Near-wall heat sources have a crucial part to play in the process of particle deposition. Thus, this study investigates the impact of the near-wall heat source on the distribution patterns of particle deposition on the vertical wall behind the heat source, taking into account the variability in heat source temperatures and distances from the vertical wall. A model based on the Eulerian–Lagrangian method was established for tracking the motion trajectories of 1000 particles with a density of 1400 kg/m3 and a particle size range of 0.01–10.0 μm. The temperature field, airflow field, and particle deposition distribution in six cases were analyzed. It was shown that the heat source temperature significantly affectis the temperature field, airflow field, and particle deposition distribution on the vertical wall behind the heat source. This study demonstrated that as the temperature rises, the quantity of particles deposited in the upper-right region of the vertical wall decreases more noticeably. The quantity of particles deposited onto the vertical wall is inversely related to the distance between the near-wall heat source and the vertical wall. On one hand, the deposition distribution law serves as a foundation for advancing the technology aimed at removing suspended particles via thermal plumes. On the other hand, it provides critical insights for addressing the challenges associated with harmful particle deposition linked to the attachment effects of thermal plumes.

The ground-motion simulation of regional-specific earthquake scenarios is crucial for the seismic design of key facilities. Herein, we considered parameter uncertainty in ground-motion simulations and the impact of multiple faults when determining the maximum credible ground-motion parameters of large earthquakes at a near-field dam. The source models of the Daju–Lijiang, Xiaozhongdian–Daju, and Longpan–Qiaohou faults were established based on geological and geophysical data. Although the method for identifying asperity is not yet mature and still faces many difficulties, it provides an opportunity to identify the non-uniform slip distribution on the rupture plane by earthquake scenarios. A multi-scheme stochastic finite-fault simulation method was then used to estimate the minimum; mean; maximum; and 50th-, 84th-, and 95th-percentile values of the peak ground acceleration and pseudo-spectral acceleration response spectra. The results showed that the Longpan–Qiaohou fault can generate the largest ground-motion parameters compared with the other two faults. Moreover, this result was supported by the statistical analysis of the results of twelve thousand simulations of these three faults. Thus, it can be concluded that the maximum credible ground-motion parameters are represented by the 84th-percentile pseudo-spectral acceleration response spectrum of the Longpan–Qiaohou fault. This finding will benefit the seismic safety design of the target dam. More importantly, this multi-scheme method can be applied to other key facilities to obtain reasonable ground-motion parameters.

Abstract Freshwater pollution and shortage have become an imminent problem. Therefore, it is necessary to develop a multi-functional membrane for the production of fresh water. In this work, the regenerated lignocellulose modified cotton fabric was developed as a novel, multi-functional and degradable membrane (LCPT@CF) for efficient oil–water separation and solar steam generation for the first time. The fabrication method has the merits of simple, environmentally friendly and cost effective. The regenerated lignocellulose was adhered on the surface of cotton fabric by tannic acid and polyvinyl alcohol complexes tightly, and the multilayered structures of the LCPT@CF can be formed, which endowed the membranes with underwater superoleophobic property and durability. The underwater superoleophobic property enabled LCPT@CF to purify various kinds of oil-in-water emulsions with a separation efficiency of more than 99.90%. Moreover, benefiting from the excellent photothermal conversion capacity of regenerated lignocellulose, the LCPT@CF achieved high evaporation rate of 1.39 kg m−2 h−1 and favorable evaporation efficiency of 84% under 1 sun illumination, and the LCPT@CF also presented excellent salt-resistance for evaporating seawater for 20 cycles, without salt accumulation. More importantly, the LCPT@CF could be naturally degradable by microorganisms in the natural condition within 3 months, which had outstanding environmental friendliness. These above results demonstrated that the green and efficient LCPT@CF could play great potential in oil–water separation and sewage purification.

