Double-sided silicon microstrip detectors (DSSDs) with integrated coupling capacitors formed by an oxide-nitride-oxide (ONO) dielectric film were fabricated using newly developed processing techniques. We report on the processing techniques and some characteristics of the detectors fabricated in the above process.

We built full-size models of a detector unit for a silicon micro-vertex detector for the KEK B factory. The model consists of four dummy double-sided, double-metal silicon micro-strip detectors and two silicon end boards with dummy readout VLSIs mounted on both sides. In this trial the Flip-Chip Bonding (FCB) method, using an anisotropic conductive film, is applied to both sides of the detector unit to bond 640 strips at a pitch of 50 ||micro~meter~. The structure using the FCB method successfully provides a new architecture for the detector unit of a vertex detector.

Despite advancements in therapeutic monoclonal antibodies (mAbs) and cell line engineering, separating host cell proteins (HCPs) from mAbs during downstream purification remains challenging. Therefore, in this study, we developed a novel multimodal chromatography (MMC) resin to enhance HCP removal during mAb polishing processes. We evaluated the impact of both ligand structure and pore size of the MMC resin by purifying a post-protein A chromatography solution in flow-through mode. We observed that the efficiency of HCP clearance depended on the hydrophobic moiety structure of the ligand and predicted the mAb purification capability of MMC through linear salt-gradient elution experiments involving a mixture of transferrin, bovine serum albumin (BSA), and pepsin. Our findings revealed that the prototype immobilized 1,12-dodecanediamine via the formyl group exhibited the best performance attributed to its long alkyl chain. Furthermore, an investigation of effects of base bead pore size on HCP capacity using cellulose base beads of five different pore sizes showed that larger pore resin base beads had the highest HCP removal capacity. Specifically, MMC resins with a pore diameter exceeding 440 nm reduced the HCP level by three orders of magnitude under high mAb loading conditions (> 1000 mg/mL-resin). Secretory pathway proteins are cotranslationally translocated into the endoplasmic reticulum (ER) of metazoan cells through the protein channel, translocon. Given that there are far fewer translocons than ribosomes in a cell, it is essential that secretory protein-translating ribosomes only occupy translocons transiently. Therefore, if translocons are obstructed by ribosomes stalled or slowed in translational elongation, it possibly results in deleterious consequences to cellular function. Hence, we investigated how translocon clogging by stalled ribosomes affects mammalian cells. First, we constructed ER-destined translational arrest proteins (ER-TAP) as an artificial protein that clogged the translocon in the ER membrane. Here, we show that the translocon clogging by ER-TAP expression activates triage of signal sequences (SS) in which secretory pathway proteins harboring highly efficient SS are preferentially translocated into the ER lumen. Interestingly, the translocon obstructed status specifically activates inositol requiring enzyme 1α (IRE1α) but not protein kinase R-like ER kinase (PERK). Given that the IRE1α-XBP1 pathway mainly induces the translocon components, our discovery implies that lowered availability of translocon activates IRE1α, which induces translocon itself. This results in rebalance between protein influx into the ER and the cellular translocation capacity.Key words: endoplasmic reticulum, translocation capacity, translocon clogging, IRE1, signal sequence.

While anaphylaxis can occur at any time during general anesthesia, 90% of cases occur at induction of anesthesia. As several drugs are administered simultaneously at this time, it is difficult to identify the causative agent. However, it has been found that rocuronium is the most common drug associated with perioperative anaphylaxis. We treated 2 cases of patients who were administered sugammadex for anaphylactic symptoms thought to be caused by rocuronium, after which the anaphylactic symptoms disappeared. One of the most important aspects of treating anaphylactic shock is improving hemodynamics. If signs indicating circulatory collapse are observed, epinephrine should be administered immediately. However, because rocuronium was suspected of being the causative agent, and taking the patients' clinical course over time into consideration, sugammadex was initially administered. As a result, symptoms improved. Therefore, we believe that the administration of sugammadex may be effective for treating anaphylaxis caused by rocuronium and also help in identifying the causative agent.

