Anionic dyes are one of the most serious contaminants in water as these molecules are known to be toxic to many living organisms. Herein, we report the development of functionalized polyvinylidene fluoride membranes modified with chitosan-coated iron oxide nanomaterials (Fe-PVDF) for the efficient treatment of anionic dye-contaminated water. Aqueous solutions of anionic dyes could be captured rapidly by passing through the functionalized membrane under reduced pressure. Under neutral conditions, Fe-PVDF showed a maximum removal capacity of 74.6 mg/g for Evans blue (EB) through the adsorption process. In addition, the adsorption capacity was significantly enhanced up to 434.78 mg/g under acidic conditions. The adsorption process for EB matched well with the Langmuir model, indicating monolayer adsorption of the dye to the membrane surface. Moreover, Fe-PVDF can be reusable by a simple washing step in an alkaline solution, and thus, the composite membrane was applied several times without a significant decrease in its adsorption performance. The same composite membrane was further applied to the removal of five other different anionic dyes with high efficiencies. The adsorption mechanism can be explained by the electrostatic interaction between the positively charged chitosan and the negatively charged dye as well as the affinity of the sulfate groups in dye molecules for the surface of the iron oxide nanoparticles. The easy preparation and rapid decolorization procedures make this composite membrane suitable for efficient water treatment.

Edible bird's nest (EBN) is a nutritious food with many beneficial effects, including protecting cells against oxidation and infection due to wounds, bacteria, or viruses. EBN has shown anti-aging, anti-inflammatory, and wound-healing properties in skin cells. Here, we investigated whether EBN has protective effects against photoaging, inflammation, and immunosenescence in hairless mice treated with UVB irradiation. The skin thickness was lower in mice on an EBN diet than in mice treated with UVB alone. The level of hydration was significantly increased, while the index of transepidermal water loss decreased, in groups on the EBN diet. EBN also reduced erythema index in UVB-irradiated mice. At the molecular level, skin elasticity and anti-aging are associated with high expression of elastin, collagen, and filaggrin and low expression of the membrane metalloproteinases, MMP-1 and MMP-2. Inflammatory markers such as interleukins, IL-1β and IL-6, and TNF- decreased significantly in the EBN groups. Caspase-3, an important factor in the apoptotic pathway and in congenital and adaptive immune responses, decreased in the EBN groups. Moreover, EBN diminished the overexpression of immunoglobulin E and elevated mast cell counts in UVB-irradiated mice. Overall, these findings suggest that EBN protects skin against aging and alleviates inflammation in UVB-irradiated hairless mice.

Bacterial exopolysaccharides (EPSs) are important alternatives to plant polysaccharides in fermented products and exhibit antioxidant activity, which is particularly desirable for functional foods. This study evaluated the use of spent media wastewater (SMW) derived from kimchi fermentation for the production of an EPS and analyzed the characterization and antioxidant activity of the resulting EPS. The EPS concentration and conversion yields of sequential purification were 7.7-9.0 g/L and 38.6-45.1%, respectively. Fourier transform infrared spectra and NMR spectra indicated that the EPS was a linear glucan with α-(1 → 6) linkages. The EPS also exhibited thermal tolerance to high temperatures. In vitro antioxidant activity analyses indicated the scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, thiobarbituric acid reactance (TBAR), and ferric ion reducing antioxidant power (FRAP) values of 71.6-79.1, 28.2-33.0%, and 0.04-0.05 mM FeCl3, respectively. These results reveal that the EPS extracted from SMW has potential as a thermally tolerant, nontoxic, and natural antioxidant for industrial applications.

Although classical metabolic engineering strategies have succeeded in developing microbial strains capable of producing desired bioproducts, metabolic imbalance resulting from extensive genetic manipulation often leads to decreased productivity. Thus, abiotic strategies for improving microbial production performance can be an alternative to overcome drawbacks arising from intensive metabolic engineering. Herein, we report a promising abiotic method for enhancing lycopene production by UV-C irradiation using a radiation-resistant ΔcrtLm/crtB+dxs+ Deinococcus radiodurans R1 strain. First, the onset of UV irradiation was determined through analysis of the expression of 11 genes mainly involved in the carotenoid biosynthetic pathway in the ΔcrtLm/crtB+dxs+ D. radiodurans R1 strain. Second, the effects of different UV wavelengths (UV-A, UV-B, and UV-C) on lycopene production were investigated. UV-C irradiation induced the highest production, resulting in a 69.9% increase in lycopene content [64.2 ± 3.2 mg/g dry cell weight (DCW)]. Extended UV-C irradiation further enhanced lycopene content up to 73.9 ± 2.3 mg/g DCW, a 95.5% increase compared to production without UV-C irradiation (37.8 ± 0.7 mg/g DCW).

