Soot particles (SP) are ubiquitous components of atmospheric particulate matter and have been shown to cause various adverse health effects. In the atmosphere, freshly emitted SP can be coated by condensed low-volatility secondary organic and inorganic species. In addition, gas-phase oxidants may react with the surface of SP. Due to the chemical and physical resemblance of SP carbon backbone with polyaromatic hydrocarbon species and their potent oxidation products, we investigated the biological responses of BEAS-2B lung epithelial cells following exposure to fresh- and photochemically aged-SP at the air–liquid interface. A comprehensive physical and chemical aerosol characterization was performed to depict the atmospheric transformations of SP, showing that photochemical aging increased the organic carbon fraction and the oxidation state of the SP. RNA-sequencing and qPCR analysis showed varying gene expression profiles for fresh- and aged-SP. Exposure to aged-SP increased DNA damage, oxidative damage, and upregulation of NRF2-mediated oxidative stress response genes compared to fresh-SP. Furthermore, aged-SP augmented inflammatory cytokine secretion and activated AhR-response, as evidenced by increased expression of AhR-responsive genes. These results indicate that oxidative stress, inflammation, and DNA damage play a key role in the cytotoxicity of SP in BEAS-2B cells, where aging leads to higher toxic responses. Collectively, our results suggest that photochemical aging may increase SP toxicity through surface modifications that lead to an increased toxic response by activating different molecular pathways.

The most commonly used construction material nowadays is steel-reinforced concrete which underlies corrosion and thus buildings are susceptible for structural collapses. Recently, a new construction material resistant to oxidation and with a higher tensile strength called carbon concrete composite (C3) was developed. The new material allows resource-saving constructions using carbon fiber instead of steel reinforcement materials embedded in a concrete matrix. C3 reinforcements consist of carbon fibers coated with an organic polymer matrix. In this study, abrasive dust from a dry cutting process of two C3 reinforcement materials, as well as a C3 material were investigated with respect to the occurrence of toxic fibers or harmful organic compounds in the inhalable particulate matter (PM) fractions PM2.5 and PM10. It could be shown that the ratio between elemental and organic carbon in PM10 is dependent on the shape of the C3 reinforcement material due to different mechanisms of PM formation. This could have an impact on the toxicity of different C3 reinforcement materials. Harmful fibers according to the World Health Organization (WHO) definition like they are found in asbestos concrete were not found. However, bisphenol A (BPA) as well as the PAHs phenanthrene, anthracene, fluoranthene and pyrene were found due to pyrolysis of the organic matrix material at the edge of the sawblade differentiating the Carcinogenic Equivalency (TEQ) of investigated materials and their PM fractions. Furthermore, derivatives of BPA occurred in abrasive dust from C3 reinforcement materials potentially leading to genotoxicity and reproductive toxicity.

Anthropogenic air pollution has a severe impact on climate and human health. The immense molecular complexity and diversity of particulate matter (PM) is a result of primary organic aerosol (POA) as well as secondary organic aerosols (SOAs). In this study, a direct inlet probe (DIP), i.e., atmospheric solids analysis probe (ASAP), with ion mobility high-resolution mass spectrometric detection is applied. Primary particulate matter emissions from three sources were investigated. Furthermore, photochemically aged emissions were analyzed. DIP introduction allowed for a direct analysis with almost no sample preparation and resulted in a complex molecular pattern. This pattern shifted through oxidation processes toward heavier species. For diesel emissions, the fuel's chemical characteristic is partially transferred to the particulate matter by incomplete combustion and characteristic alkylated series were found. Polycyclic aromatic hydrocarbons (PAHs) were identified as major contributors. Ion mobility analysis results in drift time profiles used for structural analysis. The apex position was used to prove structural changes, whereas the full-width-at-half-maximum was used to address the isomeric diversity. With this concept, the dominance of one or a few isomers for certain PAHs could be shown. In contrast, a broad isomeric diversity was found for oxygenated species. For the in-depth specification of fresh and aged spruce emissions, the ion mobility resolving power was almost doubled by allowing for three passes in the circular traveling wave design. The results prove that ASAP coupled with ion mobility spectrometry-mass spectrometry (IMS-MS) serves as a promising analytical approach for tackling the vast molecular complexity of PM.

The emissions of marine diesel engines have gained both global and regional attentions because of their impact on human health and climate change. To reduce ship emissions, the International Maritime Organization capped the fuel sulfur content of marine fuels. Consequently, either low-sulfur fuels or additional exhaust gas cleaning devices for the reduction in sulfur dioxide (SO2) emissions became mandatory. Although a wet scrubber reduces the amount of SO2 significantly, there is still a need to consider the reduction in particle emissions directly. We present data on the particle removal efficiency of a scrubber regarding particle number and mass concentration with different marine fuel types, marine gas oil, and two heavy fuel oils (HFOs). An open-loop sulfur scrubber was installed in the exhaust line of a marine diesel test engine. Fine particulate matter was comprehensively characterized in terms of its physical and chemical properties. The wet scrubber led up to a 40% reduction in particle number, whereas a reduction in particle mass emissions was not generally determined. We observed a shift in the size distribution by the scrubber to larger particle diameters when the engine was operated on conventional HFOs. The reduction in particle number concentrations and shift in particle size were caused by the coagulation of soot particles and formation/growing of sulfur-containing particles. Combining the scrubber with a wet electrostatic precipitator as an additional abatement system showed a reduction in particle number and mass emission factors by >98%. Therefore, the application of a wet scrubber for the after-treatment of marine fuel oil combustion will reduce SO2 emissions, but it does not substantially affect the number and mass concentration of respirable particulate matters. To reduce particle emission, the scrubber should be combined with additional abatement systems.

