Abstract Biochar (BC) applications in soil has positive effects on plant performance, particularly for loose soil in agricultural context. However, how biochar types affect plant performance of non-crop species and soil–plant carbon relationships is not clear. We selected five different BC types and three plant species to investigate the responses of plant performance and the soil–plant carbon relationship to BC effects. The result demonstrated that peanut shell BC led to the death of both R. tomentosa and C. edithiae, due to a reduction in nutrient uptake caused by higher soil electricity conductivity (2001.7 and 976.3 µS cm−1). However, the carbon content of S. arboricola increased by 57% in peanut shell BC-amended soil, suggesting that S. arboricola has a higher tolerance for soil salinity. Wood BC-amended soil led to better stomatal conductance (gs) and leaf area index (LAI) of both R. tomentosa and C. edithiae due to the higher water retention in the soil (22.68% and 20.79%). This illustrated that a higher amount of water retention brought by wood BC with a great amount of pore volume might be the limited factor for plant growth. The relationship between gs and LAI suggested that gs would not increase when LAI reached beyond 3. Moreover, wood and peanut shell BC caused a negative relationship between soil organic carbon and plant carbon content, suggesting that plants consume more carbon from the soil to store it in the plant. Overall, wood BC is recommended for plant growth of R. tomentosa and C. edithiae, and peanut shell BC is suggested for S. arboricola carbon storage. Graphical Abstract

Volatile components are one of the main active components of Ligusticum chuanxiong Hort and are important indicators for evaluating the medicinal quality of Ligusticum chuanxiong. To explore the effects of 60Co-γ irradiation on the volatile components of Ligusticum chuanxiong, the samples were treated with different irradiation doses, and the changes of the volatile components before and after irradiation were analyzed using an electronic nose combined with headspace solid-phase microextraction and gas chromatography-mass spectrometry. Principal component analysis (PCA), linear discriminant analysis (LDA), orthogonal partial least squares discriminant analysis (OPLS-DA), and cluster statistical analysis were used to elucidate changes in the volatile components of Ligusticum chuanxiong before and after irradiation. The electronic nose detection results and gas chromatography–mass spectrometry detection results indicated that ketone alkanes, alcohols, aldehydes, and ketones contributed to the odor of Ligusticum chuanxiong. Irradiation treatments at doses ≤10 kGy had no significant effect on the odor of Ligusticum chuanxiong. The results demonstrated that after irradiation at 0 kGy (control group), 3 kGy, 7 kGy, and 10 kGy, 60 compounds were identified in Ligusticum chuanxiong, including 23 hydrocarbons, 12 alcohols, 10 lipids, and 4 aldehydes or ketones, and 11 others. There were mainly 21 volatile substances that changed after irradiation, accounting for approximately 12.92% of the volatile components in Ligusticum chuanxiong; however, they were not the main volatile components of Ligusticum chuanxiong. No new compounds were identified in Ligusticum chuanxiong after irradiation treatment within a dose of 10 kGy. The results provide a theoretical basis and reference for the influence of 60Co-γ irradiation on the quality of Ligusticum chuanxiong samples and also lay a certain foundation for the application of irradiation in Ligusticum chuanxiong pieces and traditional Chinese medicine and simple preparation resources. The results indicate that the treatment of 60Co-γ irradiation on the Ligusticum chuanxiong samples with doses ˂10 kGy does not affect its main volatile components.

Abstract Background Postharvest quality deterioration poses a significant challenge to the commercial value of fresh lotus seeds. Low temperature storage is widely employed as the primary method for preserving postharvest lotus seeds during storage and transportation. Results This approach effectively extends the storage life of lotus seeds, resulting in distinct physiological changes compared to room temperature storage, including a notable reduction in starch, protein, H2O2, and MDA content. Here, we conducted RNA-sequencing to generate global transcriptome profiles of postharvest lotus seeds stored under room or low temperature conditions. Principal component analysis (PCA) revealed that gene expression in postharvest lotus seeds demonstrated less variability during low temperature storage in comparison to room temperature storage. A total of 14,547 differentially expressed genes (DEGs) associated with various biological processes such as starch and sucrose metabolism, energy metabolism, and plant hormone signaling response were identified. Notably, the expression levels of DEGs involved in ABA signaling were significantly suppressed in contrast to room temperature storage. Additionally, nine weighted gene co-expression network analysis (WGCNA)-based gene molecular modules were identified, providing insights into the co-expression relationship of genes during postharvest storage. Conclusion Our findings illuminate transcriptional differences in postharvest lotus seeds between room and low temperature storage, offering crucial insights into the molecular mechanisms of low temperature preservation in lotus seeds.