According to the previously described anti-photoaging effect of the enzyme-processed Panax ginseng extract and Gastrodia elata extract, we hypothesized that the combination of the two extracts would have superior effect to protect human skin from UVB radiation. Besides, the mixture of active components isolated from herbal extracts, ginsenoside F2, and α-gastrodin was investigated on the photo-protective capability. The expression of aging-related markers including matrix metalloproteinase-1 (MMP-1), interleukin-6 (IL-6), and procollagen type 1 was evaluated using ELISA kits. It was reported that the herbal extract at a Panax ginseng extract to Gastrodia elata extract ratio of 1:10 (w/w) and the compound mixture with equal proportion of ginsenoside F2 and α-gastrodin exhibited significant inhibition of MMP-1 and IL-6 production, and marked upregulation of procollagen type 1 formation. Thus, the combination of either the enzyme-processed herbal extracts or their active components would enhance the properties of prevention and treatment of UVB-induced skin damage.

Pine wilt disease, caused by Bursaphelenchus xylophilus (pine wood nematode), leads to severe environmental and economic damage. Here, we report the results of experiments on the biological control of pine wilt disease through termination of the insect vector of the nematode and the mechanism of the insecticidal action of Metarhizium anisopliae JEF-279 against Monochamus alternatus (Japanese pine sawyer). A combined treatment with a fungal conidia suspension and a fungal protease-containing culture filtrate caused 75.8% mortality of the insect vector. Additionally, the presence of destruxins was confirmed in the dead Japanese pine sawyer adults, and half of the 10 protein spots in proteomic analysis were identified as an actin related to muscle contraction. Based on proteomic and microscopic analyses, the infection cycle of the Japanese pine sawyer by M. anisopliae JEF-279 was inferred to proceed in the following sequence: (1) host adhesion and germination, (2) epicuticle degradation, (3) growth as blastospore, (4) killing by various fungal toxins (insecticidal metabolites), (5) immune response as defense mechanism, and (6) hyphal extrusion and conidiation. Consequently, the combined fungal conidia suspension and protease-containing culture filtrate treatment may be applied as an insecticidal agent, and flaccid paralysis is likely a major mechanism underlying the insecticidal action of M. anisopliae JEF-279 on host insects.

The biorefining of agricultural waste into green chemicals has clear potential for improving global environmental sustainability. In this study, we evaluated the potential of acetic acid production from carbohydrate feedstock (onion waste, OW) as a more environmentally friendly source than feedstock produced from natural gas. In particular, OW is an ideal feedstock for the biorefining process as it contains a sufficient amount of carbohydrates (69.7%). Five days of the simultaneous saccharification and two-step fermentation (SSTF) process produced acetic acid from OW more efficiently than the simultaneous saccharification and cofermentation (SSCF) process. SSTF produced 19.3 g/L acetic acid and recorded the highest conversion yield (90.5%) from OW (6% substrate loading, w/v). These results suggested that acetic acid can be efficiently and sustainably produced from OW by the SSTF process.

A Gram-stain-negative, facultatively anaerobic, non-motile and rod-shaped bacterial strain, designated THG-SD5.5T, was isolated from a soil sample collected in a tangerine field, Republic of Korea. According to the 16S rRNA gene sequence comparisons, the isolate was identified as a member of the genus Chitinophaga and to be closely related to Chitinophaga ginsengihumi KACC 17604T (97.9%) and Chitinophaga rupis KACC 14521T (97.5%). The 16S rRNA gene sequence similarities with other species of the genus Chitinophaga were in the range 92.8–95.5%. Catalase test was positive. Oxidase test was negative. The DNA G + C content was determined to be 46.1 mol%. DNA-DNA hybridization values between strain THG-SD5.5T and C. ginsengihumi KACC 17604T and C. rupis KACC 14521T were 45.1% and 15.6%, respectively. Strain THG-SD5.5T was also found to be able to grow at 24–33 °C, at 0–5% NaCl and at pH 5.5–9.0. The major fatty acids were identified as anteiso-C15:0, C16:0, anteiso-C17:0 and C18:0. The dominant respiratory quinone was identified as menaquinone-7 (MK-7). The polar lipids were found to be diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, an unidentified aminolipid, two unidentified phospholipids and three unidentified lipids. Based on these phenotypic, genotypic and phylogenetic characterisations, strain THG-SD5.5T (= KACC 19338T = CGMCC 1.16304T) is concluded to represent a novel species of the genus Chitinophaga, for which the name Chitinophaga aurantiaca sp. nov. is proposed.

As a robust radioanalytical method for tracking carbonaceous particulates in vivo, polycyclic aromatic hydrocarbons from diesel exhaust were labeled with a radioactive-iodine-tagged pyrene analogue. Single-photon emission computed tomography and biodistribution studies showed high uptake and slow clearance of this matter in the respiratory system, which may underlie its severe toxicity.

