Freshwater scarcity demands exploration of alternative resources like saline water and soils. Understanding the molecular mechanisms behind NaCl regulation in potential crop plants becomes increasingly important for promoting saline agriculture. This study investigated the euhalophyte Salicornia europaea, analyzing its gene expression, yield, and total phenolic compounds under hydroponic cultivation. We employed five salinity levels (0, 7.5, 15, 22.5, and 30 g/L NaCl) across five harvests at 15-day intervals, capturing plant development. Notably, this design deviated from conventional gene expression studies by recording organ-specific responses (shoots and roots) in plants adapted to long-term salinity treatments at various developmental stages. The highest fresh mass of S. europaea was observed four months after germination in 15 g/L NaCl. Identifying a reliable set of reference genes for normalizing gene expression data was crucial due to comparisons across shoots, roots, developmental stages, and salinity levels. A set of housekeeping genes – ubiquitin c (SeUBC), actin (SeActin) and dnaJ-like protein (SeDNAJ) – was identified for this purpose. Interestingly, plants grown without NaCl (0 g/L) displayed upregulation of certain genes associated with a NaCl deficiency related nutritional deprivation. These genes encode a tonoplast Na+/H+-antiporter (SeNHX1), a vacuolar H+-ATPase (SeVHA-A), two H+-PPases (SeVP1, SeVP2), a hkt1-like transporter (SeHKT), a vinorine synthase (SeVinS), a peroxidase (SePerox), and a plasma membrane Na+/H+-antiporter (SeSOS1). Other genes encoding an amino acid permease (SeAAP) and a proline transporter (SeProT) demonstrated marginal or dispersing salinity influence, suggesting their nuanced regulation during plants development. Notably, osmoregulatory genes (SeOsmP, SeProT) were upregulated in mature plants, highlighting their role in salinity adaptation. This study reveals distinct regulatory mechanisms in S. europaea for coping with varying salinity levels. Identifying and understanding physiological reactions and sodium responsive key genes further elucidate the relationship between sodium tolerance and the obligate sodium requirement as a nutrient in euhalophytes.

The sclerotia of Sclerotinia sclerotiorum serve as a primary inoculum source for initiating infections. This study aimed to evaluate the effects of environmental factors on sclerotial germination under field conditions by establishing sclerotia depots to monitor apothecia appearance over four consecutive years. Additionally, the effects of soil moisture content (25%, 50%, 75%, and 95%), air temperature (10 °C/10 °C, 14 °C/10 °C, and 18 °C/10 °C), and light conditions (white and UV light) on sclerotial germination and apothecial formation were investigated under controlled conditions with a 17 h day/7 h night regime. Furthermore, variability in carpogenic germination among S. sclerotiorum isolates was examined. From 2021 to 2024, significant differences were observed in both the sclerotia germination rate and timing of germination within the season. High soil moisture, particularly prolonged wetness, and soil temperatures between 10 and 14 °C were key factors for apothecial formation under field conditions. Under controlled conditions, higher soil moisture levels (75% and 95%) accelerated sclerotial germination, with sclerotia incubated at 14 °C/10 °C germinating earlier after 38 days than those at 10 °C/10 °C or 18 °C/10 °C. Additionally, the type of light significantly affected apothecial formation, which was observed only in treatments exposed to a combination of white and UV light. Furthermore, significant variations were also found in the duration until sclerotia of different S. sclerotiorum isolates produced the first stipe and the first apothecium, indicating that the genetic characteristics of each isolate affect carpogenic germination.

Salinization is a major cause of soil degradation that affects several million hectares of agricultural land, threatening food security and the sustainability of agricultural systems worldwide. Nevertheless, despite the negative impact of salinity, salt-affected land also provides several important ecosystem services, from providing habitats and nurseries for numerous species to sustainable food production. This opinion paper, written in the framework of the EU COST Action CA22144 SUSTAIN on the sustainable use of salt-affected land, therefore, focuses on the potential of halophytes and saline agriculture to transform and restore key functions of these salt-affected and marginal lands. As the current knowledge on sustainable saline agriculture upscaling is fragmented, we highlight (i) the research gaps in halophyte and salinity research and (ii) the main barriers and potentials of saline agriculture for addressing food security and environmental sustainability in terms of population growth and climate change.

Abstract The increase in mineral and ash-rich waste biomass (MWB) generation in emerging economies poses critical environmental problems and bottlenecks the solid waste and wastewater treatment systems. Transforming these MWB such as sewage sludge from wastewater treatment (SSW) to biochar can be a sustainable method for their disposal and resource recovery. However, such biochar has limited applicability due to the relatively low organic content and possibly contaminated nature of SSW. This may be offset through combined pyrolysis with other MWB, which can also support municipal solid waste management. Studies on this MWB co-pyrolysis are lacking and have not yet seen successful long-term implementation. This work is the second part of authors’ research encompassing an analytical and lab-scale investigation of biochar production from MWB through pyrolysis for the case of Chennai city, India. Here, the physicochemical properties of biochar derived from lab-scale co-pyrolysis of SSW with other MWB such as anaerobic digestate from waste to energy plants of food, kitchen or market waste fermentation, and banana peduncles (BP) collected from vegetable markets and their thermolysis mechanism are comprehensively investigated for purpose of carbon sequestration. Also, a novel preliminary investigation of the effect of sample weight (scaling effect) on the analytical pyrolysis of biomass (BP as model substrate) is undertaken to elucidate its impact on the heat of pyrolysis and carbon distribution in resultant biochar. The maximum carbon sequestration potential of the derived biochar types is 0.22 kg CO2 kg−1 biomass. The co-pyrolysis of MWB is exothermic and governed by the synergetic effects of the components in blends with emission profiles following the order CO2 > CH4 > CO > NH3. Co-pyrolysis reduced the heavy metal enrichment in SSW biochar. The derived biochars can be an immediate source of N, P and S in nutrient-deficient acidic soils. The biochar has only up to 4-ring polyaromatic compounds and a residence time longer than 1 h at 500 °C is necessary to improve carbonization. The heat released during analytical pyrolysis of the model biomass and distribution of carbon in the resultant biochar are significantly influenced by scaling effects, drawing attention to the need for a more detailed scaling investigation of biomass pyrolysis.

