La Arteritis de Células Gigantes (ACG) es considerada uno de los síndromes vasculíticos más prevalentes. Afecta predominantemente a las arterias de mediano y gran calibre siendo la complicación más temida, una neuropatía óptica isquémica anterior que puede evolucionar a una pérdida irreversible de la visión. Los síntomas son causados por isquemia local debido al daño endovascular acompañado además de daño sistémico mediado por citoquinas. En los últimos años la ecografía doppler, la resonancia magnética y el PET scan fueron tomando importancia en la búsqueda de herramientas que puedan confirmar o excluir el diagnóstico de ACG sin embargo la biopsia de la arteria temporal (BAT) continúa siendo la prueba estándar para el diagnóstico de esta enfermedad.

El Escleredema de Buschke es una enfermedad poco común del tejido conectivo, de etiología desconocida que se caracteriza por el exceso de mucopolisacáridos ácidos en la dermis. Se describen tres tipos de acuerdo a su etiología. Clínicamente se caracteriza por induración cutánea simétrica que afecta en forma progresiva la región cervical, rostro, tórax, dorso y miembros, respetando manos y pies. El diagnóstico se basa en el cuadro clínico y los hallazgos histopatológicos característicos. No se ha establecido un tratamiento específico y efectivo, aunque se cita el uso de inmunosupresores, esteroides sistémicos y locales, así como fotoquimioterapia sistémica con psoralenos y radiación ultravioleta. En este artículo presentamos el caso de una mujer adulta con Escleredema de Buschke.

9. Characterization and potential utilization of recently available milk thistle, Silybum marianum (L.) Gaertn., wild type and mutant accessions

Milk thistle, Silybum marianum (L.) Gaertn. (Asteraceae), is an economically important medicinal plant utilized for silymarin production. Moreover, the species has been positively evaluated for vegetable oil and biomass production. Despite these positive characteristics, milk thistle is still marked by traits that are typical of undomesticated species (most importantly natural fruit dispersal at maturity) and requires further genetic improvement for its complete exploitation. This manuscript summarizes all the information collected through time about a collection of nine milk thistle wild and mutant lines and it discusses the possible further utilization of these genotypes. The accessions are characterized by interesting traits related to: fruit silymarin composition (S. marianum chemotype A and B), fruit fatty acid composition (high oleic and high stearic acid lines), fruit condensed tannins content, vegetative biomass composition (modification of xylans or lignin content), vegetative biomass structure (dwarf and tall lines), modifications of leaf variegation (hypervareigated line) and different types of fruit shatter resistance at maturity. All the lines underwent subsequent generations of selfing and are stable for all the described traits. The accessions will be made available at the Genebank of the Leibniz Institute of Plant Genetics and Crop Plant Research (IPK, Gatersleben) and may prove to be a useful genetic material for the improvement of qualitative fruit traits (silymarin quality, fatty acid composition) and for the further development of shatter-resistant S. marianum genotypes towards the complete domestication of this promising species.

The Giant Cell Arteritis is considered one of the most prevalent autoimmune vasculitis syndromes, which affects arteries of medium and large caliber, being the most feared complication, anterior ischemic optic neuropathy that can evolve to an irreversible loss of vision. The symptoms are caused by local ischemia due to endovascular damage accompanied by systemic damage mediated by cytokines. In recent years, doppler ultrasound, Magnetic Resonance Imaging (MRI) and PET scans have become important in the search for tools that can confirm or exclude the diagnosis of GCA. However, the Temporal Artery Biopsy (BAT) continues to be the gold standard test for the diagnosis of this disease.