Organ-on-a-chip has the potential to replace preclinical trials which have been problematic for decades due to unaffordable cost and time. The performance of in vitro tumor-on-a-chip depends on how accurately the system represents analogous tumor-microenvironment (TME) and TME associated phenomena. In this study, we have focused on angiogenesis, one of the most significant features of TME for tumor growth and metastasis. Angiogenesis in TME is triggered through cascaded interactions among TME associated neighboring cells including immune cells, tumor cells, and fibroblast cells [1]. Therefore, temporally-controlled TME-on-a-chip is desired for an accurate representation of angiogenesis. However, conventional microfluidic devices cannot temporarily manipulate the condition of interacting cells and secreted signal molecules. Here, we proposed a hydrogel-based variable TME-on-a-chip with diffusion switch channels. The channels between hydrogel walls enable temporal diffusion control by controlling inflow. The diffusion control was observed in diffusion experiment with a fluorescent dye. Furthermore, experiment of HUVEC’s migration toward diffused VEGF also confirmed that TME-on-a-chip is capable of reproducing an angiogenic switch triggering through temporal diffusion control. Due to a simple fabrication procedure, the design of the microfluidic device can be easily modified to represent more complex variable TME models.

Cohen syndrome (CS), a rare autosomal recessive disorder, has been associated with genetic mutations in the VPS13B gene, which regulates vesicle-mediated protein sorting and transport. However, the cellular mechanism underlying CS pathogenesis in patient-derived human neurons remains unknown. We identified a novel compound heterozygous mutation, due to homozygous variation of biparental origin and heterozygous variation inherited from the father, in the VPS13B gene in a 20-month-old female patient. To understand the cellular pathogenic mechanisms, we generated induced pluripotent stem cells (iPSCs) from the fibroblasts of the CS patient. The iPSCs were differentiated into forebrain-like functional glutamatergic neurons or neurospheres. Functional annotation from transcriptomic analysis using CS iPSC-derived neurons revealed that synapse-related functions were enriched among the upregulated and downregulated genes in the CS neurons, whereas processes associated with neurodevelopment were enriched in the downregulated genes. The developing CS neurospheres were small in size compared to control neurospheres, likely due to the reduced proliferation of SOX2-positive neural stem cells. Moreover, the number of SV2B-positive puncta and spine-like structures was significantly reduced in the CS neurons, suggesting synaptic dysfunction. Taking these findings together, for the first time, we report a potential cellular pathogenic mechanism which reveals the alteration of neurodevelopment-related genes and the dysregulation of synaptic function in the human induced neurons differentiated from iPSCs and neurospheres of a CS patient.

Heterogeneity in the etiopathology of autism spectrum disorders (ASD) limits the development of generic remedies, requires individualistic and patient-specific research. Recent progress in human-induced pluripotent stem cell (iPSC) technology provides a novel platform for modeling ASDs for studying complex neuronal phenotypes. In this study, we generated telencephalic induced neuronal (iN) cells from iPSCs derived from an ASD patient with a heterozygous point mutation in the DSCAM gene. The mRNA of DSCAM and the density of DSCAM in dendrites were significantly decreased in ASD compared to control iN cells. RNA sequencing analysis revealed that several synaptic function-related genes including NMDA receptor subunits were downregulated in ASD iN cells. Moreover, NMDA receptor (R)-mediated currents were significantly reduced in ASD compared to control iN cells. Normal NMDA-R-mediated current levels were rescued by expressing wild-type DSCAM in ASD iN cells, and reduced currents were observed by truncated DSCAM expression in control iN cells. shRNA-mediated DSCAM knockdown in control iN cells resulted in the downregulation of an NMDA-R subunit, which was rescued by the overexpression of shRNA-resistant DSCAM. Furthermore, DSCAM was co-localized with NMDA-R components in the dendritic spines of iN cells whereas their co-localizations were significantly reduced in ASD iN cells. Levels of phospho-ERK1/2 were significantly lower in ASD iN cells, suggesting a potential mechanism. A neural stem cell-specific Dscam heterozygous knockout mouse model, showing deficits in social interaction and social memory with reduced NMDA-R currents. These data suggest that DSCAM mutation causes pathological symptoms of ASD by dysregulating NMDA-R function.

The present work describes the feasibility of coffee residue extracts as cryoprotective agents in the storage stability of freeze-dried lactic acid bacteria. Coffee residue extracts were extracted from coffee residue, produced after coffee extraction for coffee powder and instant coffee preparation, using an autoclave. Leuconostoc mesenteroides WiKim32 was selected to evaluate the ability of coffee residue extracts to protect bacteria during freeze-dried storage. The storage stability of freeze-dried Leu. mesenteroides WiKim32 with coffee residue extracts was comparable to those with commercial cryoprotective agents. Coffee residue extracts contributed to storage stability immediately after freeze-drying (61.2%) and subsequent storage (48.7%). Our data indicate that the protective effect of the coffee residue extracts is associated with ions, carbohydrates, and phenolic compounds. Coffee residue extracts are feasible materials, which can reduce the storage and distribution costs compared to commercial agents currently available.

