1. Do the shuffle: exploring reasons for music listening through shuffled play

Abstract

Adults listen to music for an average of 18 hours a week (with some people reaching more than double that). With rapidly changing technology, music collections have become overwhelmingly digital ushering in changes in listening habits, especially when it comes to listen- ing on personal devices. By using interactive visualizations, descriptive analysis and thematic analysis, this project aims to explore why people download and listen to music and which aspects of the music listening experience are prioritized when people talk about tracks on their device. Using a newly developed data collection method, Shuffled Play, 397 participants answered open-ended and closed research questions through a short online questionnaire after shuffling their music library and playing two pieces as prompts for reflections. The findings of this study highlight that when talking about tracks on their personal devices, people prioritise characterizing them using sound and musical features and associating them with the informational context around them (artist, album, and genre) over their emotional responses to them. The results also highlight that people listen to and download music because they like it–a straightforward but important observation that is sometimes glossed over in previous research. These findings have implications for future work in under- standing music, its uses and its functions in peoples’ everyday lives.

ISBN 978-1-4673-4953-6, International audience, In large SoCs, managing the effects of soft-errors in flip-flops is essential, however, selective mitigation is necessary to minimize the area and power costs. The identification of the optimal set of flip-flops to protect typically requires compute-intensive fault-injection campaigns. We present new techniques which group similar flip-flops into clusters to significantly reduce the number of fault injections. The number of required fault injections can be significantly lower than the total number of flip-flops and in one industrial design with over 100,000 flip-flops, by simulating only 2,100 fault injections, the technique identified a set of 4.1% of the flip-flops, which when protected, reduced the critical failure rate by a factor of 7x.

Abstract

BACKGROUND: Hearing impairment is common among older adults and affects cognitive assessments for identification of dementia which rely on good hearing function. We developed and validated a version of the Montreal Cognitive Assessment (MoCA) for people with hearing impairment.METHODS: We adapted existing MoCA 8.1 items for people with hearing impairment by presenting instructions and stimuli in written rather than spoken format. One Attention domain and two Language domain items required substitution by alternative items. Three and four candidate items respectively were constructed and field-tested along with the items adapted to written form. We used a combination of individual item analysis and item substitution to select the set of alternative items to be included in the final form of the MoCA-H in place of the excluded original items. We then evaluated the performance and reliability of the final tool, including making any required adjustments for demographic factors.RESULTS: One hundred and fifty-nine hearing-impaired participants, including 76 with normal cognition and 83 with dementia, completed the adapted version of the MoCA. A further 97 participants with normal hearing completed the standard MoCA as well as the novel items developed for the MoCA-H to assess score equivalence between the existing and alternative MoCA items and for independence from hearing impairment. Twenty-eight participants were retested between 2-4 weeks after initial testing. After the selection of optimal item set, the final MoCA-H had an area under the curve of 0.973 (95% CI 0.952-0.994). At a cut-point of 24 points or less sensitivity and specificity for dementia was 92.8% and 90.8%, respectively. The intraclass correlation for test-retest reliability was 0.92 (95%CI 0.78-0.97).CONCLUSION: The MoCA-H is a sensitive and reliable means of identifying dementia among adults with acquired hearing impairment.

Abstract

Importance: Cardiovascular accumulation of collagen (fibrosis) may contribute to the progression from ventricular dysfunction to heart failure. Galectin-3, a potential marker of pro-fibrotic activity, might identify those at greater risk.\ud Objective: To investigate the effects of spironolactone, according to serum galectin-3 concentration, on serum markers of fibrosis and on cardiac structure and function, in people at increased risk of developing heart failure.\ud Design: Prospective, randomized, open-label, blinded endpoint (PROBE) trial.\ud Setting: Clinical research facilities in ten European hospitals.\ud Participants: People with, or at high-risk of, coronary disease with increased plasma concentrations of B-type natriuretic peptides (BNP or NT-proBNP).\ud Interventions: spironolactone (up to 50 mg/day) or control for up to nine months.\ud Main Outcomes and Measures: The primary outcome was the interaction between baseline serum galectin-3 and change in serum procollagen type-III N-terminal pro-peptide (PIIINP), a by-product of type-III collagen synthesis. Serum procollagen type-I C-terminal pro-peptide (PICP) and collagen type-1 C-terminal telopeptide (CITP), respectively reflecting synthesis and degradation of type-I collagen, were also measured.\ud Results: Of 527 participants, the median age was 73 years and 26% were women. Median follow-up was 267 days. Changes in PIIINP were similar for those assigned to spironolactone and control (mean difference -0.15; 95% confidence interval [CI] -0.44 to 0.15 μg/L; p=0.32) and did not differ when serum galectin-3 was above or below median. Abstract

International audience; This study presents a detailed methodology, which combines high-precision thermogravimetry and FTIR spectroscopy, aiming to establish the most accurate and reliable means of measuring the molar absorption coefficients of adsorbed species. As the integrated molar absorption coefficients of Py complexes with Brønsted and Lewis acid sites, ɛ(Py-B) and ɛ(Py-L), are determined and the validity of the Beer-Lambert-Bouguer law for IR characterisation of solid acids is demonstrated, this work is setting a benchmark for the quantitative acidity measurements in zeolites and related materials. The following values of ɛ(Py-B) have been obtained at 150°C (band at ~1545 cm-1): 1.09±0.08 cm µmol-1 for ZSM-5; 1.12±0.16 cm µmol-1 for BEA; 1.29±0.04 cm µmol-1 for MOR and 1.54±0.15 cm µmol-1 for FAU. The value of ɛ(Py-L) (band at ~1455 cm-1 , which refers to different cations) measured at the same temperature is 1.71±0.1 cm µmol-1. Values of ɛ(Py-B) depend on the zeolite structure, in contrast to that for ɛ(Py-L). Clear correlations are presented between the temperature of the FTIR measurements and ɛ values for Py complexes and other species, which decrease by ~10% as the temperature increases by 100oC. In addition, the effects of key experimental procedures, instrumentation design and sample preparation are established and quantified.