The middle segment of the Kalawenguquan fault has been active since the Holocene, with a maximum credible earthquake magnitude of MS 7.5. We established two source models based on empirical relationships, as well as geological, geomorphological, and seismic characteristics. Taking into account the uncertainties of simulation parameters, we adopted the stochastic finite-fault method to calculate all combinations of a multi-scheme simulation. The effects of different source models, initial rupture points, and site locations on the prediction of ground motion parameters were analyzed. The results indicate that when a site is located on a smaller asperity and is a certain distance from the largest asperity, the simulation results are higher. For different sites, when the initial rupture point is located near the smaller rather than the larger asperity, the acceleration response spectra are higher. Our results show that the relationships between the initial rupture points, asperities, and sites have a significant impact on the simulation results. Therefore, our study highlights the relevance of determining the initial rupture point and source model to obtain a reasonable evaluation for near-field strong ground motion simulations at major infrastructures.

In this paper, the Internet of Things (IoT) with intelligent face perception and processing function is used to supervise online English teaching. In the intelligent learning environment, learners mainly learn by watching the information presentation screen of the learning content, i.e., the learning screen, which is the main environment for learners to learn and is the main channel for information interaction between learners and the learning content. The color matching, layout, graphic decoration, and background texture of the learning screen have a significant impact on learners’ emotions, interests, motivation, and effect in the learning process. On the contrary, the accurate identification of learners’ emotions is the basis for building a harmonious emotional interaction in the wisdom learning environment and is an important means to judge learners’ learning status, which is of great significance to promote learners’ wisdom learning. In addition to providing learners with personalized learning contents and learning paths, the learning images presented by the intelligent learning environment should also be compatible with learners’ emotional states and visual emotional preferences and can play a role in regulating and stimulating learners’ learning emotions. The system works well in the testing process, which verifies the feasibility, rationality, and effectiveness of our application of face perception to online teaching effectiveness monitoring, and can be combined with the old result-oriented effectiveness monitoring method for online teaching, with certain theoretical research significance and practical application value.

This paper analyzes a finite buffer size discrete-time Geo/G/1/N queue with multiple working vacations and different input rate. Using supplementary variable technique and embedded Markov chain method, the queue-length distribution solution in the form of formula at arbitrary epoch is obtained. Some performance measures associated with operating cost are also discussed based on the obtained queue-length distribution. Then, several numerical experiments follow to demonstrate the effectiveness of the obtained formulae. Finally, a state-dependent operating cost function is constructed to model an express logistics service center. Regarding the service rate during working vacation as a control variable, the optimization analysis on the cost function is carried out by using parabolic method.

Abstract Retinal neovascularization (RNV) disease is one of the leading causes of blindness, yet the molecular underpinnings of this condition are not well understood. To delve into the critical aspects of cell-mediated angiogenesis, we analyzed our previously published single-cell data. Our analysis revealed that retinal pigment epithelium (RPE) cells serve a crucial promotional function in angiogenesis. RPE cells were regulated by N6-methyladenosine (m6A). Next, we detected several critical m6A methylase in hypoxic ARPE-19 cells and in oxygen-induced retinopathy (OIR) mice, our results revealed a significant decrease in the level of methyltransferase like 3 (METTL3). METTL3 specific inhibitor STM2457 intravitreal injection or METTL3 conditional knockout mice both showed a significantly reduced neovascularization area of retina. Additionally, the angiogenesis-related abilities of human retinal endothelial cells (HRECs) were diminished after co-cultured with ARPE-19 treated with STM2457 or sh-METTL3 in vitro. Furthermore, through the integration of Methylated RNA immunoprecipitation (MeRIP) sequencing and RNA sequencing, we discovered that the metabolic enzyme quinolinate phosphoribosyltransferase (QPRT) was directly modified by METTL3 and recognized by the YTH N6-methyladenosine RNA binding protein C1 (YTHDC1). Moreover, after over-expressing QPRT, the angiogenic abilities of HRECs were improved through the phosphorylated phosphatidylinositol-3-kinase (p-PI3K)/ phosphorylated threonine kinase (p-AKT) pathway. Collectively, our study provided a novel therapeutic target for retinal angiogenesis.