Autism spectrum disorder (ASD) is a group of neurodevelopmental disorders that are highly heterogeneous in clinical symptoms as well as etiologies. Mutations in SHANK2 are associated with ASD and accordingly, Shank2 knockout mouse shows ASD-like behavioral phenotypes, including social deficits. Intriguingly, two lines of Shank2 knockout (KO) mouse generated by deleting different exons (exon 6–7 or exon 7) showed distinct cellular phenotypes. Previously, we compared gene expressions between Shank2 KOs lacking exon 6–7 (e6–7 KO) and KOs lacking exon 7 (e7 KO) by performing RNA-seq. In this study, we expanded transcriptomic analyses to identify novel transcriptional variants in the KO mice. We found prominent expression of a novel exon (exon 4′ or e4’) between the existing exons 4 and 5 in the Shank2 e6–7 KO model. Expression of the transcriptional variant harboring this novel exon was confirmed by RT-PCR and western blotting. These findings suggest that the novel variant may function as a modifier gene, which contributes to the differences between the two Shank2 mutant lines. Furthermore, our result further represents an example of genetic compensation that may lead to phenotypic heterogeneity among ASD patients with mutations in the same gene.

As concerns are increasing about drinking water contamination with heavy metals, we investigated the possible use of a pellicle (floating biofilm)-like biofilm-producing microorganism as a biosorbent for the treatment of Pb(II) in aqueous solutions. The bacterial pellicle-producing Methylobacterium hispanicum EM2 strain (EM2) was newly isolated from mine tailing soil, and we investigated its use as a biosorbent for treating a Pb(II)-contaminated aqueous solution. The EM2 strain was strongly resistant to Pb(II) up to a concentration of 800 mg/L, and achieved remarkable adsorption performance (adsorption rate and maximum adsorption capacity of 96% &plusmn, 3.2% and 79.84 mg/g, respectively) under optimal conditions (pH, biomass content, contact time, and initial Pb(II) concentration of 7.1 g/L, 60 min, and 10 mg/L, respectively). The adsorption of Pb(II) was characterized by scanning electron microscopy-energy dispersive x-ray spectroscopy and Fourier-transform infrared analysis. The equilibrium data matched the Freundlich isotherm model well, indicating the occurrence of multilayer adsorption of Pb(II) onto the heterogeneous surface of the EM2 strain, which was also consistent with the pseudo-second-order kinetic model (R2 = 0.98). The high Pb(II) removal efficiency was also confirmed by conducting an adsorption experiment using Pb(II)-contaminated industrial wastewater.

We herein report a convenient radiolabeling method capable of real-time in vivo imaging and biodistribution of bacteria in a living subject. Escherichia coli (E. coli), a model microorganism in this study, was incubated with technetium-99m (99mTc) tricarbonyl under physiological conditions provided radiolabeled bacterial cells. 99mTc-labeled bacterial cells were obtained in high yield (42%, n = 5) and purity (> 99%) within 30 min. In vitro radiolabeling study using cellular components revealed that 99mTc could be incorporated with cytosolic proteins and it was maintained stably in E. coli under several media including mouse serum, high salt solutions with varying pH. SPECT/CT images and organ biodistribution of 99mTc-labeled bacterial cells showed quantitative in vivo behavior and accumulation of E. coli in mice. The radiolabeling method demonstrated here will be a promising platform tool for in vivo tracking and biodistribution of infectious microorganism.

The purpose of this study was to evaluate the effects of hiziki extract on alveolar bone loss, inflammation, and osteo-biomarker expression in hPDL cells (10, 50, 100 μg/ml final concentrations in culture medium) and on a ligature-induced periodontitis rat model (50, 100, 200 mg/kg with oral administration). Hiziki extract increased alkaline phosphatase activity and mineralized nodule formation in hPDL cell. In western blot analysis, hiziki extract resulted in increased expression of osteoblast markers, including transforming growth factor beta (TGF-β), SMAD anchor for receptor activation (SARA) and runt-related transcription factor 2 (RUNX2) in hDPL cells. Additionally, expression of osteoclast markers and inflammatory cytokines was inhibited, which were receptor activator of NF-κB (RANK), RANK receptor (RANKL) and nuclear factor of activated T cells, cytoplasmic 1 (NFATc1). Hiziki extract also prevented alveolar bone loss in a ligature-induced periodontitis rat model through reducing the distance between cementoenamel junction and alveolar bone crest (CBJ-ABC) and furcation involvement. These findings suggested that hiziki extract has prophylactic potential for the prevention of periodontitis through anti-inflammation and, anti-bone resorption effects and the inhibition of alveolar bone destruction.

