Abstract Most of the sentiment analysis studies focus on the sentimental classification of pictures in the video, ignoring the spatio-temporal information of the sequence of picture frames as well as text and audio information. The multiple kernel learning is a new hotspot in the field of nuclear machine learning, capable of handling multiple modalities. For multiple kernel learning, it is easy to ignore the basic features that are not discriminative, and cannot make full use of the base features of different modes. This paper puts forward a novel multi-modal fusion model for sentiment analysis, in which a multiple kernel learning algorithm based on convolution margin-dimension constraint is proposed for feature fusion. Moreover, the 3D convolutional neural network is used to extract the features of visual information, and the multiple kernel learning algorithm based on margin-dimension constraint is used to fuse visual, text and audio sentiment features. Experiments conducted on the MOUD and IEMOCAP sentiment databases show that the proposed model outperforms existing models in the field of multi-modal sentiment analysis research.

Soil erosion is mainly affected by the rainfall characteristics and land cover conditions, and soil erosion modelling is important for evaluating land degradation status. The revised Universal Soil Loss Equation (RUSLE) have been widely used to simulate soil loss rate. Previous studies usually considered the general rainfall characteristics and direct effect of runoff with the event rainfall erosivity factor (Re) to produce event soil loss (Ae), whereas the fluctuation of rainfall intensity within the natural rainfall profile has rarely been considered. In this study, the relative amplitude of rainfall intensity (Ram) was proposed to generalize the features of rainfall intensity fluctuation under natural rainfall, and it was incorporated in a new Re (Re=RamEI30) to develop the RUSLE model considering the fluctuation of rainfall intensity (RUSLE-F). The simulation performance of RUSLE-F model was compared with RUSLE-M1 model (Re=EI30) and RUSLE-M2 model (Re=QREI30) using observations in field plots of grassland, orchard and shrubland during 2011–2016 in a loess hilly catchment of China. The results indicated that the relationship between Ae and RamEI30 was well described by a power function with higher R2 values (0.82–0.96) compared to QREI30 (0.80–0.88) and EI30 (0.24–0.28). The RUSLE-F model much improved the accuracy in simulating Ae with higher NSE (0.55–0.79 vs −0.11∼0.54) and lower RMSE (0.82–1.67 vs 1.04–2.49) than RUSLE-M1 model. Furthermore, the RUSLE-F model had better simulation performance than RUSLE-M2 model under grassland and orchard, and more importantly the rainfall data in the RUSLE-F model can be easily obtained compared to the measurements or estimations of runoff data required by the RUSLE-M2 model. This study highlighted the paramount importance of rainfall intensity fluctuation in event soil loss prediction, and the RUSLE-F model contributed to the further development of USLE/RUSLE family of models.

Abstract Precious metals are core assets for the development of modern technologies in various fields. Their scarcity poses the question of their cost, life cycle and reuse. Recently, an emerging catalysis employing contact-electrification (CE) at water-solid interfaces to drive redox reaction, called contact-electro-catalysis (CEC), has been used to develop metal free mechano-catalytic methods to efficiently degrade refractory organic compounds, produce hydrogen peroxide, or leach metals from spent Li-Ion batteries. Here, we show ultrasonic CEC can successfully drive the reduction of Ag(ac), Rh3+, [PtCl4]2-, Ag+, Hg2+, Pd2+, [AuCl4]-, and Ir3+, in both anaerobic and aerobic conditions. The effect of oxygen on the reaction is studied by electron paramagnetic resonance (EPR) spectroscopy and ab-initio simulation. Combining measurements of charge transfers during water-solid CE, EPR spectroscopy and gold extraction experiments help show the link between CE and CEC. What’s more, this method based on water-solid CE is capable of extracting gold from synthetic solutions with concentrations ranging from as low as 0.196 ppm up to 196 ppm, reaching in 3 h extraction capacities ranging from 0.756 to 722.5 mg g−1 in 3 h. Finally, we showed CEC is employed to design a metal-free, selective, and recyclable catalytic gold extraction methods from e-waste aqueous leachates.

BackgroundNon-suicidal self-injury (NSSI) is a worldwide mental health problem that deserves thorough investigation. This study aims to explore the effect of parenting styles, attachment to parents, and self-compassion on the occurrence of non-suicidal self-injury (NSSI) behavior in adolescents and whether these factors influence their recovery motivation.MethodsA total of 132 adolescents who had engaged in NSSI within the last year and 72 adolescents who had never engaged in NSSI were recruited from the Shenzhen Kangning Hospital and from primary and secondary schools and communities. Differences in the Hamilton Depression Scale (HAMD), Inventory of Parent and Peer Attachment (IPPA), Egma Minn av. Bardndosnauppforstran (EMBU), and Self-Compassion Scale (SCS) of participants were compared. A binary logistic model was established to measure the odds ratios of these variables on the occurrence of NSSI. In the NSSI adolescent sample, separate binary logistic models were created with NSSI impulse inhibition, NSSI resistance activity, and NSSI recovery motivation as dependent variables and with parenting styles, attachment to parents, and self-compassion as independent variables.ResultsCompared with adolescents with no NSSI behavior, those who had engaged in NSSI within the past year had higher scores on the HAMD, as well as higher EMBU-negative father parental behavior (punishment, excessive interference, rejection, and overprotection), EMBU-negative mother parental behavior (excessive interference, rejection, and punishment), and SCS negative self-compassion scores. Moreover, adolescents with NSSI had lower EMBU-father emotional warmth, EMBU-mother emotional warmth, IPPA-attachment to father, IPPA-attachment to mother, and SCS positive self-compassion scores. Current depressive symptoms and maternal punishment are risk factors for NSSI in adolescents, while positive self-compassion was a protective factor. Positive self-compassion can positively predict NSSI impulse inhibition, NSSI resistance activity, and NSSI recovery motivation. However, we unexpectedly found that the father’s emotional warmth negatively predicts NSSI resistance activity.ConclusionThis study found that positive self-compassion has a significant impact on the prevention of and recovery from NSSI behavior in adolescents.

Abstract Intestinal epithelial renewal, which depends on the proliferation and differentiation of intestinal stem cells (ISCs), is essential for epithelial homoeostasis. Understanding the mechanism controlling ISC activity is important. We found that death receptor 5 (DR5) gene deletion (DR5-/-) mice had impaired epithelial absorption and barrier function, resulting in delayed weight gain, which might be related to the general reduction of differentiated epithelial cells. In DR5-/- mice, the expression of ISC marker genes, the number of Olfm4+ ISCs, and the number of Ki67+ and BrdU+ cells in crypt were reduced. Furthermore, DR5 deletion inhibited the expression of lineage differentiation genes driving ISC differentiation into enterocytes, goblet cells, enteroendocrine cells, and Paneth cells. Therefore, DR5 gene loss may inhibit the intestinal epithelial renewal by dampening ISC activity. The ability of crypts from DR5-/- mice to form organoids decreased, and selective DR5 activation by Bioymifi promoted organoid growth and the expression of ISC and intestinal epithelial cell marker genes. Silencing of endogenous DR5 ligand TRAIL in organoids down-regulated the expression of ISC and intestinal epithelial cell marker genes. So, DR5 expressed in intestinal crypts was involved in the regulation of ISC activity. DR5 deletion in vivo or activation in organoids inhibited or enhanced the activity of Wnt, Notch, and BMP signalling through regulating the production of Paneth cell-derived ISC niche factors. DR5 gene deletion caused apoptosis and DNA damage in transit amplifying cells by inhibiting ERK1/2 activity in intestinal crypts. Inhibition of ERK1/2 with PD0325901 dampened the ISC activity and epithelial regeneration. In organoids, when Bioymifi’s effect in activating ERK1/2 activity was completely blocked by PD0325901, its role in stimulating ISC activity and promoting epithelial regeneration was also eliminated. In summary, DR5 in intestinal crypts is essential for ISC activity during epithelial renewal under homoeostasis.

Abstract Neoantigen peptides hold great potential as vaccine candidates for tumor immunotherapy. However, due to the limitation of antigen cellular uptake and cross‐presentation, the progress with neoantigen peptide‐based vaccines has obviously lagged in clinical trials. Here, a stapling peptide‐based nano‐vaccine is developed, comprising a self‐assembly nanoparticle driven by the nucleic acid adjuvant‐antigen conjugate. This nano‐vaccine stimulates a strong tumor‐specific T cell response by activating antigen presentation and toll‐like receptor signaling pathways. By markedly improving the efficiency of antigen/adjuvant co‐delivery to the draining lymph nodes, the nano‐vaccine leads to 100% tumor prevention for up to 11 months and without tumor recurrence, heralding the generation of long‐term anti‐tumor memory. Moreover, the injection of nano‐vaccine with signal neoantigen eliminates the established MC‐38 tumor (a cell line of murine carcinoma of the colon without exogenous OVA protein expression) in 40% of the mice by inducing potent cytotoxic T lymphocyte infiltration in the tumor microenvironment without substantial systemic toxicity. These findings represent that stapling peptide‐based nano‐vaccine may serve as a facile, general, and safe strategy to stimulate a strong anti‐tumor immune response for the neoantigen peptide‐based personalized tumor immunotherapy.

