Abstract Populus cathayana × canadansis ‘Xinlin 1’ (‘P.‘xin lin 1’) with the characteristics of rapid growth and high yield, is frequently attacked by herbivorous insects. However, little is known about how it defenses against Hyphantria cunea (H. cunea) at molecular and biochemical levels. Differences in the transcriptome and metabolome were analyzed after ‘P. ‘xin lin 1’ leaves were fed to H. cunea for 0h, 2h, 4h, 8h, 16h and 24h. In the five comparison groups including 2h vs. CK, 4h vs. CK, 8h vs. CK, 16h vs. CK, and 24h vs. CK, a total of 8925 genes and 842 metabolites were differentially expressed. A total of 825 transcription factors (TFs) were identified, which encoded 56 TF families. The results showed that the top four families with the highest number of TFs were AP2/ERF, MYB, C2C2, bHLH. Analyses of leaves which were fed to H. cunea showed that the differentially expressed genes (DEGs) and differentially accumulated metabolites (DAMs) were significantly enriched in plant hormone signal transduction pathway, MAPK signaling pathway, flavonoid, flavone and flavonol and anthocyanin biosynthesis pathway. Additionally, there were a number of genes significantly up-regulated in MAPK signaling pathway. Some compounds involved in plant hormone signal transduction and flavonoid/flavone and flavonol/ anthocyanin pathways such as jasmonic acid (JA), jasmonoyl-L-Isoleucine (JA-Ile), kaempferol and cyanidin-3-O-glucoside were induced in infested ‘P.‘xin lin 1’. This study provides a new understanding for exploring the dynamic response mechanism of poplar to the infestation of H. cunea.

Clear cell renal cell carcinoma (ccRCC) accounts for approximately 75–80% of all patients with renal cell carcinoma. Despite its prevalence, little is known regarding the key components involved in ccRCC metastasis. In this study, scRNA‐seq analysis was employed to classify CD8+ T cells into four sub‐clusters based on their genetic profiles and immunofluorescence experiments were used to validate two key clusters. Through gene set enrichment analysis, these newly identified sub‐clusters were found to exhibit distinct biological characteristics. Notably, TYMP, TOP2A, CHI3L2, CDKN3, CENPM, and RZH2 were highly expressed in these sub‐clusters, indicating a correlation with poor prognosis. Among these sub‐clusters, CD8+ T cells (MT‐ND4) were identified as potentially playing a critical role in mediating ccRCC metastasis. These results contribute to our understanding of CD8+ T cell heterogeneity in ccRCC and shed light on the mechanisms underlying the loss of immune response against cancer.

Abstract Background The purpose of this study was to investigate effect of liver Transplants (LT) with retrograde reperfusion on early postoperative recovery of liver function and its risk factors. Methods We conducted a retrospective analysis of clinical data from 136 liver transplantation (LT) patients at the 900th Hospital of the Chinese People’s Liberation Army Joint Support Army, covering the period from January 2015 to January 2021. All participants provided informed consent, adhering to medical ethics guidelines. Patients were stratified into two groups based on the liver perfusion technique used: retrograde reperfusion (RTR, n = 108) and initial portal reperfusion (IPR, n = 28). Our study focused on a subset of 23 patients from each group to compare postoperative liver function recovery. The final analysis included 86 RTR and 28 IPR cases after excluding 8 RTR patients who underwent initial hepatic artery reperfusion and 14 who received simultaneous hepatic artery and portal vein reperfusion. Further subdivision within the RTR group identified 19 patients with early hepatic allograft dysfunction (EAD) and 67 without, allowing for an assessment of the influence of preoperative and intraoperative parameters, as well as perfusion methods, on EAD incidence post-LT. Results Alanine aminotransferase (ALT) was 329 (211 ~ 548) and 176 (98 ~ 282) U/L on the 3rd and 7th day after RTR, respectively, which was significantly lower than 451 (288 ~ 918) and 251 (147 ~ 430) U/L in the IPR group (Z =-1.979, -2.299, P = 0.048, 0.021). Aspartate aminotransferase (AST) on postoperative days 3, 5, and 7 was 252 (193, 522), 105 (79, 163), and 93 (41, 135) U/L in the RTR group, respectively; it was also significantly lower than 328 (251, 724), 179 (129, 306), and 150 (91, 200)U/L in the IPR group (Z=-2.212, -3.221, -2.979; P = 0.027, 0.001, 0.003). Logistic regression analysis showed that MELD score was an independent risk factor for EAD after LT. Conclusion RTR LT is more favorable for patients’ early postoperative liver function recovery. For patients undergoing LT for RTR, preoperative MELD score was an independent risk factor for their postoperative development of EAD.

Weldability plays a crucial role in the journey of high entropy alloys towards their engineering applications. In this study, gas metal arc welding was performed to join an as-rolled CoCrFeMnNi high entropy alloy using Monel 400 as the filler wire. The present research findings demonstrate a favorable metallurgical chemical reaction between the Monel 400 filler and the CoCrFeMnNi high entropy alloy, resulting in compositional mixing within the fusion zone that promotes a solid-solution strengthening effect, counteracting the typical low hardness associated to the fusion zone of these alloys. The weld thermal cycle induced multiple microstructure changes across the joint, including variations in the grain size, existing phases and local texture. The grain size was seen to increase from the base material toward the fusion zone. An FCC matrix and finely sparse Cr-Mn-based oxides existed in both base material and heat affected zone, while in the fusion zone new FCC phases and carbides were formed upon the mixing of the Monel 400 filler. The role of the filler material on the fusion zone microstructure evolution was rationalized using thermodynamic calculations. Texture shifted from a γ-fiber (in the base material) to a strong cubic texture in the fusion zone. Digital image correlation during tensile testing to fracture coupled with microhardness mapping revealed that, stemming from the process-induced microstructure changes, the micro and macromechanical response differed significantly from the original base material. This study successfully established a correlation between the impact of the process on the developed microstructural features and the resultant mechanical behavior, effectively assessing the processing-microstructure-properties relationships towards an improved understanding of the physical metallurgy associated to these advanced engineering alloys. In conclusion, this work provides an important theoretical framework and practical guidance for optimizing the engineering applications of high entropy alloys.

BackgroundPopulus simonii, a notable native tree species in northern China, demonstrates impressive resistance to stress, broad adaptability, and exceptional hybridization potential. DOF family is a class of specific transcription factors that only exist in plants, widely participating in plant growth and development, and also playing an important role in abiotic stress response. To date, there have been no reported studies on the DOF gene family in P. simonii, and the expression levels of this gene family in different tissues of poplar, as well as its expression patterns under cold, heat, and other stress conditions, remain unclear.MethodsIn this study, DOF gene family were identified from the P. simonii genome, and various bioinformatics data on the DOF gene family, gene structure, gene distribution, promoters and regulatory networks were analyzed. Quantitative real time PCR technology was used to verify the expression patterns of the DOF gene family in different poplar tissues.ResultsThis research initially pinpointed 41 PSDOF genes in P. simonii genome. The chromosomal localization results revealed that the PSDOF genes is unevenly distributed among 19 chromosomes, with the highest number of genes located on chromosomes 4, 5, and 11. A phylogenetic tree was constructed based on the homology between Arabidopsis thaliana and P. simonii, dividing the 41 PSDOF genes into seven subgroups. The expression patterns of PSDOF genes indicated that specific genes are consistently upregulated in various tissues and under different stress conditions, suggesting their pivotal involvement in both plant development and response to stress. Notably, PSDOF35 and PSDOF28 serve as pivotal hubs in the interaction network, playing a unique role in coordinating with other genes within the family.ConclusionThe analysis enhances our comprehension of the functions of the DOF gene family in tissue development and stress responses within P. simonii. These findings provide a foundation for future exploration into the biological roles of DOF gene family.