Abstract Salt-tolerant halophytes have shown potential for biorefinery and agricultural use in salt-affected soils, increasing the value of marginal lands. They could provide a bio-based source for compounds obtained from the petrochemical industry or an alternative for biomass currently imported overseas. Salicornia europaea, Tripolium pannonicum and Crithmum maritimum were cultivated in hydroponic systems under various salinity conditions, harvested green but not food-grade, and fractionated to green juice and fibre residue. Obtained fractions were characterised for contents of carbohydrates, Klason lignin, crude protein, organic acids, lipids, and minerals to evaluate the biomass’ suitability for biorefinery. Significant differences were observed in the biomass yield and the composition of the biomass fractions from different cultivation salinities. High concentrations of crude protein were found. Thus, these species could have the potential for green protein production. Fractions rich in carbohydrates could be used for lignocellulose processing and processes utilising micro-organisms.

Seagrass meadows provide valuable ecosystem services but are fragile and threatened ecosystems all over the world. This review highlights the current advances in seagrass research from Viet Nam. One goal is to support decision makers in developing science-based conservation strategies. In recent years, several techniques were applied to estimate the size of seagrass meadows. Independent from the method used, there is an alarming decline in the seagrass area in almost all parts of Viet Nam. Since 1990, a decline of 46.5% or 13,549 ha was found. Only in a few protected and difficult-to-reach areas was an increase observed. Conditions at those sites could be investigated in more detail to make suggestions for conservation and recovery of seagrass meadows. Due to their lifestyle and morphology, seagrasses take up compounds from their environment easily. Phytoremediation processes of Thalassia hemprichii and Enhalus acoroides are described exemplarily. High accumulation of heavy metals dependent on their concentration in the environment in different organs can be observed. On the one hand, seagrasses play a role in phytoremediation processes in polluted areas; on the other hand, they might suffer at high concentrations, and pollution will contribute to their overall decline. Compared with the neighboring countries, the total Corg stock from seagrass beds in Viet Nam was much lower than in the Philippines and Indonesia but higher than that of Malaysia and Myanmar. Due to an exceptionally long latitudinal coastline of 3,260 km covering cool to warm water environments, the seagrass species composition in Viet Nam shows a high diversity and a high plasticity within species boundaries. This leads to challenges in taxonomic issues, especially with the Halophila genus, which can be better deduced from genetic diversity/population structures of members of Hydrocharitaceae. Finally, the current seagrass conservation and management efforts in Viet Nam are presented and discussed. Only decisions based on the interdisciplinary cooperation of scientists from all disciplines mentioned will finally lead to conserve this valuable ecosystem for mankind and biodiversity.

The synthesis of indole-3-acetonitrile (IAN) from the indolic glucosinolate (iGSL) glucobrassicin (GB) is a unique trait of members of the Brassicales. To assess the contribution of this pathway to indole-3-acetic acid (IAA) synthesis under stress conditions, drought stress (DS) experiments with Arabidopsis thaliana were performed in vitro. Analysis of GSLs in DS plants revealed higher contents of GB in shoots and roots compared to control plants. Deuterium incorporation experiments showed the highest turnover of GB compared to all other GSLs during drought conditions. Evidence suggests the involvement of the thioglucosidase BGLU18 in the degradation of GB. The nitrile specifier proteins NSP1 and NSP5 are known to direct the GSL hydrolysis towards formation of IAN. Nitrilases like NIT2 are able to subsequently synthesize IAA from IAN. Expression of BGLU18, NSP1, NSP5 and NIT2 and contents of GB, IAN and IAA were significantly elevated in DS plants compared to control plants suggesting the increased use of GB as IAA source. Significantly higher contents of reactive oxygen species in DS bglu18 and epithionitrile specifier protein (esp) mutants compared to Col-0 indicate higher stress levels in these mutants highlighting the need for both proteins in DS plants. Furthermore, GB accumulation in leaves was higher in both mutants during DS when compared to Col-0 indicating enhanced synthesis of GB due to a lack of breakdown products. This work provides the first evidence for the breakdown of iGSLs to IAN which seems to be used for synthesis of IAA in DS A. thaliana plants.

Moringa oleifera is considered to be one of the most valuable and beneficial crop tree species. The great nutritiousness is assigned to its high leaf protein content, and its health-promoting effect to the anti-carcinogenic properties of its genuine glucosinolates and their degradation products. From a plant physiological perception protein contents of 30% seem to be quite high. Accordingly, a reconsideration of these findings became necessary. The related inquiry unveils that also in the leaves of many other plant species such high protein contents are reported, provided that they are quantified by Kjeldahl nitrogen determinations. But, it is rather likely that the nitrogen accounting for the putative high protein contents is due to insoluble cell wall bound hydroxyproline-rich glycoproteins. Due to their extreme insolubility, these compounds cannot be digested easily, and thus, they do not contribute to the nutritiousness of M. oleifera leaves. In contrast to classical glucosinolates, those occurring in M. oleifera are characterized by an attachment of a rhamnose to the aglycone. In consequence, the products generated during the myrosinase-cata-lysed hydrolysis correspond to non-volatile rhamnosides of isothiocyanates. Since over time, olfactorily active substances emerge, the rhamnose moiety has to be cleaved off, putatively by a corresponding rhamnosidase.