Airborne particles affect air quality, weather and climate. The continuing urbanization is expected to expose a growing fraction of the world’s population to still increasing levels of anthropogenic emissions of airborne particles and precursors of secondary aerosols. These anthropogenically emitted particles are dominated in number by ultrafine particles (UFP; diameter less than 100 nm), which therefore are common in urban air. Their size and chemical composition determine whether the particles impose a risk to human health or the environment. Thus, in-depth knowledge about airborne UFPs' sources and atmospheric fate is essential for environmental risk assessment. A detailed chemical analysis of UFPs aids to better understand environmental processes in the atmosphere and possible effects on human health.Despite the need of learning more about the origin, behaviour, mobility, fate, and toxicity of UFPs, attempts to analyze their chemical composition in the atmosphere are still rare. Considering their low mass, partial volatility and dynamic character, it is still a great challenge to separate, catch and analyze UFPs in the atmosphere.Impactors are important tools to separate and collect environmental particles from the air with the aim of analyzing their chemical composition. Herein, we report our evaluation of commercially available and frequently deployed cascade impactors for their applicability of sampling airborne UFPs. We tested the following criteria: (1) A precise size separation or cut‑off in the ultrafine range to enable size-dependent chemical analysis, (2) The collection of the greatest as possible particle mass (high sampling volume) while minimizing evaporation losses of semi-volatile fractions (small pressure drop). Therefore, different impactors were connected in line between a customizable particle generation source, a flow reactor for dilution, mixing and ageing, and a mobility particle size spectrometer (MPSS). So far, our results indicate a significant variability among impactors of the same model and highlight the difficulty of combining all these requirements in one device. However, after careful physical characterization, we developed a strategy to optimize the particle sampling for atmospheric UFPs chemical composition analysis.This project is financed by the Bavarian Ministry of the Environment and Consumer Protection.

BACKGROUND: Secondary organic aerosols (SOAs) formed from anthropogenic or biogenic gaseous precursors in the atmosphere substantially contribute to the ambient fine particulate matter [PM ≤2.5μm in aerodynamic diameter (PM2.5)] burden, which has been associated with adverse human health effects. However, there is only limited evidence on their differential toxicological impact. OBJECTIVES: We aimed to discriminate toxicological effects of aerosols generated by atmospheric aging on combustion soot particles (SPs) of gaseous biogenic (β-pinene) or anthropogenic (naphthalene) precursors in two different lung cell models exposed at the air-liquid interface (ALI). METHODS: Mono- or cocultures of lung epithelial cells (A549) and endothelial cells (EA.hy926) were exposed at the ALI for 4 h to different aerosol concentrations of a photochemically aged mixture of primary combustion SP and β-pinene (SOAβPIN-SP) or naphthalene (SOANAP-SP). The internally mixed soot/SOA particles were comprehensively characterized in terms of their physical and chemical properties. We conducted toxicity tests to determine cytotoxicity, intracellular oxidative stress, primary and secondary genotoxicity, as well as inflammatory and angiogenic effects. RESULTS: We observed considerable toxicity-related outcomes in cells treated with either SOA type. Greater adverse effects were measured for SOANAP-SP compared with SOAβPIN-SP in both cell models, whereas the nano-sized soot cores alone showed only minor effects. At the functional level, we found that SOANAP-SP augmented the secretion of malondialdehyde and interleukin-8 and may have induced the activation of endothelial cells in the coculture system. This activation was confirmed by comet assay, suggesting secondary genotoxicity and greater angiogenic potential. Chemical characterization of PM revealed distinct qualitative differences in the composition of the two secondary aerosol types. DISCUSSION: In this study using A549 and EA.hy926 cells exposed at ALI, SOA compounds had greater toxicity than primary SPs. Photochemical aging of naphthalene was associated with the formation of more oxidized, more aromatic SOAs with a higher oxidative potential and toxicity compared with β-pinene. Thus, we conclude that the influence of atmospheric chemistry on the chemical PM composition plays a crucial role for the adverse health outcome of emissions. https://doi.org/10.1289/EHP9413.

Adverse health effects driven by airborne particulate matter (PM) are mainly associated with reactive oxygen species formation, pro-inflammatory effects, and genome instability. Therefore, a better understanding of the underlying mechanisms is needed to evaluate health risks caused by exposure to PM. The aim of this study was to compare the genotoxic effects of two oxidizing agents (menadione and 3-chloro-1,2-propanediol) with three different reference PM (fine dust ERM-CZ100, urban dust SRM1649, and diesel PM SRM2975) on monocytic THP-1 and alveolar epithelial A549 cells. We assessed DNA oxidation by measuring the oxidized derivative 8-hydroxy-2'-deoxyguanosine (8-OHdG) following short and long exposure times to evaluate the persistency of oxidative DNA damage. Cytokinesis-block micronucleus cytome assay was performed to assess chromosomal instability, cytostasis, and cytotoxicity. Particles were characterized by inductively coupled plasma mass spectrometry in terms of selected elemental content, the release of ions in cell medium and the cellular uptake of metals. PM deposition and cellular dose were investigated by a spectrophotometric method on adherent A549 cells. The level of lipid peroxidation was evaluated via malondialdehyde concentration measurement. Despite differences in the tested concentrations, deposition efficiency, and lipid peroxidation levels, all reference PM samples caused oxidative DNA damage to a similar extent as the two oxidizers in terms of magnitude but with different oxidative DNA damage persistence. Diesel SRM2975 were more effective in inducing chromosomal instability with respect to fine and urban dust highlighting the role of polycyclic aromatic hydrocarbons derivatives on chromosomal instability. The persistence of 8-OHdG lesions strongly correlated with different types of chromosomal damage and revealed distinguishing sensitivity of cell types as well as specific features of particles versus oxidizing agent effects. In conclusion, this study revealed that an interplay between DNA oxidation persistence and chromosomal damage is driving particulate matter-induced genome instability.

Solid fuel usage in residential heating and cooking is one of the largest sources of ambient and indoor air particulate matter, which causes adverse effects on the health of millions of peoples worldwide. Emissions from solid fuel combustion, such as biomass or coal, are detrimental to health, but toxicological responses are largely unknown. In the present study, we compared the toxicological responses regarding cytotoxicity, inflammation and genotoxicity of spruce (SPR) and brown coal briquette (BCB) combustion aerosols on human alveolar epithelial cells (A549) as well as a coculture of A549 and differentiated human monocytic cells (THP-1) into macrophages exposed at the air-liquid interface (ALI). We included both the high emissions from the first hour and moderate emissions from the third hour of the batch combustion experiment in one ALI system, whereas, in the second ALI system, we exposed the cells during the whole 4-hour combustion experiment, including all combustion phases. Physico-chemical properties of the combustion aerosol were analysed both online and offline. Both SPR and BCB combustion aerosols caused mild cytotoxic but notable genotoxic effects in co-cultured A549 cells after one-hour exposure. Inflammatory response analysis revealed BCB combustion aerosols to cause a mild increase in CXCL1 and CXCL8 levels, but in the case of SPR combustion aerosol, a decrease compared to control was observed.