Modern breeding of sunflower (Helianthus annuus L.), which started 100 years ago, increased the number and the diversity of cultivated forms. In addition, for more than 50 years, wild sunflower and other Helianthus species have been collected in North America where they all originated. Collections of both cultivated and wild forms are maintained in gene banks in many countries where sunflower is an important crop, with some specificity according to the availability of germplasm and to local research and breeding programmes. Cultivated material includes land races, open pollinated varieties, synthetics and inbred lines. The majority of wild accessions are ecotypes of wild Helianthus annuus, but also 52 other species of Helianthus and a few related genera. The activities of three gene banks, in USA, France and Serbia, are described in detail, supplemented by data from seven other countries. Past and future uses of the genetic resources for environmental adaptation and breeding are discussed in relation to genomic and improved phenotypic knowledge of the cultivated and wild accessions available in the gene banks. L’amélioration moderne du tournesol (Helianthus annuus L.) a débuté il y a un siècle, diversifiant et augmentant le nombre des formes cultivées du tournesol. De plus, des collectes de tournesols sauvages et d’espèces du genre Helianthus ont lieu depuis 50 ans en Amérique du Nord d’où ils sont tous originaires. Ainsi, des collections de tournesols cultivés et sauvages sont conservées par des centres de ressources génétiques dans de nombreux pays où le tournesol est une culture importante. Chacun d’eux présente des spécificités par rapport aux ressources génétiques maintenues, en fonction des programmes de recherche ou de sélection variétale locales. Le matériel génétique cultivé comprend des écotypes, des populations et des lignées tandis que les accessions sauvages correspondent eux écotypes d’Helianthus annuus sauvages et des 52 autres espèces apparentées du genre Helianthus. Les activités de trois centres de ressources génétiques des États-Unis, de la France et de la Serbie sont décrites en détail, complétées par des données provenant des centres de sept autres pays. L’historique de l’utilisation des ressources génétiques et les perspectives futures pour l’adaptation des variétés à l’environnement sont discutés ainsi que leur caractérisation au niveau génomique et phénotypique. Pouzdan i neometan rad plinskih turbina bitno ovisi o sustavu podmazivanja i kvaliteti mazivih ulja. U zadnje vrijeme raste tipična radna temperatura turbinskih ulja, budući da plinske turbine u proizvodnji struje postaju sve djelotvornije. Istovremeno se povećavaju zahtjevi za što dugotrajnijim uljima za plinske turbine. Kako bi se zadovoljili svi ti zahtjevi, sadašnja turbinska ulja trebaju imati odgovarajuću termooksidacijsku stabilnost. Međutim, nakon što su u sustav turbina uvedene nove kvalitete ulja za plinske turbine, pojavili su se novi izazovi s obzirom na stvaranje mulja, lakova i taloga. Ova onečišćenja i problemi vezani uz maziva ulja dovode do zastoja, gubitaka i dodatnih troškova održavanja. Kako bi se pojasnili razlozi i pozadina ove složene pojave, potrebno je provesti opsežna ispitivanja i studije o problemima vezanim za termooksidacijsku stabilnost., The reliable and trouble-free operation of the gas turbines is highly dependent upon the lubricating system and the lubricating oil performance. Recently the typical operating temperatures of turbine oils have been rising as modern gas turbines in power generation become more efficient. At the same time gas turbine oils are increasingly required to have longer service life. In order to fulfil all these requirements the current turbine oils need to possess adequate thermo-oxidation stability. However, as new quality of gas turbine oils have been introduced in turbine systems, new challenges appeared in regard of sludge, varnish and deposit formation. These contaminants and lubricating oil related problems led to downtimes, outages and extra maintenance costs. Due to clarify the reasons and backgrounds, extensive investigations and studies of the given thermo-oxidation stability problems are required for a better understanding of the complex phenomena.

Reactive oxygen species (ROS) and endothelin-1 (ET-1) contribute to vascular pathophysiology in obesity. In this context, whether ET-1 modulates hydroxyl radical (•OH) formation and the function of ROS/•OH in obesity is not known. In the present study, formation and function of ROS, including •OH, were investigated in the aorta of lean and leptin-deficient obese ob/ob mice. Hydroxyl radical formation was detected ex vivo using terephthalic acid in intact aortic rings and the involvement of ROS in ET-1-mediated vasoreactivity was analyzed using the antioxidant EPC-K1, a combination of α-tocopherol and ascorbic acid. Generation of either •OH, •O2−, and H2O2was strongly inhibited by EPC-K1 (all P < 0.05). In obese mice, basal vascular •OH formation and ROS activity were reduced by 3-fold and 5-fold, respectively ( P < 0.05 vs. lean). ET-1 markedly enhanced •OH formation in lean (6-fold, P < 0.05 vs. untreated) but not in obese mice. Obesity increased ET-1-induced contractions ( P < 0.05 vs. lean), and ROS scavenging further enhanced the response ( P < 0.05 vs. untreated). Exogenous ROS, including •OH caused stronger vasodilation in obese animals ( P < 0.05 vs. lean), whereas endothelium-dependent relaxation was similar between lean and obese animals. In conclusion, we present a sensitive method allowing ex vivo measurement of vascular •OH generation and provide evidence that ET-1 regulates vascular •OH formation. The data indicate that in obesity, vascular formation of ROS, including •OH is lower, whereas the sensitivity to ROS is increased, suggesting a novel and important role of ROS, including •OH in the regulation of vascular tone in disease status associated with increased body weight.