Abstract While high circulating tumor DNA (ctDNA) levels are associated with poor survival for multiple cancers, variant-specific differences in the association of ctDNA levels and survival have not been examined. Here we investigate KRAS ctDNA (ctKRAS) variant-specific associations with overall and progression-free survival (OS/PFS) in first-line metastatic pancreatic ductal adenocarcinoma (mPDAC) for patients receiving chemoimmunotherapy (“PRINCE”, NCT03214250), and an independent cohort receiving standard of care (SOC) chemotherapy. For PRINCE, higher baseline plasma levels are associated with worse OS for ctKRAS G12D (log-rank p = 0.0010) but not G12V (p = 0.7101), even with adjustment for clinical covariates. Early, on-therapy clearance of G12D (p = 0.0002), but not G12V (p = 0.4058), strongly associates with OS for PRINCE. Similar results are obtained for the SOC cohort, and for PFS in both cohorts. These results suggest ctKRAS G12D but not G12V as a promising prognostic biomarker for mPDAC and that G12D clearance could also serve as an early biomarker of response.

Abstract Background Cystic Fibrosis causing mutations in the gene CFTR, reduce the activity of the CFTR channel protein, and leads to mucus aggregation, airway obstruction and poor lung function. A role for CFTR in the pathogenesis of other muco-obstructive airway diseases such as Chronic Obstructive Pulmonary Disease (COPD) has been well established. The CFTR modulatory compound, Ivacaftor (VX-770), potentiates channel activity of CFTR and certain CF-causing mutations and has been shown to ameliorate mucus obstruction and improve lung function in people harbouring these CF-causing mutations. A pilot trial of Ivacaftor supported its potential efficacy for the treatment of mucus obstruction in COPD. These findings prompted the search for CFTR potentiators that are more effective in ameliorating cigarette-smoke (CS) induced mucostasis. Methods Small molecule potentiators, previously identified in CFTR binding studies, were tested for activity in augmenting CFTR channel activity using patch clamp electrophysiology in HEK-293 cells, a fluorescence-based assay of membrane potential in Calu-3 cells and in Ussing chamber studies of primary bronchial epithelial cultures. Addition of cigarette smoke extract (CSE) to the solutions bathing the apical surface of Calu-3 cells and primary bronchial airway cultures was used to model COPD. Confocal studies of the velocity of fluorescent microsphere movement on the apical surface of CSE exposed airway epithelial cultures, were used to assess the effect of potentiators on CFTR-mediated mucociliary movement. Results We showed that SK-POT1, like VX-770, was effective in augmenting the cyclic AMP-dependent channel activity of CFTR. SK-POT-1 enhanced CFTR channel activity in airway epithelial cells previously exposed to CSE and ameliorated mucostasis on the surface of primary airway cultures. Conclusion Together, this evidence supports the further development of SK-POT1 as an intervention in the treatment of COPD.

Abstract Vaccination has successfully controlled several infectious diseases although better vaccines remain desirable. Host response to vaccination studies have identified correlates of vaccine immunogenicity that could be useful to guide development and selection of future vaccines. However, it remains unclear whether these findings represent mere statistical correlations or reflect functional associations with vaccine immunogenicity. Functional associations, rather than statistical correlates, would offer mechanistic insights into vaccine-induced adaptive immunity. Through a human experimental study to test the immunomodulatory properties of metformin, an anti-diabetic drug, we chanced upon a functional determinant of neutralizing antibodies. Although vaccine viremia is a known correlate of antibody response, we found that in healthy volunteers with no detectable or low yellow fever 17D viremia, metformin-treated volunteers elicited higher neutralizing antibody titers than placebo-treated volunteers. Transcriptional and metabolomic analyses collectively showed that a brief course of metformin, started 3 days prior to YF17D vaccination and stopped at 3 days after vaccination, expanded oxidative phosphorylation and protein translation capacities. These increased capacities directly correlated with YF17D neutralizing antibody titers, with reduced reactive oxygen species response compared to placebo-treated volunteers. Our findings thus demonstrate a functional association between cellular respiration and vaccine-induced humoral immunity and suggest potential approaches to enhancing vaccine immunogenicity.

