1. Ecotoxicological testing of sediments and dredged material: an overlooked opportunity?

Abstract

Purpose: Basing decisions for the management of contaminated sediments on ecotoxicological data is still often met with skepticism by European stakeholders. These concerns are discussed as they pertain to bioassays to show how ecotoxicological data may provide added value for the sustainable management of sediment in aquatic systems., Materials and methods: Five “concerns” are selected that are often raised by stakeholders. The ecotoxicological practice is discussed in light of the knowledge gained in recent decades and compared with chemical sediment analysis and chemical data., Results and discussion: Common assumptions such as a higher uncertainty of biotest results for sediments compared to chemical analyses are not supported by interlaboratory comparisons. Some confusion also arises, because the meaning of biotest data is often misunderstood, questioning their significance in light of a limited number of organisms and altered test conditions in the lab. Because biotest results describe a sediment property, they should not be directly equated with an impact upon the biological community. To identify a hazard, however, the possibility of false-negative results due to the presence of contaminants that are not analyzed but are toxic is lower., Conclusions: The cost of increased investment in ecotoxicological tests is, in our view, small compared with that of making false-negative assessments of sediment/dredged material that can ultimately have long-term environmental costs. Abstract

Background: the built environment in urban areas may have side effects on children's respiratory health, whilst less is known for adulthood.Aim: to assess the association between increasing exposure to grey spaces and allergic status in an adult general population sample.Methods: 2070 subjects (age range 15–84 yrs), living in Pisa/Cascina, Italy, were investigated in 1991–93 through a questionnaire on health status and risk factors, skin prick test (SPT), serum Immunoglobulins E (IgE), and serum antibodies to benzo(a)pyrene diol epoxide (BPDE)-DNA adducts. Land-cover exposure within a 1000 m buffer from each subject's home address was assessed through the CORINE Land Cover program (CLC 1990) within the FP7/HEALS project (2013–2018). Participants' residential addresses were geocoded and the proportion of surrounding grey spaces was calculated. Through logistic regression models, adjusting for potential confounding factors, the effect of a 10% increase in grey spaces exposure on allergic biomarkers/conditions was assessed; the relationship with serum antibodies to BPDE-DNA adducts positivity was also analyzed.Results: A 10% increase in grey spaces coverage was associated with a higher probability of having SPT positivity (OR 1.07, 95% CI 1.02-1.13), seasonal SPT positivity (OR 1.12, 1.05-1.19), polysensitization (OR 1.11, 1.04-1.19), allergic rhinitis (OR 1.10, 1.04-1.17), co-presence of SPT positivity and asthma/allergic rhinitis (OR 1.16, 1.08-1.25), asthma/allergic rhinitis (OR 1.06, 1.00-1.12), presence of serum antibodies to BPDE-DNA adducts positivity (OR 1.07, 1.01-1.14).Conclusions: grey spaces have adverse effects on allergic status and are related to a biomarker of polycyclic aromatic hydrocarbons exposure in adulthood. Thus, they may be used as a proxy of urban environmental exposure.

Abstract

International audience; Objective: Metals have been suggested as a risk factor for amyotrophic lateral sclerosis (ALS), but only retrospective studies are available to date. We compared metal levels in prospectively collected blood samples from ALS patients and controls, to explore whether metals are associated with ALS mortality. Methods: A nested ALS case–control study was conducted within the prospective EPIC (European Prospective Investigation into Cancer and Nutrition) cohort. Cases were identified through death certificates. We analyzed metal levels in erythrocyte samples obtained at recruitment, as a biomarker for metal exposure from any source. Arsenic, cadmium, copper, lead, manganese, mercury, selenium, and zinc concentrations were measured by inductively coupled plasma–mass spectrometry. To estimate ALS risk, we applied conditional logistic regression models. Results: The study population comprised 107 cases (65% female) and 319 controls matched for age, sex, and study center. Median time between blood collection and ALS death was 8 years (range = 1–15). Comparing the highest with the lowest tertile, cadmium (odds ratio [OR] = 2.04, 95% confidence interval [CI] = 1.08–3.87) and lead (OR = 1.89, 95% CI = 0.97–3.67) concentrations suggest associations with increased ALS risk. Zinc was associated with a decreased risk (OR = 0.50, 95% CI = 0.27–0.94). Associations for cadmium and lead remained when limiting analyses to noncurrent smokers. Interpretation: This is the first study to compare metal levels before disease onset, minimizing reverse causation. The observed associations suggest that cadmium, lead, and zinc may play a role in ALS etiology. Cadmium and lead possibly act as intermediates on the pathway from smoking to ALS. Abstract

International audience; The paper presents some recent trends in multi-fidelity digital modelling for marine engineering applications. Digital modelling is achieved by machine learning methods, namely multi-fidelity surrogate models, trained by computational fluid dynamics (CFD). Adaptative approaches are discussed for radial basis functions and Gaussian process models. Simulation-based design optimisation problems are presented to discuss the use and effects of different adaptivity concepts: (1) adaptive refinement of the computational-domain discretization in CFD; (2) adaptive sampling of the design/operational space; (3) adaptive selection of the fidelity used for the surrogate model training in a multi-fidelity environment; (4) adaptivity of the models to noise. Model adaptation allows for the efficient training of machine learning models, reducing the computational cost associated to building the training sets and improving the overall accuracy of the digital representation.

