In many porous catalyst supports, the accessibility of interior catalytic sites to reactant species could be restricted due to limitations of reactant transport through pores comparable to reactant dimensions. The interplay between reaction and diffusion in porous catalysts is defined through the Thiele modulus and the effectiveness factor, with diffusional restrictions leading to high Thiele moduli, reduced effectivess factors, and a reduction in the observed reaction rate. We demonstrate a method to integrate ceramic nanostraws into the interior of ordered mesoporous silica MCM-41 to mitigate diffusional restrictions. The nanostraws are the natural aluminosilicate tubular clay minerals known as halloysite. Such halloysite nanotubes (HNTs) have a lumen diameter of 15-30 nm, which is significantly larger than the 2-4 nm pores of MCM-41, thus facilitating entry and egress of larger molecules to the interior of the pellet. The method of integrating HNT nanostraws into MCM-41 is through a ship-in-a-bottle approach of synthesizing MCM-41 in the confined volume of an aerosol droplet that contains HNT nanotubes. The concept is applied to a system in which microcrystallites of Ni@ZSM-5 are incorporated into MCM-41. Using the liquid phase reduction of nitrophenol as a model reaction catalyzed by Ni@ZSM-5, we show that the insertion of HNT nanostraws into this composite leads to a 50% increase in the effectiveness factor. The process of integrating nanostraws into MCM-41 through the aerosol-assisted approach is a one-step facile method that complements traditional catalyst preparation techniques. The facile and scalable synthesis technique toward the mitigation of diffusional restrictions has implications to catalysis and separation technologies.

The hydrophobic effect of alkyl group insertion into phospholipid bilayers is exploited in modifying and modulating vesicle structure. We show that amphiphilic polypeptoids (peptide mimics) with n-decyl side chains, which we term as hydrophobe-containing polypeptoids (HCPs), can insert the alkyl hydrophobes into the membrane bilayer of phospholipid-based vesicles. Such insertion leads to disruption of the liposomes and the formation of HCP-lipid complexes that are colloidally stable in aqueous solution. Interestingly, when these complexes are added to fresh liposomes, remnant uncomplexed hydrophobes (the n-decyl groups) bridge liposomes and fuse them. The fusion leads to the engulfing of liposomes and the formation of multilayered vesicles. The morphology of the liposome system can be changed from stopping fusion and forming clustered vesicles to the continued formation of multilayered liposomes simply by controlling the amount of the HCP-lipid complex added. The entire procedure occurs in aqueous systems without the addition of any other solvents. There are several implications to these observations including the biological relevance of mimicking fusogenic proteins such as the SNARE proteins and the development of new drug delivery technologies to impact delivery to cell organelles.

Linear-bottlebrush, amphiphilic copolymers were grafted from the surface of silica (SiO2) nanoparticles to form “nanoparticle micelles” that, regardless of the concentration, possess the capability to encapsulate and stabilize non-polar structures in aqueous media. These nanoparticles afford high water dispersibility and enhanced oil encapsulation from their inherent architectural design. Utilizing well-established techniques, such as surface-initiated ring-opening polymerization (SI-ROP) and surface-initiated atom transfer radical polymerization (SI-ATRP), to effectively fine-tune the ratio of hydrophobic (poly(caprolactone), PCL) to hydrophilic (poly[oligo(ethylene glycol)methyl methacrylate mono-methyl ether], POEGMA) copolymer composition, enhances oil encapsulation while maintaining aqueous solubility. Preliminary oil encapsulation was quantified with differential scanning calorimetry (DSC) using two different types of crude oil. These nanoparticles offer an alternative dispersant system to replace commercially used Corexit™ 9500 by effectively breaking up disastrous oil slicks and minimizing the long-term effects of dispersant additives in the environment.

Rationale: Pulmonary vascular endotheliitis, perivascular inflammation, and immune activation are observed in COVID-19 patients. While the initial SARS-CoV-2 infection mainly infects lung epithelial cells, whether it also infects endothelial cells (ECs) and to what extent SARS-CoV-2-mediated pulmonary vascular endotheliitis is associated with immune activation remain to be determined. Methods: To address these questions, we studied SARS-CoV-2-infected K18-hACE2 (K18) mice, a severe COVID-19 mouse model, as well as lung samples from SARS-CoV-2-infected nonhuman primates (NHP) and patient deceased from COVID-19. We used immunostaining, RNAscope, and electron microscopy to analyze the organs collected from animals and patient. We conducted bulk and single cell (sc) RNA-seq analyses, and cytokine profiling of lungs or serum of the severe COVID-19 mice. Results: We show that SARS-CoV-2-infected K18 mice develop severe COVID-19, including progressive body weight loss and fatality at 7 days, severe lung interstitial inflammation, edema, hemorrhage, perivascular inflammation, systemic lymphocytopenia, and eosinopenia. Body weight loss in K18 mice correlated with the severity of pneumonia, but not with brain infection. We also observed endothelial activation and dysfunction in pulmonary vessels evidenced by the up-regulation of VCAM1 and ICAM1 and the downregulation of VE-cadherin. We detected SARS-CoV-2 in capillary ECs, activation and adhesion of platelets and immune cells to the vascular wall of the alveolar septa, and increased complement deposition in the lungs, in both COVID-19-murine and NHP models. We also revealed that pathways of coagulation, complement, K-ras signaling, and genes of ICAM1 and VCAM1 related to EC dysfunction and injury were upregulated, and were associated with massive immune activation in the lung and circulation. Conclusion: Together, our results indicate that SARS-CoV-2 causes endotheliitis via both infection and infection-mediated immune activation, which may contribute to the pathogenesis of severe COVID-19 disease.