Abstract Ti15Mo alloy has been regarded as one of the most potential biomedical materials due to its excellent performance. However, the low hardness and poor wear resistance of titanium alloy limit the further application. Therefore, high temperature solid carburising technology was performed on the surface of Ti15Mo alloys to prepare titanium carbide (TiC) coating with graphene (G) as the carburising agent. The microstructure, mechanical properties, and tribological properties of TiC coating under different lubricants were investigated. Results showed that TiC coating was closely bonded to the titanium substrate. The maximum thickness of TiC coating treated with 1150°C was approximately 184.02 μm, and the microhardness of alloys treated with 1100°C can achieve 1221.5 HV. All modified Ti15Mo alloys showed improved tribological performance compared to the original samples. The wear mechanisms of modified Ti15Mo alloys were abrasive wear and adhesive wear under the SBF lubricant, and the TiC coating was slightly peeled off. The overall friction coefficient and wear rate under 25% calf serum lubricant were lower than the SBF lubricant, and surface scratches were almost absent, and slight abrasive wear and adhesive wear occurred on the surface.

Abstract Microscopic hyperspectral imaging technology is a potential non‐destructive and non‐contact method for colour measurement of micrometre‐sized textile fibres. However, specularity on the fibre surface can distort the accurate colour information and affect the accuracy of the colour measurement. This paper proposed a specular‐constrained sparse approximation (SCSA) for specular‐diffuse reflection separation from hyperspectral images of wool fibres. First, a specular prior map is generated based on the lightness dissimilarity. Then the SCSA model is used to decompose the processed hyperspectral image A into low‐rank data L, sparse specularity data S constrained by the specular prior map, sparse noise E, and Gaussian noise N. A non‐linear logistic sigmoid function and a sparse approximation of A – L – N to S are used to improve the performance of specularity removal during iterative optimization. The experimental results show that the proposed method significantly preserves diffuse reflectance and texture details in the specular highlight regions to obtain actual spectral reflectance and chromatic values from hyperspectral images of wool fibres.

Abstract A representative class of kagome materials, AV3Sb5 (A = K, Rb, Cs), hosts several unconventional phases such as superconductivity, $${{\mathbb{Z}}}_{2}$$ Z 2 non-trivial topological states, and electronic nematic states. These can often coexist with intertwined charge-density wave states. Recently, the discovery of the isostructural titanium-based single-crystals, ATi3Bi5 (A = K, Rb, Cs), which exhibit similar multiple exotic states but without the concomitant charge-density wave, has opened an opportunity to disentangle these complex states in kagome lattices. Here, we combine high-resolution angle-resolved photoemission spectroscopy and first-principles calculations to investigate the low-lying electronic structure of RbTi3Bi5. We demonstrate the coexistence of flat bands and several non-trivial states, including type-II Dirac nodal lines and $${{\mathbb{Z}}}_{2}$$ Z 2 non-trivial topological surface states. Our findings also provide evidence for rotational symmetry breaking in RbTi3Bi5, suggesting a directionality to the electronic structure and the possible emergence of pure electronic nematicity in this family of kagome compounds.

In this work, a combination of in-situ high synchrotron X-ray diffraction and electron backscattered diffraction were used to systematically investigate the activation and evolution of the deformation mechanisms in an as-cast Fe42Mn28Co10Cr15Si5 metastable high entropy alloy deformed until fracture at room temperature. This work unveils the critical role of the dual-phase γ-f.c.c. / ε-h.c.p. microstructure on the deformation response of the alloy. The different deformation modes, i.e., slip, transformation induced plasticity (TRIP) and transformation induced twinning (TWIP), were seen to initiate at different loading stresses and then to overlap. Quantitative microstructural characterization, which included the evolution of the phase fraction, stress partitioning, dislocation density, c/a ratio and lattice strain for different planes, was performed to elucidate the role of each phase on the macroscopic mechanical response of the metastable high entropy alloy. Furthermore, the magnitude of the different strengthening contributions has been quantified for the first time.

Neutrophil extracellular traps (NETs) are composed of chromatin filaments coated with granular and cytosolic proteins, which contribute to the pathogenesis and progression of immune-related diseases. NETs are frequently observed in gouty arthritis, but the related mechanisms remain poorly understood. The aim of our study was to systematically elucidate the molecular mechanisms of self-remitting effects in gouty arthritis, and the causative relationship between neutrophil autophagy and NETs. The air pouch and paw edema model were used to simulate gouty arthritis in mice. Neutrophil infiltration and the formation of NETs were found in gouty arthritis. Interestingly, monosodium urate (MSU) crystals could induce the formation of NETs, degrade inflammatory factors, and alleviate the inflammatory response in gouty arthritis. In addition, MSU crystals resulted in profound molecular alterations in neutrophils using RNA-seq analysis, including autophagy activation. MSU crystals could activate neutrophil autophagy in vitro, and autophagy activators and inhibitors could regulate the formation of NETs. Furthermore, we explored the mechanism of autophagy-induced NETs. Autophagy related protein 7 (ATG7) produced by neutrophils stimulated with MSU crystals worked synergistically with p53 to enter the nucleus, promoting peptidyl arginine deiminase 4 (PAD4) expression, and inducing the formation of NETs. Finally, we substantiated that neutrophil autophagy regulates the severity of gouty arthritis via the formation of NETs in PAD4 -/- mice. Our results indicated that the autophagy of neutrophils regulates the formation of NETs and degrades inflammatory factors. Regulating autophagy and interfering with the formation of NETs represents a potential therapeutic approach against gouty arthritis during clinical practice.

In this work, a 316L stainless steel to Inconel 625 functionally graded material (FGM) was built using different deposition strategies (named as direct and smooth-type interfaces) by Twin-Wire and Arc Additive Manufacturing (T-WAAM). This combination of materials is of interest in chemical plants, oil & gas, and nuclear applications, where high corrosion and wear resistance are essential requirements. Although these properties are superior in Inconel 625, replacing Inconel with stainless steel in strategic regions of structural components can reduce the overall costs and parts’ weight. Both direct and smooth transition interfaces were tested and characterized. Microscopic analysis revealed that each interface and the as-built samples had an austenitic matrix, and every sample was well bonded and free of defects. Different types of microstructures evolved at the interfaces due to distinct gradients in composition. Synchrotron X-ray diffraction measurements showed that the smooth-gradient produced secondary phases, such as δ-phase (Ni3Nb) and carbides, that were not present with the direct interface strategy. Overall, the properties were superior in the FGM with a direct interface, which experienced higher strengths and elongations upon failure. Moreover, neutron diffraction measurements revealed that lower residual stresses developed in the direct interface FGM than in the smooth gradient FGM.