이 연구는 토착심리학(indigenous)적인 관점에서 한국인들의 고유한 문화 변수(체면과 우리)가 갈등 상황에서 커뮤니케이션하는 방식에 어떤 영향을 주는지 살펴보았다. 또한, 감정 변수(분노와 연민) 가 문화와 갈등전략의 관계를 어떻게 매개하는지 검증하였다. 연구결과 갈등상황에서 체면은 갈등 전략에 우리보다 더 강한 영향력을 갖는 것으로 나타나, 체면이 갈등해소 전략에 미치는 영향과 관련한 선행연구 결과를 지지하였다. 또한 체면과 우리가 갈등해소 방식에 미치는 효과는 갈등 유형 에 따라 다르게 나타났다. 체면은 상대적으로 감정관여도가 높은 갈등 상황에서 영향력을 보였다. 감정변수인 분노의 매개효과 또한 갈등 유형에 따라 다르게 나타났는데, 상대적으로 감정관여도가 높은 갈등 상황에서는 연민의 매개효과가 나타났던 반면, 상대적으로 감정관여도가 낮은 갈등 상황 에서는 분노의 매개효과만이 발견되었다. 이와 같은 결과를 통해 한국인의 문화변수가 가지는 맥락 적 특성에 대한 기존의 이론적 논의를 경험적으로 입증하였다.

Purpose. In traditional Korean medicine, Artemisia apiacea H. (ART) and Scutellaria baicalensis G. (SCU) are combined for the treatment of malaria and other malaria-like diseases. Because SCU is well-known as an antibacterial agent, the antimicrobial effect of a mixture of ART and SCU was investigated. Methodology. Plant samples were purchased from Kyungdong mart and extracted with 70 % ethanol. The in vitro antimicrobial activity of ART and SCU against pathogenic fungi (Aspergillus niger, Aspergillus oryzae, Candida albicans, Candida tropicalis and Candida glabrata), Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus) was evaluated using a broth microdilution assay, modified-disc diffusion and agar dilution methods with further CH2Cl2-fractionated ART, SCU and a mixture of ART/SCU (at a ratio of 3 : 5) (THAN-1). Results. ART and SCU were effective against A. niger, C. albicans, B. subtilis and S. aureus. The range of minimum inhibitory concentration (MIC) values was 0.03125 to 4 mg ml−1 in the ART and SCU treatments. ART exhibited stronger activity than SCU. Interestingly, a 3 : 5 ratio mixture of ART and SCU (THAN-1) showed stronger antimicrobial activity than ART or SCU used individually. Conclusion. A treatment using a mixture of herbs such as THAN-1 would be useful in the suppression of the growth of pathogenic bacterial and fungal strains.

본 연구의 목적은 활동중심 평가의 하나인 브릿징 평가의 음악 및 동작영역을 우리나라 유아교육 현장에 적용해 보고, 교사의 평가 경험 및 유아의 반응을 탐색하는 데 있다. 연구참여자는 D시에 위치한 어린이집 교사 1인, 공립유치원 교사 2인과 각 학급으로부터 6명씩 총 18명의 만4, 5세 유아들이다. 연구자료는 참여관찰과 평가 후 면담, 심층면담, 전화통화 및 이메일 교신, 그리고 유아평가 기록지, 교사저널, 연구일지 등을 통해 수집, 전사, 분석되었다. 유아들에게 음악을 듣고 분석하여 동작으로 표현하는 능력을 평가하는 음악에 맞춰 동작 표현하기, 조화롭게 조율하여 리듬악기를 연주해 보는 리듬악기 연주하기, 가장 기본적인 목소리를 활용하여 리듬과 비트를 표현하는 노래 부르기라는 세 가지 활동으로 이루어진 음악 및 동작영역의 브릿징 평가활동을 실시하였다. 연구결과, 노래 부르기 활동은 쉽게 이루어졌지만, 평소 자주 하지 않던 악기 연주하기나 음악에 맞춰 동작으로 표현하기는 유아들이 다소 생소해 하는 모습이 발견되었다. 이러한 결과는 우리 유아교육현장에서 더 다양한 음악활동을 통해 유아의 음악 및 동작영역에서의 발달과 학습을 지원해 줄 수 있는 방향으로 교사 태도 및 실천에서의 변화가 요구됨을 제안하였으며 이러한 변혁 과정에서 활동중심 평가의 긍정적 가능성을 시사하였다.

