Abstract Background Gangrene and perforation are severe complications of acute appendicitis, associated with a higher mortality rate compared to uncomplicated appendicitis. Accurate preoperative identification of Gangrenous or perforated appendicitis (GPA) is crucial for timely surgical intervention. Methods This retrospective multicenter study includes 796 patients who underwent appendectomy. Univariate and multivariate logistic regression analyses are used to develop a nomogram model for predicting GPA based on laboratory tests and computed tomography (CT) findings. The model is validated using an external dataset. Results Seven independent predictors were included in the nomogram: white blood cell count, lymphocyte count, D-dimer, serum glucose, albumin, maximum outer diameter of the appendix, and presence of appendiceal fecalith. The nomogram achieved good discrimination and calibration in both the training and testing sets. In the training set, the AUC was 0.806 (95%CI: 0.763–0.849), and the sensitivity and specificity were 82.1% and 66.9%, respectively. The Hosmer-Lemeshow test showed good calibration (P = 0.7378). In the testing set, the AUC was 0.799 (95%CI: 0.741–0.856), and the sensitivity and specificity were 70.5% and 75.3%, respectively. Decision curve analysis (DCA) confirmed the clinical utility of the nomogram. Conclusion The laboratory test-CT nomogram model can effectively identify GPA patients, aiding in surgical decision-making and improving patient outcomes.

This paper introduces a low-boom aircraft optimization design method guided by equivalent area distribution, which effectively improves the intuitiveness and refinement of inverse design. A gradient optimization method based on discrete adjoint equations is proposed to achieve the fast solution of the gradient information of target equivalent area distribution relative to design variables and to drive the aerodynamic shape update to the optimal solution. An optimization experiment is carried out based on a self-developed supersonic civil aircraft configuration with engines. The results show that the equivalent area distribution adjoint equation can accurately solve the gradient information. After optimization, the sonic boom level of the aircraft was reduced by 13.2 PLdB, and the drag coefficient was reduced by 60.75 counts. Moreover, the equivalent area distribution adjoint optimization method has outstanding advantages, such as high sensitivity and fast convergence speed, and can take both the low sonic boom and the low drag force of the aircraft into account, providing a powerful tool for the comprehensive optimization design of supersonic civil aircraft by considering sonic boom and aerodynamic force.

Fluid thrust vectoring (FTV) control has obvious advantages in structural quality and stealth performance because of its fast response and light weight. However, improving FTV vector performance will cause a loss in engine performance due to the need to draw airflow from the engine. In order to alleviate the above problems and further improve the vector performance of FTV, a nozzle combined with throat skewing and shock vector control is proposed, and the secondary flow of the nozzle comes from the throat and is injected into the nozzle divergence section. The numerical results indicate that compared with the original configuration, the vector angle and vector efficiency of the new configuration are more linear with the nozzle pressure ratio (NPR), and the vector angle and vector efficiency are improved by 163% and 218%, respectively, while experiencing a maximum reduction in the thrust coefficient of 1.4%. Compared with the only bypass-type shock vector nozzle, the new configuration utilizes the diversion of the two jets to eliminate the reattachment of the separation bubble after the jet and its resulting abrupt change in vector performance, improving the performance while having good control characteristics. Additionally, a sensitivity analysis of the spacing between two jets is also carried out. The spacing between two jets should be increased to make the flow pass through two weaker shock waves to improve the vector performance while ensuring that the separation after the jet is no longer attached.

Abstract Tree peony (Paeonia Sect. Moutan) is a famous ornamental plant, with huge historical, cultural, and economic significance worldwide. In this study, we reported the ~13.79 Gb draft genome of a wide‐grown Paeonia suffruticosa cultivar “Luo shen xiao chun,” representing the largest sequenced genome in dicots to date. Phylogenetic analyses based on genome sequences demonstrated that P. suffruticosa was placed as sister to Vitales, and they together formed a clade that was sister to Rosids, weakly supporting a relationship of ((Saxifragales and Vitales) and Rosids). The identification and expression analysis of MADS‐box genes based on the genome assembly and de novo transcriptome assembly of P. suffruticosa revealed that the function of C class genes was restricted in flower development, which might be responsible for the stamen petalody in tree peony cultivars. Overall, the first sequenced genome in the family Paeoniaceae provides an important resource for the origin, domestication, and evolutionary study as well as cultivar breeding in tree peony.

