Perfluorooctanesulfonic acid (PFOS) is a neurotoxic widespread organic contaminant which affects several brain functions including memory, motor coordination and social activity. PFOS has the ability to traverse the placenta and the blood brain barrier (BBB) and cause weight gain in female mice. It’s also known that obesity and consumption of a high fat diet have negative effects on the brain, impairs cognition and increases the risk for the development of dementia. The combination effect of developmental exposure to PFOS and the intake of a high-fat diet (HFD) has not been explored. This study investigates the effect of PFOS and /or HFD on weight gain, behavior and transcriptomic and proteomic analysis of adult brain mice. We found that female mice exposed to PFOS alone showed an increase in weight, while HFD expectedly increased body weight. The combination of HFD and PFOS exacerbated generalized behavior such as time spent in the center and rearing, while PFOS alone impacted the distance travelled. These results suggest that PFOS exposure may promote hyperactivity. The combination of PFOS and HFD alter social behavior such as rearing and withdrawal. Although HFD interfered with memory retrieval, biomarkers of dementia did not change except for total Tau and phosphorylated Tau. Tau was impacted by either or both PFOS exposure and HFD. Consistent with behavioral observations, global cerebral transcriptomic analysis showed that PFOS exposure affects calcium signaling, MAPK pathways, ion transmembrane transport, and developmental processes. The combination of HFD with PFOS enhances the effect of PFOS in the brain and affects pathways related to ER stress, axon guidance and extension, and neural migration. Proteomic analysis showed that HFD enhances the impact of PFOS on inflammatory pathways, regulation of cell migration and proliferation, and MAPK signaling pathways. Overall, these data show that PFOS combined with HFD may reprogram the genome and modulate neuromotor development and may promote symptoms linked to attention deficit-hyperactivity disorders (ADHD) and autism spectrum disorders (ASD). Future work will be needed to confirm these connections.

Introduction: Autism spectrum disorder (ASD) is characterized by aberrations in social interaction and communication associated with repetitive behaviors and interests, with strong clinical heterogeneity. Genetic factors play an important role in ASD, but about 75% of ASD cases have an undetermined genetic risk.Methods: We extensively investigated an ASD cohort made of 102 families from the Middle Eastern population of Qatar. First, we investigated the copy number variations (CNV) contribution using genome-wide SNP arrays. Next, we employed Next Generation Sequencing (NGS) to identify de novo or inherited variants contributing to the ASD etiology and its associated comorbid conditions in families with complete trios (affected child and the parents).Results: Our analysis revealed 16 CNV regions located in genomic regions implicated in ASD. The analysis of the 88 ASD cases identified 41 genes in 39 ASD subjects with de novo (n = 24) or inherited variants (n = 22). We identified three novel de novo variants in new candidate genes for ASD (DTX4, ARMC6, and B3GNT3). Also, we have identified 15 de novo variants in genes that were previously implicated in ASD or related neurodevelopmental disorders (PHF21A, WASF1, TCF20, DEAF1, MED13, CREBBP, KDM6B,SMURF1, ADNP, CACNA1G, MYT1L, KIF13B, GRIA2, CHM, and KCNK9). Additionally, we defined eight novel recessive variants (RYR2, DNAH3, TSPYL2, UPF3B KDM5C, LYST, and WNK3), four of which were X-linked.Conclusion: Despite the ASD multifactorial etiology that hinders ASD genetic risk discovery, the number of identified novel or known putative ASD genetic variants was appreciable. Nevertheless, this study represents the first comprehensive characterization of ASD genetic risk in Qatar's Middle Eastern population.

Category: Bunion Introduction/Purpose: The aim of this study was to evaluate the efficacy of percutaneous hemiepiphysiodesis for JHV by investigating the available literature on the procedure. Methods: The literature search was conducted by using the following bibliographic electronic databases: EMBASE, MEDLINE, and PubMed.gov. Study screening at all stages was completed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-analyses (PRISMA) criteria. The Methodological Index for Non-Randomized Studies (MINORS) scale was used to evaluate the quality of included studies. Data on patient characteristics, radiological and clinical outcomes, and notable complications were also collected. Results: Of the 91 studies identified by our search strategy, 4 met the inclusion criteria. The identified studies included 62 patients and 109 feet. The mean hallux valgus angle (HVA) improved preoperative to postoperative at a range of Δ3.45 to Δ5.5. The mean intermetastartal angle (IMA) improved preoperative to postoperative at a range of Δ2.2 to Δ4.0. The mean HVA progression per month improved from preoperative to postoperative at a range of Δ0.07 degrees/month to Δ0.17 degrees/month. The mean IMA progression per month improved from preoperative to postoperative at a range of Δ0.046 degrees/month to Δ0.067 degrees/month. Of the studies that reported a mean American Orthopaedic Foot and Ankle Society (AOFAS) score or a mean Hallux Metatarsophalangeal Interphalangeal Scale (HMIS), there was an improvement at a range of Δ16.5 to Δ36. Conclusion: Percutaneous Hemiepiphysiodesis can serve as an effective procedure for treating Juvenile Hallux Valgus.