Abstract

Oligophrenin-1 (Ophn1) encodes a Rho GTPase activating protein whose mutations cause X-linked intellectual disability (XLID) in humans. Loss of function of Ophn1 leads to impairments in the maturation and function of excitatory and inhibitory synapses, causing deficits in synaptic structure, function and plasticity. Epilepsy is a frequent comorbidity in patients with Ophn1-dependent XLID, but the cellular bases of hyperexcitability are poorly understood. Here we report that male mice knock-out (KO) for Ophn1 display hippocampal epileptiform alterations, which are associated with changes in parvalbumin-, somatostatin- and neuropeptide Y-positive interneurons. Because loss of function of Ophn1 is related to enhanced activity of Rho-associated protein kinase (ROCK) and protein kinase A (PKA), we attempted to rescue Ophn1-dependent pathological phenotypes by treatment with the ROCK/PKA inhibitor fasudil. While acute administration of fasudil had no impact on seizure activity, seven weeks of treatment in adulthood were able to correct electrographic, neuroanatomical and synaptic alterations of Ophn1 deficient mice. These data demonstrate that hyperexcitability and the associated changes in GABAergic markers can be rescued at the adult stage in Ophn1-dependent XLID through ROCK/PKA inhibition.SIGNIFICANCE STATEMENTIn this study we demonstrate enhanced seizure propensity and impairments in hippocampal GABAergic circuitry in Ophn1 mouse model of X-linked intellectual disability (XLID). Importantly, the enhanced susceptibility to seizures, accompanied by an alteration of GABAergic markers were rescued by Rho-associated protein kinase (ROCK)/protein kinase A (PKA) inhibitor fasudil, a drug already tested on humans. Because seizures can significantly impact the quality of life of XLID patients, the present data suggest a potential therapeutic pathway to correct alterations in GABAergic networks and dampen pathological hyperexcitability in adults with XLID.

Since seizures can significantly impact the quality of life of XLID patients, the present data suggest a potential therapeutic pathway to correct alterations in GABAergic networks and dampen pathological hyperexcitability in adults with XLID.

No. of bitstreams: 0 Previous issue date: 2017-10-01 A strain of an achlorophyllic alga, named PR24T, was isolated from cow milk samples from the state of Minas Gerais, Brazil. Based on 18S rDNA, 28S rRNA, D1/D2 region of the LSU rDNA and SSU rRNA gene sequence similarities, this strain was found to be a member of the genus Prototheca and closely related to Prototheca blaschkeae SAG2064T. However, the novel strain could easily be distinguished from recognized Prototheca species by internal transcribed spacer, species-specific PCR, single-strand conformation polymorphism-PCR analysis and phenotypic characteristics. The inability to grow in Sabouraud broth at pH 4.0 and the different cellular fatty acid composition clearly distinguished PR24T from the reference strain of P. blaschkeae. The combination of genotypic and phenotypic data indicates that strain PR24T represents a subspecies of P. blaschkeae, for which the name Prototheca blaschkeae subsp. brasiliensis subsp. nov. is proposed. The respective type strain is PR24T (=DSM 103592T=IHEM 26958T). Abstract

International audience; Skeletal muscle regeneration depends on satellite cells. After injury these muscle stem cells exit quiescence, proliferate and differentiate to regenerate damaged fibres. We show that this progression is accompanied by metabolic changes leading to increased production of reactive oxygen species (ROS). Using Pitx2/3 single and double mutant mice that provide genetic models of deregulated redox states, we demonstrate that moderate overproduction of ROS results in premature differentiation of satellite cells while high levels lead to their senescence and regenerative failure. Using the ROS scavenger, N-Acetyl-Cysteine (NAC), in primary cultures we show that a physiological increase in ROS is required for satellite cells to exit the cell cycle and initiate differentiation through the redox activation of p38a MAP kinase. Subjecting cultured satellite cells to transient inhibition of P38a MAP kinase in conjunction with NAC treatment leads to their rapid expansion, with striking improvement of their regenerative potential in grafting experiments.