Our analysis of investor account data uncovers not only large inflows of new investors to the warrant market but also greatly intensified trading by existing warrant investors. This episode exemplifies the so-called “whack-a-mole” game in financial regulations.

The use of chemical dispersants is a well-established approach to oil spill remediation where surfactants in an appropriate solvent are contacted with the oil to reduce the oil-water interfacial tension and create small oil droplets capable of being sustained in the water column. Dispersant formulations typically include organic solvents, and to minimize environmental impacts of dispersant use and avoid surfactant wastage it is beneficial to use water-based systems and target the oil-water interface. The approach here involves the tubular clay minerals known as halloysite nanotubes (HNTs) that serve as nanosized reservoir for surfactants. Such particles generate Pickering emulsions with oil, and the release of surfactant reduces the interfacial tension to extremely low values allowing small droplets to be formed that are colloidally stable in the water column. We report new findings on engineering the surfactant-loaded halloysite nanotubes to be stimuli responsive such that the release of surfactant is triggered by contact with oil. This is achieved by forming a thin coating of wax to stopper the nanotubes to prevent the premature release of surfactant. Surfactant release only occurs when the wax dissolves upon contact with oil. The system thus represents an environmentally benign approach where the wax coated HNTs are dispersed in an aqueous solvent and delivered to an oil spill whereupon they release surfactant to the oil-water interface upon contact with oil.

We report the ability to place a high concentration of liposomes in a confined volume as a multicompartment cluster that mimics biological cells and allows for the modulation of release of encapsulated species. The formation of these coated multicompartmental structures is achieved by first binding liposomes into clusters before encapsulating them within a two-dimensional metal–organic framework composed of tannic acid coordinated with a metal ion. The essential feature is a molecularly thin skin over a ssystem of clustered liposomes in a pouch. The structural features of these pouches are revealed by small-angle scattering and electron microscopy. Through cryogenic electron microscopy, clusters with intact liposomes are observed that appear to be encapsulated within a pouch. Small-angle X-ray scattering shows the emergence of a relatively weak Bragg peak at q = 0.125 Å(−1), possibly indicating the attachment of the bilayers of adjacent liposomes. The metal–phenolic network (MPN) forms a nanosized conformal coating around liposome clusters, resulting in the reduced release rate of the encapsulated rhodamine B dye. We further show the possibility of communication between the adjacent nanocompartments in the cluster by demonstrating enhanced energy transfer using fluorescence resonance energy transfer (FRET) experiments where the lipophilic donor dye 3,3′-dioctadecyloxacarbocyanine perchlorate (DiO) incorporated within one liposomal compartment transfers energy upon excitation to the lipophilic acceptor dye 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate (DiI) in a neighboring liposomal compartment due to their close proximity within the multicompartmental cluster. These observations have significance in adapting these multicompartmental structures that mimic biological cells for cascade reactions and as new depot drug delivery systems.

This work develops the concepts of particle-stabilized emulsions using tubular natural clays known as halloysites to attach to the oil–water interface and stabilize oil-in-water emulsions. Such halloysite nanotubes (HNT) serve as reservoirs for surfactants and can deliver surfactants to the oil–water interface and thus lower the oil–water interfacial tension. This two-step concept of surfactant delivery and droplet stabilization by particles has significant implications to oil spill remediation. However, to deliver surfactant loaded HNTs in a water-based solvent slurry, it is important to stopper the nanotubes to prevent premature release of the surfactant. Cotton is one of the most important and widely grown crops in the world. Understanding the synthesis mechanism of cotton fiber elongation can provide valuable tools to the cotton industry for improving cotton fiber yield and quality at the molecular level. In this work, the surface and thermal characteristics of cotton fiber samples collected from a wild type (WT) and three mutant lines (Li1, Li2-short, Li2-long, Li2-mix, and liy) were comparatively investigated. Microimaging revealed a general similarity trend of WT ≥ Li2-long ≈ Li2-mix &gt, Li1 &gt, Li2 short ≈ liy with Ca detected on the surface of the last two. Attenuated total reflectance Fourier transform infrared (ATR FT-IR) spectroscopy and thermogravimetric measurements also showed that Li2-short and liy were more similar to each other, and Li2-long and Li2-mix closer to WT while Li1 was quite independent. FT-IR results further demonstrated that wax and amorphous cellulose were co-present in fiber structures during the fiber formation processes. The correlation analysis found that the FT-IR-based maturity parameter was well correlated (p ≤ 0.05) to the onset decomposition temperature and all three weight-loss parameters at onset, peak, and end decomposition stages, suggesting that the maturity degree is a better parameter than crystallinity index (CI) and other FT-IR parameters that reflect the thermal stability of the cotton fiber. In summary, this work demonstrated that genetic mutation altered the surface and thermal characteristics in the same way for Li2-short and liy, but with different mechanisms for the other three mutant cotton fiber samples.