Body cavity macrophages reside on the serous surfaces of organs and believed to participate in organ repair following injury. Here the authors show with a fate-mapping reporter system that these cells, although accumulate at the surfaces of injured liver or lung, don’t penetrate deeply into the tissue.

The nuclease activity of the CRISPR-Cas9 system relies on the delivery of a CRISPR-associated protein 9 (Cas9) and a single guide RNA (sgRNA) against the target gene. CRISPR components are typically delivered to cells as either a Cas9/sgRNA ribonucleoprotein (RNP) complex or a plasmid encoding a Cas9 protein along with a sequence-specific sgRNA. Multiple transfection reagents are known to deliver CRISPR-Cas9 components, and delivery vectors are being developed for different purposes by several groups. Here, we repurposed a dual-fluorescence (RFP-GFP-GFP) reporter system to quantify the uptake level of the functional CRISPR-Cas9 components into cells and compare the efficiency of CRISPR delivery vectors. Using this system, we developed a novel and rapid cell-based microplate reader assay that makes possible real-time, rapid, and high throughput quantification of CRISPR nuclease activity. Cells stably expressing this dual-fluorescent reporter construct facilitated a direct quantification of the level of the internalized and functional CRISPR-Cas9 molecules into the cells without the need of co-transfecting fluorescently labeled reporter molecules. Additionally, targeting a reporter gene integrated into the genome recapitulates endogenous gene targeting. Thus, this reporter could be used to optimize various transfection conditions of CRISPR components, to evaluate and compare the efficiency of transfection agents, and to enrich cells containing desired CRISPR-induced mutations.

Parkinson’s disease (PD) is a complex multifactorial disorder that is not yet fully surmised, and it is only when such a disease is tackled on multiple levels simultaneously that we should expect to see fruitful results. Gene therapy is a modern medical practice that theoretically and, so far, practically, has demonstrated its capability in joining the battle against PD and other complex disorders on most if not all fronts. This review discusses how gene therapy can efficiently replace current forms of therapy such as drugs, personalized medicine or invasive surgery. Furthermore, we discuss the importance of enhancing delivery techniques to increase the level of transduction and control of gene expression or tissue specificity. Importantly, the results of current trials establish the safety, efficacy and applicability of gene therapy for PD. Gene therapy’s variety of potential in interfering with PD’s pathology by improving basal ganglial circuitry, enhancing dopamine synthesis, delivering neuroprotection or preventing neurodegeneration may one day achieve symptomatic benefit, disease modification and eradication.

Abstract Oxygen-responsive promoters can be useful for synthetic biology applications, however, information on their characteristics is still limited. Here, we characterized a group of heterologous microaerobic globin promoters in Escherichia coli. Globin promoters from Bacillus subtilis, Campylobacter jejuni, Deinococcus radiodurans, Streptomyces coelicolor, Salmonella typhi and Vitreoscilla stercoraria were used to express the FMN-binding fluorescent protein (FbFP), which is a non-oxygen dependent marker. FbFP fluorescence was monitored online in cultures at maximum oxygen transfer capacities (OTRmax) of 7 and 11 mmol L−1 h−1. Different FbFP fluorescence intensities were observed and the OTRmax affected the induction level and specific fluorescence emission rate (the product of the specific fluorescence intensity multiplied by the specific growth rate) of all promoters. The promoter from S. typhi displayed the highest fluorescence emission yields (the quotient of the fluorescence intensity divided by the scattered light intensity at every time-point) and rate, and together with the promoters from D. radiodurans and S. coelicolor, the highest induction ratios. These results show the potential of diverse heterologous globin promoters for oxygen-limited processes using E. coli.

Abstract Background Dissolved oxygen tension (DOT) is hardly constant and homogenously distributed in a bioreactor, which can have a negative impact in the metabolism and product synthesis. However, the effects of DOT on plasmid DNA (pDNA) production and quality have not been thoroughly investigated. In the present study, the effects of aerobic (DOT ≥30% air sat.), microaerobic (constant DOT = 3% air sat.) and oscillatory DOT (from 0 to 100% air sat.) conditions on pDNA production, quality and host performance were characterized. Results Microaerobic conditions had little effect on pDNA production, supercoiled fraction and sequence fidelity. By contrast, oscillatory DOT caused a 22% decrease in pDNA production compared with aerobic cultures. Although in aerobic cultures the pDNA supercoiled fraction was 98%, it decreased to 80% under heterogeneous DOT conditions. The different oxygen availabilities had no effect on the fidelity of the produced pDNA. The estimated metabolic fluxes indicated substantial differences at the level of the pentose phosphate pathway and TCA cycle under different conditions. Cyclic changes in fermentative pathway fluxes, as well as fast shifts in the fluxes through cytochromes, were also estimated. Model-based genetic modifications that can potentially improve the process performance are suggested. Conclusions DOT heterogeneities strongly affected cell performance, pDNA production and topology. This should be considered when operating or scaling-up a bioreactor with deficient mixing. Constant microaerobic conditions affected the bacterial metabolism but not the amount or quality of pDNA. Therefore, pDNA production in microaerobic cultures may be an alternative for bioreactor operation at higher oxygen transfer rates.