Abiotic stress conditions, such as salinity, affect plant development and productivity and threaten the sustainability of agricultural production. Salt has been proven to accumulate in soil and water over time as a result of various anthropogenic activities and climatic changes. Species of the genus Salicornia thrive in the most saline environments and have a wide climatic tolerance. They can be found in a variety of subtropical, oceanic, and continental environments. This study aims to establish Salicornia europaea as a novel source of plant-based compounds that can grow in areas unsuitable for other crops. The morphological and compositional changes in the tissues of S. europaea in different consecutive developmental stages have not been investigated so far. Therefore, a comprehensive study of changes during the lifecycle of S. europaea was carried out, following changes in the plant’s composition, including biomass yield, and soluble and insoluble compounds. For this, plants were cultivated in hydroponics for 15 weeks and harvested weekly to analyze biomass production, to determine soluble and insoluble compounds, protein content, and polyphenols. According to the results, glucan, xylan, and lignin increase with plant age, while water extractives decrease. Protein content is higher in young plants, while flavonoid content depends on the phenological stage, decreasing in the early flowering stage and then increasing as plants enter early senescence. Our results can aid in finding the optimal harvesting stage of S. europaea, depending on the component of interest.

The phytotoxic mechanisms of patulin (PAT) and penicillic acid (PA) have not been identified unambiguously. This study aims to clarify their effects on thioredoxins (Trxs). Aflatoxin B1 (AFB1), PAT and PA were isolated by solvent extraction and chromatographic techniques from the cultures of Aspergillus flavus Z2 (LC171449), Penicillium vulpinum CM1 and Aspergillus ochraceus EMCC516, respectively. The three mycotoxins showed phytotoxicity to the germination of maize seeds, which was manifested by inhibiting radicle and coleoptile emergence, in addition to their toxic effects on fresh weights and root and shoot lengths. The phytotoxicity of AFB1 and PAT was greater than that of PA. Due to the central roles exhibited by plant Trxs in cellular metabolic activities, they were tested as target proteins for PAT and PA using AFB1 as positive control. In vivo studies showed that the mycotoxins significantly reduced Trx activity measured in the roots and shoots of maize seedlings. PAT showed greater Trx-inhibiting activity than PA and AFB1. In vitro studies of the mycotoxins on Trx y1 (from Arabidopsis thaliana) and thioredoxin reductase (Tr, from rat liver) activities confirm the results of in vivo studies. The inactivation of Trx with PAT and PA was reduced in the presence of glutathione (GSH). Data obtained suggest that lactone mycotoxins are more highly reactive with simple low-molecular-weight thiols (like GSH) than with complex ones (like Trx).

The implementation of digital tools into science education is a major demand of various stakeholders, such as teachers, schools and ministries of education. However, teaching innovations and the introduction of new competences need to be carefully tested and optimized for successful and sustainable application and learning success. Our aim was to develop and establish an easily adaptable teaching unit comprising the aspects of 3D printing from computer-aided modeling to slicing, printing and post-processing, which is linked to curricular learning content. The original teaching concept developed with a small group of students has been adapted to the conditions in large groups and full-size 9th grade school classes. With an increased sample size, it was now possible to investigate and analyze the teaching approach with respect to student’s motivation, learning success as well as the quality and acceptance of the teaching–learning arrangement for designing and 3D printing flower models. The goal of the study was to further optimize the existing teaching tool based on the evaluation of the student experience. While the exploration of this teaching approach ties into the current discourse of innovative biology teaching, the efficacy is evidenced by results that indicate a positive impact on student’s motivation and a high learning success regarding computer-aided modeling and 3D printing. As a result, the teaching-revised concept reported in this article is based on the students’ evaluation and can be provided as well-tested teaching material for schools.

The genus Halophila shows the highest species diversity within the seagrass genera. Southeast Asian countries where several boundary lines exist were considered as the origin of seagrasses. We hypothesize that the boundary lines, such as Wallace's and Lydekker's Lines, may act as marine geographic barriers to the population structure of Halophila major. Seagrass samples were collected at three islands in Vietnamese waters and analyzed by the molecular maker ITS. These sequences were compared with published ITS sequences from seagrasses collected in the whole region of interest. In this study, we reveal the haplotype and nucleotide diversity, linking population genetics, phylogeography, phylogenetics and estimation of relative divergence times of H. major and other members of the Halophila genus. The morphological characters show variation. The results of the ITS marker analysis reveal smaller groups of H. major from Myanmar, Shoalwater Bay (Australia) and Okinawa (Japan) with high supporting values. The remaining groups including Sri Lanka, Viet Nam, the Philippines, Thailand, Malaysia, Indonesia, Two Peoples Bay (Australia) and Tokushima (Japan) showed low supporting values. The Wallacea region shows the highest haplotype and also nucleotide diversity. Non-significant differences were found among regions, but significant differences were presented among populations. The relative divergence times between some members of section Halophila were estimated 2.15-6.64 Mya.

Resources such as fertile soil and clean water are already limited in many parts of the world. Additionally, the conventional use of arable land is becoming increasingly difficult, which is further exacerbated by climate change. Soilless cultivation systems do not only offer the opportunity to save water and cultivate without soil but also the chance to open up urban areas such as residential rooftops for food production in close proximity to consumers. In this review, applications of soilless farming systems are identified and compared to conventional agriculture. Furthermore, aspects of economic viability, sustainability and current developments are investigated. An insight into the most important soilless farming systems—hydroponics, aquaponics and vertical farming—is provided. The systems are then differentiated from each other and, as far as possible, evaluated in terms of their environmental impact and compared with conventional cultivation methods. Comparing published data analyzing the yield of hydroponic cultivation systems in comparison to soil-based cultivation methods enables a basic overview of the profitability of both methods and, thus, lays the foundation for future research and practical applications. The most important inert substrates for hydroponic applications are presented, and their degree of sustainability is compared in order to emphasize environmental impacts and affect substrate selections of future projects. Based on an assessment of the most important soilless cultivation systems, the challenges and developments of current techniques are highlighted and discussed.