We use account-level data from the Shenzhen Stock Exchange to show that daily price limits, a widely adopted market stabilization mechanism, may lead to unintended, destructive market behavior: large investors tend to buy on the day when a stock hits the 10% upper price limit and then sell on the next day; and their net buying on the limit-hitting day predicts stronger long-run price reversal. We also analyze a sample of special treatment (ST) stocks, which face tighter 5% daily price limits, and provide a causal validation from comparing market dynamics before and after they are assigned the ST status.

We use Cai et al. (2017)’s proxy of patience and find out that investors do learn from their prior trading experiences. They become more patient when they participate in more IPOs, and they tend to obtain higher returns. We also document a concave learning curve: inexperienced investors are overall less patient than more experienced ones; however, inexperienced investors’ marginal effect of learning is higher than experienced ones. This learning effect only exists in infrequent traders.

Dense nonaqueous phase chlorinated compounds such as trichloroethylene (TCE) and tetrachloroethylene (PCE) are widespread groundwater and soil contaminants which cause long-term environmental pollution. Extensive efforts have been carried out to develop materials for in situ remediation particularly using nanoscale zerovalent iron (NZVI) to reduce TCE to relatively innocuous products such as ethane and ethylene. A novel technology is described here that uses earth-abundant natural tubular aluminosilicate clays known as halloysite (HNT) to support NZVI. These systems are efficient at the reductive dechlorination of such chlorinated hydrocarbons indicating a pseudo-first order rate constant of 0.1 L g–1 h–1 with NZVI particle size between 5 and 10 nm. We report a novel approach to the instantaneous photoinitiated synthesis of mixed anatase-rutile nanocrystalline TiO2 thin films with a three-dimensional nanostructure through pulsed white light irradiation of photosensitive Ti-organic precursor films. Pulsed photoinitiated pyrolysis accompanied by instantaneous self-assembly and crystallization occurred to form graphitic oxides-coated TiO2 nanograins. Subsequent pulsed light irradiation working as in situ pulsed photothermal treatment improved the crystalline quality of TiO2 film despite its low attenuation of light. The non-radiative recombination of photogenerated electrons and holes in TiO2 nanograins, coupled with inefficient heat dissipation due to low thermal conductivity, produces enough heat to provide the thermodynamic driving force for improving the crystalline quality. The graphitic oxides were reduced by pulsed photothermal treatment and can be completely removed by oxygen plasma cleaning. This photoinitiated nanofabrication technology opens a promising way for the low-cost and high-throughput manufacturing of nanostructured metal oxides as well as TiO2 nanocrystalline thin films.

We explore the use of poly(N-isopropylacrylamide) (PNIPAm)-grafted carbon microspheres (CM) dispersed in water as a stimulus-responsive lubricant. A critical concentration between 3 and 5 mg/mL of PNIPAm-grafted CM is needed to achieve low friction (coefficient of friction ∼ 0.04) at room temperature between borosilicate and silicon surfaces. An increase in the temperature of the system above the lower critical solution temperature (LCST) causes the aggregation of PNIPAm-grafted CM which leads to an increase in friction forces. The process is not immediately reversible unless the lubricant is sonicated so as to redisperse the aggregates. This work provides insight into the rolling friction mechanism and demonstrates the importance of particle singlets in achieving effective lubrication through a rolling mechanism.

α-Fe2O3 microspheres with hollow and yolk–shell (X@Fe2O3) morphologies are fabricated through synthesis in the confined environment of aerosol droplets. The process exploits the competitive carbonization of sucrose and the recrystallization of iron salts concurrently occurring in the droplet, to control the initial placement of iron species on the external surface or within the carbon microspheres. Upon calcination, the iron salts fuse and are oxidized to form a robust α-Fe2O3 shell. A partial burnoff of the carbon during calcination leads to the formation of microspheres with a core of carbon while complete burnoff leads to hollow α-Fe2O3 particles. Integration of ceramic nanoparticles in the precursor solution results in the inclusion of clusters of such nanoparticles in the core. The flexibility of synthesis of either hollow or yolk–shell particles makes the process amenable to the rapid fabrication of functional materials. As an example, α-Fe2O3 hollow microspheres show a 20-fold improvement in photo-Fenton reactivity towards the degradation of methylene blue, in comparison to commercially available materials.