Abstract

The $^{26}$Al short-lived radioactive nuclide is the source of the observed galactic diffuse $\gamma$-ray emission at 1.8 MeV. While different sources of $^{26}$Al have been explored, such as AGB stars, massive stars winds, and supernovae, the contribution of very massive stars has never been studied. We study the stellar wind contribution of very massive stars, i.e stars with initial masses between 150 and 300 M$_\odot$, to the enrichment in $^{26}$Al of the galactic interstellar medium. We discuss the production of $^{26}$Al by studying rotating and non-rotating very massive stellar models with initial masses between 150 and 300 M$_\odot$ for metallicities Z=0.006, 0.014, and 0.020. We confront this result to a simple Milky Way model taking into account both the metallicity and the star formation rate gradients. We obtain that very massive stars in the Z=0.006-0.020 metallicity range might be very significant contributors to the $^{26}$Al enrichment of the interstellar medium. Typically, the contribution of the winds of massive stars to the total quantity of $^{26}$Al in the Galaxy increases by 150\% when very massive stars are considered. Abstract

International audience; In recent years, research on high-risk athletes has focused mainly on personality traits, highlighting a complex and often ambivalent personality structure. Although scholars pointed out emotional difficulties amongst high-risk athletes, there exists to this day no in-depth investigation of the affective aspect. In an effort to address this so-far neglected question, this study offers the first thorough exploration of patterns of romantic relationships among high-risk athletes. Two groups were selected: people who practice extreme sports (N = 189; Extreme) and those who practice other types of sports (N = 207; Non-extreme). A series of tests were administered to evaluate romantic love, personality traits and self-esteem. Results showed that Extreme tend to display less intense feelings than their Non-extreme counterparts; conversely, their sentimental relationships are characterized by positive emotions and stable feelings, avoiding negative emotions. Hence, despite stereotypes which characterize Extreme as thrill-seekers prone to impulsivity, it seems that this group do not in fact seek as many thrills in their love life, but rather seek stability and contentment. Although still in it explorative phase, this study produced preliminary data which should be considered in devising further assessment and therapeutic strategies for this population of athletes.

A global synthesis of raw seedproduction data shows a 250-fold increase in seed abundance from cold-dry to warm-wet climates, driven primarily by a 100-fold increase in seed production for a given tree size. The modest (threefold) increase in forest productivity across the same climate gradient cannot explain the magnitudes of these trends. The increase in seeds per tree can arise from adaptive evolution driven by intense species interactions or from the direct effects of a warm, moist climate on tree fecundity. Abstract

Climate change, with its increasing frequency of environmental disturbances puts pressures on the livestock sector. To deal with these pressures, more complex traits such as resilience must be considered in our management strategies and in our breeding programs. Resilient animals respond well to environmental challenges, and have a decreased probability of needing assistance to overcome them. This paper discusses the need for operational measures of resilience that can be deployed at large scale across different farm types and livestock species. Such measures are needed to provide more precise phenotypes of resilience for use in farm management, but also for use in animal breeding. Any measure of response and recovery reflects both the animals resilience and the perceived size of the environmental disturbance, which can vary over time, depending on multiple animal and farm-related contexts. Therefore, and because universal definitions of resilience are too broad to be operational, we argue that resilience should be seen as a latent construct that cannot be directly measured and selected for. This leads to the following two points: (1) any postulated operational measure of resilience to a disturbance should be constructed from a sufficient number of indicators that each individually capture different facets of the resilience, such that when combined they better reflect the full resilience response; and (2) any postulated operational measure of resilience will have to be validated against reference measures that are the accumulated consequences of good resilience (e.g. productive lifespan or ability to re-calve). In a dairy cow case study, a practical resilience definition for dairy cattle was proposed and tested based on a scoring system containing several categories. In general terms and within a given parity, a cow receives plus points for each calving, and for a shorter calving interval, fewer inseminations and a higher milk production compared to her herd peers. She will receive minus points in case the number of inseminations increases, for each curative treatment day, and if her milk production is lower compared to her herd peers. By using readily available farm data, we were able to assess a practical lifetime resilience score, based on which cows can then be ranked within the herd. Cows that reach a next parity were shown to have a higher rank than cows that are culled before the next parity. To examine the usefulness of such a score, this resilience ranking was linked to two precision livestock technology-derived measures, related to milk yield deviations and accelerometer-derived deviations. Higher resilience ranking cows had fewer drops in milk yield and a more stable activity pattern during the lactation. This case study, taking the operational approach to quantifying and defining resilience, shows the promise of a data-driven approach for identifying resilience measures when applied within a biologically logical framework. ispartof: Peer Community Journal vol:2 status: published

