Abstract Magnetic resonance imaging contrast agents can enhance diagnostic precision but often face limitations such as short imaging windows, low tissue specificity, suboptimal contrast enhancement, or potential toxicity, which affect resolution and long-term monitoring. Here, we present a protein contrast agent based on lanmodulin, engineered with a single-point mutation at position 108 from N to D to yield maximum gadolinium binding sites. After loading with Gd3+ ions, the resulting protein complex, LanND-Gd, exhibits efficient renal clearance, high relaxivity, and prolonged renal retention compared to commercial agents. LanND-Gd enables high-performance visualization of whole-body structures and brain vasculature in male mice at a resolution finer than one hundred micrometers. In male ischemia mouse models, LanND-Gd also improves kidney dysfunction monitoring while minimizing risks of neural toxicity or immunogenic reactions. This protein-based contrast agent offers superior image quality, improved biocompatibility, and extended imaging timeframes, promising significant advancements in magnetic resonance-based diagnostics and patient outcomes.

Objective To investigate the molecular mechanism of lncRNA (HOTAIR)/miR-206/TAGLN2 signaling axis in regulating the invasion of papillary thyroid carcinoma cells line TPC-1. Methods The papillary thyroid carcinoma(PTC) and paracancerous tissues were collected from the First Hospital of Lanzhou University, and the expression of lncRNA (HOTAIR), miR-206 and TAGLN2 in PTC tissues as well as paracancerous tissues was detected by RT-qPCR and Western blot; The thyroid papillary cancer cells with miR-206 over-expression and HOTAIR under-expression were constructed to detect the expression of target gene and its effect on the invasion of TPC-1 cells by Transwell assay. Results Compared with the adjacent tissue, the expression of HOTAIR in papilla-rythyroid cancer tissue was increased by 2.48 times. miR-206-5p was decreased down to 0.6,and the expression of TAGLN2 was increased (P＜0.05). After over-expression of miR-206, the protein expression of TAGLN2 was decreased in TPC-1 cell, and the invasion of TPC-1 cells was inhibited (P＜0.05). Knockdown of HOTAIR increased expression of miR-206 in TPC-1 cells and decreased the expression of TAGLN2. The Transwell assay showed that the invasion of TPC-1 cells was significantly reduced (P＜0.05). Conclusions In PTC, reducing the expression of HOTAIR results in over-expression of miR-206 then further inhibits the expression of TAGLN2. The biological outcome is inhibition of invasion of thyroid cancer.

Abstract A muon collider would enable the big jump ahead in energy reach that is needed for a fruitful exploration of fundamental interactions. The challenges of producing muon collisions at high luminosity and 10 TeV centre of mass energy are being investigated by the recently-formed International Muon Collider Collaboration. This Review summarises the status and the recent advances on muon colliders design, physics and detector studies. The aim is to provide a global perspective of the field and to outline directions for future work.

Abstract The neutron capture cross section of $$ ^{181} $$ 181 Ta is relevant to s-process of nuclear astrophysics, extraterrestrial samples analysis in planetary geology and new generation nuclear energy system design. The $$^{181}$$ 181 Ta( $$n,\gamma $$ n , γ ) cross section had been measured between 1 eV and 800 keV at the back-streaming white neutron facility (Back-n) of China spallation neutron source(CSNS) using the time-of-flight (TOF) technique and $$\hbox {C}_{6}\,\hbox {D}_{6}$$ C 6 D 6 liquid scintillator detectors. The experimental results are compared with the data of several evaluated libraries and previous experiments in the resolved and unresolved resonance region. Resonance parameters are extracted using the R-Matrix code SAMMY in the 1–700 eV region. The astrophysical Maxwell average cross section(MACS) from kT = 5 to 100 keV is calculated over a sufficiently wide range of neutron energies. For the characteristic thermal energy of an astrophysical site, at kT = 30keV the MACS value of $$^{181}$$ 181 Ta is 834 ± 75 mb, which shows an obvious discrepancy with the Karlsruhe Astrophysical Database of Nucleosynthesis in Stars (KADoNiS) recommended value 766 ± 15 mb. The new measurements strongly constrain the MACS of $$^{181}$$ 181 Ta( $$n,\gamma $$ n , γ ) reaction in the stellar s-process temperatures.

