Introduction: Schizophrenia is a major mental illness and a significant contributor to the global burden of disease. Around one-fifth of patients with Schizophrenia have significant depression during the phase of clinical remission. Aim: To evaluate the prevalence of depression in patients with Schizophrenia, the Quality of Life (QoL) of patients with depression in Schizophrenia, and to study the relationship between the two. Materials and Methods: This cross-sectional study was conducted at the Department of Psychiatry, Chettinad Hospital and Research Institute, Chennai, Tamil Nadu, India. One hundred patients aged 18 to 59 years diagnosed with Schizophrenia as per International Classification of Diseases (ICD-11), operationally in remission for a minimum of one month, were included. The Calgary Depression Scale for Schizophrenia (CDSS) and the World Health Organisation Quality of Life Brief version (WHOQoL-BREF) scale were used to measure the presence of depression and QoL in patients with Schizophrenia. Data were analysed with t-test, Chi-square tests, and Pearson correlation using Statistical Package for Social Sciences (SPSS) software version 21.0. Results: The mean age of the study participants was 31.6±5.1 years. Of the total study population, 72% were male, 31% had a high school level education, 20% were unemployed, 64% were married, 46% were from a semi-urban background, and 45% belonged to a lower-middle socio-economic background. Twenty-two percent of patients with Schizophrenia in remission were found to have depression. A longer duration of untreated psychosis (mean=9.14±2.83 years) was significantly associated with the development of depression in patients with Schizophrenia. We describe here a case of nasal polyp of the left nose due to Trichoderma harzianum, an emerging fungal infection with an often fatal outcome. Culture showed growth of T. harzianum which was identified by cultural characteristics, microscopic morphology, and molecular methods. The patient was initially treated with a combination of surgical removal of the polyp and oral antibiotics. This case points out that careful scrutiny of nasal polyp is required to ensure accurate diagnosis and appropriate management of cases without recurrence.

Abstract The present work investigates the water wave interaction with bottom-standing thick porous trapezoidal-shaped structures and shore-side vertical rigid wall in the presence of uniform ocean currents. This study has been done to understand the impact of different physical parameters like friction, porosity, and ocean currents along with different structural parameters (width and height) on different phenomena like wave energy reflection, wave forces, wave energy dissipation, etc. The quadratic boundary element method-based numerical technique has been used to solve the boundary value problem. The structural porosity is modeled using Sollitt and Cross’s model of water wave interaction with thick porous structures. Several results associated with the wave energy reflection and energy dissipation have been analyzed. Also, the wave force exerted by the incoming waves has been investigated to check the stability and sustainability of the right vertical rigid wall and porous structure. The Doppler-shift effect is observed in wave transformation characteristics due to the presence of ocean currents. The impact of following and opposing ocean currents can be seen in the graph of wave energy reflection, dissipation, and wave forces. The periodic patterns can be observed clearly in wave characteristics like wave energy reflection, dissipation, and wave forces when plotted against the non-dimensional separation gap between the porous breakwater and shore-side rigid seawall.

Abstract Studies on the neural correlates of navigation in 3D environments are plagued by several issues that need to be solved. For example, experimental studies show markedly different place cell responses in rats and bats, both navigating in 3D environments. In this study, we focus on modelling the spatial cells in rodents in a 3D environment. We propose a deep autoencoder network to model the place and grid cells in a simulated agent navigating in a 3D environment. The input layer to the autoencoder network model is the HD layer, which encodes the agent’s HD in terms of azimuth (θ) and pitch angles (ϕ). The output of this layer is given as input to the Path Integration (PI) layer, which computes displacement in all the preferred directions. The bottleneck layer of the autoencoder model encodes the spatial cell-like responses. Both grid cell and place cell-like responses are observed. The proposed model is verified using two experimental studies with two 3D environments. This model paves the way for a holistic approach using deep neural networks to model spatial cells in 3D navigation.

Abstract Background The high prevalence of osteoporosis has increased the economic burden on the health system globally. The burden of osteoporosis and its associated factors have not been adequately assessed in community settings in the Nepalese context thus far. Therefore, this study aimed to assess the prevalence of osteoporosis and its associated factors, lifestyle behaviors, and dietary calcium intake. Methods A community-based cross-sectional study was conducted among 395 people aged 50 years and older in the Madhesh Province of Nepal between July 2022 and August 2023. The Osteoporosis Self-assessment Tools for Asians (OSTA) index was used to measure osteoporosis. A structured questionnaire was used to collect sociodemographic information, anthropometric data, lifestyle behavior, daily dietary calcium intake, and frequency of calcium-rich food consumption. A food frequency questionnaire and 24-hour recall methods were used to assess dietary intake. The chi-square test, binary logistic regression and Mann‒Whitney U test were applied to measure the association between predictors and the outcome of interest. Abstract Parkinson’s disease is increasingly prevalent. It progresses from the pre-motor stage (characterised by non-motor symptoms like REM sleep behaviour disorder), to the disabling motor stage. We need objective biomarkers for early/pre-motor disease stages to be able to intervene and slow the underlying neurodegenerative process. Here, we validate a targeted multiplexed mass spectrometry assay for blood samples from recently diagnosed motor Parkinson’s patients (n = 99), pre-motor individuals with isolated REM sleep behaviour disorder (two cohorts: n = 18 and n = 54 longitudinally), and healthy controls (n = 36). Our machine-learning model accurately identifies all Parkinson patients and classifies 79% of the pre-motor individuals up to 7 years before motor onset by analysing the expression of eight proteins—Granulin precursor, Mannan-binding-lectin-serine-peptidase-2, Endoplasmatic-reticulum-chaperone-BiP, Prostaglaindin-H2-D-isomaerase, Interceullular-adhesion-molecule-1, Complement C3, Dickkopf-WNT-signalling pathway-inhibitor-3, and Plasma-protease-C1-inhibitor. Many of these biomarkers correlate with symptom severity. This specific blood panel indicates molecular events in early stages and could help identify at-risk participants for clinical trials aimed at slowing/preventing motor Parkinson’s disease.

Abstract Background There is a lack of contemporary data describing global variations in vascular access for hemodialysis (HD). We used the third iteration of the International Society of Nephrology Global Kidney Health Atlas (ISN-GKHA) to highlight differences in funding and availability of hemodialysis accesses used for initiating HD across world regions. Methods Survey questions were directed at understanding the funding modules for obtaining vascular access and types of accesses used to initiate dialysis. An electronic survey was sent to national and regional key stakeholders affiliated with the ISN between June and September 2022. Countries that participated in the survey were categorized based on World Bank Income Classification (low-, lower-middle, upper-middle, and high-income) and by their regional affiliation with the ISN. Results Data on types of vascular access were available from 160 countries. Respondents from 35 countries (22% of surveyed countries) reported that > 50% of patients started HD with an arteriovenous fistula or graft (AVF or AVG). These rates were higher in Western Europe (n = 14; 64%), North & East Asia (n = 4; 67%), and among high-income countries (n = 24; 38%). The rates of > 50% of patients starting HD with a tunneled dialysis catheter were highest in North America & Caribbean region (n = 7; 58%) and lowest in South Asia and Newly Independent States and Russia (n = 0 in both regions). Respondents from 50% (n = 9) of low-income countries reported that > 75% of patients started HD using a temporary catheter, with the highest rates in Africa (n = 30; 75%) and Latin America (n = 14; 67%). Funding for the creation of vascular access was often through public funding and free at the point of delivery in high-income countries (n = 42; 67% for AVF/AVG, n = 44; 70% for central venous catheters). In low-income countries, private and out of pocket funding was reported as being more common (n = 8; 40% for AVF/AVG, n = 5; 25% for central venous catheters). Conclusions High income countries exhibit variation in the use of AVF/AVG and tunneled catheters. In low-income countries, there is a higher use of temporary dialysis catheters and private funding models for access creation.