Abstract Background Vascular endothelial growth factor (VEGF) is one of the most powerful proangiogenic factors and plays an important role in multiple diseases. Increased glycolytic rates and lactate accumulation are associated with pathological angiogenesis. Results Here, we show that a feedback loop between H3K9 lactylation (H3K9la) and histone deacetylase 2 (HDAC2) in endothelial cells drives VEGF-induced angiogenesis. We find that the H3K9la levels are upregulated in endothelial cells in response to VEGF stimulation. Pharmacological inhibition of glycolysis decreases H3K9 lactylation and attenuates neovascularization. CUT& Tag analysis reveals that H3K9la is enriched at the promoters of a set of angiogenic genes and promotes their transcription. Interestingly, we find that hyperlactylation of H3K9 inhibits expression of the lactylation eraser HDAC2, whereas overexpression of HDAC2 decreases H3K9 lactylation and suppresses angiogenesis. Conclusions Collectively, our study illustrates that H3K9la is important for VEGF-induced angiogenesis, and interruption of the H3K9la/HDAC2 feedback loop may represent a novel therapeutic method for treating pathological neovascularization.

In this study, 334 accelerograms of 42 small-to-moderate earthquakes recorded at 36 strong-motion stations were used to investigate the ground-motion characteristics of the southwestern margin of the Sichuan-Yunnan rhombic block (SWM-SYRB), one of the most active seismic regions in China. The high-frequency attenuation parameter was estimated using the spectral decay method, and the site component (κo) was fitted. The estimates decrease with the increasing time-averaged S-wave velocity of the uppermost 30 m (κo). Using the generalized inversion technique, the source spectra, quality factor (Q), and site amplification were derived from the Fourier amplitude spectra (FAS). The obtained average stress drop for earthquakes occurred in SWM-SYRB was the second largest among various boundary areas of SYRB. The inverted Q model was. The low Q structure that extends southwestward from the Songpan-Garze block to SWM-SYRB could be responsible for the strong regional attenuation of ground motion with distance. At frequencies above 10 Hz, the average site amplifications were influenced by the high-frequency attenuation effect. The site amplification of site class D reached a factor of 6 at 0.7 Hz. Moreover, it was observed that site amplification factors can be even higher when peak ground acceleration is larger than. Finally, the obtained parameters were used in the stochastic finite-fault simulation method to reproduce the FAS, 5%-damped pseudospectral acceleration, and time series of the 2021 6.1 Yangbi earthquake. The simulated spectra properly matched the observations in a broad frequency band of 0.1-25 Hz. Abstract Activated microglia in the retina are essential for the development of autoimmune uveitis. Yin‐Yang 1 (YY1) is an important transcription factor that participates in multiple inflammatory and immune‐mediated diseases. Here, an increased YY1 lactylation in retinal microglia within in the experimental autoimmune uveitis (EAU) group is observed. YY1 lactylation contributed to boosting microglial activation and promoting their proliferation and migration abilities. Inhibition of lactylation suppressed microglial activation and attenuated inflammation in EAU. Mechanistically, cleavage under targets & tagmentation （CUT&Tag） analysis revealed that YY1 lactylation promoted microglial activation by regulating the transcription of a set of inflammatory genes, including STAT3, CCL5, IRF1, IDO1, and SEMA4D. In addition, p300 is identified as the writer of YY1 lactylation. Inhibition of p300 decreased YY1 lactylation and suppressed microglial inflammation in vivo and in vitro. Collectively, the results showed that YY1 lactylation promoted microglial dysfunction in autoimmune uveitis by upregulating inflammatory cytokine secretion and boosting cell migration and proliferation. Therapeutic effects can be achieved by targeting the lactate/p300/YY1 lactylation/inflammatory genes axis.

Abstract Autoimmune uveitis (AU) is a kind of immune‐mediated disease resulting in irreversible ocular damage and even permanent vision loss. However, the precise mechanism underlying dynamic immune changes contributing to disease initiation and progression of AU remains unclear. Here, we induced an experimental AU (EAU) model with IRBP651‐670 and found that day[D]14 was the inflammatory summit with remarking clinical and histopathological manifestations and the activation of retinal microglia exhibited a time‐dependent pattern in the EAU course. We conducted single‐cell RNA sequencing of retinal immune cells in EAU mice at four time points and found microglia constituting the largest proportion, especially on D14. A novel inflammatory subtype (Cd74high Ccl5high) of retinal microglia was identified at the disease peak that was closely associated with modulating immune responses. In vitro experiments indicated that inflammatory stimuli induced proinflammatory microglia with the upregulation of CD74 and CCL5, and CD74 overexpression in microglia elicited their proinflammatory phenotype via nuclear factor‐kappa B signaling that could be attenuated by the treatment of neutralizing CCL5 antibody to a certain extent. In‐vivo blockade of Cd74 and Ccl5 effectively alleviated retinal microglial activation and disease phenotype of EAU. Therefore, we propose targeting CD74 and CCL5 of retinal microglia as promising strategies for AU treatment.