Nominally phase-pure γ-Ga2O3 was deposited on (100) MgAl2O4 within a narrow temperature window centered at ∼470 °C using metal-organic chemical vapor deposition. The film deposited at 440 °C exhibited either poor crystallization or an amorphous structure; the film grown at 500 °C contained both β-Ga2O3 and γ-Ga2O3. A nominally phase-pure β-Ga2O3 film was obtained at 530 °C. Atomic-resolution scanning transmission electron microscopy (STEM) investigations of the γ-Ga2O3 film grown at 470 °C revealed a high density of antiphase boundaries. A planar defect model developed for γ-Al2O3 was extended to explain the stacking sequences of the Ga sublattice observed in the STEM images of γ-Ga2O3. The presence of the 180° rotational domains and 90° rotational domains of β-Ga2O3 inclusions within the γ-Ga2O3 matrix is discussed within the context of a comprehensive investigation of the epitaxial relationship between those two phases in the as-grown film at 470 °C and the same film annealed at 600 °C. The results led to the hypotheses that (i) incorporation of certain dopants, including Si, Ge, Sn, Mg, Al, and Sc, into β-Ga2O3 locally stabilizes the “γ-phase” and (ii) the site preference(s) for these dopants promotes the formation of “γ-phase” and/or γ-Ga2O3 solid solutions. However, in the absence of such dopants, pure γ-Ga2O3 remains the least stable Ga2O3 polymorph, as indicated by its very narrow growth window, lower growth temperatures relative to other Ga2O3 polymorphs, and the largest calculated difference in Helmholtz free energy per formula unit between γ-Ga2O3 and β-Ga2O3 than all other polymorphs.

Abstract Background Peste des petits ruminants (PPR) disease is a cross-species infectious disease that severely affects small ruminants and causes great losses to livestock industries in various countries. Distinguishing vaccine-immunized animals from naturally infected animals is an important prerequisite for the eradication of PPR. At present PPRV are classified into lineages I through IV, and only one vaccination strain, Nigeria/75/1, belongs to lineage II, but all of the epidemic strains in China at present are from lineage IV. Results To achieve this goal, we developed an SYBR Green I real-time qRT-PCR method for rapid detection and identification of PPRV lineages II and IV by analyzing different melting curve analyses. The negative amplification of other commonly circulating viruses such as orf virus, goat poxvirus, and foot-and-mouth disease virus demonstrated that primers targeting the L gene of PPRV were extremely specific. The sensitivity of the assay was assessed based on plasmid DNA and the detection limit achieved was 100 copies of PPRV lineages II and IV. Conclusion Since the method has high sensitivity, specificity, and reproducibility, it will be effectively differentiated PPRV lineages II from PPRV lineages IV in PPRV infected animals.

The neutron capture measurement of natural rhenium was performed with the time-of-flight technique at the Back-n facility of the China Spallation Neutron Source in the energy range from 1 to 500 eV of the resolved resonance region (RRR) and from 5 to 400 keV in the unresolved resonance region (URR). Prompt γ-rays originating from neutron-induced capture events were detected by four C_{6}D_{6} detectors. The pulse height weighting technique and the double-bunch unfolding method based on the Bayesian theory were used in the data analysis. To obtain reliable measurement results, background subtraction, normalization, and corrections were carefully considered. The multilevel R-matrix Bayesian code SAMMY was used to extract the resonance parameters in the RRR. An absence of resonance near 392 eV is observed in our measurement, which has been observed in previous works. The average cross sections in the URR of natural rhenium relative to ^{197}Au were obtained in logarithmical equidistant energy bins with 20 bins per energy decade. The talys code was used to describe the average cross sections in the URR; the Maxwellian-averaged cross sections (MACSs) of ^{185}Re and ^{187}Re are given from kT=5 to 100 keV. At a thermal energy of kT=30 keV, the MACS value for ^{185}Re (1469±127 mb) is in good agreement with the Karlsruhe Astrophysical Database of Nucleosynthesis in Stars (KADoNIS) recommended value (1535±62 mb) within the error bars. By contrast, the value of 1361±118 mb for ^{187}Re shows a discrepancy with the KADoNiS recommended value (1160±57 mb).

232Th/233U fuel cycle is an alternate candidate for future nuclear energy. Neutron-induced fission cross section of 232Th is one of the important nuclear data if using thorium as the fuel in the future. 232Th(n, f) cross sections from 1 to 300 MeV were measured at China Spallation Neutron Source (CSNS) Back-streaming neutron facility (Back-n) relative to 235U(n, f) and n-p scattering. A fast ionization chamber for fission cross section measurement (FIXM) and a proton recoil telescope (PRT) were used to perform the measurement. The neutron energy in this measurement reaches up to 300 MeV which is supposed to be the highest among the current Experimental Nuclear Reaction Data (EXFOR) library. It stands as the only experimental data in the region of 200-300 MeV in EXFOR as well. The experimental and evaluated data of 232Th(n, f) cross section above 60 MeV are scarce, the measurement data of this work will be valuable references for future evaluation in high energy. For the data below 60 MeV, most of our data are more consistent with ENDF/B-VIII.0, excepting in the region of 1-2 MeV where our data are more in favor of CENDL-3.2.

