The hemogram and hemogram-derivative ratios (HDRs) are becoming markers of the severity and mortality of COVID-19. We evaluated the hemograms and serial weekly HDRs [neutrophil-lymphocyte ratio (NLR), monocyte-lymphocyte ratio (MLR), platelet-lymphocyte ratio (PLR), neutrophil-platelet ratio (NPR) and systemic immune-inflammatory index (SII)] in the survivors and non-survivors of COVID-19.We retrospectively reviewed the medical notes and serial hemograms of real-time reverse-transcription polymerase chain reaction (RT-PCR)-confirmed COVID-19 adults hospitalized from April 2020 to March 2021 from the time of diagnosis to the 3Of the 320 adults, 257 (80.3%) were survivors and had a lower mean age than the non-survivors (57.73This study shows that serial lymphocyte and neutrophil counts, NLR, PLR, MLR, NPR and SII could serve as good and easily accessible markers of severity and predictors of outcomes in COVID-19 patients and should be used for the monitoring of treatment response.

Coronavirus disease (COVID-19) is an infectious disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Its main form of transmission is through respiratory droplets. Case reports have described the presence of this virus in biological materials such as blood, feces, urine, and tears, which generate hypotheses about other means thereby the disease is transmitted. In this report, we describe a case of SARS-CoV-2 identified on the eye surface of an asymptomatic health-care professional. The nasopharyngeal reverse transcription polymerase chain reaction test, using a sample collected on the same day, and the serological test, performed 3 months later, did not reveal any evidence of SARS-CoV-2 infection. These results alert on the possibility of a false-positive reverse transcription polymerase chain reaction result for the ocular surface or the presence of the virus in the conjunctival mucosa in individuals without infection.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can infect a broad range of human tissues by using the host receptor angiotensin-converting enzyme 2 (ACE2). Individuals with comorbidities associated with severe COVID-19 display higher levels of ACE2 in the lungs compared to those without comorbidities, and conditions such as cell stress, elevated glucose levels and hypoxia may also increase the expression of ACE2. Here, we showed that patients with Barrett's esophagus (BE) have a higher expression of ACE2 in BE tissues compared to normal squamous esophagus, and that the lower pH associated with BE may drive this increase in expression. Human primary monocytes cultured in reduced pH displayed increased ACE2 expression and higher viral load upon SARS-CoV-2 infection. We also showed in two independent cohorts of 1,357 COVID-19 patients that previous use of proton pump inhibitors is associated with 2- to 3-fold higher risk of death compared to those not using the drugs. Our work suggests that pH has a great influence on SARS-CoV-2 Infection and COVID-19 severity.

COVID-19 patients may exhibit neuropsychiatric and neurological symptoms. We found that anxiety and cognitive impairment are manifested by 28-56% of SARS-CoV-2-infected individuals with mild respiratory symptoms and are associated with altered cerebral cortical thickness. Using an independent cohort, we found histopathological signs of brain damage in 25% of individuals who died of COVID-19. All of the affected brain tissues exhibited foci of SARS-CoV-2 infection and replication, particularly in astrocytes. Infection of neural stem cell-derived astrocytes changed energy metabolism, altered key proteins and metabolites used to fuel neurons and for biogenesis of neurotransmitters, and elicited a secretory phenotype that reduces neuronal viability. Our data support the model where SARS-CoV-2 reaches the brain, infects astrocytes and triggers neuropathological changes that contribute to the structural and functional alterations in the brain of COVID-19 patients.

The antiparasitic drug nitazoxanide is widely available and exerts broad-spectrum antiviral activity in vitro. However, there is no evidence of its impact on SARS-CoV-2 infection.In a multicenter, randomized, double-blind, placebo-controlled trial, adult patients who presented up to 3 days after onset of Covid-19 symptoms (dry cough, fever, and/or fatigue) were enrolled. After confirmation of SARS-CoV2 infection by RT-PCR on nasopharyngeal swab, patients were randomized 1:1 to receive either nitazoxanide (500 mg) or placebo, TID, for 5 days. The primary outcome was complete resolution of symptoms. Secondary outcomes were viral load, general laboratory tests, serum biomarkers of inflammation, and hospitalization rate. Adverse events were also assessed.From June 8 to August 20, 2020, 1,575 patients were screened. Of these, 392 (198 placebo, 194 nitazoxanide) were analyzed. Median time from symptom onset to first dose of study drug was 5 (4-5) days. At the 5-day study visit, symptom resolution did not differ between the nitazoxanide and placebo arms. However, at the 1-week follow-up, 78% in the nitazoxanide arm and 57% in the placebo arm reported complete resolution of symptoms (p=0.048). Swabs collected were negative for SARS-CoV-2 in 29.9% of patients in the nitazoxanide arm versus 18.2% in the placebo arm (p=0.009). Viral load was also reduced after nitazoxanide compared to placebo (p=0.006). No serious adverse events were observed.In patients with mild Covid-19, symptom resolution did not differ between the nitazoxanide and placebo groups after 5 days of therapy. However, early nitazoxanide therapy was safe and reduced viral load significantly.Take home messageThis was the first study to evaluate the effect of early nitazoxanide therapy in mild Covid-19. Nitazoxanide did not accelerate symptom resolution after 5 days of therapy; however, reduced viral load significantly with no serious adverse events.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the agent of a major global outbreak of respiratory tract disease known as coronavirus disease-2019 (COVID-19). SARS-CoV-2 infects mainly lungs and may cause several immune-related complications, such as lymphocytopenia and cytokine storm, which are associated with the severity of the disease and predict mortality1,2. The mechanism by which SARS-CoV-2 infection may result in immune system dysfunction is still not fully understood. Here we show that SARS-CoV-2 infects human CD4+T helper cells, but not CD8+T cells, and is present in blood and bronchoalveolar lavage T helper cells of severe COVID-19 patients. We demonstrated that SARS-CoV-2 spike glycoprotein (S) directly binds to the CD4 molecule, which in turn mediates the entry of SARS-CoV-2 in T helper cells. This leads to impaired CD4 T cell function and may cause cell death. SARS-CoV-2-infected T helper cells express higher levels of IL-10, which is associated with viral persistence and disease severity. Thus, CD4-mediated SARS-CoV-2 infection of T helper cells may contribute to a poor immune response in COVID-19 patients.

Background: Brazil became the South American epicenter for coronavirus disease (COVID-19) soon after the first case was diagnosed in February 2020 with the highest infection rate occurring in the state of Sao Paulo. COVID-19 is characterized by marked thrombo-inflammation mechanisms, and neutrophil-lymphocyte ratio (NLR) among many clinical and laboratory data, is becoming an inflammatory marker of severity and mortality of COVID-19. We evaluated the serial weekly lymphocyte ratios, which are easily derivable from the routine blood counts, in the survivors and non-survivors of COVID-19 at the Clinical Hospital of University of Campinas (UNICAMP), Campinas, Sao Paulo, Brazil, from time of diagnosis to the 3 rd week of care. This hospital is one of the referral centers for COVID-19 patients in this state. Methods: In this retrospective study, we reviewed the medical notes of 320 adults hospitalized patients with PCR-confirmed COVID-19 at the Clinical Hospital of UNICAMP, from March 2020 to March 2021. The serial weekly hematological parameters (analyzed using automated counter - XN 9000™, Sysmex, Japan) from the time of diagnosis were analyzed and lymphocytes ratios (neutrophil-lymphocyte, NLR, platelet-lymphocyte PLR, and monocyte-lymphocyte MLR) were calculated. The survivors (n=257) were those who recovered from the disease and were discharged from the hospital, while the non-survivors (n=63) were those who died in the course of treatment. Statistical analyses were performed using SPSS (version 22). Unpaired data of Survivors and Non-survivors with COVID-19 were compared using Mann-Whitney tests. Repeated measures were compared within and between groups using univariate and multivariate tests in general linear models. On the other hand, all three ratios significantly decrease in the survivors. While neutrophilia and lymphopenia improved in the survivor, they worsen in non-survivors. These cells may have contributed towards the recovery by ameliorating the inflammatory response in survivors, and death by worsening the response in non-survivors of COVID-19. This study shows that serial lymphocyte count, neutrophil count, NLR, PLR, and MLR could serve as good and easily accessible markers of outcomes in patients with COVID-19 and could be used for monitoring of response to treatment. COVID-19 can result in severe lung injury. It remained to be determined why diabetic individuals with uncontrolled glucose levels are more prone to develop the severe form of COVID-19. The molecular mechanism underlying SARS-CoV-2 infection and what determines the onset of the cytokine storm found in severe COVID-19 patients are unknown. Monocytes/macrophages are the most enriched immune cell types in the lungs of COVID-19 patients and appear to have a central role in the pathogenicity of the disease. These cells adapt their metabolism upon infection and become highly glycolytic, which facilitates SARS-CoV-2 replication. The infection triggers mitochondrial ROS production, which induces stabilization of hypoxia-inducible factor- 1α (HIF - 1α) and consequently promotes glycolysis. HIF- 1α-induced changes in monocyte metabolism by SARS-CoV-2 infection directly inhibit T cell response and reduce epithelial cell survival. Targeting HIF-1 ɑ may have great therapeutic potential for the development of novel drugs to treat COVID-19.

