- Abstract
Thyme is a perennial herb that grows in temperate climatic regions of the world. The essential oils of the plant, which has been used for medicinal purposes since ancient times, are responsible for both its antimicrobial and antioxidant properties. The composition of volatile components differs from species to species, and these differences create specific taste and odor for each species. The GC-MS method is frequently used in the characterization of essential oils, and the method needs to be modified according to the column type and length used, the furnace temperature program, and the characteristics of the carrier gas. The fact that the essential oil components have different evaporation points has necessitated the use of different furnace temperature programs. In some studies, the essential oil components of the same sample were examined in columns with different polarities and it was determined that there was a change in the number of the detected components. Although the concentration of essential oil components varies according to the harvest season of thyme, the geographical and climatic characteristics of the region where it grows, it has been revealed in many studies that the dominant essential oil components among the species are similar. Thyme is one of the spices that is widely consumed in the regions where it grows and is used in many dishes, especially soups, and gives a regional identity to the dish where it is used. In this study, the amount of phytochemicals and antioxidant capacity of the plant Satureja hortensis L. (süpürge kekiği, çibriska) which grows spontaneously and is used as a spice in meals in the Karakoçlar settlement area of Olur district of Erzurum province was investigated. The total amounts of phenolic and flavonoid substances were determined to be 40.85±0.85 mg GAE/g and 26.52±0.46 mg QE/g, respectively. In addition, the DPPH radical scavenging activity of the plant extract was calculated as 45.24 ±3.47 μg TE/mL and the IC50 value was calculated as 177.11±0.99 μg/mL. It was determined that the Cu2+ reducing antioxidant capacity of the S. hortensis plant extract was 2162.09±113.61 μg TE/mL and the Fe3+ reducing antioxidant capacity was 24.36±4.59 μg TE/mL. As a result, it was concluded that the S. hortensis plant, which is known with different names in different regions, has antioxidant potential and can be used as a natural antioxidant source in addition to its common use.

8. Some Physicochemical Characteristics of Denizli Thyme (Origanum onites) Honey.

In this study, the number of beekeepers staying within the borders of Denizli province, which is one of the important accommodation centers of wandering beekeepers with its large thyme fields, and their accommodation places were determined and some information about thyme honey was obtained. Especially in Gözler, Uzunpınar, Belenardıç and Güzelpınar neighborhoods of Pamukkale district and Yağcılar neighborhoods of Güney district, thyme fields are dense and approximately 24 thousand bee hives stayed in 2021. Some physicochemical analyzes were carried out, for example, 9 honeys in 2021 and 6 honeys in 2022, which were collected in July when the Denizli thyme honey produced in these areas was harvested. First, pollen analysis was made in honey samples, and it was determined that it was thyme honey. Then, proline, diastase activity (DA), moisture, electrical conductivity (EC), pH, free acidity (FA), HMF, fructose (F), glucose (G), total phenolic content (TPC) was determined in 15 honey samples. Average amount of proline in thyme honey samples in 2021 and 2022, respectively; 663.5 and 708.9 mg/kg, DA 10.9 and 11.75; humidity rates 17.1% and 17.3%; EC 560 and 540 µS cm-1 ; pH 3.8 and 3.8; FA; 26.4 and 28.8 meq.kg-1 ; HMF, 9.5 and 12.4 mg/kg; F+G 69.6% and 69%; F/G was determined as 1.13 and 1.10, and the TPC was determined as 75.9 mg GAE/100g. In the future, it will be possible to reveal the descriptive features of Denizli thyme honey from other honeys by conducting more comprehensive and advanced analyzes. [ABSTRACT FROM AUTHOR]

Objective Cadmium (Cd) is a very toxic and carcinogenic heavy metal that can cause harmful effects on human health. Toxicity may develop due to Cd exposure, especially in the kidneys. Ankaferd blood stopper (ABS) is a herbal mix that is used for its hemostatic properties in surgery. Also, ABS enhances wound and tissue healing. In this study, we aimed to evaluate the possible ameliorative effects of ABS in Cd-induced renal damage. Material and Method Thirty-two male rats were randomly divided into 4 groups: control, Cd (cadmium chloride, 2.5 mg/kg single dose, ip), ABS (ABS, 1.5 ml/kg single dose ip), and Cd+ABS (cadmium chloride, 2.5 mg/kg single dose ip-ABS, 1.5 ml/kg single dose ip). At the end of the experiment, urea and creatinine levels were analyzed from the rats' serum. In addition, total oxidant status (TOS), total antioxidant status (TAS) levels, superoxide dismutase (SOD), and glutathione peroxidase (GPx) activity were measured spectrophotometrically from renal tissues. The oxidative stress index (OSI) was calculated from TOS and TAS levels. Also, we evaluated alterations in the mRNA expression of Bcl-2-associated X protein (Bax), B-cell-lymphoma-2 (Bcl- 2), silenced information regulator 1 (SIRT1), and p53 in kidney tissue of rats by using the qRT-PCR method. Results In the Cd group, serum urea, creatinine levels, and tissue oxidative stress markers, TOS and OSI were significantly higher while Gpx activity was significantly lower than in the control group (p<0.05). Also, the expression of p53 and in Bax/Bcl2 ratio significantly increased in the Cd group (p<0.05). But, ABS treatment significantly decreased urea, creatinine, TOS, OSI levels, and Bax/Bcl2 ratio, p53 expression in Cd applied group (p<0.05). Conclusion Ankaferd blood stopper showed protective effects by reducing oxidative stress and mitochondria-mediated apoptosis in acute Cd exposure. [ABSTRACT FROM AUTHOR]

Contrary to the shrinking agricultural lands, it becomes very important to minimize the pre-harvest and post-harvest losses in agricultural production due to the constantly increasing population. In this study, thyme and lavender oil were applied to the fruits of the 'Fuji' variety of apple species with high ethylene production, with the aim of minimizing post-harvest losses, and the possibilities of being an alternative to 1-MCP, which is the most widely used commercially, were investigated in terms of results. Although promising results were obtained in the short and medium term in terms of weight loss, the effectiveness of the applications decreased with the increase in storage time. At the end of the 9-month storage period, it was determined that approximately 34.6% of fruit weight loss occurred in the control group, while these values were determined as 23.6%, 26.4% and 27.5% in the promising 400 ppm thyme, 500 and 600 ppm lavender applications, and similar results were obtained with 1-MCP (27.5%). Titratable acidity (TEA), which was determined as 0.33% before storage, showed a regular decrease during the storage period and was measured as 0.13% at the end of the storage period in the control group. As expected, the water-soluble dry matter increased in the early period of storage and decreased continuously afterwards. In line with the ninemonth storage results, it was concluded that 400 ppm thyme and 500 ppm lavender applications are promising in minimizing the loss of SÇKM and TEA. In terms of all the properties examined, it can be said that 400 ppm thyme and 500 ppm lavender applications can be alternatives to 1-MCP in order to reduce post-harvest losses. The thyme (Thymus sipyleus BOISS. subsp. sipyleus BOISS. var. sipyleus L.) plant was rooted from its natural environment at an altitude of 1750 m from Beyşehir Muslu Yayla location and was cultured at 1345 m altitude in Beyşehir İslibucak. Nitrogen + Phosphorus + Potassium (K1), Micro Elements (Boron + Copper + Iron + Manganese + Molybdenum + Zinc) (K3) and Nitrogen + Phosphorus + Potassium + Micro Elements (Boron + Copper + Iron + Manganese + Molybdenum + Zinc) (K2 and K4) was used as fertilizer mixtures for thyme production. Plant samples were harvested in three separate periods in August, September and October 2019 and SPME-GC-MS was used for the analysis of its volatile components. The results showed that the applied fertilizer mixtures and the harvest period provided a variety in the amounts of the thyme plant main volatile components. The highest value of p-cymene (43.76%) in the October harvest of thyme produced using K4 fertilizer mixture application, the highest value of γ-terpinene (24.79%) in the August harvest of thyme produced using K4 fertilizer mixture application, the highest value of thymol (20.26%) in the August harvest of thyme produced using K3 fertilizer mixture application and the highest value of carvacrol (67.01%) in the September harvest of thyme produced using K1 fertilizer mixture application were observed.