Drought is one of the main abiotic stress factors affecting yield of Paeonia ostii. In this study, we conducted bioinformatics and differential expression analyses of P. ostii ‘Feng Dan’ ptc-miR396g-5p in leaf samples under different drought stress. ptc-miR396g-5p belongs to the miR396 family. Among the 271 plant species registered in the miRBase database, at least one miR396 member was found in 48 Angiospermae species, 3 in Gymnospermae species, and 1 in Pteridophy. Mature sequence alignment showed that P. ostii ‘Feng Dan’ ptc-miR396g-5p had high sequence similarity with miR396 from other species. Secondary structure prediction showed that the precursor sequence of ‘Feng Dan’ ptc-miR396g-5p could form a stable stem-loop structure, and the mature sequence was located on the 5′ arm of the secondary structure. Phylogenetic tree analysis showed that ‘Feng Dan’ was closely related to 20 species such as Glycine max, Medicago truncatula, Populus trichocarpa, Citrus sinensis, Vitis vinifera, and Theobroma cacao. The predicted target gene of the ‘Feng Dan’ ptc-miR396g-5p encodes a Signal Transducer and Activator of Transcription (STAT) transcription factor. The negative correlation of expression between the miRNA and its target gene was confirmed by qRT-PCR. Our data indicate that ‘Feng Dan’ ptc-miR396g-5p′s expression decreases under drought, leading to an expression increase of the STAT transcription factor.

The present work deals with the short-time thermal phase-stability of the as-cast eutectic AlCoCrFeNi2.1 high entropy alloy. Toward this end, the effect of different temperatures (800–1000 °C) and soaking times (15–60 min) on the stability of primary dendritic regions and formation of the ordered phases was explored. Microstructural characterization was supported by thermodynamic calculations and assessment of the subsequent mechanical properties. Upon the increase in annealing temperature and soaking time, the primary FCC dendritic areas grown and destabilized owing to elemental partitioning. This was followed by dendrite fragmentation and formation of needle shape B2 ordered phases within the primary FCC regions. Despite the unstable nature of the primary constituent phases, the material hardness increased considerably to a peak point corresponding to the moderate soaking time of 45 min. The variation of the subsequent mechanical properties was discussed relying on the characteristics of the ordered and primary phases.

Shape memory alloys (SMA) are a class of smart materials with inherent shape memory and superelastic characteristics. Unlike other SMAs, iron-based SMAs (Fe-SMA) offer cost-effectiveness, weldability, and robust mechanical strength for the construction industry. Thus, applying these promising materials to advanced manufacturing processes is of considerable industrial and academic relevance. This study aims to present a pioneer application of a Fe–Mn–Si–Cr–Ni–V-C SMA to arc-based directed energy deposition additive manufacturing, namely wire and arc additive manufacturing (WAAM), examining the microstructure evolution and mechanical/functional response. The WAAM-fabricated Fe-SMAs presented negligible porosity and high deposition efficiency. Microstructure characterization encompassing electron microscopy and high energy synchrotron X-ray diffraction revealed that the as-deposited material is primarily composed by γ FCC phase with modest amounts of VC, ε and σ phases. Tensile and cyclic testing highlighted the Fe-SMA's excellent mechanical and functional response. Tensile testing revealed a yield strength and fracture stress of 472 and 821 MPa, respectively, with a fracture strain of 26%. After uniaxial tensile loading to fracture, the γ → ε phase transformation was clearly evidenced with post-mortem synchrotron X-ray diffraction analysis. The cyclic stability during 100 load/unloading cycles was also evaluated, showcasing the potential applicability of the fabricated material for structural applications.

Obtaining a reliable NiTi to Ti6Al4V dissimilar joint exhibiting NiTi's superelasticity can provide design flexibility in aerospace and biomedical fields via integrating distinct material benefits. However, this materials couple is vulnerable to severe embrittlement due to the development of excessive intermetallic compounds (IMCs), namely Ti2Ni. Pd-free and Pd-interlayered NiTi-Ti6Al4V laser joints were evaluated for their microstructure, compositional changes, thermodynamic mechanism, and mechanical properties. The presence of Pd constrained the formation of Ti2Ni IMC, and NiTi-Ti6Al4V joints with excellent mechanical properties demonstrating superelastic behavior were achieved for the first time. The tensile strength and rupture strain of the Pd-added NiTi-Ti6Al4V joint improved more than twofold, reaching 520 MPa and 5.6%, respectively. During cyclic tensile testing, the Pd-added joint demonstrated superelasticity and a comparable irrecoverable strain to NiTi (1 versus 0.65%). Multiscale characterization revealed that the fraction of Ti2Ni decreased from 83 to 10% near the NiTi boundary and 24 to 6% at the weld center compared to the Pd-free joint. The superior thermodynamic formation tendency of Ti-Pd compounds over Ti2Ni IMC favored their development, and thus Ti-Pd and NiTi compounds dominated the fusion zone (FZ) at the expense of Ti2Ni IMC, explaining the improved mechanical performance of the Pd-added joint.

Ferroptosis resistance is vital for B cell development, especially in inflammatory diseases, yet the underlying mechanism is still unclear. In this study, based on the scRNA-seq technique and flow cytometry, we discovered a proportion of neutrophils exhibited upregulated expression of the IL-6 and correlated with the expression of IL-6 receptor and SLC7A11 from B cells in lupus kidney. Moreover, we identified that in lupus kidney, neutrophils could provide IL-6 to facilitate ferroptosis resistance in B cells via SLC7A11, and inhibition of SLC7A11 could significantly enhance ferroptosis in B cells and could decrease B cell proliferation. This study helps understand the crosstalk between neutrophils and B cells in the kidney in the development of lupus.

Liver hepatocellular carcinoma (LIHC) is a highly lethal malignant tumor originating from the digestive system, which is a serious threat to human health. In recent years, immunotherapy has shown significant therapeutic effects in the treatment of LIHC, but only for a minority of patients. The basement membrane (BM) plays an important role in the occurrence and development of tumors, including LIHC. Therefore, this study is aimed at establishing a risk score model based on basement membrane-related genes (BMRGs) to predict patient prognosis and response to immunotherapy. First, we defined three patterns of BMRG modification (C1, C2, and C3) by consensus clustering of BMRG sets and LIHC transcriptome data obtained from public databases. Survival analysis showed that patients in the C2 group had a better prognosis, and Gene Set Variation Analysis (GSVA) revealed that the statistically significant pathways were mainly enriched in the C2 group. Moreover, we performed Weighted Correlation Network Analysis (WGCNA) on the above three subgroups and obtained 179 intersecting genes. We further applied function enrichment analyses, and the results demonstrated that they were mainly enriched in metabolism-related pathways. Furthermore, we conducted the LASSO regression analysis and obtained 4 BMRGs (MPV17, GNB1, DHX34, and MAFG) that were significantly related to the prognosis of LIHC patients. We further constructed a prognostic risk score model based on the above genes, which was verified to have good predictive performance for LIHC prognosis. In addition, we analyzed the correlation between the risk score and the tumor immune microenvironment (TIM), and the results showed that the high-risk scoring group tended to be in an immunosuppressed status. Finally, we investigated the relationship between the risk score and LIHC immune function. The results demonstrated that the risk score was closely related to the expression levels of multiple immune checkpoints. Patients in the low-risk group had significantly higher IPS scores, and patients in the high-risk group had lower immune escape and TIDE score. In conclusion, we established a novel risk model based on BMRGs, which may serve as a biomarker for prognosis and immunotherapy in LIHC.