The increase of Zika virus (ZIKV) infections in Brazil in the last two years leaves a prophylactic measures on alert for this new and emerging pathogen. Concerning of our positive experience, we developed a new prototype using Neisseria meningitidis outer membrane vesicles (OMV) on ZIKV cell growth in a fusion of OMV in the envelope of virus particles. The fusion of nanoparticles resulting from outer membrane vesicles of N. meningitidis with infected C6/36 cells line were analyzed by Nano tracking analysis (NTA), zeta potential, differential light scattering (DLS), scan and scanning transmission eletronic microscopy (SEM and STEM) and high resolution mass spectometry (HRMS) for nanostructure characterization. Also, the vaccination effects were viewed by immune response in mice protocols immunization (ELISA and inflammatory chemokines) confirmed by Zika virus soroneutralization test. The results of immunizations in mice showed that antibody production had a titer greater than 1:160 as compared to unvaccinated mice. The immune response of the adjuvant and non-adjuvant formulation activated the cellular immune response TH1 and TH2. In addition, the serum neutralization was able to prevent infection of virus particles in the glial tumor cell model (M059J). This research shows efficient strategies without recombinant technology or DNA vaccines.

O bocavirus humano (HBoV) é um parvovirus recentemente identificado em associação com a presença de sintomas de infecção do trato respiratório. Esse vírus possui um genoma de aproximadamente 5217 nucleotídeos que contém 3 open reading frames que codificam 4 proteínas (NS1, NP-1, VP-1 e VP-2). HBoV tem sido detectado em amostras respiratórias de diversas partes do mundo, incluindo Austrália, América do Norte, Europa, Ásia e África, o que sugere uma distribuição global desse vírus. Entretanto, nenhum estudo longitudinal de HBoV em amostras respiratórias foi realizado na América Latina. Dessa forma, nós realizamos um estudo prospectivo de HBoV em lavados nasofaríngeos (LFNs) coletados de pacientes com sintomas de infecção do trato respiratório (IRA) atendidos em um hospital universitário de Ribeirão Preto, SP e em um hospital universitário de Salvador, BA no período entre 2005 a 2007. 1288 LFNs de 1217 pacientes foram encaminhados ao laboratório de virologia e foram testados por PCR para HBoV. Desses pacientes, 962 eram menores de 5 anos e 177 eram maiores de 5 anos. Além disso, também foram analisados 50 LFNs de crianças menores de 5 anos que não tinham sintomas respiratórios. Todas as amostras positivas para HBoV foram testadas para todos os outros vírus respiratórios, incluindo o vírus sincicial respiratório (HRSV), rinovirus humano (HRV), influenza humano (HFLU), metapneumovirus humano (HMPV), parainfluenza humano (HPIV), coronavirus humano (HCoV) e adenovirus humano (HAdV). A carga viral de HBoV foi determinada por PCR em tempo real em todas as amostras positivas e o genoma completo de 19 amostras de HBoV foi seqüenciado. Com intuito, de fazer um levantamento sorológico e determinar sítios replicativos de HBoV, nós ainda clonamos e expressamos em S. cerevisae (Y258) o gene de VP2, que codifica uma das proteínas do capsídeo viral. A prevalência desse vírus foi de 4,8% em crianças menores de cinco anos e de 1% em pacientes maiores de cinco anos. HBoV não foi detectado em crianças sem sintomas. Dos 259 pacientes analisados em 2005, 25 (10%) foram positivos para HBoV. Esse vírus circulou mais frequentemente em abril, mês de maior incidência do HRSV. Em 2006, HBoV foi detectado em apenas 10 LFNs de 334 (3%) amostras testadas, sem qualquer pico de freqüência. Em 2007 HBoV foi detectado em 13 de 552 (2%) amostras, com uma freqüência de detecção um pouco maior em junho e julho. Os sintomas mais comumente observados foram rinorréia, tosse, febre e chiado, que foram observados geralmente em mais de 50% dos casos positivos para HBoV. Não houve uma diferença significativa na prevalência desses sintomas entre as crianças positivas e negativas para HBoV. Entretanto, foi observada uma maior freqüência de diarréia entre as crianças com esse vírus. Nesse estudo também foi documentado uma alta freqüência de co-infecções virais entre os pacientes com HBoV. Os vírus mais frequentemente associados com o bocavirus humano foram: HRSV, HRV e HAdV. Além disso, foi detectado uma maior carga viral media e uma maior freqüência de diarréia nos 15 pacientes com infecção exclusiva por HBoV do que nos pacientes com co-infecção. Esses resultados mostraram que HBoV pode alcançar títulos enormes (tão grandes como1014/mL) em LFNs de pacientes com sintomas respiratórios e que isso é associado a de diarréia. O seqüenciamento do genoma inteiro de HBoV realizado nesse estudo indica que a divergência genômica entre as amostras desse vírus é muito pequena. Como conclusão, nós demonstramos que HBoV circula e é detectado em associação com sintomas de infecção respiratória e diarréia no Brasil. Novos estudos, com um longo acompanhamento em diferentes populações serão necessários para determinar a sazonalidade e o real impacto clínico de HBoV em nosso país. Human bocavirus (HBoV) is a parvovirus recently identified in association with respiratory tract infections. HBoV 5217 nt genome contains 3 open reading frames encoding four proteins (NS1, NP-1, VP-1 and VP-2). HBoV has been reported in respiratory samples from children in several parts of the world (including Australia, North America, Europe, Asia, and Africa), suggesting that the virus circulates worldwide. However, no longitudinal studies of HBoV in respiratory samples have been reported in Latin America. We report a prospective study of HBoV in nasopharyngeal aspirates (NPAs) collected from patients seen for acute respiratory tract infections (ARI) at the University of Sao Paulo Hospital in Ribeirao Preto, southeast Brazil and at the University Hospital in Salvador, Brazil. 1288 NPAs from 1217 patients was submitted to the virology lab for respiratory virus detection from 2005 to 2007 and were screened for HBoV by polymerase chain reaction (PCR), whom 962 were under 5 years of age and 177 were older than 5 years. In addition, NPAs from 50 children under 12 years without IRA was also tested to HBoV for PCR. All samples positive of HBoV was tested for others respiratory virus, including the human respiratory syncitial virus (HRSV), human rhinovirus (HRV), human influenza (HFLU), human metapneumovirus (HMPV), human parainfluenza virus (HPIV), human coronavirus (HCoV) and human adenovirus (HAdV). These samples had their HBoV viral load determined by real time PCR and the viral entire genome of nineteen HBoV sample was sequenced. We also cloned and expressed in S. cerevisae (Y258) the gene of VP2, one protein of viral capside. The prevalence of this virus was of 4,8% in children under 5 years and 1% in adults, both with IRA. HBoV was not found on the patients without symptoms. In 2005, of the 259 patients tested, 25 (10%) were positive for HBoV. Interestingly, the virus circulated more frequently in April, the month of peak activity of respiratory HRSV. In 2006 HBoV was detected in only 10 NPAs out of 334 samples (3%) tested, without any notable peak of frequency. In 2007 HBoV was detected in 13 out of 552 (2%) tested samples with little higher frequency of detection in June an July. Rhinorrhea, cough, and wheezing were observed in more than 50% of the HBoV-positive children, and no obvious respiratory clinical differences were noted between HBoV-positive and negative children. However, was noted a higher frequency of diarrhea on HBoV-positive patients. In this study was also observed a larger frequency (71%) of viral coinfections between the HBoV-positive patients. The respiratory viruses more frequently associated with human bocavirus were: HRSV, HRV and HAdV. Interestingly, on the 15 HBoV-alone patients was observed a higher viral load and a higher prevalence of diarrhea than HBoV-coinfection patients. These results showed that this virus can reach enormous titles (like 1014) in NPAs from patients with respiratory infection symptoms and this is associated with diahhrea. The entire genome sequencing of HBoV of our study indicates that the genetic divergence between the HBoV lineages is small. In conclusion, we demonstrated that HBoV circulates and is detected in association with respiratory symptoms and diarrhea in Brazil. Long term surveillance will be needed to determine whether or not an HBoV season occurs and what is the real clinical impact of this virus in our country.