The effect of adding 0.05% natural herb extracts obtained from rosemary (Rosmarinus officinalis), thyme (Thymbra spicata), and basil (Ocimum basilicum L.) on mackerel balls during 10 months of frozen storage (-18oC) were investigated concerning sensory (raw and cooked), biochemical (PV-peroxide value, FFA-free fatty acids, TVB-N -total volatile basic nitrogen, TBA- thiobarbituric acid and pH) and microbiological analyses (TVC- total viable count). The results indicated that the total phenolic compounds of plant extracts were determined as 38.13 mg GAE/g, 81.85 mg GAE/g, and 21.08 mg GAE/g for thyme, rosemary, and basil, respectively. The shelf life of raw fish balls was found as eight months for the control and basil groups and ten months for rosemary and thyme groups. 0.05% basil extract gave the fishball an intense taste and odor and a bitter taste. TVB-N, TBA, FFA, PV, and pH values remained lower than the acceptability limits during the storage period in all groups. The plant extracts, especially rosemary and thyme, were effective for inhibiting bacterial growth and the values of biochemical parameters. Therefore, it was concluded that natural extracts could be added to fish products as an antioxidant to prolong the shelf life of fish. [ABSTRACT FROM AUTHOR]

This study aimed to determine the antioxidant activity and chemical composition of essential oils obtained from thyme and rosemary. In our study, the yield for thyme essential oil was determined as 1.34% and the yield for rosemary essential oil was determined as 0.40%. It has been determined that there are components such as carvacrol, gamma-terpinen, thymol, terpineol, alpha-pinene, borneol, linalol in thyme essential oil. The most important components of rosemary essential oil are components such as eucalyptol, camphor, carvacrol, endo-borneol, delta-3-karen, terpineol, bornyl-acetate, o-simene. In addition, the total phenol content, antioxidant activity (DPPH) and p-anisidine values of essential oils were determined. Total phenol contents and DPPH IC50 values were found to be 18.72±0.37 mg GAE/g DW and 4.74 µg extract/mL for thyme, whereas 12.11±0.29 mg GAE/g DW and 2.75 µg extract/mL for rosemary. Our study also showed good antioxidant activity of thyme and rosemary essential oils. As a result, these properties show that thyme and rosemary essential oils have a very strong potential applicability as antioxidant agents for the food and pharmaceutical industries.

Food manufacturers have focused on the production of minimally processed foods as an alternative to the use of preservatives and traditional heat treatments in order to protect quality and extend shelf life. Therefore, the use of natural antimicrobials is preferred. Oleoresins which are rich in natural antimicrobials, are a mixture of essential oil and resin extracted from plants. In this study, the tomato soup was enriched with thyme oleoresin, and the minimum inhibition concentration value of oleoresin on the target microorganism, Bacillus coagulans, at heat treatment of tomato based product was determined. The changes on the physical and chemical quality characteristics of the product were determined. The D90 value of the B. coagulans in tomato soup was found as 3.67 min. while this value was found as 2.58 min. for the bacteria in the soup which contains thyme oleoresin. Bu çalışmada, biberiye ve kekik yağı ekstraktları kullanılarak elde edilen alabalık (Oncorhynchus mykiss) marinatlarının depolanması sırasında meydana gelen kimyasal değişimler araştırılmış ve kullanılan bitkisel yağ ekstraktlarının ürünün yağ asidi kompozisyonu üzerine etkisi belirlenmeye çalışılmıştır. Fileto haline getirilmiş balıklar üç farklı formülasyon kullanılarak (M grubu: %4 asetik asit-%10 NaCl, B grubu: %4 asetik asit-%10 NaCl-%0,1 biberiye yağı ekstraktı, K grubu: %4 asetik asit-%10 NaCl- %0,1 kekik yağı ekstraktı) marine edilmiş, olgunlaşma aşamasından sonra vakum paketlenerek +4°C’ deki buzdolabı koşullarında depolanmışlardır. Numunelerin 0. gün, 1., 2., 3., 4. ve 5. ayın sonunda kimyasal analizleri (pH, TBA, Peroksit Sayısı, TVB-N, Serbest yağ asitleri, yağ asitleri kompozisyonu) yapılmıştır. Deneysel örnekler TVB-N açısından değerlendirildiğinde depolamanın 3. ayında K grubu örneklerinde 14,12 mg/100 g, B grubu örneklerinde 19,2 mg/100 g ve M grubu örneklerinde ise 28,9 mg/100 g olarak tespit edilmiştir. M grubu örnekleri TBA sayısı bakımından değerlendirildiğinde muhafazanın 3. ayında 5,84 mg MDA/ kg olup, diğer gruplar ile istatistiki fark önemli bulunmuştur. Kontrol grubu (M grubu) örneklerinin 4 ve 5. aylarda duyusal olarak bozulduğu için analizleri yapılamamıştır. Kontrol grubu ve K grubu örneklerinde marinasyon işlemi sonrasında, muhafaza periyodu boyunca EPA ve DHA değerlerinde düşüş şekillenmiş olup, istatistikî açıdan fark önemli bulunmuştur. Biberiye ilave edilen gruba ait örneklerin yağ asidi değerleri incelendiğinde, kaproik, miristik ve palmitik asit miktarları marinasyon işleminden sonra artmıştır ve fark istatistiki açıdan önemli bulunmuştur.

The aim of this study is to investigate the socio-economic structure of the farms producing thyme in Denizli province and to determine their problems. We specifically selected Denizli Province which have 92.49% of total oregano acreage and 87.55% of the production in Turkey. Data were obtained by survey method face-to-face interviewing with 58 farmers producing thyme. The data-set belonged to 2019 production year. The average age of the thyme farmers was 51.47 years, the average number of their family members was 4.60 people per family, their average plant production experience 25.57 years, and their thyme production experience was 17.05 years. When the production pattern of the stakeholders is examined; the total cultivation area was 66.01 da and 81.69% of it is cultivated for thyme production. As the main problems encountered in thyme production and marketing stages were weed management, the absence of alternative buyers no other than the merchant who decide the price in the market. In addition, a SWOT analysis was conducted to determine the strengths and weaknesses of the thyme sector. [ABSTRACT FROM AUTHOR]

The aim of the present study was to investigate the usage of thyme leaves and its effects on performance, some egg quality traits and humoral immunity in laying hens. A total of 108 laying hens aged 36 weeks were randomly assigned to three dietary groups for 16 weeks. Dried and ground (Thymus vulgaris L.) thyme leaves were used at the level of 0, 1 and 2% in the diets of groups. Feed intake, body weight, egg production, egg weight and egg quality characteristics were not affected by dietary thyme supplementation. Thyme supplementation at 2% resulted in beneficial effects in laying hens due to the lowering of yolk cholesterol and total saturated fatty acids concentration while increasing omega-3 fatty acids. Furthermore, the yolk malondialdehyde, blood serum cholesterol and triglycerides levels were reduced and antibody titers against SRBC were increased with 2% thyme supplementation. In conclusion, the results showed the antioxidative and hypolipidemic effects of thyme supplementation in laying hens along with improved humoral immune response without any negative effects on performance and egg quality characteristics. Salmonella enterica serovar Enteritidis infections are among the leading causes of human foodborne illness mainly due to the consumption of contaminated poultry meat and eggs. Therefore, the aim of this study was to investigate the antimicrobial effects of essential oils (EOs) derived from Thymus vulgaris (thyme), Cinnamomum zeylanicum (cinnamon) and Zingiber officinale (ginger) on these infections. These EOs were added to Salmonella enterica ser. Enteritidis cultures in 96-well microplates in a sealed pouch and incubated at 37°C for 24 h minimum inhibitory concentration was measured to determine their antimicrobial effects. Ciprofloxacin (1 mg/mL) added to the culture medium served as the positive control to test the antibacterial effect. In this in vitro cell culture study, the cytotoxic effects of all EOs on healthy fibroblasts were investigated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium-bromide cell viability test. Cinnamon EO showed antibacterial effects at all concentrations, whereas ginger EO was effective only in combination with thyme EO at 50% concentration. Further, thyme EO was found to be bactericidal at 50% concentration and bacteriostatic at 25% concentration. None of the EOs were cytotoxic to fibroblasts. In conclusion, cinnamon EO is highly effective and safe for the treatment of Salmonella enterica ser. Enteritidis infections, followed by thyme EO. [ABSTRACT FROM AUTHOR]

