional achiral-chiral LC-LC method in heart-cut mode for ketoprofen and its enantiomeric fraction determination was proposed. A C 8 column was used in the first dimension, and the chiral column was an α 1 -acid glycoprotein. The mobile phase of the chiral system was optimized by a factorial design. The effect of temperature on retention and on enantiomeric resolution was studied. Particular attention was paid to mobile phase compatibility for the two columns and to transferring time, using ketoprofen standards. The R-(-) and S-(+)-ketoprofen retention times were 9 and 11 min, respectively; the resolution was higher than 1.1 and enantiomeric fraction close to 0.5. The method was applied to capsules and gels containing ketoprofen. Factorial design was also used to establish the best conditions for gel sample preparation. This study describes a method to determine nonsteroidal anti-inflammatory drugs (NSAIDs) in urine samples based on the use of single-drop microextraction (SDME) in a three-phase design as a preconcentration technique coupled in-line to capillary electrophoresis. Different parameters affecting the extraction efficiency of the SDME process were evaluated (e.g. type of extractant, volume of the microdroplet, and extraction time). The developed method was successfully applied to the analysis of human urine samples with LODs ranging between 1.0 and 2.5 μg/mL for all of the NSAIDs under study. This method shows RSD values ranging from 8.5 to 15.3% in interday analysis. The enrichment factors were calculated, resulting 27-fold for ketoprofen, 14-fold for diclofenac, 12-fold for ibuprofen, and 44-fold naproxen. Samples were analyzed applying the SDME-CE method and the obtained results presented satisfactory recovery values (82-115%). Polymer monolith microextraction (PMME) based on capillary monolithic column is an effective and useful technique to preconcentrate trace analytes from environmental and biological samples. Here, we report the fabrication of a novel aluminum terephthalate metal-organic framework (MIL-53(Al)) incorporated capillary monolithic column via in situ polymerization for the PMME of non-steroidal anti-inflammatory drugs (NSAIDs) (ketoprofen, fenbufen and ibuprofen) in water and urine samples. The fabricated MIL-53(Al) incorporated monolith was characterized by X-ray powder diffractometry, scanning electron microscopy, Fourier transform infrared spectrometry, and nitrogen adsorption experiment. The MIL-53(Al) incorporated monolith gave larger surface area than the neat polymer monolith. A 2-cm long MIL-53(Al) incorporated capillary monolith was applied for PMME coupled with high-performance liquid chromatography for the determination of the NSAIDs. Potential factors affecting the PMME were studied in detail. Under the optimized conditions, the developed method gave the enhancement factors of 46-51, the linear range of 0.40-200μgL(-1), the detection limits (S/N=3) of 0.12-0.24μgL(-1), and the quantification limits (S/N=10) of 0.40-0.85μgL(-1). The recoveries for spiked NSAIDs (20μgL(-1)) in water and urine samples were in the range of 77.3-104%. Besides, the MIL-53(Al) incorporated monolith was stable enough for 120 extraction cycles without significant loss of extraction efficiency. The new sample preparation concept "Parallel artificial liquid membrane extraction (PALME)" was evaluated for extraction of the acidic drugs ketoprofen, fenoprofen, diclofenac, flurbiprofen, ibuprofen, and gemfibrozil from human plasma samples. Plasma samples (250 μL) were loaded into individual wells in a 96-well donor plate and diluted with HCl to protonate the acidic drugs. The acidic drugs were extracted as protonated species from the individual plasma samples, through corresponding artificial liquid membranes each comprising 2 μL of dihexyl ether, and into corresponding acceptor solutions each comprising 50 μL of 25 mM ammonia solution (pH 10). The liquid membranes and the acceptor solutions were located in a 96-well filter plate, which was sandwiched with the 96-well donor plate during extraction. Parallel extraction of several samples was performed for 15 to 60 min, followed by high-performance liquid chromatography-ultraviolet detection of the individual acceptor solutions. Important PALME parameters including the chemical composition of the liquid membrane, extraction time, and sample pH were optimized, and the extraction performance was evaluated. Except for flurbiprofen, exhaustive extraction was accomplished from plasma. Linearity was obtained for all six drugs in the range 0.025-10 μg/mL, with r (2) values ranging between 0.998 and 1.000. Precision data were in the range 3-22% RSD, and accuracy data were within 72-130% with spiked plasma samples. Based on the current experiences, PALME showed substantial potential for future high-throughput bioanalysis of non-polar acidic drugs.

This research advances the knowledge of ion-exchange of four non-steroidal anti-inflammatory drugs (NSAIDs) - diclofenac (DCF), ibuprofen (IBP), ketoprofen (KTP), and naproxen (NPX) - and one analgesic drug-paracetamol (PCM) - by strong-base anion exchange resin (AER) in synthetic ureolyzed urine. Freundlich, Langmuir, Dubinin-Astakhov, and Dubinin-Radushkevich isotherm models were fit to experimental equilibrium data using nonlinear least squares method. Favorable ion-exchange was observed for DCF, KTP, and NPX, whereas unfavorable ion-exchange was observed for IBP and PCM. The ion-exchange selectivity of the AER was enhanced by van der Waals interactions between the pharmaceutical and AER as well as the hydrophobicity of the pharmaceutical. For instance, the high selectivity of the AER for DCF was due to the combination of Coulombic interactions between quaternary ammonium functional group of resin and carboxylate functional group of DCF, van der Waals interactions between polystyrene resin matrix and benzene rings of DCF, and possibly hydrogen bonding between dimethylethanol amine functional group side chain and carboxylate and amine functional groups of DCF. Based on analysis of covariance, the presence of multiple pharmaceuticals did not have a significant effect on ion-exchange removal when the NSAIDs were combined in solution. The AER reached saturation of the pharmaceuticals in a continuous-flow column at varying bed volumes following a decreasing order of DCF > NPX ≈ KTP > IBP. Complete regeneration of the column was achieved using a 5% (m/m) NaCl, equal-volume water-methanol solution. Results from multiple treatment and regeneration cycles provide insight into the practical application of pharmaceutical ion-exchange in ureolyzed urine using AER.