Esta Tese de Doutorado analisa distintos aspectos de respostas imunes a infecções virais e inclui 3 estudos. Na Parte I temos um estudo sobre a proteção cruzada entre os vírus Chikungunya e Mayaro onde foi possível concluir que: A resposta imunológica ao CHIKV confere proteção cruzada parcial contra a infecção secundária por MAYV, reduzindo a carga viral tecidual e os danos histopatológicos. Anticorpos anti-CHIKV de seres humanos e de camundongos possuem baixa neutralização cruzada contra a infecção por MAYV. Após a infecção secundária por MAYV em camundongos, observaram-se elevados títulos de anticorpos com atividade neutralizante cruzada. A depleção de células do sistema imune indicou que uma combinação de células imunes adaptativas podem ser relevantes na proteção cruzada contra MAYV. A redução de citocinas pró-inflamatórias, de células NK e de monócitos inflamatórios durante a infecção por MAYV de camundongos previamente infectados com CHIKV, sugere papel da imunidade inata na proteção cruzada. Na Parte II temos uma análise de epítopos antigênicos da proteína E do vírus Zika, onde foi possível: Selecionar 9 peptídeos lineares da proteína E de ZIKV com base na reatividade cruzada de anticorpos IgG anti-DENV. Dos 9 peptídeos selecionados, 3 pertencem ao domínio I (DI) da proteína E, 2 ao DII, sendo supostamente gênero ou grupo-específicos e 4 peptídeos ao DIII, correspondendo, provavelmente, a regiões antigênicas imunodominantes dos flavivirus. Observou-se soro-neutralização cruzada dos soros de pacientes convalescentes de DENV contra a infecção por ZIKV não relacionada aos 9 peptídeos lineares mencionados. A maioria dos soros de convalescentes de DENV exibiram ADE, potencializando a infecção por ZIKV e, ao ser depletar os anticorpos com afinidade pelos 9 peptídeos lineares selecionados, reduziram esta atividade, sugerindo que anticorpos contra tais peptídeos podem estar envolvidos no ADE para ZIKV. Na Parte III temos uma avaliação da resposta imune à vacina inativada de SARS-CoV-2, onde concluímos que: Indivíduos vacinados 2 vezes com o SARS-CoV-2 inativado seroconvertem, produzindo altos níveis de IgG anti-Spike (S) viral com significativa capacidade neutralizante, que é significativamente reduzida contra SARS-CoV-2 e as variantes Gamma e Zeta após 6 meses. Anticorpos de animais vacinados por vacina de vírus inativado mostraram-se menos neutralizantes contra as variantes Gamma e Zeta do SARS-CoV-2, mas com significativo aumento 3 dias após a infecção. O estímulo de células esplênicas de camundongos com antígenos de SARS-CoV-2 inativado em animais imunizados induziu linfoproliferação mesmo aos antígenos das diferentes variantes virais. A imunização prévia com SARS-CoV-2 inativado reduziu em quase 100 vezes a carga das variantes Gamma e Zeta no pulmão dos camundongos. Níveis indetectáveis de IL-6 e TNF-α nos camundongos imunizados após infecção com as linhagens WT e a variante Zeta, associaram-se com a redução do infiltrado inflamatório pulmonar, evidenciando um papel protetor da imunização prévia por SARS-CoV-2 inativado. This Doctoral Thesis analyzes different aspects of immune responses to viral infections and includes 3 studies. In Part I, we have a study on the cross-protection between Chikungunya and Mayaro viruses where it was possible to conclude that: The immune response to CHIKV provides partial cross-protection against secondary MAYV infection, reducing tissue viral load and histopathological damage. Anti-CHIKV antibodies from humans and mice have low cross-neutralization against MAYV infection. After secondary MAYV infection in mice, high titers of antibodies with cross-neutralizing activity were observed. Depletion of immune system cells indicated that a combination of adaptive immune cells might be relevant in cross-protection against MAYV. The reduction of pro-inflammatory cytokines, NK cells, and inflammatory monocytes during MAYV secondary infection, suggests a role for innate immunity in cross-protection. In Part II, we have an analysis of antigenic epitopes of Zika virus E protein, where it was possible to: Select 9 linear ZIKV protein E peptides based on the cross-reactivity of anti-DENV IgG antibodies. Of the 9 peptides selected, 3 belong to the domain I (DI) of protein E, 2 to DII, supposedly being genus or group-specific, and 4 peptides to DIII, probably corresponding to immunodominant antigenic regions of flaviviruses. Serum cross-neutralization of sera from convalescent DENV patients against ZIKV infection unrelated to the 9 linear peptides mentioned was observed. Most of the DENV convalescent sera exhibited ADE, potentiating a ZIKV infection and, when depleting those with affinity for the 9 linear peptides selected, they reduced their activity, suggesting that antibodies against such peptides may be involved in ADE for ZIKV. In Part III, we have an evaluation of the immune response to the inactivated SARS-CoV-2 vaccine, where we conclude that: Individuals vaccinated twice with the inactivated SARS-CoV-2 seroconvert, producing high levels of IgG anti-Spike (S) viral protein with significant neutralizing capacity, which is significantly reduced against SARS-CoV-2 and the Gamma and Zeta variants after 6 months. Antibodies from animals vaccinated by the inactivated virus vaccine were less neutralizing against the Gamma and Zeta variants of SARS-CoV-2, but with a significant increase 3 days after infection. The stimulation of mouse splenic cells from immunized mice with SARS-CoV-2 inactivated antigens induced lymphoproliferation even to the antigens of the different viral variants. Prior immunization with inactivated SARS-CoV-2 reduced the burden of Gamma and Zeta variants in mouse lungs by almost 100 times. Undetectable levels of IL-6 and TNF-α in mice immunized after infection with WT strains and the Zeta variant were associated with a reduction in the pulmonary inflammatory infiltrate, evidencing a protective role of previous immunization by inactivated SARS-CoV-2.

A infecção pelo vírus da chikungunya (CHIKV) em humanos é geralmente caracterizada por um breve viremia, seguida de um rápido clearence viral. Apesar do quase imediato controle viral sistêmico, estudos sugerem a persistência do CHIKV em humanos e primatas não-humanos em reservatório imunológicos. A fase crônica da infecção por CHIKV e pelo vírus do Ross River (RRV), alfavírus artritogênicos, possui características similares à desregulação da reposta imune observada na artrite reumatoide (AR). O desenvolvimento de uma poliartralgia persistente em número considerável de pacientes possui um grande impacto social e econômico. Assim, potencialmente, a permanência viral pode estar relacionada com a manutenção da resposta inflamatória exacerbada por longos períodos. Sabe-se que a migração de células T CD4+ para a cápsula articular ocorre durante a primeira semana de infecção, e correlaciona-se negativamente com o pior prognóstico da chikungunya. Contudo, pouco se sabe sobre o papel dos subtipos das células T na fisiopatologia da doença. Uma vez que não existiam modelos de camundongos que permitissem o estudo da artrite crônica induzida pelo CHIKV, nosso objetivo foi desenvolver e caracterizar a resposta de células T e persistência viral em um modelo animal crônico. Portanto, em nosso estudo, desenvolvemos um novo modelo animal para o estudo da chikungunya crônica, que é capaz de recapitular os sinais comumente observados na artrite crônica da chikungunya em humanos. Os nossos resultados demonstram a importância das células T CD4+ para a patogenia da doença crônica da chikungunya em camundongos, especificamente células Th17, dados corroborados em humanos. Alguns dados sugerem que a infecção por CHIKV regula negativamente a produção de citocinas essenciais na regulação de resposta de células Th17, tais como, IL-10, IL-27 e IL-35. Adicionalmente, nossos resultados demonstraram que, de forma dependente da expressão de IFITM3, os macrófagos e células sinoviais são reservatórios virais e passíveis a replicação viral de alfavírus artritogênicos. Macrófagos infectados mantém o microambiente extremamente inflamatório através da produção de citocinas inflamatórias como o IL-6, e quimiocinas como CCL5 e CCL20, estimulando um maior recrutamento de infiltrado de células T na sinovias infectadas com o CHIKV, mesmo após o controle viral sistêmico. Em conclusão, esses dados demonstram que no nosso modelo animal a artrite crônica induzida por CHIKV é dependente da sinalização de IL-17 pelas células Th17, e que a persistência viral em macrófagos sinoviais mantém o microambiente inflamatório propicio para a manutenção do recrutamento das células T para a região sinovial. Chikungunya virus (CHIKV) infections in humans are usually characterized by brief viremia followed by rapid viral clearance. Despite the almost immediate systemic viral control, studies suggest the persistence of CHIKV in humans and non-human primates in immunological reservoirs. The chronic phase of infection by CHIKV and by Ross River virus (RRV), arthritogenic alphaviruses, has characteristics similar to the dysregulation of the immune response seen in rheumatoid arthritis (RA). The development of persistent polyarthralgia in a considerable number of patients has a significant social and economic impact. Thus, potentially, viral permanence may be related to the maintenance of the exacerbated inflammatory response for long periods. It was previously demonstrated that the migration of CD4+ T cells to the joint capsule occurs during the first week of infection, and it correlates with the worst prognosis of chikungunya. However, little is known about the role of T cell subtypes in the pathophysiology of the disease. Since there were no mouse models that would allow the study of CHIKV-induced chronic arthritis, we aimed to develop and characterize a new chronic animal model, followed by its T cell response and viral persistence. Therefore, in our study, we developed a new animal model for the chronic chikungunya understanding. Our model can induce the signs commonly seen in chronic chikungunya arthritis in humans. Our results demonstrate the importance of CD4+ T cells for the pathogenesis of chronic chikungunya disease in mice, specifically Th17 cells, data corroborated in humans. Some data suggest that CHIKV infection negatively regulates the production of essential cytokines in the regulation of Th17 cell response, such as IL-10, IL-27, and IL-35. Additionally, our results demonstrated that macrophages and synovial cells, in an IFITM3 expression-dependent manner, are viral reservoirs and susceptible to viral replication of arthritogenic alphaviruses. Infected macrophages maintain the extremely inflammatory microenvironment through the inflammatory cytokines production such as IL-6, and chemokines such as CCL5 and CCL20, stimulating greater recruitment of T cell infiltrate in CHIKV-infected synovia, even after systemic viral control. In conclusion, these data demonstrate that in our animal model CHIKV-induced chronic arthritis is dependent on IL-17 signaling by Th17 cells and that viral persistence in synovial macrophages maintains the inflammatory microenvironment conducive to the maintenance of T cell recruitment to the synovial region.

