To the reader The Machine Vision Group of the University of Oulu has reached the honorable age of 25 years. In that time, it has developed into the most well-known community of experts in this field of research in Finland, and has also been widely recognized and respected internationally. The results of the group’s research have been applied to a great extent in the industry as well as other research groups both domestically and abroad. This 25th anniversary book presents the quarter-of-a-century success story of the group, from its foundation and early years until the leading-edge research of today. Separate chapters account for the most significant scientific accomplishments, the collaboration with the industry, and the perspectives of future research. The book also allows previous employees to explain how they now can utilize the training obtained in the group in their new challenges. A majority of the book consists of an impressive selection of the group’s most important and most merited scientific publications in their original form. The end section includes statistical data of the theses produced on machine vision. The 25th anniversary book is directed to everyone interested in machine vision and Oulu expertise in the field. The book gives an excellent example of how there is no shortcut to top-of-the-line research, but it can only be created through years of persistent research activity. Although the approach in the scientific publications is academic, the aim has been to write the initial part of the book in an easily comprehensible manner with numerous practical examples. Besides the editors, the other contributors to the making of the book were Janne Heikkilä, Olli Silvén, Timo Ahonen, Esa Rahtu, and Jukka Kontinen. Tiivistelmä. Yksinäisyys voi lapsilla ja nuorilla johtaa vakaviin psyykkisiin ja emotionaalisiin ongelmiin. Varhainen puuttuminen on erittäin tärkeää, sillä kroonistuessa yksinäisyydestä eroon pääseminen vaikeutuu. Tämän pro gradu -tutkielman tavoitteena on tarkastella yksinäisyyttä ilmiönä koulukontekstissa. Tutkielmalla pyritään selvittämään, millaisena oppilaiden yksinäisyys ilmenee koulussa peruskoulun opettajien käsitysten mukaan. Lisäksi tarkoituksena on löytää vastauksia,millaisia käsityksiä peruskoulun opettajilla on koulun mahdollisuuksista vähentää yksinäisyyttä. Teoreettisessa viitekehyksessä käsitellään yksinäisyyden ulottuvuuksia,taustatekijöitä ja seurauksia. Yksinäisyyden tehokkaimpia vähentämismenetelmiä tarkastellaan aikaisemman tutkimustiedon pohjalta. Lopuksi esitellään koulujen interventio-ohjelmia ja yhteistyötä eri tahojen kanssa. Tutkielma on laadullinen tutkimus, jossa on fenomenografinen lähestymistapa. Aineisto on kerätty sähköisellä kyselylomakkeella syyskuussa 2020. Aineisto koostuu 25 opettajan vastauksista. Aineiston analyysi on toteutettu fenomenografisen analyysimallin mukaisesti. Tulokset osoittavat, että opettajat tunnistavat yksinäisyyteen liittyviä tunnusmerkkejä. Nämä ovat samansuuntaisia aiemman tutkimustiedon kanssa. Oppilaiden yksinäisyys näyttäytyy opettajien käsitysten mukaan erityisesti ulkopuolisuutena ja ystävien puutteena. Yksinäisiä oppilaita kuvattiin ujoina ja arkoina, mutta toisaalta yksinäisyys voi näyttäytyä myös häiritsevänä käytöksenä. Opettajat pitävät oppilaan yksilöllistä kohtaamista, yhteisöllisyyttä, erilaisia pedagogisia käytäntöjä, yhteistyötä sekä eri interventio-ohjelmia yksinäisyyttä vähentävinä tekijöinä. Opettajien mukaan erityisesti kuulluksi tuleminen, ryhmähengen vahvistaminen sekä ohjattu välituntitoiminta vähentävät yksinäisyyttä. Johtopäätöksenä voidaan todeta, että opettajat pyrkivät lisäämään oppilaiden kontaktimahdollisuuksia ystävyyssuhteiden solmimiseksi. Tutkielman luotettavuutta lisäävät tutkijatriangulaatio sekä tutkimusprosessin ja tulosten yksityiskohtainen kuvaus. Tutkielman teossa on pyritty noudattamaan hyvää tieteellistä käytäntöä. Tutkielman tarkoituksena on lisätä kasvattajien tietoisuutta siitä, mitä yksinäisten oppilaiden hyväksi voidaan tehdä.