Abstract Background Guaranteeing nursing service safety and quality is a prioritized issue in the healthcare setting worldwide. However, there still lacks a valid scale to measure the quality and safety competencies of newly graduated nurses globally. Methods This scale was developed in two phases. In Phase One, a literature review and three-round e-Delphi were conducted to generate the initial item pool; while in Phase Two, five experts tested the content validity of the scale. The construct validity was evaluated using confirmatory factor analysis (CFA), and the data were collected among 1,221 newly graduated nursing students between May, 2017 and August, 2017. Finally, the internal consistency reliability and test-retest reliability were tested. Results The final version’s Competency Scale of Quality and Safety (CSQS) was confirmed by the CFA involving 64 items in six dimensions, including patient-center care, safety, evidence-based practice, collaboration and teamwork, continuous quality improvement, and informatics. The results of data showed that the data supported the modified model of CSQS (Standardized Root Mean Square Residual = 0.03, p = 0.053, Adjusted Goodness of Normed Fit Index = 1.00, Root Mean Square Error of Approximation = 0.007, Fit Index = 0.95, Goodness of Fit Index = 0.97, χ2/df = 1.06), and the standardized factor loadings of items were from 0.59 to 0.74 (p

IntroductionChronic schizophrenia has a course of 5 years or more and has a widespread abnormalities in brain functional connectivity. This study aimed to find characteristic functional and structural changes in a long illness duration chronic schizophrenia (10 years or more).MethodsThirty-six patients with a long illness duration chronic schizophrenia and 38 healthy controls were analyzed by independent component analysis of brain network functional connectivity. Correlation analysis with clinical duration was performed on six resting state networks: auditory network, default mode network, dorsal attention network, fronto-parietal network, somatomotor network, and visual network.ResultsThe differences in the resting state network between the two groups revealed that patients exhibited enhanced inter-network connections between default mode network and multiple brain networks, while the inter-network connections between somatomotor network, default mode network and visual network were reduced. In patients, functional connectivity of Cuneus_L was negatively correlated with illness duration. Furthermore, receiver operating characteristic curve of functional connectivity showed that changes in Thalamus_L, Rectus_L, Frontal_Mid_R, and Cerebelum_9_L may indicate a longer illness duration chronic schizophrenia.DiscussionIn our study, we also confirmed that the course of disease is significantly associated with specific brain regions, and the changes in specific brain regions may indicate that chronic schizophrenia has a course of 10 years or more.

Community parks in old urban areas have problems such as outdated facilities and low quality, which inhibit the development of healthy aging. However, limited research has examined the correlation between such park characteristics and park satisfaction among elderly individuals. Additionally, the mechanisms underlying this association remain unclear. In this investigation, a moderated mediation framework grounded in self-determination theory was formulated to probe the interplay between these variables, with a specific focus on the mediating influence of social self-efficacy among the elderly and the moderating effect of use intention. A questionnaire survey (N = 319) was conducted in Shamian Park, Guangzhou, employing structural equation modeling for data analysis. Findings indicate that, even after controlling for demographic factors, park characteristics not only exert a direct influence on satisfaction but also exhibit an indirect impact through the mediation of social self-efficacy, with both pathways being moderated by use intention. This study has implications for how to improve the design of community parks in old urban areas in developing countries to better meet the basic needs of the elderly and promote healthy aging.

The present study provides a wideband high-gain monopulse antenna based on a dielectric lens operating in X- and Ku-bands, in which a wideband dielectric lens is designed and employed to fulfill the radiation pattern and bandwidth necessities of a monopulse antenna. The proposed configuration has four horns allowing for the simultaneous creation of $\Delta $ and $\Sigma $ designs in two perpendicular planes. The main advantages of the proposed dielectric lens are low cost, lightweight, and easy fabrication using 3-D printing technology. The measurement findings show that the peak gain of the sum pattern is 28.9 dBi with a peak aperture efficiency of 60% over the desired frequency bandwidth. The suggested design can produce a simultaneous sum and two distinct difference patterns in orthogonal planes, meeting the rigorous demands for speed and accuracy in tracking applications.

ABSTRACT: Brown fermented milk (BFM) is favored by consumers in the dairy market for its unique burnt flavor and brown color. However, Maillard reaction products (MRP) from high-temperature baking are also noteworthy. In this study, tea polyphenols (TP) were initially developed as potential inhibitors of MRP formation in BFM. The results showed that the flavor profile of BFM did not change after adding 0.08% (wt/wt) of TP, and its inhibition rates on 5-hydroxymethyl-2-furaldehyde (5-HMF), glyoxal (GO), methylglyoxal (MGO), Nε-carboxymethyl lysine (CML), and Nε-carboxyethyl lysine (CEL) were 60.8%, 27.12%, 23.44%, 57.7%, and 31.28%, respectively. After 21 d of storage, the levels of 5-HMF, GO, MGO, CML, and CEL in BFM with TP were 46.3%, 9.7%, 20.6%, 5.2%, and 24.7% lower than the control group, respectively. Moreover, a smaller change occurred in their color and the browning index was lower than that of the control group. The significance of this study was to develop TP as additives to inhibit the production of MRP in brown fermented yogurt without changing color and flavors, thereby making dairy products safer for consumers.