Adsorption of three selected pharmaceuticals and personal care products (PPCPs) (ketoprofen (KEP), carbamazepine (CBZ), and bisphenol A (BPA)) by two reduced graphene oxides (rGO1 and rGO2) and one commercial graphene was examined under different solution conditions. Single-walled carbon nanotubes (SWCNTs), multiwalled carbon nanotubes (MWCNTs), and powdered graphite were also investigated for comparison. All adsorption isotherms followed the order of SWCNTs > rGO1 > rGO2 > MWCNTs > graphene > graphite, consistent with the orders of their surface areas and micropore volumes. After surface area normalization, adsorption affinities of the three PPCPs onto graphenes were lower than onto graphite, suggesting incomplete occupation for adsorption sites because of the aggregation of graphene sheets and the presence of oxygen-containing functional groups. The observed pH effects on adsorption correlated well with the pH-regulated distribution of the protonated neutral species of the three PPCPs. Increasing ionic strength from 0 to 20 mM increased KEP adsorption due to the electrostatic screening by Na(+) and Ca(2+). Both humic acid (HA) and sodium dodecylbenzenesulfonate (SDBS) suppressed PPCPs adsorption to all adsorbents, but their impacts onto graphenes were lower than those onto CNTs because of their lower adsorption by graphenes. More severe HA (or SDBS) effect was found on negatively charged KEP at the tested solution pH 6.50 due to the electrostatic repulsion between the same charged KEP and HA (or SDBS). The findings of the present study may have significant implications for the environmental fate assessment of PPCPs and graphene.

With the present contribution, we demonstrate that the baseline separation of ketoprofen enantiomers can be successfully achieved (α = 1.09; R(S) = 1.60) in the reversed-phase mode of elution with a commercially available anion-exchange-based chiral stationary phase, incorporating the quinine 2,6-diisopropylphenyl carbamate derivative as the enantioresolving unit. Focused modification of the eluent composition indicated a stereoselective role of hydrophobic and π-π interactions between the selector and selectand units, besides the prime ionic intermolecular interaction. The mechanistic hypotheses based on the chromatographic data were confirmed by in silico molecular dynamic simulations, which allowed us to establish the network of selector-selectand interactions underlying the stereorecognition process at a molecular level. Homochiral metal-organic frameworks with fine-tuned pore sizes/walls and large surface areas are promising porous materials for enantioseparation considering the traditional zeolite molecular sieves have no chirality. Using enantiopure pyridyl-functionalized salen [(N-(4-Pyridylmethyl)-L-leucine·HBr)] as a starting material, we have prepared a noninterpenetrated three-dimensional homochiral metal organic framework {[ZnLBr]·H2O}n, which was further used as a chiral stationary phase for high-performance liquid chromatography to enantioseparate racemic drugs, showing excellent performances in enantioseparation of drugs. The metal-organic framework can be regarded as a novel molecular sieve-like material with a chiral separation function based on the relative sizes of the chiral channels and the resolved molecules.

A model describing ibuprofen and ketoprofen biodegradation by activated sludge from three different wastewater treatment plants (WWTP) was developed in this study. This model successfully described the biodegradation profiles observed at two different initial concentrations of each compound, where a lag-phase was observed prior to the biodegradation of each compound. Twelve ibuprofen and ketoprofen degrading isolates were identified in this study from the WWTP sludge showing the best removal performance. One of these isolates was characterised via another model, where biodegradation was dependent on biomass growth rate as well as the ibuprofen concentration. The fact that different models were needed to describe the biodegradation by activated sludge and a pure culture suggests that the biodegradation of non-steroidal anti-inflammatory drugs (NSAIDs) depends on the microbial community, thus pharmaceutical biodegradation models may require adaptation depending upon the system. The enantiomers of ketoprofen were separated by capillary electrophoresis using the (2,3,6-tri-O-methyl)-derivatives of α-, β-, and γ-cyclodextrin (CyD) as chiral selectors. The affinity pattern of the ketoprofen enantiomers toward these CyDs changed depending on their cavity size. Thus, with hexakis (2,3,6-tri-O-methyl)-α-CyD and heptakis (2,3,6-tri-O-methyl)-β-CyD, the R enantiomer of the drug migrated first, whereas the enantiomer migration order was reversed in the presence of octakis(2,3,6-tri-O-methyl)-γ-CyD. A stir membrane liquid phase microextraction procedure working under the three-phase mode is proposed for the first time for the determination of six anti-inflammatory drugs in human urine. The target compounds are isolated and preconcentrated using a special device that integrates the extractant and the stirring element. An alkaline aqueous solution is used as extractant phase while 1-octanol is selected as supported liquid membrane solvent. After the extraction, all the analytes are determined by liquid chromatography (LC) with ultraviolet detection (UV). The analytical method is optimized considering the main involved variables (e.g., pH of donor and acceptor phases, extraction time, stirring rate) and the results indicate that the determination of anti-inflammatory drugs at therapeutic and toxic levels is completely feasible. The limits of detection are in the range from 12.6 (indomethacin) to 30.7 μg/L (naproxen). The repeatability of the method, expressed as relative standard deviation (RSD, n = 5) varies between 3.4% (flurbiprofen) and 5.7% (ketoprofen), while the enrichment factors are in the range from 35.0 (naproxen) to 72.5 (indomethacin).