BackgroundAdvancement in the instrumentation techniques made it possible to identify the compositions and concentrations of various chemicals, such as organic compounds or metals in combustion aerosols. However, the prediction of combustion aerosol-induced toxicity end points in lung epithelium cells is difficult due to the large number of non-linear chemical-biological interactions. It is evident that some chemicals present in the combustion aerosols upregulate certain genes/ proteins while other chemicals downregulate them, making the prediction of toxicity of a mixture of chemicals a challenging task. Also, the presence of large numbers of feedback and feedforward regulatory loops makes the entire prediction process highly dynamic. MethodsHere we present an integrative workflow to construct and analyze combustion aerosol-type specific chemical-gene regulatory network. For this, we develop an algorithm to estimate the combined regulatory effect of chemicals present in the combustion aerosol on each of the genes/proteins in the network. Further we rank the nodes by combining various network topological and non-topological parameters using a multi-objective optimization function. The top ranked nodes were used to identify aerosol-type specific key regulators that contributes toward the understanding of adverse outcomes due to the exposure of combustion aerosols. ResultsThe integrative workflow is evaluated using transcriptomics analysis carried out on human bronchial epithelium cell line BEAS-2B exposed at the air liquid interface (ALI) to the combustion aerosols generated by ship engine running either on distillate Diesel Fuel (DF) or on Heavy Fuel Oil (HFO) along with chemometric analysis. Based on the prioritized chemicals, we prepared DF and HFO combustion aerosols-specific chemical-gene regulatory network. Our study reveals the large differences in the observed biological effects caused by operating the same ship engine on the different fuels.ConclusionThe presented workflow can be used to investigate key regulatory processes associated with the toxicity outcomes of mixture of chemicals and also for the hazard classification and assessment of various combustion aerosols.

The study investigates the spatial pattern of black carbon (BC) at a high spatial resolution in Augsburg, Germany. Sixty two walks were performed to assess the concentrations of equivalent black carbon (eBC), ultraviolet particulate matter (UVPM), and equivalent brown carbon (eBrC) in different seasons and at different times of the day with a mobile platform (i.e., trolley). Along with BC measurements, images of street microenvironments were recorded. Meteorological parameters, including temperature, relative humidity, and wind speed, were monitored. The BC concentrations showed significant spatial heterogeneity and diurnal variations peaking in the morning and at night. The highest BC concentrations were observed near dense traffic. The correlations between BC and street views (buildings, roads, cars, and vegetation) were weak but highly significant. Moreover, meteorological factors also influenced the BC concentration. A model based on street view images and meteorological data was developed to examine the driving factors of the spatial variability of BC concentrations at a higher spatial resolution as different microenvironments based on traffic density. The best results were obtained for UVPM and eBC (71 and 70% explained variability). eBrC (53%), to which other sources besides road traffic can also make significant contributions, is modeled less well.

Allergic diseases are an increasing global burden. Epidemiological and in vivo studies showed that farming environments could protect from allergic asthma. Studies explaining this protective effect mainly focused on the influence of chemical compounds in the molecular size range of proteins and endotoxins. Our study aimed at deciphering the possible role of small-sized semi-volatile organic compounds (SVOCs) of farming aerosols in immunomodulation processes. Bronchial epithelial BEAS-2B cells were exposed to aerosol extracts of particulate matter (PM2.5) from farming environments. These cell exposures revealed a decisive effect of the smaller sized fraction (TLR2). Additionally, we performed a comprehensive chemical investigation of two typical farming aerosols (cow vs. sheep) by applying comprehensive gas chromatography coupled to time-of-flight mass spectrometry. We were able to identify several SVOCs characteristic for the protective cow sheds environment including four key components. Cell exposure with the two farming extracts showed a distinct regulation of the E3 ubiquitin ligase PELI2 gene and TLR2 by cow shed extracts. Finally, the regulation of TLR2 corresponded to the regulation that was observed after exposing cells to an artificial mixture of the four key components identified in the cow sheds. In summary, we were able to demonstrate the importance of smaller particle-bound SVOCs found in farming environments concerning their possible contribution to a protective farm effect.

We investigated concentrations of organic carbon (OC), elemental carbon (EC) and a wide range of particle-bound organic compounds in daily sampled PM2.5 at the remote Pha Din (PDI) – Global Atmosphere Watch (GAW) monitoring station in Northwestern Vietnam during an intense 3-week sampling campaign from 23rd March to 12th April 2015. The site is known to receive trans-regional air masses during large-scale biomass burning episodes, but OC composition studies are missing in the scientific literature. We quantified 51 organic compounds simultaneously by in-situ derivatization thermal desorption gas chromatography time-of-flight mass spectrometry (IDTD-GC-TOFMS). Anhydrosugars, methoxyphenols, n-alkanes, fatty acids, polycyclic aromatic hydrocarbons, oxygenated polycyclic aromatic hydrocarbons, nitrophenols as well as OC were used in a hierarchical cluster analysis highlighting distinctive patterns for periods under low, medium and high biomass burning (BB) influence. The highest particle phase concentration of the typical primary organic aerosol (POA) and possible secondary organic aerosol (SOA) constituents, especially nitrophenols, were found on 5th and 6th April. We linked the trace gases CH4, CO2, CO, O3 mixing ratios to the statistical classification of BB events based on OA composition and found increased CO and O3 levels during medium and high BB influence. Likewise, a backward trajectory analysis indicates different source regions for the identified periods based on the OA cluster, with cleaner air masses arriving from northeast, i.e., mainland China and Yellow sea. The more polluted periods are characterized by trajectories from southwest, with more continental recirculation of the medium cluster, and more westerly advection for the high cluster. These findings highlight that BB activities in Northern Southeast Asia cannot only significantly enhance the regional organic aerosol loading, but also affect the carbonaceous PM2.5 constituents and the trace gases in northwestern Vietnam. The presented analysis adds valuable data on the carbonaceous and in particular OC chemical composition of PM2.5 in a region of scarce data availability.