Abstract

CHEOPS Consortium: V. Bourrier et al., Much remains to be understood about the nature of exoplanets smaller than Neptune, most of which have been discovered in compact multi-planet systems. With its inner ultra-short period planet b aligned with the star and two larger outer planets d-c on polar orbits, the multi-planet system HD 3167 features a peculiar architecture and offers the possibility to investigate both dynamical and atmospheric evolution processes. To this purpose we combined multiple datasets of transit photometry and radial velocimetry (RV) to revise the properties of the system and inform models of its planets. This effort was spearheaded by CHEOPS observations of HD 3167b, which appear inconsistent with a purely rocky composition despite its extreme irradiation. Overall the precision on the planetary orbital periods are improved by an order of magnitude, and the uncertainties on the densities of the transiting planets b and c are decreased by a factor of 3. Internal structure and atmospheric simulations draw a contrasting picture between HD 3167d, likely a rocky super-Earth that lost its atmosphere through photo-evaporation, and HD 3167c, a mini-Neptune that kept a substantial primordial gaseous envelope. We detect a fourth, more massive planet on a larger orbit, likely coplanar with HD 3167d-c. Dynamical simulations indeed show that the outer planetary system d-c-e was tilted, as a whole, early in the system history, when HD 3167b was still dominated by the star influence and maintained its aligned orbit. RV data and direct imaging rule out that the companion that could be responsible for the present-day architecture is still bound to the HD 3167 system. Abstract

Sex-ratio distorters based on X-chromosome shredding are more efficient than sterile male releases for population suppression. X-shredding is a form of sex distortion that skews spermatogenesis of XY males towards the preferential transmission of Y-bearing gametes, resulting in a higher fraction of sons than daughters. Strains harboring X-shredders on autosomes were first developed in the malaria mosquito Anopheles gambiae, resulting in strong sex-ratio distortion. Since autosomal X-shredders are transmitted in a Mendelian fashion and can be selected against, their frequency in the population declines once releases are halted. However, unintended transfer of X-shredders to the Y-chromosome could produce an invasive meiotic drive element, that benefits from its biased transmission to the predominant male-biased offspring and its effective shielding from female negative selection. Indeed, linkage to the Y-chromosome of an active X-shredder instigated the development of the nuclease-based X-shredding system. Here, we analyze mechanisms whereby an autosomal X-shredder could become unintentionally Y-linked after release by evaluating the stability of an established X-shredder strain that is being considered for release, exploring its potential for remobilization in laboratory and wild-type genomes of An. gambiae and provide data regarding expression on the mosquito Y-chromosome. Our data suggest that an invasive X-shredder resulting from a post-release movement of such autosomal transgenes onto the Y-chromosome is unlikely.

Abstract

(Abridged) Soft and hard X-ray excesses, compared to the continuum power-law shape between ~2-10 keV, are common features observed in the spectra of active galactic nuclei (AGN) and are associated with the accretion disc-corona system around the supermassive black hole. However, the dominant process at work is still highly debated and has been proposed to be either relativistic reflection or Comptonisation. We aim to characterise the main X-ray spectral physical components from the bright bare Broad Line Seyfert 1 AGN Mrk 110, and the physical process(es) at work in its disc-corona system viewed almost face-on. We perform the X-ray broad-band spectral analysis thanks to two simultaneous XMM-Newton and NuSTAR observations performed on November 16-17 2019 and April 5-6 2020, we also use for the spectral analysis above 3 keV the deep NuSTAR observation obtained in January 2017. The broad-band X-ray spectra of Mrk 110 are characterised by the presence of a prominent and absorption-free smooth soft X-ray excess, moderately broad OVII and FeKalpha emission lines and a lack of a strong Compton hump. The continuum above ~3keV is very similar at both epochs, while some variability (stronger when brighter) is present for the soft X-ray excess. A combination of soft and hard Comptonisation by a warm and hot corona, respectively, plus mildly relativistic disc reflection reproduce the broadband X-ray continuum very well. The inferred warm corona temperature, kT_warm~0.3 keV, is similar to the values found in other sub-Eddington AGN, whereas the hot corona temperature, kT_hot~21-31 keV (depending mainly on the assumed hot corona geometry), is found to be in the lower range of the values measured in AGN., Comment: 11 pages (including appendix), 8 figures. Abstract

We present 17 transit light curves of seven known warm-Jupiters observed with the CHaracterising ExOPlanet Satellite (CHEOPS). The light curves have been collected as part of the CHEOPS Guaranteed Time Observation (GTO) program that searches for transit-timing variation (TTV) of warm-Jupiters induced by a possible external perturber to shed light on the evolution path of such planetary systems. We describe the CHEOPS observation process, from the planning to the data analysis. In this work, we focused on the timing performance of CHEOPS, the impact of the sampling of the transit phases, and the improvement we can obtain by combining multiple transits together. We reached the highest precision on the transit time of about 13-16 s for the brightest target (WASP-38, G = 9.2) in our sample. From the combined analysis of multiple transits of fainter targets with G ≥ 11, we obtained a timing precision of ∼2 min. Additional observations with CHEOPS, covering a longer temporal baseline, will further improve the precision on the transit times and will allow us to detect possible TTV signals induced by an external perturber., The early support for CHEOPS by Daniel Neuenschwander is gratefully acknowledged. Abstract