The halophyte plant species Salicornia europaea and Salicornia ramosissima were investigated for their potential to serve as a substrate for biogas production. Salicornia europaea was cultivated in hydroponic systems under varying salt concentrations (0, 10, 20, and 30 g/L NaCl), while S. ramosissima was grown in greenhouse farming with aquaculture effluent irrigation. The biomethane potential of the two halophyte feedstocks was determined through batch experiments, and correlations to the plant biochemical composition were investigated. Ash and mineral content of S. europaea was correlated to the increasing salt concentration used for plant cultivation in hydroponic systems. No indication of inhibition of the anaerobic digestion process was detected for sodium concentrations of up to 2400 mg/L in the anaerobic batch-test assays. The highest biomethane yield of S. europaea of 250 mL CH4/gVS was obtained when grown under 20 g/L NaCl and up to 300 mL CH4/gVS for S. ramosissima. By concentrating the dry matter content, the biomethane yield per ton of feedstock could be increased from 24 m3 CH4/t of the fresh halophyte plant to 74 m3 CH4/t by fractionation into a pulp fraction and to 149 m3 CH4/t by drying of the plant at room temperature for 1 week.

Abstract Background Biofilms have great significance in healthcare-associated infections owing to their inherent tolerance and resistance to antimicrobial therapies. New approaches to prevent and treat unwanted biofilms are urgently required. To this end, three seagrass species (Enhalus acoroides, Halophila ovalis and Halodule pinifolia) collected in Vietnam and in India were investigated for their effects in mediating non-lethal interactions on sessile bacterial (Escherichia coli) and fungal (Candida albicans) cultures. The present study was focused on anti-biofilm activities of seagrass extracts, without killing cells. Methods Methanolic extracts were characterized, and major compounds were identified by MS/MS analysis. The antibiofilm properties of the seagrass extracts were tested at sub-lethal concentrations by using microtiter plate adhesion assay. The performance of the most promising extract was further investigated in elegant bioreactors to reproduce mature biofilms both at the solid/liquid and the solid/air interfaces. Dispersion and bioluminescent assays were carried out to decipher the mode of action of the bioactive extract. Results It was shown that up to 100 ppm of crude extracts did not adversely affect microbial growth, nor do they act as a carbon and energy source for the selected microorganisms. Seagrass extracts appear to be more effective in deterring microbial adhesion on hydrophobic surfaces than on hydrophilic. The results revealed that non-lethal concentrations of E. acoroides leaf extract: i) reduce bacterial and fungal coverage by 60.9 and 73.9%, respectively; ii) affect bacterial biofilm maturation and promote dispersion, up to 70%, in fungal biofilm; iii) increase luminescence in Vibrio harveyi by 25.8%. The characterization of methanolic extracts showed the unique profile of the E. acoroides leaf extract. Conclusions E. acoroides leaf extract proved to be the most promising extract among those tested. Indeed, the selected non-lethal concentrations of E. acoroides leaf extract were found to exert an antibiofilm effect on C. albicans and E. coli biofilm in the first phase of biofilm genesis, opening up the possibility of developing preventive strategies to hinder the adhesion of microbial cells to surfaces. The leaf extract also affected the dispersion and maturation steps in C. albicans and E. coli respectively, suggesting an important role in cell signaling processes.

Abstract Sulphotransferases are a diverse group of enzymes catalysing the transfer of a sulfuryl group from 3′-phosphoadenosine 5′-phosphosulphate (PAPS) to a broad range of secondary metabolites. They exist in all kingdoms of life. In Arabidopsis thaliana (L.) Heynh. twenty-two sulphotransferase (SOT) isoforms were identified. Three of those are involved in glucosinolate (Gl) biosynthesis, glycosylated sulphur-containing aldoximes containing chemically different side chains, whose break-down products are involved in stress response against herbivores, pathogens, and abiotic stress. To explain the differences in substrate specificity of desulpho (ds)-Gl SOTs and to understand the reaction mechanism of plant SOTs, we determined the first high-resolution crystal structure of the plant ds-Gl SOT AtSOT18 in complex with 3′-phosphoadenosine 5′-phosphate (PAP) alone and together with the Gl sinigrin. These new structural insights into the determination of substrate specificity were complemented by mutagenesis studies. The structure of AtSOT18 invigorates the similarity between plant and mammalian sulphotransferases, which illustrates the evolutionary conservation of this multifunctional enzyme family. We identified the essential residues for substrate binding and catalysis and demonstrated that the catalytic mechanism is conserved between human and plant enzymes. Our study indicates that the loop-gating mechanism is likely to be a source of the substrate specificity in plants.

Watercress, Nasturtium officinale R. Br., is a native water or semiaquatic plant that has a high nutrient density. Physiologically relevant are the various glucosinolates, which possess positive health effects in form of their thio- and isothiocyanates. In an interdisciplinary project, we aim to develop a hydroponic, and finally an aquaponic, circulatory cultivation system and to study the health effects of watercress. In humans, there is a lack of data-based knowledge on potential beneficial health effects of watercress. Growth of watercress was followed during one season in an open-door hydroponic system. Watercress was also cultivated in the greenhouse in different substrates with different concentrations of nutrients and salt. The biomass production is strongly dependent on the temperature. The glucosinolate contents differ significantly during the growing season, especially during flowering. Watercress naturally grows in nutrient rich fresh waters, however, when cultivated at NaCl concentrations of up to 120 mM the gain in biomass is still high. In a human proof of-concept study, indications for antioxidant and anti-inflammatory effects of fresh watercress were observed already after a single dose intake of fresh watercress (85 g). Further in vivo and in vitro studies are planned to study health beneficial effects of watercress and its metabolic activity.