Following the emergence of antibiotic-resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-resistant Staphylococcus pseudintermedius (MRSP), phage therapy has attracted significant attention as an alternative to antibiotic treatment. Bacteriophages belonging to kayvirus (previously known as Twort-like phages) have broad host range and are strictly lytic in Staphylococcus spp. Previous work revealed that kayvirus ɸSA039 has a host-recognition mechanism distinct from those of other known kayviruses: most of kayviruses use the backbone of wall teichoic acid (WTA) as their receptor; by contrast, ɸSA039 uses the β-N-acetylglucosamine (β-GlcNAc) residue in WTA. In this study, we found that ɸSA039 could switch its receptor to be able to infect S. aureus lacking the β-GlcNAc residue by acquiring a spontaneous mutation in open reading frame (ORF) 100 and ORF102. Moreover, ɸSA039 could infect S. pseudintermedius, which has a different WTA structure than S. aureus. By comparison, with newly isolated S. pseudintermedius-specific phage (SP phages), we determined that glycosylation in WTA of S. pseudintermedius is essential for adsorption of SP phages, but not ɸSA039. Finally, we describe a novel strategy of S. aureus which protects the bacteria from infection of SP phages. Notably, glycosylation of ribitol phosphate (RboP) WTA by TarM or/and TarS prevents infection of S. aureus by SP phages. These findings could help to establish a new strategy for the treatment of S. aureus and S. pseudintermedius infection, as well as provide valuable insights into the biology of phage-host interactions.

Tongue pressure is reportedly associated with dysphagia. This study investigated relationships among characteristics of head and neck cancer, tongue pressure and dysphagia screening tests performed in patients with head and neck cancer during the acute phase after surgical resection. Fifty-seven patients (36 men, 21 women; age range 26-95 years) underwent surgical resection and dysphagia screening tests (Repetitive Saliva Swallowing Test, Water Swallowing Test, Modified Water Swallowing Test and Food Test) and pre- and postoperative measurement of tongue pressure at 5 time points (preoperatively, and 1-2 weeks and 1, 2, and 3 months postoperatively). Progression of cancer (stage), tracheotomy, surgical reconstruction, chemotherapy, radiotherapy and neck dissection were factors associated with postoperative tongue pressure. Data were analyzed by linear mixed-effect model, Spearman correlation coefficient and receiver operating characteristic (ROC) curve. Tongue pressure was significantly reduced 1-2 weeks after surgery, and recovered over time. Changes in tongue pressure were significantly associated with stage, radiotherapy and reconstruction. All screening tests showed a significant relationship with tongue pressure. Analysis of ROC and area under the effect curve suggested that a tongue pressure of 15 kPa can be used as a cut-off value to detect dysphagia after surgery for head and neck cancer. Our results suggest that tongue pressure evaluation might offer a safe, useful and objective tool to assess dysphagia immediately postoperatively in patients with head and neck cancer.

Influenza virus RNA-dependent RNA polymerase is composed of three viral proteins, PB1, PB2, and PA. The host protein Ebp1 (ErbB3-binding protein1) interacts with PB1, and inhibits both in vitro RNA synthesis and virus replication. On Western blotting, the induction of Ebp1 was observed after influenza virus infection. To understand the induction of Ebp1 by influenza virus infection, we introduced a series of deletions within the 981-nucleotide long sequence located upstream of the Ebp1 gene (-664 to +317 nt from the transcription initiation site) and ligated them to the GFP gene. GFP expression assays indicated that the 981-nt upstream region was required for expression of GFP in not all cells but some cells. Microscopic analysis of the transformants showed that GFP expression was up-regulated by the influenza virus infection. Furthermore, quantitative real-time PCR indicated that influenza virus infection induced Ebp1 mRNA expression. Our data showed that (i) the newly synthesized vRNP of influenza virus induces Ebp1 expression; (ii) the Ebp1 promoter localizes between -664 nt and the initiation site of the Ebp1 gene, +317-nt long sequence in the noncoding region is required for regulation of Ebp1 gene expression; and (iii) Ebp1 expression level is correlated with virus protein expression level., (© 2011 The Authors. A novel method of monocyte culture in vitro was developed. The fraction of monocytes was obtained by density centrifugation of heparinised human venous blood samples. Monocytes were suspended in a modified Rosewell Park Memorial Institute medium (RPMI)-1640 (mRPMI) supplemented with 10% non-inactivated autologous serum added to the feeder cells. An avian cell line was used for feeder cells. Only those monocytes that settled on feeder cells grew rapidly at 37°C-38°C into a formation of clumped masses within two to three days. The cell mass was harvested and subcultures were made without feeder cells. A stable cell line (K63) was established from subcultures using a limited dilution method and cell cloning in microplates. K63 cells were adapted for later growth in the mRPMI medium supplemented with 10% foetal calf serum. The cells were well maintained at over 50th passage levels. This method proved to be applicable for monocyte cultures of animals as well.