To overcome the strength-toughness inversion relationship in steel/Fe matrix composites, a novel W-WC/Fe composite fiber-reinforced Fe matrix composite ((W-WC/Fe)f-IMC) with exceptional strength and toughness was fabricated by an in-situ fabrication method combining infiltration and in-situ solid-phase diffusion. The (W-WC/Fe)f-IMC consists of aligned W-WC/Fe composite fibers embedded in an Fe matrix. The W-WC/Fe composite fiber exhibits a characteristic core-shell structure on its cross section, with a resilient metal W fiber serving as the core and a robust annular WC-Fe layer acting as the shell. Due to the inward-outward growth and the gradient microstructure of the WC-Fe layer, the interface bonding strength between the W fiber/WC-Fe layer and the WC-Fe layer/matrix remains excellent. As a result, the (W-WC/Fe)f-IMC achieves an exceptional fracture strain of 35.5 ± 1.3 %, while maintaining high yield strength of 701 ± 16 MPa and ultimate compression strength of 1100 ± 18 MPa, surpassing those of cast Fe, which had values of 26.9 ± 1.1 %, 365 ± 20 MPa, and 834 ± 15 MPa, respectively. The simultaneous enhancement of the strength and toughness in (W-WC/Fe)f-IMC can be primarily attributed to the synergistic coupling effect between the high strength W-WC/Fe composite fibers and the tough matrix. The strategy proposed in this study holds promise as a method to alleviate the trade-off between strength and ductility in steel/Fe matrix composites.

IntroductionThe components of nighttime sap flux (En), which include transpiration (Qn) and stem water recharge (Rn), play important roles in water balance and drought adaptation in plant communities in water-limited regions. However, the quantitative and controlling factors of En components are unclear.MethodsThis study used the heat balance method to measure sap flow density in Vitex negundo on the Loess Plateau for a normal precipitation year (2021) and a wetter year (2022).ResultsThe results showed that the mean values were 1.04 and 2.34 g h-1 cm-2 for Qn, 0.19 and 0.45 g h-1 cm-2 for Rn in 2021 and 2022, respectively, and both variables were greater in the wetter year. The mean contributions of Qn to En were 79.76% and 83.91% in 2021 and 2022, respectively, indicating that the En was mostly used for Qn. Although the vapor pressure deficit (VPD), air temperature (Ta) and soil water content (SWC) were significantly correlated with Qn and Rn on an hourly time scale, they explained a small fraction of the variance in Qn on a daily time scale. The main driving factor was SWC between 40-200 cm on a monthly time scale for the Qn and Rn variations. Rn was little affected by meteorological and SWC factors on a daily scale. During the diurnal course, Qn and Rn initially both declined after sundown because of decreasing VPD and Ta, and Qn was significantly greater than Rn, whereas the two variables increased when VPD was nearly zero and Ta decreased, and Rn was greater than Qn.DiscussionThese results provided a new understanding of ecophysiological responses and adaptation of V. negundo plantations to increasing drought severity and duration under climate changes.

BackgroundParvimonas micra (P. micra), a Gram-positive anaerobic bacterium, exhibits colonization tendencies on oral mucosal and skin surfaces, potentially evolving into a pathogenic entity associated with diverse diseases. The diagnostic trajectory for P. micra-related diseases encounters delays, often with severe consequences, including fatality, attributed to the absence of symptom specificity and challenges in culture. The absence of a consensus on the diagnostic and therapeutic approaches to P. micra exacerbates the complexity of addressing associated conditions. This study aims to elucidate and scrutinize the clinical manifestations linked to P. micra, drawing insights from an extensive literature review of pertinent case reports.Case presentationA 53-year-old male sought medical attention at our institution presenting with recurrent hemoptysis. Empirical treatment was initiated while awaiting pathogen culture results; however, the patient’s symptoms persisted. Subsequent metagenomic next-generation sequencing (mNGS) analysis revealed a pulmonary infection attributable to P. micra. Resolution of symptoms occurred following treatment with piperacillin sulbactam sodium and moxifloxacin hydrochloride. A comprehensive literature review, utilizing the PubMed database, was conducted to assess case reports over the last decade where P. micra was identified as the causative agent.ConclusionThe literature analysis underscores the predilection of P. micra for immunocompromised populations afflicted by cardiovascular diseases, diabetes, orthopedic conditions, and tumors. Risk factors, including oral and periodontal hygiene, smoking, and alcohol consumption, were found to be associated with P. micra infections. Clinical manifestations encompassed fever, cough, sputum production, and back pain, potentially leading to severe outcomes such as Spondylodiscitis, septic arthritis, lung abscess, bacteremia, sepsis, and mortality. While conventional bacterial culture remains the primary diagnostic tool, emerging technologies like mNGS offer alternative considerations. In terms of treatment modalities, β-lactam antibiotics and nitroimidazoles predominated, exhibiting recovery rates of 56.10% (46/82) and 23.17% (19/82), respectively. This case report and literature review collectively aim to enhance awareness among clinicians and laboratory medicine professionals regarding the intricacies of P. micra-associated infections.

Large-scale rotating mechanical equipment in the mining arena plays a pivotal role in mining production, where vibration issues directly influence production efficiency and safety. This Review aims to provide a comprehensive review of the latest advancements and methodologies related to the generation mechanisms, identification, and applications of vibrational characteristics in large-scale mining rotating mechanical equipment. Semi-autogenous mills, ball mills, and coal mills are selected as archetype equipment, and the Lagrangian motion equation is employed to unveil the generation mechanisms of vibrations and the embedded physical information in the signals of these machines. Initially, the research delves deeply into the acquisition, extraction, and identification of vibrational signal features, emphasizing that while mechanical vibration signals can reveal the internal operational state and fault information of machinery, there remains a need to enhance their capability to depict complex vibrational signals. Subsequently, this Review discusses in depth the studies focused on predicting the vibrational state of equipment by establishing accurate and reliable soft measurement models, pointing out that current models still have room for improvement in prediction accuracy and generalization capabilities. Conclusively, based on the elucidation of mechanical vibration mechanisms and the collation and outlook of the existing research study, the importance of on-site monitoring, deep learning, Internet of Things technology, and full lifecycle management is accentuated. To better support practical engineering applications, further exploration into the physical properties of vibrational signals and the mechanisms of mechanical vibrations is essential.

The Loess Plateau of China is facing increasingly serious challenges related to water shortage issues despite the successful vegetation restoration. Surface soil water (0–40 cm) is crucial to water transfer and discharge in the soil-plant-atmosphere continuum, which is controlled by precipitation (P), air temperature (T a ), and plant components. This study selected one typical grass species and set different treatments of natural condition (NC), removing litter (CR), and only roots (OR), to analyze the responses of soil water storage (W) and its change (∆W) to P and T a at different time scales (including daily, weekly and monthly scales). The results showed that P had significantly positive effects on W and ∆W, while T a negatively influenced W and ∆W. Meanwhile, T a also controlled the significance of P affecting W. P contributed more (53–63%) to ∆W variation at the daily scale, but T a showed more effect (73–80%) at the weekly scale, and both of them contributed more than 77% at the monthly scale. P and T a existed a threshold phenomenon for increasing soil water, where P thresholds were higher than 0.93, 4.98, and 28.17 mm and Ta threshold were lower than 28.69, 28.89, and 29.96°C at the daily, weekly and monthly scales, respectively. Moreover, grass components also affected the responses of ∆W to P and T a , and the OR treatment displayed the lowest sensitivity and had the lowest loss rate of soil water. The total infiltration, total loss amount, and rainwater retention rate exhibited: NC > CR > OR. Overall, grass canopy and litter improved soil water condition, and the roots most strongly protected soil water after infiltration. This study can provide a better understanding of surface soil water responding to climate change under drought stress with water shortage implications, and may strength the management of grassland in semi-arid region.