Bu tez çalışması Batman Üniversitesi Bilimsel Araştırma Projeleri Koordinatörlüğü tarafından BTÜBAP-2022-YL-08 nolu proje ile desteklenmiştir., Bu çalışmada, siyah sarımsak ile kekiğin zatürre patojen bakterileri üzerine antimikrobiyal ve antioksidatif sinerjik etkileri araştırılmıştır. Çalışmamızda su ve etil alkol ekstrelerinin antimikrobiyal aktivitelerini belirmek için agar kuyu difüzyon, minimum inhibisyon konsantrasyonu (MIC) ve minimum bakterisidal konsantrasyonu (MBC) testleri yapılmıştır. Bu şekilde toplam fenolik, flavonoid, ve antioksidan kapasite (DPPH-2,2-diphenyl-1-picrylhydrazyl, CUPRAC-Cupric Reducing Antioxidant Capacity, FRAP-Fluorescence recovery after photobleaching) belirlenmiştir. Agar kuyu difüzyon testinde beyaz sarımsağın (BS) su ekstraktında (SE) en yüksek etki gösterdiği bakteri Staphylococcus aureus (34.64 mm zon çapı) iken, BS etil alkol(EE) ekstraktı uygulanan bakterilerde en yüksek Pseudomonas aeruginosa ve Klebsiella pneumoniae (30.48mm zon çapı) olduğu görülmüştür. Siyah sarımsak su ekstraktı (SSSE) uygulanan bakterilerde sadece Staphylococcus aureus (9.44 mm zon çapı) üzerinde etkili olurken, siyah sarımsak etil alkol ekstraktının (SSEE’nin) en etkili olduğu bakteri Pseudomonas aeruginosa (18.17mm zon çapı aralığı) olduğu belirlenmiştir. Kekiğin su ekstraktının (KSE) en etkili olduğu bakteri Klebsiella pneumoniae (23.50mm zon çapı) iken, etil alkol ekstraktının (KEE) en etkili olduğu bakteri ise Pseudomonas aeruginosa (20.62 mm zon çapı) olarak tespit edilmiştir. BS+KSE en etkili olduğu bakteri Klebsiella pneumoniae (22.34mm zon çapı) iken, BS+KEE’nin (23.79mm zon çapı) en etkili olduğu bakteri yine Klebsiella pneumoniae olduğu verisine ulaşılmıştır. SS+KSE’nin sadece Pseudomonas aeruginosa bakterisine etkili (8.52 mm zon çapı) iken, SS+KEE’nin en etkili olduğu bakteri Acinetobacter baumannii (38.50 mm zon çapı) olarak tespit edilmiştir. MBC testi verilerine göre, BSSE (0.012 mg/mL) tüm bakteri izolatlarında etkili iken, MIC testi için en etkili olduğu bakteri Staphylococcus aureus (0.094 mg/mL) olarak bulunmuştur. MBC testinin BSEE (0.012 mg/mL) Pseudomonas aeruginosa hariç diğer tüm bakterilerde etki gösterirken, MIC BSEE’nin (0.023 mg/mL) en etkili olduğu bakteri Staphylococcus aureus bilgisi elde edilmiştir. MBC testinin SSSE’de (0,188 mg/mL) en etkili olduğu bakteri Pseudomonas aeruginosa iken, MIC SSSE’de (0.750 mg/mL) sadece Klebsiella pneumoniae bakterisine etki etmiştir. MBC testinin SSEE (0.012 mg/mL) tüm bakteri izolatlarında etkili iken, MIC SSEE’nin (0.047 mg/mL) en etkili olduğu bakteri Staphylococcus aureus olarak bulunmuştur. MBC testinin KSE‘de (0.375 mg/mL) en etkili olduğu bakteriler Pseudomonas aeruginosa, Staphylococcus aureus ve Acinetobacter baumannii iken, MIC KEE’nin hiçbir bakteri izolatında etkili olmadığı görülmüştür. MBC testinin KEE’de (0.375 mg/mL) tüm bakterilerde etkili iken, MIC KEE’nin (0.023 mg/mL) en etkili olduğu bakteri Staphylococcus aureus olduğu verisine ulaşılmıştır. MBC testinin BS+KSE (0.012 mg/mL) en etkili olduğu bakteriler Klebsiella pneumoniae ve Staphylococcus aureus iken, MIC BS+KSE (0.375 mg/mL) en etkili olduğu bakteri Staphylococcus aureus tespiti yapılmıştır. Biyokimyasal testlerde araştırılan ekstraktlar arasında, en yüksek fenolik madde miktarı değerleri KSE 1.184 mgGAE/mL, en yüksek flavonoid madde miktarı değerleri KSE 0.129 mg rutin eşdeğeri/mL olduğu görülmüştür. Antioksidan testlerde ise, en yüksek DPPH radikal giderme aktivitesi değerleri BS+KEE kombinasyonunda 0.198 mg Troloks eşdeğeri/mL, en yüksek CUPRAC aktivitesi değerleri KSE 4.318 mg Troloks eşdeğeri/mL ve en yüksek FRAP aktivitesi değerleri KSE 2.633 mg Troloks eşdeğeri/mL olarak tespit edilmiştir. Yapılan araştırma sonucunda, zengin biyokimyasal ve antioksidatif özellikleri belirlenen tıbbi bitkilerin, yalnız ve birlikte kullanımının patojen pnömoni izolatlarına karşı antimikrobiyal özellikleri yorumlanmıştır. Bu tıbbi bitkilerin pnömoni kökenli hastalarda kullanılan ilaçları destekleyici olarak kullanılması faydalı olabileceği kanısına varılmıştır., In this study, antimicrobial and antioxidative synergistic effects of black garlic and thyme on pneumonia pathogenic bacteria were investigated. In our study, agar well diffusion, minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) tests were performed to determine the antimicrobial activities of water and ethyl alcohol extracts. In this way, total phenolic, flavonoid, and antioxidant capacity (DPPH-2,2-diphenyl-1-picrylhydrazyl, CUPRAC (Cupric Reducing Antioxidant Capacity), FRAP-(Fluorescence recovery after photobleaching) were determined. Staphylococcus aureus (34.64 mm zone diameter) was the bacteria in which white garlic (WG) showed the highest effect in water extract (WE) in the agar well diffusion test, while Pseudomonas aeruginosa and Klebsiella pneumoniae (30.48 mm zone diameter) had the highest effect in bacteria applied WG ethyl alcohol extract (EE) has been found. While black garlic water extract (BGWE) was effective only on Staphylococcus aureus (9.44 mm zone diameter) in bacteria applied, it was determined that black garlic ethyl alcohol extract (BGEE) was the most effective bacteria Pseudomonas aeruginosa (18.17 mm zone diameter range). The most effective bacterium was Klebsiella pneumoniae (23.50 mm zone diameter), while water extract (TWE) of thyme was most effective, while Pseudomonas aeruginosa (20.62 mm zone diameter) was determined to be the most effective bacterium by ethyl alcohol extract (TEE). While the bacteria that WG+TWE was most effective was Klebsiella pneumoniae (22.34mm zone diameter), the bacteria that WG+TEE (23.79mm zone diameter) was most effective was again Klebsiella pneumoniae. While BG+TWE was effective only against Pseudomonas aeruginosa bacteria (8.52 mm zone diameter), the bacteria that BG+TEE was most effective was Acinetobacter baumannii (38.50 mm zone diameter). According to MBC test data, WGWE (0.012 mg/mL) was effective in all bacterial isolates, while Staphylococcus aureus (0.094 mg/mL) was found to be the most effective bacteria for MIC test. While MBC test was effective on all bacteria except WGEE (0.012 mg/mL) Pseudomonas aeruginosa, Staphylococcus aureus information was obtained for which MIC WGEE (0.023 mg/mL) was the most effective. While Pseudomonas aeruginosa was the most effective bacteria in MBC test in BGWE (0.188 mg/mL), it only affected Klebsiella pneumoniae bacteria in MIC BGWE (0.750 mg/mL). While BGEE (0.012 mg/mL) of the MBC test was effective in all bacterial isolates, the bacteria that MIC BGEE (0.047 mg/mL) was most effective was found to be Staphylococcus aureus. While the bacteria that the MBC test was most effective in TWE (0.375 mg/mL) were Pseudomonas aeruginosa, Staphylococcus aureus and Acinetobacter baumannii, MIC TEE was not found to be effective in any bacterial isolate. While the MBC test was effective in TEE (0.375 mg/mL) in all bacteria, it was found that Staphylococcus aureus was the most effective bacteria in MIC TEE (0.023 mg/mL). While the bacteria Klebsiella pneumoniae and Staphylococcus aureus were the most effective in MBC test with WG+TWE (0.012 mg/mL), the bacteria Staphylococcus aureus was detected for which MIC WG+TWE (0.375 mg/mL) was the most effective. While the bacteria that MBC test WG+TEE (0.012 mg/mL) is the most effective are Klebsiella pneumoniae and Staphylococcus aureus, MIC WG+TEE (0.023 mg/mL) is the most effective bacteria Staphylococcus aureus and Acinetobacter baumannii. Staphylococcus aureus was the most effective bacteria in MBC test BG+TWE (0.012 mg/mL), while MIC BG+TWE (0.188 mg/mL) was effective only on Staphylococcus aureus bacteria. Staphylococcus aureus and Acinetobacter baumannii were found to be the most effective bacteria in MBC test BG+TEE (0.012 mg/mL), while Pseudomonas aeruginosa and Staphylococcus aureus were found to be the most effective bacteria with MIC BG+TEE (0.023 mg/mL). Among the extracts investigated in biochemical tests, the highest phenolic content values were found to be TWE 1.184 mg GAE/mL, and the highest flavonoid content values were TWE 0.129 mg routine equivalent/mL. In antioxidant tests, the highest DPPH radical scavenging activity values were determined as 0.198 mg Trolox equivalent/mL in the WG+TEE combination, the highest CUPRAC activity values were found to be 4.318 mg Trolox equivalent/mL for TWE and the highest FRAP activity values for TWE were 2.633 mg Trolox equivalent/mL. . As a result of the research, the antimicrobial properties of medicinal plants, whose rich biochemical and antioxidant properties were determined, against pathogen pneumonia isolates were interpreted when used alone or together. It has been concluded that the use of these medicinal plants as a supplement to the drugs used in patients with pneumonia may be beneficial.