Abstract Connectome studies have shown how Alzheimer’s disease (AD) disrupts functional and structural connectivity among brain regions. But the molecular basis of such disruptions is less studied, with most genomic/transcriptomic studies performing within-brain-region analyses. To inspect how AD rewires the correlation structure among genes in different brain regions, we performed an Inter-brain-region Differential Correlation (Inter-DC) analysis of RNA-seq data from Mount Sinai Brain Bank on four brain regions (frontal pole, superior temporal gyrus, parahippocampal gyrus and inferior frontal gyrus, comprising 264 AD and 372 control human post-mortem samples). An Inter-DC network was assembled from all pairs of genes across two brain regions that gained (or lost) correlation strength in the AD group relative to controls at FDR 1%. The differentially correlated (DC) genes in this network complemented known differentially expressed genes in AD, and likely reflects cell-intrinsic changes since we adjusted for cell compositional effects. Each brain region used a distinctive set of DC genes when coupling with other regions, with parahippocampal gyrus showing the most rewiring, consistent with its known vulnerability to AD. The Inter-DC network revealed master dysregulation hubs in AD (at genes ZKSCAN1, SLC5A3, RCC1, IL17RB, PLK4, etc.), inter-region gene modules enriched for known AD pathways (synaptic signaling, endocytosis, etc.), and candidate signaling molecules that could mediate region-region communication. The Inter-DC network generated in this study is a valuable resource of gene pairs, pathways and signaling molecules whose inter-brain-region functional coupling is disrupted in AD, thereby offering a new perspective of AD etiology.

Agricultural management practices alter soil characteristics and influence soil biological properties. Hence, a field trial was carried out to assess the 14-year long-term impact of tillage and residue management practices on soil biological activities and microbial population in a rice-wheat cropping system in two depths viz., 0-15 and 15-30 cm. Soil organic carbon levels differed significantly (p> 0.05) across various treatments. Microbial biomass carbon, Microbial quotient, and soil enzymatic activities were significantly greater (10-82%) in crop residue incorporation/retention treatments. Zero tillage with residue retention (ZT+R) had the greatest bacterial, actinomycetes, and fungi population, next to zero tillage with residue incorporation (RT+R). The ZT+R treatment had the greatest value of K-strategist and r-strategist, and was equivalent to RT+R across both soil depths. When compared to Conventional tillage (CT), zero tillage (ZT) increased wheat yield by 9%. However, compared to CT, rice and rice-wheat systems had lower grain yields, whereas crop residue increased wheat and rice-wheat system yields by 10% and 6%, respectively. The findings of this long-term study show that residue management and tillage practices can enhance soil biological attributes while also supporting microbial diversity.

Cardiolipin (CL) is a unique, four-chain phospholipid synthesized in the inner mitochondrial membrane (IMM). The acyl chain composition of CL is regulated through a remodeling pathway, whose loss causes mitochondrial dysfunction in Barth syndrome (BTHS). Yeast has been used extensively as a model system to characterize CL metabolism, but mutants lacking its two remodeling enzymes, Cld1p and Taz1p, exhibit mild structural and respiratory phenotypes compared to mammalian cells. Here, we show an essential role for CL remodeling in the structure and function of the IMM in yeast grown under reduced oxygenation. Microaerobic fermentation, which mimics natural yeast environments, caused the accumulation of saturated fatty acids and, under these conditions, remodeling mutants showed a loss of IMM ultrastructure. We extended this observation to HEK293 cells, where phospholipase A2 inhibition by Bromoenol lactone resulted in respiratory dysfunction and cristae loss upon mild treatment with exogenous saturated fatty acids. In microaerobic yeast, remodeling mutants accumulated unremodeled, saturated CL, but also displayed reduced total CL levels, highlighting the interplay between saturation and CL biosynthesis and/or breakdown. We identified the mitochondrial phospholipase A1 Ddl1p as a regulator of CL levels, and those of its precursors phosphatidylglycerol and phosphatidic acid, under these conditions. Loss of Ddl1p partially rescued IMM structure in cells unable to initiate CL remodeling and had differing lipidomic effects depending on oxygenation. These results introduce a revised yeast model for investigating CL remodeling and suggest that its structural functions are dependent on the overall lipid environment in the mitochondrion.

Malaria is still a public health problem in tropical countries like India; major malaria parasite species are Plasmodium falciparum and P. vivax. Of which, P. vivax is responsible for ∼40% of the malaria burden at least in the Indian scenario. Unfortunately, there is limited data on the population structure and genetic diversity of P. vivax parasites in India. In this study, we investigated the genetic diversity of P. vivax strains in the South-west district, Delhi and, Nuh district, Haryana [National Capital Region (NCR)], using a polymorphic marker- P. vivax merozoite surface protein-3α (PvMSP-3α) gene. Dried blood spots from microscopically confirmed P. vivax patients were used for investigation of the PvMSP-3α gene. PCR-RFLP was performed on the PvMSP-3α gene to investigate the genotypes and allelic variability with HhaI and AluI restriction enzymes. In total, 40 successfully PCR amplified PvMSP-3α gene segments were subjected to RFLP analysis. Amplified products showed three different base pair size variations viz. genotype A in 31(77.5%), genotype B in 4(10%) and genotype C in 5(12.5%) P. vivax specimens. RFLP with HhaI and AluI revealed 17 (H1-H17) and 25 (A1-A25) allelic variants, respectively. Interestingly, two similar sub-allelic variants, ie. H8 (with HhaI), and A4 (with AluI) clustered within the rural area of Nuh district, Haryana in two samples. With this study, we propose to commission such type of genetic diversity analysis of P. vivax to investigate the circulating genotypes of the parasites from distinct geographical locations across India, that can have significant implications in understanding the population structures of P. vivax.

Abstract Apple is an important fruit crop that is always in demand due to its commercial and nutraceutical value. Also, the requirement for quality planting material for this fruit crop for new plantations is increasing continuously. In-vitro propagation is an alternative approach, which may help to produce genetically identical high grade planting material. In this study, for the first time, an efficient and reproducible propagation protocol has been established for apple root stock MM 104 via axillary bud. Culturing axillary buds on Murashige and Skoog apple rootstock (MM 104) resulted in better in-vitro propagation. (MS) basal medium supplemented with 3.0% (w/v) sucrose and 0.8% (w/v) agar. The axillary buds were established in MS basal medium with BA (5.0 µM), NAA (1.0 µM) and further used to establish invitro propagation protocol. Plant Growth Regulators (PGRs), BA (1.0 µM) in combination with NAA (1.0 µM) was found most efficient for shoot multiplication (100%) and produced 9.8 shoots/explants with an average shoot length of (2.4 ± cm). All the shoots produced roots in 0.1 µM IBA with a 5-day dark period. Acclimatization of in-vitro raised plantlets was obtained with vermiculite: perlite: sand: soil (2:2:1:1) resulting in 76% survival under field conditions. The study showed that the use of axillary bud is efficient for multiple-shoot production of apple rootstock (MM 104). This is the first comprehensive report on in-vitro growth of apple root stock MM 104 with an assessment of genetic stability using DNA fingerprinting profiles based on Inter Simple Sequence Repeats (ISSR) and Start Codon Targeted (SCoT). The genetic stability of in-vitro-produced plants, as determined by SCoT and ISSR primers, demonstrated genetic closeness to the mother plant.