BackgroundAnal cancer, mainly attributed to human papillomavirus (HPV) infection, is rising in prevalence among the general population in Pakistan. This study aimed to examine the knowledge, attitudes, and practices (KAP) towards anal cancer screening and HPV of the general population in Pakistan.MethodWe surveyed anal cancer KAP using social media and snowball sampling from December 2022 to May 2023. The questionnaire had 16 knowledge, 12 attitudes, 6 practice questions, and socio-demographic variables. We applied validity criteria for inclusion and exclusion and used cutoffs ≥50% for each KAP category. We analyzed data in R with Guttman’s λ2 for reliability, did univariate and bivariate analysis, and reported frequencies, percentages, p-values, coefficients, odds ratios, and 95% confidence intervals.ResultsWe surveyed 1620 people and discovered low awareness of HPV and anal cancer causes prevention, and screening (11%-24%), high stigma and embarrassment for screening (54%-70%), strong moral beliefs (89%), condom nonuse (91%), and low engagement in health services and programs (9.1%-14%). Knowledge (75.23%, OR = 1.0984, p = 0.05) was shaped by socio-demographic factors, attitude, and practice, with higher education enhancing knowledge (OR = 1.0984, p = 0.05). Attitude (78.45%, OR = 6.6052, p< 0.001) was influenced by socio-demographic factors, practice, and knowledge as well. Younger females, single, unemployed, students, living with more family members, earning more income, and residing in Islamabad had a more positive attitude (ORs from 1.0115 to 6.6052, p< 0.05), while religion did not affect attitude (p = 0.51). Practice (9.16%, OR = 0.1820, p< 0.001) was determined by socio-demographic factors, knowledge, and attitude. Older males, employed teachers, living with more family members, earning less income, and residing in Islamabad had better practice (ORs from 0.1323 to 3.8431, p< 0.05), but marital status and religion did not influence practice (p > 0.05).ConclusionPakistani young adults need more education, awareness, health services, and programs on HPV and anal cancer, as they have low awareness, high stigma, and socio-cultural challenges. In addition, it is recommended for more research and policy initiatives are needed to address socio-cultural factors and increase anal Pap to overcome anal cancer.

Abstract In filter bank multi‐carrier (FBMC) with offset quadrature amplitude modulation (OQAM) systems, the major limitation of FBMC‐OQAM signaling is the high peak‐to‐average power ratio (PAPR). High PAPR results in increased hardware complexity and is also prone to non‐linear effects of high‐power amplifiers (HPA). The existing HPA energy‐efficient optimization strategies are constrained severely by the scarcity and heterogeneity of signal distribution in the degree of freedom. This paper theorizes the optimal FBMC‐OQAM signal distribution, which has a global and steady feature and evaluates the maximal HPA efficiency‐suited multi‐carrier modulation (MCM) signal. This paper first proposes that the Binary Particle Swarm Optimization based Partial Transmit Sequence (BPSO‐PTS) with the Input‐Back Off Modulation Error Ratio (IBO‐MER) scheme traces the signal distribution for maximal HPA efficiency to save supply‐side energy. This paper further proposes Continuous‐Unconstrained Particle Swarm Optimization based PTS(CUPSO‐PTS) with IBO‐MER to obtain the theoretical boundaries and drastically accelerate convergence in the continuous‐unconstrained searching space. To decrease the computational complexity of MER calculation, this paper proposes the CUPSO with IBO Signal‐to‐Distortion Ratio (IBO‐SDR), enabling it to meet more practical applications. The simulation results show that the proposed scheme outperforms conventional IBO and HPA efficiency schemes.

The cement industry, as one of the primary contributors to global greenhouse gas emissions, accounts for 7% of the world’s carbon dioxide emissions. There is an urgent need to establish a rapid method for detecting cement plants to facilitate effective monitoring. In this study, a comprehensive method based on YOLOv5-IEG and the Thermal Signature Detection module using Google Earth optical imagery and SDGSAT-1 thermal infrared imagery was proposed to detect large-scale cement plant information, including geographic location and operational status. The improved algorithm demonstrated an increase of 4.8% in accuracy and a 7.7% improvement in MAP@.5:95. In a specific empirical investigation in China, we successfully detected 781 large-scale cement plants with an accuracy of 90.8%. Specifically, of the 55 cement plants in Shandong Province, we identified 46 as operational and nine as non-operational. The successful application of advanced models and remote sensing technology in efficiently and accurately tracking the operational status of cement plants provides crucial support for environmental protection and sustainable development.

Geochemical composition changes in groundwater related to earthquakes have been documented in previous studies, and most such studies focused on the changes in major ions, hydrogen, oxygen isotopes, and geochemical gases. Changes in trace elements were suggested to be more sensitive to small earthquakes than the commonly used chemical constituents such as major ions, yet they received less attention. Beijing is located in the Zhangjiakou-Bohai seismic belt and experiences frequent occurrences of small earthquakes. In this paper, we collected groundwater samples from a hot spring in Yanqing district of Beijing weekly from August 2021 to August 2022. Each water sample contained 41 trace chemical compositions. During the sampling, an earthquake with a magnitude of ML3.3 occurred in the Chaoyang district of Beijing on 3 February 2022, so these trace elements changes were systematically monitored before and after the earthquake: Li, Sc, Ti, and Pb elements had upward changes before the earthquake, while Cu, Nb, Th, Zn, Tl, and U elements had downward changes before the earthquake. Eu (rare earth elements) had upward changes after the earthquake. At the same time, the earthquake caused no significant changes in the groundwater level in the seismic monitoring well near the Songshan spring. Such responses indicate that trace elements are likely to be more sensitive to crustal strain than groundwater level. We considered that the earthquake-induced rock cracks before or after the earthquake caused enhancing water-rock interaction and led to the migration of trace elements between the water column and rocks, which is the mechanism to explain the trace elemental changes. This study probably provides a comprehensive assessment of the sensitivity of trace element constituents to the earthquake. Furthermore, we suggest that more long-term continuous monitoring and research of trace elements in Beijing and Zhangjiakou-Bohai Fault Zone should be considered to explore the response mechanism of groundwater geochemistry to earthquakes in the future.

BackgroundBreast Cancer (BC) stands out as the widely prevalent malignancy among all the types of cancer affecting women worldwide. There is significant evidence that the pathogenicity of BC may be altered by Human Papillomavirus (HPV) infection; however, conclusive data are not yet available.MethodsBy searching five databases, including EMBASE, IBECS, PubMed, Scopus, Science Direct, Google Scholar, and Web of Science, a thorough systematic analysis was conducted on the prevalence of HPV in BC patients from 1990 to June 30, 2022. After applying extensive eligibility criteria, we selected 74 publications for further analysis based on the prevalence of HPV infections in breast tissues. All of the data were analyzed using a random-effects meta-analysis, Cochran Q test and I2 statistic were used to calculate the heterogeneity of the prevalence among these studies using subgroup analysis. Variations in the HPV prevalence estimates in different subgroups were evaluated by subgroup meta-analysis.ResultsIn total, 3156 studies were initially screened, resulting in 93 full-text studies reviewed, with 74 meeting inclusion criteria. Among a total of 7156 BC biopsies, the pool prevalence of HPV was 25.6% (95% CI= 0.24-0.33, τ2 = 0.0369 with significant heterogeneity between estimates (I2 = 97% and p< 0.01). Consequently, 45 studies with available controls were further studied, and the prevalence of HPV in case-control studies was 26.2% with overall odds 5.55 (95% CI= 3.67-8.41, I2 = 38%, τ2 = 1.4878, p< 0.01). Further subgroup analysis of HPV revealed HPV-16 had a maximum prevalence of 9.6% (95% CI= 3.06-11.86, I2 = 0%, τ2 = 0.6111, p< 0.01). Among different geographical regions, Europe reported the maximum prevalence of HPV, i.e., 39.2% (95% CI=1.29-7.91, I2 = 18%, τ2 = 1.2911, p< 0.01). Overall distribution showed HPV-18 was a frequent HPV subtype reported in Australia.ConclusionCurrent study provides a global estimate of HPV prevalence in BC patients and demonstrates a significant association between this virus and BC etiology. Nevertheless, we recommend further investigation into the underlying mechanism is essential to validate this hypothesis.

