Hepatocellular carcinoma (HCC) is one of the most aggressive and refractory cancers due to its high propensity to metastasize and the unavailability of efficacious treatments. Circular RNAs (circRNAs) participate in diverse biological activities in human cancers. Here, we detected the upregulation of a novel circRNA, circZCCHC2 (hsa_circ_0000854), in HCC samples and cells. The upregulation indicated an unfavorable prognosis in HCC patients. CircZCCHC2 accelerated cell growth and metastasis in vitro and tumorigenicity in vivo. Mechanistic investigations revealed that circZCCHC2 regulated BTBD7 expression by sponging miR-936. Moreover, the suppression of malignancy caused by circZCCHC2 knockdown could be sufficiently reversed by miR-936 inhibition. Additionally, the suppressed Rho/ROCK2 pathway conferred by circZCCHC2 knockdown could be restored by inhibiting miR-936 expression. Collectively, our findings reveal that circZCCHC2 plays an oncogenic role of in HCC progression by modulating the miR-936/BTBD7/Rho/ROCK2 pathway.

Background PEBP (phosphatidyl ethanolamine-binding protein) is widely found in eukaryotes including plants, animals and microorganisms. In plants, the PEBP family plays vital roles in regulating flowering time and morphogenesis and is highly associated to agronomic traits and yields of crops, which has been identified and characterized in many plant species but not well studied in Tartary buckwheat (Fagopyrum tataricum Gaertn.), an important coarse food grain with medicinal value. Methods Genome-wide analysis of FtPEBP gene family members in Tartary buckwheat was performed using bioinformatic tools. Subcellular localization analysis was performed by confocal microscopy. The expression levels of these genes in leaf and inflorescence samples were analyzed using qRT-PCR. Results Fourteen Fagopyrum tataricum PEBP (FtPEBP) genes were identified and divided into three sub-clades according to their phylogenetic relationships. Subcellular localization analysis of the FtPEBP proteins in tobacco leaves indicated that FT- and TFL-GFP fusion proteins were localized in both the nucleus and cytoplasm. Gene structure analysis showed that most FtPEBP genes contain four exons and three introns. FtPEBP genes are unevenly distributed in Tartary buckwheat chromosomes. Three tandem repeats were found among FtFT5/FtFT6, FtMFT1/FtMFT2 and FtTFL4/FtTFL5. Five orthologous gene pairs were detected between F. tataricum and F. esculentum. Seven light-responsive, nine hormone-related and four stress-responsive elements were detected in FtPEBPs promoters. We used real-time PCR to investigate the expression levels of FtPEBPs among two flowering-type cultivars at floral transition time. We found FtFT1/FtFT3 were highly expressed in leaf and young inflorescence of early-flowering type, whereas they were expressed at very low levels in late-flowering type cultivars. Thus, we deduced that FtFT1/FtFT3 may be positive regulators for flowering and yield of Tartary buckwheat. These results lay an important foundation for further studies on the functions of FtPEBP genes which may be utilized for yield improvement.

Abstract Plant disease is a threat to global food security. Breeding crops carrying broad-spectrum resistance loci is an effective way to control infectious disease. Disease-resistant mutants are valuable resources for deciphering the underlying mechanisms of plant immunity and could provide genetic loci to generate disease-resistant crops. Here, we identified a rice mutant, rbr7 (rice blast resistance 7), that confers resistance against different strains of Magnaporthe oryzae. Disease-mimicking necrotic lesions started to appear on the leaves of rbr7 four weeks after sowing. Histochemical analysis revealed reactive oxygen species accumulation and cell death accompanied by spontaneous lesion formation in rbr7. Map-based cloning and bulk segregation analysis showed a 2855 bp fragment deletion on chromosome 5, leading to the disruption of the LOC_Os05g28480-coding protein. Transgenic rbr7 complementation plants showed compromised resistance to rice blast, indicating that LOC_Os05g28480, or Rbr7, regulates the rice immune response. Rbr7 encodes a small protein of unknown function with 85 amino acids. Transcriptomic analysis revealed that disruption of RBR7 led to the upregulation of genes responding to salicylic acid, systemic acquired resistance and pathogenesis-related genes. Taken together, our findings reveal insights into a novel small protein involved in regulating plant resistance to rice blast and provide a potential target for crop breeding.