High-intensity focused ultrasound (HIFU) can locally ablate biological tissues such as tumors, i.e., induce their rapid heating and coagulative necrosis without causing damage to surrounding healthy structures. It is widely used in clinical practice for minimally invasive treatment of prostate cancer. Nonablative, low-power HIFU was established as a promising tool for triggering the release of chemotherapeutic drugs from temperature-sensitive liposomes (TSLs). In this study, we combine ablative HIFU and thermally triggered chemotherapy to address the lack of safe and effective treatment options for elderly patients with high-risk localized prostate cancer. DU145 prostate cancer cells were exposed to chemotherapy (free and liposomal Sorafenib) and ablative HIFU, alone or in combination. Prior to cell viability assessment by trypan blue exclusion and flow cytometry, the uptake of TSLs by DU145 cells was verified by confocal microscopy and cryogenic scanning electron microscopy (cryo-SEM). The combination of TSLs encapsulating 10 μM Sorafenib and 8.7W HIFU resulted in a viability of less than 10% at 72 h post-treatment, which was significant less than the viability of the cells treated with free Sorafenib (76%), Sorafenib-loaded TSLs (63%), or HIFU alone (44%). This synergy was not observed on cells treated with Sorafenib-loaded nontemperature sensitive liposomes and HIFU. According to cryo-SEM analysis, cells exposed to ablative HIFU exhibited significant mechanical disruption. Water bath immersion experiments also showed an important role of mechanical effects in the synergistic enhancement of TSL-mediated chemotherapy by ablative HIFU. This combination therapy can be an effective strategy for treatment of geriatric prostate cancer patients.

Progenitors derived from the stromal vascular fraction (SVF) of white adipose tissue (WAT) possess the ability to form clonal populations and differentiate along multiple lineage pathways. However, the literature continues to vacillate between defining adipocyte progenitors as “stromal” or “stem” cells. Recent studies have demonstrated that a nonpericytic subpopulation of adipose stromal cells, which possess the phenotype, CD45−/CD31−/CD146−/CD34+, are mesenchymal, and suggest this may be an endogenous progenitor subpopulation within adipose tissue. We hypothesized that an adipose progenitor could be sorted based on the expression of CD146, CD34, and/or CD29 and when implanted in vivo these cells can persist, proliferate, and regenerate a functional fat pad over serial transplants. SVF cells and culture expanded adipose stromal/stem cells (ASC) ubiquitously expressing the green fluorescent protein transgene (GFP-Tg) were fractionated by flow cytometry. Both freshly isolated SVF and culture expanded ASC were seeded in three-dimensional silk scaffolds, implanted subcutaneously in wild-type hosts, and serially transplanted. Six-week WAT constructs were removed and evaluated for the presence of GFP-Tg adipocytes and stem cells. Flow cytometry, quantitative polymerase chain reaction, and confocal microscopy demonstrated GFP-Tg cell persistence, proliferation, and expansion, respectively. Glycerol secretion and glucose uptake assays revealed GFP-Tg adipose was metabolically functional. Constructs seeded with GFP-Tg SVF cells or GFP-Tg ASC exhibited higher SVF yields from digested tissue, and higher construct weights, compared to nonseeded controls. Constructs derived from CD146− CD34+ -enriched GFP-Tg ASC populations exhibited higher hemoglobin saturation, and higher frequency of GFP-Tg cells than unsorted or CD29+ GFP-Tg ASC counterparts. These data demonstrated successful serial transplantation of nonpericytic adipose-derived progenitors that can reconstitute adipose tissue as a solid organ. These findings have the potential to provide new insights regarding the stem cell identity of adipose progenitor cells.

The melting of the permanent ice cover in the arctic region due to the climate change is increasing industrial activity in the region and also consequently there is the enhanced risk for oil-spills. Therefore, there is a need for the development of green chemicals for oil-spill treatment in the arctic region. Carboxymethylated chitosan nanoparticles were cross-linked with Mg, Ca or Sr and the stability of nanoparticles in increased pH and salinity was studied. The carboxymethylation of chitosan was investigated with FTIR and the nanoparticles were studied with dynamic light scattering and SEM. The nanoparticles cross-linked with Ca ions were found to be most stable in increased salinity and pH. The interaction of Ca cross-linked nanoparticles with dodecane in water was also demonstrated. The nanoparticles showed promising potential for applications in oil-spill treatment.