The altered expression of ARMCX1 in patients with gastric cancer has been reported frequently, yet its correlation to prognosis and chemotherapy needs to be unveiled. In combination of the gene expression data retrieved from TCGA database and bioinformatic analysis, this study discovered 590 differentially expressed genes in the cancerous biopsies isolated from gastric patients, compared with controls. Among which, ARMCX1 exhibited great potential to serve as a prognostic biomarker for gastric patients; furthermore, patients with low expression of ARMCX1 could be more sensitive to these 9 chemotherapeutic agents: A-770041, AMG-706, ATRA, BEZ235, bortezomib, CGP60474, dasatinib, HG-64-1, and pazopanib, rather than the other chemotherapeutic agents. This study helps the improvement of evaluating the prognosis of gastric cancer patients, and would help optimize chemotherapeutic strategies in consideration of the expression of ARMCX1.

Purpose: Microglia-neuron crosstalk is critically involved in synaptic plasticity and degeneration by releasing diverse mediators in Alzheimer’s disease (AD). Therefore, determining contributors that modulate the systemic microenvironment is essential. Cordycepin (CCS) is a novel neuroprotective compound obtained from Cordyceps militaris. However, the anti-AD efficacy and potential mechanism of CCS treatment remain unclear. This study aimed to elucidate the microglia-neuron symphony in AD after CCS treatment and to explore the possible mechanisms of its neuroprotective efficacy. Methods and results: CCS treatment improved learning and memory impairment in 9-month-old APP/PS1 mice by behavioral tests. CCS polarized the microglia from M1 to M2, inhibited neuronal apoptosis and promoted synaptic remodeling accompanied by in vivo and in vitro upregulation of NGF. The cAMP-response element-binding protein (CREB) was also activated after MG-M2 polarization. Further, we verified that the sg3 promoter region of NGF (–1018 to –1011) is the key binding site for CREB-induced NGF transcription, which increased NGF expression and secretion. Finally, microglia-derived NGF was confirmed as an important mediator in microglia-neuron symphony to improve the neuronal microenvironment after CCS treatment. Conclusions: CCS improved the neuronal synaptic plasticity and senescence by promoting MG-M2 activation driven by CREB-induced NGF upregulation and facilitated symphony communication between the microglia and neuron in AD. This study provides a new perspective on the development of a novel strategy for anti-AD therapy and offers new targets for anti-AD drug development.

The flowering period is the most important ornamental trait of tree peony, while industrial development of tree peony has been limited by short flowering period. miR319 plays an important regulatory role in plant flowering. In the current study, the expression characteristics and evolution of PsmiR319 in tree peony flowering was explored using ‘Feng Dan’ and ‘Lian He’, which are early-flowering and late-flowering varieties of tree peony, respectively. The structure, evolution, and target(s) of PsmiR319 were analyzed by bioinformatics. Evolution analysis showed that pre-PsmiR319 was distributed in 41 plant species, among which the length of the precursor sequence exhibited marked differences (between 52 and 308 bp). Pre-PsmiR319 of tree peony was located close to the corresponding sequences of Linum usitatissimum and Picea abies in the phylogenetic tree, and in addition, could form a typical hairpin structure including a mature body with a length of 20 bp located on the 3p arm and part of the loop sequence. The mature sequence of miR319 was highly conserved among different species. Target genes of PsmiR319 include MYB-related transcription factor in tree peony. Expression of PsmiR319, assayed by qRT-PCR, differed between ‘Feng Dan’ and ‘Lian He’ during different flower development periods. PsmiR319 and its target gene showed a negative expression regulation relationship during the periods of CE (color exposure), BS (blooming stage), IF (initial flowering), and HO (half opening) in the early-flowering ‘Feng Dan’, and the same in FB (Full blooming) periods of late-flowering ‘Lian He’. Findings from this study provide a reference for further investigation into the mechanism of miR319 in the development of different varieties of tree peony.

Post-stroke depression (PSD) is a common cerebrovascular complication characterized by complex pathogenesis and poor treatment effects. Here, we tested the influence of differentially expressed genes (DEGs), non-targeted metabolites, and intestinal microbes on the occurrence and development of PSD. We acquired gene expression profiles for stroke patients, depression patients, and healthy controls from the Gene Expression Omnibus database. After screening for DEGs using differential expression analysis, we identified common DEGs in stroke and depression patients that were considered to form the molecular basis of PSD. Functional enrichment analysis of DEGs also revealed that the majority of biological functions were closely related to metabolism, immunity, the nervous system, and microorganisms, and we also collected blood and stool samples from healthy controls, stroke patients, and PSD patients and performed 16S rDNA sequencing and untargeted metabolomics. After evaluating the quality of the sequencing data, we compared the diversity of the metabolites and intestinal flora within and between groups. Metabolic pathway enrichment analysis was used to identify metabolic pathways that were significantly involved in stroke and PSD, and a global metabolic network was constructed to explore the pathogenesis of PSD. Additionally, we constructed a global regulatory network based on 16S rDNA sequencing, non-targeted metabolomics, and transcriptomics to explore the pathogenesis of PSD through correlation analysis. Our results suggest that intestinal flora associates the dysregulation of cerebral cortex gene expression and could potentially promote the occurrence of depression by affecting the metabolism of stroke patients. Our findings may be helpful in identifying new targets for the prevention and treatment of PSD.

Head and neck squamous cell carcinoma (HNSCC) usually has a poor prognosis and is associated with a high mortality rate. Its etiology is mainly the result from long-term exposure to either alcohol, tobacco or human papillomavirus (HPV) infection or a combination of these insults. However, HNSCC patients with HPV have been found to show a survival advantage over those without the virus, but the mechanism that confers this advantage is unclear. Due to the large number of HPV-independent HNSCC cases, there is a possibility that the difference in prognosis between HPV-positive (HPV+) and negative (HPV-) patients is due to different carcinogens. To clarify this, we used scRNA data and viral tracking methods in order to identify HPV+ and HPV- cells in the tumour tissues of patients infected with HPV. By comparing HPV+ and HPV- malignant cells, we found a higher level of tumour stemness in HPV- tumour cells. Using tumour stemness-related genes, we established a six-gene prognostic signature that was used to divide the patients into low- and high-risk groups. It was found that HPV patients who were at low-risk of contracting HNSCC had a higher number of CD8+ T-cells as well as a higher expression of immune checkpoint molecules. Correspondingly, we found that HPV+ tumour cells expressed higher levels of CCL4, and these were highly correlated with CD8+ T cells infiltration and immune checkpoint molecules. These data suggest that the stemness features of tumour cells are not only associated with the prognostic risk, but that it could also affect the immune cell interactions and associated signalling pathways.