Summary: Cells cease to proliferate above their growth-permissible temperatures, a ubiquitous phenomenon generally attributed to heat damage to cellular macromolecules. We here report that, in the presence of rapamycin, a potent inhibitor of Target of Rapamycin Complex 1 (TORC1), the fission yeast Schizosaccharomyces pombe can proliferate at high temperatures that usually arrest its growth. Consistently, mutations to the TORC1 subunit RAPTOR/Mip1 and the TORC1 substrate Sck1 significantly improve cellular heat resistance, suggesting that TORC1 restricts fission yeast growth at high temperatures. Aiming for a more comprehensive understanding of the negative regulation of high-temperature growth, we conducted genome-wide screens, which identified additional factors that suppress cell proliferation at high temperatures. Among them is Mks1, which is phosphorylated in a TORC1-dependent manner, forms a complex with the 14-3-3 protein Rad24, and suppresses the high-temperature growth independently of Sck1. Our study has uncovered unexpected mechanisms of growth restraint even below the temperatures deleterious to cell physiology.

In deregulated electricity markets, predicting price and load is a common practice. However, market participants and shareholders often seek deeper insights into other system statuses associated with price prediction, such as power flow and market share of generation companies (GenCos). These insights are challenging to obtain using purely data-driven methods. This paper proposes a physics-based solution for the probabilistic prediction of market-clearing outcomes, using real sanitized offer data from the National Electricity Market of Singapore (NEMS). Our approach begins with approximating the generator offers that have been historically cleared. Using this pool of offer data, we propose a probabilistic market-clearing process. This process allows for the probabilistic prediction of market prices. By considering the power system network and its constraints, we also naturally obtain probabilistic predictions of power flow and market shares. We validate our approach using actual NEMS data. Our findings show that while the overall performance of price prediction is comparable to existing methods, our proposed method can also provide probabilistic predictions of other associated system operating conditions. Furthermore, our method enables scenario studies, such as the impact of demand-side participation and the penetration of rooftop photovoltaic (PV) systems on the Uniform Singapore Energy Price (USEP).

We introduce a novel method that combines satellite data, advanced clustering techniques, machine learning feature extraction, and statistical models to enhance fishery forecasting accuracy. Focusing on jumbo flying squid in the southeast Pacific Ocean near Peru, we utilize MODIS-Aqua and MODIS-Terra satellite data on sea surface temperature (SST) to construct a deep convolutional embedded clustering (DCEC) model and extract the monthly SST features (FM) based on an optimized number of clusters determined by the Davies–Bouldi index (DBI). We use the extracted FM to construct a series of Generalized Additive Models (GAM) to forecast the catch per unit effort (CPUE) of jumbo flying squid within a spatial resolution of 0.5° × 0.5°. Our results demonstrate the following findings: (1) The SST feature clusters obtained through the DCEC model could capture the SST monthly variations; (2) The GAM models with FM outperform the models with the traditional monthly average SST in terms of predictive accuracy; (3) Using both FM and average SST together can further improve model performance. This study demonstrates the effectiveness of the DCEC combined with DBI in extracting marine environmental features and highlights the ocean environment feature extraction method to enhance the precision and reliability of fishery forecasting models.

Background: The exponential increase in childhood obesity poses a global health problem including Malaysia. Increased snacking behaviours could be one of the contributing factors to unhealthy eating habits and excessive energy intake. This study aimed to investigate the influence of snacking pattern (portion, frequency, energy contribution) and preference on the energy intake and weight status among Malaysian Chinese children aged 7–12 years in Klang Valley, Malaysia. Methods: A total of 99 Chinese children (37 boys, 62 girls) with a mean age of 10.4 ± 1.5 years old were recruited from three local primary schools. Weight and height were measured. Snacking behaviours (frequency, portion size, and preference) were assessed using a questionnaire. Two days of 24-h dietary recall were used to assess the children's dietary intake. Results: Most of the children (62.6%) had normal body weight status. Snacking contributed to 23.3% and 21.6% of total daily energy intake on weekdays and weekends respectively. The most preferred snacks were ice cream, yoghurt, and chocolate drinks, whereas the least preferred snacks were syrup, traditional kuih, and breads. There were no significant differences in snacking frequency and portion size by the amount of pocket money given for snacks (Pearson χ2 = 1.52, p = 0.47; Pearson χ2 = 3.19, p = 0.53, respectively). Conclusion: Education regarding the right choice of snacks is important to ensure better diet quality to optimise children's growth.