Magnetically responsive oil-in-water emulsions are effectively stabilized by a halloysite nanotube supported superparamagnetic iron oxide nanoparticle system. The attachment of the magnetically functionalized halloysite nanotubes at the oil-water interface imparts magnetic responsiveness to the emulsion and provides a steric barrier to droplet coalescence leading to emulsions that are stabilized for extended periods. Interfacial structure characterization by cryogenic scanning electron microscopy reveals that the nanotubes attach at the oil-water interface in a side on-orientation. The tubular structure of the nanotubes is exploited for the encapsulation and release of surfactant species that are typical of oil spill dispersants such as dioctyl sulfosuccinate sodium salt and polyoxyethylene (20) sorbitan monooleate. The magnetically responsive halloysite nanotubes anchor to the oil-water interface stabilizing the interface and releasing the surfactants resulting in reduction in the oil-water interfacial tension. The synergistic adsorption of the nanotubes and the released surfactants at the oil-water interface results in oil emulsification into very small droplets (less than 20μm). The synergy of the unique nanotubular morphology and interfacial activity of halloysite with the magnetic properties of iron oxide nanoparticles has potential applications in oil spill dispersion, magnetic mobilization and detection using magnetic fields.

Nanoscale capsule-type particles with stimuli-respondent transport of chemical species into and out of the capsule are of significant technological interest. We describe the facile synthesis, properties, and applications of a temperature-responsive silica-poly( N-isopropylacrylamide) (PNIPAM) composite consisting of hollow silica particles with ordered mesoporous shells and a complete PNIPAM coating layer. These composites start with highly monodisperse, hollow mesoporous silica particles fabricated with precision using a template-driven approach. The particles possess a high specific surface area (1771 m2/g) and large interior voids that are accessible to the exterior environment through pore channels of the silica shell. An exterior PNIPAM coating provides thermoresponsiveness to the composite, acting as a gate to regulate the uptake and release of functional molecules. Uptake and release of a model compound (rhodamine B) occurs at temperatures below the lower critical solution temperature (LCST) of 32 °C, while the dehydrated hydrophobic polymer layer collapses over the particle at temperatures above the LCST, leading to a shutoff of uptake and release. These transitions are also manifest at an oil-water interface, where the polymer-coated hollow particles stabilize oil-in-water emulsions at temperatures below the LCST and destabilize the emulsions at temperatures above the LCST. Cryogenic scanning electron microscopy indicates patchlike particle structures at the oil-water interface of the stabilized emulsions. The silica-PNIPAM composite therefore couples advantages from both the hollow mesoporous silica structure and the thermoresponsive polymer.

The wetting behavior of a surface depends on both its surface chemistry and the characteristics of surface morphology and topography. Adding structure to a flat hydrophobic or oleophobic surface increases the effective contact angle and thus the hydrophobicity or oleophobicity of the surface, as exemplified by the lotus leaf analogy. We describe a simple strategy to introduce micropatterned roughness on surfaces of soft materials, utilizing the template of hexagonally packed pores of breath figures as molds. The generated inverse replicas represent micron scale patterned beadlike protrusions on hydrogel surfaces. This added roughness imparts superoleophobic properties (contact angle of the order of 150° and greater) to an inherently oleophobic flat hydrogel surface, when submerged. The introduced pattern on the hydrogel surface changes morphology as it swells in water to resemble morphologies remarkably analogous to the compound eye. Analysis of the wetting behavior using the Cassie-Baxter approximation leads to estimation of the contact angle in the superoleophobic regime and in agreement with the experimental value.

The carbonization of hydrophilic particle surfaces provides an effective route for tuning particle wettability in the preparation of particle-stabilized emulsions. The wettability of naturally occurring halloysite clay nanotubes (HNT) is successfully tuned by the selective carbonization of the negatively charged external HNT surface. The positively charge chitosan biopolymer binds to the negatively charged external HNT surface by electrostatic attraction and hydrogen bonding, yielding carbonized halloysite nanotubes (CHNT) on pyrolysis in an inert atmosphere. Relative to the native HNT, the oil emulsification ability of the CHNT at intermediate levels of carbonization is significantly enhanced due to the thermodynamically more favorable attachment of the particles at the oil-water interface. Cryogenic scanning electron microscopy (cryo-SEM) imaging reveals that networks of CHNT attach to the oil-water interface with the particles in a side-on orientation. The concepts advanced here can be extended to other inorganic solids and carbon sources for the optimal design of particle-stabilized emulsions.

Iron-carbon (Fe-C) composite microspheres prepared through a facile aerosol-based process are effective remediation agents for the simultaneous adsorption and reduction of chlorinated hydrocarbons. Complete dechlorination was achieved for the class of chlorinated ethenes that include tetrachloroethylene (PCE), trichloroethylene (TCE), cis- and trans-1,2-dicloroethylene (c-DCE, t-DCE), 1,1-dichloroethylene (1,1-DCE) and, vinyl chloride (VC). The Fe-C particles potentially provides multi-functionality with requisite characteristics of adsorption, reaction, and transport for the effective in situ remediation of chlorinated hydrocarbons. The carbon support immobilizes the ferromagnetic iron nanoparticles onto its surface, thereby inhibiting aggregation. The adsorptive nature of the carbon support prevents the release of toxic intermediates such as the dichloroethylenes and vinyl chloride. The adsorption of chlorinated ethenes on the Fe-C composites is higher (>80%) than that of humic acid ( 90%). The aerosol-based process is an efficient method to prepare adsorptivereactive composite particles in the optimal size range for transport through the porous media and as effective targeted delivery agents for the in situ remediation of soil and groundwater contaminants.