Summary: This study focused on examining the exact role of circulating immune cells in the development of diabetic retinopathy (DR). In vitro co-culture experiments showed that peripheral blood mononuclear cells (PBMCs) from patients with type 1 DR crucially modulated the biological functions of human retinal microvascular endothelial cells (HRMECs), consequently disrupting their normal functionality. Single-cell RNA sequencing (scRNA-seq) study revealed unique differentially expressed genes and pathways in circulating immune cells among healthy controls, non-diabetic retinopathy (NDR) patients, and DR patients. Of significance was the observed upregulation of JUND in each subset of PBMCs in patients with type 1 DR. Further studies showed that downregulating JUND in DR patient-derived PBMCs led to the amelioration of HRMEC dysfunction. These findings highlighted the notable alterations in the transcriptomic patterns of circulating immune cells in type 1 DR patients and underscored the significance of JUND as a key factor for PBMCs in participating in the pathogenesis of DR.

Abstract Background This research aimed to investigate the roles of fanconi anemia complementation group D2 (FANCD2) on the regulation of ferroptosis in osteosarcoma progression. Methods The function of FANCD2 on cell viability, invasion, migration, and tumor growth were explored. FANCD2 and pathway-related genes were determined by western blot. Ferroptosis-associated markers were determined, including lipid peroxidation, labile iron pool (LIP), ferrous iron (Fe2+), and ferroptosis-related genes. Results FANCD2 expression was increased in osteosarcoma cells. FANCD2 knockdown reduced cell viability, invasion, and migration of osteosarcoma cells. FANCD2 knockdown regulated ferroptosis-related gene expression, and distinctly increased the levels of LIP, Fe2+, and lipid peroxidation, and these effects were reversed by a ferroptosis inhibitor Fer-1. In addition, JAK2 and STAT3 expression were reduced by silencing of FANCD2, and STAT3 activator (colivelin) distinctly reversed tumor suppressor effects of FANCD2 silencing on osteosarcoma development. Conclusion These findings suggested that FANCD2 silencing could suppress osteosarcoma cell viability, migration, invasion, and tumor growth, and induced ferroptosis by regulating the JAK2/STAT3 axis. These findings may provide novel therapeutic ideas for clinical treatment of osteosarcoma.

This study used 306 accelerograms recorded at 22 strong-motion stations to investigate the source parameters, quality factor (Q), and site effects of S-wave Fourier acceleration amplitude spectrum (FAS) of the 2019 MS 6.0 Changning earthquake sequence in China with surface-wave magnitudes (MS) of 4.1-6.0. The generalized inversion technique (GIT) was adopted. The inverted stress drop of the mainshock was 1.15 MPa, and those of the aftershocks varied from 0.11 to 1.04 MPa with an average value of 0.43 MPa. The MS of these earthquakes were larger than Mw with an average magnitude difference of 0.22. The inverted Q values increase rapidly with frequencies at 0.5-4.0 Hz from 62 to 2920 and become less dependent at 4.0-25.0 Hz. Such a phenomenon indicates that the propagation path attenuation mechanism transited to intrinsic at high frequencies. A bilinear Q(f) model for which Q(f)=237.6f1.27 (Q<1280) and Q=1280 at higher frequencies was obtained. The high-frequency attenuation model of the study area was κ=0.0420+0.0001262R. The inverted site responses of the 22 stations were compared with those calculated using the horizontal-to-vertical spectral ratio (HVSR) method. In general, the amplification curves of most stations obtained with the GIT were similar to those of HVSR, and the amplification levels were relatively higher. Contrarily, obvious discrepancies were observed between the results estimated from the two methods at several stations. Such effects were attributed to the limitation that the majority of the stations were distributed along the boundary of the basin and mountainous areas, and the inverted Q values were representative of the specific area rather than the pure basin and mountainous areas. Finally, a nonlinear soil site effect was observed at 51GXT in earthquakes with peak ground acceleration greater than 300 cm/s². The nonlinearity obviously aggravated the site amplification at 1.0-5.0 Hz. [ABSTRACT FROM AUTHOR]