The regulatory network of R2R3 MYB transcription factors in anthocyanin biosynthesis is not fully understood in blue-coloured berries containing delphinidin compounds. We used blue berries of bilberry (Vaccinium myrtillus) to comprehensively characterise flavonoid-regulating R2R3 MYBs, which revealed a new type of co-regulation in anthocyanin biosynthesis between members of MYBA-, MYBPA1- and MYBPA2-subgroups. VmMYBA1, VmMYBPA1.1 and VmMYBPA2.2 expression was elevated at berry ripening and by abscisic acid treatment. Additionally, VmMYBA1 and VmMYBPA1.1 expression was strongly downregulated in a white berry mutant. Complementation and transient overexpression assays confirmed VmMYBA1 and VmMYBA2 to induce anthocyanin accumulation. Promoter activation assays showed that VmMYBA1, VmMYBPA1.1 and VmMYBPA2.2 had similar activity towards dihydroflavonol 4-reductase (DFR) and anthocyanidin synthase (ANS), but differential regulation activity for UDP-glucose flavonoid 3-O-glucosyltransferase (UFGT) and flavonoid 3′5′-hydroxylase (F3′5′H) promoters. Silencing of VmMYBPA1.1 in berries led to the downregulation of key anthocyanin and delphinidin biosynthesis genes. Functional analyses of other MYBPA regulators, and a member of novel MYBPA3 subgroup, associated them with proanthocyanidin biosynthesis and F3′5′H expression. The existence of 18 flavonoid-regulating MYBs indicated gene duplication, which may have enabled functional diversification among MYBA, MYBPA1 and MYBPA2 subgroups. Our results provide new insights into the intricate regulation of the complex anthocyanin profile found in blue-coloured berries involving regulation of both cyanidin and delphinidin branches.

Tutkielman tarkoituksena on kartoittaa, kuvailla, selittää, tulkita, vertailla ja ymmärtää potilas- ja asiakas-sanojen käyttöä ja siten tuottaa uutta ja jäsenneltyä tietoa terveydenhuollossa käytettävän kielen merkityksistä ja funktioista etenkin terveydenhuollon johto- ja kehitystehtävissä toimiville. Tutkielmassa hahmotellaan sanojen käyttöä ylläpitäviä ja lisääviä tekijöitä. Aineisto muodostuu viiden julkis- ja viiden yksityissektorin terveydenhuollon organisaation verkkosivuilta kerätyistä potilas- ja asiakas-sanan sisältävistä virkkeistä (n = 400). Menetelmä on kielitieteellinen semanttisten roolien (kaikkiaan kymmenen) analyysi. Tulosten mukaan niin julkis- kuin yksityissektorin aineistossa selvästi yleisin semanttinen rooli on HYÖTYJÄ, joka ilmentää passiivista ja staattista hoidettavana olemista ja terveydenhuollollisten toimenpiteiden kohteena olemista. Potilas- ja asiakas-sanat sijoittuvat TEKIJÄN rooliin suurin piirtein samassa suhteessa, eli asiakas-sanan käyttö ei ilmennä mittavampaa tekijyyttä, saati luo sitä. Asiakkaan liikkumavara näyttäisi olevan suurempi kuin potilaan, joskaan sanojen käytössä ei usein ollut eroja. Sen sijaan julkis- ja yksityissektorin aineistojen välisiä eroja oli enemmän: esimerkiksi erityisesti asiakkaan osallistumista kielennettiin julkissektorin aineistossa selvästi enemmän, mikä kertonee julkissektorilla vallitsevasta arvon yhteistuottajuuden ideasta. VASTAANOTTAJAN rooliin sijoittuu niin ikään lähinnä asiakas, joka on aktiivinen ja osaa muun muassa hyödyntää tietotekniikkaa hoitonsa tukena. Tulosten perusteella voidaan tehdä johtopäätös, jonka mukaan potilas- ja asiakas-sanoja käytetään toisaalta rinnasteisesti ja synonyymisesti, toisaalta ambivalentisti ja epätarkasti. Potilas-sanan asema terveydenhuollon kielessä on asiakas-sanan yleistymisestä huolimatta vakaa. Merkitykset elävät jatkuvasti, ja ne ovat neuvoteltavissa. Potilas- ja asiakas-sanojen konnotatiiviset merkitykset olisivat samankaltaistumassa: kun potilas-sanaan liitetyt kielteiset merkitykset sementoituvat asiakas-sanaan, syntyy tarve alkaa miettiä sille korvaavaa ilmausta. Terveyspalvelujen käyttäjiä voi kohdella asiakkaina, vaikka heitä kutsuisi potilaiksi. The purpose of this study was to chart, describe, explain, interpret, compare, and comprehend the way words patient and client/customer are written and, thus, generate new and analyzed information on the meanings and functions of the language used in health care. The factors maintaining and increasing the usage of both words were outlined. The research material comprised sentences (n = 400), including the words patient and client/customer gathered from five public- and five private-sector health care organization web sites. The data was analyzed using linguistic semantic role (altogether 10) analysis. According to the results