4. Antibody-Free Glycogen Nanoparticles Engage Human Immune T Cells for Intracellular Delivery of Small Drugs or mRNA

Abstract

T cells play a major role in immune defense against viral infections and diseases such as cancer. Accordingly, developing nanoparticle (NP) systems to effectively deliver therapeutics to T cells is of interest. However, NP-mediated delivery of drugs to T cells is challenging because of the nonphagocytic nature of T cells. To engage T cells and induce cellular internalization, NPs are typically decorated with specific receptor-targeting antibodies, often using laborious and costly procedures. Herein, we report that natural glycogen NPs (i.e., nanosugars) with different sizes (20–80 nm) and surface charges (neutral and positively charged) engage Jurkat T cells, undergo intracellular trafficking, and release encapsulated drug without the use of receptor-targeting antibodies. Specifically, glycogen–resveratrol constructs are employed to reactivate HIV-1 latently infected Jurkat T cells (J-Lat A2) and trigger proviral expression. Both neutral and positively charged glycogen NPs engage with J-Lat A2 cells. Large (84 ± 29 nm) and positively charged (23 ± 5 mV) NPs, denoted phytoglycogen-ethylenediamine (PGEDA) NPs, readily associate with the cell membrane and are internalized (60%) in J-Lat A2 cells but remain confined in the endocytic vesicles, with moderate reactivation of latent HIV-1 (4.7 ± 0.5%). Conversely, small (21 ± 5 nm) and positively charged (10 ± 6 mV) NPs, bovine glycogen-EDA (BGEDA) NPs, associate slowly with T cells but show nearly 100% internalization and efficient endosomal escape properties, resulting in 1.5-fold higher reactivation of latent HIV-1 in T cells. PGEDANPs and BGEDANPs are also internalized by primary human T cells (>90% cell association) and enable the transfection of mRNA, with BGEDANPs showing a 2-fold higher transfection than PGEDANPs. This work highlights the potential of BGEDANPs for the effective intracellular delivery of small-molecule drugs and mRNA in T cells.

Abstract

Background: Calorie restriction (CR) retards aging and increases longevity in many animal models. However, it is unclear whether CR can be implemented in humans without adverse effects on body composition.Objective: We evaluated the effect of a 2-y CR regimen on body composition including the influence of sex and body mass index (BMI; in kg/m2) among participants enrolled in CALERIE-2 (Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy), a multicenter, randomized controlled trial.Design: Participants were 218 nonobese (BMI: 21.9-28.0) adults aged 21-51 y who were randomly assigned to 25% CR (CR, n = 143) or ad libitum control (AL, n = 75) in a 2:1 ratio. Measures at baseline and 12 and 24 mo included body weight, waist circumference, fat mass (FM), fat-free mass (FFM), and appendicular mass by dual-energy X-ray absorptiometry; activity-related energy expenditure (AREE) by doubly labeled water; and dietary protein intake by self-report. Values are expressed as means ± SDs.Results: The CR group achieved 11.9% ± 0.7% CR over 2-y and had significant decreases in weight (-7.6 ± 0.3 compared with 0.4 ± 0.5 kg), waist circumference (-6.2 ± 0.4 compared with 0.9 ± 0.5 cm), FM (-5.4 ± 0.3 compared with 0.5 ± 0.4 kg), and FFM (-2.0 ± 0.2 compared with -0.0 ± 0.2 kg) at 24 mo relative to the AL group (all between-group P < 0.001). Moreover, FFM as a percentage of body weight at 24 mo was higher, and percentage of FM was lower in the CR group than in the AL. AREE, but not protein intake, predicted preservation of FFM during CR (P < 0.01). Men in the CR group lost significantly more trunk fat (P = 0.03) and FFM expressed as a percentage of weight loss (P < 0.001) than women in the CR group.Conclusions: Two years of CR had broadly favorable effects on both whole-body and regional adiposity that could facilitate health span in humans. The decrements in FFM were commensurate with the reduced body mass; although men in the CR group lost more FFM than the women did, the percentage of FFM in the men in the CR group was higher than at baseline. Abstract

This study compares the utility of two measures of psychopathic traits, the Antisocial Features (ANT) scale of the Personality Assessment Inventory and the Psychopathy Checklist-Revised (PCL-R), to predict serious institutional misconduct among incarcerated sex offenders over a 2-year follow-up period. Archival disciplinary data for 58 offenders were classified as major infractions involving physical aggression, verbal aggression/acts of defiance, or nonaggressive offenses. Significant correlations were obtained between both measures of psychopathy and each type of disciplinary offense except physical aggression, the occurrence of which was rare in this sample. Regression analyses indicated that each measure accounted for unique--or incremental--variance in one of the criterion measures. Overall classification accuracy based on standard cut scores was somewhat more positive for ANT than for the PCL-R.