Abstract The underlying mechanisms that determine gene expression and chromatin accessibility in retinogenesis are poorly understood. Herein, single‐cell RNA sequencing and single‐cell assay for transposase‐accessible chromatin sequencing are performed on human embryonic eye samples obtained 9–26 weeks after conception to explore the heterogeneity of retinal progenitor cells (RPCs) and neurogenic RPCs. The differentiation trajectory from RPCs to 7 major types of retinal cells are verified. Subsequently, diverse lineage‐determining transcription factors are identified and their gene regulatory networks are refined at the transcriptomic and epigenomic levels. Treatment of retinospheres, with the inhibitor of RE1 silencing transcription factor, X5050, induces more neurogenesis with the regular arrangement, and a decrease in Müller glial cells. The signatures of major retinal cells and their correlation with pathogenic genes associated with multiple ocular diseases, including uveitis and age‐related macular degeneration are also described. A framework for the integrated exploration of single‐cell developmental dynamics of the human primary retina is provided.

Uveitis causes blindness and critical visual impairment in people of all ages, and retinal microglia participate in uveitis progression. Unfortunately, effective treatment is deficient. Icariin (ICA) is a bioactive monomer derived from Epimedium. However, the role of ICA in uveitis remains elusive. Our study indicated that ICA alleviated intraocular inflammation in vivo. Further results showed the proinflammatory M1 microglia could be transferred to anti-inflammatory M2 microglia by ICA in the retina and HMC3 cells. However, the direct pharmacological target of ICA is unknown, to this end, proteome microarrays and molecular simulations were used to identify the molecular targets of ICA. Data showed that ICA binds to peroxiredoxin-3 (PRDX3), increasing PRDX3 protein expression in both a time- and a concentration-dependent manner and promoting the subsequent elimination of H2O2. In addition, GPX4/SLC7A11/ACSL4 pathways were activated accompanied by PRDX3 activation. Functional tests demonstrated that ICA-derived protection is afforded through targeting PRDX3. First, ICA-shifted microglial M1/M2 phenotypic polarization was no longer detected by blocking PRDX3 both in vivo and in vitro. Next, ICA-activated GPX4/SLC7A11/ACSL4 pathways and downregulated H2O2 production were also reversed via inhibiting PRDX3 both in vivo and in vitro. Finally, ICA-elicited positive effects on intraocular inflammation were eliminated in PRDX3-deficient retina from experimental autoimmune uveitis (EAU) mice. Taking together, ICA-derived PRDX3 activation has therapeutic potential for uveitis, which might be associated with modulating microglial M1/M2 phenotypic polarization.

Abstract Background Phylogenetic species trees are widely used in inferring evolutionary relationships. Existing software and algorithms mainly focus on phylogenetic inference. However, less attention has been paid to intermediate steps, such as processing extremely large sequences and preparing configure files to connect multiple software. When the species number is large, the intermediate steps become a bottleneck that may seriously affect the efficiency of tree building. Results Here, we present an easy-to-use pipeline named PhySpeTree to facilitate the reconstruction of species trees across bacterial, archaeal, and eukaryotic organisms. Users need only to input the abbreviations of species names; PhySpeTree prepares complex configure files for different software, then automatically downloads genomic data, cleans sequences, and builds trees. PhySpeTree allows users to perform critical steps such as sequence alignment and tree construction by adjusting advanced options. PhySpeTree provides two parallel pipelines based on concatenated highly conserved proteins and small subunit ribosomal RNA sequences, respectively. Accessory modules, such as those for inserting new species, generating visualization configurations, and combining trees, are distributed along with PhySpeTree. Conclusions Together with accessory modules, PhySpeTree significantly simplifies tree reconstruction. PhySpeTree is implemented in Python running on modern operating systems (Linux, macOS, and Windows).