The AlCoCrFeNi2.1 eutectic high entropy alloy is of great interest due to its unique mechanical properties combining both high strength and plasticity. Here, gas tungsten arc welding was performed for the first time on an as-cast AlCoCrFeNi2.1 alloy. The microstructural evolution of the welded joints was assessed by combining electron microscopy with electron backscatter diffraction, synchrotron X-ray diffraction analysis and thermodynamic calculations. Microhardness mapping and tensile testing coupled with digital image correlation were used to investigate the strength distribution across the joint. The base material, heat affected zone and fusion zone are composed of an FCC + B2 BCC eutectic structure, although the relative volume fractions vary across the joint owing to the weld thermal cycle. The BCC nanoprecipitates that existed in the base material started to dissolve into the matrix in the heat affected zone and closer to the fusion zone boundary. Compared to the as-cast base material, the fusion zone evidenced grain refinement owing to the higher cooling rate experienced during solidification. This translates into an increased hardness in this region. The joints exhibit good strength/ductility balance with failure occurring in the base material. This work establishes the potential for using arc-based welding for joining eutectic high entropy alloys.

miRNA plays an important role in plant growth and development and in response to various stresses. Quantitative real-time PCR (qRT-PCR) technology is often used to detect the expression level of miRNAs and genes by comparing with reference genes. In order to screen out the optimal reference miRNAs in different tree peony varieties, the petals of 42 different early- and late-flowering tree peony varieties were used as experimental materials, and geNorm, NormFinder, Bestkeeper, and RefFinder software were used to evaluate the stability of 16 candidate reference miRNAs. The results showed that the average Ct values of all candidate reference miRNAs were between 15.34 ± 0.29 and 32.64 ± 0.38. The optimal number of reference miRNAs was four, which were PsPC-5p-19095, PsPC-3p-51259, PsmiR159a, and PsPC-3p-6660 in geNorm. The stability of PsPC-3p-6660 was the highest in the analysis results of NormFinder software. Among the analysis results of Bestkeeper software, PsMIR319-p5 has the highest stability. Among the results of comprehensive evaluation and analysis of several software using RefFinder, the candidate reference miRNA with the highest stability was PsPC-3p-6660. When PsPC-3p-6660 was used as the reference miRNA, the expression of PomiR171 and PomiR414 in response to different flowering times of tree peony was relatively stable in 42 tree peony varieties, indicating that PsPC-3p-6660 was stable and reliable. The results of this study provide a reference miRNA for studying the expression changes of miRNA in different tree peony varieties and further exploring the regulatory mechanism of miRNA in different peony varieties.

This paper investigates the wettability of Kovar alloys with high-borosilicate glass and microscopically analyses the mechanism of wettability and diffusion between Kovar and borosilicate glass. First, Kovar was oxidised at 800 °C for 5, 15, 25, 35, and 60 min to observe the oxide morphology of the Kovar surface layer and to analyse the composition of the oxide layer. To investigate the wetting pattern formations of Kovar and high-borosilicate glass under different wetting temperatures, times, and preoxidation conditions, Kovar and high-borosilicate glass obtained from different oxidation treatments were held at 1060 °C for 20 min for wetting experiments, and the glass–metal wetting interface morphology and elemental distribution were observed using SEM and EDS. The elemental diffusion at the wetting interface between the borosilicate glass and the Kovar with different preoxidation and at the glass spreading boundary was investigated. The longitudinal diffusion of the liquid glass in the metal oxide layer formed a new tight chemical bond of Fe2SiO4, and the lateral diffusion of the liquid glass in the Kovar surface layer formed a black halo.

Dissimilar joining involving high entropy alloys is currently being explored to evaluate the suitability of these novel advanced engineering materials in structural applications. Recently, joining of a CoCrFeMnNi high entropy alloy to 316 stainless steel was successfully attempted. However, the joint ductility was limited by the lack of deformation experienced by the cold-rolled CoCrFeMnNi base material during tensile loading. In this work, it is shown that by simply changing the base material condition, from cold-rolled to annealed, it is possible to significantly improve the joint fracture strain from ≈ 5 to ≈ 10 %, while preserving the strength at ≈ 450 MPa. Using electron microscopy, high energy synchrotron X-ray diffraction and mechanical testing aided by digital image correlation, the microstructure evolution across the welded joint is assessed and correlated to its mechanical performance. Moreover, thermodynamic calculations considering the compositional changes across the fusion zone were used to predict the microstructure evolution of this region.

Clear cell renal cell carcinoma (ccRCC) usually affects multiple organs (e.g., bone and brain), and patient prognosis is usually poor. Although it is known that CD8+ T cell infiltration can potentially alleviate ccRCC progression, few studies have concentrated on the correlation between CD8+ T cell infiltration and ccRCC prognosis. In this study, ten genes expressed by infiltrated CD8+ T cells (i.e., AMD1, CCSER2, CIB1, DRAP1, HMGB2, HMGN1, NPIPB5, PTP4A2, RORA, and SAP18) were suggested as potential ccRCC prognostic biomarkers, by using next-generation sequencing (i.e. bulk sequencing and single-cell sequencing) of ccRCC, papillary renal cell carcinoma (papRCC), and control kidney biopsies. Specifically, we identified four genes (i.e., CCSER2, DRAP1, NPIPB5, and SAP18) as potential novel prognostic biomarkers for ccRCC. It is noteworthy that SAP18 derived from CD8+ T cells negatively correlates to Atg7+ neutrophils in ccRCC, compared with papRCC, indicating a potential decreased neutrophil metabolic function in autophagy and fatty acids. This study elucidated the protective role of infiltrated CD8+ T cells in ccRCC and identified ten candidate genes related to an improved prognosis in patients with ccRCC.

High-risk human papillomavirus (hrHPV) infection has been associated with a higher probability of progression to cervical cancer. However, several extensive studies have reported that the presence of hrHPV can lead to a better prognosis, but the mechanism of how this occurs is unclear. In this study, microbiological analysis was used to identify HPV infection as a factor for the prognosis of patients with cervical squamous cell carcinoma (CSCC). Comparing the interactions of HPV+ and HPV- malignant cells with immune cells as well as the trajectory of malignant cells either with or without HPV, we found that most of the HPV+ cells are well differentiated while HPV- cells appear to be hypo-fractionated. Using transcriptomic and immunostaining data, we validated a set of unfavourable molecules in the HPV- CSCC cells, including KRT16, ITGB1, CXCR1, VEGFA, CRCT1 and TNFRSF10B/DR5. This study provides a basis for the development of a rational post-operative follow-up programme and the development of an appropriate treatment plan for patients with cervical cancer.