This year we want to celebrate the 100th birthday of our journal, which was first published as Angewandte Botanik (= Applied Botany) in 1919. It started as journal for members of the “Vereinigung für Angewandte Botanik (German Society for Applied Botany)” only. On a monthly basis, it delivered the newest research results in the field of applied botany as well as short communications, literature reviews and reports from scientific meetings to the German speaking research community. After a short publication hiatus in the 1940ies, society members continued to report their findings, quickly exhausting the available print pages. Throughout the years, English titles and summaries where added, making the journal accessible for a wider, non-German speaking audience. The final step into the international research community was the opening of the journal for authors, who were not society members in the 1980ies. In 2004, the “Deutsche Gesellschaft für Qualitätsforschung - Pflanzliche Lebensmittel (German Society for Quality Research in Plant Foods)” joined the journal due to partial overlap in their research focus, thus, making it the Journal of Applied Botany and Food Quality as we know it today. The latest change happened in 2013, with the switch from a subscription based print journal to an online-only gold open access journal. This once again broadened the readership and gained the attention of even more international researchers, which nowadays make up the majority of our journal authors. The first article in "Angewandte Botanik" published in 1919 was submitted by Otto Appel, a phytopathologist well known for his discoveries on bacterial and fungal diseases of crops such as potato and cereals. His work ranged from fundamental research to applied research and reflected in his long-term membership in the board of the Association for Applied Botany. In the first contribution of this special jubilee section Hartwig Schulz and Heike Riegler report about "Otto Appel and his contributions to food quality and safety at the beginning of the 20th century" and give insights into the major challenges in phytopathology at the early years of the 20th century and discoveries by Otto Appel in this field. Maik Kleinwächter and Dirk Selmar prepared a contribution about "Modern Applied Botany - Changes in the perception of applied botanists to themselves and others during the last century. Three recent examples of the scientific potential of this field". They give a short outline on the history of applied research in plant biology in Germany and illustrate the relevance of modern Applied Botany in three relevant post-harvest processes. They state that interdisciplinary work and intensive cooperation with growers and producers is an integral part of developing feasible and economically acceptable solutions that can be successfully transferred into practice. The major challenge in Applied Botany today is the implementation of new concepts and ideas into product-related research. In the third article in the special section Claudia Bahmann, Thomas Rinas, Nicolas Niemenak, Elsa Hegmann, Bernward Bisping, David Seigler and Reinhard Lieberei describe how "Cacao seed tests controls transport processes during germination and fermentation-like incubation". The contribution of Muna Ali Abdalla and Karl H. Mühling leads us to "Plant-derived sulfur containing natural products produced as a response to biotic and abiotic stresses. A review of their structural diversity and medicinal importance" and reveals the diversity in structure and function of secondary compounds in plants. In the article written by Wolfgang Kreis "Exploiting plant cell culture for natural product formation" we get insights into the high expectations researchers and industry had on the exploitation of plant cell cultures from a historical and personal point of view. Imke Hutter and Carolin Schneider report on the current state of "Commercial micropropagation in Germany" from the applied site. In an interdisciplinary project, Jan Philipp Schuchardt, Andreas Hahn, Theresa Greupner, Paulina Wasserfurth, María Rosales-López, Johann Hornbacher and Jutta Papenbrock work on different aspects concerning "Watercress – cultivation methods and health effects". Georg Langenkämper and Christian Zörb submit an article about "Modern aspects of wheat grain storage proteins". Finally, on the interaction of plants with microorganisms, in this case fungi, is shed light on by Oluwatosin Abdulsalam, Erika Kothe and Katrin Krause in "The parasitic-neutral-mutual continuum of plant-fungal interactions".

The vegetable watercress (Nasturtium officinale R.Br.) is, besides being a generally nutritious food, a rich source of glucosinolates. Gluconasturtiin, the predominant glucosinolate in watercress, has been shown to have several health beneficial properties through its bioactive breakdown product phenethyl isothiocyanate. Little is known about the immunoregulatory effects of watercress. Moreover, anti-inflammatory effects have mostly been shown in in vitro or in animal models. Hence, we conducted a proof-of-concept study to investigate the effects of watercress on the human immune system. In a cross-over intervention study, 19 healthy subjects (26.5 ± 4.3 years; 14 males, 5 females) were given a single dose (85 g) of fresh self-grown watercress or a control meal. Two hours later, a 30 min high-intensity workout was conducted to promote exercise-induced inflammation. Blood samples were drawn before, 5 min after, and 3 h after the exercise unit. Inflammatory blood markers (IL-1β, IL-6, IL-10, TNF-α, MCP-1, MMP-9) were analyzed in whole blood cultures after ex vivo immune cell stimulation via lipopolysaccharides. A mild pro-inflammatory reaction was observed after watercress consumption indicated by an increase in IL-1β, IL-6, and TNF-α, whereas the immune response was more pronounced for both pro-inflammatory and anti-inflammatory markers (IL-1β, IL-6, IL-10, TNF-α) after the exercise unit compared to the control meal. During the recovery phase, watercress consumption led to a stronger anti-inflammatory downregulation of the pro-inflammatory cytokines IL-6 and TNF-α. In conclusion, we propose that watercress causes a stronger pro-inflammatory response and anti-inflammatory counter-regulation during and after exercise. The clinical relevance of these changes should be verified in future studies.

The present study investigated the changes in total and individual glucosinolates (GSLs) in roots and leaves of different clubroot-resistant and -susceptible oilseed rape cultivars following artificial inoculation with Plasmodiophora brassicae isolates with different virulence. The results showed significant differences in clubroot incidence and severity as well as in the amount of total and individual glucosinolates between oilseed rape cultivars in response to virulence of the pathogen. Single among with total aliphatic and total indolic glucosinolate contents were significantly lower in leaves of susceptible cultivars compared to resistant ones due to the infection. Similarly, single and total aliphatic as well as indolic glucosinolate contents in roots were lower in susceptible cultivars compared to resistant cultivars analyzed. The different isolates of P. brassicae seem to differ in their ability to reduce gluconasturtiin contents in the host. The more aggressive isolate P1 (+) might be able to suppress gluconasturtiin synthesis of the host in a more pronounced manner compared to the isolate P1. A possible interaction of breakdown products of glucobrassicin with the auxin receptor transport inhibitor response 1 (TIR1) is hypothesized and its possible effects on auxin signaling in roots and leaves of resistant and susceptible cultivars is discussed. A potential interplay between aliphatic and indolic glucosinolates that might be involved in water homeostasis in resistant cultivars is explained.