O vírus Oropouche (OROV) pertence à ordem Bunyavirales, família Peribunyaviridae, gênero Orthobunyavirus, e está entre as mais frequentes causas de arbovirose no Brasil. OROV é endêmico da região Amazônica, onde já causou mais de meio milhão de casos em surtos publicados, no entanto recentemente tem ganhado atenção devido à ocorrência de infecções em seres humanos e primatas não humanos fora daquela região. OROV é transmitido pela picada do vetor Culicoides paraensis e causa doença febril aguda. OROV tem genoma composto por três segmentos de RNA (L, M e S) que codificam as proteínas estruturais, bem como duas não estruturais: NSm e NSs. A proteína NSs é um fator de virulência reconhecido em alguns vírus do gênero Orthobunyavirus, mas quase nada se sabe sobre funções da proteína NSm. Evidências obtidas com outros vírus da família mostram que NSm tem papel em processos de montagem e morfogênese de partículas virais. Todavia, não existem conhecimentos sobre funções da NSm de OROV. A partir do desenvolvimento deste trabalho foi possível conhecer que a proteína NSm de OROV não é essencial à produção de progênie viral, mas está envolvida com a montagem do vírus pela via canônica no complexo de Golgi. A ausência de NSm modifica o recrutamento de organelas envolvidas em fábricas virais, com diminuição da área dessas fábricas, atrasando a produção de progênie infecciosa durante a fase exponencial de replicação. Outra importante descoberta é que NSm está envolvida na atenuação da virulência de OROV in vivo, já que na sua ausência OROV é altamente letal em modelo murino. O contrário foi observado para o vírus deletado da proteína NSs, que é menos letal que o OROVwt. Mais ainda, OROV deletado simultaneamente de NSm e NSs é quase completamente atenuado. OROV sem NSs e NSm infectam células do sangue periférico humano das linhagens mielóide, de modo similar ao OROVwt, mas com produção alterada da citocina TNF-? sugerindo que essas proteínas não estruturais estão envolvidas na patogenicidade de OROV. A expressão heteróloga de NSm demonstra que NSm sozinha tem distribuição diferente da NSm do vírus nativo, indicando que a expressão de elementos da poli-proteína M viral é essencial para o seu direcionamento final. Em conjunto, essas observações mostram que NSm de OROV está envolvida com a montagem de fábricas virais e com a morfogênese viral, e tanto NSm quanto NSs estão relacionadas com a patogenicidade do vírus em modelo murino. O mutante de OROV com deleção de ambas as proteínas é significantemente atenuado, e pode ser uma importante ferramenta para o desenvolvimento de vacina. Esses dados inéditos contribuem para o conhecimento de OROV e podem auxiliar na descoberta de possíveis alvos terapêuticos. Oropouche virus is classified in Bunyavirales order, family Peribunyaviridae, genus Orthobunyavirus and it\'s one of the most frequent causes of arboviruses in Brazil. OROV is endemic in the Amazon region, where it infected half million cases in documented outbreaks. However, it\'s recently gaining more attention due to an occurrence in humans and primates outside this region. OROV is transmitted by the bite of Culicoides paraensis vector and causes a febrile disease. Its genome is composed of three negative RNA strands (L, M and S) that codes the structural and non-structural viral proteins: NSm and NSs. NSs is a known virulence factor in the Orthobunyavirus genus, but nearly nothing is known about NSm function. Evidence obtained with other orthobunyaviruses showed that NSm is involved with viral particle assembly and morphogenesis. But there are not information\'s about OROVs NSm function. With the development of this work it was possible to discover that NSm protein it is not essential for progeny production, but it is involved with the canonical assembly pathway in Golgi complex. The absence of NSm modifies organelles recruitment to viral factories, diminishing viral factories area, affecting progeny production in the exponential growth phase in the replication curve. Another important discovery is that NSm is a virulence attenuator factor since the OROV NSm mutant is highly pathogenic in a mouse model. The opposite was observed in OROV NSs mutant, being less lethal compared to OROV wild type. Moreover, OROV mutant lacking both non-structural proteins infect human peripheral blood cells from myelocytic lineage, in similar ways of OROV wild-type, but with altered TNF-a cytokine production, suggesting that these proteins are involved with OROV pathogenicity. Heterologous NSm expression demonstrated that its coding region by itself it is not functional, indicating that is necessary to express elements from the M polyprotein to the correct destination. Taking together, these observations show that OROVs NSm is involved with viral factories assembly and viral morphogenesis, and both NSm and NSs are related to pathogenicity in a mouse model. OROV mutant lacking both non-structural proteins its highly attenuated in vivo, and it can be an essential tool to develop a vaccine to this virus. These new data contribute to the discovery of possible therapeutic targets in treating this virus.