The vesicle trafficking apparatus is a fundamental machinery to maintain the homeostasis of membrane-enclosed organelles in eukaryotic cells. Thus, it is broadly conserved in eukaryotes including plants. Intensive studies in the model organisms have produced a comprehensive picture of vesicle trafficking in yeast and human. However, with respect to the vesicle trafficking of plants including rice, our understanding of the components and their coordinated regulation is very limited. At present, several vesicle trafficking apparatus components and cargo proteins have been identified and characterized in rice, but there still remain large unknowns concerning the organization and function of the rice vesicle trafficking system. In this review, we outline the main vesicle trafficking pathways of rice based on knowledge obtained in model organisms, and summarize current advances of rice vesicle trafficking. We also propose to develop methodologies applicable to rice and even other crops for further exploring the mysteries of vesicle trafficking in plants.

The process of cold chain of fruits and vegetables can easily cause damage to the internal structure of fruits and vegetables and arouse quality deterioration due to problems such as extensive control and large losses. Meanwhile, the moisture and heat inside the fruits and vegetables would dynamically migrate, which is not a beneficial effect of the storage and sales on fruits and vegetables. Therefore, it is an urgent problem for studying multi-scale heat and mass transfer mechanism in the cold chain process of fruits and vegetables. This paper clarifies the numerical imaging technology for multi-scale modeling of fruits and vegetables, discusses the multi-scale computational fluid dynamics modeling method of heat and mass transfer mechanism in the cold chain process of fruits and vegetables, as well as the basic problems and solutions in modeling, and optimizes the heat and mass transfer models and designs more efficient and reasonable cooling systems, in order to provide a theoretical basis for the study of the multi-scale heat and mass transfer mechanism, loss reduction and preservation during the cold chain process of fruits and vegetables.

Underwater images are frequently subject to color distortion and loss of details. However, previous enhancement methods did not tackle these mixed degradations by dividing them into sub-problems that could be effectively addressed. Moreover, the parameters and computations required for these methods are usually costly for underwater equipment, which has limited power supply, processing capabilities, and memory capacity. To address these challenges, this work proposes a Lightweight Parallel Strategy Network (LPS-Net). Firstly, a Dual-Attention Enhancement Block and a Mirror Large Receptiveness Block are introduced to, respectively, enhance the color and restore details in degraded images. Secondly, we employed these blocks on parallel branches at each stage of LPS-Net, with the goal of achieving effective image color and detail rendering simultaneously. Thirdly, a Gated Fusion Unit is proposed to merge features from different branches at each stage. Finally, the network utilizes four stages of parallel enhancement, achieving a balanced trade-off between performance and parameters. Extensive experiments demonstrated that LPS-Net achieves optimal color enhancement and superior detail restoration in terms of visual quality. Furthermore, it attains state-of-the-art underwater image enhancement performance on the evaluation metrics, while using only 80.12 k parameters.