The first Oregon case of New Delhi metallo-β-lactamase-1 (NDM-1)-producing Escherichia coli was reported during November 2013. Epidemiologic investigation revealed only local outpatient medical care and no travel outside Oregon for both the patient and his household contact. Environmental sampling discovered a matching isolate from the patient’s household vacuum cleaner, suggesting environmental persistence.

Introducing the management of non traumatic rhegmatogenous retinal detachment in pediatric patient in the context of a genetic syndrome associated with failure of type 2 collagen synthesis which manifests itself in ocular , hearing , facial , bone and joint abnormalities. We describe the clinical presentation of the picture and surgical repaire technique consisted of lensectomy , vitrectomy , peeling of vitreoretinal proliferation membranes , giant retinal tear management and prolonged tamponade by silicone oil. Long term clinical follow-up was carried out. We highlight this case report to be a therapeutic challenge in a deaf and high myopic child , with motor disabilities and atypical vitreoretinal disorders have generated difficulties in surgical resolution , there is also little scientific reference in world literature and represent a clinical model of abnormal vitreoretinal interaction

Educational resources can be easily found on the Web. Most search engines base their algorithms on a resource's text or popularity, requiring teachers to navigate the results until they find an appropriate resource. This makes searching for resources a tedious and cumbersome task. Specialized repositories contain resources that are annotated with metadata that aim to facilitate the discovery of quality resources. Nevertheless, the abundance and variety of content make searching a complex task. Recommender systems can assist teachers in finding the proper content by determining clusters of similar users and inferring users interest in a resource, assembling clusters of similar resources, or a mix of both. Probabilistic model-based (PMB) techniques, on the other hand, make it possible to classify resources into more than one cluster with various degrees of probability. In this paper, we used recommender systems and PMB techniques to analyze a dataset produced by primary and secondary level teachers over the course of four years and under natural conditions. We found that a hybrid recommendation, the Collaborative Topic Regression (CTR) technique, performs better than other approaches despite the high sparseness of the dataset. In addition, learning resources annotated with curriculum metadata had a positive impact on recommender's accuracy whereas free-text or other metadata negatively impacted the results.

This article introduces a new family of orthogonal moments, the fractional Racah moments (FrROMs), developed to meet the demands of copyright protection for medical images, a field requiring robust and secure methods that preserve the integrity of the original image. The FrROMs are derived from a matrix of fractional Racah orthogonal polynomials (FrROPs), which in turn are obtained from the matrix of classical Racah polynomials (ROPs) using the spectral decomposition theorem. To compute the ROPs, we enhanced a method for calculating the initial terms of these polynomials to avoid instability issues associated with higher-order ROPs. Subsequently, the FrROMs are used as descriptors for embedding and extracting a zero-watermark. Experimental results demonstrate that the proposed zero-watermarking scheme efficiently leverages the FrROMs, surpassing classical ROMs in terms of robustness and reconstruction capability. This scheme provides strong protection against various image processing attacks while maintaining the original content intact. Compared to existing methods, it stands out for its enhanced robustness and reliability, making a significant contribution to the secure protection of medical images.