As doenças causadas pelos vírus da chikungunya (CHIKV), dengue (DENV) e Zika (ZIKV), principalmente na fase aguda, induzem sintomatologias muito semelhantes entre si. O CHIKV pertence ao gênero Alphavirus da família Togaviridae, enquanto os DENV e ZIKV pertencem ao gênero Flavivirus da família Flaviviridae. A rápida implementação de tratamento, especialmente nos pacientes com dengue, diminui drasticamente o número de casos fatais. O objetivo deste trabalho foi desenvolver métodos de transcrição reversa seguida da reação em cadeia da polimerase (RT-PCR) em tempo real para diagnóstico diferencial e precoce de chikungunya, dengue e Zika. Sondas e primers específicos para CHIKV, ZIKV e os quatro sorotipos de DENV (DENV-1 a -4) foram desenhados a partir dos primeiros nucleotídeos da extremidade 5\' dos genomas virais para amplificação de fragmentos com 150, 185 e 170 pares de bases para DENV, ZIKV e CHIKV, respectivamente. Diversos protocolos foram realizados para otimização da reação de RT-PCR em tempo real considerando as concentrações de sondas, íon magnésio e temperatura de anelamento de sondas e primers. Análises de reação cruzada mostraram que os primers e as sondas selecionadas foram altamente específicos para os vírus em questão, sem reconhecimento cruzado de outros flavivírus e alfavírus, com exceção das sondas para os DENV-3 e DENV-4 que apresentaram reação cruzada com outros sorotipos. A sonda para DENV-3 reconheceu cruzadamente o DENV-1 e a sonda para DENV-4 o DENV-2. O limite de detecção da RT-PCR para dengue com sondas foi de 9,90x104, 2,42x102, 6,55x106 e 3,70x106 cópias/mL para os DENV-1 a -4, respectivamente. Contudo, o limite de detecção da RT-PCR para dengue com SYBR Green foi de 9,90x101, 2,42x101, 6,55x103 e 3,70x101 cópias/mL para os DENV-1 a -4, respectivamente, inferior àquela utilizando sondas. Os limites de detecção da RT-PCR com sondas para o ZIKV e o CHIKV foram de 1,25x100 e 5,25x100 PFU/mL, respectivamente, similares aos resultados encontrados quando realizada a RT-PCR com SYBR Green para estes mesmos vírus que foram de 1,25x101 e 5,25x100 PFU/mL, respectivamente. Amostras de soro de indivíduos assintomáticos e amostras de soro, urina ou saliva armazenadas no biorrepositório \"Banco de amostras de arbovírus\" (FCFRP Nº 006) do Laboratório de Virologia da FCFRP-USP, foram testadas com a RT-PCR com SYBR Green para dengue, observando-se uma sensibilidade de 54%, especificidade de 100%, valores preditivos positivo e negativo de 100% e 75%, respectivamente. Entretanto, quando avaliadas apenas as amostras de soro, observou-se uma sensibilidade de 82%, especificidade de 100%, valores preditivos positivo e negativo de 100% e 71% respectivamente, sugerindo que as amostras de soro são as mais indicadas para utilização no diagnóstico da dengue. Estas amostras também foram analisadas com as RT-PCRs com sondas para os vírus chikungunya e Zika, sendo todas negativas. As amostras detectadas com a RT-PCR com SYBR Green para dengue foram analisadas com a RT-PCR com sondas, sendo observada uma concordância de mais de 90% com os sorotipos previamente identificados. Conclui-se, portanto, que a detecção diferencial dos vírus é eficaz e fiel em uma única etapa e o método possui alta especificidade e sensibilidade Diseases caused by the viruses chikungunya (CHIKV), dengue (DENV) and Zika (ZIKV), specially in the acute phase, induced very similar symptoms. The CHIKV belongs to de genus Alphavirus, family Togaviridae, while the DENV and ZIKV belong to the genus Flavivirus, family Flaviviridae. The aim of this work was to develop a real time reverse transcription followed by polymerase chain reaction (RT-PCR) for early and differential diagnosis of chikungunya, dengue and Zika. Specific primers and probes for CHIKV, ZIKV and the four DENV (DENV-1 a -4) serotypes were designed from the first nucleotides on the 5\'end of the viral genomes to amplify fragments 150, 185, 170 base pairs fragments for DENV, ZIKV and CHIKV, respectively. Several protocols were analyzed for the optimization of the real time RTPCR, considering probes concentrations, magnesium ion and the primers and probes annealing temperature. Cross-reactivity tests showed that the selected primers and probes were highly specific to the target virus without recognition of other flaviviruses and alphaviruses, except for the probes for DENV-3 and DENV-4, which showed cross-reaction with other serotypes. The probe for DENV-3 cross-reacted with DENV-1 and the probe DENV-4 with DENV-2. The limit of detection of the RT-PCR with probes for dengue was 9,90x104, 2,42x102, 6,55x106 and 3,70x106 copies/mL for DENV-1, DENV-2, DENV-3 and DENV-4, respectively. However, the limit of detection of the RT-PCR with SYBR Green for dengue was 9,90x101, 2,42x101, 6,55x103 and 3,70x101 copies/mL for DENV-1, DENV-2, DENV-3 and DENV-4 respectively. The limit of detection of the RT-PCR with probes for ZIKV and CHIKV was 1,25x100 and 5,25x100 PFU/mL, respectively, similar to that found for the RT-PCR with SYBR Green, which was 1,25x101 e 5,25x100 PFU/mL for ZIKV and CHIKV, respectively. Serum samples from asymptomatic participants and serum, urine and saliva samples stored at the samples biorepository named \"Banco de amostras de arbovírus\" (FCFRP Nº 006) of the FCFRP-USP Virology laboratory were tested with RT-PCR with SYBR Green for dengue and showed a 54% sensitivity, 100% specificity and 100% and 75% positive and negative predictive values, respectively. However, when only serum samples were analysed, it showed 82% sensitivity, 100% specificity and 100% and 71% positive and negative predictive values, respectively, suggesting that the serum samples are more suitable for dengue diagnosis. Those samples were also analyzed with the RT-PCR with probes for CHIKV and ZIKV, being all negatives. The samples detected with the RT-PCR with SYBR Green for dengue were analyzed with the RTPCR with probes, showing to detect the expected serotype in more than 90% of them. Therefore, it\'s concluded that differential virus detection is effective and faithful in a single step and the method has high specificity and sensitivity

A Dengue é uma doença viral sistêmica, transmitida por mosquitos, que afeta anualmente cerca de 100 milhões de pessoas em todo o mundo. Causada por quatro sorotipos do vírus da Dengue (DENV), suas manifestações clínicas podem variar de assintomáticas à formas que podem levar a óbito. Curiosamente, os pacientes com Dengue apresentam uma resposta exacerbada das células secretoras de anticorpos (ASCs) no sangue cerca de sete dias após o início dos sintomas. A frequência dessas ASCs induzidas pelo DENV representa mais de 50% de todas as células B circulantes no sangue. Essa quantificação é maior que aquelas encontradas em outras infecções virais, contextos de imunização e até mesmo em pacientes com neoplasias de ASCs. Além disso, a magnitude dessa resposta transitória se correlaciona com a gravidade da doença. Então, como a infecção pelo DENV induz essa resposta enorme? Para responder à essa pergunta, combinamos abordagens in vitro e in silico. Células mononucleares do sangue periférico (PBMCs) obtidas de indivíduos saudáveis foram cultivadas in vitro durante sete dias na presença do DENV ou mitógenos. Após a estimulação pelo DENV, as células B presentes nas PBMCs foram capazes de se diferenciarem em ASCs, tanto fenotipicamente quanto funcionalmente, em magnitude similar àquelas estimuladas com mitógenos. Essa diferenciação demonstrou ser dependente da presença de outras células contidas nas PBMCs, assim como do contato célula-célula. Embora ambos os estímulos tenham sido capazes de induzir a diferenciação de ASCs, eles diferiram metabolicamente e transcricionalmente. PBMCs estimuladas pelo DENV apresentaram um maior consumo de triptofano, associado à maior expressão de IDO1 e IDO2 e maior síntese de quinurenina, bem como maiores expressões de IL-10, BAFF e SYK. Ainda, as concentrações de quinurenina foram positivamente correlacionadas com a enumeração de ASCs nessas culturas. Dados de transcriptoma públicos de pacientes com Dengue também suportam esses achados. Outros flavivírus, como o vírus Zika e a cepa vacinal da Febre Amarela não foram capazes de induzir a mesma magnitude de diferenciação das células B em ASCs in vitro. Tão pouco apresentaram correlação entre a enumeração de ASCs e a síntese de quinurenina. Por fim, através da construção de uma hipotética via de diferenciação de células B em ASCs durante infecção pelo DENV, através da combinação de dados da literatura e transcriptomas públicos, demonstramos que moléculas relacionadas à via do STAT3 (IL-10, IL-6, IRF4 e BLIMP1) estão mais expressas nos pacientes infectados e moléculas que respondem aos sinais de cálcio (Calcineurina, NFATC1, DOK3 e GRB2) estão menos expressas nos pacientes infectados. Esses dados proporcionam um melhor entendimento da resposta de células B durante a infeção pelo DENV, particularmente sobre como o metabolismo e a sinalização das células B estão conectados nesse processo. Dengue is a mosquito-borne viral disease that affects annually about 100 million people worldwide. Caused by four Dengue virus (DENV) serotypes, it ranges from asymptomatic to life threatening forms. Curiously, Dengue patients present an exacerbated blood antibody-secreting cell (ASCs) response around seven days after the symptoms onset. The frequency of those DENV-induced ASCs represents more than 50% of all circulating blood B cells. This is greater than found in others viral infections, immunization contexts and even in ASCs related-leukemia patients. Moreover, the magnitude of that transitory response correlates with the disease severity. So, how does the DENV infection induce this enormous response? In order to answer this question we have combined in vitro and in silico approaches. Peripheral blood mononuclear cells (PBMC) obtained from healthy individuals were cultured in vitro during seven days in the presence of DENV or mitogens. Upon the DENV stimulation, PBMC-contained B cells were able to differentiate phenotypically and functionally into ASCs, both phenotypically and functionally, in a similar magnitude than mitogen-stimulated cells. This differentiation was demonstrated to be dependent of the presence of the remaining PBMCs, as well as of the cell-cell contact. Although both stimuli were able to induce the ASCs differentiation, they differed metabolically and transcriptionally. DENV-stimulated PBMCs showed higher tryptophan consumption, associated with higher IDO1 and IDO2 expression and higher kynurenine synthesis, as well as higher IL-10, BAFF and SYK expressions compared to mitogen-exposed counterparts. Additionally, the kynurenine concentrations were positively correlated with the ASCs-enumeration in those cultures. Public transcriptome data supports these findings as well. Other flaviviruses, such as Zika virus and the attenuated vaccine Yellow Fever were not able to induce the same magnitude of ASCs differentiation in vitro. Hence, they did not present a correlation between the number of generated ASCs and the supernatant kynurenine levels. Based on the combination of the literature and public transcriptome data, we have constructed a hypothetical B cell differentiation pathway that might be occurring during DENV infection. It displays that STAT3 pathway-related molecules (IL-10, IL-6, IRF4 and BLIMP1) are more expressed in Dengue patients and molecules that respond to calcium signals (Calcineurin, NFATC1, DOK3 and GRB2) are less expressed in Dengue patients than in control. These data provide a better understanding of the B cell response elicited by DENV infection, particularly about how the B cell metabolism and signaling can be connected into this process.

