Quantum computing leverages the quantum resources of superposition and entanglement to efficiently solve computational problems considered intractable for classical computers. Examples include calculating molecular and nuclear structure, simulating strongly interacting electron systems, and modeling aspects of material function. While substantial theoretical advances have been made in mapping these problems to quantum algorithms, there remains a large gap between the resource requirements for solving such problems and the capabilities of currently available quantum hardware. Bridging this gap will require a co-design approach, where the expression of algorithms is developed in conjunction with the hardware itself to optimize execution. Here we describe an extensible co-design framework for solving chemistry problems on a trapped-ion quantum computer and apply it to estimating the ground-state energy of the water molecule using the variational quantum eigensolver (VQE) method. The controllability of the trapped-ion quantum computer enables robust energy estimates using the prepared VQE ansatz states. The systematic and statistical errors are comparable to the chemical accuracy, which is the target threshold necessary for predicting the rates of chemical reaction dynamics, without resorting to any error mitigation techniques based on Richardson extrapolation.

Circulating tumor cells (CTCs) have the potential of becoming the gold standard marker for cancer diagnosis, prognosis and monitoring. However, current methods for its isolation and characterization suffer from equipment variability and human operator error that hinder its widespread use. Here we report the design and construction of a fully automated high-throughput fluorescence microscope that enables the imaging and classification of cancer cells that were labeled by immunostaining procedures. An excellent agreement between our machine vision-based approach and a state-of-the-art microscopy equipment was achieved. Our integral approach provides a path for operator-free and robust analysis of cancer cells as a standard clinical practice.

The detection and analysis of circulating tumor cells (CTCs) may enable a broad range of cancer-related applications, including the identification of acquired drug resistance during treatments. However, the non-scalable fabrication, prolonged sample processing times, and the lack of automation, associated with most of the technologies developed to isolate these rare cells, have impeded their transition into the clinical practice. This work describes a novel membrane-based microfiltration device comprised of a fully automated sample processing unit and a machine-vision-enabled imaging system that allows the efficient isolation and rapid analysis of CTCs from blood. The device performance was characterized using four prostate cancer cell lines, including PC-3, VCaP, DU-145, and LNCaP, obtaining high assay reproducibility and capture efficiencies greater than 93% after processing 7.5 mL blood samples from healthy donors, spiked with 100 cancer cells. Cancer cells remained viable after filtration due to the minimal shear stress exerted over cells during the procedure, while the identification of cancer cells by immunostaining was not affected by the number of non-specific events captured on the membrane. We were also able to identify the androgen receptor (AR) point mutation T878A from 7.5 mL blood samples spiked with 50 LNCaP cells using RT-PCR and Sanger sequencing. Finally, CTCs were detected in 8 of 8 samples from patients diagnosed with metastatic prostate cancer (mean $\pm$ SEM = 21 $\pm$ 2.957 CTCs/mL, median = 21 CTC/mL), thereby validating the potential clinical utility of the device., Comment: 13 pages, 6 figures, under review

We actively stabilize the harmonic oscillation frequency of a laser-cooled atomic ion confined in a radiofrequency (rf) Paul trap by sampling and rectifying the high voltage rf applied to the trap electrodes. We are able to stabilize the 1 MHz atomic oscillation frequency to be better than 10 Hz or 10 ppm. This represents a suppression of ambient noise on the rf circuit by 34 dB. This technique could impact the sensitivity of ion trap mass spectrometry and the fidelity of quantum operations in ion trap quantum information applications.

We sense the motion of a trapped atomic ion using a sequence of state-dependent ultrafast momentum kicks. We use this atom interferometer to characterize a nearly-pure quantum state with $n=1$ phonon and accurately measure thermal states ranging from near the zero-point energy to $\bar{n}\sim 10^4$, with the possibility of extending at least 100 times higher in energy. The complete energy range of this method spans from the ground state to far outside of the Lamb-Dicke regime, where atomic motion is greater than the optical wavelength. Apart from thermometry, these interferometric techniques are useful for characterizing ultrafast entangling gates between multiple trapped ions.