Particulate nitrate (pNO3) is now becoming the principal component of PM2.5 during severe winter haze episodes in many cities of China. To gain a comprehensive understanding of the key factors controlling pNO3 formation and driving its trends, we reviewed the recent pNO3 modeling studies which mainly focused on the formation mechanism and recent trends of pNO3 as well as its responses to emission controls in China. The results indicate that although recent chemical transport models (CTMs) can reasonably capture the spatial–temporal variations of pNO3, model-observation biases still exist due to large uncertainties in the parameterization of dinitrogen pentoxide (N2O5) uptake and ammonia (NH3) emissions, insufficient heterogeneous reaction mechanism, and the predicted low sulfate concentrations in current CTMs. The heterogeneous hydrolysis of N2O5 dominates nocturnal pNO3 formation, however, the contribution to total pNO3 varies among studies, ranging from 21.0% to 51.6%. Moreover, the continuously increasing PM2.5 pNO3 fraction in recent years is mainly due to the decreased sulfur dioxide emissions, the enhanced atmospheric oxidation capacity (AOC), and the weakened nitrate deposition. Reducing NH3 emissions is found to be the most effective control strategy for mitigating pNO3 pollution in China. This review suggests that more field measurements are needed to constrain the parameterization of heterogeneous N2O5 and nitrogen dioxide (NO2) uptake. Future studies are also needed to quantify the relationships of pNO3 to AOC, O3, NOx, and volatile organic compounds (VOCs) in different regions of China under different meteorological conditions. Research on multiple-pollutant control strategies involving NH3, NOX, and VOCs is required to mitigate pNO3 pollution, especially during severe winter haze events.

Global food security is threatened by rice blast disease caused by the filamentous fungus Magnaporthe oryzae. An understanding of rice resistance mechanisms is fundamental to developing strategies for disease control. In this review, we summarize recent advances in pathogen-associated molecular pattern-triggered immunity, effector-triggered immunity, defense regulator-mediated immunity, and effects of nutrient elements on rice blast resistance. We outline strategies used for breeding rice cultivars with improved disease resistance. We also present the major research challenges for rice blast disease resistance and propose approaches for future investigation.

Chlorophyll–a (Chl–a) concentration is an indicator of phytoplankton pigment, which is associated with the health of marine ecosystems. A commonly used method for the determination of Chl–a is satellite remote sensing. However, due to cloud cover, sun glint and other issues, remote sensing data for Chl–a are always missing in large areas. We reconstructed the Chl–a data from MODIS and VIIRS in the Arabian Sea within the geographical range of 12–28° N and 56–76° E from 2020 to 2021 by combining the Data Interpolating Convolutional Auto–Encoder (DINCAE) and the Bayesian Maximum Entropy (BME) methods, which we named the DINCAE–BME framework. The hold–out validation method was used to assess the DINCAE–BME method’s performance. The root–mean–square–error (RMSE) and the mean–absolute–error (MAE) values for the hold–out cross–validation result obtained by the DINCAE–BME were 1.8824 mg m−3 and 0.4682 mg m−3, respectively; compared with in situ Chl–a data, the RMSE and MAE values for the DINCAE–BME–generated Chl–a product were 0.6196 mg m−3 and 0.3461 mg m−3, respectively. Moreover, DINCAE–BME exhibited better performance than the DINEOF and DINCAE methods. The spatial distribution of the Chl–a product showed that Chl–a values in the coastal region were the highest and the Chl–a values in the deep–sea regions were stable, while the Chl–a values in February and March were higher than in other months. Lastly, this study demonstrated the feasibility of combining the BME method and DINCAE.

The development trend of high shares of renewables and power electronics has increased the demand for new energy converters in the power system, but there is a lack of systematic research on the stability of different types of converters when transmitting power, which is worth exploring in depth. In this study, the power transfer capabilities of grid-forming and grid-following converters are investigated separately through an equivalent circuit diagram and phasor diagram when connected to the grid, and a quantitative relationship between converters’ power transmission limit and short circuit ratio under static stability conditions is obtained, leading to the conclusion that, in terms of power transmission, grid-forming converters are more suitable for weak grids with high damping and low inertia, whereas grid-following converters are more suitable for strong grids with high inertia. The conclusions are further verified by constructing the converter grid-connected models for different grid strengths through the PLECS simulation platform and the real-time simulation RTBOX1 and F28379D launchpad platform.