ABSTRACT Peste des petits ruminants virus (PPRV) causes a highly contagious disease in small ruminants and severe economic losses in developing countries. PPRV infection can stimulate high levels of interferon (IFN) and many IFN-stimulated genes (ISGs), such as ISG15, which may play a key role in the process of viral infection. However, the role of ISG15 in PPRV infection and replication has not yet been reported. In this study, we found ISG15 expression to be significantly upregulated after PPRV infection of caprine endometrial epithelial cells (EECs), and ISG15 inhibits the proliferation of PPRV. Further analysis showed that free ISG15 could inhibit PPRV proliferation. Moreover, ISG15 does not affect the binding, entry, and transcription but does suppress the replication of PPRV. A detailed analysis revealed that ISG15 interacts and colocalizes with both viral N and P proteins and that its interactive regions are all located in the N-terminal domain. Further studies showed that ISG15 can competitively interact with N and P proteins and significantly interfere with their binding. Finally, through the construction of the C-terminal mutants of ISG15 with different lengths, it was found that amino acids (aa) 77 to 101 play a key role in inhibiting the binding of N and P proteins and that interaction with the P protein disappears after the deletion of 77 to 101 aa. The present study revealed a novel mechanism of ISG15 in disrupting the activity of the N0-P complex to inhibit viral replication. IMPORTANCE PPRV, a widespread and fatal disease of small ruminants, is one of the most devastating animal diseases in Africa, the Middle East, and Asia, causing severe economic losses. IFNs play an important role as a component of natural immunity against pathogens, yet the role of ISG15, an IFN-stimulated gene, in protecting against PPRV infection is currently unknown. We demonstrated, for the first time, that free ISG15 inhibits PPRV proliferation by disrupting the activity of the N0-P complex, a finding that has not been reported in other viruses. Our results provide important insights that can further understand the pathogenesis and innate immune mechanisms of PPRV.

Peste des petits ruminants (PPR) is an acute and highly pathogenic infectious disease caused by peste des petits ruminants virus (PPRV), which can infect goats and sheep and poses a major threat to the small ruminants industry. The innate immune response plays an important role as a line of defense against the virus. The effect of PPRV on the active innate immune response has been described in several studies, with different conclusions. We infected three goat-derived cell lines with PPRV and tested their innate immune response. PPRV proliferated in caprine endometrial epithelial cells (EECs), caprine skin fibroblasts cells (GSFs), and goat fibroblast cells (GFs), and all cells expressed interferon (IFN) by poly (I: C) stimulation. PPRV infection stimulated expression of type I and type III IFN on EECs, and expression of the latter was significantly stronger, but IFN was not stimulated in fibroblasts (GSFs and GFs). Our results suggested that the effect of PPRV on IFN was cell-type specific. Nine IFN-stimulated genes (ISGs) were detected in EECs, but only ISG15 and RSAD2 were significantly upregulated. The effects of PPRV on IFN and IFN-induced ISGs were cell-type specific, which advances our understanding of the innate immune response induced by PPRV and creates new possibilities for the control of PPRV infection.

Following the discovery of the Higgs boson at the LHC in 2012, new large colliders are being considered and studied by the international high-energy community to explore the Higgs boson in details and to probe new physics beyond the Standard Model. In China, a two-stage circular collider project, CEPC-SPPC was proposed and is under study. The first stage, CEPC (Circular Electron Positron Collier, a so-called Higgs factory) is focused on the Higgs physics, and the second stage, SPPC (Super Proton-Proton Collider) will be an energy frontier collider and a discovery machine beyond the LHC. The two colliders will share a same tunnel of 100 km in circumference, with a goal of 250 GeV in center-of-mass for CEPC and 75 TeV for SPPC Phase-I and 125–150 TeV for the SPPC ultimate goal. This article presents the design concept of the SPPC and some study results about the key accelerator physics problems and technical issues, which include luminosity optimization, beam collimation, beam-beam effects, longitudinal beam dynamics, high-field magnets and beam screen.

Bunch splitting is an RF manipulation method of changing the bunch structure, bunch numbers and bunch intensity in a high-intensity synchrotron that serve as the injector for a particle collider. An efficient way to realize bunch splitting is to use the combination of different harmonic RF systems, such as the two-fold bunch splitting of a bunch with a combination of fundamental harmonic and higher harmonic RF systems. The two-fold bunch splitting and three-fold bunch splitting methods have been experimentally verified and successfully applied in the CERN PS. In this paper, a generalized multifold bunch splitting method is given. The five-fold bunch splitting method using specially designed multi-harmonic RF systems was studied and tentatively applied to the medium-stage synchrotron (MSS), the third accelerator of the injector chain of the Super Proton-Proton Collider (SPPC), to mitigate the pileup effects and collective instabilities of a single bunch in the SPPC. The results show that the five-fold bunch splitting is feasible and both the bunch population distribution and longitudinal emittance growth after the splitting are acceptable, e.g., a few percent in the population deviation and less than 10% in the total emittance growth. Intensity-related effects and beam feedback issues during the splitting process have also been addressed.