A Gram-stain-negative, aerobic, non-motile, yellow and rod-shaped bacterial strain was isolated from forest mud located at Kyung Hee University, South Korea. Strain THG-AG6.4T grew at 10–35 °C, pH 6.0–9.5 and in the presence of 0–1.5 % (w/v) NaCl. Phylogenetic analysis, based on 16S rRNA gene sequencing, showed that strain THG-AG6.4T was most closely related to Flavobacterium gyeonganense HME 7524T (97.66 %), Flavobacterium defluvii EMB 117T (96.93 %) and Flavobacterium arsenitoxidans S2-3HT (96.80 %). The DNA G+C content of strain THG-AG6.4T was 30.2 mol%. The DNA–DNA relatedness values between strain THG-AG6.4T and its closest phylogenetic neighbour, F. gyeonganense HME 7524T, were below 61.0 %. The predominant quinone of strain THG-AG6.4T was MK-6. The major polar lipids were phosphatidylethanolamine, an unidentified phospholipid, five unidentified glycolipids, phosphatidylserine, an unidentified lipid, an unidentified aminophospholipid, two unidentified aminolipids and two unidentified aminoglycolipids. The major fatty acids were C16 : 0, C18 : 0 10-methyl, summed feature 3 and C18 : 1ω9c. The major polyamine was homospermidine. On the basis of the phenotypic, genotypic and phylogenetic characterization of strain THG-AG6.4T, it is concluded that the isolate represents a novel species of the genus Flavobacterium , for which the name Flavobacterium limi sp. nov. is proposed, with THG-AG6.4T as the type strain (=KACC 18851T=CGMCC 1.16060T).

Radix Scutellariae (RS) has long been used in the treatment of inflammatory and allergic diseases. Its main flavonoids, baicalin (BG) and wogonoside (WG), can be hydrolyzed into their corresponding aglycones, baicalein (B) and wogonin (W). In this study, we developed a safe and effective method of transforming these glycosides using Peclyve PR. The transformation rate of BG and WG reached 98.5 and 98.1%, respectively, with 10% enzyme at 40 °C for 60 h. Furthermore, we compared the anti-photoaging activity of RS before and after enzyme treatment, as well as their respective main components, in UVB-irradiated HaCaT cells. Results found that enzyme-treated RS (ERS) appeared to be much better at preventing UVB-induced photoaging than RS. ERS significantly inhibited the upregulation of matrix metalloproteinase-1 and IL-6 caused by UVB radiation by inactivating the MAPK/AP-1 and NF-κB/IκB-α signaling pathways. ERS treatment also recovered UVB-induced reduction of procollagen type I by activating the TGF-β/Smad pathway. In addition, ERS exhibited an excellent antioxidant activity, which could increase the expression of cytoprotective antioxidants such as HO-1 and NQ-O1, by facilitating Nrf2 nuclear transfer. These findings demonstrated that the photoprotective effects of RS were significantly improved by enzyme-modified biotransformation.

A Gram-stain-positive, aerobic, non-motile, short-rod shaped actinobacterium, designated THG-T2.14T, was isolated from soil sampled from the rhizosphere of mugunghwa. Growth occurred at 10–40 °C (optimum 30 °C), at pH 5.0–10.0 (optimum 7.0) and at 0–7.0 % NaCl (optimum 3.0 %). Based on 16S rRNA gene sequence analysis, the nearest phylogenetic neighbours of strain THG-T2.14T were identified as Microbacterium yannicii DSM 23203T (98.8 %), Microbacterium trichothecenolyticum DSM 8608T (98.8 %), Microbacterium arthrosphaerae DSM 22421T (98.7 %) and Microbacterium jejuense KACC 17124T (98.4 %). The major fatty acids were anteiso-C15 : 0, anteiso-C17 : 0 and iso-C16 : 0. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, one unidentified lipid, two unidentified phospholipids and two unidentified phosphoglycolipids. The menaquinones were MK-12, and MK-13. The major polyamine was spermidine. The peptidoglycan contained ornithine, alanine, glycine, homoserine and glutamic acid. The diagnostic diamino acid was ornithine. The acyl type of the muramic acid was glycolyl. The whole-cell sugars were rhamnose, ribose, galactose, arabinose, xylose and glucose. The DNA G+C content of strain THG-T2.14T was 71.2 mol%. The DNA–DNA relatedness between strain THG-T2.14T and its closest reference strains were significantly lower than the threshold value of 70 %. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA–DNA hybridization data, strain THG-T2.14T represents a novel species of the genus Microbacterium , for which the name Microbacterium hibisci sp. nov. is proposed. The type strain is THG-T2.14T (=KACC 18931T=CCTCC AB 2016180T).

This study presents two science teachers, Catherine and Jennifer, who took their first steps toward teaching socioscientific issues through collaborative action research. The teachers participated in the collaborative action research project because they wanted to address socioscientific issues but had limited experience in teaching them. The research questions included what kinds of challenges the teachers encountered when implementing socioscientific issues and to what extent they resolved the challenging issues as participating in collaborative action research. The primary data source consisted of audiotapes of regular group meetings containing information on the process of constructing and implementing lesson plans and reflecting on their teaching of socioscientific issues. We also collected classroom videotapes of the teachers’ instruction and audiotapes of students’ small group discussions and their worksheets. The findings indicated that when addressing socioscientific issues in the classes, the teachers encountered several challenging issues. We categorized them into four: (1) restructuring classroom dynamics and culture, (2) scaffolding students’ engagement in socioscientific issues, (3) dealing with values, and (4) finding their niche in schools. However, this study showed that collaborative action research could be a framework for helping the teachers to overcome such challenges and have successful experiences of teaching socioscientific issues. These experiences became good motivation, to gradually develop their understanding of teaching socioscientific issues and instructional strategies for integrating the knowledge and skills that they had accumulated over the years.