Optical imaging systems are used extensively in the life and physical sciences because of their ability to non-invasively capture details on the microscopic and nanoscopic scales. Such systems are often limited by source or detector noise, image distortions and human operator misjudgement. Here, we report a general, quantitative method to analyse and correct these errors. We use this method to identify and correct optical aberrations in an imaging system for single atoms and realize an atomic position sensitivity of ∼0.5 nm Hz−1/2 with a minimum uncertainty of 1.7 nm, allowing the direct imaging of atomic motion. This is the highest position sensitivity ever measured for an isolated atom and opens up the possibility of performing out-of-focus three-dimensional particle tracking, imaging of atoms in three-dimensional optical lattices or sensing forces at the yoctonewton (10−24 N) scale. The position of a single Yb atomic ion is determined with a minimum uncertainty of 1.7 nm for 0.2 s integration time — the highest position sensitivity reported to date for an isolated atom.

CORRIGENDUM: InterMOD: integrated data and tools for the unification of model organism research

Hybrid quantum systems can be formed that combine the strengths of multiple platforms while avoiding the weaknesses. Here we report on progress toward a hybrid quantum system of neutral atom spins coupled to superconducting qubits. We trap laser-cooled rubidium atoms in the evanescent field of an ultrathin optical fiber, which will be suspended a few microns above a superconducting circuit that resonates at the hyperfine frequency of the Rb atoms, allowing magnetic coupling between the atoms and superconductor. As this will be done in a dilution refrigerator environment, the technical demands on the optical fiber is severe. We have developed and optimized a tapered fiber fabrication system, achieving optical transmission in excess of 99.95% , and fibers that can sustain 400 mW of optical power in a UHV environment. We have also optimized tapered fibers that can support higher order optical modes with high transmission (> 97%), which may be useful for different optical potential geometries. We have developed an in-situ tunable high-Q superconducting microwave resonator that can be tuned to within the resonator linewidth of the 6.8 GHz frequency of the Rb hyperfine transition.

Model organisms are widely used for understanding basic biology and have significantly contributed to the study of human disease. In recent years, genomic analysis has provided extensive evidence of widespread conservation of gene sequence and function amongst eukaryotes, allowing insights from model organisms to help decipher gene function in a wider range of species. The InterMOD consortium is developing an infrastructure based around the InterMine data warehouse system to integrate genomic and functional data from a number of key model organisms, leading the way to improved cross-species research. So far including budding yeast, nematode worm, fruit fly, zebrafish, rat and mouse, the project has set up data warehouses, synchronized data models and created analysis tools and links between data from different species. The project unites a number of major model organism databases, improving both the consistency and accessibility of comparative research, to the benefit of the wider scientific community.

We present an experimental and theoretical study of the energy transfer between modes during the tapering process of an optical nanofiber through spectrogram analysis. The results allow optimization of the tapering process, and we measure transmission in excess of 99.95% for the fundamental mode. We quantify the adiabaticity condition through calculations and place an upper bound on the amount of energy transferred to other modes at each step of the tapering, giving practical limits to the tapering angle., Comment: 29 pages, 17 figures

Ninety-two strains of Yersinia pestis recovered over a 21-year period were evaluated for susceptibility to traditional and newer antimicrobial agents. In vitro resistance was noted only against rifampin and imipenem (∼20% of strains). The most active compounds (MIC at which 90% of the isolates tested are inhibited) against Y. pestis were cefixime, ceftriaxone, trimethoprim-sulfamethoxazole, and trovafloxacin.

Although pronouncements of the ¿health care crisis¿ have been made for thirty years, in many ways the fundamental underpinnings of the U.S. health care system have changed very little. However, calls for health care reform have progressed through four distinct phases. From before the 1965 passage of the Medicare and Medicaid programs until about 1972, the primary issue was expanding access to care. From 1972 through 1976, health care reform proposals experienced a transition period. From 1976 until about 1992, cost containment became the dominant theme. Since 1992 there has been a renewed emphasis on improving access to care. But Americans have mixed feelings about health care reform. On the one hand, Americans want improved access to care, increased efficiency, and high quality care. On the other hand, Americans are leery of higher taxes and expanding the government's role in health care decision-making. In the final analysis, it is ultimately politics which governs the form of health care reform by effecting compromises between what is ideologically desirable and what is economically plausible.