Patients with pancreatic walled-off necrosis (WON) treated by minimally invasive approaches often require repeated necrosectomy instead of conventional open necrosectomy. A 55-year-old woman with lateral infected pancreatic WON was successfully treated by an infrequently used technique involving an ultrasonic pneumatic lithotripsy system (UPLS) and double-catheter aspirated lavage through a percutaneous drainage tract. No perioperative complications occurred. The mean operating time of the repeated procedures was 35 minutes (range, 25–48 minutes). The total hospitalization stay was 46 days, and no recurrence of the fluid collection was observed during the 15-month follow-up period. UPLS-assisted necrosectomy and double-catheter aspirated lavage is an alternative technique for lateral WON that fails to improve by percutaneous drainage. It efficiently combines disintegration and aspiration procedures and can be safely repeated under total intravenous anesthesia through sinus tracts.

The coal industry contributes significantly to the social economy, but the emission of greenhouse gases puts huge pressure on the environment in the process of mining, transportation, and power generation. In the integrated energy system (IES), the current research about the power-to-gas (P2G) technology mainly focuses on the injection of hydrogen generated from renewable energy electrolyzed water into natural gas pipelines, which may cause hydrogen embrittlement of the pipeline and cannot be repaired. In this paper, sufficient hydrogen energy can be produced through P2G technology and coal-to-hydrogen (C2H) of coal gasification, considering the typical scenario of coal district is rich in coal and renewable energy. In order to transport the mined coal to the destination, hydrogen heavy trucks have a broad space for development, which can absorb hydrogen energy in time and avoid potentially dangerous hydrogen injection into pipelines and relatively expensive hydrogen storage. An optimized scheduling model of electric-gas IES is proposed based on second-order cone programming (SOCP). In the model proposed above, the closed industrial loop (including coal mining, hydrogen production, truck transportation of coal, and integrated energy systems) has been innovatively studied, to consume renewable energy and coordinate multi-energy. Finally, an electric-gas IES study case constructed by IEEE 30-node power system and Belgium 24-node natural gas network was used to analyze. The results show that by introducing the proposed hydrogen production technology, typical daily operating costs are effectively reduced by 7.7%. Under China’s carbon emissions trading system, the operating costs of hydrogen heavy trucks have been reduced by 0.95 and 4.68% respectively, compared with electric vehicles and diesel trucks. Under Europe’s stricter carbon emissions trading system, the percentages of cost reduction are 2.56 and 9.12%, respectively. The above technical results verify the feasibility, economy, low carbon, and effectiveness of the proposed mechanism.

Rice grain yield consists of several key components, including tiller number, grain number per panicle (GNP), and grain weight. Among them, GNP is mainly determined by panicle branches and spikelet formation. In this study, we identified a gene affecting GNP and grain yield, OsSPL9, which encodes SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE (SPL) family proteins. The mutation of OsSPL9 significantly reduced secondary branches and GNP. OsSPL9 was highly expressed in the early developing young panicles, consistent with its function of regulating panicle development. By combining expression analysis and dual-luciferase assays, we further confirmed that OsSPL9 directly activates the expression of RCN1 (rice TERMINAL FLOWER 1/CENTRORADIALIS homolog) in the early developing young panicle to regulate the panicle branches and GNP. Haplotype analysis showed that Hap3 and Hap4 of OsSPL9 might be favorable haplotypes contributing to high GNP in rice. These results provide new insights on high grain number breeding in rice.

Patients with acute pancreatitis (AP) often suffer tough complications, some of which are fatal. The early diagnosis and definite treatment of central nervous system (CNS) complications have not been fully achieved yet, which seriously affects the mortality of severe acute pancreatitis (SAP). We present a case of infected pancreatic necrosis (IPN) in a 62-year Chinese man who developed acute herpes simplex encephalitis (HSE) caused by herpes simplex virus type 1 (HSV-1) after favorable minimally invasive retroperitoneal approaches (MIRAs). The patient was successfully treated with 115 days stayed in our hospital. The MIRAs included image-guided retroperitoneal percutaneous catheter drainage (PCD), nephroscopic pancreatic necrosectomy (NPN), and ultrasonic pneumatic lithotripsy system (UPLS) assisted non-narcotic sinus track necrosectomy (NSN). HSE is relatively rare and potentially life threatening. We attempt to discuss the probable risk factors and how the relatively rare HSE are related to the patients of SAP with latent HSV.