29. The Effects of Using Oregano and L-Carnitine on Second Phase Laying Performance Parameters of Force Molting Programs in Laying Hens.

This study was carried out to use Oregano leaf and L-carnitine as an alternative feed additives in alfalfa flour or low Sodium-Calcium (Na-Ca) diet and to determine their effect on second phase laying period after force molting in commercial laying hens. A total of 1170 birds were divided into 13 groups which were further subdivided into 6 replicates having 15 birds in each. A negative control group (K) which was neither forced molted nor provided with any feed additive substances in their diets Moreover, other three treatment groups (M: Marbel powder, A: Alfalfa, F: Low Ca -Na diet), were forced molted and one withhold feeding group was used for the study; the forced molted groups were supplied L-carnitine (C) (100 ppm) and Oregano leaves (O) (5%) as a feed additive in designed individual diets (MC, AC, FC, MO, AO, FO) alone and in combination (50 ppm; 2.5%) in groups (MCO, ACO, FCO). All hens were feeding with commercial feeds during the second laying phase. The trial was 98 days after 22 days of forcing stage. Performance parameters (except egg quality) were measured during 14 days periods. In the results, eggs yield values were found significantly higher (P<0.01) among the treatment groups over control group. It was concluded that supplementing L-carnitine and Oregano leaves could be used as an alternative feed additives in alfalfa meal during feed withdrawal period and their combination also showed a positive impact on laying performance in laying hens. Breakfast zahter is a traditional food which is prepared by mixing ground roasted nuts and various spices and consumed with olive oil in breakfast in the eastern mediterranean region of our country, especially in Kilis, Gaziantep and Hatay. In this study, it was aimed to develop new breakfast zahter formulas by adding different rates of thyme and menengic fruit. In this direction, 15 different formulations Breakfast Zahter were produced in. In addition, the production of the control sample was carried using the same formulation with the exception of thyme and menengic fruit. The sample (M4 + K4) with 2% menengic paste and 2% thyme plant got the higest score in sensory analysis. Moisture, ash, protein, fat, carbohydrate, raw fiber, phytic acid, antioxidant activity, total phenolic substance and energy values were determined on that sample. Lamiaceae/Labiatae family includes the highest number of commercially used species such as mint, rosemary, thyme, oregano, lavender, marjoram and sage. These species are generally used as flavoring additives in meat dishes, sausage products, seafood, stews, salads, canned foods, sauces and soups. In this study, the species diversity of Lamiaceae family members in Akseki-İbradı-Manavgat districts of Antalya in southern Turkey and their ethnobotanical utilization in the region and in the literatures were presented. 149 taxa (131 at species level) belonging to 27 genera of Lamiaceae were identified in the study area. Of these, 50 taxa (33.56%) are endemic to Turkey. The native people harvest species from wild populations and generally air-dry them under sun or on shadow places. They consume and sell mainly air-dried Origanum onites (Turkish oregano) leaves. They obtain essential oil traditionally by steam distillation from Origanum majorana (white marjoram) due to its high oil yield and efficiency. Herbal treatments are used for cure various diseases such as strong cough, chronic cold, wounds, gastrointestinal disorders and skin problems. Wild-crafting of populations may cause genetic erosion of the species. The sustainability of these species should be maintained by genetic resource conservation programs by in situ and ex situ conservation strategies. [ABSTRACT FROM AUTHOR]

In this study, the effects of some herbal wastes such as daphne (Laurus nobilis), thyme (Origanum onites) and cumin (Cuminum cyminum) on the yield of Pleurotus ostreatus were investigated. While compost formula consisted 43% wheat straw+ 43% poplar dust+ 10% wheat bran+ 4% gyps mixture was used as the control group, 20% to 40% of herbal waste was added to this formula in the experiment groups. Total number of mushroom was counted in both the experiment and the control groups. Then, the harvested mushrooms were weighed and morphological features of caps were measured and recorded. As a result of obtained data, while it was detected that compost contained 20% cumin waste had the highest number of primordium (34), the control group had the lowest number of primordium (21 items). The highest biological yield was detected as 35.8% in the group contained 20% cumin waste. The lowest biological yield was detected in the control group with 24.6%.

The effects of commercial thyme (Origanum vulgare L.), sage (Salvia officinalis L.), garlic (Allium sativum L.), sesame (Sesame indicum L.) , rosemary (Rosmarinus officinalis L., syn. Salvia rosmarinus Spenn.), lemon (Citrus limon (L.) Osbeck) and mustard (Brassica nigra L.) essential oils (Botalife Natural and Aromatic Products Inc., Türkiye) on disease severity were investigated in simultaneous inoculation of Meloidogyne incognita (Kofoid and White, 1919) Chitwood, 1949 and Fusarium oxysporum f.sp. radicis lycopersici (Jarvis & Shoemaker) (FORL) on tomato. Nematicide (Velum®, Fluopyram, Bayer Crop Production Inc., Türkiye) and fungicide (Cebir®, Fludioxonil + Metalaxyl, Hektaş Crop Production Inc., Türkiye) were used as positive controls. The negative control was only plants with nematode and fungus inoculation. The study was set up in a randomized plot design with 5 replications for each essential oil. In simultaneous inoculations, 1000 M. incognita second juvenile larvae/1ml and 3x106 spore/ml FORL were used for each seedling. The essential oil applications were applied to the soil at a dose of 1000 ppm for each pot, one day after the nematode and fungus inoculation. The study was terminated after 60 days, and the evaluation was based on gall, egg mass, and disease severity. Fungal growth and nematode development on roots were found lower in all tested oils applications than in negative control but fungicidal and nematicidal activity varied. Thyme and garlic essential oils had the highest control effect on nematode and fungus with 55.20% in simultaneous inoculation and this effect was higher than only nematicide (38.84%) and only fungicide (33.20%) applications. Sage (38.84%), rosemary (33.28%), and mustard (38.92%) essential oils were found to suppress disease severity higher than sesame (22.16%) and lemon (22.16%). Uçucu yağ uygulamaları, nematod ve fungus inokulasyonundan bir gün sonra her saksı için 1000 ppm dozunda toprağa uygulanmıştır. Çalışma 60 gün sonra sonlandırılmış ve değerlendirme gal, yumurta paketi ve hastalık şiddetine göre yapılmıştır. Köklerde fungus büyümesi ve nematod gelişimi, test edilen tüm yağ uygulamalarında negatif kontrole göre daha düşük bulunmuştur, ancak fungus gelişimi ve nematod aktivitesi değişiklik göstermiştir. Kekik ve sarımsak uçucu yağları eş zamanlı inokulasyonda nematod ve fungus üzerinde %55.20 ile en yüksek kontrol etkisine sahip olmuş ve bu etki sadece nematod (%38.84) ve sadece fungisit (%33.20) uygulamalarından daha yüksek bulunmuştur. Adaçayı (%38.84), biberiye (%33.28) ve hardal (%38.92) esansiyel yağlarının, susam (%22.16) ve limona (%22.16) göre hastalık şiddetini daha fazla baskıladığı bulunmuştur. Kekik ve sarımsak esansiyel yağlarının kök ur nematodu ve FORL hastalık komplekslerini yönetmek için iyi birer alternatif olduğu belirlenmiştir.