The effect of electron–phonon interactions on transport properties of 2D materials is unclear. Transport measurements on atomically thin Nb3SiTe6 crystals now show that reduced dimensionality results in the suppression of electron–phonon coupling. It is now well established that many of the technologically important properties of two-dimensional (2D) materials, such as the extremely high carrier mobility in graphene1 and the large direct band gaps in MoS2 monolayers2, arise from quantum confinement. However, the influence of reduced dimensions on electron–phonon (e–ph) coupling and its attendant dephasing effects in such systems has remained unclear. Although phonon confinement3,4,5,6,7 is expected to produce a suppression of e–ph interactions in 2D systems with rigid boundary conditions6,7, experimental verification of this has remained elusive8. Here, we show that the e–ph interaction is, indeed, modified by a phonon dimensionality crossover in layered Nb3SiTe6 atomic crystals. When the thickness of the Nb3SiTe6 crystals is reduced below a few unit cells, we observe an unexpected enhancement of the weak-antilocalization signature in magnetotransport. This finding strongly supports the theoretically predicted suppression of e–ph interactions caused by quantum confinement of phonons.

The objective of this study is to develop and compare several Sorafenib-loaded biocompatible nanoparticle models in order to optimize drug delivery and tumor cellular kill thereby improving the quality of Sorafenib-regimented chemotherapy. Sorafenib-loaded poly (lactic-co-glycolic) acid (PLGA), 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) liposomes, and hydrophobically modified chitosan (HMC)-coated DPPC liposomes were evaluated for several characteristics including zeta potential, drug loading, and release profile. Cytotoxicity and uptake trials were also studied using cell line RCC 786-0, a human metastatic clear cell histology renal cell carcinoma cell line. Sorafenib-loaded PLGA particles and HMC-coated DPPC liposomes exhibited significantly improved cell kill compared to Sorafenib alone at lower concentrations, namely 10-15 and 5-15 μM from 24 to 96 h, respectively. At maximum dosage and time (15 μM and 96 h), Sorafenib-loaded PLGA and HMC-coated liposomes killed 88.3 ± 1.8% and 98 ± 1.1% of all tumor cells, significant values compared with Sorafenib 81.8 ± 1.7% (p < 0.01). Likewise, HMC coating substantially improved cell kill for liposome model for all concentrations (5-15 μM) and at time points (24-96 h) (p < 0.01). PLGA and HMC-coated liposomes are promising platforms for drug delivery of Sorafenib. Because of different particle characteristics of PLGA and liposomes, each model can be further developed for unique clinical modalities.

We report the ability of hydrophobically modified polypeptoids (HMPs), which are amphiphilic pseudopeptidic macromolecules, to connect across lipid bilayers and thus form layered structures on liposomes. The HMPs are obtained by attaching hydrophobic decyl groups at random points along the polypeptoid backbone. Although native polypeptoids (with no hydrophobes) have no effect on liposomal structure, the HMPs remodel the unilamellar liposomes into structures with comparable diameters but with multiple concentric bilayers. The transition from single-bilayer to multiple-bilayer structures is revealed by small-angle neutron scattering (SANS) and cryo-transmission electron microscopy (cryo-TEM). The spacing between bilayers is found to be relatively uniform at ∼6.7 nm. We suggest that the amphiphilic nature of the HMPs explains the formation of multibilayered liposomes; i.e., the HMPs insert their hydrophobic tails into adjacent bilayers and thereby serve as the connective glue between bilayers. At higher HMP concentrations, the liposomes are entirely disrupted into much smaller micellelike structures through extensive hydrophobe insertion. Interestingly, these small structures can reattach to fresh unilamellar liposomes and self-assemble to form new two-bilayer liposomes. The two-bilayer liposomes in our study are reminiscent of two-bilayer organelles such as the nucleus in eukaryotic cells. The observations have significance in designing new nanoscale drug delivery carriers with multiple drugs on separate lipid bilayers and extending liposome circulation times with entirely biocompatible materials.