Abstract Bi-allelic pathogenic variants in PRKN are the most common cause of autosomal recessive Parkinson’s disease (PD). 647 patients with PRKN-PD were included in this international study. The pathogenic variants present were characterised and investigated for their effect on phenotype. Clinical features and progression of PRKN-PD was also assessed. Among 133 variants in index cases (n = 582), there were 58 (43.6%) structural variants, 34 (25.6%) missense, 20 (15%) frameshift, 10 splice site (7.5%%), 9 (6.8%) nonsense and 2 (1.5%) indels. The most frequent variant overall was an exon 3 deletion (n = 145, 12.3%), followed by the p.R275W substitution (n = 117, 10%). Exon3, RING0 protein domain and the ubiquitin-like protein domain were mutational hotspots with 31%, 35.4% and 31.7% of index cases presenting mutations in these regions respectively. Background: Percutaneous nephrolithotomy (PCNL) is the preferred treatment for the removal of large kidney stones, sized >20 mm. However, there is still an ongoing debate concerning the best anesthesia for PCNL. Aims and Objectives: The objective of this study was to assess and compare the efficacy and safety of spinal anesthesia (SA) versus general anesthesia (GA) in PCNL. Materials and Methods: A prospective randomized study included eighty-two patients who were enrolled to receive either SA or GA for PCNL. The study assessed patients’ characteristics, vital parameters, requirements for additional analgesia, and patient satisfaction. In addition, intraoperative and post-operative complications were documented. Patients’ satisfaction was also analyzed. Results: In both groups, the vital parameters were consistently maintained at safe levels during the procedures. Societal Impact Statement Preserving and conserving crop landraces, tended by indigenous farming communities, is crucial for future food security. This research focused on rice landrace diversity in the north‐eastern Himalayan region of Nagaland, India, where Naga communities cultivate rice according to their dietary and cultural preferences. Rice diversity is closely linked to the region's ethnolinguistic and ecological variety. On‐farm conservation, in collaboration with indigenous communities, is imperative to protect these germplasm resources. Involving these communities actively in conservation efforts will safeguard their traditional knowledge, endorse sustainable farming practices, and enhance the resilience of local agricultural systems. Summary Understanding the genetic diversity and cultural significance of crop landraces is crucial for their conservation and sustainable utilization. This study focused on rice landraces from Nagaland in north‐eastern India to assess their genetic diversity and explore their associations with ethnolinguistic groups. We collected 78 rice landraces from Nagaland and a small part of Manipur and conducted microsatellite genotyping for genetic analysis. We integrated social anthropology and population genetics analyses of rice landraces to glean insights into the genetic diversity, population structure, and ethnolinguistic history of rice cultivation in Nagaland. The study revealed the rich cultural significance of rice landraces among the Nagas. Farmers practiced small‐scale subsistence farming, maintaining diverse rice landraces. Naming conventions were based on factors such as seed source, color, grain type, and ecological suitability. Rice landraces played important roles in ethnic cultures, festivals, and religious ceremonies. Genetic analysis identified significant diversity, with 277 alleles across 69 loci and a moderate gene diversity of 0.57. Two distinct sub‐populations were identified, with one dominated by Chakhesang and Angami Nagas and the other by Sümi and Lotha accessions. Differentiation was observed between lowland and upland cultivars, with one sub‐population comprising exclusively lowland varieties. Cultural factors and cultivation practices influenced population differentiation, with ethnicity and ecotype having a significant impact. The study also highlighted the correlation between ethnolinguistic differentiation and the indica–japonica structuring of rice landraces. Different ethnic groups in Nagaland had distinct cultivation practices, contributing to genetic differentiation. Overall, this research emphasizes the need to preserve rice landraces and associated traditional knowledge for future improvements and cultural heritage conservation. It provides insights into genetic diversity, cultural significance, and the relationship between genetic diversity, cultural practices, and agricultural traditions.

Purpose – This paper aims to examine the influence of board characteristics on the integrated reporting quality (IRQ) of Indian-listed companies. Design/methodology/approach – The study uses a sample of 197 firms from the BSE 500 for the years 2017–2018 to 2019–2020. The proposed hypotheses are tested using two-stage least squares regression. Findings – The study documents a positive influence of board size, board independence and gender diversity on IRQ. The study also finds that board activity and role duality are insignificant with IRQ. Among the firm-specific characteristics, variables such as firm size, profitability and capital intensity positively influence IRQ. Originality/value – The current study presents the first investigation in the context of India on the various board characteristics influencing IRQ. The study reiterates the role that gender-diverse boards have in improving information transparency. Policymakers can therefore drive adoption by recommending changes in board characteristics and increasing the quota for women on boards.

The quest for happiness is highly esteemed in every culture. Based on the theory of the positive activity model, the current study examined the role of psychological hardiness in happiness among youth. The study also investigated the parallel mediating role of vitality and materialistic values in the relationship between psychological hardiness and happiness. Data were collected from 414 youths selected through purposive sampling techniques. The variables of the study were measured through standardized instruments. Findings generated through structural equation modelling with the maximum likelihood estimation technique revealed that psychological hardiness was a significant predictor of happiness. Additionally, vitality and materialistic values partially mediated the relationship between psychological hardiness and happiness. The present study is one of the first to include vitality and materialistic values as a parallel intermediary mechanism to explain the relationship between psychological hardiness and happiness. Implications of the study are discussed in light of the study findings.

Fungal diseases cause substantial loss to agricultural crops, affecting both quantities and quality. Although several methods are used for preventing disease incidence, fungicides remain crucial for higher yields and better quality. But in the past, the efficacy of several fungicides has decreased due to increased cases of fungicide resistant. In our pursuit of new effective fungicides, we synthesised a series of twenty 2-Indazol-1-yl-chromen-4-one derivatives (6a- 6t). The characterization of synthesized compounds was performed by several spectroscopic methods including Infrared, Nuclear Magnetic Resonance (1H and 13C) and HRMS. Out of 20 synthesised compounds, 19 (6b- 6t) were found to be novel. All synthesised indazolylchromones showed very good antifungal activity against Sclerotium rolfsii and Fusarium oxysporum. Among the tested compounds, 6t and 6f exhibited very good fungicidal activity against S. rolfsii with an ED50 of 10.10 mg L-1 and 16.18 mg L-1, respectively. In case of Fusarium oxysporum compound 6f displayed good’ activity with an ED50 value of 27.82 mg L-1. Molecular docking study was done to predict the binding sites of most active compounds, 6t and 6f with Cytochrome P450 14alpha -sterol demethylase (CYP51) enzyme using molsoft software. The acute toxicity predictions the of synthesized compounds for fish (LC50,96 Hr), daphnid (LC50, 48 Hr) and green algae (EC50, 96Hr) and the chronic toxicity predictions (ChV) were assessed using Ecological Structure Activity Relationship (ECOSAR) model. As per ECOSAR prediction, all the chemicals are inside AD and not missing predictions.

The agricultural productivity and sustainability in Eastern Gangetic Plain Zones of India are threatened because of the inefficiency of current production practices, shortage of resources, and socioeconomic constraints. We hypothesized the potential impact of intensified cereal systems with mung bean as a third crop within the annual cropping cycle. We assessed economic, social, and environmental indicators for intensified and current cropping system management practiced by different farm types in the region using the FarmDESIGN model. Building on a farm typology constructed for the region in our past research, we used five types of farmers: part-time (PT), well-endowed (WE), small-scale (SS) crop and livestock mix, medium-scale (MS), and resource-poor farmers (RP) in this study. The performance indicators of the 229 original cropping systems cultivated within the 43 farms varied strongly in the eight performance indicators. This variability of cropping systems performance within the farm types resulted in the absence of significant differences between the types. Compared to the original cropping systems, the intensified cropping systems with mung bean not only performed high in dietary energy (DE) production and organic matter (OM) inputs into the soil but also had high application rates of biocides and minimized losses of nitrogen (N). The intervention systems were low in labor requirement and scored at an intermediate level for crop gross margin, water use, and greenhouse gas (GHG) emissions. The ranges of areas of maize- and rice-based systems that could be replaced by intensified systems were largest for the WE and RP farm types. This was reflected in large ranges of change in the performance indicators, but no significant differences in response were found between the farm types. The intensification of maize- and rice-based systems with the proposed intervention cropping systems involving mung bean would result in increased profitability, higher DE yield, and lower requirements for labor and water as the proportion of the farms being converted increases. However, the use of biocides would increase, while the intervention cropping systems would have no significant effect on OM input, GHG emissions, and soil N losses.