The adsorption of two acidic pharmaceutically active compounds (PhACs), clofibric acid and ketoprofen, onto powdered activated carbon (PAC) was investigated with a particular focus on the influence of natural organic matter (NOM) on the adsorption of the PhACs. Suwannee River humic acids (SRHAs) were used as a substitute for NOM. Batch adsorption experiments were conducted to obtain adsorption kinetics and adsorption isotherms with and without SRHAs in the system. The adsorption isotherms and adsorption kinetics showed that the adsorption ofclofibric acid was not significantly affected by the presence of SRHAs at a concentration of 5 mg (as carbon) L(-1). An adsorption capacity of 70 to 140 mg g(-1) was observed and equilibrium was reached within 48 h. In contrast, the adsorption of ketoprofen was markedly decreased (from about 120 mg g(-1) to 70-100 mg g(-1)) in the presence of SRHAs. Higher initial concentrations of clofibric acid than ketoprofen during testing may explain the different behaviours that were observed. Also, the more hydrophobic ketoprofen molecules may have less affinity for PAC when humic acids (which are hydrophilic) are present. The possible intermolecular forces that could account for the different behaviour of clofibric acid and ketoprofen adsorption onto PAC are discussed. One monomer molecularly imprinted polymer coatings were first synthesized in fused silica capillary columns with 2-methacrylamidopropyl methacrylate (MAM) as single functional monomer in addition to a cross-linking monomer. Since MAM may generate no or little EOF, a strategy of precursor of polymerization, which does not interfere with the formation of defined imprints, was used to introduce an ionizable functional monomer to generate a stable electroosmotic flow for electrochromatography (CEC) by post-polymerization hydrolization. The resulting MAM-based open-tubular imprinted capillary was able to separate enantiomers by means of CEC. The resolution of enantiomers separation achieved on S-amlodipine-imprinted capillary was up to 16.1. The strong recognition ability (selectivity factor was 3.23) and high column performance (theory plates was 26,053 plates m(-1)) of template were obtained. The MIP coatings were also prepared using either S-naproxen or S-ketoprofen as template molecule. The resolutions of enantiomers separation were 2.20 and 4.56, respectively. The results illustrate that the synthesis of MIP using one monomer is not only an experimental-simplified process, but also an approach to producing chiral stationary phase with high efficiency and selectivity., (Copyright © 2011 Elsevier B.V. The fate of pharmaceuticals in a wastewater treatment plant (WWTP) in Kumamoto, Japan with activated sludge treatment is reported. Selected pharmaceuticals were detected in influent. Results from the present study confirmed that Acetaminophen, Amoxicillin, Ampicillin and Famotidine were removed at a high rate (>90% efficiency). In contrast, removal efficiency of Ketoprofen, Losartan, Oseltamivir, Carbamazepine, and Diclofenac was relatively low (<50%). The selected pharmaceuticals were also detected in raw sludge. In digestive process, Indomethacin, Atenolol, Famotidine, Trimethoprim and Cyclofosamide were removed at a high (>70% efficiency). On the other hand, removal of Carbamazepine, Ketoprofen and Diclofenac was not efficient (<50%).

We designed and synthesized a cinchonine derivative to be used as a novel chiral monomer. It was employed in a dual role of functional monomer and cross-linking monomer, displaying multi-binding sites for the template (S)-ketoprofen. Monodisperse molecularly imprinted core-shell microspheres were prepared using surface imprinting method on silica gel. The results show a substantial synergistic effect in the enantioselective recognition, confirming our initial hypothesis. Computational simulation of the monomer and template pre-arrangement strongly supports our proposed chiral recognition mechanism for the imprinted microspheres., (Copyright © 2011 Elsevier B.V. We screened a strain NK13 for a certain extent asymmetric hydrolysis the rac-ketoprofen Chloroethyl ester to (S)-Ketoprofen. As identified, NK13 was Bacillus megaterium. Digested NK13 genomic DNA with Sau3AI partially and recovered the fragment from 2 kb to 6 kb, cleaved the plasmid of pUC18 with BamH I, ligated the 2-6 kb fragment of NK13 genomic DNA into pUC18 plasmid, and then transformed an Escherichia coli strain DH5alpha. We created the gene library of NK13 and obtained a positive clone, pUC-NK1 in the library from the tributyrin flat. The result of sequencing showed that there was a whole open read frame (ORF) of 633 bp lipase gene in the plasmid of pUC-NK1. To compare with the genes of GenBank, this lipase gene was reported firstly (GenBank Accession No. EU381317). The lipase gene was amplified by PCR, using pUC-NK1 plasmid as template, and subcloned into the high expression vector pET21b(+) under the control of T7 promoter. The recombinant plasmid, pET-NKest1, was then transformed into an Escherichia coli strain BL21 (DE3) for the production of recombinant lipase protein. After 3 hours of induction by isopropyl-beta-D-thiogalactoside (IPTG), lipase was expressed. SDS-PAGE analysis showed that the relative molecular mass of the lipase protein was about 20 kDa. The result of high performance liquid chromatography (HPLC) showed that the conversion rate of the recombinant strain was fifty times than the wild strain NK13's. The (S)-Ketoprofen enantiomeric excess of the recombinant strain was 75.28%, which indicated that the lipase could hydrolyze (S)-Ketoprofen Chloroethyl ester firstly. If we research the conditions of the hydrolysis rac-ketoprofen Chloroethyl ester of this lipase further, maybe it could offer a foundation to product (S)-Ketoprofen industrially.