The accuracy of theoretical mass, radius and effective temperature values for M-dwarf stars is an active topic of debate. Differences between observed and theoretical values have raised the possibility that current theoretical stellar structure and evolution models are inaccurate towards the low-mass end of the main sequence. To explore this issue we use the CHEOPS satellite to obtain high-precision light curves of eclipsing binaries with low mass stellar companions. We use these light curves combined with the spectroscopic orbit for the solar-type companion to measure the mass, radius and effective temperature of the M-dwarf star. Here we present the analysis of three eclipsing binaries. We use the pycheops data analysis software to fit the observed transit and eclipse events of each system. Two of our systems were also observed by the TESS satellite -- we similarly analyse these light curves for comparison. We find consistent results between CHEOPS and TESS, presenting three stellar radii and two stellar effective temperature values of low-mass stellar objects. These initial results from our on-going observing programme with CHEOPS show that we can expect to have ~24 new mass, radius and effective temperature measurements for very low mass stars within the next few years., Comment: 12 pages, 8 figures, accepted for publication in MNRAS

Abstract

Zeolite dealumination and desilication are common methods, which improve accessibility of the active sites located inside the zeolite crystallites and tune the zeolite acidity. The exact mechanism of these post-synthesis zeolite treatments remains under discussion. In this paper, a series of dealuminated and desilicated ZSM-5 zeolites are prepared using single or combined post-synthesis dealumination and desilication of large zeolite single crystals with oxalic acid and sodium hydroxide. The ZSM-5 zeolite has been synthesized using silica extracted from fly ash. Zeolite desilication results in the potholes, leaf-type structures and other clearly visible defects on the surface of zeolite crystallites, while the effect of dealumination is less pronounced. The low temperature nitrogen adsorption suggests the formation of mesopores in the zeolites formed by the voids between and in the irregular zeolite crystallites produced during post-synthesis treatments. 27Al and 29Si MAS NMR in combination with XRD data are indicative of only minor modifications of the bulk zeolite structure during dealumination and desilication. The total Bronsted acidity only slightly decreases and is not affected by the type of zeolite post-synthesis treatment. After desilication of large single crystals of ZSM-5, noticeable enhancement of the Bronsted acidity on the zeolite external surface has been detected by FTIR of adsorbed collidine. The zeolites with higher acidity on the external surface have shown higher activity in the anisole acylation with hexanoic acid. A combination of characterization techniques suggests that the dealumination and desilication of the large crystals of ZSM-5 zeolite using post-synthesis treatments respectively with acid and alkali selectively starts at the zeolite external surface.

Abstract

About 40% of foodborne infections are acquired in the home. The aim of the present study was to track contamination of pathogens during domestic food preparation and link the contamination to preparation practices. Research participants from 87 households in six European countries were observed and interviewed during shopping and preparation of a chicken and vegetable meal. The presence of Salmonella spp., Campylobacter spp. and norovirus on raw chicken, kitchen surfaces, cloths and sponges was determined.\ud \ud \ud \ud The prevalence of Campylobacter on raw chicken varied from 8.3% in Norway (NO) to 80% in France (FR) and Portugal (PT), with a mean prevalence of 57%. Campylobacter was found on half of the products that had been frozen and appeared to be less prevalent on chicken from supermarkets than other sources. Salmonella was found in 8.6% of raw chicken samples, exclusively from Hungary (HU).\ud \ud \ud \ud A relationship between observed practices and spread of pathogens to kitchen surfaces was found only for the use of cutting boards for chicken and/or vegetables. After food preparation, Campylobacter and Salmonella were isolated from 23% (samples derived from HU, RO, UK) and 8.7% (HU), respectively of cutting boards. Research participants in France and Portugal were more likely to buy products that fitted their recipe, with less need for using cutting boards. Using the same board and knife for vegetables after using it for chicken and without washing with detergent was common in Portugal and Romania, but not in the other countries. Contamination with Campylobacter to other kitchen surfaces or washing utensils were found in five households (UK, RO, PT). Rinsing chicken in sinks was common in three countries (PT, HU, RO), and washing vegetables in the same sink was also usual. Prevalence of Norovirus was low, with detection in one out of 451 samples. The participants' awareness of the risk posed by pathogens from raw chicken differed among the six countries, with higher awareness in Norway and the UK than the other countries studied.\ud \ud \ud \ud In conclusion, practices intended to avoid cross-contamination from chicken to kitchen surfaces and washing utensils are not established among consumers in all European countries. Nevertheless, cross-contamination events that disseminate infectious doses of pathogens seems to be rare, probably due to the relatively low levels of pathogens in food combined with food preferences. Food safety interventions must consider the national food culture, preferences, practices and the prevalence and levels of pathogens in food. Emphasis should be on providing and promoting chicken products with lower risk (prevalence of pathogens, ready-to-cook) and safe use of cutting boards.

Abstract

Aims Uncontrolled blood pressure (BP) increases the risk of developing heart failure (HF). The effect of spironolactone on BP of patients at risk of developing HF is yet to be determined. To evaluate the effect of spironolactone on the BP of patients at risk for HF and whether renin can predict spironolactone’s effect. Methods and results HOMAGE (Heart OMics in Aging) was a prospective multicentre randomized open-label blinded endpoint (PROBE) trial including 527 patients at risk for developing HF randomly assigned to either spironolactone (25–50 mg/day) or usual care alone for a maximum of 9 months. Sitting BP was assessed at baseline, Months 1 and 9 (or last visit). Analysis of covariance (ANCOVA), mixed effects models, and structural modelling equations was used. The median (percentile25–75) age was 73 (69–79) years, 26% were female, and >75% had history of hypertension. Overall, the baseline BP was 142/78 mmHg. Patients with higher BP were older, more likely to have diabetes and less likely to have coronary artery disease, had greater left ventricular mass (LVM), and left atrial volume (LAV). Compared with usual care, by last visit, spironolactone changed SBP by −10.3 (−13.0 to −7.5) mmHg and DBP by −3.2 (−4.8 to −1.7) mmHg (P Conclusion Spironolactone had a clinically important BP-lowering effect. Spironolactone should be considered for lowering blood pressure in patients who are at risk of developing HF.