Two cell lines, named KCEK and KCEL, were established from chicken embryonic kidney and lung. The basal culture medium was newly developed and the cell growth medium consisted of K1999 supplemented with 10% heat inactivated chicken serum. Both cells were well adapted to grow in vitro and more than 50 passages have been made so far. Once the cell lines were established the cells were easily adapted to grow in other growth media supplemented with fetal calf serum. Neither tumor formation in chicks nor P52 avian leucosis common antigen was detected in these cells. However, the oncogene analysis on these cells has not been performed yet. Both cells were permissive hosts for the Aujeszky's disease virus, Newcastle disease virus, and vesicular stomatitis virus.

A feline monocytic cell line was established from a venous blood sample obtained from a healthy male donor cat. The cloned cells, temporarily named FLMo/K02, were successively passaged in vitro with cell growth medium consisting of RPMI-1640 supplemented with 10% heat-inactivated fetal calf serum. Non-specific esterase and acid phosphatase, as marker enzymes, were clearly demonstrated by cytochemical examinations. The cells treated with allogeneic serum for two hours in advance showed enhanced reactivity to monoclonal antibodies, feline CD1a, canine CD11b, feline CD11c, canine CD11d, and feline MHC-II.

A strain of porcine epidemic diarrhea virus (PEDV), P-5V, utilized as a live virus vaccine in Japan was infected to a swine cell lines, KSEK6 and IB-RS-2 cells. Clear CPE, characterized by cellular destruction, started to appear in the infected cells on 2-3 days post infection (DPI) and affected cells was completely degenerated on 4 DPI. The virus was serially passaged in the cells even without addition of trypsin. Small but clear plaques were formed under an agar overlay medium on the cells. The infective titer in the order of 10(7.00-7.50) TCID50 per ml was obtained at usual incubation temperature.

The propagation of seventeen virus strains, classified into five virus families, in a swine monocyte cell line (SW/K99) was studied in the point of infective progeny production. The viruses examined were adapted to grow in a swine epithelial cell line (KSEK6) and were proved to show clear CPE in advance. These viruses were successively passaged in the monocyte cultures regardless of CPE occurrence. The cultures at 3rd passage level were titrated for their infectivity using KSEK6 cells. Out of the viruses examined, only Aujeszky's disease virus (ADV) was able to propagate in the monocytes. Four ADV strains, two virulent and two attenuated strains, were compared for their growth in two cell lines. The final amount of infective progeny measured at 72 hours incubation at 36.5 degrees C was almost in a similar order between 2 cell lines. However, the amount of infective progeny produced at 24 hours incubation was higher in SW/K99 cells than that in KSEK6 cells. The occurrence of CPE was also more evident in SW/K99 cells in an early incubation. The data indicate that viral susceptibility of the monocytes to ADV is higher than that of epithelium. Other viruses were abortive in the monocytes.

A swine monocyte cell line was established from peripheral blood sample collected from a healthy adult male pig. The cloned cells grow actively in forming monolayers in both glass and plastic cell culture flasks with the growth medium reported previously (Kadoi, 2000) at 36.5 degrees C incubation. The plating efficiency is more than 95%. Densely grown cells in flasks show an epithelioid morphology. The fundamental properties of the cells were examined for cytological definition as monocytes. A positive property detected was guinea pig complement receptor, porcine IgG receptor, non-specific esterase, and acid phosphatase. A significant phagocytic activity proved by the inoculation of Saccharomyces cerevisiae is also one of the characteristics observed in the LPS-activated cells.

Five conventional Beagle dogs were intravenously injected with ten million canine monocyte cells (Cn/K99) cultured in vitro (Kadoi, 2000) adsorbed with a strain of calicivirus originally isolated from lions (Kadoi et al., 1997). Another two Beagle dogs were injected similarly with the virus suspension solely as control. Serum samples were collected from these dogs at intervals and specific seroneutralizing antibody production against the virus was measured in vitro. A significantly higher antibody production was demonstrated in the five dogs group. A clear booster effect was also proved in the sera of the dogs after the second virus inoculation made on day 100. A possibility of antigen presentation function of non-self monocytes is suggested.