IntroductionRainfall events can determine a cascade of plant physiological and ecological processes, and there is considerable interest in the way that rainfall modifies plant water flux dynamics.MethodsThe sap flow density (SF) of the planted species of Vitex negundo and Hippophae rhamnoides, on the Loess Plateau of China was monitored using the heat balance method from 2015 to 2017.Results and discussionThe results showed that SF responded differently to rainfall classes because of the changing meteorological and soil water content (SWC) conditions. For class 1: 0.2–2 mm, SF increased by 14.36–42.93% for the two species, which were mainly attributable to the effect of solar radiation and vapor pressure deficit after rainfall. For class 2: 2–10 mm, SF remained nearly stable for V. negundo and decreased for H. rhamnoides because of the relative humidity’s effect. For class 3: > 10 mm, SF increased significantly because of increased SWC and the increasing response to solar radiation. The increased percentage of SF was relatively higher for V. negundo when rainfall was less than 20 mm, while the value was higher for H. rhamnoides when rainfall was greater than 10 mm. Further, V. negundo’s water potential increased at the soil–root interface (ψ0) and ψL, indicating that the plant, which has shallower roots and a coarser of leaf and bark texture, considered as anisohydric species and used precipitation-derived upper soil water to survive. The relatively consistent ψL and ψ0 for H. rhamnoides, which has deep roots and leathery leaves, indicated that this species was considered as isohydric species and insensitive to the slight change in the soil water status. The differed response patter and water use strategies between the two species showed that species as V. negundo are more susceptible to frequent, but small rainfall events, while larger, but less frequent rainfall events benefit such species as H. rhamnoides. This study quantified the effect of environmental factors for SF variation. The results could help formulate a selection process to determine which species are more suitable for sustainable management in the afforestation activities under the context of more frequent and intense rainfall events.

Mg-based alloys are regard to be suitable hydrolysis materials for hydrogen generation. The mechanico-chemical reaction (MCR) strategy was employed to effectively activate Mg-based binary alloys as well as elevate their environmental stability. MCR is proved to be an effective way to activate Mg-based binary alloys and enhance the ability to resist environmental toxicity. The hydrolysis H2 generation process of MCR Mg25Ni, Mg30Ce and Mg30La samples after environmental exposure are elevated and the final H2 capacities can increase to 186.1, 204.8 and 341.4 mL·g−1 with the fastest H2 generation rates of 26.0, 24.2 and 41.5 mL·g−1·min−1, respectively. The generation capacities of the above-mentioned samples are 297, 370 and 629 mL·g−1 at 318 K within 10 min, respectively. Mg-based binary alloys can be effectively activated by MCR and Maintain high reactivity at the same time. The hydrogen generation activation energies of MCR Mg25Ni, Mg30Ce and Mg30La after environmental exposure are 26.6, 30.8 and 23.3 kJ·mol−1, respectively. The above-mentioned work may provide a feasible strategy to activate the as-cast Mg-based alloys and elevate the Resistance to environmental toxicity, which may pave the way for applying the portable hydrogen generators.

A challenge for synthetic biology is the design and construction of prototissue. Here, the authors spatially segregate layers of enzyme-decorated coacervate protocells as a model prototissue capable of chemical signal processing and modulating outputs of nitric oxide to inhibit blood clot formation.

(1) Background: DNA damage in cumulus cells hinders oocyte maturation and affects steroid hormone secretion. It is crucial to identify the key factors that regulate cellular DNA damage and steroid hormone secretion. (2) Methods: Treatment of bovine cumulus cells with bleomycin to induce DNA damage. The effects of DNA damage on cell biology were determined by detecting changes in DNA damage degree, cell cycle, viability, apoptosis, and steroid hormones. It was verified that mir-302d targeted regulation of CDKN1A expression, and then affected DNA damage and steroid hormone secretion in cumulus cells. (3) Results: Bleomycin induced increased DNA damage, decreased G1-phase cells, increased S-phase cells, inhibited proliferation, promoted apoptosis, affected E2 and P4 secretion, increased CDKN1A expression, and decreased miR-302d expression. Knockdown of CDKN1A reduced DNA damage, increased G1-phase cells, decreased G2-phase cells, promoted proliferation, inhibited apoptosis, increased E2 and P4 secretion, and increased the expression of BRCA1, MRE11, ATM, CDK1, CDK2, CCNE2, STAR, CYP11A1, and HSD3B1. The expression of RAD51, CCND1, p53, and FAS was decreased. Overexpression of CDKN1A resulted in the opposite results. miR-302d targets CDKN1A expression to regulate DNA damage and then affects the cell cycle, proliferation, apoptosis, steroid hormone secretion, and the expression of related genes. (4) Conclusions: miR-302d and CDKN1A were candidate molecular markers for the diagnosis of DNA damage in bovine cumulus cells.

Soil water shortage has become a severe issue in ecological restoration and sustainable development in the Loess Plateau, facing the challenges of climate change and vegetation restoration. This study monitored the soil water content in surface soil (0–40 cm) with different sub-shrub component treatments, including the natural condition (NC), the canopy plus the roots (CR) and only the roots (OR), to analyze the change in soil water storage (∆W) and its response to precipitation (P) and air temperature (Ta) on a daily scale. P was the main factor controlling the daily ∆W, contributing 49–52% to the variation in the daily ∆W, and Ta only explained 6–21% of the variation. Minimum P amounts of 0.74–1.12 mm and maximum Ta of 29.09–32.00 °C were the thresholds required to increase soil water storage (W). Sub-shrub components showed significant influences on soil water conservation. We found that the ∆W hierarchy for each sub-shrub treatment was NC (1.73 mm) > CR (0.71 mm) > OR (0.56 mm) on rainy days and NC (−0.53 mm) < CR (−0.36 mm) < OR (−0.06 mm) on no-rain days. Additionally, the hierarchy of the rainwater retention rate was NC (26.43%) > OR (13.71%) > CR (4.58%). Thus, a canopy could increase infiltration and hugely consume soil water at the same time, while litter could weaken or offset the canopy’s effects and the roots promote infiltration with little evaporation loss.

Autophagy plays an important role in the infectious processes of diverse pathogens. For instance, cellular autophagy could be harnessed by viruses to facilitate replication. However, it is still uncertain about the interplay of autophagy and swine acute diarrhea syndrome coronavirus (SADS-CoV) in cells. In this study, we reported that SADS-CoV infection could induce a complete autophagy process both in vitro and in vivo, and an inhibition of autophagy significantly decreased SADS-CoV production, thus suggesting that autophagy facilitated the replication of SADS-CoV. We found that ER stress and its downstream IRE1 pathway were indispensable in the processes of SADS-CoV-induced autophagy. We also demonstrated that IRE1-JNK-Beclin 1 signaling pathway, neither PERK-EIF2S1 nor ATF6 pathways, was essential during SADS-CoV-induced autophagy. Importantly, our work provided the first evidence that expression of SADS-CoV PLP2-TM protein induced autophagy through the IRE1-JNK-Beclin 1 signaling pathway. Furthermore, the interaction of viral PLP2-TMF451-L490 domain and substrate-binding domain of GRP78 was identified to activate the IRE1-JNK-Beclin 1 signaling pathway, and thus resulting in autophagy, and in turn, enhancing SADS-CoV replication. Collectively, these results not only showed that autophagy promoted SADS-CoV replication in cultured cells, but also revealed that the molecular mechanism underlying SADS-CoV-induced autophagy in cells.

IntroductionA study in 2006 showed that the clinical course of PEDV disease was markedly aggravated by transplacental infection of PCV2. Therefore, we investigated whether the small intestine supports PCV2 replication and the effect of PCV2 infection on PEDV replication in epithelial cells in vitro.MethodsTo confirm the intestinal tropism of PCV2, the viral loads in the small-intestinal tissues after PCV2 infection were determined with virus titration, and the viral titers in the infected pig jejunum, ileum, ileocecal valve, and colon were 104.86, 104.09, 102.52, and 102.35 TCID50/g, respectively. We then determined the propagation characteristics of PCV2 in ileal epithelial cells (IPI-FX) and jejunal epithelial cells (IPEC-J2) with an immunoperoxidase monolayer assay, virus titration, and an immunofluorescence assay. Both IPI-FX and IPEC-J2 cells supported the replication of PCV2, with titers of 105.5 and 105.0 TCID50/ml, respectively. We established an infection model of PCV2 and PEDV in IPI-FX cells and found that PEDV and PCV2 infected the cells individually and together. The effects of PCV2 infection on PEDV replication were determined with reverse transcription–quantitative PCR (qPCR), western blotting, and virus titration. When PCV2 infected IPI-FX cells before PEDV, PCV2 significantly inhibited the replication of PEDV in a dose- and time-dependent manner and that the mRNAs of IFN-β, TNF-α, IL1β, and OASL were downregulated (detected with qPCR). Surprisingly, when IPI-FX cells were co-infected with PCV2 and PEDV, PCV2 promoted the replication of PEDV, the expression of the host IFN-β, TNF-α, IL1β, and OASL mRNAs was upregulated.DiscussionThese findings demonstrate that the co-infection of IPI-FX cells with PCV2 and PEDV represents an excellent in vitro model in which to investigate their combined pathogenic mechanisms.