A Gram-stain-positive, pink-pigmented, coccus-shaped, strictly aerobic, non-motile bacterium, strain THG-AG1.5T, was isolated from rhizosphere of Hibiscus syriacus L. (Mugunghwa flower) located in Kyung Hee University, Yongin, Gyeonggi, Republic of Korea. The isolated strain grew optimally at 25–30 °C, at pH 6.0–7.5 and in the presence of additional 0–1.5 % (w/v) NaCl. Strain THG-AG1.5T exhibited tolerance to UV radiation (>1500 J m−2) and to gamma radiation (>12 kGy). Based on 16S rRNA gene sequence comparisons, strain THG-AG1.5T was closely related to Deinococcus daejeonensis MJ27T (98.03 %), Deinococcus radiotolerans C1T (97.61 %) and Deinococcus grandis DSM 3963T (97.32 %). The genomic DNA G+C content of strain THG-AG1.5T was 74.8 mol%. The DNA–DNA hybridization values between strain THG-AG1.5T and its closest phylogenetically neighbours were below 63.0 %. The peptidoglycan amino acids were alanine, valine, glutamic acid, glycine, ornithine, lysine and aspartic acid. Strain THG-AG1.5T contained ribose, mannose and glucose as whole-cell-wall sugars and menaquinone-8 (MK-8) as the only isoprenoid quinone. The major component in the polyamine pattern was spermidine. The major polar lipids of strain THG-AG1.5T were a phosphoglycolipid, six unidentified glycolipids and an unidentified aminophospholipid. The major fatty acids were identified as iso-C15 : 0, C15 : 1ω6c, C16 : 0, iso-C17 : 0, C17 : 0, C18 : 0 and summed feature 3. On the basis of our polyphasic taxonomy study, strain THG-AG1.5T represents a novel species within the genus Deinococcus , for which the name Deinococcus hibisci sp. nov. is proposed. The type strain is THG-AG1.5T (=KACC 18850T=CCTCC AB 2016078T).

A Gram-stain-negative, facultatively anaerobic, non-motile, rod-shaped and yellow-pigmented bacterium, designated strain THG-DN6.14T, was isolated from a freshwater sample near Donghaksa temple in Daejeon, South Korea. On the basis of the results of 16S rRNA gene sequence comparisons, THG-DN6.14T was found to be most closely related to Emticicia sediminis JBR12T (99.1 % sequence similarity), Emticicia oligotrophica DSM 17448T (97.6 %), Emticicia aquatica HMF2925T (96.5 %), and Emticicia ginsengisoli Gsoil 085T (94.4 %). The DNA–DNA relatedness between THG-DN6.14T and its phylogenetically closest neighbours was below 65.0 %. The DNA G+C content was 43.3 mol%. The major polar lipids were found to be phosphatidylethanolamine, an unidentified glycolipid and an unidentified aminoglycolipid. The major fatty acids were identified as C16 : 0, iso-C15 : 0, iso-C17 : 0 3-OH, and summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c). The respiratory quinone was menaquinone MK-7. These data supported the affiliation of THG-DN6.14T to the genus Emticicia . THG-DN6.14Tcould be distinguished from related species of the genus Emticicia by physiological and biochemical tests. Therefore, the novel isolate represents a novel species, for which the name Emticicia aquatilis sp. nov. is proposed, with THG-DN6.14T (=KACC 18540T=CGMCC 1.15958T) as the type strain.

The extremophilic bacterium Deinococcus radiodurans R1 has been considered as an attractive microorganism due to its remarkable tolerance to various external stresses. Considering the nature of D. radiodurans R1, it has potential as a platform microorganism for industrial applications, including biorefinery and bioremediation process. However, D. radiodurans R1 is well known for its hard genetic manipulation. Thus, much effort has been made to develop efficient genetic engineering tools for making D. radiodurans R1 suitable for industrial platform microorganism. Although a plasmid-based single gene knockout method has been reported, development of multiple gene knockout system has not yet been reported. Here we report, for the first time, Cre-lox based rapid and efficient multiple knockout method for metabolic engineering of D. radiodurans R1. Also, deletion of dr0053 gene was successfully achieved within seven days to make biofilm overproducing strain.