Abstract Coronavirus disease-19 (Covid-19) pandemic have demonstrated the importantance of vaccines in disease prevention. Self-amplifying mRNA vaccines could be another option for disease prevention if demonstrated to be safe and immunogenic. Phase 1 of this randomized, double-blinded, placebo-controlled trial (N = 42) assessed the safety, tolerability, and immunogenicity in healthy young and older adults of ascending levels of one-dose ARCT-021, a self-amplifying mRNA vaccine against Covid-19. Phase 2 (N = 64) tested two-doses of ARCT-021 given 28 days apart. During phase 1, ARCT-021 was well tolerated up to one 7.5 μg dose and two 5.0 μg doses. Local solicited AEs, namely injection-site pain and tenderness were more common in ARCT-021vaccinated, while systemic solicited AEs, mainly fatigue, headache and myalgia were reported in 62.8% and 46.4% of ARCT-021 and placebo recipients, respectively. Seroconversion rate for anti-S IgG was 100% in all cohorts, except for the 1 μg one-dose in younger adults and the 7.5 μg one-dose in older adults. Anti-S IgG and neutralizing antibody titers showed a general increase with increasing dose, and overlapped with titers in Covid-19 convalescent patients. T-cell responses were also observed in response to stimulation with S-protein peptides. Taken collectively, ARCT-021 is immunogenic and has favorable safety profile for further development.

Abstract Remarkable potency has been demonstrated for mRNA vaccines in reducing the global burden of the ongoing COVID-19 pandemic. An alternative form of the mRNA vaccine is the self-amplifying mRNA (sa-mRNA) vaccine, which encodes an alphavirus replicase that self-amplifies the full-length mRNA and SARS-CoV-2 spike (S) transgene. However, early-phase clinical trials of sa-mRNA COVID-19 vaccine candidates have questioned the potential of this platform to develop potent vaccines. We examined the immune gene response to a candidate sa-mRNA vaccine against COVID-19, ARCT-021, and compared our findings to the host response to other forms of vaccines. In blood samples from healthy volunteers that participated in a phase I/II clinical trial, greater induction of transcripts involved in Toll-like receptor (TLR) signalling, antigen presentation and complement activation at 1 day post-vaccination was associated with higher anti-S antibody titers. Conversely, transcripts involved in T-cell maturation at day 7 post-vaccination informed the magnitude of eventual S-specific T-cell responses. The transcriptomic signature for ARCT-021 vaccination strongly correlated with live viral vector vaccines, adjuvanted vaccines and BNT162b2 1 day post-vaccination. Moreover, the ARCT-021 signature correlated with day 7 YF17D live-attenuated vaccine transcriptomic responses. Altogether, our findings show that sa-mRNA vaccination induces innate immune responses that are associated with the development of adaptive immunity from other forms of vaccines, supporting further development of this vaccine platform for clinical application.

Abstract The development of immunotherapy agents has revolutionized the field of oncology. The only FDA-approved immunotherapeutic approach in breast cancer consists of immune checkpoint inhibitors, yet several novel immune-modulatory strategies are being actively studied and appear promising. Innovative immunotherapeutic strategies are urgently needed in triple negative breast cancer (TNBC), a subtype of breast cancer known for its poor prognosis and its resistance to conventional treatments. TNBC is more primed to respond to immunotherapy given the presence of more tumor infiltrating lymphocytes, higher PD-L1 expression, and higher tumor mutation burden relative to the other breast cancer subtypes, and therefore, immuno-oncology represents a key area of promise for TNBC research. The aim of this review is to highlight current data and ongoing efforts to establish the safety and efficacy of immunotherapeutic approaches beyond checkpoint inhibitors in TNBC.