A rapid approach has been developed for screening trace level compounds with antitumor activities based on their interactions with microtubules. This interaction can be quantified with liquid chromatography (LC) by measuring the difference of bioactive compound's concentration before and after the formation of compound-microtubule complexes in the fast dialyzers. To test the effectiveness of this approach, several antitumor drugs such as colchicine, taxol, daunorubicin, and a non-antitumor reagent ketoprofen were used. Results indicate that the antitumor constituents can be identified without any disturbance, and the inactive components can be excluded. This screening method was then successfully applied to some potential antitumor compound mixtures, and the active compounds could be separated and screened rapidly. The binding activities measured were consistent with their cytotoxicity assays. This integrative approach is rapid and convenient for screening, isolating, and analyzing potential antitumor active compounds from a mixture.

The aim of this study was to develop an analytical procedure which allows the quantification of pharmaceuticals in water at the ng L(-1) level. Hence, it is reported research on the application of a rapid, inexpensive and simple continuous hollow fiber liquid-phase micro extraction (CHF-LPME) for the pre-concentration and determination of non-steroidal anti-inflammatory drugs (NSAIDs), such as ibuprofen (IBP), naproxen (NAP), and ketoprofen (KEP), in wastewater. In this method, a 2.50 cm end sealed piece of a polypropylene hollow fiber was immersed into the organic solvent, octanol, for 30 s. After solvent impregnation with the pores of the fiber, the excess amounts of solvent were removed from inside the fiber, and 4.0 microL of octanol, as the acceptor phase, was injected into the fiber carefully. The fiber was settled using a microsyringe into a 10.0 mL glass test tube, and 20.00 mL of the aqueous solution (the donor phase), was circulated by a pump around it. After analyte extraction for an optimized period of time (20 min), 2 microL of the organic solvent was withdrawn into the microsyringe and injected into the GC-FID for further analysis. Finally, based on the optimized analytical conditions, the method was linear in the range of 2.5-500 ng L(-1). The limits of detection were 1-2 ng L(-1). Repeatability of this method on an intra-day scale was 3.4-10.2% (RSD%). NSAIDs have been detected in several municipal wastewater samples, and the concentration range was 9.0-19.0 ng L(-1).

Experiments were performed to evaluate whether counter-current chromatography (CCC) could function as an alternative purification method to reversed-phase high-performance liquid chromatography (RP-HPLC) and normal-phase supercritical fluid chromatography (SFC). RP-HPLC and SFC are the routine methods currently used in our high-throughput purification (HTP) facility for the purification of high-throughput organic synthesis (HTOS) libraries and medicinal chemistry reaction mixtures. Pre-equilibration of the solvent mixture layers was not mandatory for effective chromatography when hexanes-ethyl acetate-methanol-water (HEMW) solvent mixtures were used. Key to the use of CCC for high-throughput applications is the ability to effectively select a solvent system appropriate to each library member. Pilot-scale CCC elution time was used to estimate a starting solvent ratio and RP-HPLC retention time was then used to adjust solvent ratios within a particular library. It was also found that dimethyl sulfoxide (DMSO) and DMSO-methanol were suitable as sample injection solvents when using the HEMW solvent systems.

Some racemic nonsteroidal anti-inflammatory drugs, namely naproxen, indoprofen, ketoprofen, flurbiprofen, carprofen, cicloprofen, flunoxaprofen and suprofen were separated into their enantiomers by nano-LC. Chiral recognition was achieved adding to the mobile phase heptakis (2,3,6-tri-O-methyl)-beta-cyclodextrin (TM-beta-CD). Capillary columns of 100 microm id, packed with different RP particles were used for experiments. Effect of experimental parameters such as mobile phase composition, stationary phase type and length of packed capillary column on retention factor and chiral resolution of analytes were studied. The stationary phase type played a very important role in the enantiorecognition process. Best results in terms of highest enantioresolution factor and largest number of separated enantiomers were obtained reducing the particles size to 3 microm with RP(18) stationary phase. Most favourable mobile phase for enantiodiscrimination was obtained using relatively low concentrations of ACN (30%, v/v), 30 mM of TM-beta-CD and pH value of 3.0. The retention time of all studied enantiomers decreased by increasing the CD derivative concentration. The retention factors of selected studied compounds, specifically flurbiprofen, naproxen and suprofen, were measured employing TM-beta-CD concentrations in the range 0-40 mM. Assuming a 1:1 enantiomer/CD ratio, the apparent association constants of the studied enantiomers were calculated.

Enhanced chiral separation performance has been observed for ketoprofen enantiomers in capillary electrochromatography (CEC) with an open-tubular (OT) column prepared with a specific molecule imprinted polymer (MIP) on the innerwall of 50mum ID capillary. The column was prepared by in situ thermal polymerization inside the pretreated and silanized fused silica capillary. A specific diluted monomer mixture composed of S-ketoprofen, methacrylic acid (MAA, functional monomer), ethylene glycol dimethacrylate (EDMA, cross-linker), and 4-styrenesulfonic acid (4-SSA) dissolved in 9/1 (v/v) acetonitrile/2-propanol was used to fabricate the OT-MIP layer. 4-SSA was added to form a MIP layer capable of stable and strong electro-osmotic flow (EOF) over the pH range of this study securing CEC elution of ketoprofen having partial negative charge near the optimized pH. Various parameters such as buffer pH, organic modifier composition, salt concentration, and applied potential have been optimized for CEC chiral separation of ketoprofen enantiomers. Very good separation selectivity and efficiency were observed, thus the chromatographic resolution of ketoprofen enantiomers was as high as 10.5, and the number of theoretical plates of R-ketoprofen, 156,000/m (40,000/m for S-ketoprofen), which proves that the OT-MIP-CEC type approach is a promising strategy in MIP study.