Abstract

Tri-ortho-cresyl phosphate (ToCP) is an anti-wear, flame retardant additive used in industrial lubricants, hydraulic fluids and gasoline. The neurotoxic effects of ToCP arise from the liver-activated metabolite 2-(o-cresyl)-4H-1,3,2-benzodioxaphosphoran-2-one (cresyl saligenin phosphate or CBDP), which inhibits esterase enzymes including butyrylcholinesterase (BChE). Following BChE adduction, CBDP undergoes hydrolysis to form the aged adduct ortho-cresyl phosphoserine (oCP-BChE), thus providing a biomarker of CBDP exposure. Previous studies have identified ToCP in aircraft cabin and cockpit air, but assessing human exposure has been hampered by the lack of a laboratory assay to confirm exposure. This work presents the development of an immunomagnetic-UHPLC-MS/MS method for the quantitation of unadducted BChE and the long-term CBDP biomarker, oCP-BChE, in human serum. The method has a reportable range from 2.0 ng/ml to 150 ng/ml, which is consistent with the sensitivity of methods used to detect organophosphorus nerve agent protein adducts. The assay demonstrated high intraday and interday accuracy (≥85%) and precision (RSD ≤ 15%) across the calibration range. The method was developed for future analyses of potential human exposure to CBDP. Analysis of human serum inhibited in vitro with CBDP demonstrated that the oCP-BChE adduct was stable for at least 72 h at 4, 22 and 37 °C. Compared to a previously reported assay, this method requires 75% less sample volume, reduces analysis time by a factor of 20 and demonstrates a threefold improvement in sensitivity. Published 2015. Abstract

Cancer is the second most common cause of death in the Western world. Yet despite extensive research into the disease, most improvements in survivability have been accredited to advances in early detection than advances in treatment. A problem with the treatment of the disease stems from the genetic individuality of any given cancer. While cells can become transformed with only a few mutations, their genetics evolves into an increasingly aberrant state in which it can be impossible to distinguish initial driver mutations from subsequent passenger mutations. But knowing the earliest events in oncogenesis may prove useful for the understanding and treatment of cancer. I have tried to discover some such events in a Drosophila model of epithelial transformation. In this model, known oncogenes were overexpressed in conjunction with a simultaneous knockdown in potential tumour suppressor genes. If a synergy existed, the epithelium being modified would no longer resemble the phenotype of the oncogene alone, but would be significantly larger and / or disorganised in its appearance. The work presented here begins with the previously discovered result that depletion of held out wings cooperates with epithelial growth factor receptor in a different Drosophila model - one specific for transformation leading to metastasis. From this point I discovered that held out wings synergises with the oncoproteins Yorkie and Notch in epithelia; and in an effort to uncover the nature of this synergy, I discovered that Yorkie also synergises with the depletion of myospheroid and blistery.

Abstract

Organophosphorus nerve agents (OPNAs) are toxic compounds that are classified as prohibited Schedule 1 chemical weapons. In the body, OPNAs bind to butyrylcholinesterase (BChE) to form nerve agent adducts (OPNA-BChE). OPNA-BChE adducts can provide a reliable, long-term protein biomarker for assessing human exposure. A major challenge facing OPNA-BChE detection is hydrolysis (aging), which can continue to occur after a clinical specimen has been collected. During aging, the o-alkyl phosphoester bond hydrolyzes, and the specific identity of the nerve agent is lost. To better identify OPNA exposure events, a high-throughput method for the detection of five aged OPNA-BChE adducts was developed. This is the first diagnostic panel to allow for the simultaneous quantification of any Chemical Weapons Convention Schedule 1 OPNA by measuring the aged adducts methyl phosphonate, ethyl phosphonate, propyl phosphonate, ethyl phosphoryl, phosphoryl and unadducted BChE. The calibration range for all analytes is 2.00-250. ng/mL, which is consistent with similar methodologies used to detect unaged OPNA-BChE adducts. Each analytical run is 3 min, making the time to first unknown results, including calibration curve and quality controls, less than 1 h. Analysis of commercially purchased individual serum samples demonstrated no potential interferences with detection of aged OPNA-BChE adducts, and quantitative measurements of endogenous levels of BChE were similar to those previously reported in other OPNA-BChE adduct assays. Abstract

This Supplemental Environmental Impact Statement has been prepared in accordance with the National Environmental Policy Act to analyze the potential environmental consequences of the Proposed Action and No-Action Alternative. The environmental consequences of testing the ABL were analyzed in the Final Environmental Impact Statement for the Program Definition and Risk Reduction Phase of the Airborne Laser Program, dated April 1997. Since that date, the proposed test activities have been refined sufficiently to warrant analysis in a supplemental EIS. Changes to the test activities that support a supplemental analysis include the addition of a second ABL aircraft, refinement of both ground- and flight-test activities, and analysis of the potential for laser energy to continue off the test ranges. The document includes analysis of local community, airspace, health and safety, hazardous materials and hazardous waste management air quality, noise, biological resources, cultural resources, and socioeconomics. The Proposed Action involves both ground-level and flight testing of the ABL systems. Two ABL aircraft {Block 04 and Block 08 aircraft) would be utilized during test activities. Software upgrades to the Block 2004 aircraft would be tested and added to that test article under a Block 2006 effort. Once upgraded with the newer operating system the Block 2004 aircraft would be designated as the Block 2006 aircraft. Ground-testing activities would be conducted at Edwards AFB within the installations' boundaries and on existing test ranges. Kirtland AFB and WSMR/Holloman AFB have been identified as alternative ground-test locations in the event ground tests cannot be conducted at Edwards AFB. Flight test activities would be conducted at WSMR including FAA-coordinated airspace and airspace utilized by Fort Bliss, at R-2508 Airspace Complex utilized by Edwards AFB, and at the Western Range over the Pacific Ocean off the coast of Vandenberg AFB.