Microbiome research has gained much attention in recent years as the importance of gut microbiota in regulating host health becomes increasingly evident. However, the impact of radiation on the microbiota in the murine bone marrow transplantation model is still poorly understood. In this paper, we present key findings from our study on how radiation, followed by bone marrow transplantation with or without T cell depletion, impacts the microbiota in the ileum and caecum. Our findings show that radiation has different effects on the microbiota of the two intestinal regions, with the caecum showing increased interindividual variation, suggesting an impaired ability of the host to regulate microbial symbionts, consistent with the Anna Karenina principle. Additionally, we observed changes in the ileum composition, including an increase in bacterial taxa that are important modulators of host health, such as Akkermansia and Faecalibaculum. In contrast, radiation in the caecum was associated with an increased abundance of several common commensal taxa in the gut, including Lachnospiraceae and Bacteroides. Finally, we found that high doses of radiation had more substantial effects on the caecal microbiota of the T-cell-depleted group than that of the non-T-cell-depleted group. Overall, our results contribute to a better understanding of the complex relationship between radiation and the gut microbiota in the context of bone marrow transplantation and highlight the importance of considering different intestinal regions when studying microbiome responses to environmental stressors.

Background: Circular RNAs (circRNAs) hold potential as diagnostic markers for colorectal cancer (CRC); however, their functional mechanisms remain incompletely elucidated. This work investigates the clinical implications of a unique set comprising six circRNAs derived from serum in CRC. Furthermore, we delve into the role of exosomal circ_0084043, originating from colorectal cancer-associated fibroblasts (CAFs), with a specific focus on its contribution to endothelial cell angiogenesis. Methods: The study analyzed circRNA levels in serum samples obtained from both CRC and control groups using qRT-PCR. Additionally, exosomes originating from colorectal CAFs and normal fibroblasts (NFs) were purified and confirmed by electron microscopy and Western blotting techniques. The proangiogenic effects of CAF-derived exosomal circ_0084043 were assessed in endothelial cells through proliferation, migration, and in vitro capillary tube formation assays. Gain- and loss-of-function experiments were employed to clarify the role of the circ_0084043/miR-140–3p/HIF-1α axis in endothelial cell angiogenesis, utilizing luciferase reporter assay, Western blotting, and ELISA for mechanism elucidation. Results: The candidate circRNAs (circ_0060745, circ_001569, circ_007142, circ_0084043, Circ_BANP, and CiRS-7) exhibited notably elevated expression in CRC patient sera compared to the levels observed in healthy individuals. Except for CiRS-7, all circRNAs showed elevated expression in CRC patients with positive lymph node metastasis and advanced tumor stages. Exosomes released by colorectal CAFs augmented endothelial cell proliferation, migration, and angiogenesis by upregulating VEGF expression and secretion. Circ_0084043 was highly detected in endothelial cells treated with CAF-derived exosomes. Silencing circ_0084043 reduced VEGFA expression and diminished CAF exosome-induced endothelial cell processes, indicating its pivotal role in angiogenesis. Circ_0084043 sponges miR-140–3p, regulating HIF-1α, and a reverse relationship was also identified between miR-140–3p and VEGFA in endothelial cells. Inhibiting miR-140–3p mitigated circ_0084043 knockdown effects in CAF exosome-treated endothelial cells. Co-transfection of si-circ_0084043 and a miR-140–3p inhibitor reversed the inhibited migration and angiogenesis caused by circ_0084043 knockdown in CAF exosome-treated endothelial cells. Inhibiting miR-140–3p rescued reduced VEGFA expression due to circ_0084043 knockdown in endothelial cells exposed to CAF-derived exosomes, indicating modulation of the circ_0084043/miR-140–3p/VEGF signaling in CAF-derived exosome-induced angiogenesis. Conclusions: This study unveiled a distinctive signature of six serum-derived circular RNAs, indicating their potential as promising diagnostic biomarkers for CRC. Importantly, exosomal circ_0084043 originating from colorectal CAFs was identified as playing a crucial role in endothelial cell angiogenesis, exerting its influence through the modulation of the miR-140–3p/HIF-1α/VEGF signaling axis.

To address environmental pollution caused by dyes and heavy metals, it is crucial to fabricate effective adsorbent material to remove contaminants from the environment. In this research, a biogenic-assisted treatment was applied to kaolin clay to eliminate arsenic ions (As(V)) and malachite green (MG) using an adsorption technique. An environmentally friendly ultrasonic method was used to incorporate Pterocarpus mildraedii into the mesoporous structure of kaolin clay (PMKC) material with a unique flake-like morphology characterized by hexagonal structures formed by quartz and kaolin. Various analytical techniques were employed to analyze the functional groups, crystallographic structures, and surface properties of PMKC. The findings reveal that PMKC, an eco-friendly adsorbent, effectively removed As(V) and MG from wastewater under specific conditions: a solution pH of 6, a PMKC dosage of 40 mg/L, a contact time of 60 min, an initial concentration of As(V)/MG of 100 mg/L, and an adsorption temperature of 40 °C. Furthermore, PMKC exhibited high adsorption capacities of 337.22 mg/g towards As(V) and 274.73 mg/g towards MG. Dubinin-Radushkevich and the pseudo-first-order models were found to best describe the experimental data obtained from the isotherm and kinetic studies. Because PMKC can be reused effectively and remains stable for up to ten cycles, it holds promise as a valuable material for removing dye and heavy metal pollutants.

Background: Peninsular India, being completely under the influence of monsoonal climate, suffers crop yield variability due to rainfall distribution-induced soil moisture constraints. Timely and appropriate assessment of this rainfall and soil moisture-induced crop yield variability serves as a key for exemplary relief assistance. Per cent available soil moisture (PASM) is one among several drought declaration indices followed by stakeholders in India for declaration of drought, needs re-evaluation as the existing criteria in unable to capture the yield loss due to ineffective classification of PASM categories. This study attempts to revise the agricultural drought classes by PASM based on relationships established between yield of major rainfed crops of the study region and PASM.Methods: Analysis of yield variability due to PASM was carried out based on long term observations in experiments conducted at five dry farming locations (Akola, Parbhani, Kovilpatti, Ananthapuramu and Bengaluru) of peninsular India. The average yield for each category of PASM was calculated and tabulated for regression analysis. The PASM versus yield in each group was correlated and regression equations were developed if significant positive correlations were established.Results: The range of available soil moisture to obtain at least 50 percent of optimum yield in cereals (maize: 26 and finger millet: 52.9 PASM), pulses (pigeon pea: 37.2 PASM), oilseeds (soybean: 26.8 to 30.5, groundnut: 53.8 to 61.7 PASM) and commercial crops (cotton: 26.3 PASM) was 26–61 percent.Conclusion: The revised PASM-based drought classes (0–50 severe; 51–75 mild and 76–100 no drought) would help in drought declaration and precise identification of drought-hit areas for meaningful relief assistance. However, there is further investigation is needed to include a soil component for further fine-tuning of the criteria.