obtained, the most common semantic role was clearly BENEFICIARY, both in the public- and the private-sector material. It embodies passive and static as nursed and medically cured. Patient and client/customer (words) are placed in an AGENT role approximately relatively: the usage of client/customer (word) did not embody more large-scale authorship, let alone create it. The leeway of client/consumer (word) seemed to be more extensive than patient (word), even though there were no differences worth mentioning in the usage of the words in general. Instead, there were more differences in the public- and private-sector material: for example, the involvement of clients/customers (words) in particular, was obviously more noticeable in the public sector material than in the private sector, which may indicate the idea of collaboration and active engagement with value-creation in the public sector. Client/customer (word) was also often seen as RECIPIENT, which is active and, for example, able to utilize communication technology as support in his/her care. The key findings demonstrate that the patient and client/customer words are used, however, appositively and synonymously and, then, ambivalently and imprecisely. The status of patient (word) in health care language is stable, despite the fact that client/customer (word) has become more common. The meanings differentiate constantly and are negotiable. The connotative meanings of patient (word) and client/consumer (word) seem to have become reminiscent: when the negative meanings attached to patient (word) affect client/consumer (word), an expression to replace it will be needed. Health care service users can be treated as a client/customer but be called patients. The results of this thesis can be most effectively utilized in health care management, planning, and development.

Luottamus vuorovaikutuksen pohjana on tärkeää huomioida opettajan työssä. Usein opettajien oletetaan tietävän, miten luottamusta rakennetaan suhteessa oppilaiden kanssa tai luottamuksen oletetaan syntyvän ikään kuin muun koulutyön ohessa. Tässä tutkimuksessa opettajalla ajatellaan kuitenkin olevan tärkeä ja aktiivinen asema luottamuksen rakentamisessa. Luottamuksesta olevat tutkimukset usein painottuvat sen rakentumisen edellytyksiin, eri elementteihin ja yleisempiin vaiheisiin, mutta niistä jää kokonaan pois näkökulma siitä, kuinka luottamusta todella rakennetaan arkikontekstissa konkreettisin teoin ja mitä välineitä opettajalla on rakentaa luottamusta. Teoreettinen viitekehys, välittämisen etiikka, pedagoginen rakkaus ja dialogisuus, antaa opettajalle myös paljon voimavaroja luottamuksellisen ilmapiirin rakentamisessa, jossa aidolla ja välittävällä kohtaamisella, avoimella keskustelulla ja lapsen huomioimisella on tärkeä merkitys. Tämän tutkimuksen aineistonkeruu on tapahtunut etnografisen menetelmän kautta havainnoiden erästä luokkayhteisöä kuukauden ajan syksyllä 2013. Erityisen mielenkiintoisen tästä asetelmasta tekee se, että luokan uusi opettaja ei ollut tavannut tulevia oppilaitaan aikaisemmin ja näin pääsin tutkijana havainnoimaan vuorovaikutuksen avausta ja luottamuksen rakentamista heti alusta. Kuukauden havainnointijakso päättyi opettajan haastatteluun, joka toimi ikään kuin tarkistuksena havainnoilleni, sillä siinä hänen äänensä tuli kuuluviin. Analyysissä puolestaan korostuu kokonaisuuden hahmottaminen, kielellisyyden tarkka pohdinta kentän sosiaalisen kontekstin vuoksi, aineiston kriittinen pohdinta ja analyysikysymysten johdattamat ajatusketjut. Tutkimuksesta käy ilmi, että ensimmäinen kuukausi vuorovaikutuksesta on luottamuksen kannalta erittäin tärkeää aikaa: siinä luodaan pohjaa myös tuleville arjen kohtaamisille. Tutkimuksen tuloksena on luottamuksen rakentumisen alun kolme vaiheitta, jotka ovat toisiin orientoituminen, arjessa asemoituminen ja yhteisen ymmärryksen tila. Jokaisessa vaiheessa opettajan on tärkeää saavuttaa tietyt asiat vuorovaikutuksessa, jotta seuraava vaihe voi alkaa. Kussakin vaiheessa opettajalla on käytössään erilaisia välineitä niiden asioiden täyttämiseen. Niitä ovat esimerkiksi toisen aktiivinen vastaanottaminen, sanavalinnat, yhdessä neuvottelu, jolloin oppilaisiinkin luotetaan ja symbolit. Luottamus vuorovaikutuksessa on erittäin tärkeää. Se auttaa tekemään toisten kohtaamisesta syvällisempää ja vaikuttaa yhteenkuuluvuuden tunteen, sitoutumisen, kunnioituksen ja luotetuksi tulemisen kautta monin tavoin hyvinvointiin. Luottamuksen pohja on menneisyydessä, mutta sen merkitys vain korostuu tulevaisuudessa.