The effects of selenium on serum cytokines, immune cell composition and spleen proteome in mice are still unclear. In this study, male mice were fed with selenium-containing diets for eight weeks, and serum cytokines were assayed using Luminex liquid suspension chip. The spleen was obtained for histological morphology, immunohistochemistry and proteome sequencing. The results indicated that the spleen index and histological structure of the spleen were reduced and damaged with low and high selenium. Serum cytokines were regulated by selenium. Additionally, compared with medium selenium, there were 180 and 189 differential proteins with high and low selenium, respectively. The differential proteins were mainly associated with metabolic processes, regulation of response to oxidative stress, cell maturation and differentiation, cytokines and receptor binding, and were verified using Western blotting. In conclusion, it was suggested that low and high selenium affected the secretion of cytokines and impaired the immune function of the spleen.

Embryos contain a large number of lipid droplets, and lipid metabolism is gradually activated during embryonic development to provide energy. However, the regulatory mechanisms remain to be investigated. Stearoyl-CoA desaturase 1 (Scd1) is a fatty acid desaturase gene that is mainly involved in intracellular monounsaturated fatty acid production, which takes part in many physiological processes. Analysis of transcripts at key stages of embryo development revealed that Scd1 was important and expressed at an increased level during the cleavage and blastocyst stages. Knockout Scd1 gene by CRISPR/Cas9 from zygotes revealed a decrease in lipid droplets (LDs) and damage in the inner cell mass (ICM) formation of blastocyst. Comparative analysis of normal and knockout embryo transcripts showed a suppression of ribosome protein (RPs) genes, leading to the arrest of ribosome biogenesis at the 2-cell stage. Notably, the P53-related pathway was further activated at the blastocyst stage, which eventually caused embryonic development arrest and apoptosis. In summary, Scd1 helps in providing energy for embryonic development by regulating intra-embryonic lipid droplet formation. Moreover, deficiency activates the RPs-Mdm2-P53 pathway due to ribosomal stress and ultimately leads to embryonic development arrest. The present results suggested that Scd1 gene is essential to maintain healthy development of embryos by regulating energy support.

This study aimed to investigate the biological effects of supplementation of bovine lactoferricin (BLFc) at the rate of 100 mg/kg/day (LF-1) or 200 mg/kg/day (LF-2) in lactating dairy goats. Dietary BLFc supplementation increased the concentration of lactoferrin (LF) in the milk and serum (p < 0.05) without affecting the feed intake. In the LF-1 group, serum Fe, total antioxidant (T-AOC), and immunoglobulin A (IgA) were increased (p < 0.05), while malondialdehyde (MDA) was decreased (p < 0.05). In the LF-2 group, ruminal fluid pH value was decreased (p < 0.05), and the composition of ruminal microflora on day 42 was more diversified. Firmicutes phylum in the LF-2 group was the most abundant phyla. In contrast, Bacteroidetes phylum in the control group and the LF-1 group were the most abundant. Lower milk somatic cell count and higher IgA were observed in the LF-1 group and the LF-2 group than those in the control group (p < 0.05). These results suggested beneficial effects of supplementation of 100 mg/kg/day BLFc on reducing the oxidative stress and altering diversity of ruminal microflora.

Constructing heterojunction is an attractive strategy for promoting photoelectrochemical (PEC) performance in water splitting and organic pollutant degradation. Herein, a novel porous BiVO4/Boron-doped Diamond (BiVO4/BDD) heterojunction photoanode containing masses of ultra-micro electrodes was successfully fabricated with an n-type BiVO4 film coated on a p-type BDD substrate by magnetron sputtering (MS). The surface structures of BiVO4 could be adjusted by changing the duration of deposition (Td). The morphologies, phase structures, electronic structures, and chemical compositions of the photoanodes were systematically characterized and analyzed. The best PEC activity with the highest current density of 1.8 mA/cm2 at 1.23 VRHE was achieved when Td was 30 min, and the sample showed the highest degradation efficiency towards tetracycline hydrochloride degradation (TCH) as well. The enhanced PEC performance was ascribed to the excellent charge transport efficiency as well as a lower carrier recombination rate, which benefited from the formation of BiVO4/BDD ultra-micro p-n heterojunction photoelectrodes and the porous structures of BiVO4. These novel photoanodes were expected to be employed in the practical PEC applications of energy regeneration and environmental management in the future.