Residential wood combustion (RWC) emits large amounts of gaseous and particulate organic aerosol (OA). In the atmosphere, the emission is transformed via oxidative reactions, which are under daylight conditions driven mainly by hydroxyl radicals (OH). This continuing oxidative ageing produces secondary OA and may change the health- and climate-related properties of the emission. However, it is not well known how the composition of RWC-originated OA changes as the function of OH exposure. In this work, emissions from two modern residential logwood combustion appliances were photochemically aged in an oxidation flow reactor (OFR) with various OH exposure levels, reaching up to 6×1011 s cm−3 (equivalent to 1 week in the atmosphere). Gaseous organic compounds were analysed by proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS), while particulate OA was analysed online by a high-resolution soot particle aerosol mass spectrometer (SP-HR-ToF-AMS) and offline by in situ derivatization thermal desorption–gas chromatography–time-of-flight mass spectrometry (IDTD-GC-ToF-MS). Photochemical reactions increased the mass of particulate organic carbon by a factor of 1.3–3.9. The increase in mass took place during the first atmospheric equivalent day of ageing, after which the enhancement was independent of the extent of photochemical exposure. However, ageing increased the oxidation state of the particulate OA linearly throughout the assessed range, with ΔH:C/ΔO:C slopes between −0.17 and −0.49 in van Krevelen space. Ageing led to an increase in acidic fragmentation products in both phases, as measured by the IDTD-GC-ToF-MS for the particulate and PTR-ToF-MS for the gaseous phase. For the gaseous organic compounds, the formation of small carbonylic compounds combined with the rapid degradation of primary volatile organic compounds such as aromatic compounds led to a continuous increase in both the O : C and H : C ratios. Overall, the share of polycyclic aromatic compounds (PACs) in particles degraded rapidly during ageing, although some oxygen-substituted PACs, most notably naphthaldehydic acid, increased, in particular during relatively short exposures. Similarly, the concentrations of particulate nitrophenols rose extensively during the first atmospheric equivalent day. During continuous photochemical ageing, the dominant transformation mechanisms shifted from the initial gas-phase functionalization/condensation to the transformation of the particulate OA by further oxidation reactions and fragmentation. The observed continuous transformation of OA composition throughout a broad range of OH exposures indicates that the entire atmospheric lifetime of the emission needs to be explored to fully assess the potential climate and health effects of RWC emissions.

A proton-transfer-reaction time-of-flight mass spectrometer combined with the novel CHARON (“chemical analysis of aerosol online”) aerosol inlet was used for characterization of submicrometer particulate organic matter in ship engine exhaust. Particles were sampled from diluted and cooled exhaust of a marine test bench engine that was operated on residual heavy fuel oil (HFO) and low-sulfur distillate marine gas oil (MGO), respectively. In both fuel operation modes, exhaust particle mass spectra were dominated by polycycloalkanes in the C20-to-C39 range, which are typical main constituents of lubricating oils. Exhaust particle mass spectra were closely reproduced when the engine’s lubricant oil was directly measured in aerosolized form. We report emission profiles of lubricant oil hydrocarbons as a function of their volatility and as a function of their carbon atom number. The combustion of spruce logwood in a modern residential stove was found to emit polycyclic aromatic hydrocarbons (PAH) and oxygenated polycyclic aromatic hydrocarbons (OPAH) with emission factors of 404 μg MJ−1 of 35 analysed PAH, 317 μg MJ−1 of 11 analysed Oxy-PAH and 12.5 μg MJ−1 of 5 analysed OH-PAH, most of which are known as potential mutagens and carcinogens. Photochemical ageing in an oxidation flow reactor (OFR) degraded particle-bound PAH, which was also reflected in declining PAH toxicity equivalent (PAH-TEQ) values by 45 to 80% per equivalent day of photochemical ageing in the atmosphere. OPAH concentrations decreased less than PAH concentrations during photochemical ageing, supposedly due to their secondary formation, while 1-hydroxynaphthalene, 1,5-dihydroxynaphthalene and 1,8-naphthalaldehydic acid were significantly increased after ageing. Furthermore, secondary organic aerosol (SOA) formation and aromatic compounds not included in targeted analysis were investigated by thermal-optical carbon analysis (TOCA) hyphenate to resonance-enhanced multi-photon ionisation time-of-flight mass spectrometry (REMPI-TOFMS). The commonly used PAH-source indicators phenanthrene/anthracene, fluoranthene/pyrene, retene/chrysene, and indeno[cd]pyrene/benzo[ghi]perylene remained stable during photochemical ageing, enabling identification of wood combustion emissions in ambient air. On the other hand, benz[a]pyrene/benz[e]pyrene and benz[a]anthracene/chrysene were found to decrease with increasing photochemical age. Retene/chrysene was not a proper classifier for the wood combustion emissions of this study, possibly due to more efficient combustion than in open wood burning, from which this diagnostic ratio was initially derived. This study motivates in-depth investigation of degradation kinetics of particle-bound species on different combustion aerosol as well as the consequences of photochemical ageing on toxicity and identification of wood combustion emissions in ambient air.