This study is about the preparation of an open tubular capillary column of molecularly imprinted polymer (MIP) and its application to chiral separation by microLC. A non-covalent in-situ molecular imprinting polymerization protocol was used to synthesize the S-ketoprofen MIP. A special procedure was employed to secure formation of an open tubular and rigid MIP layer in a silica capillary of 100 microm id. The capillary was filled with the reaction mixture, sealed, and placed in a water bath at 50 degrees C for 3 h. Then it was flushed with a 0.5 MPa nitrogen flow for 5 min, and was again placed in the water bath for 2 h to complete MIP formation. Methacrylic acid (MAA) has been known to be an inefficient functional monomer in preparation of MIP of an acid molecule. However, MAA was used with ethylene glycol dimethacrylate in preparation of the S-ketoprofen MIP in this study. The open tubular structure and the microLC mode of separation enabled free optimization without any restriction, thus a very good resolution (R=4.7) of ketoprofen enantiomers was achieved when a mobile phase composed of 30% acetonitrile and 70% acetate buffer at pH 4.5 was used with 5 mbar inlet pressure. This may be partially attributed to the open tubular structure of our MIP, enabling low column back-pressure and free optimization of eluent composition, as well as to the small capillary dimensions. Our MIP capillary column also showed some versatility in chiral separation, thus a good chiral separation was observed for naproxen, ibuprofen, and fenoprofen enantiomers.

An enzymatic membrane reactor (EMR) for enantioseparation of (R,S)-ketoprofen via Candida antarctica lipase B (CALB) as biocatalyst was investigated. A comparative study of free and immobilized CALB was further conducted. The catalytic behaviour of CALB in an EMR was affected by the process parameters of enzyme load, substrate concentration, substrate molar ratio, lipase solution pH, reaction temperature, and substrate flow rate. Immobilization of CALB in the EMR was able to reduce the amount of enzyme required for the enantioseparation of (R,S)-ketoprofen. Immobilized CALB in the EMR assured higher reaction capacity, better thermal stability, and reusability. It was also found to be more cost effective and practical than free CALB in a batch reactor.

Removal property of nine pharmaceuticals (clofibric acid, diclofenac, fenoprofen, gemfibrozil, ibuprofen, indomethacin, ketoprofen, naproxen and propyphenazone) by chlorination, coagulation-sedimentation and powdered activated carbon treatment was examined by laboratory-scale experiments under the conditions close to actual drinking water treatment processes. Indomethacin and propyphenazone were completely degraded by chlorination within 30 minutes, but others remained around 30% (naproxen and diclofenac) or more than 80% of the initial concentration after 24 hours. A couple of unidentified peaks in a chromatogram of the chlorinated samples suggested the formation of unknown chlorination by-products. Competitive adsorption was observed when the mixed solution of the target pharmaceuticals was subjected to batch adsorption test with powdered activated carbon. Clofibric acid and ibuprofen, which were relatively less hydrophobic among the nine compounds, persisted around 60% of the initial concentration after 3 hours of contact time. Removal performance in actual drinking water treatment would become lower due to existence of other competitive substances in raw water (e.g. natural organic matter). Coagulation-sedimentation using polyaluminium chloride hardly removed most of the pharmaceuticals even under its optimal dose for turbidity removal. Electrokinetic cross membrane extraction of acidic drugs was demonstrated for the first time. The acidic drugs were extracted from an alkaline aqueous donor solution (300 microl), through a thin supported liquid membrane of 1-heptanol sustained in the pores of the wall of a porous hollow fiber, and into an aqueous alkaline acceptor solution (30 microl) present inside the lumen of the hollow fiber by the application of a d.c. electrical potential. The negative electrode was placed in the donor solution, and the positive electrode was placed in the acceptor solution. Optimal extractions were accomplished with 1-heptanol as the supported liquid membrane, with 50 V as the driving force, and with pH 12.0 in both the donor and acceptor solutions, respectively (NaOH). Equilibrium extraction conditions were obtained after 5 min of operation with the whole assembly agitated at 1200 rpm. Eleven different acidic drugs were extracted with recovery values between 8 and 100%, and initial data supported that electrokinetic cross membrane extraction provided repeatable data and linear response between original donor concentration and final acceptor concentration of the acidic model compounds.

The elimination of six acidic pharmaceuticals (clofibric acid, diclofenac, ibuprofen, ketoprofen, mefenamic acid, and naproxen) in a real wastewater treatment plant (WWTP) using an activated sludge system and membrane bioreactors (MBRs) was investigated by using a gas chromatography/mass spectrometry (GC/MS) system for measurement of the compounds. Limited information is available for some of the tested pharmaceuticals at present. Solid retention times (SRTs) of the WWTP and the two MBRs were 7, 15, and 65 days, respectively. The elimination rates varied from compound to compound. The MBRs exhibited greater elimination rates for the examined pharmaceuticals than did the real plant. Dependency of the elimination rates of the pharmaceuticals on SRTs was obvious; the MBR operated with a longer SRT of 65 days clearly showed better performance than did the MBR with a shorter SRT of 15 days. The difference between the two MBRs was particularly significant in terms of elimination of ketoprofen and diclofenac. Measurements of the amounts of adsorbed pharmaceuticals on the sludge and aerobic batch elimination experiments were carried out to investigate the elimination pathways of the pharmaceuticals. Results of the batch elimination tests revealed that the sludges in the MBRs had large specific sorption capacities mainly due to their large specific surface areas. Despite the sorption capacities of sludges, the main mechanism of elimination of the pharmaceuticals in the investigated processes was found to be biodegradation. Biodegradation of diclofenac, which has been believed to be refractory to biodegradation, seemed to occur very slowly.