αS1-Casein (encoded by the CSN1S1 gene) is associated with food allergy more than other milk protein components. Milk allergy caused by αS1-casein is derived from cow milk, goat milk and other ruminant milk. However, little is known about the transcription regulation of αS1-casein synthesis in dairy goats. This study aimed to investigate the regulatory roles of signal transducer and activator of transcription 5 (STAT5) on αS1-casein in goat mammary epithelial cells (GMEC). Deletion analysis showed that the core promoter region of CSN1S1 was located at −110 to −18 bp upstream of transcription start site, which contained two putative STAT5 binding sites (gamma-interferon activation site, GAS). Overexpression of STAT5a gene upregulated the mRNA level and the promoter activity of the CSN1S1 gene, and STAT5 inhibitor decreased phosphorylated STAT5 in the nucleus and CSN1S1 transcription activity. Further, GAS site-directed mutagenesis and chromatin immunoprecipitation (ChIP) assays revealed that GAS1 and GAS2 sites in the CSN1S1 promoter core region were binding sites of STAT5. Taken together, STAT5 directly regulates CSN1S1 transcription by GAS1 and GAS2 sites in GMEC, and the mutation of STAT5 binding sites could downregulate CSN1S1 expression and decrease αS1-casein synthesis, which provide the novel strategy for reducing the allergic potential of goat milk and improving milk quality in ruminants.

Lactoferrin (LF) is a prominent protein in milk, and is present in many other secretory fluids and white blood cells. The variation of LF content in milk is possibly regulated by multiple environmental factors such as milk composition and endogenous factors. In this study, we analysed the variations of LF in both milk and plasma of 20 Xinong Saanen goats and its related factors. Firstly, LF content in colostrum was 222.6 ± 41.57 μg/mL, and then throughout the lactation period the mean concentrations remained stable, ranged between 34.61 and 51.94 μg/mL. The LF concentration in plasma fluctuated between 173.56 and 246.20 μg/mL during the entire lactational period. There was no correlation between the milk and plasma LF concentrations (P > .05). EGF acts as a ligand of the EGF receptor (EGFR) to activate the EGFR-mediated signaling pathways and is involved in the regulation of cell physiology. However, the roles of EGFR mediated signaling pathways in the regulation of lipid metabolism in goat mammary epithelial cells (GMECs) are poorly understood. To evaluate the impact of EGF on GMECs, the triglyceride (TG) content and lipid droplet were detected, using TG assay and immunofluorescence. Further, expression of lipogenic genes, the protein kinase B (Akt), phospholipase C-γ1 (PLC-γ1) and extracellular signal-regulated kinases (ERK)1/2 signaling pathways were measured by real-time polymerase chain reaction and Western blot, respectively. The results showed that the mRNA expression of EGFR gene was significantly upregulated in lactating goat mammary gland tissues compared to non-lactation period (p < 0.05). TG contents in EGF-treated GMECs were significantly increased (p < 0.05), and an increase of lipid droplets was also detected. In vitro studies demonstrated that the mRNA levels of lipogenesis-related FASN, ACC, SCD1, LXRa, LXRb and SP1 genes were positively correlated to the mRNA level of EGFR gene shown by gene overexpression and silencing (p < 0.05). The phosphorylations of Akt, ERK1/2 and PLC-γ1 in GMECs were greatly upregulated in the presence of EGF, and specific inhibitors were capable of blocking the phosphorylation of Akt, ERK1/2 and PLC-γ1. Compared with EGF-treated GMECs, the mRNA levels of FASN, ACC and SCD1 were significantly decreased in GMECs co-treated with PLC-γ1 and Akt inhibitor and EGF (p < 0.05), and TG content was also dropped significantly. These observations implied that EGFR plays an important role in regulating de novo fatty acid synthesis in GMECs, mainly mediated by Akt and PLC-γ1 signaling pathways.