A cytopathic virus was isolated from young pigs suffering from severe diarrhea in Okinawa, Japan in 1986. The disease was highly contagious among young pigs. The physico-chemical properties of the virus indicated an enterovirus, but, no of serological relationship was detected with reference strains of porcine enteroviruses. With the aid of Genbank for genomic sequence data, RT-PCR and hybridization method was performed. The viral isolate was identified as the Coxsackievirus (CV) B5 of human enterovirus. This is the first report of the isolation of CV B5 from pigs in Japan.

Since human caliciviruses are responsible for viral gastroenteritis transmitted by contaminated foods and the viruses barely propagate in cell culture, feline caliciviruses were employed as a model for the measurement of their stability in marine water. Survival of four strains of feline calicivirus in marine water was measured when the seed viruses were diluted 1/10 with marine water and maintained at 4 degrees C, 10 degrees C, and 20 degrees C respectively. Among the virus strains studied, a considerable amount of infective viruses remained at 10 degrees C or lower temperature conditions even for a period of 30 days.

A canine monocyte cell line was established from the peripheral blood sample collected from a healthy young male Beagle dog. The cloned cells grew easily and were serially passaged in vitro in the medium, a slight modification further made on Dulbecco's modified Eagle medium, supplemented with 10% fetal bovine serum. The morphology of single cell was shown in triangular or round form, however, it became epithelioid in a densely grown monolayer. Non-specific esterase was detected in all cells by a cytochemical examination. The cells reacted rapidly to the addition of a small amount of LPS and differentiated to the cells of morphologically typical macrophages. Both complement receptor (CD35) and Fc gamma receptor (CD64) were demonstrated on the cell membrane.

Two SPF kittens were experimentally infected via the intranasal route with a strain of calicivirus originally isolated from sick lion suffering from vesicular disease. Fever, 40.3 degrees C and 40.7 degrees C, and vesicular formation in tongue and snout were reproduced in both kittens. The infected virus was recovered from nasal, oral and rectal swabs. A longer duration of virus recovery was proved with oral swab samples taken from 1 to 10 and 12 days post infection. This suggests that tongue and oral tissues are the main tissues for virus multiplication.

One of the major viral respiratory diseases of cattle is parainfluenza type-3 virus infection. Attenuated live virus vaccines have been used in most countries. An experimental attempt was made to clarify the antibody response of cattle inoculated with a live vaccine in a post-tracheal position by a spray method. A significantly higher titers of antibodies in both hemagglutination inhibition and neutralization were obtained in the serum samples of animals vaccinated in this manner than those vaccinated by intramuscular injection or intranasal instillation.

The majority of lions and tigers in a Safari park in Japan were suffering from acute vesicular disease caused by a calicivirus infection. All of these animals were injected subcutaneously with an inactivated calicivirus vaccine experimentally prepared with one of the seed viruses originally isolated from sick lions. Seroneutralizing antibody of paired sera collected from ten female lions at two week intervals was measured. A significant elevation of specific antibody was detected in the serum samples and no local or systemic reactions associated with vaccination were observed.

We studied the epidemiology of influenza viruses in Thailand by isolating them and comparing their antigenic features with those of Japanese isolates. Between 1991 and 1994, 32 strains were isolated from 186 throat swab specimens. Twenty-one of the 32 isolates were of type A, subtype H3N2, and 11 strains were type B. It was suggested that the isolates of type A, subtype H3N2, drifted antigenically from A/Beijing/352/89-like to A/Kitakyusyu/159/93-like variants used as reference strains for comparison. The type B isolates in 1991 were suspected to be antigenically different from those of B/Bangkok/163/90, Thailand, in HI tests. These 1991 isolates were similar to B/Mie/1/93, which was isolated in the latter half of the epidemic in Japan in winter 1992/1993.