The pharmacological activities in bioactive plant extracts play an increasing role in sustainable resources for valorization and biomedical applications. Bioactive phytochemicals, including natural compounds, secondary metabolites and their derivatives, have attracted significant attention for use in both medicinal products and cosmetic products. Our review highlights the pharmacological mode-of-action and current biomedical applications of key bioactive compounds applied as anti-inflammatory, bactericidal with antibiotics effects, and pain relief purposes in controlled clinical studies or preclinical studies. In this systematic review, the availability of bioactive compounds from several salt-tolerant plant species, mainly focusing on the three promising species Aster tripolium, Crithmum maritimum and Salicornia europaea, are summarized and discussed. All three of them have been widely used in natural folk medicines and are now in the focus for future nutraceutical and pharmacological applications.

The importance of green technologies is steadily growing. Salt-tolerant plants have been proposed as energy crops for cultivation on saline lands. Halophytes such as Salicornia europaea, Tripolium pannonicum, Crithmum maritimum and Chenopodium quinoa, among many other species, can be cultivated in saline lands, in coastal areas or for treating saline wastewater, and the biomass might be used for biogas production as an integrated process of biorefining. However, halophytes have different salt tolerance mechanisms, including compartmentalization of salt in the vacuole, leading to an increase of sodium in the plant tissues. The sodium content of halophytes may have an adverse effect on the anaerobic digestion process, which needs adjustments to achieve stable and efficient conversion of the halophytes into biogas. This review gives an overview of the specificities of halophytes that needs to be accounted for using their biomass as feedstocks for biogas plants in order to expand renewable energy production. First, the different physiological mechanisms of halophytes to grow under saline conditions are described, which lead to the characteristic composition of the halophyte biomass, which may influence the biogas production. Next, possible mechanisms to avoid negative effects on the anaerobic digestion process are described, with an overview of full-scale applications. Taking all these aspects into account, halophyte plants have a great potential for biogas and methane production with yields similar to those produced by other energy crops and the simultaneous benefit of utilization of saline soils.

Reduction in atmospheric sulfur and intensified agriculture have led to sulfur deficiency, often correlated with a higher susceptibility to pathogens. The spread of fungal pathogens, such as the soil-born Verticillium longisporum, was observed. Defense responses of infected plants are linked to sulfur-containing compounds including glucosinolates (GSLs). Some pathogens infect their hosts at specific time periods during the day. To investigate the relation of sulfur-containing metabolites with diurnal effects of infection time points, Brassica napus plants cultivated at two different sulfur supplies, were infected with V. longisporum at four different time points during the day. It was demonstrated that 3, 7 and 14 days after inoculation the infected plants differed in their infection rate depending on the time point of infection. Additionally, infected plants had higher contents of sulfur-containing metabolites, such as specific GSLs, in comparison to non-infected plants. Sufficient sulfur fertilization was always reflected in higher contents of sulfur-containing compounds as well as a lower rate of infection compared to sulfur-deprived plants. On the microscopic level vascular occlusions in the hypocotyl were visible and the amount was dependent on the time point of infection. The results might be used to optimize sulfur fertilization to reduce susceptibility to V. longisporum.

Adverse environmental conditions limit crop yield and better understanding of plant response to stress will assist the development of more tolerant cultivars. Maize and common bean plantlets were evaluated under salinity, high temperature, drought and waterlogged conditions to identify biochemical markers which could be useful for rapid identification of putative stress tolerant plants. The levels of phenolics (free, cell wall-linked, total), aldehydes including malondialdehyde and chlorophylls (a, b, total) were measured on stressed plantlets. Only two indicators were statistically non-significant: chlorophyll b in maize plantlets stressed with sodium chloride and malondialdehyde content in drought stressed maize. The most remarkable effects of abiotic stresses can be summarized as follows: (i) salinity increased levels of free phenolics in maize plantlets and chlorophylls (a, b, total) in common bean; (ii) high temperature (40 °C) elevated levels of chlorophylls (a, b, total) in maize but decreased chlorophylls (a, b, total) and free phenolics in common bean; (iii) drought increased phenolics and decreased chlorophylls (a, b, total) in maize and increased chlorophyll pigments (a, b, total) in common bean; (iv) waterlogging increased free phenolics and decreased chlorophylls (a, b, total) in maize and increased chlorophyll (a, total) in common bean. Free phenolics and chlorophylls, especially a, were the most responsive indicators to stress and can, therefore, be considered putative biochemical markers for abiotic stress tolerance in maize and common bean. The use of Fisher’s linear discriminant analysis to differentiate non-stressed and stressed plants in breeding programs is also a novel aspect of this report. Fisher’s linear discriminant functions classified correctly 100% of non-stressed or stressed originally grouped plants.

Mycotoxins are secondary fungal metabolites, toxic to humans, animals and plants. Among the hundreds of known mycotoxins, aflatoxins, citrinin, patulin, penicillic acid, tenuazonic acid, ochratoxin A, cytochalasins, deoxynivalenol, fumonisins, fusarin C, fusaric acid, and zearalenone are considered the types that most contaminate cereal grain. The majority of the mycotoxins in these groups are produced by three fungal genera: Aspergillus, Penicillium and Fusarium. These metabolites primarily affect the seed quality, germination, viability, seedling vigour, growth of root and cleoptile. Additionally, since the fungi responsible for the production of these mycotoxins are often endophytes that infect and colonize living plant tissues, accumulation of mycotoxins in the plant tissues may at times be associated with development of plant disease symptoms. The presence of mycotoxins, even in the absence of disease symptoms, may still have subtle biological effects on the physiology of plants. Several studies highlight the toxic effects of mycotoxins on animals and cell lines but little is known about the mode of action of most of these metabolites on plant cells. The most important mycotoxins with phytotoxic effects and their producers in addition to their discovery are briefly outlined below and will be addressed in this article.