Patients with burn wounds are susceptible to wound infection and sepsis. This research introduces a novel burn wound dressing that contains silver nanoparticles (SNPs) to treat infection in a 2-acrylamido-2-methylpropane sulfonic acid sodium salt (AMPS-Na(+) ) hydrogel. Silver nitrate was dissolved in AMPS-Na(+) solution and then exposed to gamma irradiation to form SNP-infused hydrogels. The gamma irradiation results in a cross-linked polymeric network of sterile hydrogel dressing and a reduction of silver ions to form SNPs infused in the hydrogel in a one-step process. About 80% of the total silver was released from the hydrogels after 72 h immersion in simulated body fluid solution; therefore, they could be used on wounds for up to 3 days. All the hydrogels were found to be nontoxic to normal human dermal fibroblast cells. The silver-loaded hydrogels had good inhibitory action against Pseudomonas aeruginosa and methicillin-resistant Staphylococcus aureus. Results from a pilot study on a porcine burn model showed that the 5-mM silver hydrogel was efficient at preventing bacterial colonization of wounds, and the results were comparable to the commercially available silver dressings (Acticoat(TM) , PolyMem Silver(®) ). These results support its use as a potential burn wound dressing.

This study examines the antitumor potential of curcumin and C6 ceramide (C6) against osteosarcoma (OS) cell lines when both are encapsulated in the bilayer of liposomal nanoparticles. Three liposomal formulations were prepared: curcumin liposomes, C6 liposomes and C6-curcumin liposomes. Curcumin in combination with C6 showed 1.5 times enhanced cytotoxic effect in the case of MG-63 and KHOS OS cell lines, in comparison with curcumin liposomes alone. Importantly, C6-curcumin liposomes were found to be less toxic on untransformed primary human cells (human mesenchymal stem cells) in comparison to OS cell lines. In addition, cell cycle assays on a KHOS cell line after treatment revealed that curcumin only liposomes induced G2/M arrest by upregulation of cyclin B1, while C6 only liposomes induced G1 arrest by downregulation of cyclin D1. C6-curcumin liposomes induced G2/M arrest and showed a combined effect in the expression levels of cyclin D1 and cyclin B1. The efficiency of the preparations was tested in vivo using a human osteosarcoma xenograft assay. Using pegylated liposomes to increase the plasma half-life and tagging with folate (FA) for targeted delivery in vivo, a significant reduction in tumor size was observed with C6-curcumin-FA liposomes. The encapsulation of two water insoluble drugs, curcumin and C6, in the lipid bilayer of liposomes enhances the cytotoxic effect and validates the potential of combined drug therapy.

Liposomal coatings are formed on films of a biopolymer, hydrophobically modified chitosan (hm-chitosan), containing dodecyl groups as hydrophobes along the polymer backbone. The alkyl groups insert themselves into the liposome bilayer through hydrophobic interactions and thus tether liposomes, leading to a densely packed liposome layer on the film surface. Such liposomal surfaces exhibit effective lubrication properties due to their high degree of hydration, and reduce the coefficient of friction to the biologically-relevant range. The compliancy and robustness of these tethered liposomes allow retention on the film surface upon repeated applications of shear. Such liposome coated films have potential applications in biolubrication.

Water-soluble gold nanoparticles (AuNP) stabilized with cavitands having carboxylic acid groups have been synthesized and characterized by a variety of techniques. Apparently, the COOH groups similar to thiol are able to prevent aggregation of AuNP. These AuNP were stable either as solids or in aqueous solution. Most importantly, these cavitand functionalized AuNP were able to include organic guest molecules in their cavities in aqueous solution. Just like free cavitands (e.g., octa acid), cavitand functionalized AuNP includes guests such as 4,4'-dimethylbenzil and coumarin-1 through capsule formation. The exact structure of the capsular assembly is not known at this stage. Upon excitation there is communication between the excited guest present in the capsule and gold atoms and this results in quenching of phosphorescence from 4,4'-dimethylbenzil and fluorescence from coumarin-1.

A rapid solvothermal method for the fabrication of hexaniobate nanoscrolls in high-yield is reported. Surfactant-containing organic solutions of HxK4−xNb6O17 crystallites treated above 200 °C result in complete conversion to nanoscrolls in 6 h. Both simple nanoscroll structures (SNS) as well as intercalated nanoscrolls (INS) with various surfactants can be readily obtained. Nanoscrolls prepared by this route appear to be more rigid and symmetric compared to those made by standard aqueous methods. Further, on exchange of surface groups, nanoscrolls can be suspended in either hydrophobic or hydrophilic solvents. This approach is significant to bulk nanoscroll production and should be effective in the development of new composite materials.

The stability of crude oil droplets formed by adding chemical dispersants can be considerably enhanced by the use of the biopolymer, hydrophobically modified chitosan. Turbidimetric analyses show that emulsions of crude oil in saline water prepared using a combination of the biopolymer and the well-studied chemical dispersant (Corexit 9500A) remain stable for extended periods in comparison to emulsions stabilized by the dispersant alone. We hypothesize that the hydrophobic residues from the polymer preferentially anchor in the oil droplets, thereby forming a layer of the polymer around the droplets. The enhanced stability of the droplets is due to the polymer layer providing an increase in electrostatic and steric repulsions and thereby a large barrier to droplet coalescence. Our results show that the addition of hydrophobically modified chitosan following the application of chemical dispersant to an oil spill can potentially reduce the use of chemical dispersants. Increasing the molecular weight of the biopolymer changes the rheological properties of the oil-in-water emulsion to that of a weak gel. The ability of the biopolymer to tether the oil droplets in a gel-like matrix has potential applications in the immobilization of surface oil spills for enhanced removal.