According to the World Health Organization particulate emissions from the combustion of solid fuels caused more than 110,000 premature deaths worldwide in 2010. Log wood combustion is the most prevalent form of residential biomass heating in developed countries, but it is unknown how the type of wood logs used in furnaces influences the chemical composition of the particulate emissions and their toxicological potential. We burned logs of birch, beech and spruce, which are used commonly as firewood in Central and Northern Europe in a modern masonry heater, and compared them to the particulate emissions from an automated pellet boiler fired with softwood pellets. We determined the chemical composition (elements, ions, and carbonaceous compounds) of the particulate emissions with a diameter of less than 1 µm and tested their cytotoxicity, genotoxicity, inflammatory potential, and ability to induce oxidative stress in a human lung epithelial cell line. The chemical composition of the samples differed significantly, especially with regard to the carbonaceous and metal contents. Also the toxic effects in our tested endpoints varied considerably between each of the three log wood combustion samples, as well as between the log wood combustion samples and the pellet combustion sample. The difference in the toxicological potential of the samples in the various endpoints indicates the involvement of different pathways of toxicity depending on the chemical composition. All three emission samples from the log wood combustions were considerably more toxic in all endpoints than the emissions from the pellet combustion. © 2016 Wiley Periodicals, Inc. Ambient particulate matter (PM) is a leading global environmental health risk. Current air quality regulations are based on airborne mass concentration. However, PM from different sources have distinct chemical compositions and varied toxicity. Connections between emission control measures, air quality, PM composition, and toxicity remain insufficiently elucidated. The current study assessed the composition and toxicity of PM collected in Nanjing, China before, during, and after an air quality intervention for the 2014 Youth Olympic Games. A co-culture model that mimics the alveolar epithelium with the associated macrophages was created using A549 and THP-1 cells. These cells were exposed to size-segregated inhalable PM samples. The composition and toxicity of the PM samples were influenced by several factors including seasonal variation, emission sources, and the air quality intervention. For example, we observed a size-dependent shift in particle mass concentrations during the air quality intervention with an emphasized proportion of smaller particles (PM2.5) present in the air. The roles of industrial and fuel combustion and traffic emissions were magnified during the emission control period. Our analyses revealed that the PM samples demonstrated differential cytotoxic potencies at equal mass concentrations between sampling periods, locations, and time of day, influenced by variations in the predominant emission sources. Coal combustion and industrial emissions were the most important sources affecting the toxicological responses and displayed the least variation in emission contributions between the sampling periods. In conclusion, emission control mitigated cytotoxicity and oxidative stress for particles larger than 0.2 μm, but there was inadequate evidence to determine if it was the key factor reducing the harmful effects of PM0.2.

The impact of wood combustion on ambient aerosols was investigated in Augsburg, Germany during a winter measurement campaign of a six-week period. Special attention was paid to the high time resolution observations of wood combustion with different mass spectrometric methods. Here we present and compare the results from an Aerodyne aerosol mass spectrometer (AMS) and gas chromatographic – mass spectrometric (GC-MS) analysed PM1 filters on an hourly basis. This includes source apportionment of the AMS derived organic matter (OM) using positive matrix factorisation (PMF) and analysis of levoglucosan as wood combustion marker, respectively. During the measurement period nitrate and OM mass are the main contributors to the defined submicron particle mass of AMS and Aethalometer with 28% and 35%, respectively. Wood combustion organic aerosol (WCOA) contributes to OM with 23% on average and 27% in the evening and night time. Conclusively, wood combustion has a strong influence on the organic matter and overall aerosol composition. Levoglucosan accounts for 14% of WCOA mass with a higher percentage in comparison to other studies. The ratio between the mass of levoglucosan and organic carbon amounts to 0.06. This study is unique in that it provides a one-hour time resolution comparison between the wood combustion results of the AMS and the GC-MS analysed filter method at a PM1 particle size range. The comparison of the concentration variation with time of the PMF WCOA factor, levoglucosan estimated by the AMS data and the levoglucosan measured by GC-MS is highly correlated (R2 = 0.84), and a detailed discussion on the contributors to the wood combustion marker ion at mass-to-charge ratio 60 is given. At the end, both estimations, the WCOA factor and the levoglucosan concentration estimated by AMS data, allow to observe the variation with time of wood combustion emissions (gradient correlation with GC-MS levoglucosan of R2 = 0.84). In the case of WCOA, it provides the estimated magnitude of wood combustion emission. Quantitative estimation of the levoglucosan concentration from the AMS data is problematic due to its overestimation in comparison to the levoglucosan measured by the GC-MS.

Denuders are gas-particle partitioning tools and can be used in combination with filters and adsorbers for phase-separated collection of hazardous semi-volatile organic compound (SVOC) aerosols. Here, we investigated the suitability of carbonaceous monolithic multi-channel denuders for this approach. Particle transmission efficiency through the denuders was investigated using particles of polystyrene latex (PSL) and droplets of n-hexadecane and diethylene glycol. The time-dependent vapour collection efficiency was analysed for n-hexadecane and diethylene glycol vapours and also compared to calculated predictions. Our measurements showed an averaged transmission efficiency of 97 +/- 4.4% for PSL particles with diameters of 0.51, 0.99, 1.93, and 3.00 mu m. Measurements with one denuder and 1.08, 1.98, and 2.97 mu m particles consisting of n-hexadecane or diethylene glycol resulted in an averaged transmission efficiency of 99 +/- 6.5%. Regarding the vapour collection efficiency at a flow rate of 5 l min(-1), n-hexadecane vapour could be adsorbed to a similar extent (91 +/- 1.4% for one denuder, 98 +/- 0.3% for two denuders) as diethylene glycol vapour (93 +/- 1.8% for one denuder, 97 +/- 0.9% for two denuders). The comparison between experimental and theoretical vapour collection efficiencies revealed differences around 2.8% for n-hexadecane and around 12.3% for diethylene glycol.The results show that the tested denuders can be used as vapour collection tools for SVOCs, and can be integrated in currently used personal air samplers for separated vapour and particle collection.

The consumption of wood fuel is markedly increasing in developing and industrialized countries. Known side effects of wood smoke inhalation manifest in proinflammatory signaling, oxidative stress, DNA damage and hence increased cancer risk. In this study, the composition and acute biological impact of emissions of state-of-the-art wood combustion compliances: masonry heater (MH) and pellet boiler (PB) were investigated. Therefore A549 cells were exposed to emission aerosols in an automated air-liquid interface exposure station followed by cytotoxicity, transcriptome and proteome analyses. In parallel, aerosols were subjected to a chemical and physical haracterization. Compared to PB, the MH combustion at the same dilution ratio resulted in a 3-fold higher particle mass concentration (PM2.5) and deposited dose (PB: 27 $\pm$ 2 ng/cm2, MH; 73 $\pm$ 12 ng/cm2). Additionally, the MH aerosol displayed a substantially larger concentration of aldehydes, polycyclic aromatic hydrocarbons (PAH) or oxidized PAH. Gene ontology analysis of transcriptome of A549 cells exposed to MH emissions revealed the activation of proinflammatory response and key signaling cascades MAP kinase and JAK-STAT. Furthermore, CYP1A1, an essential enzyme in PAH metabolism, was induced. PB combustion aerosol activated the proinflammatory marker IL6 and different transport processes. The proteomics data uncovered induction of DNA damage-associated proteins in response to PB and DNA doublestrand break processing proteins in response to MH emissions. Taking together, the MH produces emissions with a higher particle dose and more toxic compounds while causing only mild biological responses. This finding points to a significant mitigating effect of antioxidative compounds in MH wood smoke.