36. Effects of supplementation of oregano and rosemary essential oils to broiler rations on performance and meat quality

Bu çalışmada, yemlere katılan kekik (300 ppm), biberiye (300 ppm) uçucu yağlarının tek başlarına ve 300 ppm ile 400 ppm karışımlarının broyler performansı, piliç etinin oksidasyonu ve duyusal özellikleri üzerine etkileri incelenmesi, amaçlanmıştır. Toplam 450 adet Ross 308 erkek broyler civciv kontrol, kekik, biberiye, kekik ve biberiye karışımı-1 ve kekik ve biberiye karışımı-2 olarak beş gruba ayrılmıştır. Her grup, dokuz civcivden oluşan 10 replikasyona ayrılmıştır. Kontrol hariç diğer grupların yemlerine, sırasıyla 300 ppm kekik uçucu yağı, 300 ppm biberiye uçucu yağı, 300 ppm kekik ve biberiye uçucu yağ karışımı (150+150), 400 ppm kekik ve biberiye uçucu yağ karışımı (200+200) katılmıştır. Denemenin 42. gününde kesilen piliçlerin göğüs ve but etlerinde, sırasıyla 7-8 gün depolamadan (+4˚C'de) önce ve sonra, malondialdehit düzeyleri ölçülmüştür. Lezzet panelinde etin duyusal özellikleri incelenmiştir. Denemenin 1., 7., 14., 28., 35. günlerinde canlı ağırlık, canlı ağırlık kazancı, yem tüketimi, ve yemden yararlanma oranı açısından farklar önemsizdir. Denemenin 21. gününde, biberiye uçucu yağ grubunun canlı ağırlık değeri karışım-300 ve karışım-400 grubundan yüksek bulunmuştur. Deneme sonunda yem tüketimi, karkas randımanı ve göğüs ile but etlerinin duyusal özellikleri açısından önemli fark görülmemiştir. 42. günde biberiye uçucu yağı ve karışım-400 gruplarının canlı ağırlık değerleri ile karışım-400 grubunun canlı ağırlık kazancı değeri kontrole göre yüksek bulunmuştur. Karışım-400 grubunun yemden yararlanma oranı değeri kontrole göre iyileşmiştir. Göğüs ve but etlerinin MDA (Malondialdehit) düzeylerinde, depolama (+4C) başlangıcında önemli fark bulunmamıştır. Depolamanın 7-8. günlerinde biberiye-300 ve karışım-400 gruplarının MDA düzeylerinin kontrol göre düşük olduğu saptanmıştır. This study aimed to determine the effect of individual (300 ppm) and combined (300, 400 ppm) oregano and rosemary volatile oil supplementation to broiler performance, oxidation and sensory attributes of broiler meat. A total of 450 Ross 308 male chicks were divided into 5 groups: control, oregano, rosemary, oregano and rosemary mix-1 and oregano and rosemary mix-2, with 9 chicks of 10 replications. Except control, other groups' feeds were supplemented with volatile oils of 300 ppm oregano, 300 ppm rosemary, 300 ppm oregano and rosemary mix (150+150), and 400 ppm oregano and rosemary mix (200+200), respectively. On 42nd day, malondialdehyde levels were measured in breast and thigh muscles of slaughtered chickens, before and after 7-8 day storage at +4˚C. Meat sensory attributes were investigated in a tasting panel. No significant difference in live weight, live weight gain, feed consumption, and feed conversion ratio on days 1, 7, 14, 28, and 35 was observed. On 21 st day, rosemary group's live weight was higher than mix-300 and mix-400 groups. At trial's end, no significant difference was determined in feed consumption, carcass yield, sensory attributes of breast and thigh muscles. On 42nd day, rosemary and mix-400 groups' live weight, and mix-400 group's live weight gain was found higher than control. Mix-400 group's feed conversion ratio was better than control. There was no significant difference between breast and thigh muscles' malondialdehyde levels before +4˚C storage. Malondialdehyde levels were lower in rosemary and mix-400 groups compared to control on the 7-8 days of storage.

37. Increasing the active anti-bacterial properties of wood material with laurel and thyme extracts

Wooden products come into our lives more and more day by day, but they also bring some question marks about whether the wooden surfaces that are in contact with are hygienically safe. Two different anti-bacterial properties of wood material, passive and active, can be mentioned. The passive antibacterial property is due to the inherent porosity and hygroscopicity of the wood. However, the active anti-bacterial property of wood is related to the extractive substances in the wood. The aim of this study is to investigate the antibacterial properties of woods treated with laurel (Laurus nobilis) and thyme (Origanum onites) extracts for use in hygienically sensitive areas by direct diffusion method. Ten different bacterial strains were used to perform the experiments. The wooden discs used in the experiments were sterilized by autoclave after they were prepared from poplar (Populus tremula) wood with low active anti-bacterial properties. Bacterial strains prepared with 0.5 McFarland concentration were inoculated on Mueller-Hinton agar Petri dishes by swabbing method. The wood discs were then placed directly on the inoculated agar surface. After 24 hours of incubation at 37 °C, the zone of inhibition on the agar around the discs was measured in mm. As a result, it was determined that thyme extracts applied to poplar wood reduced bacterial growth. Thus, it has been determined that the use of wood species with low antibacterial properties by treating them with thyme extracts will give positive results in hygienically sensitive areas., Ahşap ürünler gün geçtikçe hayatımıza daha çok girmekle birlikte temas edilen ahşap yüzeylerin hijyenik olarak güvenli olup olmadığı ile ilgili bazı soru işaretlerini de beraberinde getirmektedir. Ahşap malzemenin pasif ve aktif olmak üzere iki farklı anti-bakteriyel özelliğinden bahsedilebilir. Burada, pasif anti-bakteriyel özellik ahşabın doğası gereği olan porozif yapısı ve higroskopisiteden kaynaklanmaktadır. Ancak, ahşabın aktif anti-bakteriyel özelliği ise odun içeriğinde bulunan ekstraktif maddeler ile ilişkilidir. Bu çalışmanın amacı, hijyenik açıdan hassas alanlarda kullanılmak üzere defne (Laurus nobilis) ve kekik (Origanum onites) ekstraktları ile muamele edilmiş odunların anti-bakteriyel özelliklerinin direk difüzyon yöntemi ile incelenmesidir. Deneyleri gerçekleştirmek için 10 farklı bakteri suşu kullanılmıştır. Deneylerde kullanılan ahşap diskler aktif anti-bakteriyel özellikleri düşük kavak (Populus tremula) ağacından hazırlandıktan sonra otoklav ile sterilize edilmiştir. Bakteri suşları 0,5 McFarland’da hazırlanarak sürüntü sürme yöntemiyle Mueller-Hinton agar petri kaplarına aşılanmıştır. Daha sonra odun diskleri doğrudan aşılanmış agar üzerine yerleştirilmiştir. 37 °C’de 24 saat inkübasyondan sonra, disklerin etrafındaki agar üzerindeki inhibisyon bölgesi mm olarak not edilmiştir. Elde edilen sonuçlara göre, kavak odununa uygulanan kekik ekstraktlarının bakteriyel büyümeyi azalttığı tespit edilmiştir. Böylece, anti-bakteriyel özelliği düşük olan ağaç türlerinin kekik ekstraktları ile muamele edilerek hijyenik açıdan hassas alanlarda kullanımının olumlu sonuçlar vereceği tespit edilmiştir.

This study was carried out in vitro and in vivo to determine the therapeutic effects of Thymus kotschyanus and Thymus collinus in the territory of Nakhchivan Autonomous Republic against the gastrointestinal parasites in sheep. Essential oils and extracts of these plants were used as antihelminthic preparation in the research. An antihelminthic effect was determined against helminth species (Trichostrongylus axei, Nematodirus abnormalis, Trichocephalus ovis, Haemonchus contortus) collected from the digestive system in in vitro conditions. Furthermore, in vivo conditions, the antihelminthic effect against gastrointestinal helminthes in naturally infected sheep was investigated. Comparing to the control group in the in vitro conditions, helminthes were killed in a shorter time in the experimental groups. In vivo conditions, the antihelminthic effect of Th. kotschyanus essential oil was 88.1% and 82.9% of Th. collinus essential oil. The antihelminthic effects of the extracts prepared from these plants were 69.1% and 63.5%, respectively. The result obtained from Th. kotschyanus volatile oil preparation was 21.6%, the result of Th. collinus volatile oil preparation was 23.2% which was more specific than the first one. The results of the study show that both herbal oils and extracts can be used in the treatment of gastrointestinal parasites of sheep by preparing antihelminthic preparations. [ABSTRACT FROM AUTHOR]

Satureja cilicica P.H. Davis is an endemic species of Lamiaceae, distributed in Turkey. In this study, the essential oil of S. cilicica was obtained by hydrodistillation from the aerial parts. The essential oil was analyzed by GC/MS and the main constituents are identified. Radical scavenging capacities of oil obtained from S. cilicica were determined by using DPPH and ABTS radical scavenging assays. In addition, ferric-reducing power of the essential oil was detected. Cytotoxic activity of the essential oil was determined by MTT assay after 24 h treatment of MCF-7. The major components of the essential oil are p-cymene (17.68%), carvacrol (14.02%), γ-terpinene (11.23 %) and thymol (8.76%). Radical scavenging capacities of the essential oil were measured as 3.28±0.02 and 238.15±3.59 mg Trolox equivalent per milliliter of oil in term of DPPH and ABTS radical scavenging, respectively. Ferric-reducing power of essential oil was found 39.76±3.66 mg Trolox equivalent per milliliter of essential oil. Growth inhibition effect of essential oil against MCF-7 cancer cell is established. According to the results, essential oil of S. cilicica exhibit low cytotoxic activity, with IC50 values, with IC50 value of 268 μg/mL. 42. Effect of Thyme and Garlic Aromatic Waters on Microbiological Properties of Raw Milk Cheese.