O Zika vírus (ZIKV) emergiu como problema de saúde mundial e ainda demanda esforços da comunidade científica para o entendimento dos mecanismos moleculares envolvidos em sua infecção na célula hospedeira. O ZIKV faz parte do gênero dos Flavivirus, como o vírus da Dengue, cujo mecanismo de invasão celular é descrito na literatura. Neste, é evidenciado o envolvimento de receptores de membrana da família TAM, como Tyro3, Axl, e Mer, assim como de seus ligantes solúveis, Gas6 e Proteína S num fenômeno chamado de mimetismo apoptótico. Nesse mecanismo a fosfatidilserina presente no envelope viral interage com os receptores TAM e, assim, resulta na internalização da partícula. Estudos conduzidos pelo nosso grupo demonstram uma possível relação entre a expressão aumentada dos receptores TAM e a infecção por ZIKV, bem como a suscetibilidade da linhagem de camundongos SJL à infecção. Entretanto, os mecanismos moleculares dependentes de receptores TAM na infecção por ZIKV ainda não são compreendidos por completo. Objetivo. Avaliar a relevância dos receptores TAM na infecção por ZIKV em modelos experimentais in vitro e in vivo. Resultados e Discussão. Observamos que animais SJL, susceptíveis, possuem níveis de expressão de Tyro3, Axl e Gas6 elevados em comparação aos C57BL/6, resistentes. Neste contexto, demonstramos que a combinação ZIKV&#43 rmGas6 leva ao aumento da carga viral no baço e no soro de SJL e C57BL/6. Contudo, o tratamento dos animais com bloqueador da função quinase de Axl, R428, não reduziu a quantidade de partículas virais. Interessantemente, verificamos que a infecção de prenhes C57BL/6 com ZIKV&#43rmGas6 permitiu o aparecimento de alterações fenotípicas nos fetos, o que não foi observado no controle com ZIKV puro. Os experimentos in vitro utilizando células dendríticas e macrófagos não apresentaram diferenças na replicação viral ou na liberação de Gas6 após a infecção, mesmo com a utilização de células nocaute. Conclusão. Em suma, nossos dados corroboram com a literatura em dois pontos distintos. O primeiro deles na dispensabilidade dos TAM para infecção in vitro de células imunocompetentes. O segundo, evidencia o papel dos receptores dependente da presença de Gas6 e na sindrome congênita. Introduction. Zika virus (ZIKV) has emerged as a global health problem and still demands efforts by scientific community to understand the molecular mechanisms involved in its infection in host cells. ZIKV belongs to Flavivirus genus, such as the Dengue virus, whose mechanism of cell invasion is elucidated in the literature. In this, it is clear the involvement of membrane receptors as TAM family, such as Tyro3, Axl, and Mer, as well as their soluble ligands, Gas6 and Protein S in a phenomenon called apoptotic mimicry. In this mechanism, phosphatidylserine exposed in the viral envelope interacts with TAM receptors and, thus, resulting in the internalization of viral particle. Studies conducted by our group demonstrated a possible relationship between increased TAM receptor expression and ZIKV infection, as well as the susceptibility of SJL mouse line to infection. Despite this, the molecular mechanisms related to TAM receptors in ZIKV infection are still not fully understood. Aim. Evaluate the relevance of TAM receptors in ZIKV infection in experimental models in vitro and in vivo Results and discussion. We observed that susceptible SJL animals present higher Tyro3, Axl and Gas6 expression levels compared to resistant C57BL/6. In this context, we demonstrated that the combination of ZIKV&#43rmGas6 in SJL and C57BL/6 mice leads to increased viral load in spleen and serum. Meanwhile, treatment of animals with Axl kinase blocker, R428, did not reduced the amount of viral particles. Interestingly, we found that C57BL/6 pregnancy infection with ZIKV&#43rmGas6 allowed the appearance of phenotypic changes in the fetuses, which was not observed in the ZIKV control. In vitro experiments using dendritic cells and macrophages showed no difference in viral replication or Gas6 release after infection, even with the use of Axl knockout cells. Conclusion. In short, our data corroborate with the literature in two distinct points. The first one in the dispensability of TAM for in vitro infection of immunocompetent cells. The second, evidences the role of TAM receptors in a dependent on the presence of Gas6.

Rinossinusite crônica (RSC), com ou sem pólipo nasal é uma inflamação crônica da mucosa nasal e seios paranasais, cujo tratamento muitas vezes pode ser a cirurgia. Vários vírus respiratórios são detectados em biópsias de pólipo nasal, concha média e mucosa do seio maxilar de pacientes com a doença, mas seu papel no desencadeamento dessa inflamação não é claro. O Objetivo do presente estudo teve como objetivo localizar e caracterizar in situ sítios de infecção por rinovírus (HRV), metapneumovírus (HMPV) e vírus sincicial respiratório (HRSV), detectar se há replicação dos mesmos, caracterizar as células, em tecidos de pólipo nasal, concha média e mucosa do seio maxilar removidas cirurgicamente de pacientes com rinossinusite crônica. Tecidos de 17 pacientes positivos por qPCR foram utilizados para identificar a localização e caracterização dos sítios de infecção, além de avaliar a atividade replicativa, onde proteínas de capsídeo viral foram detectadas por imunohistoquimica (IHQ). Para identificação das células infectadas pelos vírus estudados, foi realizada a técnica de SIMPLE. Os resultados revelaram positividade em 7 de 11 amostras positivas para HRV analisadas por PCR em tempo real, sendo encontrada a proteína viral em 5 amostras de pólipo nasal, uma amostras da mucosa do seio maxilar e uma em concha média. Cinco amostras tiveram seu genoma detectado no PCR em tempo real para HMPV, sendo que 3 delas tiveram marcação da proteína por IHQ, uma amostra positiva em cada tecido estudado. Em um total de 4 amostras, houve detecção da proteína F de HRSV em 3 delas, sendo 2 em pólipo nasal e uma concha média. Em todas as reações de IHQ foram incluídos controles positivos e negativos adequados. Depois da detecção das proteínas desses vírus respiratórios, as amostras tiveram seus fenótipos testados, células produtoras de muco e epiteliais ciliadas, mastócitos, células dendríticas, eosinófilos e linfócitos através da técnica SIMPLE. Foi demonstrado que há evidência de replicação de rinovírus, metapneumovírus e vírus sincicial respiratório no epitélio e no estroma tecidual de pólipo nasal, concha média e mucosa do seio maxilar. Foram detectadas células CD4+ e CD123+ infectadas por HRSV, evidenciadas pela marcação da proteína estrutural viral F. Adicionalmente, observamos células CD4+, CD14+ e CD20+, além de células residentes em glândulas produtoras de muco, infectadas com HRV utilizando anticorpo anti-VP1 viral. Para HMPV a infecção se limitou ao epitélio superficial dos tecidos e glândulas produtoras de muco. Esses dados revelam uma possível persistência de HRV, HRSV e HMPV em tecidos de pacientes com rinossinusite crônica, que podem assim serem reservatórios de vírus na ausência de sintomas de infecção aguda. Chronic rhinosinusitis (CRS) with or without nasal polyp is a chronic inflammation of the nasal mucosa and paranasal sinuses, whose treatment can often be surgery. Several respiratory viruses are detected in biopsies of nasal polyp, middle shell and maxillary sinus mucosa of patients with the disease, but their role in triggering this inflammation is unclear. The objective of the present study was to locate and characterize in situ sites of infection by rhinovirus (HRV), metapneumovirus (HMPV) and respiratory syncytial virus (HRSV), to detect if there is replication of the same, to characterize the cells in nasal polyp tissues , middle shell and maxillary sinus mucosa surgically removed from patients with chronic rhinosinusitis. Tissues from 17 patients positive for qPCR were used to identify the location and characterization of infection sites, and to evaluate the replicative activity, where viral capsid proteins were detected by immunohistochemistry (IHC). To identify the cells infected by the viruses studied, the SIMPLE technique was performed. The results revealed positivity in 7 of 11 HRV positive samples analyzed by real-time PCR. Viral protein was found in 5 nasal polyp samples, one in the maxillary sinus mucosa and one in the middle shell. Five samples had their genome detected in the realtime PCR for HMPV, 3 of them had protein marking by IHC, a positive sample in each tissue studied. In a total of 4 samples, HRSV F protein was detected in 3 of them, 2 in nasal polyp and 1 medium concha. All positive and negative controls were included in all IHC reactions. After the detection of the proteins of these respiratory viruses, the samples had their tested phenotypes, mucus-producing cells and ciliated epithelial cells, mast cells, dendritic cells, eosinophils and lymphocytes through the SIMPLE technique. It has been demonstrated that there is evidence of rhinovirus, metapneumovirus and respiratory syncytial virus replication in the epithelium and in the tissue stroma of the nasal polyp, middle concha and maxillary sinus mucosa. CD4 +, CD14 + and CD20 + cells were detected, as well as cells residing in mucus-producing glands, infected with HRV using anti-viral VP1 antibody. For HMPV the infection was limited to the superficial epithelium of the tissues and mucus producing glands. These data reveal a possible persistence of HRV, HRSV and HMPV in tissues of patients with chronic rhinosinusitis, who may thus be virus reservoirs in the absence of symptoms of acute infection.