Abstract

Background General practitioners (GPs) should regularly review patients’ medications and, if necessary, deprescribe, as inappropriate polypharmacy may harm patients’ health. However, deprescribing can be challenging for physicians. This study investigates GPs’ deprescribing decisions in 31 countries. Methods In this case vignette study, GPs were invited to participate in an online survey containing three clinical cases of oldest-old multimorbid patients with potentially inappropriate polypharmacy. Patients differed in terms of dependency in activities of daily living (ADL) and were presented with and without history of cardiovascular disease (CVD). For each case, we asked GPs if they would deprescribe in their usual practice. We calculated proportions of GPs who reported they would deprescribe and performed a multilevel logistic regression to examine the association between history of CVD and level of dependency on GPs’ deprescribing decisions. Results Of 3,175 invited GPs, 54% responded (N = 1,706). The mean age was 50 years and 60% of respondents were female. Despite differences across GP characteristics, such as age (with older GPs being more likely to take deprescribing decisions), and across countries, overall more than 80% of GPs reported they would deprescribe the dosage of at least one medication in oldest-old patients (> 80 years) with polypharmacy irrespective of history of CVD. The odds of deprescribing was higher in patients with a higher level of dependency in ADL (OR =1.5, 95%CI 1.25 to 1.80) and absence of CVD (OR =3.04, 95%CI 2.58 to 3.57). Interpretation The majority of GPs in this study were willing to deprescribe one or more medications in oldest-old multimorbid patients with polypharmacy. Willingness was higher in patients with increased dependency in ADL and lower in patients with CVD.

Abstract

Nearly 40% foodborne outbreaks in the European Union are attributable to food practices in domestic homes that include handling and preparation of raw chicken. Hand washing is an important way to prevent cross-contamination with pathogens during chicken preparation. This study, which is part of the EU Horizon 2020 funded consortium SafeConsume, aimed at quantifying and understanding hand washing practices in three categories of households and five European countries. A quantitative survey (n = 1889) was combined with qualitative research, during which 75 participants from France, Norway, Portugal, Romania, and the United Kingdom were observed and interviewed. An original method for analysing video with “The Observer XT” software was developed to identify when and how risk arises. The quantitative survey and qualitative research data revealed that touching raw chicken was more frequent in Romania and Portugal. Practices to avoid touching raw chicken were declared and observed, although observations revealed that these practices were not always consistently followed. Only a third of the participants washed their hands with soap after handling raw chicken with important variations among countries (a majority in Norway and in the UK, a few in France and Portugal, none in Romania), in contrast to the results of the survey. Observations and interviews suggested that rinsing hands with water only and washing hands with soap are considered equivalent by many people. Barriers to washing hands due to improper equipment were mainly observed in Romania. Washing hands after touching raw chicken was motivated by food safety concerns for some participants in Norway and the UK, but not in France and Portugal, where it was motivated by unpleasant feelings on hands, or presented as a habit. Participants not washing their hands after touching the chicken did it after other actions they presumably perceived as unsafe (e. g. touching the bin, handling pets, and blowing the nose), indicating that they did not specifically consider touching raw chicken as risky. Knowledge, habits, and equipment with regard to chicken and hand washing differed among European countries, resulting in safe and risky practices. Abstract

International audience; CRISPR guide RNAs (gRNAs) can be programmed with relative ease to allow the genetic editing of nearly any DNA or RNA sequence. Here, we propose novel molecular architectures to achieve RNA-dependent modulation of CRISPR activity in response to specific RNA molecules. We designed and tested, in both living Escherichia coli cells and cell-free assays for rapid prototyping, cis-repressed RNA-interacting guide RNA (igRNA) that switch to their active state only upon interaction with small RNA fragments or long RNA transcripts, including pathogen-derived mRNAs of medical relevance such as the human immunodeficiency virus infectivity factor. The proposed CRISPR-igRNAs are fully customizable and easily adaptable to the majority if not all the available CRISPR-Cas variants to modulate a variety of genetic functions in response to specific cellular conditions, providing orthogonal activation and increased specificity. We thereby foresee a large scope of application for therapeutic, diagnostic, and biotech applications in both prokaryotic and eukaryotic systems.

Abstract

Tropical forests constitute a prolific sanctuary of unique floral diversity and potential medicinal sources, however, many of them remain unexplored. The scarcity of rigorous scientific data on the surviving Mascarene endemic taxa renders bioprospecting of this untapped resource of utmost importance. Thus, in view of valorizing the native resource, this study has as its objective to investigate the bioactivities of endemic leaf extracts. Herein, seven Mascarene endemic plants leaves were extracted and evaluated for their in vitro antioxidant properties and antiproliferative effects on a panel of cancer cell lines, using methyl thiazolyl diphenyl-tetrazolium bromide (MTT) and clonogenic cell survival assays. Flow cytometry and comet assay were used to investigate the cell cycle and DNA damaging effects, respectively. Bioassay guided-fractionation coupled with liquid chromatography mass spectrometry (MS), gas chromatography-MS, and nuclear magnetic resonance spectroscopic analysis were used to identify the bioactive compounds. Among the seven plants tested, Terminaliabentzo&euml, was comparatively the most potent antioxidant extract, with significantly (p &lt, 0.05) higher cytotoxic activities. T. bentzo&euml, extract further selectively suppressed the growth of human hepatocellular carcinoma cells and significantly halted the cell cycle progression in the G0/G1 phase, decreased the cells&rsquo, replicative potential and induced significant DNA damage. In total, 10 phenolic compounds, including punicalagin and ellagic acid, were identified and likely contributed to the extract&rsquo, s potent antioxidant and cytotoxic activities. These results established a promising basis for further in-depth investigations into the potential use of T. bentzo&euml, as a supportive therapy in cancer management.