Background Light is one of the most significant environmental factors affecting to the accumulation of flavonoids in fruits. The composition of the light spectrum has been shown to affect the production of phenolic compounds during fruit ripening. However, specific information on the biosynthesis of flavonoids in fruits in response to different wavelengths of light is still scarce. In the present study bilberry (Vaccinium myrtillus L.) fruits, which are known to be rich with anthocyanin compounds, were illuminated with blue, red, far-red or white light during the berry ripening process. Following the illumination, the composition of anthocyanins and other phenolic compounds was analysed at the mature ripening stage of fruits. Results All the three monochromatic light treatments had significant positive effect on the accumulation of total anthocyanins in ripe fruits compared to treatment with white light or plants kept in darkness. The elevated levels of anthocyanins were mainly due to a significant increase in the accumulation of delphinidin glycosides. A total of 33 anthocyanin compounds were detected in ripe bilberry fruits, of which six are novel in bilberry (cyanidin acetyl-3-O-galactose, malvidin acetyl-3-O-galactose, malvidin coumaroyl-3-O-galactose, malvidin coumaroyl-3-O-glucose, delphinidin coumaroyl-3-O-galactose, delphinidin coumaroyl-3-O-glucose). Conclusions Our results indicate that the spectral composition of light during berry development has significant effect on the flavonoid composition of ripe bilberry fruits. Hypericum perforatum L. (St. John’s wort) is a medicinal plant widely utilized for the treatment of depression. The antidepressant activity is mainly attributed to the phenolic compounds hypericins and hyperforins, which also have a wide range of other pharmacologically interesting properties. The biosynthetic routes leading to hypericins and hyperforins are poorly understood, although a polyketide pathway including type III polyketide synthases (PKSs) has been suggested to be involved. Furthermore, a gene called hyp-1 is assumed to attend to the final stages of the hypericin biosynthesis. In the present work, the biosynthesis of hypericins and hyperforins in H. perforatum was further studied by focusing on the elucidation of the precursors and genes involved. The incorporation of isotopically labelled branched-chain amino acids into hyperforins was investigated as well as the possibilities to enhance the production of hyperforins in H. perforatum in vitro cultures by feeding them with amino acid precursors. Furthermore, two novel cDNAs encoding for type III PKSs were isolated from H. perforatum. The functions of these new genes, designated HpPKS1 and HpPKS2, as well as the role of hyp-1 were elucidated by comparing their expression with the levels of hypericins and hyperforins in H. perforatum tissues. The enzymatic activity of the recombinant HpPKS2 protein was also analyzed. To study Hyp-1 at a protein level, a protein extraction method was optimized for tissues of Hypericum species. The results show the incorporation of valine and isoleucine into the acyl side chain of hyperforin and adhyperforin, respectively. Through the biotransformation of the amino acid precursors, it is possible to enhance the levels of adhyperforin, but not hyperforin, in H. perforatum shoot cultures, which demonstrates the tight regulation of the hyperforin biosynthesis. A correlation between HpPKS1 expression and hyperforins was detected in H. perforatum tissues. The localization of HpPKS2 mRNA in dark glands in which hypericins accumulate as well as the octaketide synthase activity of the recombinant HpPKS2 suggest that HpPKS2 is associated with possible co-operating tailoring enzymes in the biosynthesis of hypericins. The presence of both hyp-1 mRNA and Hyp-1 protein in distinct places compared with hypericins in H. perforatum tissues does not support the idea that Hyp-1 would be involved in the biosynthesis of hypericins in dark glands, although mobility of the Hyp-1 protein was shown to be possible. The present thesis extends knowledge about the biosynthesis of hypericins and hyperforins in H. perforatum by providing new candidate genes for their biosynthesis and by identifying precursors for hyperforins. Moreover, new information was obtained about the role of hyp-1 in H. perforatum.