This study investigated whether intrarenal endothelin-1(ET-1) contributes to sodium excretion in aged rats. Metabolic function studies were performed in male Wistar rats (3 and 24 months) treated with placebo or the orally active ET(A) receptor antagonist darusentan (20 mg/kg/d) for 4 weeks. Mean arterial pressure was measured using an intra-arterial catheter. Electrolytes, aldosterone levels, renin activity, and angiotensin converting enzyme activity were determined in plasma, and mRNA expression of epithelial sodium channel (ENaC) and Na(+), K(+)-ATPase subunits was measured in the renal cortex and medulla. Aging was associated with a marked decrease in urinary excretion of sodium, chloride, and potassium (all P < 0.001) as well as renin activity (P < 0.05), but had no significant effect on gene expression of ENaC or Na(+), K(+)-ATPase subunits. In aged rats, darusentan treatment increased ion excretion (P < 0.05), reduced cortical gene expression of alphaENaC and alpha(1)-Na(+), K(+)-ATPase (both P < 0.05), and increased plasma aldosterone levels (P < 0.01). These data demonstrate a decrease of sodium and potassium excretion in aged rats, changes that are partly sensitive to ETA receptor blockade. Treatment with darusentan also reduced cortical expression of alphaENaC and alpha(1)-Na(+), K(+)-ATPase and increased plasma aldosterone levels independently of blood pressure, electrolytes, renin activity, or angiotensin converting enzyme activity. These findings may provide new pathogenetic links between aging and sodium sensitivity.

The article focuses on the underlying religious authoritative concern over superstition in the late Middle Ages in Europe. It explores developments in late medieval religious and intellectual culture, causing authorities to focus their concern on lay appropriations of ecclesiastical prayers, rites and blessings. It implies that the nature of spiritual being combined with certain intellectual developments and the growth of specific intellectual structures in the early 15th century will caused religious authorities to examine common beliefs and practices more closely than had previously been the case. Roman authorities had coined the term "superstitio" in anquity and had used it to describe excessive forms of divination, as well as the actions of suspicious foreign cults.

Reactive oxygen species (ROS) and endothelin-1 (ET-1) contribute to vascular pathophysiology in obesity. In this context, whether ET-1 modulates hydroxyl radical (.OH) formation and the function of ROS/·OH in obesity is not known. In the present study, formation and function of ROS, including ·OH, were investigated in the aorta of lean and leptin-deficient obese ob/ob mice. Hydroxyl radical formation was detected ex vivo using terephthalic acid in intact aortic rings and the involvement of ROS in ET-1-mediated vasoreactivity was analyzed using the antioxidant EPC-K1, a combination of α-tocopherol and ascorbic acid. Generation of either ·OH, ·O
2 - , and H2 O2 was strongly inhibited by EPC-K1 (all P < 0.05). In obese mice, basal vascular ·OH formation and ROS activity were reduced by 3-fold and 5-fold, respectively (P < 0.05 vs. lean). ET-1 markedly enhanced ·OH formation in lean (6-fold, P < 0.05 vs. untreated) but not in obese mice. Obesity increased ET-1-induced contractions (P < 0.05 vs. lean), and ROS scavenging further enhanced the response (P < 0.05 vs. untreated). Exogenous ROS, including ·OH caused stronger vasodilation in obese animals (P < 0.05 vs. lean), whereas endothelium-dependent relaxation was similar between lean and obese animals. In conclusion, we present a sensitive method allowing ex vivo measurement of vascular ·OH generation and provide evidence that ET-1 regulates vascular ·OH formation. The data indicate that in obesity, vascular formation of ROS, including ·OH is lower, whereas the sensitivity to ROS is increased, suggesting a novel and important role of ROS, including ·OH in the regulation of vascular tone in disease status associated with increased body weight. AIMS: To assess the performance of 2-sucrose-phosphate based transport medium (2-SP) for the detection of Chlamydia trachomatis and Neisseria gonorrhoeae by an automated commercial polymerase chain reaction (PCR) and ligase chain reaction (LCR) compared to centrifugation culture on McCoy cells for C trachomatis. Second, to compare both amplification systems for initial diagnostic testing of a low prevalence population for sexually transmitted diseases. METHODS: Four hundred and eighty one consecutive urogenital and conjunctival specimens were examined. All tests were performed on the same specimen collected with a dacron swab and transported in 2-SP medium. Samples that were positive by culture or by both PCR and LCR were considered to be true positives. RESULTS: The prevalences of C trachomatis and of N gonorrhoeae were 2.7% and 0.4%, respectively. PCR had a resolved sensitivity and specificity of 100% and 99.8%, respectively, for C trachomatis, and 100% and 98.9%, respectively, for N gonorrhoeae. LCR was 100% sensitive and specific for both pathogens. The resolved sensitivity of the shell vial assay was 85%. No culture positive sample would have been missed by PCR or LCR. The inhibition rate for PCR was 4.8%. CONCLUSIONS: 2-SP medium proved to be suitable for both PCR and LCR. It is not limited to any one test manufacturer and allows the performance of amplification techniques and viral and chlamydia culture from the same specimen. The LCR was more reliable than PCR on initial testing. However, hands on time is longer, and no amplification control is available for LCR. [ABSTRACT FROM PUBLISHER]