Background: Palatal rugae are asymmetrical elevations present on hard palate and are significant due to their uniqueness, stability and thus, aid in forensic identification. Objective: The objective is to determine palatal rugae morphology between parents and their offsprings and to determine their genetic inheritance. Materials and Methods: This randomized controlled cross-sectional single-blinded study was conducted on 50 families (randomly selected) who visited Vyas Dental College and Hospital. A total of 150 maxillary study models were examined and rugae were delineated and shape, size, and pattern of rugae were recorded. Results: Weak but significant correlation was seen in the nonspecific and angular type of palatal rugae between child and mother and child and father as P < 0.05. Matching with either or both of their parent was seen for curved, wavy, divergent, straight, horseshoe, angular, and nonspecific type of rugae with the higher % of wavy pattern matching (52% with mother and 54% with father). A considerable number of samples showed high heritability of primary and secondary rugae from both parents, but more predominantly to their mothers (branched, nonspecific, and straight). Conclusion: Both environmental and genetic factors can influence the rugae pattern. Further, studies should be conducted on a larger sample size to get more significant results.

Abstract The present study investigated two single nucleotide polymorphisms (SNPs)—rs479200 and rs516651 in the host EGLN1/PHD2 gene for their association with COVID-19 severity. A retrospective cohort of 158 COVID-19 patients from the Indian population (March 2020 to June 2021) was enrolled. Notably, the frequency of C allele (0.664) was twofold higher than T allele (0.336) in severe COVID-19 patients. Here, we report a novel finding that the C allele of rs479200 in the EGLN1 gene imparts a high risk of severe COVID-19 (odds ratio—6.214 (1.84–20.99) p = 0.003; 9.421 (2.019–43.957) p = 0.004), in additive inheritance model (adjusted and unadjusted, respectively).

This article presents a systematic literature review on hybrid recommendation systems (HRS) in the e-commerce sector, a field characterized by constant innovation and rapid growth. As the complexity and volume of digital data increases, recommendation systems have become essential in guiding customers to services or products that align with their interests. However, the effectiveness of single-architecture recommendation algorithms is often limited by issues such as data sparsity, challenges in understanding user needs, and the cold start problem. Hybridization, which combines multiple algorithms in different methods, has emerged as a dominant solution to these limitations. This approach is utilized in various domains, including e-commerce, where it significantly improves user experience and sales. To capture the recent trends and advancements in HRS within e-commerce over the past six years, we review the state-of-the-art overview of HRS within e-commerce. This review meticulously evaluates existing research, addressing primary inquiries and presenting findings that contribute to evidence-based decision-making, understanding research gaps, and maintaining transparency. The review begins by establishing fundamental concepts, followed by detailed methodologies, findings from addressing the research questions, and exploration of critical aspects of HRS. In summarizing and incorporating existing research, this paper offers valuable insights for researchers and outlines potential avenues for future research, ultimately providing a comprehensive overview of the current state and prospects of HRS in e-commerce.

Background: Osteoarthritis (OA) is one of the common causes of knee disability. Depending on the stages of the disease, various treatment modalities are available. OA, being a degenerative and gradually progressive disease compliance with the treatment, has a major role in the outcome. Materials and Methods: The study was carried out at a tertiary care orthopedic center to assess the level of compliance of tablet glucosamine and chondroitin sulfate in patients with early OA of the knee. 100 consecutive patients visiting the outpatient department of orthopedics were included after clinical and radiological confirmation of diagnosis. Patients were given tablet glucosamine and chondroitin sulfate and followed up for 12 weeks. Compliance was assessed by the Morisky Medication Adherence Scale (MMAS). Results: One hundred patients consisting of 44 males and 56 females were enrolled. The average age of the study population was 51.06 ± 9.44 years. Response recorded by the MMAS showed good compliance in 46%, moderate in 36%, and poor in 18% of cases. A significant positive correlation was found with education, family support, and awareness of the disease, whereas a negative correlation was found with age, functional impairment, comorbidities, and polypharmacy. Conclusion: The study confirmed the existence of good compliance to glucosamine and chondroitin sulfate in patients with early OA of the knee. The variation in the reported level of compliance in the study population was due to factors such as literacy level, family status, functional status, associated medical illnesses, daily medicinal load, as well as awareness of the disease. Only a few studies have been conducted in developing countries on medication compliance, factors influencing it, and magnitude of the impact of noncompliance; hence, there is a need for more systematic studies.

Abstract There is extensive synaptic loss from frontotemporal lobar degeneration, in preclinical models and human in vivo and post mortem studies. Understanding the consequences of synaptic loss for network function is important to support translational models and guide future therapeutic strategies. To examine this relationship, we recruited 55 participants with syndromes associated with frontotemporal lobar degeneration and 24 healthy controls. We measured synaptic density with positron emission tomography using the radioligand [11C]UCB-J, which binds to the presynaptic vesicle glycoprotein SV2A, neurite dispersion with diffusion magnetic resonance imaging, and network function with task-free magnetic resonance imaging functional connectivity. Synaptic density and neurite dispersion in patients was associated with reduced connectivity beyond atrophy. Functional connectivity moderated the relationship between synaptic density and clinical severity. Our findings confirm the importance of synaptic loss in frontotemporal lobar degeneration syndromes, and the resulting effect on behaviour as a function of abnormal connectivity.

Non-linear neutrosophic numbers (NLNNs) are different kinds of neutrosophic numbers with at least one non-linear membership function (either of truthiness, falsity or indeterminacy part) of the information. Furthermore, a linear programming problem with non-linear neutrosophic numbers as coefficients/parameters is a special type of programming problem known as a non-linear linear programming problem (NLN-LPP). This paper elaborates on the concepts of non-linear neutrosophic number (NLNN) sets, different forms of non-linear neutrosophic numbers (NLNNs), cuts on non-linear neutrosophic numbers (NLNNs), possibility mean, possibility standard deviation, and possibility variance of non-linear neutrosophic numbers (NLNNs). In this paper, we also propose the solution technique for non-linear neutrosophic linear programming problems (NLN-LPPs) in which all coefficients/parameters are non-linear neutrosophic numbers (NLNNs). In this continuation, we suggest a new modified possibility score function for non-linear NNs in terms of possibility means and possibility standard deviations of non-linear neutrosophic numbers (NLNNs) for better use of all parts of information. This modified score function is used to convert non-linear neutrosophic number (NLNN) coefficients/parameters of non-linear neutrosophic linear programming problem (NLN-LPP) into equivalent crisp values. Thereafter, the equivalent crisp problem is solved with the usual method to obtain the optimal solution of non-linear neutrosophic linear programming problem (NLN-LPP). The proposed solution algorithm is unique and new for solving non-linear neutrosophic linear programming problems. A numerical example is solved with the proposed algorithm to legitimate the research output. A case study is also discussed to show its applicability in solving real-life problems.

Background: In Radiotherapy, computation of dose is important since in a small field with heterogeneity, dose is usually computed with discrepancies.Objective: The present study was aimed to evaluate the dosimetry of treatment planning algorithms in lung equivalent heterogeneous medium for Volumetric Modulated Arc Therapy (VMAT) with step and shoot Intensity-Modulated Radiation Therapy (ss-IMRT), and dynamic Intensity-Modulated Radiation Therapy (d-IMRT).Material and Methods: In this experimental study, Computerized Imaging Reference System (CIRS) phantom was used with an inhomogeneous Racemosa wood cylinder for two types of tumors, namely, Left Lung Central Tumor (LCT) and Left Lung Peripheral Tumor (LPT) in the CIRS left lung cavity. The computed tomography (CT) datasets were employed with the generation of VMAT, d-IMRT and ss-IMRT plans for the LCT and LPT irradiated with 6 MV photon beams. In this study, the accuracy and efficacy of two algorithms: Monte Carlo (MC) and the Pencil Beam (PB), from the Monaco treatment planning system (TPS), were tested by using Gafchromic EBT3 films and CIRS thorax phantom.Results: Regardless of treatment techniques, both algorithms exhibited higher divergence in LPT than LCT. In both LCT and LPT, the highest deviation was near the tumor-lung junction. However, the deviation was higher in the PB algorithm than MC algorithm, with a minimally acceptable variation of -0.8%. Conclusion: The MC algorithm shows more consistency for EBT3 measured dose in lung equivalent heterogeneous medium. However, accurate dose predictions are complicated due to electronic disequilibrium within and at the interface of inhomogeneity. These constraints may cause variations from the anticipated outcomes of the treatments.