In situ characterization techniques have made a significant progress in recent years, especially in the electrochemical field. For Li-ion batteries, in situ characterization techniques refer to using analytical equipment to directly characterize electrode materials during electrochemical measurements. At present, most in situ batteries are developed from commercial simulated batteries, of which the cost is very high and the cycle life is quite short. In this work, two kinds of coin-cell-based in situ batteries were designed as in situ X-ray diffraction (XRD) and Raman coin cells which exhibit many admirable advantages, such as low cost, long cycle life, easy to carry, and so on. In the designing process, in situ XRD and Raman coin cell have been tested with two electrode materials of Li4Ti5O12 and LiFePO4, and we solved many technical problems of assembling and measuring these two kinds of cells. Finally, in situ coin cells could be improved to investigate a variety of electrode materials, and this technique would arouse wide interests in the electrochemical field.

BackgroundConsiderable controversy exists on the association between retinal vein occlusion (RVO) and stroke risk. Therefore, we conducted a meta‐analysis to assess the relationship between RVO and stroke risk. Methods and ResultsPubMed, EMBASE, and the Cochrane library databases were searched for cohort studies with data on RVO and stroke risk. Studies that reported adjusted relative risks (RRs) with 95% CIs of stroke associated with RVO were included. Stratified analyses were conducted according to key characteristics. A total of 5 articles including results from 6 prospective cohort studies with 431 cases of stroke and 37 471 participants were included in the meta‐analysis. Overall, after adjustment for established cardiovascular risk factors, participants with RVO at baseline were considerably more associated with a greater incidence of stroke risk (combined RR: 1.50, 95% CI: 1.19–1.90), compared to participants without RVO. The results were more pronounced for stroke (RR: 1.72, 95% CI: 1.24–2.37) in the stratified with a stroke history. The risk of stroke was nonsignificant in male subjects (RR: 1.20, 95% CI: 0.96–1.49) and in female subjects (RR: 0.93, 95% CI: 0.64–1.34). The presence of both central RVO (RR: 1.90, 95% CI: 1.46–2.48) and branch RVO (RR: 1.79, 95% CI: 1.18–2.72) was associated with increased risk of stroke. Stratifying by age, the associations between RVO and risk of stroke were similar between the age range in the cohorts that ranged from 50 to 59 years and 60 to 69 years. ConclusionsExposure to RVO was associated with an increased risk of stroke, especially in subjects aged between 50 and 69 years. Future studies on the effect of RVO treatment and modifiable risk factor reduction on stroke risk in RVO patients are warranted.

Magnetic resonance imaging (MRI) is by nature a multi-modality technique that provides complementary information about different aspects of diseases. So far no attempts have been reported to assess the potential of multi-modal MRI in discriminating individuals with and without migraine, so in this study, we proposed a classification approach to examine whether or not the integration of multiple MRI features could improve the classification performance between migraine patients without aura (MWoA) and healthy controls. Twenty-one MWoA patients and 28 healthy controls participated in this study. Resting-state functional MRI data was acquired to derive three functional measures: the amplitude of low-frequency fluctuations, regional homogeneity and regional functional correlation strength; and structural MRI data was obtained to measure the regional gray matter volume. For each measure, the values of 116 pre-defined regions of interest were extracted as classification features. Features were first selected and combined by a multi-kernel strategy; then a support vector machine classifier was trained to distinguish the subjects at individual level. The performance of the classifier was evaluated using a leave-one-out cross-validation method, and the final classification accuracy obtained was 83.67% (with a sensitivity of 92.86% and a specificity of 71.43%). The anterior cingulate cortex, prefrontal cortex, orbitofrontal cortex and the insula contributed the most discriminative features. In general, our proposed framework shows a promising classification capability for MWoA by integrating information from multiple MRI features.