Enteroviruses affect millions of people worldwide and are of significant clinical importance. The standard method for enterovirus identification and genotyping still relies on Sanger sequencing of short diagnostic amplicons. In this study, we assessed the feasibility of nanopore sequencing using the new flow cell "Flongle" for fast, cost-effective, and accurate genotyping of human enteroviruses from clinical samples. PCR amplification of partial VP1 gene was performed from multiple patient samples, which were multiplexed together after barcoding PCR and sequenced multiple times on Flongle flow cells. The nanopore consensus sequences obtained from mapping reads to a reference database were compared to their Sanger sequence counterparts. Using clinical specimens sampled over different years, we were able to correctly identify enterovirus species and genotypes for all tested samples, even when doubling the number of barcoded samples on one flow cell. Average sequence identity across sequencing runs was >99.7%. Phylogenetic analysis showed that the consensus sequences achieved with Flongle delivered accurate genotyping. We conclude that the new Flongle-based assay with its fast turnover time, low cost investment, and low cost per sample represents an accurate, reproducible, and cost-effective platform for enterovirus identification and genotyping. Las doce contribuciones que integran el libro, escritas por un equipo de reconocidos expertos internacionales en la materia, plantea un ambicioso recorrido que tiene la virtud de cubrir gran parte del arco temporal que abarcó la caza de brujas europea, desde los entusiasmos iniciales de los siglos XIV y XV hasta las tardías polémicas de finales del siglo XVII. En efecto, el recorrido se inicia en la década de 1320 con la bula'Super illius specula'de Juan XXII y culmina en la década de 1690, con'De Betoverde Weereld'de Balthasar Bekker. Este trayecto tiene poco de casual. No cabe atribuir al azar el hecho de que el punto de partida del itinerario sea la curia pontificia, por entonces asentada en la cálida sede provenzal de Aviñón, ni que el punto de llegada sea la marítima y fría Ámsterdam, trinchera de la libertad de consciencia en vísperas del Siglo de las Luces. Entre estos dos extremos cronológicos, que encierran la fase de apogeo de la demonología radical y de la caza de brujas europea, Satán desplegó una panoplia infinita de rostros, máscaras y semblantes, propios de su naturaleza angélica proteiforme. Ésta, capaz de adoptar cualquier disfraz o apariencia, se mantuvo omnipresente como dispositivo cultural diseñado para otorgar sentido al angustiante universo de los hombres y mujeres del Medioevo tardío y de la Modernidad temprana.

Im Einklang mit den jüngeren curricularen Entwicklungen offenbart die institutionelle Segmentierung des allgemein- und berufsbildenden Schulwesens eine insbesondere für die Leitfächer der politischen Bildung problematische Verkürzung des (Allgemein-)Bildungsanspruchs. Statt die gesellschaftspolitische Wirkmächtigkeit politischer Bildung zu akzentuieren, dominieren vielfach entpolitisierende „Kammerprüfungen“ den Politikunterricht. Angesichts der wachsenden Kluft zwischen der „politischen Elite“ einerseits und den sozioökonomisch weniger privilegierten Bevölkerungsgruppen andererseits, ist diese Entwicklung nicht nur pädagogisch, sondern auch politisch in besonderer Weise bedenklich. Vor diesem Hintergrund werden im vorliegenden Buch konstitutive Merkmale des Politikunterrichts in der dualen Ausbildung erörtert. Dazu werden historisch gewachsene Strukturen sowie aktuelle curriculare Entwicklungen der politischen Bildung an Berufsschulen problematisiert, um anschließend – ausgehend vom politikdidaktischen Prinzip der Lebensweltorientierung – die Gesellschaftsdeutungen von Auszubildenden hinsichtlich ihrer Teilhabeerfahrungen und -vorstellungen auszuwerten.