Abstract Large cardamom (Amomum subulatum Roxb.) is an economically important cash crop that provides a livelihood option for the rural communities in Sikkim Himalaya. However, its production has declined drastically over the past few decades due to climate change and other factors affecting the livelihood of marginal cardamom-dependent farmers in the region. Climate change causes a shift in elevational distributions of mountain species, and it is pivotal to understand its effect on yield and yield-related traits for economically important plant species like large cardamom. For this, we randomly studied 41 large cardamom cultivation sites in Sikkim (India) with elevations ranging between 975 and 2069 m asl and evaluated the yield-related traits (number of capsules per spike, capsule length, capsule width, fresh capsule weight, dry capsule weight, number of seeds per locule, fresh seed weight, and dry seed weight) in five cultivars (Dzongu Golsey, Sawney, Seremna, Ramsey, and Varlangey). We observed a significant variability (P 1515 m), while cultivation of high-elevation Ramsey cultivar (suitable in elevation > 1515 m) has shifted downward ( 1515 m) and showed moderate to high negative correlation between elevation and yield-related traits, indicating the negative effect of elevation on their yield. Likewise, Ramsey and Varlangey cultivated in high elevation (> 1515 m) performed better than the one cultivated in mid-elevation (975–1515 m) and depicted moderate to high positive correlation between elevation and yield-related traits, suggesting a positive influence of elevation on their yield. Although there is an elevational shift in the cultivation of large cardamom cultivars, the elevation influences the performance of the large cardamom cultivars, and it also suggests cultivating the cultivars in their suitable elevation range for better productivity.

Background: Recently, a wide range variety of manifestations, including a self‑limiting to severe illness, has been increasingly reported in dengue. Few studies attract attention to severe dengue, mainly observed in secondary infection. With this background, this study aims to provide a comprehensive overview to differentiate primary from secondary dengue using serology (IgG) and the possible association of severity of illness in secondary dengue. Methods: Present retrospective cross-sectional study was conducted at a North Indian tertiary care center from September 2021 to January 2022. Clinical data of confirmed dengue patients from the medicine department were collected and assigned as primary and secondary dengue. Results: Of the 220 dengue patients, 22 (10 %) had secondary dengue infection. Hemorrhagic manifestations were reported in 58/220 (26.4 %) cases while 7/22 (31.8 %) in secondary dengue. Prevalent hemorrhagic manifestations in secondary dengue include purpura (27.3 %), vaginal bleeding (4.5 %), melaena (9.1 %), and epistaxis (4.5 %). In addition, 42 (19.1 %) patients had pancytopenia, and 8 (36.6 %) cases were of secondary dengue. Hepatic dysfunction was noted in 164 (74.5 %) cases. Notably, all secondary dengue cases (22;100 %) had hepatic dysfunction and severe in 9 (40.9 %) cases. In addition, in secondary dengue patients, evidence of plasma leakages such as hypoproteinemia 7 (31.8 %) and ascites (35 %) were statistically more frequent. Overall, two deaths (0.9 %) were reported, and were one in each group. Conclusions: Many parameters, including hemorrhagic manifestation (melaena), hematological characteristic (pancytopenia), evidence of plasma leakage (hypoproteinemia and ascites), gastrointestinal (GB wall thickening and hepatic dysfunction) and reduction in mean hemoglobin and platelet count were found to be statistically significant in secondary dengue infection. Additionally, early classification of secondary dengue may help to anticipate its severity and allow for early strategic intervention/management to lower morbidity and mortality.

ABSTRACT: Objectives: Artemisinin (ART) resistance in Plasmodium is threatening the artemisinin combination therapies—the first line of defence against malaria. ART resistance has been established to be mediated by the Plasmodium Kelch13 (PfK13) protein. For the crucial role of PfK13 in multiple pathways of the Plasmodium life cycle and ART resistance, it is imperative that we investigate its interacting partners. Methods: We recombinantly expressed PfK13-p (Bric a brac/Poxvirus and zinc finger and propeller domains), generating anti-PfK13-p antibodies to perform co-immunoprecipitation assays and probed PfK13 interacting partners. Surface plasmon resonance and pull-down assays were performed to establish physical interactions of representative proteins with PfK13-p. Results: The co-immunoprecipitation assays identified 17 proteins with distinct functions in the parasite life cycle– protein folding, cellular metabolism, and protein binding and invasion. In addition to the overlap with previously identified proteins, our study identified 10 unique proteins. Fructose-biphosphate aldolase and heat shock protein 70 demonstrated strong biophysical interaction with PfK13-p, with KD values of 6.6 µM and 7.6 µM, respectively. Additionally, Plasmodium merozoite surface protein 1 formed a complex with PfK13-p, which is evident from the pull-down assay. Conclusion: This study adds to our knowledge of the PfK13 protein in mediating ART resistance by identifying new PfK13 interacting partners. Three representative proteins—fructose-biphosphate aldolase, heat shock protein 70, and merozoite surface protein 1—demonstrated clear evidence of biophysical interactions with PfK13-p. However, elucidation of the functional relevance of these physical interactions are crucial in context of PfK13 role in ART resistance.

Since the early 1970s, the study of Black–Scholes (BS) partial differential equations (PDEs) under the Efficient Market Hypothesis (EMH) has been a subject of active research in financial engineering. It has now become obvious, even to casual observers, that the classical BS models derived under the EMH framework fail to account for a number of realistic price evolutions in real-time market data. An alternative approach to the EMH framework is the Fractal Market Hypothesis (FMH), which proposes better and clearer explanations of market behaviours during unfavourable market conditions. The FMH involves non-local derivatives and integral operators, as well as fractional stochastic processes, which provide better tools for explaining the dynamics of evolving market anomalies, something that classical BS models may fail to explain. In this work, using the FMH, we derive a time-fractional Black–Scholes partial differential equation (tfBS-PDE) and then transform it into a heat equation, which allows for ease of implementing a high-order numerical scheme for solving it. Furthermore, the stability and convergence properties of the numerical scheme are discussed, and overall techniques are applied to pricing European put option problems.

Encysted hydrocele of the spermatic cord occurs due to the persistence of the Cranford section, which is a segment of the patent processus vaginalis. This results in localised fluid collection, causing an encysted hydrocele. The present case reports a rare cause of groin swelling that mimics an incarcerated inguinal hernia. A 55-year-old male presented with complaints of swelling over his right groin for the past 6 months and dull-aching pain for the past 1 week. Clinically diagnosed as a case of a right incarcerated inguinal hernia. High-frequency ultrasound revealed an anechoic cystic lesion of size 8×3.8 cm in the subcutaneous plane of the right inguinal region with no bowel or omentum as content. Intraoperatively, a 9×7 cm ovoid encysted hydrocele of the cord was identified. No intraoperative findings were suggestive of an inguinal hernia. The hydrocele sac was carefully excised from cord contents. Prompt anticipation and surgical management are required for the diagnosis and management of an encysted hydrocele.