With the development of wireless communication systems, it is particularly essential to maximize the quality of experience (QoE) of machine-to-machine (M2M) communication. In this paper, we propose a new QoE-oriented uplink rate control and resource allocation scheme for the Internet of Things (IoT) network, by introducing an evaluation model based on mean opinion score (MOS) for different machine-type communication (MTC) devices. The existing works are only dedicated to solving the short-term resource allocation problems by considering the current transmission time slots, which cannot handle long-standing problems. To this end, based on the recently developed Lyapunov optimization, we convert the original long-term optimization problem into the admission rate control subproblem and the resource allocation subproblem in each time slot. To solve the joint power optimization and sub-channel selection subproblems, Gale-Shapley algorithm is utilized to formulate it as a two-dimensional matching problem, and the preference lists are established by the transmission rate and signal to interference plus noise ratio (SINR). In the proposed algorithms, a priority mechanism is employed to ensure fairness. The simulation results demonstrate that without prior knowledge of the data arrivals and sub-channel statistics, the proposed algorithms can significantly improve the overall perceived quality from the users' perspective.

Background/Aims: The expression of PRAME and its role in hepatocellular carcinoma (HCC) remain unknown. The aim of this study was to examine the functional role of PRAME in HCC development and exploring the molecular mechanism. Methods: We first detected PRAME expression in 96 human HCC tissue samples and correlated with clinicopathological characteristics and prognosis of the patients. We then established stable HCC cell lines with PRAME overexpression and knockdown followed by functional analysis in vitro. Further, we examined the relationship between PRAME and p53 pathway in vitro by using Western blotting. Finally, PRAME expression was detected to evaluate its correlation with p-p53 and p53 pathway related apoptotic proteins in xenograft tumor mouse model using immunohistochemistry. Results: PRAME expression was significantly higher in HCC tissues than in adjacent non-tumor tissues and their expression was positively correlated with alpha fetoprotein levels and tumor size. In addition, PRAME expression was associated with AJCC stage and is a potential biomarker of poor prognosis regarding 5-year overall survival in HCC. In vitro studies, we found that PRAME expression was higher in HCC cell lines than in normal hepatic cell line. Inhibited cell proliferation and increased cell apoptosis was observed in PRAME knockdown HCC cells. Futher, increased cell apoptosis was correlated with the proportion of cells in G0/G1 stage, activated p53 mediated apoptosis, and increased cyclin p21 expression. Xenograft analysis in nude mice also found that PRAME knockdown inhibited tumorigenesis while PRAME overexpression had opposite effect. Conclusions: In HCC, PRAME serves as a potential biomarker for poor prognosis and novel therapeutic target in treating this cancer. PRAME is a potential biomarker of poor prognosis in HCC. PRAME surpresses HCC cell death in vitro and in vivo by regulating p53 apoptotic signaling and may serve as a potential therapeutic target in HCC.

Multiple sclerosis (MS) is an autoimmune disease characterized by demyelination, axonal damage and progressive neurologic dysfunction in central nervous system (CNS). Many evidences show that B cells play an important role in the pathogenesis of MS. Follicular helper T cells (Tfh) secrete IL-21 to prompt the proliferation and differentiation of B cells in germinal center (GC) through clonal proliferation, somatic hypermutation, antibody class switching, antibody affinity maturation process. AG490 is a synthetic inhibitor to JAK-STAT signal pathway, which has been studied in inflammatory, tumor and autoimmune diseases. In the present study, the experimental mice were divided into 3 groups, vehicle group and AG490 group were given MOG35-55 to induce EAE model, from the third day after immunization, the mice were given vehicle or AG490 by intraperitoneal injection every other day. All mice were assessed clinical scores after immunization. On twentieth day, all mice were sacrificed, HE staining and solochrome cyanine staining were performed to evaluate inflammatory cells infiltration and demyelination, spleen sections were stained with PNA-FITC to analyze the difference in germinal center. Compared with vehicle group, the incidence of AG490 group was deceased, onset time was delayed, the severity was significantly reduced. The inflammatory cells and demyelination in AG490 group were lower than those in vehicle group. Immunofluorescence showed the fluorescence intensity of AG490 group was significantly lower than in the vehicle group, but higher than that of control group.