Abstract

International audience; Plant-based proteins are generally characterised by lower Indispensable Amino Acid (IAA) content, digestibility, and anabolic properties, compared to animal-based proteins. However, they are environmentally friendlier, and wider consumption is advocated. Older adults have higher dietary protein needs to prevent sarcopenia, a disease marked by an accelerated loss of muscle mass and function. Given the lower environmental footprint of plant-based proteins and the importance of optimising dietary protein quality among older adults, this paper aims to assess the net peripheral Amino Acid (AA) appearance after ingestion of three different plant protein and fibre (PPF) products, compared to whey protein with added fibre (WPF), in healthy older adults. In a randomised, single-blind, crossover design, nine healthy men and women aged ≥65 years consumed four test meals balanced in AA according to the FAO reference protein for humans, matched for leucine, to optimally stimulate muscle protein synthesis in older adults. A fasted blood sample was drawn at each visit before consuming the test meal, followed by postprandial arterialise blood sampling every 30 min for 3 h. The test meal was composed of a soup containing either WPF or PPF 1–3. The PPF blends comprised pea proteins with varying additional rice, pumpkin, soy, oat, and/or almond protein. PPF product ingestion resulted in a lower maximal increase of postprandial leucine concentration and the sum of branched-chain AA (BCAA) and IAA concentrations, compared to WPF, with no effect on their incremental area under the curve. Plasma methionine and cysteine, and to a lesser extent threonine, appearance were limited after consuming the PPF products, but not WPF. Despite equal leucine doses, the WPF induced greater postprandial insulin concentrations than the PPF products. In conclusion, the postprandial appearance of AA is highly dependent on the protein source in older adults, despite providing equivalent IAA levels and dietary fibre. Coupled with lower insulin concentrations, this could imply less anabolic potential. Further investigation is required to understand the applicability of plant-based proteins in healthy older adults.

Abstract

Megalencephalic Leukoencephalopathy with subcortical Cysts (MLC) is a type of vacuolating leukodystrophy, which is mainly caused by mutations in MLC1 or GLIALCAM. The two MLC-causing genes encode for membrane proteins of yet unknown function that have been linked to the regulation of different chloride channels such as the ClC-2 and VRAC. To gain insight into the role of MLC proteins, we have determined the brain GlialCAM interacting proteome. The proteome includes different transporters and ion channels known to be involved in the regulation of brain homeostasis, proteins related to adhesion or signaling as several G protein-coupled receptors (GPCRs), including the orphan GPRC5B and the proposed prosaposin receptor GPR37L1. Focusing on these two GPCRs, we could validate that they interact directly with MLC proteins. The inactivation of Gpr37l1 in mice upregulated MLC proteins without altering their localization. Conversely, a reduction of GPRC5B levels in primary astrocytes downregulated MLC proteins, leading to an impaired activation of ClC-2 and VRAC. The interaction between the GPCRs and MLC1 was dynamically regulated upon changes in the osmolarity or potassium concentration. We propose that GlialCAM and MLC1 associate with different integral membrane proteins modulating their functions and acting as a recruitment site for various signaling components as the GPCRs identified here. We hypothesized that the GlialCAM/MLC1 complex is working as an adhesion molecule coupled to a tetraspanin-like molecule performing regulatory effects through direct binding or influencing signal transduction events.

Abstract

The present study used steady-state visual evoked potentials (SSVEPs) to examine the spatio-temporal dynamics of reading morphologically complex words and test the neurophysiological activation pattern elicited by stems and suffixes. Three different types of target words were presented to proficient readers in a delayed naming task: truly suffixed words (e.g., farmer), pseudo-suffixed words (e.g., corner), and non-suffixed words (e.g., cashew). Embedded stems and affixes were flickered at two different frequencies (18.75 Hz and 12.50 Hz, respectively). The stem data revealed an earlier SSVEP peak in the truly suffixed and pseudo-suffixed conditions compared to the non-suffixed condition, thus providing evidence for the form-based activation of embedded stems during reading. The suffix data also showed a dissociation in the SSVEP response between suffixes and non-suffixes with an additional activation boost for truly suffixed words. The observed differences are discussed in the context of current models of complex word recognition.

Abstract

Local interactions and communication are key features in swarm robotics, but they are most often fixed at design time, limiting flexibility and causing a stiff and inefficient response to changing environments. Motivated by the need for higher adaptation abilities, we propose that information about emergent collective structures should percolate onto the individual behavior, modifying it in a way that determines suitable responses in the face of new working conditions and organizational challenges. Indeed, complex societies are driven by an evolving set of individual and social norms subject to cultural propagation, which contribute to determining the individual behaviors. We leverage ideas from the evolution of natural language – an undoubtedly efficient cultural trait – and exploit the resulting social dynamics to select and propagate microscopic behavioral parameters that adapt continuously to macroscopic conditions, which in turn affect the agents’ communication topography, and, therefore, feed back onto the social dynamics. This concept is demonstrated on a self-organized aggregation behavior, which is a building block for most swarm robotics behaviors and a striking example of how collective dynamics are sensitive to experimental parameters. By means of experiments with simulated and real robots, we show that the cultural evolution of aggregation rules outperforms conventional approaches in terms of adaptivity to multiple experimental settings.