In the present study, it was aimed to investigate the effect of brine solutions containing thyme and garlic extracts on physicochemical, microbiological and textural properties of Turkish white cheese made from raw milk during ripening. For this aim, garlic aromatic water (GAW), thyme hydrosol (TH) or their mixture (1:1 v/v) were incorporated into the brine with different salt concentrations (10%, 13% and 16%) at the ratio of 10% and used in the cheese ripening for 90 days. Addition of TH and GAW into the brine caused higher acidity and lower pH values while increase in salt level resulted in higher dry matter (DM) of cheese. Counts of total mesophilic aerobic bacteria (TMAB), yeast-mold (YM), lactoccocci and lactobacilli were fluctuatingly influenced from brine combinations while coagulase positive staphylococci was completely inhibited during the ripening. In general, TH, GAW or their mixture increased hardness, gumminess and chewiness of the cheese at the 1st day while cohesiveness, resilience and springiness values were not significantly (P>0.05) affected. This study aimed to determine the antioxidant activity and chemical composition of essential oils obtained from thyme and rosemary. In our study, the yield for thyme essential oil was determined as 1.34% and the yield for rosemary essential oil was determined as 0.40%. It has been determined that there are components such as carvacrol, gamma-terpinen, thymol, terpineol, alpha-pinene, borneol, linalol in thyme essential oil. The most important components of rosemary essential oil are components such as eucalyptol, camphor, carvacrol, endo-borneol, delta-3-karen, terpineol, bornylacetate, o-simene. In addition, the total phenol content, antioxidant activity (DPPH) and p-anisidine values of essential oils were determined. Total phenol contents and DPPH IC50 values were found to be 18.72±0.37 mg GAE/g DW and 4.74 µg extract/mL for thyme, whereas 12.11±0.29 mg GAE/g DW and 2.75 µg extract/mL for rosemary. Our study also showed good antioxidant activity of thyme and rosemary essential oils. As a result, these properties show that thyme and rosemary essential oils have a very strong potential applicability as antioxidant agents for the food and pharmaceutical industries. Bu çalışmada, kekik ve biberiyeden elde edilen bu uçucu yağların antioksidan aktivitesinin ve kimyasal bileşiminin belirlenmesi amaçlamıştır. Çalışmamızda kekik uçucu yağı için verim %1.34 ve biberiye uçucu yağı için verim %0.40 olarak belirlenmiştir. Kekik uçucu yağında karvakrol, gamma terpinen, timol, terpineol, alfa-pinen, borneol, linalol gibi bileşenler mevcut olduğu belirlenmiştir. Biberiye uçucu yağının en önemli bileşenleri ise ökaliptol, kamfor, karvakrol, endo-borneol, delta-3- karen, terpineol, bornil-asetat, o-simen olarak tespit edilmiştir. Ayrıca, uçucu yağlara ait toplam fenol içeriği, serbest radikalleri yakalama aktivitesi (DPPH) ve p-anisidin değerleri belirlenmiştir. Toplam fenol içerikleri ve DPPH IC50 değerleri, kekik için sırasıyla 18,72±0,37 mg GAE/g DW ve 4,74 µg ekstrakt/mL bulunurken biberiye için 12,11±0,29 mg GAE/g DW ve 2,75 µg ekstrakt/mL olarak hesaplanmıştır. Sonuç olarak elde edilen bulgular, kekik ve biberiye uçucu yağlarının iyi antioksidan aktivite değerine sahip olduğunu göstermektedir. Bu özellikler kekik ve biberiye uçucu yağlarını gıda ve ilaç endüstrileri için antioksidan ajanlar olarak çok güçlü bir potansiyel sahip olduğunu göstermektedir.