In capillary electrochromatography (CEC), magnetic particles (MPs) were packed in a fused silica capillary by using the magnetic field to be retained without frits. For a chiral CEC separation, avidin was immobilized onto the surface of the MPs (AVI-MPs) as a stationary phase by using the physical adsorption technique. The injected AVI-MPs into the capillary were stably captured with the magnet (surface magnetic flux density, 250 mT) under the separation voltage of 10 kV (190 V/cm). By employing the fritless AVI-MPs packed capillary, the chiral separation of ketoprofen was successfully attained with the packing length of only 5 cm. Effects of the modification condition of avidin, pH of background solution, and the packing length on the enantioseparation were also investigated. Under the optimal condition, furthermore, the repeatability for the retention time of ketoprofen was better than 1.5% in the relative standard deviation and the capillary-to-capillary reproducibility was also acceptable in the prepared fritless capillaries.

In the present study, penicillin G acylase (PGA), an enzyme belonging to the family of hydrolases, has been investigated as chiral selector in CE using the partial filling technique. Owing to the strong disposition of PGA to be adsorbed by the inner capillary wall, permanently coated capillaries were used to diminish both the protein-wall interactions and the EOF. In particular, the silica surface of the capillary was chemically coated by an antiadhesive and an hydrophilic layer of pullulan, a high-molecular-mass homopolysaccharide. The coating procedure consisted in the silanization with glycidoxypropyltrimethoxysilane and the subsequent coupling of the hydroxyl groups of pullulan onto the silanized capillary. Using this approach, a significant EOF suppression was obtained within a wide pH range (pH 3.0-9.0); this result was very important in order to find the suitable conditions for the application of partial filling technique. The optimization of partial filling was carried out by considering the effects of different experimental conditions (buffer pH, PGA concentration, and loading duration), on the migration time and enantioresolution of rac-ketoprofen. Under the selected conditions as: 100 mM sodium phosphate buffer (pH 5.5) containing 240 microM of PGA (partial filling of 120 s at a pressure of 50 mbar), a series of acidic compounds resulted to be enantioresolved in about 10 min. The long-term stability of the proposed coating was evaluated; more than 100 injections were performed without significant loss of reproducibility.

A high-performance liquid chromatographic method for chiral separation of ketoprofen racemate was developed. (R)- and (S)-ketoprofen enantiomers were separated on a Hypersil BDS C8 column (150 mm x 4.6 mm i.d., 5 microm) at 25 degrees C, using acetonitrile-triethylamine acetate (TEAA) buffer (pH 5.2, 20 mM) (35:65, v/v) containing 2.0 mM norvancomycin as the mobile phase. Effects of norvancomycin concentration, content of acetonitrile and TEAA buffer pH on the enantioseparation were investigated. The method was validated for linearity, repeatability, limits of detection (LOD) and limits of quantification (LOQ). Calibration curves (r2 = 0.999) were constructed in the range of 2.01-200.8 microg ml(-1) for (S)-ketoprofen and 2.04-152.4 microg ml(-1) for (R)-ketoprofen, respectively. Repeatability (n = 5) showed less than 2% relative standard deviation (R.S.D.). LOD and LOQ for the two enantiomers were found to be 0.20 and 0.78 ng for (S)-ketoprofen, 0.20 and 0.86 ng for (R)-ketoprofen, respectively. Norvancomycin and vancomycin as chiral mobile phase additives (CMPAs) in the chiral separation showed similar abilities of enantioseparation. However, to obtain the optimum enantioseparation, a lower concentration of norvancomycin than that of vancomycin is required.

A Bacillus strain having a certain extent of asymmetric resolution for (S)-ketoprofen was studied for its culture and conversion condition. The distribution and properties of lipases/esterases from its bacterial cell and the change of asymmetric resolution ability were also investigated. After purification by hydrophobic interaction chromatography and gel filtration, the lipase/esterase terase activity of active fraction was 61.7 times higher than the crude enzyme and the purity of 23 kDa protein increased more than 400 times. Hydrolytic activity of lipase/esterase using ketoprofen chloroethyl ester and pNPA as substrate remained essentially constant in the reaction system during the purification procedure. The above result clearly indicated that the application of hydrophobic interaction chromatography-gel filtration was fast, useful and effective when compared to the difficulties in purification of low content crude enzyme prepared directly from bacterial cells. Experimental results also indicated that there are several lipases/esterases in the bacillus cell selective to the optically active ketoprofen and the conversion result was derived from the comprehensive function of all these enzymes. This is a new way to understand the bacterial asymmetric resolution with high conversion rate and low optical purity. The ESI-QUAD-TOF mass spectrum was used to analyze 23 kDa protein. Protein identities were revealed by searching sequence databases (MSDB) with peptide sequence tags. The result showed that 23 kDa protein is a novel protein.

A new fabrication procedure for integration of ultraviolet transparent pure-silica planar waveguides, fiber couplers and high-aspect ratio submicrometer channels is presented. Only a single photolithographic mask step is required. The channels are 80-90 microm deep and the width can be reduced to about 0.5 microm, corresponding to a height-to-width ratio of more than 150. The core of the waveguides consists of pure silicon dioxide, which is favorable over doped silica, due to the absence of absorption centers associated with the dopants. This furthermore improves the long-term stability of the waveguides, because of an increased radiation resistance of the glass. The propagation loss decreases from 1.0 dB/cm at 200 nm to 0.2 dB/cm at 800 nm, which, to our knowledge, is the lowest propagation loss reported for integrated planar waveguides in the ultraviolet wavelength region to date. The effective optical path length is 1.2 mm for an absorbance cell with a nominal length of 1.0 mm, indicating effective suppression of stray light. The limit of detection for paracetamol when present in the entire channel network was determined to 3 microg/mL. Finally, the applicability of the fabricated devices for capillary electrophoresis was evaluated by separation of caffein, paracetamol and ketoprofone using absorbance detection at 254 nm.