Abstract

Current global change substantially threatens pollinators, which directly impacts the pollination services underpinning the stability, structure and functioning of ecosystems. Amongst these threats, many synergistic drivers, such as habitat destruction and fragmentation, increasing use of agrochemicals, decreasing resource diversity, as well as climate change, are known to affect wild and managed bees. Therefore, reliable indicators for pollinator sensitivity to such threats are needed. Biological traits, such as phenotype (e.g. shape, size and asymmetry) and storage reserves (e.g. fat body size), are important pollinator traits linked to reproductive success, immunity, resilience and foraging efficiency and, therefore, could serve as valuable markers of bee health and pollination service potential. This data paper contains an extensive dataset of wing morphology and fat body content for the European honeybee (Apis mellifera) and the buff-tailed bumblebee (Bombus terrestris) sampled at 128 sites across eight European countries in landscape gradients dominated by two major bee-pollinated crops (apple and oilseed rape), before and after focal crop bloom and potential pesticide exposure. The dataset also includes environmental metrics of each sampling site, namely landscape structure and pesticide use. The data offer the opportunity to test whether variation in the phenotype and fat bodies of bees is structured by environmental factors and drivers of global change. Overall, the dataset provides valuable information to identify which environmental threats predominantly contribute to the modification of these traits.

Abstract

Reactive astrocytes are astrocytes undergoing morphological, molecular, and functional remodeling in response to injury, disease, or infection of the CNS. Although this remodeling was first described over a century ago, uncertainties and controversies remain regarding the contribution of reactive astrocytes to CNS diseases, repair, and aging. It is also unclear whether fixed categories of reactive astrocytes exist and, if so, how to identify them. We point out the shortcomings of binary divisions of reactive astrocytes into good-vs-bad, neurotoxic-vs-neuroprotective or A1-vs-A2. We advocate, instead, that research on reactive astrocytes include assessment of multiple molecular and functional parameters—preferably in vivo—plus multivariate statistics and determination of impact on pathological hallmarks in relevant models. Abstract

The reproducibility of research using laboratory animals requires reliable management of their quality, in particular of their genetics, health and environment, all of which contribute to their phenotypes. The point at which these biological materials are transferred between researchers is particularly sensitive, as it may result in a loss of integrity of the animals and/or their documentation. Here, we describe the various aspects of laboratory animal quality that should be confirmed when sharing rodent research models. We also discuss how repositories of biological materials support the scientific community to ensure the continuity of the quality of laboratory animals. Both the concept of quality and the role of repositories themselves extend to all exchanges of biological materials and all networks that support the sharing of these reagents.

Abstract

Simple Summary We aimed to validate the correlation between tumour glycolysis/acidosis and inflammation in osteosarcoma-associated mesenchymal stromal cells and investigate the role of acidity-induced inflammation in the development of metastasis in this very aggressive cancer. We confirmed the presence of an acidic microenvironment in osteosarcoma xenografts, both subcutaneous and orthotopic, using state-of-the-art imaging technologies; corroborated the correlation between tumour glycolysis, acidosis, and inflammatory markers in human patients; and finally, explored the use of anti-IL6 antibody to target these pathogenic pathways, using advanced 3D microfluidic models. In the future, advanced imaging systems for the measurement of tumour glycolysis and/or pH may help identify osteosarcoma patients who would benefit from anti-IL6 therapies to complement conventional therapy. Abstract Osteosarcoma is the most frequent primary malignant bone tumour with an impressive tendency to metastasise. Highly proliferative tumour cells release a remarkable amount of protons into the extracellular space that activates the NF-kB inflammatory pathway in adjacent stromal cells. In this study, we further validated the correlation between tumour glycolysis/acidosis and its role in metastases. In patients, at diagnosis, we found high circulating levels of inflammatory mediators (IL6, IL8 and miR-136-5p-containing extracellular vesicles). IL6 serum levels significantly correlated with disease-free survival and 18F-FDG PET/CT uptake, an indirect measurement of tumour glycolysis and, hence, of acidosis. In vivo subcutaneous and orthotopic models, co-injected with mesenchymal stromal (MSC) and osteosarcoma cells, formed an acidic tumour microenvironment (mean pH 6.86, as assessed by in vivo MRI-CEST pH imaging). In these xenografts, we enlightened the expression of both IL6 and the NF-kB complex subunit in stromal cells infiltrating the tumour acidic area. The co-injection with MSC also significantly increased lung metastases. Finally, by using 3D microfluidic models, we directly showed the promotion of osteosarcoma invasiveness by acidosis via IL6 and MSC. In conclusion, osteosarcoma-associated MSC react to intratumoural acidosis by triggering an inflammatory response that, in turn, promotes tumour invasiveness at the primary site toward metastasis development.