Very thin free-flowing liquid sheets are promising targets for high-repetition-rate laser-ion acceleration. In this work, we report the generation of micrometer-thin free-flowing liquid sheets from the collision of two liquid jets, and study the vibration and jitter in their surface normal direction. The dependence of their motion amplitudes on the generation parameters is studied in detail. The origins of the vibration and jitter are discussed. Our results indicate that when the generation parameters are optimized, the motion amplitudes in the stable region can be stabilized below 3.7 μm to meet the stringent requirement of sheet position stability for a tight-focusing setup in laser-ion acceleration experiments.

BackgroundThe cerebellum is involved in the control and coordination of movements but it remains unclear whether stimulation of the cerebellum could improve the recovery of upper limb motor function. Therefore, this study aimed to explore whether cerebellar transcranial direct current stimulation (tDCS) therapy could promote the recovery of upper limb motor function in patients who suffered a stroke.MethodsIn this randomized, double-blind, and sham-controlled prospective study, 77 stroke patients were recruited and randomly assigned to the tDCS group (n = 39) or the control group (n = 38). The patients received anodal (2 mA, 20 min) or sham tDCS therapy for 4 weeks. The primary outcome was the change in the Fugl-Meyer Assessment-Upper Extremity (FMA-UE) score from baseline to the first day after 4 weeks of treatment (T1) and 60 days after 4 weeks of treatment (T2). The secondary outcomes were the FMA-UE response rates assessed at T1 and T2. Adverse events (AEs) related to the tDCS treatment were also recorded.ResultsAt T1, the mean FMA-UE score increased by 10.7 points [standard error of the mean (SEM) = 1.4] in the tDCS group and by 5.8 points (SEM = 1.3) in the control group (difference between the two groups was 4.9 points, P = 0.013). At T2, the mean FMA-UE score increased by 18.9 points (SEM = 2.1) in the tDCS group and by 12.7 points (SEM = 2.1) in the control group (the difference between the two groups was 6.2 points, P = 0.043). At T1, 26 (70.3%) patients in the tDCS group had a clinically meaningful response to the FMA-UE score compared to 12 (34.3%) patients in the control group (the difference between the two groups was 36.0%, P =0.002). At T2, 33 (89.2%) patients in the tDCS group had a clinically meaningful response to the FMA-UE score compared with 19 (54.3%) patients in the control group (the difference between the two groups was 34.9%, P = 0.001). There was no statistically significant difference in the incidence of adverse events between the two groups. In the subgroup analysis of different hemiplegic sides, the rehabilitation effect of patients with right hemiplegia was better than that of patients with left hemiplegia (P < 0.05); in the age subgroup analysis, different age groups of patients did not show a significant difference in the rehabilitation effect (P > 0.05).ConclusionCerebellar tDCS can be used as an effective and safe treatment to promote recovery of upper limb motor function in stroke patients.Trial registrationChiCTR.org.cn, identifier: ChiCTR2200061838.

BackgroundColon cancer represents one of the most pervasive digestive malignancies worldwide. Translocase of the outer mitochondrial membrane 34 (TOMM34) is considered an oncogene and is implicated in tumor proliferation. However, the correlation between TOMM34 and immune cell infiltration in colon cancer has not been investigated.Materials and methodsBased on multiple open online databases, we performed integrated bioinformatics analysis of TOMM34 to evaluate the prognostic value of TOMM34 and its correlation with immune cell infiltration.ResultsTOMM34 gene and protein expression levels were elevated in tumor tissues compared with normal tissues. Survival analysis revealed that upregulation of TOMM34 was significantly associated with poorer survival time in colon cancer. High TOMM34 expression was dramatically related to low levels of B cells, CD8+ T cells, neutrophils, dendritic cells, PD-1, PD-L1 and CTLA-4.ConclusionsOur results confirmed that high expression of TOMM34 in tumor tissue correlates with immune cell infiltration and worse prognosis in colon cancer patients. TOMM34 may serve as a potential prognostic biomarker for colon cancer diagnosis and prognosis prediction.

BackgroundFor breast cancer (BC) with sentinel lymph node micrometastases (SLNMs), there are limited data to guide the selection of postoperative adjuvant therapy. This study aimed to identify target populations who might benefit most from adjuvant therapy and examine prognostic factors among patients with T1-2N1miM0 BC with one or two SLNMs who underwent sentinel lymph node biopsy (SLNB) alone.MethodsThere were 7,423 patients diagnosed with T1-2N1miM0 BC between 2010 and 2015, and patients with one or two SLNMs were extracted from the Surveillance, Epidemiology, and End Results database. All the patients underwent SLNB alone without further axillary lymph node dissection, and they were stratified according to adjuvant therapy. The statistical significance of categorical variables was analyzed using the χ2 test. Univariable and multivariable Cox analyses were used to analyze characteristics predictive of Breast-cancer-specific survival and overall survival (OS). Kaplan–Meier methods with the log-rank test was analyzed to compare survival difference between the different treatments.ResultsAdjuvant chemotherapy and radiotherapy improved 5-year OS rates. Multivariate analysis revealed that age ≥70 years, high grade, T2 stage, triple-negative subtype, and absence of radiotherapy were poor prognostic factors for OS. Patients who received breast-conserving surgery (BCS), and those with invasive ductal carcinoma (IDC), luminal A, luminal B, or basal-like subtype, and T1c or T2 stage benefited from adjuvant radiotherapy. Patients who received BCS, and those with IDC, luminal A subtype, and T1b, T1c, or T2 stage benefited from adjuvant chemotherapy.ConclusionOur findings provide a clinical evaluation of treatment choice after surgery, which may help clinicians make individualized clinical decisions.

IntroductionIt has been believed that breast-conserving therapy (lumpectomy plus adjuvant radiation, Lum + RT) and mastectomy without radiation (Mast + NoRT) have equivalent survival outcomes. However, there is a need to re-evaluate the role of lumpectomy plus adjuvant radiation due to changed breast cancer management over time. This study aimed to conduct a population-based study that compare long-term oncologic survival outcomes after Lum + RT vs Mast + NoRT.MethodsThe Surveillance, Epidemiology and End Results database was used to identify female breast cancer patients with a primary localized breast cancer diagnosis from 1988 to 2018. The standardized incidence/mortality ratio (SIR/SMR) for breast cancer recurrence (BCR) and breast cancer-specific death (BSD) was estimated by the SEER*Stat program. Cumulative incidences of BCR and BSD were assessed using Gray’s method. We evaluated the effects of Lum + RT vs. Mast + NoRT on breast cancer recurrence-free survival (BRFS) and breast cancer-specific survival (BCSS). Fine-Gray competing risk model analyses, propensity score-adjusted Kaplan-Meier analyses and Cox proportional hazards model analyses were applied.ResultsA total of 205,788 women were included in the study. Patients who underwent Lum + RT had higher SIR of BCR (4.14 [95% confidence interval, CI: 3.94-4.34] vs. 1.11 [95% CI: 1.07-1.14]) and lower SMR (9.89 [95% CI: 9.71-10.08] vs. 17.07 [95% CI: 16.82-17.33]) than patients who underwent Mast + NoRT. Lum + RT was associated with higher competing risk of BCR (adjusted hazard ratio [HR]: 1.996, 95% CI: 1.925-2.069, p < 0.001) and lower competing risk of BSD when compared to Mast + RT (adjusted HR: 0.584, 95% CI: 0.572-0.597, p < 0.001). Multivariate Cox regression analysis revealed similar results (adjusted HR after PSW for BRFS: 1.792, 95% CI 1.716-1.871, p < 0.001; adjusted HR after PSW for BCSS: 0.706, 95% CI 0.688-0.725, p < 0.001). These findings persisted in the sensitivity and subgroup analyses.DiscussionThe present study further confirmed superior long-term survival with lumpectomy plus adjuvant radiation over mastectomy independent of patient characteristics including age, race, time period, historic subtype, tumor size, historic grade and stage, indicating that this benefit may result from the treatment itself.

Colorectal cancer (CRC) is one of the most common cancers worldwide. Accurate early detection and diagnosis, comprehensive assessment of treatment response, and precise prediction of prognosis are essential to improve the patients’ survival rate. In recent years, due to the explosion of clinical and omics data, and groundbreaking research in machine learning, artificial intelligence (AI) has shown a great application potential in clinical field of CRC, providing new auxiliary approaches for clinicians to identify high-risk patients, select precise and personalized treatment plans, as well as to predict prognoses. This review comprehensively analyzes and summarizes the research progress and clinical application value of AI technologies in CRC screening, diagnosis, treatment, and prognosis, demonstrating the current status of the AI in the main clinical stages. The limitations, challenges, and future perspectives in the clinical implementation of AI are also discussed.