The Invar alloy is widely used for aircraft wing mould manufacturing. In this work, keyhole-tungsten inert gas (K-TIG) butt welding was used to join 10 mm thick Invar 36 alloy plates. The effect of heat input on the microstructure, morphology and mechanical properties was studied by using scanning electron microscopy, high energy synchrotron X-ray diffraction, microhardness mapping, tensile and impact testing. It was shown that regardless of the selected heat input, the material was solely composed of austenite, although the grain size changed significantly. The change in heat input also led to texture changes in the fusion zone, as qualitatively determined with synchrotron radiation. With increases in heat input, the impact properties of the welded joints decreased. The coefficient of thermal expansion of the joints was measured, which demonstrated that the current process is suitable for aerospace applications.

ABSTRACT: Dietary fiber (DF) improves gastrointestinal health and has important associations with the alleviation of intestinal diseases and metabolic syndrome. However, due to DFs complex characteristics, such as solubility, viscosity, and fermentability, the mechanism in these was not consistent. As an herbivore, the goose has a prominent digestive ability to DF.Therefore, we choose low, medium, and high viscosity DFs (respectively resistant starch-3 []RS], inulin [INU], and β–glucan [GLU]) as Magang goose diet treatment for 4 wk, to investigate the effect and potential mechanism of different viscosities DFs on the growth and development process of goose.In summary, three degrees of viscous DFs could decrease the mechanismic lipid level of geese by promoting acid-producing bacteria and short-chain fatty acid (SCFA) production, therefore, activating AMPK pathway-related genes through the gut-liver axis. High viscous DF has a greater lipid-lowering effect on geese, while medium viscous DF has preferable intestinal mucosal protection.

Abstract Background Lysine succinylation is an emerging posttranslational modification that has garnered increased attention recently, but its role in gastric cancer (GC) remains underexplored. Methods Proteomic quantification of lysine succinylation was performed in human GC tissues and adjacent normal tissues by mass spectrometry. The mRNA and protein levels of lactate dehydrogenase A (LDHA) in GC and adjacent normal tissues were analyzed by qRT-PCR and western blot, respectively. The expression of K222-succinylated LDHA was measured in GC tissue microarray by the K222 succinylation-specific antibody. The interaction between LDHA and sequestosome 1 (SQSTM1) was measured by co-immunoprecipitation (co-IP) and proximity ligation assay (PLA). The binding of carnitine palmitoyltransferase 1A (CPT1A) to LDHA was determined by co-IP. The effect of K222-succinylated LDHA on tumor growth and metastasis was evaluated by in vitro and in vivo experiments. Results Altogether, 503 lysine succinylation sites in 303 proteins were identified. Lactate dehydrogenase A (LDHA), the key enzyme in Warburg effect, was found highly succinylated at K222 in GC. Intriguingly, this modification did not affect LDHA ubiquitination, but reduced the binding of ubiquitinated LDHA to SQSTM1, thereby decreasing its lysosomal degradation. We demonstrated that CPT1A functions as a lysine succinyltransferase that interacts with and succinylates LDHA. Moreover, high K222-succinylation of LDHA was associated with poor prognosis in patients with GC. Finally, overexpression of a succinylation-mimic mutant of LDHA promoted cell proliferation, invasion, and migration. Conclusions Our data revealed a novel lysosomal pathway of LDHA degradation, which is mediated by the binding of K63-ubiquitinated LDHA to SQSTM1. Strikingly, CPT1A succinylates LDHA on K222, which thereby reduces the binding and inhibits the degradation of LDHA, as well as promotes GC invasion and proliferation. This study thus uncovers a new role of lysine succinylation and the mechanism underlying LDHA upregulation in GC.

Background: Ginsenoside Rb1 (Rb1), one of the most abundant protopanaxadiol-type ginsenosides, exerts excellent neuroprotective effects even though it has low intracephalic exposure. Purpose: The present study aimed to elucidate the apparent contradiction between the pharmacokinetics and pharmacodynamics of Rb1 by studying the mechanisms underlying neuroprotective effects of Rb1 based on regulation of microflora. Methods: A pseudo germ-free (PGF) rat model was established, and neuroprotective effects of Rb1 were compared between conventional and PGF rats. The relative abundances of common probiotics were quantified to reveal the authentic probiotics that dominate in the neuroprotection of Rb1. The expressions of the gamma-aminobutyric acid (GABA) receptors, including GABAA receptors (α2, β2, and γ2) and GABAB receptors (1b and 2), in the normal, ischemia/reperfusion (I/R), and I/R+Rb1 rat hippocampus and striatum were assessed to reveal the neuroprotective mechanism of Rb1. Results: The results showed that microbiota plays a key role in neuroprotection of Rb1. The relative abundance of Lactobacillus helveticus (Lac.H) increased 15.26 fold after pretreatment with Rb1. I/R surgery induced effects on infarct size, neurological deficit score, and proinflammatory cytokines (IL-1β, IL-6, and TNF-α) were prevented by colonizing the rat gastrointestinal tract with Lac.H (1 × 109 CFU) by gavage 15 d before I/R surgery. Both Rb1 and Lac.H upregulated expression of GABA receptors in I/R rats. Coadministration of a GABAA receptor antagonist significantly attenuated neuroprotective effects of Rb1 and Lac.H. Conclusion: In sum, Rb1 exerts neuroprotective effects by regulating Lac.H and GABA receptors rather than through direct distribution to the target sites. In this work, a functionally graded material (FGM) part was fabricated by depositing a Cu-based alloy on top of a high strength low alloy (HSLA) steel by twin-wire and arc additive manufacturing (T-WAAM). Copper and steel parts are of interest in many industries since they can combine high thermal/electrical conductivity, wear resistance with excellent mechanical properties. However, mixing copper with steel is difficult due to mismatches in the coefficient of thermal expansion, in the melting temperature, and crystal structure. Moreover, the existence of a miscibility gap during solidification, when the melt is undercooled, causes serious phase separation and segregation during solidification which greatly affects the mechanical properties. Copper and steel control samples and the functionally graded material specimen were fabricated and investigated using optical microscopy, scanning electron microscopy, and high energy synchrotron X-ray diffraction. Retained δ-ferrite was found in a Cu matrix at the interface region due to regions with mixed composition. A smooth gradient of hardness and electric conductivity along the FGM sample height was obtained. An ultimate tensile strength of 690 MPa and an elongation at fracture of 16.6% were measured in the FGM part.

Inhibition of triple-negative breast cancer metastasis has long been a challenge, mainly due to the difficulty in identifying factors that contribute to this process. In this study, freshly isolated triple-negative breast cancer biopsied cells obtained from consenting patients were subjected to flow cytometry and bioinformatic analysis to identify three endothelial cell subclusters: EC (ATP1B3), EC (HSPA1B), and EC (KRT7) in the tumor microenvironment. These endothelial cell subclusters exhibited distinguishing biological features. Based on differentially expressed genes derived from the subclusters, gene set enrichment analysis showed that EC (ATP1B3) and EC (HSPA1B) contribute to the process of metastasis, for example, in fibrosarcoma and anaplastic carcinoma. In this study, we identified the heterogeneity of endothelial cells in the human breast cancer and have provided insights into its role in metastasis.