We demonstrated the interaction (causes and consequences) of the hormone 17 α-ethinylestradiol (EE2) with lipid monolayers (prepared as Langmuir films) and bilayers prepared as small unilamellar vesicles (SUVs) and giant unilamellar vesicles (GUVs). Both fluidity and majority chemical composition of real plasma cell membrane were guaranteed using the phospholipid 1-palmitoil-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC). Surface pressure-mean molecular area (π-A) isotherms and PM-IRRAS measurements highlighted the strong interaction of EE2 with POPC monolayers, leading the hormone to remain at the air/water interface and promoting its penetration into the phospholipid hydrophobic chains. In the case of bilayers, the entrance of the hormone inside the SUV is likely facilitated by their high curvature. In GUVs, EE2 was responsible for changes in the spherical shape, forming structures like buds and lipid protrusions. The set of results indicates the strong effects of EE2 on fluid membranes, which is an important feature to predict its damage in human cells. In the euphotic zone, the concentration and quality of DOM are mostly dependent on primary production by phytoplankton. The effect of photodegradation on algal DOM has not been investigated as much as on terrestrial DOM. In this study, we explored the effect of different spectral regions (i.e., full sun spectrum, visible light, 295&ndash, 800 nm, 305&ndash, 800 nm, and 320&ndash, 800 nm) on algal exudates by Emiliania huxleyi, a ubiquitous coccolithophore. The optical properties (absorption and fluorescence) of algal DOM were investigated before and after irradiation with the different spectral regions. The absorption and fluorescence spectra were compared before and after irradiation. The results showed an increase in the effect of photobleaching with increasing irradiation energy for all of the absorbance indices. Similarly, the protein-like fluorescence decreased at increasing irradiation energy. The humic-like fluorescence, which was the most affected, did not show a linear trend between photobleaching and irradiation energy, which suggested that irradiation mainly determined a change in these molecules&rsquo, quantum yield.

Abstract

Being able to replicate real experiments with computational simulations is a unique opportunity to refine and validate models with experimental data and redesign the experiments based on simulations. However, since it is technically demanding to model all components of an experiment, traditional approaches to modeling reduce the experimental setups as much as possible. In this study, our goal is to replicate all the relevant features of an experiment on motor control and motor rehabilitation after stroke. To this aim, we propose an approach that allows continuous integration of new experimental data into a computational modeling framework. First, results show that we could reproduce experimental object displacement with high accuracy via the simulated embodiment in the virtual world by feeding a spinal cord model with experimental registration of the cortical activity. Second, by using computational models of multiple granularities, our preliminary results show the possibility of simulating several features of the brain after stroke, from the local alteration in neuronal activity to long-range connectivity remodeling. Finally, strategies are proposed to merge the two pipelines. We further suggest that additional models could be integrated into the framework thanks to the versatility of the proposed approach, thus allowing many researchers to achieve continuously improved experimental design.

Abstract

International audience; An adaptive-fidelity approach to metamodeling from noisy data is presented for design-space exploration and design optimization. Computational fluid dynamics (CFD) simulations with different numerical accuracy (spatial discretization) provides metamodel training sets affected by unavoidable numerical noise. The-fidelity approximation is built by an additive correction of a low-fidelity metamodel with metamodels of differences (errors) between higher-fidelity levels whose hierarchy needs to be provided. The approach encompasses two core metamodeling techniques, namely: i) stochastic radial-basis functions (SRBF) and ii) Gaussian process (GP). The adaptivity stems from the sequential training procedure and the auto-tuning capabilities of the metamodels. The method is demonstrated for an analytical test problem and a CFD-based optimization of a NACA airfoil, where the fidelity levels are defined by an adaptive grid refinement technique of a Reynolds-averaged Navier-Stokes (RANS) solver. The paper discusses: i) the effect of using more than two fidelity levels; ii) the use of least squares regression as opposed to exact interpolation; iii) the comparison between SRBF and GP; and iv) the use of two sampling approaches for GP. Results show that in presence of noise, the use of more than two fidelity levels improves the model accuracy with a significant reduction of the number of high-fidelity evaluations. Both least squares SRBF and GP provide promising results in dealing with noisy data.