Ambient inhalable particulate matter (PM) is a serious health concern worldwide, but especially so in China where high PM concentrations affect huge populations. Atmospheric processes and emission sources cause spatial and temporal variations in PM concentration and chemical composition, but their influence on the toxicological characteristics of PM are still inadequately understood. In this study, we report an extensive chemical and toxicological characterization of size-segregated urban air inhalable PM collected in August and October 2013 from Nanjing, and assess the effects of atmospheric processes and likely emission sources. A549 human alveolar epithelial cells were exposed to day- and nighttime PM samples (25, 75, 150, 200, 300 μg/ml) followed by analyses of cytotoxicity, genotoxicity, cell cycle, and inflammatory response. PM10–2.5 and PM0.2 caused the greatest toxicological responses for different endpoints, illustrating that particles with differing size and chemical composition activate distinct toxicological pathways in A549 cells. PM10–2.5 displayed the greatest oxidative stress and genotoxic responses; both were higher for the August samples compared with October. In contrast, PM0.2 and PM2.5–1.0 samples displayed high cytotoxicity and substantially disrupted cell cycle; August samples were more cytotoxic whereas October samples displayed higher cell cycle disruption. Several components associated with combustion, traffic, and industrial emissions displayed strong correlations with these toxicological responses. The lower responses for PM1.0–0.2 compared to PM0.2 and PM2.5–1.0 indicate diminished toxicological effects likely due to aerosol aging and lower proportion of fresh emission particles rich in highly reactive chemical components in the PM1.0–0.2 fraction. Different emission sources and atmospheric processes caused variations in the chemical composition and toxicological responses between PM fractions, sampling campaigns, and day and night. The results indicate different toxicological pathways for coarse-mode particles compared to the smaller particle fractions with typically higher content of combustion-derived components. to study the sources contributing to quasi-ultrafine particle (UFP) organic carbon and the spatial temporal variability of the sources.24h quasi-UFP (particulate matter0.36μm in this study) was sampled at a reference site continuously and at one of 5 other sites (T1, T2, T3, T4 and B1) in parallel in Augsburg, Germany from April 11th, 2014 to February 22nd, 2015, attempting to conduct 2-week campaigns at each site in 3 different seasons. Positive matrix factorization (PMF) was applied to measured organic tracers for source apportionment analyses. Pearson correlation coefficient r and coefficient of divergence (COD) were calculated to investigate spatial temporal variation of source contributions.5 sources were identified comprising biomass burning (BB), traffic emissions (Traffic), biogenic secondary organic aerosol (bioSOA), isoprene originated secondary organic aerosol (isoSOA) and biomass burning related secondary organic aerosol (bbSOA). In general, good temporal correlation and uniform distribution within the study area are found for bioSOA and bbSOA, probably resulting from regional formation/transport. Lower temporal correlation and spatial heterogeneity of isoSOA were found at the city background site with local influence from green space and less traffic impact. BB demonstrated very good temporal correlation, but higher contributions at sites influenced by local residential heating emissions were observed. Traffic showed the least seasonality and lower correlation over time among the sources. However, it demonstrated low spatial heterogeneity of absolute contribution, and only a few days of elevated contribution was found at T3 when wind came directly from the street nearby.temporal correlation and spatial variability of sources contributing to the organic fraction of quasi-UFP vary among sites and source types and show source-specific characteristics. Therefore, caution should be taken when using one monitor site measurement to assess human exposure in health effect studies of quasi-UFP.

Children are particularly susceptible to air pollution and schools are examples of urban microenvironments that can account for a large portion of children's exposure to airborne particles. Thus this paper aimed to determine the sources of primary airborne particles that children are exposed to at school by analyzing selected organic molecular markers at 11 urban schools in Brisbane, Australia. Positive matrix factorization analysis identified four sources at the schools: vehicle emissions, biomass burning, meat cooking and plant wax emissions accounting for 45%, 29%, 16% and 7%, of the organic carbon respectively. Biomass burning peaked in winter due to prescribed burning of bushland around Brisbane. Overall, the results indicated that both local (traffic) and regional (biomass burning) sources of primary organic aerosols influence the levels of ambient particles that children are exposed at the schools. These results have implications for potential control strategies for mitigating exposure at schools.

The presented wood combustion emission study employing a logwood stove showed that four burning phases of different aerosol compositions and different amounts of emitted particulate matter (PM) occurred during a combustion batch. As a novel approach, in this study, the burning phases were defined by chemical changes in the aerosol gas phase during combustion, instead of being linked to predefined time periods or the amount of PM emissions. This deeper view into the aerosol chemistry of the different burning phases was possible by employing online mass spectrometry techniques with high time resolution. The impact of combustion conditions on emission factors and characteristics of log wood combustion was investigated. Two different kinds of log woods (spruce and beech) and one kind of briquette (spruce sawdust) were used to study differences in emission behavior depending upon the wood type. Beech wood was used to examine additionally the impact of different moisture contents and maloperation on emissions of fine particulate matter (PM). Therefore, wood logs with three different levels of moisture content were used. Maloperation was simulated by an overload scenario and an air deficiency scenario. Toxicity equivalent (TEQ) values were calculated for the different combustion conditions. According to the World Health Organization particulate emissions from the combustion of solid fuels caused more than 110,000 premature deaths worldwide in 2010. Log wood combustion is the most prevalent form of residential biomass heating in developed countries, but it is unknown how the type of wood logs used in furnaces influences the chemical composition of the particulate emissions and their toxicological potential. We burned logs of birch, beech and spruce, which are used commonly as firewood in Central and Northern Europe in a modern masonry heater, and compared them to the particulate emissions from an automated pellet boiler fired with softwood pellets. We determined the chemical composition (elements, ions, and carbonaceous compounds) of the particulate emissions with a diameter of less than 1 µm and tested their cytotoxicity, genotoxicity, inflammatory potential, and ability to induce oxidative stress in a human lung epithelial cell line. The chemical composition of the samples differed significantly, especially with regard to the carbonaceous and metal contents. Also the toxic effects in our tested endpoints varied considerably between each of the three log wood combustion samples, as well as between the log wood combustion samples and the pellet combustion sample. The difference in the toxicological potential of the samples in the various endpoints indicates the involvement of different pathways of toxicity depending on the chemical composition. All three emission samples from the log wood combustions were considerably more toxic in all endpoints than the emissions from the pellet combustion. © 2016 Wiley Periodicals, Inc. Gaseous and particulate emissions from a ship diesel research engine were elaborately analysed by a large assembly of measurement techniques. Applied methods comprised of offline and online approaches, yielding averaged chemical and physical data as well as time-resolved trends of combustion by-products. The engine was driven by two different fuels, a commonly used heavy fuel oil (HFO) and a standardised diesel fuel (DF). It was operated in a standardised cycle with a duration of 2 h. Chemical characterisation of organic species and elements revealed higher concentrations as well as a larger number of detected compounds for HFO operation for both gas phase and particulate matter. A noteworthy exception was the concentration of elemental carbon, which was higher in DF exhaust aerosol. This may prove crucial for the assessment and interpretation of biological response and impact via the exposure of human lung cell cultures, which was carried out in parallel to this study. Offline and online data hinted at the fact that most organic species in the aerosol are transferred from the fuel as unburned material. This is especially distinctive at low power operation of HFO, where low volatility structures are converted to the particulate phase. The results of this study give rise to the conclusion that a mere switching to sulphur-free fuel is not sufficient as remediation measure to reduce health and environmental effects of ship emissions.