The efficient treatment of radioactive waste arising from nuclear accidents and overuse of radioisotopes has become an important issue, and much effort has been devoted to protecting the environment from such toxic radioactive materials. Among them, radioactive iodine in water is regarded as one of the most palpable threats to humans, as it is generated by post-use in medical and industrial applications. Although a few adsorbents and purification processes have been reported, the development of a better desalination method that is able to provide high removal efficiency, ion-selectivity, and reusability is still in demand. Herein, we report a hybrid composite membrane, composed of gold nanoparticles and cellulose acetate membranes, which has the potential to be used for efficient desalination of radioactive iodine. A single filtration process using this membrane provided high removal efficiency with more than two orders of magnitude iodide anion removal within a minute and excellent ion-selectivity. Moreover, the same operation can be applied several times without significant decrease of its performance. Therefore, it is anticipated that our work will be a new avenue for desalination of radioactive iodine in various aqueous media.

According to 16S rRNA gene sequence comparisons, strain THG-DN6.20T was found to be most closely related to Novosphingobium rosa IFO 15208T (97.6 % sequence similarity), Novosphingobium sediminicola HU1-AH51T (97.5 %) and Novosphingobium barchaimii LL02T (96.9 %). The DNA-DNA relatedness between strain THG-DN6.20T and its phylogenetically closest neighbours was below 60.0 %. The respiratory quinone and polyamine detected in strain THG-DN6.20T were ubiquinone Q-10 and spermidine, respectively. The DNA G+C content was 63.1 mol%. The major polar lipids were found to be phosphatidylethanolamine, diphosphatidylglycerol, sphingoglycolipid and phosphatidylcholine. The major fatty acids were identified as C16 : 0, summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c), summed feature 8 (C18 : 1ω7c and/or C18 : 1ω6c) and C14 : 0 2-OH. These data supported the affiliation of strain THG-DN6.20T to the genus Novosphingobium. Strain THG-DN6.20T could be distinguished from related species of the genus Novosphingobium by physiological and biochemical characteristics. Therefore, the novel isolate represents a novel species, for which the name Novosphingobium lotistagni sp. nov. is proposed, with THG-DN6.20T as the type strain (=KACC 18541T=CCTCC AB 2015354T).

This paper describes a novel chromatographic method for efficient removal of anionic dyes from aqueous solutions. Chitosan-coated Fe3O4 nanoparticles can easily be immobilized on a dextran gel column. Single elution of Evans Blue (EB) solution to the nanoadsorbent-incorporated columns provides high removal efficiency with a maximum adsorption capacity of 243.9 mg/g. We also investigated the influence of initial concentration and solution pH on the removal efficiency of EB. The electrostatic interaction between the adsorbent surface and negatively charged sulfate groups on EB molecules promotes the efficient adsorption of dyes. The equilibrium data matched well with the Langmuir isotherm model, which indicated monolayer dye adsorption onto the adsorbent surface. To extend the application of the current method, we performed further adsorption experiments using other anionic dyes of different colors (Cy5.5, Acid Yellow 25, Acid Green 25, and Acid Red 1). All of these molecules can efficiently be captured under continuous flow conditions, with higher removal efficiency obtained with more negatively charged dyes. These findings clearly demonstrate that the present approach is a useful method for the removal of anionic dye contaminants in aqueous media by adsorption.

Purpose: To investigate the protective effect of Gastrodia elata Blume ( G. elata , GE) and cyclodextrin glucanotransferase (CGTase) enzyme-treated G. elata extract (EGE) against premature skin aging using ultraviolet B (UVB)-exposed normal human dermal fibroblasts (NHDFs). Methods: The extract was characterized by liquid chromatography with tandem mass spectrometry (LC-MS/MS), ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC–QToF–MS) and nuclear magnetic resonance spectroscopy (NMR). The expression of matrix metalloproteinases (MMP-1,3), interleukin-6 (IL-6), transforming growth factor (TGF-β1) and procollagen type I was assayed using ELISA kits. Safety evaluation of EGE’s dietary administration and topical application was performed by in vivo acute oral toxicity and local lymph node tests. Results: Lower MMP-1 and IL-6 and higher procollagen type I and TGF-β1 levels were observed after treatment with EGE than with GE, indicating that EGE was more effective than GE in treating UVBinduced photoaging. With respect to phenolic composition, EGE had lower 4-hydroxybenzaldehyde (4- HBA) level and higher α-gastrodin level than GE. In UVB-irradiated NHDFs, α-gastrodin exhibited higher anti-aging activity than 4-HBA and β-gastrodin based on the expression of MMP-1, MMP-3, and procollagen type I. The in vivo data indicate that EGE was safe at concentrations of up to 2000 mg/kg for dietary administration and 0.1 % for topical application. Conclusion: EGE protects UVB-induced photoaged human skin better than GE owing to its higher α- gastrodin content. Thus, EGE may be potentially useful agent in anti-aging cosmetic products.