For those patients with hepatocellular carcinoma (HCC), it is really a heavy burden. Herein, the immune genes of HCC were analyzed in groups to determine prognostic biomarkers related to immune genes in HCC. The mRNA data, clinical data in TCGA-LIHC dataset, and immune gene in the ImmPort database were collected for the combining usage with K-means concordance clustering to cluster HCC patients according to the immune gene matrix. Based on ssGSEA analysis result, HCC patients were sorted into high- and low-immune subtypes, and survival curve presented that patients in high-immune subtypes had a better prognosis. Subsequently, differential expression analysis was performed to obtain immune-related differentially expressed genes (IRGs). Cox and lasso analyses were performed for obtaining five optimal immune genes related to prognosis, and a risk assessment model was then established. Patient samples in the training and validation sets were, respectively, divided into high- and low-risk groups. K-M survival curves presented a better prognosis of patients in the low-risk group than in the high-risk group. The ROC curve indicated that this model was finely used for the prediction of prognosis. In addition, immune infiltration assessment revealed that NR0B1 and FGF9 had potential to impact the tumor immune microenvironment. Finally, using qRT-PCR and transwell assays, it was demonstrated that the macrophage chemotaxis was enhanced when NR0B1 and FGF9 were highly expressed in HCC cells. In general, a 5-gene prognostic risk assessment model was constructed based on immune genes and bioinformatics analysis methods, which provides some reference for the prognosis of HCC as well as immunotherapy.

Cardiac rehabilitation, which combines cardiology and preventive medicine, is an important part of treatment for cardiovascular diseases. Systematically, cardiac rehabilitation, including simultaneously inhibiting endothelial injury and promoting endothelial repair, is beneficial for physical and mental recovery and reduces the risks of recurrence and death in patients with cardiovascular diseases. Cardiac rehabilitation has developed rapidly in the last 50 years. A preliminary system for cardiac rehabilitation has been developed in China. The present article mainly focuses on the progress of cardiac rehabilitation from the aspects of goals, measures, and modes of research in the current scenario.

Liquid-phase exfoliated graphene sheets are promising candidates for printing electronics. Here, a high-performance printed 2.4 GHz graphene-based antenna is reported. Graphene conductive ink prepared by using liquid-phase exfoliation process is printed onto a water-transferable paper by using blade printing technique, which is then patterned as dipole antenna and transferred onto a target substrate. The fabricated dipole antenna (43 × 3 mm), exhibiting typical radiation patterns of an ideal dipole antenna, achieves −10 dB bandwidth of 8.9% and a maximum gain of 0.7 dBi. The printed graphene-antennas satisfy the application requirements of the Internet of Things and suggest its feasibility of replacing conventional metallic antennas in those applications.

High temperature shape memory alloys of the Ni-Ti-Hf system are potential candidates for aerospace applications where powder bed additive manufacturing technologies are being increasingly used. In this work, a Ti-rich NiTiHf high temperature shape memory alloy powder was processed by laser powder bed fusion. The standard heat treatment of 550 ⁰C for 3 hours was imposed to promote H-phase precipitation. At room temperature, the material has a dual-phase microstructure composed of martensite, the matrix, and H-phase, as a strengthening precipitate. High energy synchrotron X-ray diffraction is used to evaluate, in-situ, the elastic behaviour of the fabricated part. The deformation anisotropy of several (h k l) families of planes of both phases is evidenced. No major texture changes were observed upon macroscopic elastic loading. We illustrate the potential of using high energy synchrotron X-ray diffraction for detailed analyses of minority phases in additively manufactured components.

BackgroundCD8+ T cells, vital effectors pertaining to adaptive immunity, display close relationships to the immunization responses to kill tumor cells. Understanding the effect exerted by tumor infiltration CD8+ T cells in papillary renal cell carcinoma (papRCC) is critical for assessing the prognosis process and responses to immunization therapy in cases with this disease.Materials and ApproachesThe single-cell transcriptome data of papRCC were used for screening CD8+ T-cell-correlated differentially expressed genes to achieve the following investigations. On that basis, a prognosis gene signature associated with tumor infiltration CD8+ T cell was built and verified with The Cancer Genome Atlas data set. Risk scores were determined for papRCC cases and categorized as high- or low-risk groups. The prognosis significance for risk scores was assessed with multiple-variate Cox investigation and Kaplan–Meier survival curves. In addition, the possible capability exhibited by the genetic profiles of cases to assess the response to immunization therapy was further explored.ResultsSix hundred twenty-one cell death-inhibiting RNA genes were screened using single-cell RNA sequencing. A gene signature consisting of seven genes (LYAR, YBX1, PNRC1, TCF25, MYL12B, MINOS1, and LINC01420) was then identified, and this collective was considered to be an independent prognosis indicator that could strongly assess overall survival in papRCC. In addition, the data allowed papRCC cases to fall to cohorts at high and low risks, exhibiting a wide range of clinically related features as well as different CD8+ T-cell immunization infiltration and immunization therapy responses.ConclusionsOur work provides a possible explanation for the limited response of current immunization checkpoint-inhibiting elements for combating papRCC. Furthermore, the researchers built a novel genetic signature that was able to assess the prognosis and immunotherapeutic response of cases. This may also be considered as a promising therapeutic target for the disease.

BackgroundCD8+ T cells, which play a vital role in response to adaptive immunity, are closely related to the immunization responses to kill tumor cells. Understanding the effects exerted by tumor-infiltrated CD8+ T cells in HPV+ and HPV- head and neck squamous cell carcinoma (HNSCC) patients is critical for predicting their prognosis as well as their responses towards immunization-related therapy.Materials and MethodsHNSCC single cell transcriptome was used to screen for differentially expressed genes (DEGs) based on CD8+ T cells. A gene signature associated with CD8+ T cells was built and verified with the cancer genome atlas dataset with a view to predicting the prognosis of HNSCC patients. Risk scores were calculated for HNSCC cases and categorized into either high- or low-risk cohorts. The prognosis-correlated data of the risk scores were analyzed by using Kaplan-Meier survival curves and multi-variate Cox regression plots. In addition, the possibility of using the genetic profiles to predict responses toward immunization-related therapy was explored.ResultsFrom the DEGs screened from the sequencing of single-cell RNA, a gene signature of 4 genes (ACAP1, ANKRD28, C12orf75, and M6PR) were identified. It was seen that these genes could predict overall survival in HPV+ HNSCC patients. In addition, high- and low-risk HPV+ HNSCC patients showed marked differences in their CD8+ T-cell infiltration due to immunization when clinical characteristics were taken into consideration. This correlated with their immunization therapy responses.ConclusionsOur work provides insights into explaining the restricted responses of current immunization checkpoint inhibiting substances in HPV+ HNSCC patients. A novel genetic signature to predict the prognosis and immunization-correlated therapeutic responses is presented. This will provide potential new therapeutic opportunities for HPV+ HNSCC patients.