A polyethylene glycol treatment was given to fuse KSEK6 cells, an established cell line derived from porcine embryo kidney, with the lymphocytes, separated from spleens of 35 apparently healthy slaughtered pigs. Eight cytopathic virus strains were isolated from the lymphocytes of these pigs. Two virus strains were isolated by inoculating the spleen tissue homogenates to KSEK6 monolayer cultures. All of viruses were identified as porcine adenoviruses according to their physicochemical, serological and immunological properties. One of these virus strains was serologically proved to be independent from six serotypes of porcine adenoviruses ever known. The electrophoretic property of viral DNA of this strain was indicated to be different from those of other reference porcine adenoviruses. This means the presence of a 7th serotype in porcine adenoviruses.

In December 1992, 17 African lions and 7 Siberian tigers in a Safari park in Japan became sick with characteristic clinical symptoms of acute vesicular formations on tongue and snout. The disease was highly contagious since all of these animals showed similar symptoms within two days after the onset of the first case. Swabs were taken from affected animals in rubbing tongues, snouts and some from rectums. Cytopathic viruses were isolated on CRFK cell culture by virological tests. The physicochemical property of a representative virus strain, named Arthur/L, isolated from a male lion was identified as a member of Caliciviridae. However, seroneutralization test indicated that this virus strain was antigenically distinct from Japanese isolates of feline caliciviruses used for comparison. Viral capsid proteins of the present isolate, Arthur/L, and of a feline calicivirus, strain FC7, were compared in an electrophoresis in SDS-PAGE gel. The major viral capsid polypeptide of them were proved to be significantly different in molecular weight. The polypeptide of FC7 was estimated to be ca. 63 KDa whereas that of Arthur/L consisted of 2 components of ca. 65 and 62 KDa. The viral proteins of these two strains were also proved to be distinct by an immunoblotting test.

Viral susceptibility of a newly established cell line, named KMP, derived from the peritoneal cavity of BALB/C mouse is described. The cells were originally cloned from the in vitro culture of ascites of the mouse injected with Ehrlich ascitic tumor cells in advance. The electrophoretic pattern of cellular DNAs, extracted from KMP, normal BALB/C mouse spleen, and Ehrlich tumor cells respectively were compared after triple digestions with restriction endonucleases. This cell line was proved to be of mouse origin, but not the sub-line of Ehrlich tumor cells. The strains of Coxsackie virus B group, swine enterovirus, influenza virus, encephalomyocarditis virus, vesicular stomatitis virus, and Aujeszky's disease virus were able to multiply well in the cell line with considerably high infectious titers in showing clear CPE and circular plaques.

An inactivated adenovirus vaccine experimentally prepared was inoculated into young pigs against the adenovirus infection newly appeared in Japan. Sero-neutralizing titers of the paired sera collected from 50 pigs in the interval of 3 weeks was measured. The mean of antibody titers of post-sera was 45 times higher than that of pre-sera.

Serotyping of porcine adenoviruses needs type-specific antiserum. The injections of purified virions to SPF Balb/C mice, followed by an intraperitoneal injection of Ehrlich ascitic tumour cells, gave enough ascitic fluid containing practically applicable type-specific antibody.

An established cell line, KSEK6, derived from swine embryo kidney proved to be a suitable host for the propagation of a hemagglutinating encephalomyelitis virus, strain 67N. Infected cells showed clear cytopathic effect as early as 24 hours incubation at 37 degrees C. Plaques easily visible to the naked eye were also produced under agar overlay medium. The infective titer of 3rd passage level in the cells was in the order of 10(8) PFU per ml. Detecting antibodies against this virus strain in swine sera was considered to be more accurate by neutralization test than by hemagglutination inhibition.

A mechanism involved in the plaque enhancement effect of foot-and-mouth disease viruses (FMDV) by the addition of sodium thiosulfate (Hypo) in the agar overlay medium (AOM) previously reported was studied. It was experimentally proved that the diffusion of virus particles through agar overlay medium was enhanced when this salt was incorporated. Accordingly, the enlarged plaque formation was assumed to be caused by the enhanced diffusion of viral progenies produced in infectious centers during plaque assays.

The viral susceptibility of a cell line, named KSEK6, newly established from the kidney cortex of swine embryo was tested for the indication of CPE occurrences and also plaque formations. The multiplication of porcine adenoviruses was considerably high in the cells among the virus strains tested though plaques of these viruses were hardly visible under agar overlay medium. Two strains of swine enteroviruses, Aujeszky's disease virus and hemagglutinating encephalomyelitis virus also multiplied well in a similar order to those received in the other cells employed.