Today, a heightened awareness of the applicability of enantiomers in medicine and clinical practice has been gene-rated due to the continuous evolvement of the field of chirality. In this context, this article provides a review of separation of ketoprofen, an important drug, in a popular class of non-steroidal anti-inflammatory drugs (i.e. profens). This review highlights various methodologies, logistical considerations for separation and provides an exhaustive list of applications mainly focusing on the pharmacokinetic aspects. Clearly, the application of enantioselective methods for drug racemates paves the way to understand the in vivo behavior of individual enantiomer and hence an opportunity for an alternate and/or better option for treating the disease., (Copyright 2003 John Wiley & Sons, Ltd.)

Simultaneous enantioseparations of nine profens for their accurate chiral discrimination were achieved by capillary electrophoresis (CE) in the normal polarity (NP) mode with a single cyclodextrin (CD) system and in the reversed polarity (RP) mode with a dual CD system. The single CD system in the NP mode employed heptakis(2,3,6-tri-O-methyl)-beta-cyclodextrin (TMbetaCD) added at 75 mM-100 mM 2-(N-morpholino)ethanesulfonic acid buffer (pH 6.0) as the optimum run buffer. The dual CD system operated in the RP mode used 30 mM TMbetaCD and 1.0% anionic carboxymethyl-beta-cyclodextrin dissolved in pH 3.0, 100 mM phosphoric acid-triethanolamine buffer containing 0.01% hexadimethrine bromide added to reverse the electroosmotic flow. Fairly good enantiomeric resolutions and the opposite enantiomer migration orders were achieved in the two modes. Relative migration times to internal standard under respective optimum conditions were characteristic of each enantiomer with good precision (< 2% relative standard deviation, RSD), thereby enabling to crosscheck the chemical identification of profens and also their accurate chiralities. The method linearity in the two modes was found to be adequate (r > or = 0.9991) for the chiral assay of the profens investigated. Simultaneous enantiomeric purity test of ibuprofen, ketoprofen and flurbiprofen in a mixture was feasible in a single analysis by the present method.

Aim: To establish HPLC chiral separation method for ketoprofen enantiomers by using Chirobiotic V chiral seperation phase (CSP) (A) and vancomycin as chiral mobile phase additives (B)., Methods: The separation was first performed on Chirobiotic V CSP with the mobile phase of terahydrofran (THF)-0.5% triethylanine acetate(TEAA) buffer (15:85) at the flow rate of 0.7 mL.min-1. When using vancomycin as chiral mobile phase additive, the separation was carried out on C8 column (150 mm x 4.6 mm), the mobile phase was methanol-0.25% TEAA buffer (50:50), the flow rate was 0.7 mL.min-1. The effects of the concentration of vancomycin, organic modifier and the pH of the buffer on the resolution of ketoprofen enantiomers were investigated. Also, the feasibility of these two methods to be used as quantitative method was studied., Results: Ketoprofen enantiomers were separated at a baseline level under the chromatographic condition of both methods A and B, the resolution was 2.28 and 2.22, respectively. In method A the linearity of enantiomer was obtained from 0.5 mg.L-1 to 100 mg.L-1, the detectionlimit was 1 microgram.L-1. When using vancomycin as mobile phase additive the system was shown to have a high efficiency. In this system, the assay of enantiomer is linear from 2.5 mg.L-1 to 250 mg.L-1. The detection limit was 14.5 micrograms.L-1., Conclusion: Both methods can be used to detect optical purity of S-(+)-ketoprofen.

The enantiomeric separation of some nonsteroidal anti-inflammatory drugs (NSAIDs) was investigated in capillary electrophoresis (CE) using dual systems with mixtures of charged cyclodextrin (CD) derivatives. A significant enhancement of selectivity and resolution could be achieved in the enantioseparation of these analytes in their uncharged form by the simultaneous addition of two oppositely charged CD derivatives to the background electrolyte. The combination of the single-isomer cationic CD, permethyl-6-monoamino-6-monodeoxy-beta-CD (PMMAbetaCD) and the single-isomer polyanionic CD, heptakis-6-sulfato-beta-cyclodextrin (HSbetaCD) in a pH 2.5 phosphoric acid-triethanolamine buffer, was designed and employed for the enantioseparation of profens. The improvement in selectivity and resolution can be attributed to the fact that the two CDs, which lead to independent and enantioselective complexation with the analyte enantiomers, have not only opposite effects on the electrophoretic mobility of these compounds but also opposite affinity patterns towards the enantiomers of these compounds. Binding constants for these enantiomers with each CD were determined using linear regression approach, in order to be able to predict the effect of the concentrations of the two CDs on enantiomeric selectivity and resolution in such dual systems.