Abstract

The identification of causal variants in sequencing studies remains a considerable challenge that can be partially addressed by new gene-specific knowledge. Here, we integrate measures of how essential a gene is to supporting life, as inferred from viability and phenotyping screens performed on knockout mice by the International Mouse Phenotyping Consortium and essentiality screens carried out on human cell lines. We propose a cross-species gene classification across the Full Spectrum of Intolerance to Loss-of-function (FUSIL) and demonstrate that genes in five mutually exclusive FUSIL categories have differing biological properties. Most notably, Mendelian disease genes, particularly those associated with developmental disorders, are highly overrepresented among genes non-essential for cell survival but required for organism development. After screening developmental disorder cases from three independent disease sequencing consortia, we identify potentially pathogenic variants in genes not previously associated with rare diseases. We therefore propose FUSIL as an efficient approach for disease gene discovery., Discovery of causal variants for monogenic disorders has been facilitated by whole exome and genome sequencing, but does not provide a diagnosis for all patients. Here, the authors propose a Full Spectrum of Intolerance to Loss-of-Function (FUSIL) categorization that integrates gene essentiality information to aid disease gene discovery.

Celiac disease (CD) affects a growing number of individuals worldwide. To elucidate the causes for this increase, future multidisciplinary collaboration is key to understanding the interactions between immunoreactive components in gluten-containing cereals and the human gastrointestinal tract and immune system and to devise strategies for CD prevention and treatment beyond the gluten-free diet. During the last meetings, the Working Group on Prolamin Analysis and Toxicity (Prolamin Working Group, PWG) discussed recent progress in the field together with key stakeholders from celiac disease societies, academia, industry and regulatory bodies. Based on the current state of knowledge, this perspective from the PWG members provides recommendations regarding clinical, analytical and legal aspects of CD. The selected key topics that require future multidisciplinary collaborative efforts in the clinical field are to collect robust data on the increasing prevalence of CD, to evaluate what is special about gluten-specific T cells, to study their kinetics and transcriptomics and to put some attention to the identification of the environmental agents that facilitate the breaking of tolerance to gluten. Abstract

International audience; Integrated forest management (IFM) can help reconcile critical trade-offs between goals in forest management, such as nature conservation and biomass production. The challenge of IFM is dealing with these trade-offs at the level of practical forest management, such as striving for compromises between biomass extraction and habitat retention. This paper reviews some of the driving factors that influence the integration of nature conservation into forest management. The review was conducted in three steps-a literature review, an expert workshop and an expert-based cooperative analysis. Of 38 driving factors identified, three were prioritised by more of the participants than any of the others: two are socio-cultural factors, identity (how people identify with forest) as well as outreach and education, and one is economic-competitiveness in forest value chains. These driving factors correspond to what are considered in the literature as enablers for IFM. The results reveal that targeted, group-oriented, adaptive and innovative policy designs are needed to integrate nature conservation into forest management. Further, the results reveal that a "one-size-fits-all" governance approach would be ineffective, implying that policy instruments need to consider contextually specific driving factors. Understanding the main driving factors and their overall directions can help to better manage trade-offs between biodiversity conservation and biomass production in European forests.

Abstract

International audience; Sovjan, a pile-dwelling settlement located in southeastern Albania with a stratigraphic sequence extending from the Early Bronze Age to the Early Iron Age attesting continuous habitation, serves as a starting point for a discussion on the applicability of Network Analysis (NA) in the southwestern Balkans. The excavations of several settlements in the Lakes Region divided between modern Albania, Greece, and North Macedonia, together with an extensive survey of the area around the former Lake Maliq where Sovjan is located, permit an analysis of connectivity conducted at different spatial (from micro- to macro-regional) and temporal levels. Important changes in connectivity patterns can be discerned on the basis of technological and stylistic changes observable in the ceramics and the different interregional influences at play in Sovjan. The Early Bronze Age, with its contact networks that extend from the Carpathian region to Greece, is separated from the Late Bronze Age by the hitherto poorly researched Middle Bronze Age. Following this interim period in which networks shrink and change, far-reaching networks arise once again demonstrating influences from the Danube region and, above all, central Macedonia.

Abstract

Este artículo contiene 18 páginas, 4 figuras., Good datasets of geo-referenced records of alien species are a prerequisite for assessing the spatio-temporal dynamics of biological invasions, their invasive potential, and the magnitude of their impacts. However, with the exception of first records on a country level or wider regions, observations of species presence tend to remain unpublished, buried in scattered repositories or in the personal databases of experts. Through an initiative to collect, harmonize and make such unpublished data for marine alien and cryptogenic species in the Mediterranean Sea available, a large dataset comprising 5376 records was created. It includes records of 239 alien or cryptogenic taxa (192 Animalia, 24 Plantae, 23 Chromista) from 19 countries surrounding the Mediterranean Sea. In terms of records, the most reported Phyla in descending order were Chordata, Mollusca, Chlorophyta, Arthropoda, and Rhodophyta. The most recorded species was Caulerpa cylindracea, followed by Siganus luridus, Magallana sp. (cf. gigas or angulata) and Pterois miles. Abstract