Abstract

If we take seriously literal interpretations of quantum theory and general relativity then the question posed in the title is no. This article highlights where incompatibilities arise by considering our experience in the context of modern physics. Implied nonmaterial minds possess properties that no physical entities do by being both transtemporal and localised in the configuration space. The principle of localisation (PL) is proposed, and is used to show that there is no need to resort to the Everett interpretation to show that minds exist outside of physics, post relativity physics is enough. However, in this regime, there is no free will, minds are destined to experience a pre-determined but unknown future. Free will is restored when we reinstate pure wave theories in line with Everett.

Abstract

Accurately quantifying species’ area requirements is a prerequisite for effective area‐based conservation. This typically involves collecting tracking data on species of interest and then conducting home‐range analyses. Problematically, autocorrelation in tracking data can result in space needs being severely underestimated. Based on the previous work, we hypothesized the magnitude of underestimation varies with body mass, a relationship that could have serious conservation implications. To evaluate this hypothesis for terrestrial mammals, we estimated home‐range areas with global positioning system (GPS) locations from 757 individuals across 61 globally distributed mammalian species with body masses ranging from 0.4 to 4000 kg. We then applied block cross‐validation to quantify bias in empirical home‐range estimates. Area requirements of mammals 1, meaning the scaling of the relationship changed substantially at the upper end of the mass spectrum., Article impact statement: Due to autocorrelation‐induced bias, conventional methods severely underestimate the area requirements of GPS‐tracked large mammals.

Abstract

BackgroundThe best management of hypertension in frail oldest-old (≥80 years of age) remains unclear and we still lack guidelines that provide specific recommendations. Our study aims to investigate guideline use in general practitioners (GPs) and to examine if guideline use relates to different decisions when managing hypertension in frail oldest-old.Design/settingCross-sectional study among currently active GPs from 29 countries using a case-vignettes survey.MethodsGPs participated in a survey with case-vignettes of frail oldest-olds varying in systolic blood pressure (SBP) levels and cardiovascular disease (CVD). GPs from 26 European countries and from Brazil, Israel and New Zealand were invited. We compared the percentage of GPs reporting using guidelines per country and further stratified on the most frequently mentioned guidelines. To adjust for patient characteristics (SBP, CVD and GPs' sex, years of experience, prevalence of oldest-old and location of their practice), we used a mixed-effects regression model accounting for clustering within countries.ResultsOverall, 2,543 GPs from 29 countries were included. 59.4% of them reported to use guidelines. Higher guideline use was found in female (p = 0.031) and less-experienced GPs (pConclusionMany GPs reported using guidelines to manage hypertension in frail oldest-old patients, however guideline users did not decide differently from non-users concerning hypertension treatment decisions. Instead of focusing on the fact if GPs use guidelines or not, we as a scientific community should put an emphasis on what guidelines suggest in frail and oldest-old patients.

Abstract

We present new observations of the transmission spectrum of the hot Jupiter WASP-6b both from the ground with the Very Large Telescope (VLT) FOcal Reducer and Spectrograph (FORS2) from 0.45-0.83 $\mu$m, and space with the Transiting Exoplanet Survey Satellite (TESS) from 0.6-1.0 $\mu$m and the Hubble Space Telescope (HST) Wide Field Camera 3 from 1.12-1.65 $\mu$m. Archival data from the HST Space Telescope Imaging Spectrograph (STIS) and Spitzer is also reanalysed on a common Gaussian process framework, of which the STIS data show a good overall agreement with the overlapping FORS2 data. We also explore the effects of stellar heterogeneity on our observations and its resulting implications towards determining the atmospheric characteristics of WASP-6b. Independent of our assumptions for the level of stellar heterogeneity we detect Na I, K I and H$_2$O absorption features and constrain the elemental oxygen abundance to a value of [O/H] $\simeq -0.9\pm0.3$ relative to solar. In contrast, we find that the stellar heterogeneity correction can have significant effects on the retrieved distributions of the [Na/H] and [K/H] abundances, primarily through its degeneracy with the sloping optical opacity of scattering haze species within the atmosphere. Our results also show that despite this presence of haze, WASP-6b remains a favourable object for future atmospheric characterisation with upcoming missions such as the James Webb Space Telescope., Comment: Accepted to MNRAS