Capillary electrophoresis (CE) has become a powerful tool for enantiomer separations during the last decade. Since 1993, the author has investigated enantiomer separations by affinity capillary electrophoresis (affinity CE) with some proteins and by cyclodextrin electrokinetic chromatography (CDEKC) with some charged cyclodextrins (CDs). Many successful enantiomer separations are demonstrated from our study in this review article. In the enantiomer separations by affinity CE, the deterioration of detection sensitivity was observed under high concentration of the protein in running solutions. The partial filling technique was practically useful to solve the serious problem. It allowed operation at high protein concentrations, such as 500 mumol/L, without the detection problem. Charged CDs had several advantages for the enantiomer separations over neutral ones. Strong electrostatic interactions between a charged CD and oppositely charged analytes should be effective for the formation of the complex. A large difference in electrophoretic mobility between the free analyte and the inclusion complex should also enhance the enantiomeric resolution. In CE-mass spectrometry (CE-MS), the partial filling technique was applied to avoid the introduction of nonvolatile chiral selectors into the CE-MS interface. By replacing the nonvolatile electrolytes in the running buffer by volatile ones, the separation conditions employed in CE with the UV detection method could be transferred to CE-MS.

The yeast strain CGMCC 0573 was identified as Citeromyces matriensis and shown to be capable of enantioselectively hydrolyzing ethyl ester of ( R)-Ketoprofen (2-(3-benzoylphenyl)propionic acid). The strain was isolated for the first time from soil samples through a new and efficient screening procedure in which the probability of obtaining active strains was greatly increased by using ethanol and Tween-80 alternatively as additives during the enrichment culture. Studies of the culture conditions and catalytic performance of Citeromyces matriensis CGMCC 0573 showed that the enzyme occurs constitutively in the cells and its production is enhanced by feeding with Tween-80 during the early period of cultivation. Yeast extract was found to be beneficial both for growth and for esterase production. The optimal temperature and pH for the bioconversion were 40 degrees C and pH 8.0, respectively. Biotransformation using resting cells cultured in a flask with baffles and magnetic stirring and in the presence of 50 mM substrate resulted in the production of ( R)-ketoprofen at 93% ee (enantiomeric excess) and at 42.6% conversion.

A method based on the adsorption of bovine serum albumin (BSA) on the strong anion-exchange packing (SAX) was applied to prepare chiral stationary phases for capillary electrochromatography. It was found that the electroosmotic mobility only decreased 26.3% when the BSA was adsorbed. The run-to-run reproducibilities of void time and the migration times of D- and L-tryptophan enantiomers were rather good with the relative standard deviation (RSD) values of 0.90%, 0.87% and 0.96%, respectively, when mobile phase containing 7% (volume fraction) acetonitrile. The merit of this system is that the direction of electroosmotic flow is still from cathode to anode, preferable to separate acidic enantiomers. Four anionic enantiomers, tryptophan, ketoprofen, fenoprofen and warfarin, as well as the neutral enantiomers of benzoin were successfully resolved. In order to reduce the analysis time, n-hexanoic acid was added to the mobile phase as a competitive agent. The effect of acetonitrile volume fraction on the resolution of enantiomers was also investigated.

Vancomycin is an amphoteric, glycopeptide, macrocyclic antibiotic. When attached to 5 microspherical silica gel, vancomycin proved to be an effective chromatographic chiral stationary phase that could be used in the reversed-phase mode. In this study, a bonded vancomycin chiral stationary phase (Chirobiotic Vtrade mark) was investigated for the chiral liquid chromatography analysis of ketoprofen and flurbiprofen. The selectivity factor (alpha) and the chiral resolution factor (RS) of Chirobiotic Vtrade mark were evaluated first as a function of the buffer pH and molarity, and second as a function of organic modifier type and composition of the mobile phase. Four organic modifiers (tetrahydrofuran, 2-propanol, 1,4-dioxane and methanol) have been tested for their selectivity. Optimized conditions using 20% of tetrahydrofuran in ammonium nitrate (100 mM, pH 5) were selected for the enantioseparation of flurbiprofen and ketoprofen from their racemic forms. At pH 5, these acidic compounds are almost negatively charged, while the chiral selector possesses a positive charge allowing it to interact electrostatistically with the analytes. A new, automatic method for the clean-up, preconcentration, separation, and quantitation of nonsteroidal anti-inflammatory drugs (NSAIDs) in biological samples (human urine and serum) using solid-phase extraction coupled on-line to capillary electrophoresis is proposed. Automatic pretreatment is carried out by using a continuous flow system operating simultaneously with the capillary electrophoresis equipment, to which it is linked via a laboratory-made mechanical arm. This integrated system is controlled by an electronic interface governed via a program developed in GWBasic. Capillary electrophoresis is conducted by using a separation buffer consisting of 20 mM NaHPO4, 20 mM beta-cyclodextrin and 50 mM SDS at pH 9.0, an applied potential of 20 kV and a temperature of 20 degrees C. The analysis time is 10 min and the detection limits were between 0.88 and 1.71 microg mL(-1). Automatic clean-up and preconcentration is accomplished by using a C-18 minicolumn and 75% methanol as eluent. The limit of detection of NSAIDs can be up to 400-fold improved when using sample clean-up. The extraction efficiency for these compounds is between 71.1 and 109.7 microg mL(-1) (RSD 2.0-7.7%) for urine samples and from 77.2 to 107.1 microg mL(-1) (RSD 3.5-7.1%) for serum samples.

The enantiomeric separation of ketoprofen in three mobile phase systems on an ovomucoid (OVM) chiral stationary phase (CSP) column was investigated. Suitable chromatographic conditions for simultaneous separation of the two pairs of enantiomers of ketoprofen and its methyl ester were adopted. All of the four compounds occuring in the process of enzymatic stereoselective hydrolysis or esterification can be simultaneously separated.

Egg white protein-bonded columns were developed for HPLC. These columns can be used under aqueous mobile phase and separate various kinds of drug enantiomers. Hyphenated techniques using protein-bonded columns with LC/MS and/or column switching have been recognized as integral methods in pharmaceutical research to analyze drug enantiomers. Therefore, these methods are very useful for research fields of pharmacokinetics and pharmacology.