Objective To evaluate standardized training simulation examination for residents trainees in ultrasound medicine. Methods From 2018 to 2020, all the 17 resident trainees participated in the training at the standardized training base for residents located in the department of ultrasound medicine of China-Japan Friendship Hospital.The examination was conducted in stages for capacity building for clinical reasoning, the equipment operation and humanity, three rounds.The assessment performance was analyzed in groups based on academic degree and grade respectively. Questionnaire survey was implemented for trainees' subjective evaluation for simulation examination. Results Both clinical reasoning and the ultrasonic operation dealing with heart check by the graduate group and the undergraduate group showed a progressing trend for three grades, and the postgraduate group had higher performance than the undergraduate group {[clinical reasoning (342.10±26.73) points vs (269.55±30.35) points, (336.00±22.58) points vs (287.18±22.18) points, (352.00±31.00) points vs (327.27±22.31) points], [untrasonic operation for heart check (89.67±7.59) points vs (62.00±2.00) points, (83.00±7.79) points vs(67.50±7.50) points, (97.33±2.05) points vs (89.50±2.50) points]}. There was no statistically significant difference among grades. Performance was evident between grades 1 and 3 with the machine operation [(243.19±18.39) points vs (268.00±25.06) points]. In the undergraduate group, the 1st and 3rd examination progressed significantly [(234.59±15.24) points vs (262.00±33.34) points]. The questionnaire showed that 17 trainees believed that the simulation examination effectively improved their ultrasound skills. Conclusions The simulation examination can improve the clinical skill and increase trainees' confidence, so this method should be shared by other institutions for standardized residents training.

Clostridium perfringens (C. perfringens) is a significant foodborne pathogen and a common cause of intestinal diseases in both animals and humans. Our study investigated MLST, phenotypic antimicrobial resistance profiles, and resistance genes among isolates from human, animal and food. 186 C. perfringens isolates were obtained from nine provinces in China between 2013 and 2021. Additionally, some specific ST complexes were analyzed by cgMLST and cgSNP to investigate genetic relatedness. MLST indicated the most prevalent STs of C. perfringens of human and animal origin were as follows: ST221 (5/147), ST62 (4/147), ST408 (4/147), and ST493 (4/147) were predominant in humans, while ST479 (5/25) was the major type in animals. Within the same ST complex, genetically unrelated relationships or potential clustering/transmission events were further recognized by cgMLST and cgSNP, illustrating that these two methods are valuable in defining outbreaks and transmission events. All tested isolates were susceptible to vancomycin and meropenem. The rates of resistance to metronidazole, penicillin, cefoxitin, moxifloxacin, and chloramphenicol were low (metronidazole: 1.08%; penicillin: 9.68%; cefoxitin: 0.54%; moxifloxacin: 6.45%; and chloramphenicol: 3.76%). Interestingly, 49.66% of human origin were clindamycin-resistant, and 18.2% were penicillin-insensitive. Importantly, the portion of MDR isolates was significantly lower than in previous reports. The study provides an overview of the epidemiological characteristics of C. perfringens with different origins and hosts in China. C. perfringens demonstrated remarkable genetic diversity and distinct molecular features compared to antibiotic-resistance profiles from other studies.

Pneumothorax is a potentially life-threatening disease that requires urgent diagnosis and treatment. Clinically, a chest X-ray examination is the first choice for diagnosing pneumothorax. However, it is difficult to diagnose pneumothorax by only frontal chest X-ray imaging when the lesion area is only composed of a small amount of air. Therefore, we propose a pneumothorax diagnosis neural network based on feature fusion, where frontal and lateral X-ray information are fused. In this network, there are two inputs and three outputs. The two inputs are the frontal chest X-ray image and the lateral chest X-ray image. The three outputs are the classification results of the frontal chest X-ray image, the classification results of the lateral chest X-ray image, and the classification results integrating the characteristics of the fused frontal chest X-ray image and lateral chest X-ray image. Our algorithm considers the vanishing gradient problem in the pneumothorax recognition model and introduces the residual block to alleviate this problem. Because of the large number of channels in this model, we also utilize channel attention mechanisms to improve the model’s performance. Our comparative experiments show that neural network fusion of frontal and lateral chest image features can achieve higher accuracy than the single task model. Using only image-level annotation, our pneumothorax model can achieve high recognition accuracy.

Summary: The emergence of SARS-CoV-2 variants raises concerns of reduced COVID-19 vaccine efficacy. We investigated the humoral immunity in uninfected and previously infected ChAdOx1 nCoV-19, BNT162b2 and CoronaVac vaccinees, who have received complete regimes of vaccines by means of a SARS-CoV-2 surrogate virus blocking test. The ChAdOx1 nCoV-19 (p = 0.0013) and BNT162b2 (p = 0.0005) vaccines induced significant higher blocking activity with longer durability against the Spike (S) protein receptor binding domain (RBD) of wild type SARS-CoV-2 than the CoronaVac vaccine in uninfected vaccinees. Prior infection improved protection in the CoronaVac vaccinees. Subsequent investigation on the breadth of SARS-CoV-2 vaccine-induced antibody blocking responses, revealed that all vaccine platforms cross-protected uninfected vaccinees against all variant of concerns, except Omicron. Prior infection protected the ChAdOx1 nCoV-19 and BNT162b2 vaccinees against Omicron but not CoronaVac vaccinees. Our study suggests that vaccines that induce broader sterilizing immunity are essential to fight against fast-emerging variants.