Abstract Background Previous cluster-randomized controlled trials evaluating the impact of implementing evidence-based guidelines for nutrition therapy in critical illness do not consistently demonstrate patient benefits. A large-scale, sufficiently powered study is therefore warranted to ascertain the effects of guideline implementation on patient-centered outcomes. Methods We conducted a multicenter, cluster-randomized, parallel-controlled trial in intensive care units (ICUs) across China. We developed an evidence-based feeding guideline. ICUs randomly allocated to the guideline group formed a local "intervention team", which actively implemented the guideline using standardized educational materials, a graphical feeding protocol, and live online education outreach meetings conducted by members of the study management committee. ICUs assigned to the control group remained unaware of the guideline content. All ICUs enrolled patients who were expected to stay in the ICU longer than seven days. The primary outcome was all-cause mortality within 28 days of enrollment. Results Forty-eight ICUs were randomized to the guideline group and 49 to the control group. From March 2018 to July 2019, the guideline ICUs enrolled 1399 patients, and the control ICUs enrolled 1373 patients. Implementation of the guideline resulted in significantly earlier EN initiation (1.20 vs. 1.55 mean days to initiation of EN; difference − 0.40 [95% CI − 0.71 to − 0.09]; P = 0.01) and delayed PN initiation (1.29 vs. 0.80 mean days to start of PN; difference 1.06 [95% CI 0.44 to 1.67]; P = 0.001). There was no significant difference in 28-day mortality (14.2% vs. 15.2%; difference − 1.6% [95% CI − 4.3% to 1.2%]; P = 0.42) between groups. Conclusions In this large-scale, multicenter trial, active implementation of an evidence-based feeding guideline reduced the time to commencement of EN and overall PN use but did not translate to a reduction in mortality from critical illness. Biomolecular channels on the cell membrane are essential for transporting substances across the membrane to maintain cell physiological activity. Artificial transmembrane channels used to mimic biological membrane channels can regulate intra/extracellular ionic and molecular homeostasis, and they elucidate cellular structures and functionalities. Due to their program design, facile preparation, and high biocompatibility, DNA nanostructures have been widely used as scaffolds for the design of artificial transmembrane channels and exploited for ionic and molecular transport and biomedical applications. DNA-based artificial channels can be designed from two structural modules: DNA nanotubes/nanopores as transport modules for mass transportation and hydrophobic segments as anchor modules for membrane immobilization. In this review, various lipophilic modification strategies for the design of DNA channels and membrane insertion are outlined. Several types of DNA transmembrane channels are systematically summarized, including DNA wireframe channels, DNA helix bundle channels, DNA tile channels, DNA origami channels, and so on. We then discuss efforts to exploit them in biosensor and biomedical applications. For example, ligand-gated and environmental stimuli-responsive artificial transmembrane channels have been designed for transmembrane signal transduction. DNA-based artificial channels have been developed for cell mimicry and the regulation of cell behaviors. Finally, we provide some perspectives on the challenges and future developments of artificial transmembrane channel research in biomimetic science and biomedical applications.

Abstract Background This study investigated an association between schizoid tendencies and aggressive behaviors in Chinese adolescents, and explored the underlying mechanism. Methods The data of 3094 adolescents aged 12 to 16 years were collected from an epidemiological survey in China. All the subjects or their parents completed the Achenbach’s Child Behavior Checklist, the Childhood Trauma Questionnaire-Short Form, and the Adolescent Self-rating Life Event Checklist (ASLEC). Results Relative to the non-schizoid group, adolescents with schizoid tendencies (male or female) showed significantly higher scores for aggressive behaviors, emotional abuse, and ASLEC. Regarding females only, those with schizoid tendencies had significantly higher (lower) scores for physical abuse and emotional neglect (physical neglect). The aggressive behaviors score was predicted by scores for schizoid tendencies (βmale = 0.620, βfemale = 0.638, both P

Abstract Chemotherapy-induced intestinal mucositis (CIM) is a common adverse reaction to antineoplastic treatment with few appropriate, specific interventions. We aimed to identify the role of the G protein coupled estrogen receptor (GPER) in CIM and its mechanism. Adult male C57BL/6 mice were intraperitoneally injected with 5-fluorouracil to establish the CIM model. The selective GPER agonist G-1 significantly inhibited weight loss and histological damage in CIM mice and restored mucosal barrier dysfunction, including improving the expression of ZO-1, increasing the number of goblet cells, and decreasing mucosal permeability. Moreover, G-1 treatment did not alter the antitumor effect of 5-fluorouracil. In the CIM model, G-1 therapy reduced the expression of proapoptotic protein and cyclin D1 and cyclin B1, reversed the changes in the number of TUNEL+ cells, Ki67+ and bromodeoxyuridine+ cells in crypts. The selective GPER antagonist G15 eliminated all of the above effects caused by G-1 on CIM, and application of G15 alone increased the severity of CIM. GPER was predominantly expressed in ileal crypts, and G-1 inhibited the DNA damage induced by 5-fluorouracil in vivo and vitro, as confirmed by the decrease in the number of γH2AX+ cells in the crypts and the comet assay results. Referring to the data from GEO dataset we verified GPER activation restored ERK1/2 activity in CIM and 5-fluorouracil-treated IEC-6 cells. Once the effects of G-1 on ERK1/2 activity were abolished with the ERK1/2 inhibitor PD0325901, the effects of G-1 on DNA damage both in vivo and in vitro were eliminated. Correspondingly, all of the manifestations of G-1 protection against CIM were inhibited by PD0325901, such as body weight and histological changes, the mucosal barrier, the apoptosis and proliferation of crypt cells. In conclusion, GPER activation prevents CIM by inhibiting crypt cell DNA damage in an ERK1/2-dependent manner, suggesting GPER might be a target preventing CIM.

Wheat has a specific preference for NO3- and shows toxicity symptoms under high NH4+ concentrations. Increasing the nitrate supply may alleviate ammonium stress. Nevertheless, the mechanisms underlying the nitrate regulation of wheat root growth to alleviate ammonium toxicity remain unclear. In this study, we integrated physiological and weighted gene co-expression network analysis (WGCNA) to identify the hub genes involved in nitrate alleviation of ammonium toxicity at the wheat seedling stage. Five NH4+/NO3- ratio treatments, including 100/0 (Na), 75/25 (Nr1), 50/50 (Nr2), 25/75 (Nr3), and 0/100 (Nn) were tested in this study. The results showed that sole ammonium treatment (Na) increased the lateral root number but reduced root biomass. Increasing the nitrate supply significantly increased the root biomass. Increasing nitrate levels decreased abscisic acid (ABA) content and increased auxin (IAA) content. Furthermore, we identified two modules (blue and turquoise) using transcriptome data that were significantly related to root physiological growth indicators. TraesCS6A02G178000 and TraesCS2B02G056300 were identified as hub genes in the two modules which coded for plastidic ATP/ADP-transporter and WRKY62 transcription factors, respectively. Additionally, network analysis showed that in the blue module, TraesCS6A02G178000 interacts with downregulated genes that coded for indolin-2-one monooxygenase, SRG1, DETOXIFICATION, and wall-associated receptor kinase. In the turquoise module, TraesCS2B02G056300 was highly related to the genes that encoded ERD4, ERF109, CIGR2, and WD40 proteins, and transcription factors including WRKY24, WRKY22, MYB30, and JAMYB, which were all upregulated by increasing nitrate supply. These studies suggest that increasing the nitrate supply could improve root growth and alleviate ammonium toxicity through physiological and molecular regulation networks, including ROS, hormonal crosstalk, and transcription factors.

The Wangcun fossil Lagerstätte in Hunan, South China, has yielded hundreds of fossilized embryos of Markuelia hunanensis representing different developmental stages. Internal tissues have only rarely been observed, impeding further understanding of the soft tissue anatomy, phylogenetic affinity and evolutionary significance of Markuelia. In this study, we used synchrotron radiation X-ray tomographic microscopy (SRXTM) to study a new collection of fossil embryos from the Wangcun fossil Lagerstätte. We describe specimens exhibiting a spectrum of preservation states, the best of which preserves palisade structures underneath the cuticle of the head and tail, distinct from patterns of centripetal mineralization of the cuticle and centrifugal mineralization of hypha-like structures, seen elsewhere in this specimen and other fossils within the same assemblage. Our computed tomographic reconstruction of these mineralization phases preserves the gross morphology of (i) longitudinal structures associated with the tail spines, which we interpret as the proximal ends of longitudinal muscles, and (ii) a ring-shaped structure internal to the introvert, which we interpret as a ring-shaped brain, as anticipated of the cycloneuralian affinity of Markuelia. This is the first record of a fossilized nervous system in a scalidophoran, and the first instance in Orsten-style preservation, opening the potential for further such records within this widespread mode of high-fidelity three-dimensional preservation.