O vírus ZIKA pertence ao gênero Flavivírus e diversos trabalhos têm demonstrado que a infecção por esse vírus está associada com o desenvolvimento de anomalias congênitas fetais e doenças neurológicas como casos de microcefalia e de síndrome de Guillan-Barré. Com intuito de entender os mecanismos imunológicos de controle da replicação viral e na patogênese da doença, o objetivo do trabalho foi estabelecer um modelo experimental de infecção por vírus ZIKV em animais imunocompetentes e investigar a migração de leucócitos para o sistema nervoso central durante a infecção por esse vírus. Os animais C57BL/6 infectados por ZIKV apresentaram um aumento da carga viral no sistema nervoso central (SNC) após 24 horas de infecção e uma redução significativa após 48 e 72 horas de infecção. No entanto, os animais deficientes do receptor de IFN tipo I (IFNAR-/-) apresentaram uma alta carga viral após 48 e 72 horas de infecção. Nossos dados mostram que os animais IFNAR-/- são altamente suscetíveis a infecção com 100% de mortalidade na fase aguda da doença, enquanto os animais C57BL/6 são resistentes a infecção com 100% sobrevida por mais de 30 dias de infecção. A infecção com ZIKA induziu o recrutamento de duas subpopulações de monócitos CD11b+Ly6Chi e CD11b+Ly6Clo para o SNC de maneira dependente da sinalização de IFN tipo I. Uma vez que, os animais C57BL/6 apresentaram um aumento de monócitos no SNC após 48 horas de infecção, enquanto que os animais IFNAR-/- apresentaram o recrutamento dessas células somente após 72 horas de infecção. De maneira interessante, o recrutamento dos monócitos culminou com um aumento da expressão das citocinas IL-6 e IL-18 no SNC de animais C57BL/6, enquanto que a expressão dessas citocinas não foi encontrada no SNC de animais IFNAR-/-, sugerindo a importância da via de sinalização de IFN tipo I para a produção dessas citocinas. E por fim, demostramos que a migração dos monócitos para SNC de animais infectados com vírus ZIKA é dependente da produção de CCL2 produzida no cérebro de animais infectados e da expressão dos receptores de quimiocinas CCR2 e CCR5. Uma vez que, animais deficientes para CCR2 (CCR2-/-) e CCR5 (CCR5-/-) apresentaram uma redução da frequência dos monócitos CD11b+Ly6Chi para o SNC em comparação aos animais C57BL/6 após a infecção. De maneira geral, nossos dados mostram que os animais C57BL/6 são resistentes à infecção por ZIKV, uma vez que a via de sinalização de IFN do tipo I está envolvida no recrutamento de monócitos inflamatórios para SNC e consequente controle da replicação viral. O desenvolvimento de um modelo experimental de infecção por vírus ZIKV abre perspectivas para o entendimento dos mecanismos de controle da replicação viral e sobre a patogênese da doença. The ZIKA virus belongs to the genus Flavivirus, several studies have demonstrated that the infection by this virus is associated with the development of fetal congenital anomalies and neurological diseases, such as microcephaly and Guillan-Barré syndrome. In order to elucidate the immunological mechanisms of the viral replication control and the disease pathogenesis, the study aim was to establish an experimental model of ZIKV infection in immunocompetent animals and investigate the leukocytes migration to the central nervous system during this virus infection. ZIKV infected C57BL/6 animals demonstrated an increased viral load in the central nervous system (CNS) after 24 hours of infection and a significant reduction after 48 and 72 hours of infection. However, type I INF receptor deficient mice (IFNAR-/-) had a high viral load after 48 and 72 hours of infection. Our data demonstrated that IFNAR-/- animals are highly susceptible to infection with 100% mortality during the acute phase of the disease, while C57BL/6 mice are resistant with 100% survival for more than 30 days of infection. ZIKA infection induced the recruitment of two monocytes subpopulation CD11b+Ly6Chi and CD11b+Ly6Clo to the CNS in a dependent manner of type I INF signaling. C57BL/6 mice showed an increase of monocytes in the CNS after 48 hours of infection, whereas IFNAR-/- mice recruited these cells only after 72 hours of infection. Interestingly, the monocytes recruitment culminated in an increased expression of IL-6 and IL-18 in C57BL/6 mice CNS, while these cytokines expression was not observed in the CNS of IFNAR-/- animals, thus suggesting the importance of type I INF signaling pathway for these cytokines production. Finally, we have demonstrated that the monocytes migration to the CNS from animals infected with ZIKA virus is dependent of CCL2 production in the brain of infected animals, as well as the expression of the chemokine receptors CCR2 and CCR5. Given that CCR2 (CCR2-/-) and CCR5 (CCR5-/-) deficient animals demonstrated a frequency reduction of CD11b+Ly6Chi monocytes to the CNS compared to C57BL/6 mice after infection. Overall, our data demonstrate that C57BL/6 animals are resistant to ZIKV infection, since the type I INF signaling pathway is involved in the inflammatory monocytes recruitment to the CNS and subsequent viral replication control. The development of an experimental model of ZIKV infection open perspectives for the understanding of viral replication control mechanisms and disease pathogenesis.

Hantavirus é um gênero na família Hantaviradae, incluindo agentes polimórficos, envelopados e com genoma de RNA fita simples, polaridade negativa e trissegmentado. Os Hantavírus são zoonóticos e mantidos na natureza em reservatórios das ordens Rodentia, Soricomorpha e Chiroptera. No Brasil, foram descritos 8 Hantavírus e destes, 6 causam doença humana grave e de alta letalidade, a Síndrome Pulmonar e Cardiovascular, sendo dentre eles o Araraquara (ARQV) o vírus de ocorrência nas regiões de Cerrado do Sudeste (incluindo Ribeirão Preto) e Planalto Central. ARQV destaca-se por produzir a maior letalidade dentre todos os Hantavírus existentes e possuir como reservatório o roedor silvestre Necromys lasiurus que o transmite ao homem por inalação dos aerossóis contaminados de suas excretas. Neste trabalho, detectamos genoma de Hantavírus em tecidos de Necromys lasiurus e em tecidos de morcego Desmodus rotundus, ambos capturados no Nordeste do Estado de São Paulo. Destes animais, foram feitas tentativas de isolamento de Hantavírus a partir de amostras de tecido de Necromys lasiurus e Desmodus rotundus. Os procedimentos foram realizados em laboratório de nivel de biossegurança 3, onde fizeram-se as inoculações em cultura de células VERO E6, com detecção viral após uma única passagem de 4 dias. Desta forma, foi possível isolar um Hantavírus a partir de sobrenadante do lisado de coração do roedor, o que foi confirmado pela presença do genoma viral do isolado por RT-PCR do sobrenadante de cultura celular e dos antígenos virais nas células Vero E6, por imunofluorescência indireta e western blot. O processo de isolamento mostrou-se reprodutível, por 3 vezes, para o mesmo tecido do roedor. Tais resultados encorajam que esta metodologia para isolamento de Hantavírus deva ser experimentada por mais vezes. O Hantavírus isolado (provavelmente ARQV) deverá fornecer importantes informações sobre seu genoma completo, além de propiciar vários estudos futuros. Hantavirus is a genus in the Hantaviradae family, including polymorphic agents, it is enveloped and with a single stranded RNA genome, negative and tri-polarity polarity. Hantaviruses are zoonotic and kept in nature in reservoirs of the orders Rodentia, Soricomorpha and Chiroptera. There are 8 hantaviruses recorded in Brazil, 6 of them cause severe human disease and high lethality, a Pulmonary and Cardiovascular Syndrome, among which Araraquara (ARQV) is the virus that occurs in the Southeastern Cerrado (including Ribeirão Preto) and Central Plateau. ARQV stands out for producing higher lethality among all existing hantaviruses and has as reservoir the wild rodent Necromys lasiurus, which transmits to humans by inhalation of the contaminated aerosols of their excreta. In this work, we detected the genome of hantavirus in Necromys lasiurus and Desmodus rotundus, bat tissues, both captured in the Northeast of the State of São Paulo. Of these animals, attempts were made to isolate hantavirus from tissue samples of Necromys lasiurus and Desmodus rotundus. The laboratory tests of biosafety level 3, where inoculations in culture of VERO cells E6 were done, with viral detection after a single passage of 4 days. Thus, it was possible to isolate hantavirus from the rodent heart lysate supernatant, which was confirmed by the presence of the viral genome of the isolate by RT-PCR of the cell culture supernatant and the viral antigens in the Vero E6 cells by Indirect Immunofluorescence and Western Blot. The isolation process was reproducible, 3 times, for the same tissue of the rodent. These results indicate that this methodology for hantavirus isolation should be tested more often. The isolated Hantavirus (ARQV), should provide important information about its complete genome, as well as providing several future studies.