Magnetite nanoparticles in the size range of 3.2-7.5 nm were synthesized in high yields under variable reaction conditions using high-temperature hydrolysis of the precursor iron(II) and iron(III) alkoxides in diethylene glycol solution. The average sizes of the particles were adjusted by changing the reaction temperature and time and by using a sequential growth technique. To obtain γ-iron(III) oxide particles in the same range of sizes, magnetite particles were oxidized with dry oxygen in diethylene glycol at room temperature. The products were characterized by DLS, TEM, X-ray powder diffractometry, TGA, chemical analysis, and magnetic measurements. NMR r(1) and r(2) relaxivity measurements in water and diethylene glycol (for OH and CH(2) protons) have shown a decrease in the r(2)/r(1) ratio with the particle size reduction, which correlates with the results of magnetic measurements on magnetite nanoparticles. Saturation magnetization of the oxidized particles was found to be 20% lower than that for Fe(3)O(4) with the same particle size, but their r(1) relaxivities are similar. Because the oxidation of magnetite is spontaneous under ambient conditions, it was important to learn that the oxidation product has no disadvantages as compared to its precursor and therefore may be a better prospective imaging agent because of its chemical stability.

The morphology of hollow, double-shelled submicrometer particles is generated through a rapid aerosol-based process. The inner shell is an essentially hydrophobic carbon layer of nanoscale dimension (20 nm), and the outer shell is a hydrophilic silica layer of approximately 40 nm, with the shell thickness being a function of the particle size. The particles are synthesized by exploiting concepts of salt bridging to lock in a surfactant (CTAB) and carbon precursors together with iron species in the interior of a droplet. This deliberate negation of surfactant templating allows a silica shell to form extremely rapidly, sealing in the organic species in the particle interior. Subsequent pyrolysis results in a buildup of internal pressure, forcing carbonaceous species against the silica wall to form an inner shell of carbon. The incorporation of magnetic iron oxide into the shells opens up applications in external stimuli-responsive nanomaterials.

In this study, polyacrylamide nanocomposite hydrogels were formulated in combination with sodium montmorillonite (MMT) in the presence of a 2-T magnetic field. This top-down nanomanufacturing approach led to interesting changes in the internal structure of the gel and ultimately to a dramatic improvement in the ability of the composite to separate the two protein probes, ovum serum albumin and carbonic anhydrase. The morphology of the nanocomposite hydrogel was analyzed with cryogenic scanning and transmission electron microscopy, wide-angle X-ray diffraction, and small-angle scattering to determine whether morphological changes would correlate with this improved separation. As the volume fractions of MMT were well under 1% in this study (because of aqueous swelling), scattering data were dominated by the polymer structure. Significant morphological changes were noted at two length scales: (1) the hydrogel cell structure, at hundreds of nanometers, appeared to exhibit changes in the anisotropic orientation with magnetization, and (2) the polyamide structure, at tens of nanometers, exhibited decreasing pore size (small-angle X-ray scattering). The separation data correlated most closely with a reduction in pore size; however, an additional contribution to separation from local electrostatic effects from the presence of charged MMT in the cell walls could not be discounted. The change in the pore size associated with processing may have been due to the MMT presence altering the diffusion rates of the reactants during polymer formation. The method demonstrated herein could be used ultimately to separate proteins in their native state, with the potential retention of function for downstream applications, such as novel detection techniques or purification. © 2012 Wiley Periodicals, Inc. The delivery of curcumin, a broad-spectrum anticancer drug, has been explored in the form of liposomal nanoparticles to treat osteosarcoma (OS). Curcumin is water insoluble and an effective delivery route is through encapsulation in cyclodextrins followed by a second encapsulation in liposomes. Liposomal curcumin's potential was evaluated against cancer models of mesenchymal (OS) and epithelial origin (breast cancer). The resulting 2-Hydroxypropyl-γ-cyclodextrin/curcumin - liposome complex shows promising anticancer potential both in vitro and in vivo against KHOS OS cell line and MCF-7 breast cancer cell line. An interesting aspect is that liposomal curcumin initiates the caspase cascade that leads to apoptotic cell death in vitro in comparison with DMSO-curcumin induced autophagic cell death. In addition, the efficiency of the liposomal curcumin formulation was confirmed in vivo using a xenograft OS model. Curcumin-loaded γ-cyclodextrin liposomes indicate significant potential as delivery vehicles for the treatment of cancers of different tissue origin. From the Clinical Editor Curcumin-loaded γ-cyclodextrin liposomes were demonstrated in vitro to have significant potential as delivery vehicles for the treatment of cancers of mesenchymal and epithelial origin. Differences between mechanisms of cell death were also evaluated.