In the study, weed species, densities and frequency of thyme (Origanum onites L.) located in the production areas in Manisa Province were investigated. Surveys; It was carried out in two different periods in 705 decare area in 21 fields randomly selected in 2013. In weed counts, 1/4 m-2 frames were thrown according to the size of the field and counts of weeds were made on the basis of species. From the data obtained, weed density and frequency of occurrence in m-2 were determined. As a result of the surveys, 50 different weed species belonging to 26 families were identified. Among these species, it was determined that two of them are parasite (Cuscuta campestris L., Orabanche gracilis sm.) Species, four of them are narrow-leaved, others are broad-leaved weed species. It has been determined that among the broad-leaved weeds, the most intense one is Convolvulus arvensis L. (1.15 plant m-2) and it is followed consecutively by Lactuca serriola L. (1.03 plant m-2), Lactuca saligna(0.75 plant m-2), Soncus asper (0.62 plant m-2), Portulaca oleracea L. (0.58 plant m-2); In narrow leaf weeds, themost intense one is Cyperus rotundus L. (2.67 plant m-2) and it is followed consecutively by Cynedon dactylon L. (0.39 plant m-2) and Sorghum halapense L. (0.13 plant m-2). On the other hand, C. arvensis (99%), L. serriola (63%), C. rotundus (49%), S. asper (36%) were among the most common weeds., Çalışmada, Manisa İli kekik (Origanum spp.) üretim alanlarında bulunan yabancı ot türleri,yoğunlukları ve rastlanma sıklıkları araştırılmıştır. Surveyler; 2013 yılında,tesadüfü olarak seçilen 21 tarlada 705 dekar alanda, iki farklı dönemdeyürütülmüştür. Yabancı ot sayımlarında tarla büyüklüğüne göre, 1/4 m2’likçerçeveler atılarak yabancı otların tür bazında sayımları gerçekleştirilmiştir.Sayımlarda geniş yapraklı yabancı otlar tüm bitki olarak, dar yapraklıların isesapları sayılarak değerlendirilmiştir. Elde edilen verilerden m2’dekiyabancı ot yoğunluğu ve rastlanma sıklığı belirlenmiştir. Surveyler sonucunda 25 familyaya ait 46 farklı yabancı ot türü saptanmıştır. Söz konusu türler içerisindeikisinin parazit (Cuscuta campestrisL., Orabanche gracilis sm.) tür,dördünün dar yapraklı, diğerlerinin geniş yapraklı yabancı ot türlerindenolduğu belirlenmiştir. Genişyapraklı yabancı otlar içerisinde en yoğun Convolvulus arvensis L.(1,15 adet/m2), bunu sırasıylaLactuca serriola (1,03 adet/m2), Lactuca saligna L. (0,75 adet/m2), Soncus asper (0,62 adet/m2), Portulaca oleracea L. (0,58 adet/m2)türlerinin takip ettiği; dar yapraklı yabancı otlarda ise en yoğun Cyperus rotundus L. (2,67 adet/m2)türünün olduğu, bunu Cynedon dactylon(0,39 adet/m2) ve Sorghumhalapense L., (0,13 adet/m2)’nin takip ettiği belirlenmiştir.Yabancı otlardan en sık Convolvulusarvensis L. Bu tez çalışmasının amacı, Origanum onites L. (İzmir kekiği) bitkisinin elit çeşitlerinde in vitro klonal mikroçoğaltım prosedürü için başarılı bir protokol oluşturmaktır. Ege Üniversitesi Ziraat Fakültesi Tarla Bitkileri Bölümü tarafından tescil ettirilen Origanum onites L. türüne ait 2 çeşidin (Ceylan 2002 ve Tayşi 2002) nod eksplantları, ön yıkama ve farklı sterilizasyon yöntemlerine tabi tutulmuşlardır. Ceylan 2002 çeşidinde en yüksek sterilizasyon başarısı (%100); 1 dk boyunca %70’lik etil alkol uygulaması ardından 15 dk boyunca %20’lik çamaşır suyu uygulaması, 1 dk boyunca %70’lik etil alkol uygulaması ardından 20 dk boyunca %20’lik çamaşır suyu uygulaması ve 1 dk boyunca %70’lik etil alkol uygulaması ardından 20 dk boyunca %30’luk çamaşır suyu uygulamasından elde edilmiştir. Tayşi 2002 çeşidinde ise en yüksek sterilizasyon başarısına (%90); 1 dk boyunca %70’lik etil alkol uygulaması ardından 20 dk boyunca %30’luk çamaşır suyu uygulamasında ulaşılmıştır. Farklı konsantrasyon ve kombinasyonlarda BAP, IBA ve NAA ile desteklenmiş MS besin ortamlarının sürgün rejenerasyonu, ortalama sürgün uzunluğu ve ortalama yaprak sayısı üzerinde etkisinin belirlenmesi amacıyla steril edilen nod eksplantları 20 farklı besin ortamında kültüre alınmıştır. Ceylan 2002 çeşidinde en yüksek sürgün rejenerasyon yüzdesi (%13,33), en yüksek ortalama sürgün uzunluğu (0,12 cm) ve en yüksek ortalama yaprak sayısı (0,87 adet), 2,0 mg/L BAP ile desteklenmiş 0,3 g/L aktif karbon içeren MS besin ortamında elde edilmiştir. Tayşi 2002 çeşidinde herhangi bir sürgün rejenerasyonu elde edilememiştir. Ceylan 2002 çeşidinde altkültür sayısının çoğaltım katsayısına, ortalama sürgün uzunluğuna, ortalama yaprak sayısına, ortalama kök sayısı ve uzunluğuna ve nod başına elde edilen sürgün sayısına etkisini değerlendirmek amacıyla, sürgün rejenerasyon denemelerinden elde edilen eksplantlar 4 haftalık aralıklarla 9 kez altkültüre alınmıştır. En yüksek çoğaltım katsayısı (3,40), 4. altkültürde elde edilmiştir. Eksplant başına elde edilen ortalama sürgün uzunluğu (3,82 cm) ve yaprak sayısı (13,85 adet) 3. altkültürde en yüksek düzeye ulaşmıştır. Eksplant başına elde edilen en yüksek ortalama kök sayısı (0,81 adet) ve kök uzunluğu (1,99 cm) değerleri 6. altkültürde elde edilmiştir. En yüksek nod başına elde edilen sürgün sayısı (0,47 adet) değerine 8. altkültürde ulaşılmıştır. Ceylan 2002 çeşidine ait rejenere sürgünlerde farklı konsantrasyonlarda IBA ve aktif karbon içeren MS ve ½ MS temelli besin ortamlarının kök rejenerasyonuna, eksplant başına elde edilen ortalama kök sayısına ve ortalama kök uzunluğuna etkisini değerlendirmek amacıyla, eksplantlar 16 farklı besin ortamında kültüre alınmıştır. En yüksek kök rejenerasyon yüzdesi (%100), 0,5 mg/L IBA ile desteklenmiş 1,0 g/L aktif karbon içeren MS besin ortamında, 1,0 mg/L IBA ile desteklenmiş 1,0 g/L aktif karbon içeren ½ MS besin ortamında ve 2,0 mg/L IBA ile desteklenmiş 1,0 g/L aktif karbon içeren ½ MS besin ortamında kültüre alınan rejenere sürgünlerden elde edilmiştir. Ceylan 2002 çeşidine ait rejenere sürgünlerde aktif karbon miktarının gövde kalınlığı ve sürgün uzunluğu üzerine etkisini değerlendirmek amacıyla, eksplantlar 16 farklı besin ortamında kültüre alınmıştır. En yüksek ortalama gövde kalınlığı değeri (2,02 mm) ve en yüksek ortalama sürgün uzunluğu (10,02 cm) değeri 1,0 g/L aktif karbon içeren besin ortamlarında elde edilmiştir. Köklendirilmiş bitkicikler, %16,25 başarı yüzdesi ile aklimatize edilmiştir. Ceylan 2002 çeşidine ait rejenere sürgünlerde farklı konsantrasyonlarda IBA ve 1,0 g/L aktif karbon içeren besin ortamlarının uçucu yağ miktarı ve bileşenleri üzerine etkisini değerlendirmek amacıyla, iyi gelişmiş köklü bitkiciklerin uçucu yağ analizi GC-MSD cihazı ile gerçekleştirilmiştir. En yüksek uçucu yağ oranı (%2,00) değeri, 2,0 mg/L IBA ile desteklenmiş MS besin ortamlarından elde edilen rejenere sürgünlerde gözlemlenmiştir. En yüksek timol oranı (%65,33) değerine, 0,5 mg/L IBA ile desteklenmiş ½ MS besin ortamlarından elde edilen rejenere sürgünlerde; en yüksek karvakrol oranı (%6,82) değerine ise 2,0 mg/L IBA ile desteklenmiş MS besin ortamlarından elde edilen rejenere sürgünlerde ulaşılmıştır., This thesis study aims to establish a successful protocol for in vitro clonal micropropagation procedure in elite cultivars of Origanum onites L. (İzmir oregano). Node explants of 2 cultivars (Ceylan 2002 and Tayşi 2002) belonging to Origanum onites L. species registered by Ege University Faculty of Agriculture Department of Field Crops are subjected to pre-washing and different sterilization methods. The highest sterilization success (100%) in Ceylan 2002 cultivar is obtained from the application of 70% ethyl alcohol for 1 minute, followed by the application of 20% bleach for 15 minutes, application of 70% ethyl alcohol for 1 minute, followed by the application of 20% bleach for 20 minutes and application of 70% ethyl alcohol for 1 minute, followed by the application of 30% bleach for 20 minutes. The highest sterilization success (90%) in Tayşi 2002 variety is obtained from the application of 70% ethyl alcohol for 1 minute, followed by the application of 30% bleach for 20 minutes. In order to determine the effect of MS nutrient media supplemented with BAP, IBA and NAA in different concentrations and combinations on shoot regeneration, average shoot length and average leaf number, sterilized node explants are cultured in 20 different nutrient media. In Ceylan 2002 cultivar, the highest shoot regeneration percentage (13,33%), the highest average shoot length (0,12 cm) and the highest average leaf number (0,87), is achieved in MS nutrient medium containing 0,3 g/L activated carbon supplemented with 2,0 mg/L BAP. No shoot regeneration is achieved in Tayşi 2002 cultivar. Explants obtained from shoot regeneration experiments are subcultured 9 times in order to evaluate the effect of subculture number on propagation coefficient, average shoot length, average number of leaves, average root number and length and shoot number per node in Ceylan 2002 cultivar. The highest propagation coefficient (3,40) is obtained in the 4th subculture. Average shoot length (3,82 cm) and number of leaves (13,85) per explant obtained reached the highest level in the 3rd subculture. The highest average root number (0,81) and root length (1,99 cm) per explant are obtained in the 6th subculture. The highest number of shoots per node (0,47) is reached in the 8th subculture. In order to evaluate the effect of MS and ½ MS based nutrient media containing different concentrations of IBA and activated carbon on root regeneration, average root number per explant and average root length in regenerated shoots of Ceylan 2002 cultivar, explants were cultured in 16 different media. Highest percentage of root regeneration (100%) is obtained from regenerated shoots cultured in MS nutrient medium containing 1,0 g/L activated carbon supplemented with 0,5 mg/L IBA, in ½ MS nutrient medium containing 1,0 g/L activated carbon supplemented with 1,0 mg/L IBA and in ½ MS nutrient medium containing 1,0 g/L activated carbon supplemented with 2,0 mg/L IBA. The highest average root number per explant (12,03) value is obtained in MS nutrient medium containing 1,0 g/L activated carbon supplemented with 2,0 mg/L IBA. The highest average root length (2,72 cm) is achieved in MS medium containing 1,0 g/L activated carbon supplemented with 0,5 mg/L IBA. In the experiments carried out to determine the effect of activated carbon concentration on stem thickness and shoot length in regenerated shoots of Ceylan 2002, explants are cultured in 16 different media. The highest average stem thickness value (2,02 mm) and the highest average shoot length (10,02 cm) are obtained in nutrient media containing 1,0 g/L activated carbon. The rooted plantlets are acclimatized with 16,25% success percentage. In order to evaluate the effect of nutrient media containing different concentrations of IBA and 1,0 g/L activated carbon on the amount and components of essential oil in regenerated shoots of Ceylan 2002 variety, essential oil analysis of well-developed rooted plantlets is carried out with GC-MSD device. The highest essential oil content (2.00%) is observed in regenerated shoots obtained from MS nutrient media supplemented with 2,0 mg/L IBA. The highest thymol ratio (65.33%) is found in regenerated shoots obtained from ½ MS nutrient media supplemented with 0,5 mg/L IBA; the highest carvacrol ratio (6.82%) is achieved in regenerated shoots obtained from MS nutrient media supplemented with 2,0 mg/L IBA.