The rapid development of location-based social networks (LBSNs) produces the increasing number of check-in records and corresponding heterogeneous information which bring big challenges of points-of-interest (POIs) recommendation in our daily lives. The emergence of various recommender techniques bridges the gap between the numerous heterogeneous check-ins and the personalized POI recommendation. However, due to the differences between LBSNs and conventional recommendation tasks, besides the user feedback, the spatio-temporal information is also significant to precisely capture the user preferences. In this paper, we propose a multi-task learning model based POI recommender system which exploits a structure of generative adversarial networks (GAN) simultaneously considering temporal check-ins and geographical locations. The GAN-based model is capable of relieving the sparsity of check-in data in POI recommender systems. The temporal check-ins not only present the preference but also show the lifestyle of an individual while the geographical locations describe the active region of users which further filters POIs far from the feasible region. The multi-task learning strategy is capable of combining the information of temporal check-ins and geographical locations to improve the performance of personalized POI recommendation. We conduct the experiments on two real-world LBSNs datasets and the experimental results show the effectiveness of our proposed approach.

The course of myasthenia gravis (MG) is complicated by increased reports of cognitive defects in both human and animal models, which suggests potential central nervous system (CNS) damage. We conducted a systematic review of the relationships between MG and cognitive function. This systematic review followed Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Major databases were searched to examine the neuropsychological studies of adults with MG. Weighted effect sizes were pooled by cognitive domain. Eight studies representing 300 subjects were included. Eight cognitive domain categories were identified: (i) Mini-Mental State Examination (MMSE), (ii) language, (iii) processing speed, (iv) verbal learning and memory, (v) visual learning and memory, (vi) attention span, (vii) response fluency, and (viii) motor performance. Nine (cognitive domain categories, MMSE, language, processing speed, verbal learning and memory (except for delayed recall memory), and motor performance) of 16 cognitive tasks revealed significant moderate effect sizes. Verbal logical-delayed memory, finger tapping with the preferred hand, and the Symbol Digit Modalities Test showed a greater magnitude relationship to cognitive function than did other specific cognitive domains. Verbal learning and memory seems to be the most significant affected according to cognitive domain categories. For MG, the ability of attention, response fluency, visual learning, and memory seems to be reserved. The MG patients seem to perform significantly worse than the non-MG controls in a range of cognitive domains. Our findings should be interpreted with caution because of the clinical and methodological heterogeneity of included studies.

The brassinosteroid-SIGNALING KINASE (BSK) belongs to the receptor-like cytoplasmic kinase XII subgroup. BSK1 regulates development and immunity in Arabidopsis. However, the function of rice (Oryza sativa) BSK1 is largely unknown. Here, we report that the expression level of OsBSK1-2 is induced after a chitin or fagellin22 (flg22) treatment. Silencing OsBSK1-2 in rice results in compromised responses to chitin- or flg22-triggered immunity and resistance to Magnaporthe oryzae, but does not alter the plant’s architecture nor reduce plant responses to brassinosteroid signaling. Our study reveals that OsBSK1-2 functions as a major regulator in rice plant immunity.