BackgroundThe COVID-19 pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is a global crisis. Although many people recover from COVID-19 infection, they are likely to develop persistent symptoms similar to those of myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) after discharge. Those constellations of symptoms persist for months after infection, called Long COVID, which may lead to considerable financial burden and healthcare challenges. However, the mechanisms underlying Long COVID and ME/CFS remain unclear.MethodsWe collected the genes associated with Long COVID and ME/CFS in databases by restricted screening conditions and clinical sample datasets with limited filters. The common genes for Long COVID and ME/CFS were finally obtained by taking the intersection. We performed several advanced bioinformatics analyses based on common genes, including gene ontology and pathway enrichment analyses, protein–protein interaction (PPI) analysis, transcription factor (TF)–gene interaction network analysis, transcription factor–miRNA co-regulatory network analysis, and candidate drug analysis prediction.ResultsWe found nine common genes between Long COVID and ME/CFS and gained a piece of detailed information on their biological functions and signaling pathways through enrichment analysis. Five hub proteins (IL-6, IL-1B, CD8A, TP53, and CXCL8) were collected by the PPI network. The TF–gene and TF–miRNA coregulatory networks were demonstrated by NetworkAnalyst. In the end, 10 potential chemical compounds were predicted.ConclusionThis study revealed common gene interaction networks of Long COVID and ME/CFS and predicted potential therapeutic drugs for clinical practice. Our findings help to identify the potential biological mechanism between Long COVID and ME/CFS. However, more laboratory and multicenter evidence is required to explore greater mechanistic insight before clinical application in the future.

PurposeFluorine-18 (18F)-fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) and gallium-68 (68Ga)-somatostatin analog (SSA) PET/CT imaging have been increasingly used in ectopic adrenocorticotropic hormone syndrome (EAS); however, the diagnostic efficacies of these two methods in patients with EAS remain unclear. Our study aimed to compare the diagnostic efficacies of 18F-FDG PET/CT and 68Ga-DOTANOC PET/CT in EAS.MethodsThe clinical and imaging data of 68 patients with EAS who underwent 18F-FDG PET/CT and 68Ga-DOTANOC PET/CT examinations from December 2016 to April 2021 were analyzed retrospectively, and the diagnostic efficacies of these methods were compared.ResultsIn 37 cases, imaging was performed to locate the primary tumor lesion (localization group), and in 31 to evaluate tumor load or metastasis (staging group). Primary tumors were detected in 48.65% (18/37) of the localization group patients. According to scan-based analysis, the tumor lesion detection rates and false positive rates of 18F-FDG PET/CT imaging and 68Ga-DOTANOC PET/CT imaging were 18.92% vs. 45.95% (p < 0.05) and 21.62% vs. 2.70% (p < 0.05) respectively. For lesion-based analysis, the tumor lesion detection rates and false positive rates were 24.13% vs. 58.62% (p >0.05) and 31.04% vs. 3.45% (p < 0.05). In 90.32% (28/31) of the staging group patients, 286 of 292 lesions were confirmed as tumor lesions. Based on scan analysis, the detection rates and false positive rates of 18F-FDG PET/CT imaging and 68Ga-DOTANOC PET/CT imaging were 83.87% vs. 67.74% (p > 0.05) and 12.90% vs. 9.68% (p > 0.05) respectively. Based on lesion analysis, the detection rate and false positive rates were 93.84% vs. 54.80% (p < 0.05) and 1.37% vs. 1.03%(p > 0.05).Conclusion68Ga-DOTANOC PET/CT imaging may be more suitable than 18F-FDG PET/CT for identifying the primary tumor in patients with EAS, while 18F-FDG PET/CT may be more advantageous than 68Ga-DOTANOC PET/CT for patients with suspected metastasis.

AimsThis study aimed to evaluate the predictive value of the combined prognostic nutritional index (PNI) and GGT/ALT for the postoperative prognosis of patients with hepatocellular carcinoma (HCC) within Milan criteria undergoing radical hepatectomy.MethodsThis single-center retrospective study included 283 patients with HCC within the Milan criteria who underwent hepatectomy. The receiver operating characteristic (ROC) curve was used to calculate the optimal PNI and GGT/ALT cut-off values. Pre-treatment PNI, GGT/ALT, and PNI-GGT/ALT grades were calculated. Overall survival (OS) and recurrence-free survival (RFS) were estimated using the Kaplan–Meier method, and multivariate analysis was used to identify prognostic factors.ResultsMultivariate Cox regression analysis identified that the PNI, GGT/ALT, tumor number were significant prognostic markers for OS, and that the GGT/ALT, tumor number were significant prognostic markers for OS. The survival curves showed that low PNI, high GGT/ALT ratio, and high PNI-GGT/ALT grade were associated with poorer OS and DFS. With an area under the curve (AUC) of 0.690, PNI-GGT/ALT outperformed each individual score.ConclusionPNI-GGT/ALT, a new prognostic scoring model, qualifies as a novel prognostic predictor for patients with HCC within the Milan criteria after curative resection.

Variation in the relative abundance of the stable carbon isotopes has been widely used to correlate Ordovician marine successions over the past two decades. To date, only a few of studies of Ordovician carbon chemostratigraphy have been conducted in South China. Most of the previous studies in this field have focused on specific time intervals and/or events in the Middle and Upper Ordovician. The Lower and Middle Ordovician of the Yangtze Platform is typically represented by a sedimentary succession dominated by carbonate rocks, which is ideal for studying the carbon chemostratigraphy. Three sections spanning the Nantsinkuan/Lunshan, Fenhsiang, Hunghuayuan, and Dawan/Zitai formations, corresponding to the TremadocianâDapingian in age, have been sampled for high-resolution δ13C chemostratigraphy. Our new δ13C data reveal five tie-points with the potential for global correlation: (1) a positive δ13C excursion in the lower Nantsinkuan Formation within the Tremadocian Rossodus manitouensis Zone; (2) an excursion with two peaks roughly within the late Tremadocian Paltodus âdeltiferâ Zone; (3) a positive δ13C shift in the lower Hunghuayuan Formation, within the early Floian Serratognathus diversus Zone; (4) a gradual positive δ13C shift in the late Floian, ranging from the uppermost S. diversus Zone to the basal Oepikodus evae Zone; (5) a minor negative shift in the lower Dawan/Zitai Formation, within the early Dapingian Baltoniodus triangularis Zone. These excursions are herein used for correlation of the Yangtze Platform strata with successions from South China, North China, the Argentine Precordillera, North America and Baltica. From a palaeogeographical perspective, the Gudongkou, Xiangshuidong and Daling sections represent depositional environments along an inner to outer ramp profile. The δ13C data from these sections show successively heavier (higher) δ13C values with increasing depositional depth, which can be interpreted as due to remineralization of organic carbon within the carbonate rocks formed in the shallow-water environment.

Abstract Backgrounds Associations between attention deficit hyperactivity disorder (ADHD) subtypes and suicidal behaviors remains unclear. The current study explored the prevalence of suicidal behaviors, and its association with ADHD among Chinese medical students. Methods Five thousand six hundred ninety-three medical college students participated. Symptoms of suicidal behaviors, ADHD, anxiety, depression, tobacco and alcohol use were assessed using online questionnaires. Results The prevalence of lifetime suicidal ideation, suicide plans, and suicide attempts among medical college students were 27.5, 7.9 and 14.8% respectively. Participants with ADHD predominantly inattentive type (ADHD-I) had more than fivefold increased odds of suicidal behaviors, the adjusted odds ratios (ORs) of ADHD-I and ADHD combined type (ADHD-C) remained significant after controlling for confounding factors. Conclusions ADHD is associated with high risk of suicidal behaviors. ADHD-I and ADHD-C were strongly associated with suicidal behaviors independent of comorbidities. The finding suggests the importance of addressing ADHD symptoms in suicide prevention.