Leaf size is a crucial trait in eggplant breeding, as it influences photosynthesis, plant biomass and management. However, little is known about the molecular mechanism regulating leaf size in eggplant. This study reports a small leaf mutant (slf) generated with the mutagen ethyl methane sulfonate (EMS). The slf mutant showed restricted cell proliferation and an increased content of auxin. Transcriptome analysis revealed that several genes involved in auxin signaling are upregulated in slf. Exogenous application of auxinole, an auxin antagonist of TIR1/AFB receptors, repressed the expression of these genes and restored leaf growth of slf, suggesting that the small leaf size of slf is likely associated with auxin signaling. This study provides essential clues to unveil the molecular mechanism of leaf size regulation in eggplant.

COVID-19 pneumonia requires effective medical therapies. However, it is a challenge to find therapeutic drugs that not only inhibit viral replication, but also inhibit the accompanying cytokine storm and maintain an appropriate immune response. In this study, the effects of SARS-CoV-2 on gene expression in lung epithelial cells from patients with COVID-19 were systematically evaluated with bioinformatics analysis methods. Transcriptome expression specific to bystander (exposed but uninfected) and infected cells were found, and the vital pathways were identified by conducting differentially expressed gene analysis regarding the relationship between gene signatures of COVID-19 infection and disease severity. We found that a high viral load did not necessarily imply a low response of epithelial cells or a poor disease convalescence. The ability to distinguish the role of virus-correlated genes facilitates the development of potential new medicines and therapies for COVID-19 infection.

Laser welding of a Ni-rich NiTi-20Zr (at.%) high temperature shape memory alloy was performed. The starting base material was aged for 3 h at 550 °C followed by air cooling prior to welding to induce H-phase precipitation. Advanced microstructure characterization encompassing scanning and transmission electron microscopy, coupled with synchrotron X-ray diffraction, were used. Defect-free welds were obtained with a conduction welding mode. The weld thermal cycle altered the microstructure across the heat affected and fusion zones of the joints. The heat affected zone exhibited partial H-phase dissolution, causing a decrease in hardness. In the fusion zone, the H-phase fully dissolved, and the non-equilibrium rapid solidification conditions prevented the H-phase from re-precipitating during cooling, leading to a microstructure resembling that of an as-cast alloy with the same material composition. Mechanical testing revealed that the laser welded samples sustained stresses in the order of 500 MPa and exhibited stress-strain responses comparable to those of the unwelded base material. Thus, this initial study shows new possibilities for using advanced laser joining methods in these alloys.

Based on the background of the reconstruction project from Changqing Chenzhuang-Pingyin section of G220 east-deep line in China, a special tunnel structure and construction plan was carried out according to the construction measures of the shallow-buried small spacing tunnel passing underneath cultural relic buildings, and a comprehensive deformation control scheme of “CRD construction method single-arm excavation + surface grouting prereinforcement + advanced large pipe shed presupport” was put forward. The results of numerical simulation and on-site construction monitoring showed that the overall deformation of aqueduct foundation generally increases first, then decreases and increases again, and finally tends to be stable. The effects of surface grouting prereinforcement and advance large pipe shed presupport are obvious. The comprehensive deformation control scheme can ensure the safety of the existing construction and meet the safety prevention and control requirements.

A group of peptide metabolites (1–4), designated as mintaimycins, were isolated from Micromonospora sp. C-3509. The planar structures of mintaimycins were determined by combination of mass spectrometry, 1D and 2D NMR spectroscopy, and the stereochemistry of mintaimycins were partially resolved by Marfey’s or Mosher’s method. Mintaimycins featured a central β-methylphenylalanine or phenylalanine linked at its amino group with 5-methyl-2-hexenoic acid, and at its carboxyl group with 5-hydroxy-norleucine or leucine that combined a derivative of hexanoic acid or 4-methylpentanoic acid. Mintaimycin A1 (1), the principal component, was found to exhibit the biological activity of inducing pre-adipocyte differentiation of 3T3-L1 fibroblast cells at 10.0 μmol/L.

In order to explore the sound absorption and noise reduction performance of closed-cell aluminum foam in the tunnel, the field test of the sound absorption performance of aluminum foam board was carried out based on the installation of aluminum foam board in the whole line of Haoshanyu Tunnel on Qinglan Expressway. Combined with the existing loudspeaker test and typical tunnel measurements, a new field test method for the noise reduction performance of closed-cell aluminum foam board was proposed for two different working conditions including fixed-point pure tone sound source condition and mobile vehicle sound source condition. The testing results of the new methods were analyzed, and it showed that the closed-cell aluminum foam has good sound absorption property at the frequency spectra between 250 Hz and 1000 Hz, and the farther away from the sound source, the better the sound absorption effect. In the research on the noise reduction effect of actual vehicle, it was found that the insertion loss of the closed-cell foam aluminum board is about 4 dB(A), which indicated that the closed-cell aluminum foam can play a certain noise reduction effect in the tunnel.

Dwarfism is a desirable trait in eggplant breeding, as it confers higher lodging resistance and allows simplified management and harvest. However, a few dwarf mutants have been reported, and the molecular mechanism underlying dwarfism in eggplant is completely unknown. Here, we report a dwarf mutant (dwf) isolated from an ethyl methyl sulfonate (EMS)-induced mutant library. The hypocotyl length, plant height, and length of internode cells of dwf were significantly decreased compared to those of the wild-type parent ‘14-345’ (WT). Differential gene expression analysis revealed that GA-related genes, including GA2ox and DELLA, were up-regulated whereas the gibberellin (GA3) content decreased in dwf. Moreover, exogenous GA3 treatment significantly increased the relative growth rate of dwf compared to WT, further indicating the important roles of GA in regulating the dwarf phenotype of dwf. Collectively, our findings shed light on GA-mediated dwarfism in dwf plants and offer a good germplasm that could be used for eggplant dwarfism breeding in the future.

The thermal regime and microstructural phenomenon are studied by using finite-element (FE) modelling and the analytical Rosenthal equation during laser welding of aluminum alloy 5456 (AA5456) components. A major goal is to determine the merits and demerits of this analytical equation which can be an alternative to FE analysis, and to evaluate the effect of imperative assumptions on predicted consequences. Using results from the analytical and numerical approaches in conjunction with experiments, different physical features are compared. In this study, the results obtained from experiments in terms of melt pool shapes are compared with the predicted ones achieved from the numerical and analytical approaches in which the FE model is more accurate than the Rosenthal equation in the estimation of the melt pool dimensions. Furthermore, as to the partially melted zones, the estimations achieved from the numerical modeling are more genuine than ones from the analytical equation with regards to the experimental results. At high energy density, near keyhole welding mode, the reported results show that experimental melt widths are supposed to be narrower than the fusion widths estimated by the analytical solution. The primary explanation could be the influence of thermal losses that occurred during convection and radiation, which are neglected in the Rosenthal equation. Additionally, the primary dendrite arm spacing (PDAS) estimated with the numerical modeling and the analytical Rosenthal solution is comparable with the experimental results obtained.