Previous studies have shown that multivesicular bodies (MVBs)/endosomes-mediated vesicular trafficking may play key roles in plant immunity and cell death. However, the molecular regulation is poorly understood in rice. Here we report the identification and characterization of a MVBs-localized AAA ATPase LRD6-6 in rice. Disruption of LRD6-6 leads to enhanced immunity and cell death in rice. The ATPase activity and homo-dimerization of LRD6-6 is essential for its regulation on plant immunity and cell death. An ATPase inactive mutation (LRD6-6E315Q) leads to dominant-negative inhibition in plants. The LRD6-6 protein co-localizes with the MVBs marker protein RabF1/ARA6 and interacts with ESCRT-III components OsSNF7 and OsVPS2. Further analysis reveals that LRD6-6 is required for MVBs-mediated vesicular trafficking and inhibits the biosynthesis of antimicrobial compounds. Collectively, our study shows that the AAA ATPase LRD6-6 inhibits plant immunity and cell death most likely through modulating MVBs-mediated vesicular trafficking in rice.

To protect themselves from pathogens, plants have developed an effective innate immune system. Plants recognize pathogens and then rapidly alter signaling pathways within individual cells in order to achieve an appropriate immune response, including the generation of reactive oxygen species, callose deposition, and transcriptional reprogramming. Post-translational modifications (PTMs) are versatile regulatory changes critical for plant immune response processes. Significantly, PTMs are involved in the crosstalk that serves as a fine-tuning mechanism to adjust cellular responses to pathogen infection. Here, we provide an overview of PTMs that mediate defense signaling perception, signal transduction in host cells, and downstream signal activation.

Objective: Rare coexistence of disease or pathology. Background: Multiple sclerosis (MS) and neuromyelitis optica spectrum disorders (NMOSDs) are 2 similar but distinct diseases. These diseases were difficult to distinguish from each other until aquaporin-4-IgG (AQP-4-IgG) was discovered. The accurate identification of these 2 diseases is crucial for appropriate drug treatment in clinical practice. Herein, we report a case of AQP-4-IgG seroconversion with magnetic resonance imaging (MRI) findings suggestive of MS. Case Report: A 54-year-old woman developed weakness in her right lower extremity that gradually returned to normal 4 years ago. Recently, she was admitted to the hospital for numbness and weakness of both lower limbs and the right upper limb for more than 10 days. The clinical and MRI features of the patient suggested a high susceptibility for misdiagnosis of MS. However, careful observation of the MRI revealed the presence of atypical MS lesions ("red flag" signs), indicating the possibility of other diagnoses in this patient. After further examination, serum AQP-4-IgG was detected, suggesting the potential presence of another disorder, NMOSD, in the patient. Conclusions: Attention should be given to the identification of MS MRI "red flag" signs. Even for patients with a high suspicion of MS, it is necessary to conduct antibody tests for AQP-4-IgG, MOG-IgG and other relevant markers to screen for associated diseases because MS disease-modifying therapy approaches may lead to a deterioration in the state of NMOSD patients. Abstract Early-life human gut microbiome is a pivotal driver of gut homeostasis and infant health. However, the viral component (known as “virome”) remains mostly unexplored. Here, we establish the Early-Life Gut Virome (ELGV), a catalog of 160,478 non-redundant DNA and RNA viral sequences from 8130 gut virus-like particles (VLPs) enriched or bulk metagenomes in the first three years of life. By clustering, 82,141 viral species are identified, 68.3% of which are absent in existing databases built mainly from adults, and 64 and 8 viral species based on VLPs-enriched and bulk metagenomes, respectively, exhibit potentials as biomarkers to distinguish infants from adults. With the largest longitudinal population of infants profiled by either VLPs-enriched or bulk metagenomic sequencing, we track the inherent instability and temporal development of the early-life human gut virome, and identify differential viruses associated with multiple clinical factors. The mother-infant shared virome and interactions between gut virome and bacteriome early in life are further expanded. Together, the ELGV catalog provides the most comprehensive and complete metagenomic blueprint of the early-life human gut virome, facilitating the discovery of pediatric disease-virome associations in future.