The primary objective of this study is to leverage the integration of surface mapping data derived from optical, radar, and historic topographical studies with archaeological sampling to identify ancient settlement areas in the Northern Nile Delta, Egypt. This study employed the following methods: digitization of topographic maps, band indices techniques on optical data, the creation of a 3D model from SRTM data, and Sentinel-1 interferometric wide swath (IW) analysis. This type of study is particularly relevant to the search for evidence of otherwise hidden ancient settlements. Due to its geographical situation and the fertility of the Nile, Egypt witnessed the autochthonous development of predynastic and dynastic civilizations, as well as an extensive history of external influences due to Greek, Roman, Coptic, Islamic, and Colonial-era interventions. Excavation work at Buto (Tell el-Fara’in) in 2017–18, carried out by the Kafrelsheikh University (KFS) in cooperation with the Ministry of Antiquities, demonstrated that remote sensing data offers considerable promise as a tool for developing regional settlement studies and excavation strategies. This study integrates the mission work in Buto with the satellite imagery in and around the area of the excavation. The results of the initial Buto area research serve as a methodological model to expand the study area to the North Delta with the goal of detecting the extent of the ancient kingdoms of Buto and Sakha. The results of this research include the creation of a composite historical database using ancient references and early topographical maps (1722, 1941, 1950, and 1997), Optical Corona (1965), Landsat MSS (Multispectral Scanner System) (1973, 1978, and 1988), TM (Thematic Mapper) (2005) data, and Radar SRTM (2014) and Sentinel1 (2018 and 2019) data. The data in this study have been analyzed using the ArcMap, Envi, and SNAP software. The results from the current investigation highlight the rapid changes in the land use/land cover in the last century in which many ancient sites were lost due to agriculture and urban development. Three potential settlement areas have been identified with the Sentinel1 Radar data, and have been integrated with the early maps. These discoveries will help develop excavation strategies aimed at elucidating the ancient settlement dynamics and history of the region during the next phase of research.

Abstract

The aim of metamodelling in CFD-based automatic shape optimization is to replace expensive CFD computations in the optimization process by evaluations of a cheap surrogate model, created from a limited training set of simulations. Multi-fidelity metamodels [1] make this process even more efficient by basing a part of the metamodel on inexpensive low-fidelity simulations and introducing a correction based on a few high-fidelity simulations. To adaptively define the training sets of the low-fidelity and correction metamodels, different sampling strategies can be used [2]. However, the CFD results on which the metamodels are based contain numerical errors. Especially if new grids are generated for each simulation, the numerical errors for two similar geometries may be different. These differences manifest themselves as numerical noise in the training sets for the metamodels. Adaptive sampling strategies which determine new training points by evaluating the uncertainty of the metamodel, may react to this noise by placing new points in noisy regions, rather than in places where the overall metamodel is unreliable. This deteriorates the metamodel quality. In this paper, we study multifidelity metamodeling approaches which eliminate a part of the numerical noise in the metamodel. For metamodels based on Dynamic Radial Basis Functions [3], this is obtained by introducing less RBF degrees of freedom than training points and computing the RBF fits using least-squares approximation. The number of DoF will be defined adaptively as the training progresses, based on a metric like leave-one-out cross-validation combined with in-the-loop optimization of the metamodel. Applications to the shape optimisation of 2D NACA four-digit airfoils and a destroyer-type vessel will be presented. The CFD simulations are based on a Reynolds-averaged Navier Stokes solver that uses adaptive grid refinement to create the meshes.