Amaç: Bu tez çalışmasında, Türkiye'nin Doğu ve Güneydoğu Anadolu bölgesinden toplanan ve halk arasında yaygın bir şekilde çay ve baharat olarak kullanılan bazı türlerin Clevenger aparatı ile uçucu yağları hazırlanmıştır. Bu türlere özgü bileşenler piyasadan temin edilerek bu türleri içeren bir GC-MS miktarsal tayin yöntemi geliştirilip valide edilmesi amaçlanmıştır. Ayrıca bu türlerin bu bileşenler bakımından içerikleri belirlenmiştir. Gereç ve Yöntem: α-Pinen, kamfen, α-fellandren, p-simen, limonen, ökaliptol (1,8-sineol), β-osimen, γ-terpinen, linalol, kafur, sitronellal, menton, borneol, mentol, α-terpineol, cis-geraniol, pulegon, L-karvon, linalil asetat, bornil asetat, timol, karvakrol, β-karyofillen bileşenleri ile bir GC-MS yöntemi geliştirilmiş ve doğrulanmıştır. Clevenger aparatı kullanılarak bazı türlerin hidrodestilasyon yöntemi ile uçucu yağları elde edilmiştir. Daha sonra GC-MS ile bu uçucu yağların içerikleri belirlenmiştir. Bulgular: Çalışılan Thymbra türlerinin hepsinde ortak ana bileşenin karvakrol, Salvia ve Mentha türlerinde ökaliptol (1,8-sineol), Origanum türlerinde p-simen ve Lavandula türlerinde linalol olduğu görülmüştür. Thymus pubescens var. crateicola türünün timol, Cyclotrichium origanifolium türünün pulegon, Achillea vermicularis türünün ökaliptol, Teucrium polium türünün β-karyofillen, Zizphora clinopdioides türünün pulegon bileşenleri açısından zengin olduğu tespit edilmiştir. Sonuç: Ülkemizin Doğu ve Güneydoğu bölgelerinde yetişen ve halk arasında yaygın olarak kullanılan Thymus, Thymbra, Salvia, Satureja, Lavandula, Origanum, Mentha, Achillea, Teucrium, Ziziphora, Cyclotrichium, Stachys cinslerine ait bazı türlerin uçucu yağlarının majör bileşenleri tespit edilip kaynak tür olma potansiyelleri yönünden analizleri gerçekleştirilmiştir. Özellikle Thymbra türlerinin karvakrol, Cyclotrichium origanifolium türünün pulegon ve Lavandula türlerinin linalol bileşenleri için kaynak tür olabileceği bulunmuştur. Aim: In this thesis, the essential oils of some species collected from the Eastern and Southeastern Anatolia regions of Turkey and widely used as tea and spice among the people were prepared with the Clevenger apparatus. It is aimed to develop and validate a GC-MS quantification method that includes special components are provided from the market to these species. In addition, the contents of these species in terms of these components were determined. Material and Method: A GC-MS method was developed and validated with the components which including α-Pinene, camphene, α-fellandrene, p-cymene, limonene, eucalyptol (1,8-cineol), β-ocimene, γ-terpinene, linalool, camphor, citronellal, menthone, borneol, menthol, α-terpineol, cis -geraniol, pulegon, L-carvone, linalyl acetate, bornyl acetate, thymol, carvacrol, β-caryophyllene. Essential oils of some species were obtained by hydrodistillation method using Clevenger apparatus. Then, the contents of these essential oils were determined by GC-MS. Results: It was seen that, the common main component's in all studied Thymbra species were carvacrol, in all Salvia and Mentha species were eucalyptol (1,8-cineol), in all Origanum species were p-cimen, and in all Lavandula species were linalool found.It was detected that, Thymus pubescens var. crateicola is rich in thymol, Cyclotrichium origanifolium is rich in pulegon, Achillea vermicularis is rich in eucalyptol, Teucrium polium is rich in β-caryophyllene, Zizphora clinopdioides is rich in pulegon components. Conclusion: The major components of the essential oils of some species of Thymus, Thymbra, Salvia, Satureja, Lavandula, Origanum, Mentha, Achillea, Teucrium, Ziziphora, Cyclotricium, Stachys which are grown in the Eastern and Southeastern regions of our country and are widely used among the people, were determined and analyzed in terms of their potential to be source species. It was found that especially Thymbra spp. carvacrol, Cyclotricium origanifolium spp. pulegon and Lavandula spp. can be source species for linalool components. Bu araştırma, standart beç tavuklarının (Numida meleagris) organik sistemde diyetlerine kuru kekik (Origanum vulgare L. KY) yaprak tozu ilavesinin refah parametrelerini ve kan serumunda bazı biyokimyasal özelliklerini saptamak amacıyla yürütülmüştür. Standart beç tavuğu genotipinden günlük yaşta toplam 240 karışık cinsiyette civcivler rastgele 3 tekerrürlü olmak üzere 4 deneme grubuna dağıtılmıştır. Hayvanlar on altı haftalık süre boyunca temel karma yemde kontrol (0 g/kg), 5 g/kg, 10 g/kg ve 15 g/kg KY içeren karma yemler ile beslenmişlerdir. Tonik immobilite (TI), göreceli asimetri (GA), dalgalı asimetri (DA), rektal sıcaklık (RS) ve kan serumunda bazı biyokimyasal parametreler ölçülmüştür. 6, 10, 14. haftalarda ölçülen tonik immobilite (TI), tonik immobilite induksiyon sayısı (TIS) ve rektal sıcaklık (RS) bakımından gruplar arasında önemli bir farklılık bulunmamıştır (P>0.05). Grupların, parmak phalanx uzunluğu ve tarsometatarsus genişlik ve uzunluğuna ait GA ve DA ile ilgili ortalama verileri arasında istatistiki bir farklılık gözlenmemiştir (P>0.05). Sonuç olarak, beç tavukları diyetine kekik ilavesinin tonik immobilite, rektal sıcaklık, göreceli ve dalgalı asimetri gibi refah parametreleri üzerine etkisi gözlenmemiştir.

Bu çalışmada, Akseki-İbradı-Manavgat (Antalya) ilçelerinde yayılış gösteren Lamiaceae ailesine ait türlerin çeşitliliği ile bunların bölgedeki ve kaynaklardaki etnobotanik kullanımları sunulmuştur. Çalışma alanında, Lamiaceae’nin 27 cinsine ait 149 takson (131 tür) tespit edilmiştir. Bunlardan 50 takson (%33.56) Türkiye’ye özgü olan endemik bitkilerdir. Yöre halkı, kullanacakları türleri doğal popülasyonlardan toplayıp genellikle güneş altında veya gölge yerlerde kurutmaktadır. Yöre halkı, başlıca kuru Origanum onites (Türk Kekiği)’i tüketir ve satar. Yüksek yağ verimi ve etkinliği nedeniyle geleneksel olarak Origanum majorana (Akkekik)’dan damıtma ile yağ elde edilmektedir. Elde edilen bitkisel ürünler, yöre halkı tarafından şiddetli öksürük, kronik soğuk algınlığı, yaralar, mide-bağırsak bozuklukları ve cilt problemleri gibi çeşitli hastalıkların tedavisinde kullanılmaktadır. Bitki örneklerinin aşırı bir şekilde doğadan yabanıl populasyonlardan toplanması türlerin genetik erozyona uğramasına sebep olabilir. Doğal popülasyonların sürdürülebilirliğini sağlamak için, in situ ve ex situ koruma yöntemleri gibi farklı genetik kaynak koruma programları uygulanmalıdır.

In Turkey there are much herbs there is an herb called ”Kekik”, which is used for cooking, drunk as tea, or widely used as a preservative in every day. However, when we are looking for kekik, we find much kekik, there are many Latin names such as thyme and oregano and others. I tried to find out what calls Kekik in Aegean area. And I also report traditional usage and new research trends.

Effect of essential oil against bacterial knot disease caused by Pseudomonas savastanoi pv. savastanoi in in vitro conditions The antibacterial effect of essential oils of thyme (Thymus vulgaris), bergamot (Citrus bergamia), garlic (Allium sativum), french lavender (Lavandula stoechas) clove (Caryophyllus aromaticum), eucalyptus (Eycalyptus globus) against Pseudomonas savastanoi pv. savastanoi were investigated. When essential oils were applied by dropping on absorbent paper disks and mixing with bacterial suspension thyme, bergamot, garlic, french lavender, clove, eucalyptus essential oils have been found effective more than others. It is detected that as the dose of essential oils increased, the antibacterial effect increased. Bacterial population did not change after 24 h growth in different treatment of essential oils at different times, but relative decrease in bacterial population at 0, 1, 3, and 6 h growing were observed. Especially, thyme essential oil showed the highest antibacterial effect, in all of the applications in different doses. Thyme was the most effective essential oil. Bergamot, lavender and eucalyptus essential oils followed the thyme essential oil, respectively. [ABSTRACT FROM AUTHOR]