Swine acute diarrhea syndrome coronavirus (SADS-CoV) is an emerging swine enteric coronavirus that causes vomiting, severe diarrhea, dehydration and death in suckling piglets. NS7a is putative accessory protein that is predicted to be encoded by SADS-CoV, but still to be confirmed experimentally. In the present study, recombinant NS7a protein was expressed in a prokaryotic expression system and used as an antigen to prepare monoclonal antibodies (mAbs) specific to NS7a protein. We obtained two anti-NS7a mAbs, termed AH5 and EH3, that were shown by western blotting to react with the natural NS7a protein in Vero E6 cells infected with SADS-CoV. Using the produced mAbs, we observed by confocal microscopy that NS7a protein was expressed in the cytoplasm. Further studies revealed that the motif 31VNTWQEFA38 was the minimal unit of the linear B-cell epitope recognized by mAb AH5, and the motif 82FDLFERF88 was the minimal unit of the linear B-cell epitope recognized by mAb EH3. Alignment of amino acids showed that these two epitopes were highly conserved among different SADS-CoV strains and SADS-related coronaviruses from bats, but with one substitution in these two motifs in bat coronavirus HKU2. In summary, we generated and characterized two mAbs against SADS-CoV NS7a protein, and demonstrated NS7a expression in SADS-CoV-infected cells for the first time.

Forest disturbance monitoring can provide scientific data for the decision making and management of nature reserves. LandTrendr algorithm has been applied to identify forest disturbances on a long-time scale through appropriate segmentation and linear fitting. In this study, 23 nature reserves were detected using LandTrendr during 1987–2020, and the vegetation loss was quantified by years and pixel numbers. The results illustrated that (1) most disturbances occurred in the 1990s and early 21st century. (2) From the spatial distribution of forest loss, the area of forest vegetation disturbance in the coastal zone was larger than the protected area in the internal Hainan Island, the area disturbed in the coastal zone protected area was 97.12 km2, and the area disturbed in the internal area of Hainan Island protected area was 63.02 km2. (3) In terms of different levels of nature reserves, the disturbed area of national nature reserves was 28.39 km2 and the total disturbed area of provincial nature reserves was 131.75 km2. (4) In terms of different types of nature reserves, forest ecological nature reserves had the largest disturbed area of 102.96 km2, followed by marine coastal nature reserves with a disturbed area of 36.99 km2, wildlife nature reserves with a disturbed area of 10.22 km2, and wild plant nature reserves with the smallest disturbed area of 9.96 km2. The results are hoped to provide scientific support and data for the management and planning of nature reserves in Hainan Island.

Abstract SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) is a novel coronavirus causing the COVID-19 pandemic in 2020. High adaptive plasticity on the spike protein of SASR-CoV-2 enables it to transmit across different host species. In the present study, we collected 2092 high-quality genome sequences of SARS-CoV-2 from 160 regions in over 50 countries and reconstructed their phylogeny. We also analyzed the polymorphic interaction between spike protein and human ACE2 (hACE2). Phylogenetic analysis of SARS-CoV-2 suggests that SARS-CoV-2 is probably originated from a recombination event on the spike protein between a bat coronavirus and a pangolin coronavirus that endows it humans infectivity. Compared with other regions in the S gene of SARS-CoV-2, the direct-binding sites of the receptor-binding domain (RBD) is more conserved. We focused on 3,860 amino acid mutations in spike protein RBD (T333-C525) of SARS-CoV-2 and simulated their differential stability and binding affinity to hACE2 (S19-D615). The results indicate no preference for SARS-CoV-2 infectivity on people of different ethnic groups. The variants in the spike protein of SARS-CoV-2 may also be a good indicator demonstrating the transmission route of SARS-CoV-2 from its natural reservoir to human hosts.

Different from ship detection from synthetic aperture radar (SDSAR) and ship detection from spaceborne optical images (SDSOI), ship detection from visual image (SDVI) has better detection accuracy and real-time performance, which can be widely used in port management, cross-border ship detection, autonomous ship, safe navigation, and other real-time applications. In this paper, we proposed a new SDVI algorithm, named enhanced YOLO v3 tiny network for real-time ship detection. The algorithm can be used in video surveillance to realize the accurate classification and positioning of six types of ships (including ore carrier, bulk cargo carrier, general cargo ship, container ship, fishing boat, and passenger ship) in real-time. Based on the original YOLO v3 tiny network, we have made the following fine tunings. 1) The preset anchors trained on Seaship annotation data have the similar “dumpy” shape as the normal ships, helping the network to achieve faster and better training; 2) Convolution layer instead of max-pooling layer and expanding the channels of prediction network improve the small target detection ability of the algorithm. 3) Due to the problem that large-scale ships are easily disturbed by the onshore building, complex waves and light on the water surface, we introduced attention module named CBAM into the backbone network, which make the model more focused on the target. The detection accuracy of the proposed algorism is obviously better than that of the original YOLO v3 tiny work. Although it is slightly inferior to the Yolo v3 network, it has faster speed than Yolo v3. However, the proposed algorithm is a better trade-off between real-time performance and detection accuracy, and is more suitable for actual scenes. Compared with the SOAT algorithm in Z. Shao et al. (2020), our algorithm has a 9.6% improvement in mAP and a faster speed.

In the field of SAR-to-optical image synthesis, current methods based on conditional generative adversarial networks (CGANs) have satisfying performance under simple scenarios, but the performance drops severely under complicated scenarios. Considering that SAR images can form a robust time series due to SAR’s all-weather imaging ability, we take advantage of this and extract a temporal correlation from bi-temporal SAR images to guide the translation. To achieve this, we introduce a co-attention mechanism into the CGAN that learns the correlation between optically-available and optically-absent time points, selectively enhances the features of the former time point, and eventually guides the model to a better optical image synthesis on the latter time point. Additionally, we adopt a strategy to balance the weight of optical and SAR features to extract better features from the SAR input. With these strategies, the quality of synthesized images is notably improved in complicated scenarios. The synthesized images can increase the spatial and temporal resolution of optical imagery, greatly improving the availability of data for the applications of crop monitoring, change detection, and visual interpretation.

Patulin (PAT), a type of mycotoxin existing in foodstuffs, is harmful to food safety and human health. Thus, it is necessary to develop sensitive, selective and reliable analytical methods for PAT detection. In this study, a sensitive aptasensor based on a dual-signaling strategy was fabricated, in which a methylene-blue-labeled aptamer and ferrocene monocarboxylic acid in the electrolyte acted as a dual signal, for monitoring PAT. To improve the sensitivity of the aptasensor, an in-plane gold nanoparticles–black phosphorus heterostructure (AuNPs-BPNS) was synthesized for signal amplification. Due to the combination of AuNPs-BPNS nanocomposites and the dual-signaling strategy, the proposed aptasensor has a good analytical performance for PAT detection with the broad linear range of 0.1 nM–100.0 μM and the low detection limit of 0.043 nM. Moreover, the aptasensor was successfully employed for real sample detection, such as apple, pear and tomato. It is expected that BPNS-based nanomaterials hold great promise for developing novel aptasensors and may provide a sensing platform for food safety monitoring.

Soil remediation agents (SRAs) such as biochar and hydroxyapatite (HAP) have shown a promising prospect in in situ soil remediation programs and safe crop production. However, the effects of SRAs on soil microbial communities still remain unclear, particularly under field conditions. Here, a field case study was conducted to compare the effects of biochar and HAP on soil bacterial communities in a slightly Cd-contaminated farmland grown with sweet sorghum of different planting densities. We found that both biochar and HAP decreased the diversity and richness of soil bacteria, but they differently altered bacterial community structure. Biochar decreased Chao1 (−7.3%), Observed_species (−8.6%), and Shannon indexes (−1.3%), and HAP caused Shannon (−2.0%) and Simpson indexes (−0.1%) to decline. The relative abundance (RA) of some specific taxa and marker species was differently changed by biochar and HAP. Overall, sweet sorghum cultivation did not significantly alter soil bacterial diversity and richness but caused changes in the RA of some taxa. Some significant correlations were observed between soil properties and bacterial abundance. In conclusion, soil remediation with biochar and HAP caused alterations in soil bacterial communities. Our findings help to understand the ecological impacts of SRAs in soil remediation programs.

Coronavirus disease-2019 (COVID-19) pandemic started from Wuhan, China has infected more than 6.7 million individuals and killed more than 390,000 individuals globally. Due to the higher transmissibility and infectiousness, asymptomatic infection, and lack of effective treatment options and vaccine, fatalities and morbidities are increasing day by day globally. Despite physical health consequences, COVID-19 pandemic has created stress and anxiety, as result there is an increased risk of mental illnesses both in the infected and normal individuals. To eradicate these risks, it is necessary to determine the COVID-19 zoonotic source of transmission to humans and clinical manifestations in infected individuals. Although, identification or development of the highly effective therapeutic agents is necessary, however, development of protective strategies against the COVID-19 by enhancing immune responses will be an asset in the current scenarios of the COVID-19 pandemic. In this paper, we discuss the transmission, health consequences, and potential management (therapeutic and preventive) options for COVID-19 disease.