Abstract

A transdisciplinary observational study, coupled with a web-based survey, was conducted to investigate refrigerated storage of food, in five European countries. The investigated consumer groups in this study were: young families with small children and/or pregnant women, elderly people, persons with an immunodeficient system, and young single men. The refrigerator temperature was monitored for approximately two weeks using a temperature data logger. Variables such as country, income, age of refrigerators, education, living area, refrigerator loading practices had no significant effect (p > 0.05) on the overall average fridge temperature, whereas consumers' practices showed a significant influence (p < 0.05) on registered temperature values. Compared to temperatures inside the fridges belonging to young families and young single men group, the temperatures inside refrigerators belonging to elderly was in the temperature danger zone (5–63 °C). The lowest temperatures were recorded in UK consumers’ refrigerators, whereas the highest were in French households. Presence of Listeria monocytogenes was confirmed in three refrigerators out of 53 sampled (two in Romania and one in Portugal). The most vulnerable category to food safety risks is represented by elderly persons with low education, unaware of safe refrigeration practices and the actual temperature their fridges are running. Abstract

Peru Balsam, a resinous substance derived from Myroxylon balsamum var. pereirae, has historically been used as a topical ointment for various skin conditions such as scabies, poorly healing wounds, eczema, and haemorrhoids. The ingredients responsible for these properties are not fully elucidated. We investigated the chemical composition of two Peru Balsam samples, one historical and one modern, using gas chromatography/mass spectrometry to identify the active ingredients responsible for its pharmaceutical properties. Both Peru Balsam specimens investigated had similar compositions, showing the stability of the substance. Components identified are effective against scabies, exhibit antimicrobial activity and aid skin penetration. These properties are consistent with historical uses of Peru Balsam. Several ingredients are also known allergens. This study, combining chemical information with scientific literature related to pharmaceutical properties of natural substances, represents a breakthrough in the elucidation of active ingredients in Peru Balsam.

Abstract

International audience; Methane is the main component of natural and shale gas, methane clathrates and biogas, and is a potent greenhouse gas. About 90% of methane is currently burnt in various combustion processes, releasing carbon dioxide into the atmosphere 1-7. Finding ways to efficiently convert methane into fuels and chemicals is important for the rational utilization of fossil and renewable energy feedstocks, as well as for reducing the emission of greenhouse gases. Methane is a highly stable molecule; it has no functional groups and a very high C-H bond enthalpy (439 kJ mol −1). Methane is also inert relative to acid attack and has a very low proton affinity (544 kJ mol −1) and acidity (pK a = 40). Therefore, direct chemical conversion of methane to value-added chemicals and fuels remains a formidable challenge for modern science 8-10. Commercial technologies of methane utilization, other than combustion, are rather limited. They involve methane steam reforming, partial oxidation, autothermal reforming or the Andrussow reaction 11-13. The non-commercial routes for methane conversion can be divided into oxidative and non-oxidative routes 8-21. The non-oxidative routes, such as methane aromatization, result in substantial carbon deposition, while the oxidative routes, such as methane thermocatalytic coupling and methane partial oxidation, usually suffer from insufficient selectivity and abundant production of CO 2. Most of the known methane conversion reactions require very high temperatures (>800 °C). Generally, these thermochemical processes are accompanied by CO 2 emissions arising from the combustion of fossil fuels utilized to maintain the reactor at high temperatures 18-23. Photocatalytic non-oxidative coupling of methane with very low quantum efficiencies and yields was observed over silica, highly dispersed Ga or Ce species 24-26. Activation of methane over Zn 2+ / ZSM-5 and Ga 3+ /EST-10 photocatalysts has been reported, with the photocatalytic activity attributed to the presence of extra-framework zinc cations and Ti-OH groups on titanate wires 27,28. Abstract

In the adult auditory organ, mechanoelectrical transducer (MET) channels are essential for transducing acoustic stimuli into electrical signals. In the absence of incoming sound, a fraction of the MET channels on top of the sensory hair cells are open, resulting in a sustained depolarizing current. By genetically manipulating the in vivo expression of molecular components of the MET apparatus, we show that during pre-hearing stages the MET current is essential for establishing the electrophysiological properties of mature inner hair cells (IHCs). If the MET current is abolished in adult IHCs, they revert into cells showing electrical and morphological features characteristic of pre-hearing IHCs, including the re-establishment of cholinergic efferent innervation. The MET current is thus critical for the maintenance of the functional properties of adult IHCs, implying a degree of plasticity in the mature auditory system in response to the absence of normal transduction of acoustic signals., Mechanoelectrical transducer (MET) channels on the tips of inner hair cells are essential for transducing auditory sensory information. Here, the authors show that disrupting MET channel function also prevents the preservation of normal inner hair cell identity in adult mice.

Abstract

Nano-crystalline synthetic gismondine modified via cation exchange has been utilised as a highly active and selective catalyst for the production of biofuel. In comparison with low silica zeolites FAU and LTA, K-form of gismondine, based on the maximum aluminium P (MAP) zeolite, exhibited a significant improvement in catalytic performance in the methanolysis of bio-oil, which can be attributed to its nano-particle morphology and high basicity associated with the high Al content and high degree of ion exchange, as demonstrated by the TEM, XRD, TGA, NMR, XPS and CO2-TPD studies. To the best of our knowledge, this is the first report on a successful catalytic application of basic K-MAP zeolite. Abstract

The influence of molecular confinement on the melting transition of oriented Na-DNA fibers submerged in poly(ethylene glycol) (PEG) solutions has been studied. The PEG solution exerts an osmotic pressure on the fibers which, in turn, is related to the DNA intermolecular distance. Calorimetry measurements show that the melting temperature increases and the width of the transition decreases with decreasing intermolecular distance. Neutron scattering was used to monitor the integrated intensity and width of a Bragg peak from the B-form of DNA as a function of temperature. The data were quantitatively analyzed using the Peyrard-Bishop-Dauxois model. The experiments and analysis showed that long segments of double-stranded DNA persist until the last stages of melting and that there appears to be a substantial increase of the DNA dynamics as the melting temperature of the DNA is approached.