The pathogenicity of bacteria is closely related to their tolerance to environmental stress. When the pathogenic Vibrio bacteria is exposed to external stressosome, a protein-signaling hub within Vibrio bacteria senses and transmits stress signals through reversible phosphorylation. In order to explore the influence of ultra-high pressure (UHP) on the conformation of RsbT protein kinase in Vibrio and on its downstream signaling transduction, three pressure points of 100 MPa, 300 MPa and 500 MPa are used in this experiment.Each pressure point is kept for 5 minutes. After western blotting, in the RsbT protein kinase, phosphorylation ability decreases as the pressure increases. The RsbT conformation alters and its phosphorylation ability almost disappears when the pressure is above 300 MPa. This study offers new theoretical support for the influence of pressure on RsbT protein kinase conformation and biological activity in stressosome, which provide insights into treatment of food-bone bacteria using UHP.

This research aimed to describe the functions of vascular endothelial cells (VECs) in protecting target organs and the anti-atherosclerotic effects of different enantiomers of amlodipine on a rabbit model of atherosclerosis. Thirty male New Zealand white rabbits were randomly allocated to four groups (nA = 9, nB = 7, nC = 7, and nD = 7 rabbits): rabbits in group-A (control group) were fed a high-fat diet, group-B rabbits were fed a high-fat diet plus 2.5 mg/kg/day S-amlodipine, group-C rabbits were fed a high-fat diet plus 2.5 mg/kg/day R-amlodipine, and group-D rabbits were fed a high-fat diet plus 5 mg/kg/day racemic amlodipine. Different enantiomers of amlodipine did not influence lipid profiles and serum level of eNOS in the rabbit atherosclerosis model but decreased ET-1 expression to some extent. The serum NO and iNOS levels in the drug intervention groups were significantly reduced. No significant differences in the rabbits’ body weights were observed. At the 4th and 8th weeks, the serum lipid profiles significantly increased in high cholesterol diet groups. The serum ET-1 level was significantly increased in each group of rabbits at the 8th week. Both S-amlodipine and R-amlodipine may protect the endothelium by reducing the serum ET-1 level, downregulating iNOS expression.

This paper explored the the method, path and content of "One Syndrome and One Product" Traditional Chinese Medicine (TCM) nursing service in the specialized spine department, and summarized the construction and practice of "One Syndrome and One Product" nursing plan. The nursing plan consisted of several important steps as follows: establishing standardized nurse etiquette and service behavior; creating the clinical ward with comfort and warm surroundings; carrying out preparation services for discharged patients; improving nursing service experience to satisfy patients; Strengthening basic nursing and creating a humanistic care service model for patients. Taking low back pain with blood-stasis and Qi-stagnation syndrome as an example, syndrome differentiation nursing was carried out and related TCM nursing program was optimized. Implementation of "One Syndrome and One Product" nursing plan was associate with a continued increase of patient satisfaction, establishment of a humanistic care service model, transformation of nurses’ service concepts, and the formation of caring culture in the department. Through the development of syndrome differentiation nursing, an optimized TCM nursing program with low back pain of blood-stasis and Qi-stagnation syndrome was formed. NAC (NAM, ATAF, and CUC) is a ubiquitously expressed plant-specific transcription factor (TF) family which is involved in the regulation of various biological processes. However, a systematic characterization of NAC gene family is yet to be reported in lotus. Here, 82 NnNAC genes which included five predicted membrane-bound NAC proteins were identified in the lotus genome. Phylogenetic analysis revealed seven-subfamily clusters (I–VII) of NnNAC proteins, with homologous gene pairs displaying similar conserved motifs and gene structure characteristics. Transactivation assay of NnNAC proteins revealed an extensive transcriptional activation capacity which is mediated by the highly divergent C-terminal activation domain (AD). Expression analysis of NnNAC genes in lotus tissues showed high transcript levels in root, stamen, petal and seed coat. In addition, 30 and 29 differentially expressed NnNAC candidate genes putatively involved in lotus seed development and response to complete submergence stress, respectively, were identified. Overall, our study provides potentially useful candidate gene resources for future molecular breeding of lotus varieties with novel agronomic traits.