O vírus Oropouche (OROV) é um arbovírus que ocorre na região amazônica causando surtos de doenças febris agudas e que, ocasionalmente, podem ser associados a meningoencefalite. Aproximadamente 500.000 casos de Oropouche teriam ocorrido no Brasil. Entretanto, não existe vacina contra o OROV. O objetivo deste trabalho foi desenvolver um modelo animal de infecção por OROV para estudar a patogênese da doença e um modelo para testar candidatas vacinais. Protótipo vacinal utilizando a proteína recombinante do nucleocapsídeo (N) de OROV (NrOROV), que é o principal antígeno viral, foi usado como potencial candidato para vacina. Neste estudo utilizou-se um modelo animal em camundongos Balb/c de 12 semanas de idade, inoculados intracerebralmente com 8x105 PFU de OROV, capaz de induzir 100% de letalidade após o terceiro dia da infecção. Altos títulos virais foram encontrados no cérebro e na medula espinhal dos animais. Surpreendentemente, 12 e 24 horas pós-infecção foi possível detectar vírus no fígado e baço (3 Log10 PFU/g) dos camundongos. Com este modelo foram testados os candidatos vacinais. Grupos de camundongos foram imunizados 3 vezes com OROV, OROV e FCA, NrOROV, NrOROV e FCA, NrOROV, Poli I:C e Montanide ISA 720. Após 3 imunizações, os animais foram desafiados com 10 LD50 de OROV e observados por 20 dias. Os animais imunizados com NrOROV e adjuvantes, não foram capazes de produzir anticorpos neutralizantes e adquirir imunidade protetora contra OROV enquanto que os imunizados com OROV apresentaram altos níveis de anticorpos neutralizantes e completa proteção in vivo. Ainda, os anticorpos produzidos pelos animais imunizados permitiram estudar o ciclo de replicação celular do OROV utilizando imunofluorescência. Oropouche (OROV) is an arbovirus that occurs in the South American, Amazon region, producing outbreaks of acute febrile illness occasionally associated to meningoencephalitis. Approximately 500,000 cases of Oropouche have been reported in Brazil in the last 60 years. However, there is no available vaccine for OROV. We show here the development of an animal model of OROV suitable for studies on pathogenesis and vaccine testing. A vaccine prototype based on recombinant OROV nucleocapsid protein (NrOROV), an important viral antigen, was evaluated in the animal model. Initialy, we observed that all 12-week-old Balb/c mice inoculated intracerebrally with 8x105 PFU died after the third day of infection. Surprisingly, OROV genome was detectable in the liver as early as 12 hours post infection (pi) and in the spleen at 24 hours pi at 3 log10 PFU/g. Besides, high viral titers were found in brain and spinal cord. To test the NrOROV as a vaccine candidate, animals divided in 5 groups were immunized subcutaneously 3 times, two weeks apart with either OROV, OROV and Freud complete Adjuvant (FCA), NrOROV, NrOROV and FCA, NrOROV and Poly I:C and Montanide ISA 720. The experiment also included a group of naïve animals. After the third immunization, the animals were challenged with 10LD50 by intracerebral route and followed for 20 days. The animals immunized with NrOROV and adjuvants developed specific antibodies that were not able to neutralize the virus or confer protective immunity against OROV. Nevertheless, mice immunized with OROV showed high levels of neutralizing and protective antibodies. Despite the discouraging results with NrOROV as a vaccine, the mouse model is suitable to study pathogenesis, and to test other vaccines for OROV.

Por representar a mais importante arbovirose em nível mundial, as infecções causadas pelos vírus da dengue são de grande importância em nosso país, apresentando uma ampla variedade de sintomas clínicos que vão desde infecção assintomática até formas mais graves da doença. O título de anticorpos neutralizantes produzidos frente à infecção por dengue parece ser determinante na forma de apresentação da doença no paciente. Atualmente, a forma com que o teste de neutralização é realizado demanda tempo para sua realização. O objetivo deste trabalho foi padronizar um ensaio de neutralização viral por RT-PCR em tempo real em cepas virais dos quatro sorotipos dengue para, posteriormente, ser empregada na detecção rápida e em grande escala de anticorpos em soro de pacientes e de candidatos vacinais. Para isso, foram construídas curvas padrão, para cada sorotipo viral, por meio da transcrição in vitro do RNA viral. O ensaio de neutralização padronizado nesse estudo reduziu o número de dias de detecção da neutralização em 48 horas quando comparada com a técnica de neutralização tradicional (PRNT), além de utilizar técnicas moleculares sensíveis e específicas para detecção como a RT-PCR em tempo real que garantem maior aplicabilidade do teste. Because Dengue virus is the most important arboviral disease worldwide, infections caused by this pathogen are of great importance in Brazil, producing a wide variety of clinical symptoms ranging from asymptomatic infection to more serious forms of the disease. The title of neutralizing antibodies produced against the dengue infection appears to be determinant in the outcome of the disease. Currently, neutralization tests that have been performed take time for its execution. The aim of this study was to standardize a viral neutralization assay by real-time RT-PCR of viral strains of the four Dengue serotypes to subsequently be used for rapid detection and large-scale using antibodies of patients and for vaccine candidates. For this matter, standard curves were constructed for each Dengue serotype, by in vitro transcription of viral RNA. The neutralization assay in this study reduced the period of neutralization to 48 hours, compared to traditional neutralization test (PRNT), and it uses a more sensitive and specific molecular technique for detection of neutralizing antibodies, such as real time RT-PCR, to ensure greater applicability of the test

A abordagem do tema mudanças climáticas por dois jornais diários e duas revistas semanais de informação em 2006 e 2007 e a percepção deste tema por formadores de opinião constituem o tema central deste trabalho. Uma pesquisa de campo foi realizada com assinantes dos jornais Folha de S. Paulo e O Estado de S. Paulo e das revistas Veja e Época, com jornalistas desses veículos e com cientistas da área de mudanças climáticas. Mostrou-se elevado o interesse do público pelos temas de ciência, em geral, e meio ambiente em particular. Nos periódicos analisados foram detectados conceitos incorretos do ponto de vista técnico e a cobertura voltou-se principalmente às pesquisas que mostravam impactos das mudanças climáticas. Segurança energética, outro fator fortemente relacionado à questão climática, orientou a pesquisa exploratória em busca de aferir a relação percebida pelo público entre mudanças climáticas e energia nuclear. Também pode ser traçado um paralelo entre o colapso ambiental e a catástrofe nuclear, presente no imaginário popular desde as explosões das bombas atômicas em Hiroshima e Nagasaki. Considerando o papel das ciências de modo geral, é imprescindível destacar que a melhor pesquisa não pode estar dissociada da comunicação eficiente e eficaz, capaz de mobilizar os cidadãos e sensibilizar tomadores de decisão. O diálogo entre cientistas e público, saberes tradicionalmente compartimentados, pode e deve ser praticado e a imprensa tem um papel fundamental neste processo. The climate change approach by two daily newspapers and two weekly magazines in 2006 and 2007, and this theme perception by opinion-makers, constitute the major target of this work. A survey was conducted with subscribers to Folha de S. Paulo and O Estado de S. Paulo newspapers, Veja and Época magazines, with their journalists, as well as with climate change scientists. The survey showed that is equally high the public interest in general science subjects and in specific environmental themes. In the analyzed periodicals, some incorrect technical concepts were detected and the press coverage focused, mainly, on research into climate change impacts. Energy security, another factor strongly related to climate, was explored by the research to evaluate public view of a relation between climate change and nuclear energy. A parallel may be made between climate change and nuclear catastrophe, present in the popular imaginary, since the atomic bomb explosions in Hiroshima and Nagasaki. Considering the science role in general, it is essential to highlight the fact that the state of the art research should not be dissociated from efficient and effective communication, able to mobilize citizens and touch decision-makers. Although the dialogue between scientists and the public was considered, traditionally, as related to separated fields of awareness, it may be achieved and the media has a fundamental role in this process.