Plants and animals have evolved various potential mechanisms to surmount the adverse effects of heavy metal toxicity. Plants possess low molecular weight compounds containing sulfhydryl groups (-SH) that actively react with toxic metals. For instance, glutathione (γ-Glu-Cys-Gly) is a sulfur-containing tripeptide thiol and a substrate of cysteine-rich phytochelatins (γ-Glu-Cys)2–11-Gly (PCs). Phytochelatins react with heavy metal ions by glutathione S-transferase in the cytosol and afterwards they are sequestered into the vacuole for degradation. Furthermore, heavy metals induce reactive oxygen species (ROS), which directly or indirectly influence metabolic processes. Reduced glutathione (GSH) attributes as an antioxidant and participates to control ROS during stress. Maintenance of the GSH/GSSG ratio is important for cellular redox balance, which is crucial for the survival of the plants. In this context, sulfurtransferases (Str), also called rhodaneses, comprise a group of enzymes widely distributed in all phyla, paving the way for the transfer of a sulfur atom from suitable sulfur donors to nucleophilic sulfur acceptors, at least in vitro. The best characterized in vitro reaction is the transfer of a sulfane sulfur atom from thiosulfate to cyanide, leading to the formation of sulfite and thiocyanate. Plants as well as other organisms have multi-protein families (MPF) of Str. Despite the presence of Str activities in many living organisms, their physiological role has not been clarified unambiguously. In mammals, these proteins are involved in the elimination of cyanide released from cyanogenic compounds. However, their ubiquity suggests additional physiological functions. Furthermore, it is speculated that a member of the Str family acts as arsenate reductase (AR) and is involved in arsenate detoxification. In summary, the role of Str in detoxification processes is still not well understood but seems to be a major function in the organism.

Sulfurtransferases (Strs) and thioredoxins (Trxs) are members of large protein families. Trxs are disulfide reductases and play an important role in redox‐related cellular processes. They interact with a broad range of proteins. Strs catalyze the transfer of a sulfur atom from a suitable sulfur donor to nucleophilic sulfur acceptors in vitro, but the physiological roles of these enzymes are not well defined. Several studies in different organisms demonstrate protein–protein interactions of Strs with members of the Trx family. We are interested in investigating the specificity of the interaction between Str and Trx isoforms. In order to use the bimolecular fluorescence complementation (BiFC), several Str and Trx sequences from Arabidopsis thaliana were cloned into the pUC‐SPYNE and pUC‐SPYCE split‐YFP vectors, respectively. Each couple of plasmids containing the sequences for the putative interaction partners were transformed into Arabidopsis protoplasts and screened using a confocal laser scanning microscope. Compartment‐ and partner‐specific interactions could be observed in transformed protoplasts. Replacement of cysteine residues in the redox‐active site of Trxs abolished the interaction signal. Therefore, the redox site is not only involved in the redox reaction but also responsible for the interaction with partner proteins. Biochemical assays support a specific interaction among Strs and certain Trxs. Based on the results obtained, the interaction of Strs and Trxs indicates a role of Strs in the maintenance of the cellular redox homeostasis.

There is a great demand of salt-tolerant sugarcane planting material in Cuba. Temporary immersion bioreactors (TIB) are effective to significantly increase sugarcane in vitro shoot proliferation rate from 1:4 in conventional containers to about 1:35. Sugarcane micropropagation in TIBs under NaCl stress may help screen mutants with salinity tolerance. We developed the experiment shown here to identify a NaCl concentration able to stress shoot in TIBs. At 30 days of culture initiation with different NaCl levels (0 - 200 mM), explant multiplication rate, shoot cluster fresh mass, and levels of aldehydes, chlorophylls, carotenoids and phenolics were determined in the plant material. Content of soluble phenolics in the culture medium was also evaluated. Addition of NaCl decreased shoot multiplication rate and fresh mass. Other statistically significant differences were recorded but the most important were noted in the increased contents of carotenoids, malondialdehyde, other aldehydes and soluble phenolics in the plants, and in the soluble phenolics in the culture medium. This research may be useful for future experiments of in vitro selection of new sugarcane genetic materials with NaCl tolerance. Fifty percent of multiplication rate was reduced with 89 mM NaCl which can be used to stress shoots during micropropagation in TIBs and eventually detect mutants with salt tolerance.

It is assumed that salinity enhances the concentration of valuable metabolites in halophytes. The objective was to find a salt concentration and a point in time at which the yield for the valuable metabolites was maximal. Therefore, three different halophyte species were grown under different salinities and harvested over a period from shortly after stress induction up to three weeks. Various reaction patterns were found in the metabolite composition of the analyzed plant material. Halimione portulacoides showed a "short term response", indicated by an increase in all metabolites analyzed after a few hours, whereas Crithmum maritimum showed a "long term response" through accumulation of proline starting after days. Triglochin maritima did not change in metabolite concentration, but like the other plant species the biomass was reduced by salinity. Generally, a higher production in secondary metabolites by higher salinity was outbalanced by a reduction in biomass production. Concentrations of analyzed antioxidants showed a similar reaction and correlated with each other.