Open biomass burning from wildfires and the prescribed burning of forests and farmland is a frequent occurrence in South-East Queensland (SEQ), Australia. This work reports on data collected from 10 to 30 September 2011, which covers the days before (10–14 September), during (15–20 September) and after (21–30 September) a period of biomass burning in SEQ. The aim of this project was to comprehensively quantify the impact of the biomass burning on air quality in Brisbane, the capital city of Queensland. A multi-parameter field measurement campaign was conducted and ambient air quality data from 13 monitoring stations across SEQ were analysed. During the burning period, the average concentrations of all measured pollutants increased (from 20% to 430%) compared to the non-burning period (both before and after burning), except for total xylenes. The average concentration of O3, NO2, SO2, benzene, formaldehyde, PM10, PM2.5 and visibility-reducing particles reached their highest levels for the year, which were up to 10 times higher than annual average levels, while PM10, PM2.5 and SO2 concentrations exceeded the WHO 24-hour guidelines and O3 concentration exceeded the WHO maximum 8-hour average threshold during the burning period. Overall spatial variations showed that all measured pollutants, with the exception of O3, were closer to spatial homogeneity during the burning compared to the non-burning period. In addition to the above, elevated concentrations of three biomass burning organic tracers (levoglucosan, mannosan and galactosan), together with the amount of non-refractory organic particles (PM1) and the average value of f60 (attributed to levoglucosan), reinforce that elevated pollutant concentration levels were due to emissions from open biomass burning events, 70% of which were prescribed burning events. This study, which is the first and most comprehensive of its kind in Australia, provides quantitative evidence of the significant impact of open biomass burning events, especially prescribed burning, on urban air quality. The current results provide a solid platform for more detailed health and modelling investigations in the future. Direct thermal desorption and in-situ derivatization thermal desorption methods in conjunction with gas chromatography time-of-flight mass spectrometry have been characterized and evaluated for analysis of trace components from filters loaded with ambient particulate matter (PM). The limits of quantification were in the range of 7-24 pg for n-alkanes, 20 pg for hopanes, and 4-22 pg for polycyclic aromatic hydrocarbons (PAH). The limit of quantification was defined as the minimum amount of substance that conforms to the minimum distinguishable signal plus 9 times the standard deviation of this background signal from PM-loaded filters. The method has been successfully applied to low-volume samples from ambient PM collected with stationary and personal samplers. Stationary samples were collected in winter 2008 and 2010 in Augsburg, Germany. Sample aliquots of 0.2-0.3 m³ from stationary sampling were analyzed. High diurnal variation in concentration and source contribution was found especially during periods with low wind speed and low mixing layer height. High contributions of solid fuel combustion (wood and coal) were found in evening and nighttime samples, leading to peak PAH concentrations at midnight more than 10 times higher than at noon. Finally, the method was applied to samples collected by means of a personal sampler, i.e. a micro aethalometer, in Xi'an, China. Quantitative data on n-alkanes, hopanes, and PAH were obtained from sample volumes of 17 and 24 l. The impact of different sources such as vehicular and biogenic emissions could be distinguished.

Ambient PM10 concentration monitoring as well as dispersion calculations were conducted to determinethe influence of emissions from domestic heating on ambient PM10 concentrations in Augsburg,Germany. Based on the Augsburg emission inventory for domestic heating an average emission factor forparticulate emissions from the combustion of different solid fuels (wood logs, pellets, briquettes) indifferent types of stoves under various combustion conditions was found to be 120 mg MJ1 related toenergy input. Hence an emission model as well as a wind field model were created for dispersioncalculation of the emitted PM from wood combustion within Augsburg. The results of the dispersioncalculation concurred with the ambient PM10 monitoring data measured during the heating period 2007/2008. In this paper results of various measurement campaigns at different municipal waste incineration (MWI) plants concerning the change of the PCDD/PCDF isomer distribution in the crude gas during transiently impaired combustion conditions are presented. The focus is on the Cl4DD isomer distributions exemplarily for all other homologue groups to demonstrate the change in PCDD/PCDF formation mechanism at transient combustion conditions. Additionally to crude gas samples, at one plant filter and boiler ash were investigated simultaneously to determine if there is any difference in the isomer distribution between the matrices. For the ash from an electrostatic precipitator (ESP ash), the boiler ash and the corresponding crude gas sample, nearly identical changes in the Cl4DD isomer distribution under transient combustion conditions in relation to the normal operation process could be detected. By comparing the Cl4DD isomer distributions from different incineration plants (two municipal waste incinerators and one little incinerator burning wood chips for heating domestic household) under transient combustion conditions, in all cases the 1,3,6,8- and 1,3,7,9-Cl4DD were dominating the isomer distribution, whereas under normal operation other isomers were predominant. Obviously PCDD/PCDF formation mechanisms under transient combustion conditions are independent from the type of incinerator and of the burned fuel, respectively. Data sets were analyzed with respect to the possible reaction mechanism via chlorophenols and a good correlation of 2,4,6-trichlorophenol during the second phase of a start-up process and during a CO experiment was found. To get more detailed information about possible formation mechanisms, at one plant the dependence of the PCDD/PCDF isomer distribution on the different matrices was studied. Separate analysis of fly ash collected at the boiler exit, subsequent gas phase, ESP ash and boiler ash under normal operation conditions showed that, apart from the fly ash, the Cl4DD isomer distributions are nearly the same in the different matrices. Surprisingly, the Cl4DD isomer distribution of the fly ash was more similar to the distributions found under transient combustion conditions.