Recent genome-wide association studies (GWAS) have identified a link between single-nucleotide polymorphisms (SNPs) near the MBOAT7 gene and advanced liver diseases. Specifically, the common MBOAT7 variant (rs641738) associated with reduced MBOAT7 expression is implicated in non-alcoholic fatty liver disease (NAFLD), alcohol-associated liver disease (ALD), and liver fibrosis. However, the precise mechanism underlying MBOAT7-driven liver disease progression remains elusive. Previously, we identified MBOAT7-driven acylation of lysophosphatidylinositol lipids as key mechanism suppressing the progression of NAFLD (Gwag et al., 2019). Here, we show that MBOAT7 loss of function promotes ALD via reorganization of lysosomal lipid homeostasis. Circulating levels of MBOAT7 metabolic products are significantly reduced in heavy drinkers compared to healthy controls. Hepatocyte- (Mboat7-HSKO), but not myeloid-specific (Mboat7-MSKO), deletion of Mboat7 exacerbates ethanol-induced liver injury. Lipidomic profiling reveals a reorganization of the hepatic lipidome in Mboat7-HSKO mice, characterized by increased endosomal/lysosomal lipids. Ethanol-exposed Mboat7-HSKO mice exhibit dysregulated autophagic flux and lysosomal biogenesis, associated with impaired transcription factor EB-mediated lysosomal biogenesis and autophagosome accumulation. This study provides mechanistic insights into how MBOAT7 influences ALD progression through dysregulation of lysosomal biogenesis and autophagic flux, highlighting hepatocyte-specific MBOAT7 loss as a key driver of ethanol-induced liver injury.

Background Pyroptosis executor GsdmD (gasdermin D) promotes atherosclerosis in mice and humans. Disulfiram was recently shown to potently inhibit GsdmD, but the in vivo efficacy and mechanism of disulfiram's antiatherosclerotic activity is yet to be explored. Methods and Results We used human/mouse macrophages, endothelial cells, and smooth muscle cells and a hyperlipidemic mouse model of atherosclerosis to determine disulfiram antiatherosclerotic efficacy and mechanism. The effects of disulfiram on several atheroprotective pathways such as autophagy, efferocytosis, phagocytosis, and gut microbiota were determined. Atomic force microscopy was used to determine the effects of disulfiram on the biophysical properties of the plasma membrane of macrophages. Disulfiram‐fed hyperlipidemic apolipoprotein E−/− mice showed significantly reduced interleukin‐1β release upon in vivo Nlrp3 (NLR family pyrin domain containing 3) inflammasome activation. Disulfiram‐fed mice showed smaller atherosclerotic lesions (~27% and 29% reduction in males and females, respectively) and necrotic core areas (~50% and 46% reduction in males and females, respectively). Disulfiram induced autophagy in macrophages, smooth muscle cells, endothelial cells, hepatocytes/liver, and atherosclerotic plaques. Disulfiram modulated other atheroprotective pathways (eg, efferocytosis, phagocytosis) and gut microbiota. Disulfiram‐treated macrophages showed enhanced phagocytosis/efferocytosis, with the mechanism being a marked increase in cell‐surface expression of efferocytic receptor MerTK. Atomic force microscopy analysis revealed altered biophysical properties of disulfiram‐treated macrophages, showing increased order‐state of plasma membrane and increased adhesion strength. Furthermore, 16sRNA sequencing of disulfiram‐fed hyperlipidemic mice showed highly significant enrichment in atheroprotective gut microbiota Akkermansia and a reduction in atherogenic Romboutsia species. Conclusions Taken together, our data show that disulfiram can simultaneously modulate several atheroprotective pathways in a GsdmD‐dependent as well as GsdmD‐independent manner.

Rapid, inexpensive, and low-power/solar light-driven photocatalytic degradation of organic pollutants to deal with annually produced trillion tons of synthetic dye wastewater to prevent water scarcity issues, ecotoxicological risks, and human health has always been challenging. To overcome this limitation, the present study synthesized earth-abundant, inexpensive copper oxide nanosheets using a simple single-step hydrothermal route. The structural, physicochemical, and functional properties of the nanosheets have been characterized using several characterization techniques. The photocatalytic activity was studied for two commonly industrially used organic dyes, Methylene Blue (MB) and Rhodamine B (RhB). The importance of this work is the usage of a cheap commercially available Phillips UV light (11 W) as well as direct sunlight. With several optimized conditions, almost complete degradation of both dyes was achieved within 35 minutes under low-power UV light and within 70 minutes by the direct illumination of natural sunlight. The enhanced photocatalytic performance can be correlated to the synergetic effect of a higher charge transfer mechanism, good catalytic ‘active surface area’ availability (13.2 m2/g), and several optimized parameters that affect the reaction efficacy. Additionally, five repeated uses of nanosheets without sacrificing performance confirmed their stability and sustainability as a promising candidate for large-scale industrial textile wastewater remedies.

Introduction: Congenital heart disease in children are a major cause of infant mortality and morbidity. It is important to detect and manage these disorders timely as they are preventable. The objective of this study was to find out proportion of congenital heart disease in children in paediatric department in a tertiary hospital. Methods: This is a descriptive cross-sectional study carried out in the Department of Paediatric at Nepal Medical College and Teaching Hospital where all children (0-18 years) suspected to have congenital heart disease who underwent echocardiography were studied over a period of 1 year (2020-2021). The presence or absence of congenital heart disease were confirmed by echocardiography performed by paediatric cardiologist. The socioeconomic variables,clinical features and echocardiography findings were noted. Results: Out of total 249 patients,the proportion of patients diagnosed to have cardiac disorders was 73 with male predominance of 165 (66.26%). The most common age group was found to be neonates 111 (44%).The notable clinical features were murmur 47 (18.87%), tachypnoea 27 (10.84%) ,tachycardia 27 (10.84%) and cyanosis 9 (3.61%), clubbing 2 (0.80%), oedema 1 (0.40%), hypertension 9 (3.65%), murmur 47 (18.87%).Out of the total, there were 49 (19.67%) cases of acyanotic congenital heart disease, and 27 (10.84%) cases of cyanotic congenital heart disease. Conclusions: Our study focuses on early recognition of cardiac diseases which is crucial for preventing morbidity and mortality.

Mushrooms are traditionally used for various medicinal purposes in traditional oriental medicine. The Japanese and Chinese are familiar with the medicinal macro fungus Lentinula edodes (Shiitake mushroom). This study aims to evaluate the role of chemical compounds from L. edodes using network pharmacology and in-vitro studies for management of Obesity. Bioactive compounds in extracts of L. edodes were identified by GC-MS analysis. Compounds were later screened for their drug-like property by Lipinski's rule. In addition, public databases (SEA, STP, Omim and DisGenet) were searched to identify genes associated with selected molecules and obesity, as well as genes that overlap obesity target genes with genes related to L. edodes. Additionally, analysis was performed using Enrichr KG to predict the disease targets of L. edodes. Finally, network was constructed between the overlapping genes and bioactive molecules using Rstudio. Further in-vitro studies were carried out using 3T3-L1 cell line. The genes related to the selected compounds and obesity were identified and overlapped. The disease targets of L. edodes was predicted by enrichment analysis and was found to be linked to obesity. Furthermore, the hub gene was found to be fatty acid amide hydrolase, and the key bioactive compound was hexadecanoic acid methyl ester. The in-vitro cell culture studies confirmed the inhibition of adipogenesis in mushroom extract-treated 3T3-L1 cells and the augmentation of adiponectin. The study suggests that the hub gene fatty acid amide hydrolase might alleviate obesity by inhibiting arachidonoyl ethanolamide signaling, which would enhance the action of fatty acid amide hydrolase and limit appetite in L. edodes extract.