Ensuring high vaccination and even booster vaccination coverage is critical in preventing severe Coronavirus Disease 2019 (COVID-19). Among the various COVID-19 vaccines currently in use, the mRNA vaccines have shown remarkable effectiveness. However, systemic adverse events (AEs), such as postvaccination fatigue, are prevalent following mRNA vaccination, and the underpinnings of which are not understood. Herein, we found that higher baseline expression of genes related to T and NK cell exhaustion and suppression were positively correlated with the development of moderately severe fatigue after Pfizer-BioNTech BNT162b2 vaccination; increased expression of genes associated with T and NK cell exhaustion and suppression reacted to vaccination were associated with greater levels of innate immune activation at 1 day postvaccination. We further found, in a mouse model, that altering the route of vaccination from intramuscular (i.m.) to subcutaneous (s.c.) could lessen the pro-inflammatory response and correspondingly the extent of systemic AEs; the humoral immune response to BNT162b2 vaccination was not compromised. Instead, it is possible that the s.c. route could improve cytotoxic CD8 T-cell responses to BNT162b2 vaccination. Our findings thus provide a glimpse of the molecular basis of postvaccination fatigue from mRNA vaccination and suggest a readily translatable solution to minimize systemic AEs. Systemic adverse events, such as post-vaccination fatigue, are prevalent consequences of mRNA vaccination; why is this? This study shows that higher baseline expression of T and NK cell genes increases susceptibility to fatigue after mRNA vaccination, and that altering the route of vaccination may reduce the incidence of mRNA vaccine-associated systemic adverse events.

Lotus plumule is a green tissue in the middle of seeds that predominantly accumulates bisbenzylisoquinoline alkaloids (bis-BIAs) and chlorophyll (Chl). However, the biosynthetic mechanisms of these two metabolites remain largely unknown in lotus. This study used physiological and RNA sequencing (RNA-Seq) approaches to characterize the development and molecular mechanisms of bis-BIAs and Chl biosynthesis in lotus plumule. Physiological analysis revealed that exponential plumule growth occurred between 9 and 15 days after pollination (DAP), which coincided with the onset of bis-BIAs biosynthesis and its subsequent rapid accumulation. Transcriptome analysis of lotus plumule identified a total of 8,725 differentially expressed genes (DEGs), representing ~27.7% of all transcripts in the lotus genome. Sixteen structural DEGs, potentially associated with bis-BIAs biosynthesis, were identified. Of these, 12 encoded O-methyltransferases (OMTs) are likely involved in the methylation and bis-BIAs diversity in lotus. In addition, functionally divergent paralogous and redundant homologous gene members of the BIAs biosynthesis pathway, as well as transcription factors co-expressed with bis-BIAs and Chl biosynthesis genes, were identified. Twenty-two genes encoding 16 conserved enzymes of the Chl biosynthesis pathway were identified, with the majority being significantly upregulated by Chl biosynthesis. Photosynthesis and Chl biosynthesis pathways were simultaneously activated during lotus plumule development. Moreover, our results showed that light-driven Pchlide reduction is essential for Chl biosynthesis in the lotus plumule. These results will be useful for enhancing our understanding of alkaloids and Chl biosynthesis in plants.

Academic burnout seriously affects the academic performance and mental health of college students. This study developed a multi-mediation model to investigate the relationship between academic self-efficacy and academic burnout. A total of 1431 undergraduate students (51.85% female) were recruited to participate in this study. And we used SPSS PROCESS developed by Hayes to examine the mediating role of academic self-efficacy and academic burnout. The results indicated that academic self-efficacy can negatively predict academic burnout. Moreover, intrinsic motivation and learning engagement have a significant mediating effect between academic self-efficacy and academic burnout. Finally, we constructed a model comprising four variables and found that it explained 47.62% of the variance in academic burnout. These findings can be conducive to a suitable intervention path to reduce academic burnout of Chinese college students, so as to develop effective instructional strategies to improve their learning performance.

The interplay of topology, magnetism, and correlations gives rise to intriguing phases of matter. In this study, through state-of-the-art angle-resolved photoemission spectroscopy, density functional theory, and dynamical mean-field theory calculations, we visualize a fourfold degenerate Dirac nodal line at the boundary of the bulk Brillouin zone in the antiferromagnet YMn2Ge2. We further demonstrate that this gapless, antiferromagnetic Dirac nodal line is enforced by the combination of magnetism, space-time inversion symmetry, and nonsymmorphic lattice symmetry. The corresponding drumhead surface states traverse the whole surface Brillouin zone. YMn2Ge2 thus serves as a platform to exhibit the interplay of multiple degenerate nodal physics and antiferromagnetism. Interestingly, the magnetic nodal line displays a d-orbital dependent renormalization along its trajectory in momentum space, thereby manifesting Hund's coupling. Our findings offer insights into the effect of electronic correlations on magnetic Dirac nodal lines, leading to an antiferromagnetic Hund nodal line.