Indoor air pollution is associated with increased morbidity and mortality. Specifically, the health impact of emissions from domestic burning of biomass and coal is most relevant and is estimated to contribute to over 4 million premature deaths per year worldwide. Wood is the main fuel source for biomass combustion and the shift towards renewable energy sources will further increase emissions from wood combustion even in developed countries. However, little is known about the constituents of wood smoke and biological mechanisms that are responsible for adverse health effects. We exposed A549 lung epithelial cells to collected wood smoke particles and found an increase in cellular reactive oxygen species as well as a response to bioavailable polycyclic aromatic hydrocarbons. In contrast, cell vitality and regulation of the pro-inflammatory cytokine interleukin-8 were not affected. Using a candidate approach, we could recapitulate WSP toxicity by the combined actions of its constituents soot, metals and PAHs. The soot fraction and metals were found to be the most important factors for ROS formation, whereas the PAH response can be mimicked by the model PAH benzo[a]pyrene. Strikingly, PAHs adsorbed to WSPs were even more potent in activating target gene expression than B[a]P individually applied in suspension. As PAHs initiate multiple adverse outcome pathways and are prominent carcinogens, their role as key pollutants in wood smoke and its health effects warrants further investigation. The presented results suggest that each of the investigated constituents soot, metals and PAHs are major contributors to WSP toxicity. Mitigation strategies to prevent adverse health effects of wood combustion should therefore not only aim at reducing the emitted soot and PAHs but also the metal content, through the use of more efficient combustion appliances, and particle precipitation techniques, respectively.

In Europe, wood combustion in stoves and boilers is widely applied for residential heating. In Germany, approximately 15 million of 40 million households own small-scale furnaces, which deliver 7 % of Germany’s heat consumption. Using state-of-the-art small-scale combustion systems, we investigated how the air quality changes due to the emissions of harmful elements and organic pollutants during the combustion of wood and straw.

An in-situ derivatization thermal desorption method followed by gas chromatography and time-of-flight mass spectrometry (IDTD-GC-TOFMS) was developed for determination of polar organic compounds. Hydroxyl and carboxyl groups of compounds such as anhydrous sugars, alcohols and phenols, fatty acids and resin acids are targets of the derivatization procedure. Derivatization is based on silylation with N-Methyl-N-trimethylsilyl-trifluoroacetamide (MSTFA) during the step of thermal desorption. The high temperature of 300 °C during desorption is utilized for the in-situ derivatization on the collection substrate (quartz fibre filters) accelerating the reaction rate. Thereby, the analysis time is as short as without derivatization. At first the filter surface is dampened with derivatization reagent before insertion of the sample into the thermal desorption unit. To ensure ongoing derivatization during thermal desorption the carrier gas is saturated with MSTFA until the desorption procedure is finished. The method introduced here was compared with direct thermal desorption gas chromatography time-of-flight-mass spectrometry (DTD-GC-TOFMS) and with solvent extraction (SE) procedures followed by gas chromatography and mass spectrometry. Comparisons were carried out with field samples originating from ambient aerosol collected on quartz fibre filters. Moreover, the methods have been applied on NIST Standard Reference Material Urban Dust (SRM 1649a).

Urban aerosol was collected in a summer and a winter campaign for 7 and 3 days, respectively. Low volume samples were taken with a time resolution of 160 min using a filter/sorption cartridge system extended by an ozone scrubber. Concentrations of mainly particle associated polycyclic aromatic hydrocarbons (PAH) and oxidised PAH (O-PAH) were determined by gas chromatography/high resolution mass spectrometry. The sampling site was located in the city centre of Augsburg, Germany, near major roads with high traffic volume. The daily concentrations and profiles were mainly governed by local emissions from traffic and domestic heating, as well as by the meteorological conditions. During the winter campaign, concentrations were more than 10 fold higher than during the summer campaign. Highest concentrations were found concurrent with low boundary layer heights and low wind speeds. Significant diurnal variation of the PAH profiles was observed. Enhanced influences of traffic related PAH on the PAH profiles were evident during daytime in summer, whereas emissions from hot water generation and domestic heating were obvious during the night time of both seasons. A general idea about the global meteorological situation was acquired using back trajectory calculations (NOAA ARL HYSPLIT4). Due to high local emissions in combination with low air exchange during the two sampling campaigns, effects of mesoscale transport were not clearly observable.

The concentrations of some polycyclic aromatic hydrocarbons (PAH) and oxygenated PAH (O-PAH) can be changed by oxidation reactions during sampling. This can lead to an over- or underestimation of the corresponding adverse health effects. The aim of this study was the evaluation of these sampling artefacts. The potential of using an oxidant denuder was shown by parallel low-volume sampling with and without MnO2 ozone denuder. Twenty-three PAH and 11 O-PAH in ambient air were analysed, both in the vapour and particulate phase. The denuder was proven to be highly efficient for stripping ozone from air while causing no significant particle losses. In general, the concentrations of 5- to 7-ring PAH, which are predominantly associated with particles, were underestimated in non-denuded samples. The highest losses due to reaction with ozone and other atmospheric oxidants were observed for benzo[a]pyrene and perylene. Concurrently, the concentrations of most of the mainly particle-associated 4- to 5-ring O-PAH were higher in the non-denuded samples. The denuder did not only remove ozone, moreover other gaseous species such as more volatile PAH and O-PAH were partially oxidized on the catalytic surface, too. Degradation of PAH and concurrent degradation/formation reactions of O-PAH occurred. The corresponding reactivities of selected PAH and O-PAH are discussed.