Abstract Introduction Direct oral anticoagulant (DOAC) dose adjustment is based on age, renal function, and body weight. There is a paucity of data describing the factors associated with the prescription of inappropriate dosage and their impact on clinical outcomes among patients receiving transcatheter aortic valve implantation (TAVI). Methods In a single-center study, 432 patients who were on long-term DOAC therapy and underwent TAVI between 2015 and 2022 were included. We analyzed the predictors and outcomes of inappropriate dosing of DOACs; namely apixaban, dabigatran, edoxaban, and rivaroxaban. A composite endpoint, including all-cause mortality, life-threatening/major bleeding, stroke, peripheral thromboembolic complications, or myocardial infarction, was assessed after 1 year. Results In this TAVI cohort, inappropriate DOAC dosing was observed in 20.6% of patients. Abstract Background The introduction of quantitative SPECT/CT allows more objective assessments of tracer accumulation in SPECT. However standardized uptake values (SUV) still do not play a big role for orthopedic or oncologic questions. With a more reliable normalization, the use of quantitative measures might also be of use for a more objective assessment of lesions. We retrospectively included patients that received a quantitative [99mTc]-HDP bone SPECT/CT scan of the lumbar spine for 4 body weight (BW) categories. Measurements of bone activity (kBq/cc) and bone density in Hounsfield Units (HU) in a standard volume of interest in the femur, the first and the fifth lumbar vertebra of all patients, without active disease within these regions was made. Correlations between tracer uptake and clinical parameters (BW, height, age, gender) were assessed with a multiple regression and based on the model coefficients, a correction formula was calculated and applied. The dietary recommendations for individuals with diabetes focus on maintaining a balanced nutritional intake to manage blood sugar levels. This study suggests a nutritional strategy to improve glycemic control based on an analysis of a dietary optimization problem. The goal is to minimize the overall glycemic loads (GLs) of specific foods. Two variations of the particle swarm optimization (PSO) method, as well as random quantum process optimization (GQPSO), are introduced. The findings demonstrate that the quantum and random methods are more effective than the traditional techniques in reducing the glycemic loads of diets and addressing nutritional deficiencies while also aligning nutrient intake with the recommended levels. The resolution of this diet optimization model, executed multiple times with adjustments to the parameters of both methods, enables dynamic exploration and provides a wide range of diverse and effective food choices.

Abstract Waste and energy sectors have significant contributions to the greenhouse gas (GHG) emissions caused primarily by the population expansion. Waste-to-Energy (WtE) is introduced to address the issue raised by both sectors simultaneously through utilization of the potential energy stored in municipal solid waste (MSW) as well as offsetting GHG emissions. Limited research have been conducted in Egypt to assess the current situation of MSW management and associated methane emissions. The current study focused on estimating the baseline methane emissions for six Egyptian governorates and determining the energy production potential from WtE projects. To achieve this aim, three scenarios have been assessed: Baseline, Landfill Gas to Energy (LFGE), and Incineration scenarios. Key results revealed that a total of 3.7 million tonnes of methane would be emitted from all studied governorates generated over 50 years. Incineration also found to be more favorable in all governorates in terms of energy production, quantity of avoided GHG emissions, and in terms of economic viability over LFGE. Implementing incineration in all governorates would generate about 5.6 TWh energy annually and could avoid about 5 Mt CO2 eq annually in comparison to LFGE that would generate about 0.6 TWh annually and could avoid about 2.5 Mt CO2 eq annually. In terms of economic viability of WtE projects, while they were generally not economically viable under the assumptions made in the current study, incineration technology deemed promising, but policy adjustments, such as competitive Feed-in Tariff (FiT) rates and the inclusion of gate fees, are necessary. Specific minimum gate fees and FiT were identified for each governorate, providing essential guidance for decision makers to ensure the viability of WtE implementation. This study would support the decision makers in assessing technically and financially feasible options for WtE technologies in the selected governorates.

Environmental stress poses serious threats to animal welfare and production, particularly in poultry, which are susceptible to such stress. It can increase susceptibility to diseases and infections, reduce growth rates and reproductive performance, and increase behavioral issues. Environmental stress caused by conventional housing conditions can negatively affect well-being and productivity. High temperature, overcrowding, poor ventilation, insufficient lighting, and wire cages are some of the most prominent stressors in conventional housing systems. To address environmental stress in chicken farms, some strategies and tools, such as using anti-stress feed additives and enriching cages, can help improve bird behavioral activities and welfare. Breeders can improve overall bird performance by implementing these strategies and creating a more enriched and comfortable environment. Thus, this review discusses the importance of using different feed additives and environmental enrichment materials to reduce stress in chicken farms (broiler and layer) and improve bird productivity and well-being.

Abstract There are many extensions of the Standard Model with a dark matter (DM) candidate obtained via the freeze-out mechanism. It can happen that after all experimental and theoretical constraints are taken into account, all parameter points have a relic density below the experimentally measured value. This means that the models solve only partially the DM problem, and at least one more candidate is needed. In this work we show that it is possible to further extend the model with a DM candidate obtained via the freeze-in mechanism to be in agreement with the relic density experimental measurement. Once the relic density problem is solved with this addition, new questions are raised. This new model with at least two DM candidates could have a freeze-out undetectable DM particle both in direct and indirect detection. This could happen if the freeze-out DM particle would have a very low density. Hence, a collider DM hint via excess in the missing energy with no correspondence in direct and indirect detection experiments, could signal the existence of a Feebly Interacting Massive Particle (FIMP). Conversely, if a DM particle is found and a particular model can explain all observables except the correct relic density, an extension with an extra FIMP would solve the problem. The freeze-in DM candidate, due to the small portal couplings, will not change the remaining phenomenology.