Abstract

International audience; The state of copper species stabilized in nanosized LTL zeolite subjected to various post-synthesis treatments was unveiled by a range of spectroscopic techniques. FTIR and UV-Vis studies demonstrated that the reduction process of copper in the LTL nanosized zeolite leads to the formation of different species including Cu2+, Cu+ and Cu nanoparticles (Cu NPs). The adsorption of probe molecules (NO and CO) was used to selectively monitor the copper species in the LTL nanosized zeolite upon oxidation and reduction post-synthesis treatments. Both the Cu2+ and Cu+ species were probed by NO and CO, respectively. The amount of Cu+ in the LTL zeolite nanocrystals was about 43% as determined by FTIR, while the amount of Cu NPs was about 55% determined by the UV-Vis spectroscopic characterization. These results were complemented by EPR, 29Si and 63Cu MAS NMR spectroscopic data. The EPR spectroscopy was further applied to monitor the effective reduction of the Cu2+ species and their re-oxidation, while the 63Cu MAS NMR verified the presence of Cu NPs in the LTL nanosized zeolite crystals.

Abstract

International audience; Seed extracts from Moringa oleifera are of wide interest for use in water purification where they can play an important role in flocculation; they also have potential as anti-microbial agents. Previous work has focused on the crude protein extract. Here we describe the detailed biophysical characterization of individual proteins from these seeds. The results provide new insights relating to the active compounds involved. One fraction, designated Mo-CBP3, has been characterized at a molecular level using a range of biochemical and biophysical techniques including liquid chromatography, X-ray diffraction, mass spectrometry, and neutron reflection. The interfacial behavior is of particular interest in considering water purification applications and interactions with both charged (e.g. silica) and uncharged (alumina) surfaces were studied. The reflection studies show that, in marked contrast to the crude extract, only a single layer of the purified Mo-CBP3 binds to a silica interface and that there is no binding to an alumina interface. These observations are consistent with the crystallographic structure of Mo-CBP3-4, which is one of the main isoforms of the Mo-CBP3 fraction. The results are put in context of previous studies of the properties of the crude extract. This work shows possible routes to development of separation processes that would be based on the specific properties of individual proteins.

Abstract

In the era of large high-resolution spectroscopic surveys, high-quality spectra can contribute to our understanding of the Galactic chemical evolution, providing chemical abundances belonging to the different nucleosynthesis channels, and also providing constraints to stellar age. Some abundance ratios have been proven to be excellent indicators of stellar ages. We aim at providing an empirical relationship between stellar ages and [C/N] using, as calibrators, open star clusters observed by both the Gaia-ESO and APOGEE surveys. We use stellar parameters and abundances from the Gaia-ESO and APOGEE of the Galactic field and open cluster stars. Ages of star clusters are retrieved from the literature sources and validated using a common set of isochrones. We use the same isochrones to determine, for each age and metallicity, the surface gravity at which the first dredge-up and red giant branch bump occur. We study the effect of extra-mixing processes in our sample of giant stars, and we derive the mean [C/N] in evolved stars, including only stars without evidence of extra-mixing. Combining the Gaia-ESO and APOGEE samples of open clusters, we derive a linear relationship between [C/N] and logarithmic cluster ages. We apply our relationship to selected giant field stars in both Gaia-ESO and APOGEE. We find an age separation between thin and thick disc stars and age trends within their populations, with an increasing age towards lower metallicity populations. With such empirical relationship, we are able to provide an age estimate for giant stars in which C and N abundances are measured. Isochrone fitting is less sensitive for giant than dwarf stars at the turn off. The present method can be thus considered as an additional tool to give an independent estimate of the age of giant stars, with uncertainties in their ages comparable to those obtained using isochrone fitting for dwarf stars., Comment: 28 pages, 24 figures. Abstract

We present the analysis of stellar binary microlensing event OGLE-2015-BLG-0060 based on observations obtained from 13 different telescopes. Intensive coverage of the anomalous parts of the light curve was achieved by automated follow-up observations from the robotic telescopes of the Las Cumbres Observatory. We show that, for the first time, all main features of an anomalous microlensing event are well covered by follow-up data, allowing us to estimate the physical parameters of the lens. The strong detection of second-order effects in the event light curve necessitates the inclusion of longer-baseline survey data in order to constrain the parallax vector. We find that the event was most likely caused by a stellar binary-lens with masses $M_{\star1} = 0.87 \pm 0.12 M_{\odot}$ and $M_{\star2} = 0.77 \pm 0.11 M_{\odot}$. The distance to the lensing system is 6.41 $\pm 0.14$ kpc and the projected separation between the two components is 13.85 $\pm 0.16$ AU. Abstract

Breakthroughs in Medicinal Chemistry: New Targets and Mechanisms, New Drugs, New Hopes is a series of Editorials which is published on a biannual basis by the Editorial Board of the Medicinal Chemistry section of the journal Molecules. In these Editorials, we highlight in brief reports (of about one hundred words) a number of recently published articles that describe crucial findings, such as the discovery of novel drug targets and mechanisms of action or novel classes of drugs, which may inspire future medicinal chemistry endeavors devoted to addressing prime unmet medical needs.