Analysis of gene expression is one of the major ways to better understand plant reactions to changes in environmental conditions. The comparison of many different factors influencing plant growth challenges the gene expression analysis for specific gene-targeted experiments, especially with regard to the choice of suitable reference genes. The aim of this study is to compare expression results obtained by Northern blot, semi-quantitative PCR and RT-qPCR, and to identify a reliable set of reference genes for oilseed rape (Brassica napus L.) suitable for comparing gene expression under complex experimental conditions. We investigated the influence of several factors such as sulfur deficiency, different time points during the day, varying light conditions, and their interaction on gene expression in oilseed rape plants. The expression of selected reference genes was indeed influenced under these conditions in different ways. Therefore, a recently developed algorithm, called GrayNorm, was applied to validate a set of reference genes for normalizing results obtained by Northern blot analysis. After careful comparison of the three methods mentioned above, Northern blot analysis seems to be a reliable and cost-effective alternative for gene expression analysis under a complex growth regime. For using this method in a quantitative way a number of references was validated revealing that for our experiment a set of three references provides an appropriate normalization. Semi-quantitative PCR was prone to many handling errors and difficult to control while RT-qPCR was very sensitive to expression fluctuations of the reference genes.

Due to the changing climatic conditions, there is an enlargement of land areas with insufficient rainfall and therefore a reduction in the cultivated area for common crops. Hence, it is now important to find plants that are adapted to these drought conditions. The focus of our research was to apply and compare different methods to quantify the impact of drought stress on plants. Lablab purpureus is considered to be drought tolerant. Therefore, we used L. purpureus genotypes from three continents CPI 36903 (Europe), CPI 52508 (Africa) and HA-4 (Asia) as examples for our study. All genotypes were screened for their tolerance to drought stress by various methods to obtain quantitative data on the drought stress tolerance of individual genotypes and to find out which methods are especially suitable for the measurement of drought tolerance. Classical methods such as leaf size, plant height, biomass, and plant water content were investigated. In addition, by chlorophyll fluorescence measurement effects of drought on the photosynthetic system were examined. Infrared thermography was used in order to make the changes in leaf temperature in plants stressed by drought compared to unstressed plants visible. The methods were complemented by the measurement of leaf conductivity. Results indicate a difference in the usability of the methods for the determination of drought stress. Finally, a set of methods is assembled based on suitability for drought tolerance analysis in plants. The methods include classical growth parameters, including dry weight biomass, PWC and leaf area determination, as well as height measurements of the plants. The stomata behavior is analyzed by leaf conductivity and infrared thermography, both methods complete the set for drought tolerance identification. Based on the results of these methods a ranking of the examined genotypes with respect to their drought tolerance is created.

Of late, decrease in mineral oil supplies has stimulated research on use of biomass as an alternative energy source. Climate change has brought problems such as increased drought and erratic rains. This, together with a rise in land degeneration problems with concomitant loss in soil fertility has inspired the scientific world to look for alternative bio-energy species. Euphorbia tirucalli L., a tree with C3/CAM metabolism in leaves/stem, can be cultivated on marginal, arid land and could be a good alternative source of biofuel. We analyzed a broad variety of E. tirucalli plants collected from different countries for their genetic diversity using AFLP. Physiological responses to induced drought stress were determined in a number of genotypes by monitoring growth parameters and influence on photosynthesis. For future breeding of economically interesting genotypes, rubber content and biogas production were quantified. Cluster analysis shows that the studied genotypes are divided into two groups, African and mostly non-African genotypes. Different genotypes respond significantly different to various levels of water. Malate measurement indicates that there is induction of CAM in leaves following drought stress. Rubber content varies strongly between genotypes. An investigation of the biogas production capacities of six E. tirucalli genotypes reveals biogas yields higher than from rapeseed but lower than maize silage.

The phenotypic features of the Azotobacter vinelandii RhdA mutant MV474 (in which the rhdA gene was deleted) indicated that defects in antioxidant systems in this organism were related to the expression of the tandem-domain rhodanese RhdA. In this work, further insights on the effects of the oxidative imbalance generated by the absence of RhdA (e.g. increased levels of lipid hydroperoxides) are provided. Starting from the evidence that glutathione was depleted in MV474, and using both in silico and in vitro approaches, here we studied the interaction of wild-type RhdA and Cys(230)Ala site-directed RhdA mutant with glutathione species. We found that RhdA was able to bind in vitro reduced glutathione (GSH) and that RhdA-Cys(230) residue was mandatory for the complex formation. RhdA catalyzed glutathione-disulfide formation in the presence of a system generating the glutathione thiyl radical (GS, an oxidized form of GSH), thereby facilitating GSH regeneration. This reaction was negligible when the Cys(230)Ala RhdA mutant was used. The efficiency of RhdA as catalyst in GS-scavenging activity is discussed on the basis of the measured parameters of both interaction with glutathione species and kinetic studies.

Seagrasses, a unique group of submerged flowering plants, profoundly influence the physical, chemical and biological environments of coastal waters through their high primary productivity and nutrient recycling ability. They provide habitat for aquatic life, alter water flow, stabilize the ground and mitigate the impact of nutrient pollution. at the coast region. Although on a global scale seagrasses represent less than 0.1% of the angiosperm taxa, the taxonomical ambiguity in delineating seagrass species is high. Thus, the taxonomy of several genera is unsolved. While seagrasses are capable of performing both, sexual and asexual reproduction, vegetative reproduction is common and sexual progenies are always short lived and epimeral in nature. This makes species differentiation often difficult, especially for non-taxonomists since the flower as a distinct morphological trait is missing. Our goal is to develop a DNA barcoding system assisting also non-taxonomists to identify regional seagrass species. The results will be corroborated by publicly available sequence data. The main focus is on the 14 described seagrass species of India, supplemented with seagrasses from temperate regions. According to the recommendations of the Consortium for the Barcoding of Life (CBOL) rbcL and matK were used in this study. After optimization of the DNA extraction method from preserved seagrass material, the respective sequences were amplified from all species analyzed. Tree- and character-based approaches demonstrate that the rbcL sequence fragment is capable of resolving up to family and genus level. Only matK sequences were reliable in resolving species and partially the ecotype level. Additionally, a plastidic gene spacer was included in the analysis to confirm the identification level. Although the analysis of these three loci solved several nodes, a few complexes remained unsolved, even when constructing a combined tree for all three loci. Our approaches contribute to the understanding of the morphological plasticity of seagrasses versus genetic differentiation.