Abstract

Information transmission in the human brain is a fundamentally dynamic network process. In partial epilepsy, this process is perturbed and highly synchronous seizures originate in a local network, the so-called epileptogenic zone (EZ), before recruiting other close or distant brain regions. We studied patient-specific brain network models of 15 drug-resistant epilepsy patients with implanted stereotactic electroencephalography (SEEG) electrodes. Each personalized brain model was derived from structural data of magnetic resonance imaging (MRI) and diffusion tensor weighted imaging (DTI), comprising 88 nodes equipped with region specific neural mass models capable of demonstrating a range of epileptiform discharges. Each patient’s virtual brain was further personalized through the integration of the clinically hypothesized EZ. Subsequent simulations and connectivity modulations were performed and uncovered a finite repertoire of seizure propagation patterns. Across patients, we found that (i) patient-specific network connectivity is predictive for the subsequent seizure propagation pattern; (ii) seizure propagation is characterized by a systematic sequence of brain states; (iii) propagation can be controlled by an optimal intervention on the connectivity matrix; (iv) the degree of invasiveness can be significantly reduced via the proposed seizure control as compared to traditional resective surgery. To stop seizures, neurosurgeons typically resect the EZ completely. We showed that stability analysis of the network dynamics, employing structural and dynamical information, estimates reliably the spatiotemporal properties of seizure propagation. This suggests novel less invasive paradigms of surgical interventions to treat and manage partial epilepsy., Author summary Epilepsy is characterized by perturbed dynamics that originate in a local network before spreading to other brain regions. We studied patient-specific brain network models of epilepsy patients, comprising 88 nodes equipped with region specific neural mass models capable of demonstrating epileptiform discharges. Applying stability analysis led to a seizure control strategy that is significantly less invasive than the traditional surgery, which typically resects the epileptogenic regions. The invasiveness of the procedure correlates with graph theoretical importance of the nodes. The novel method subsequently removes the most unstable links, a procedure possible by advent of novel surgery techniques. Our approach is entirely based on structural data, allowing creation of a brain model based on purely non-invasive data prior to any surgery.

Abstract

Objectives: Type 2 diabetes mellitus (T2DM) and osteoarthritis (OA) are common diseases that frequently co-exist, along with overweight/obesity. While the mechanical impact of excess body weight on joints may explain lower limb OA, we sought to explore whether T2DM is linked to OA outside of excess weight and whether T2DM may play a role in OA pathophysiology. The consequence of T2DM on OA outcomes is a question of research interest. Methods: We conducted a critical review of the literature to explore the association between T2DM and OA, whether any association is site-specific for OA, and whether the presence of T2DM impacts on OA outcomes. We also reviewed the literature to assess the safety of anti-OA treatments in patients with T2DM. Results: T2DM has a pathogenic effect on OA through 2 major pathways involving oxidative stress and low-grade chronic inflammation resulting from chronic hyperglycemia and insulin resistance. T2DM is a risk factor for OA progression and has a negative impact on arthroplasty outcomes. Evidence is mounting for safety concerns with some of the most frequently prescribed anti-OA medications, including paracetamol, non-steroidal anti-inflammatory drugs, and corticosteroid injections, while other anti-OA medications may be safely prescribed in OA patients with T2DM, such as glucosamine and intra-articular hyaluronic acid. Conclusions: Future research is needed to better understand whether diabetes control and prevention can modulate OA occurrence and progression. The selection of therapy to treat OA symptoms in patients with T2DM may require careful consideration of the evidence based to avoid untoward safety issues., The meeting was funded by the ESCEO, a Belgian not-for-profit organization. Abstract

The European Academy of Allergy and Clinical Immunology (EAACI) organized the first European Strategic Forum on Allergic Diseases and Asthma. The main aim was to bring together all relevant stakeholders and decision-makers in the field of allergy, asthma and clinical Immunology around an open debate on contemporary challenges and potential solutions for the next decade. The Strategic Forum was an upscaling of the EAACI White Paper aiming to integrate the Academy's output with the perspective offered by EAACI's partners. This collaboration is fundamental for adapting and integrating allergy and asthma care into the context of real-world problems. The Strategic Forum on Allergic Diseases brought together all partners who have the drive and the influence to make positive change: national and international societies, patients’ organizations, regulatory bodies and industry representatives. An open debate with a special focus on drug development and biomedical engineering, big data and information technology and allergic diseases and asthma in the context of environmental health concluded that connecting science with the transformation of care and a joint agreement between all partners on priorities and needs are essential to ensure a better management of allergic diseases and asthma in the advent of precision medicine together with global access to innovative and affordable diagnostics and therapeutics. © 2019 EAACI and John Wiley and Sons A/S. Abstract

International audience; The paper presents how to efficiently and effectively solve stochastic shape optimization problems by combing Reynolds-averaged Navier-Stokes (RANS) equation solvers with design-space augmented dimensionality reduction (ADR). This study has been conducted within the NATO Science and Technology Organization, Applied Vehicle Technology, Task Group AVT-252 "Stochastic Design Optimization for Naval and Aero Military Vehicles." The application pertains to the robust and the reliability-based robust design optimization of a destroyer hull-form for resistance in calm water and waves and seakeeping performance, under stochastic environmental and operating conditions (speed, sea state, heading). The current work extends previous research by the authors, presented at earlier AIAA conferences [1-3], where only potential flow solvers were used. In the present work, the expected value of the total resistance is reduced respectively by 4.4 and 3% in calm water and waves. An 8% improvement of the seakeeping performance is also achieved. Design-space assessment by ADR is demonstrated to be a viable option in solving the curse of dimensionionality in shape optimization, especially when high-fidelity CPU-expensive solvers are used.