IntroductionThe diagnosis of infectious reservoirs in malaria (gametocytes) is necessary, especially in low-density infections and asymptomatic malaria patients. The gametocyte stage is a surrogate marker for infection of P. falciparum malaria in healthy individuals. The early detection of infectious gametocytes and treatment will strengthen our efforts in curbing transmission. The nested PCR and real-time quantitative PCR (RT-qPCR) methods have been demonstrated for the diagnosis of infectious gametocyte reservoirs. In this study, RDT, blood smear microscopy, and nested-PCR were used for the detection of P. falciparum and P. vivax, and compared with RT-qPCR detection of Pfg27 gametocyte biomarker gene.MethodsIn the present cross-sectional study, 356 human blood samples were collected from endemic areas of Kokrajhar Assam (asymptomatic and symptomatic malaria patients) for malaria diagnosis.ResultsA total of 8.42%(30/356) incidence of malaria was observed. Introduction: Suicide is a global public health problem. Sensible media reporting on suicide could be useful in suicide prevention. This study aimed to evaluate the knowledge, attitude, and practice regarding media reporting on suicide among media persons of a province. Methods: A qualitative study was done among media persons of the province after obtaining ethical approval from the Institutional Review Committee. Data was collected through online platform using Google form from 15 April 2022 and 15 January 2023. A convenience sampling method was used. Descriptive statistics were used for the data analysis. Results: Among 165 media persons, 54 (32%) of the participants either agreed to or were neutral about prominently reporting suicide news, and 47 (28.48%) were undecided or agreed about mentioning the details of the event. Only 50 (30.30%) thought that it is always possible to help a person with suicidal thoughts. A total of 48 (29.09%) always reported providing information about where to seek help in case one is suicidal. Conclusions: Media personnel were found to possess better knowledge about suicide reporting but exhibited inadequate practice in terms of providing method and site details and promoting support services, alongside maintaining an unfavorable attitude towards suicide. There is an urgent need to focus on coordination, standardisation, evidence generation and capacity building of media persons on suicide.

Natural arch is one of the most fascinating geomorphological features formed by erosional process and many of them are formed within gently dipping sandstone. A natural arch (Location: 21°54′32.20″ N, 83°43′46.20″ E, RL: ∼370 m) of such type is present in the rocks of Upper Kamthi Formation of Middle Jurassic age (160–184 Ma), Upper Gondwana Group, Ib River Coalfield within dense Chhengapahar Reserve Forest near Brahmani Village, Sundargarh District, Odisha. This oval shaped natural arch with a maximum length and height of ∼30 m at the base and ∼12 m respectively. The alcove of the natural arch has maximum height and width of ∼7 m and ∼15 m respectively, is opened along NE-SW direction on the top of the hill. Flesh red colored, ripple topped, trough to planar cross stratified, coarse to gritty, texturally and mineralogically immature, ferruginous sandstone of Upper Kamthi Formation forms the hill. Kaolinisation of feldspathic members makes the sandstone loose and porous. Two orthogonal set of faults-joints (130°/60°W and 40°/60°E) initiated the formation of natural arch while the porosity of the immature, arkosic sandstone accelerate the process through subaerial weathering where surface/rain water percolation–temperature fluctuation played effective roles. Therefore, the formation of natural arch is a result of structurally triggered weathering and erosion. Efforts have already been taken to preserve this place as national geo-heritage site by Geological Survey of India, Government of India.

Aim: This study was intended to assess and relate the clinical anesthetic effectiveness of tramadol hydrochloride and lidocaine. Methodology: A clinical randomized split-mouth study was piloted among 40 patients who were otherwise healthy but needed to have their maxillary first premolars extracted bilaterally for orthodontic reasons were considered in the research. In each subject on one side (quadrant) of the arch 1.8 milliliters of lidocaine was given and on other side of the arch 1.8 milliliters of tramadol was given. On the basis of a list that was prepared by a computer, the randomization was done and the sides (quadrant) of the arches were decided for all the subjects, and the injections were given as local infiltration. Results: Lidocaine was found to be statistically more effective for overall anesthesia duration, despite the fact that there was no significant variance between the two drugs at the time when anesthetic effect began to take effect. Tramadol was found to be statistically more efficient than lidocaine when compared to the anesthetic activity at the 5th minute before extraction. Tramadol was found to be statistically more successful than lidocaine for both the patient's degree of satisfaction and the rate at which wounds healed. Conclusion: Based on the findings of this research, it appears that tramadol and epinephrine could be used as a substitute to conventional local anesthetics during extractions of maxillary first premolar teeth during oral-maxillofacial surgery.

Objective: Using cone beam computed tomography (CBCT), this study compared three rotary file systems—XP Endoshaper, Hyflex CM, and K3 XF—for centering, canal transportation, and root canal area growth. Materials and Methods: The rotating file system randomly separated 60 removed human mandibular molars into three groups (n = 20). Manufacturer-recommended root canal instrumentation was done. Specialized software examined pre- and post-instrumentation CBCT scans. Centering ability was measured by canal transportation and root canal area expansion at different levels from the apex. Results: The Hyflex CM file system had the best centering and the least canal transit at all levels (P < 0.05). XP Endoshaper increased the root canal area the most (P < 0.05). The K3 XF file system had average centering and canal transfer results. Conclusion: The Hyflex CM system centered well, whereas the XP Endoshaper system increased the root canal area the most. The clinical context and therapeutic goals should determine the rotating file system.

Background: In this research study, we delve into the effects of smoking cessation programs on the progression of periodontal disease, particularly focusing on the potential benefits experienced by individuals who successfully quit smoking. Materials And Methods: The research involved the participation of 200 individuals, all of whom were active smokers and exhibited varying degrees of periodontal disease. These participants enrolled in a comprehensive smoking cessation program that included regular counseling sessions and, when necessary, the provision of nicotine replacement therapy to facilitate smoking cessation. Over a 12-month period, the progression of their periodontal disease was systematically monitored through a series of dental check-ups and measurements of critical clinical parameters. These parameters included probing depth (PD), which assesses the spaces between teeth and gums, and clinical attachment level (CAL), which evaluates the attachment of gum tissue to tooth surfaces. Results: After 12 months of active participation in the smoking cessation program, several noteworthy results were observed among the participants. On average, participants experienced a reduction in PD by approximately 1.5 mm, indicating a shallower space between the teeth and gums and, thus, healthier periodontal tissues. In addition, the CAL improved by an average of 1.2 mm, signifying enhanced attachment of gum tissue to the tooth surface, which is essential for dental stability. Participants also exhibited a significant reduction in plaque accumulation on tooth surfaces, indicative of improved oral hygiene practices. Furthermore, gingival inflammation, a common symptom of periodontal disease, notably decreased among participants, suggesting an overall improvement in gum health. Conclusion: In conclusion, this study provides compelling evidence supporting the positive impact of smoking cessation programs on the progression of periodontal disease.

Abstract The trans-Himalayan region of India, although have xeric features, still supports a unique assemblage of biodiversity, including some of the charismatic and endemic species. In the present study, we studied blue sheep (Pseudois nayaur) across the distribution range in the Western trans Himalayas of India and found about 18,775 km2 area suitable for blue sheep. The explicit Bayesian based spatial and non-spatial population structure analysis assigned blue sheep into two genetic populations, i.e., Ladakh and Lahaul-Spiti. We found relatively high genetic divergence in blue sheep which is also supported by the low current flow in Circuitscape model. With the multiple evidences, we explain landscape resistance facilitated by the landscape heterogeneity, and large patches of unsuitable habitats forced population divergence and poor functional connectivity. We found that blue sheep population has been demographically stable in the past, but showed a slight decline within the last few decades. This study is the first range-wide attempt to exhibit landscape features in shaping the spatial distribution, genetic structure and demography patterns of blue sheep in Western Himalayas, and will be of use in the conservation and management planning of blue sheep.