Short bunch proton beams are of great significance for the applications of white neutron beams and muon beams. The accelerator complex of the China Spallation Neutron Source (CSNS) was designed to support the applications mainly based on neutron scattering techniques where the proton pulse length is not very sensitive. Some theoretical and experimental studies have been performed to see if one can extract a short-bunch proton beam by bunch rotation from the rapid cycling synchrotron (RCS) at CSNS. The experimental results at RCS have evidently displayed the bunch lengthening and rotation process, which demonstrates the effectiveness of this method even with a very short available time for the rf gymnastic processes and a high-intensity beam. With a beam power of 50 kW and normal longitudinal emittance at the injection, the proton beam with a bunch length of about 53% with respect to the one in the normal operation mode was obtained and transported to the spallation target. With a reduced longitudinal emittance at injection and the beam power of 30 kW, the shortest extraction bunch length obtained is about 26% of the one in the normal operation mode. Different machine settings have also been tested to show the impact of the desynchronization between the rf and magnetic fields, the influence of the nonadiabatic rise time, and the adiabatic decay time of the rf voltage on the extraction bunch length. The experimental results are well consistent with the theoretical and simulated ones. It is interesting to observe that space charge has a beneficial effect on the bunch lengthening which will result in a shorter bunch at the extraction with the later bunch rotation. The controlled desynchronization method between the rf and magnetic fields in an RCS was also proven successful.

Abstract Backgroud Duck Tembusu virus (DTMUV), a pathogenic flavivirus, emerged in China since 2010 and causing huge economic loss in the Chinese poultry industry. Although several vaccines have been reported to control DTMUV disease, few effective vaccines are available and new outbreaks were continuously reported. Thus, it is urgently to develop a new effective vaccine for prevention of this disease. Methods In this study, a suicidal DNA vaccine based on a Semliki Forest virus (SFV) replicon and DTMUV E glycoprotein gene was constructed and the efficacy of this new vaccine was assessed according to humoral and cell-mediated immune responses as well as protection against the DTMUV challenge in ducklings. Results Our results showed that the recombinant SFV replicon highly expressed E glycoprotein in DEF cells. After intramuscular injection of this new DNA vaccine in ducklings, robust humoral and cellular immune responses were observed in all immunized ducklings. Moreover, all ducklings were protected against challenge with the virulent DTMUV AH-F10 strain. Conclusions In conclusion, we demonstrate that this suicidal DNA vaccine is a promising candidate facilitating the prevention of DTMUV infection.

In nuclear reactors, inelastic neutron scattering is a significant energy-loss mechanism which has deep impacts on designments of nuclear reactor and radiation shielding. Iron is an important material in reactor. However, for the existing nuclear data for iron, there exists an obvious divergence for the inelastic scattering cross sections and the related gamma production sections. Therefore the precise measurements are urgently needed for satisfying the demanding to design new nuclear reactors (fast reactors), Accelerator Driven Subcritical System (ADS), and other nuclear apparatus. In this paper, we report a new system with an array of HPGe detectors, electronics and acquisition system. Experiments had been carried out on three neutron facilities.

Rabbit hemorrhagic disease virus (RHDV) is an important member of the Caliciviridae family and a highly lethal pathogen in rabbits. Although the cell receptor of RHDV has been identified, the mechanism underlying RHDV internalization remains unknown. In this study, the entry and post-internalization of RHDV into host cells were investigated using several biochemical inhibitors and RNA interference. Our data demonstrate that rabbit nucleolin (NCL) plays a key role in RHDV internalization. Further study revealed that NCL specifically interacts with the RHDV capsid protein (VP60) through its N-terminal residues (aa 285-318), and the exact position of the VP60 protein for the interaction with NCL is located in a highly conserved region (472Asp-Val-Asn474; DVN motif). Following competitive blocking of the interaction between NCL and VP60 with an artificial DVN peptide (RRTGDVNAAAGSTNGTQ), the internalization efficiency of the virus was markedly reduced. Moreover, NCL also interacts with the C-terminal residues of clathrin light chain A, which is an important component in clathrin-dependent endocytosis. In addition, the results of animal experiments also demonstrated that artificial DVN peptides protected most rabbits from RHDV infection. These findings demonstrate that NCL is involved in RHDV internalization through clathrin-dependent endocytosis.
