6. Rapid assembly of oligosaccharides : reactivity tuning of glycosyl fluorides

Abstract

Chapter 1 comprises a general introduction to the field of chemical carbohydrate synthesis. The work since 1984 that has had a profound influence on the development of 1,2-diacetal mediated reactivity tuning is summarised. Chapter 2 examines the applicability of 1,2-diacetal mediated reactivity tuning to oligosaccharides assembly. In particular, the use of the butanediacetal protecting group (BDA) to tune the reactivity of glycosyl fluorides is disclosed. The application of this methodology enabled the first one-pot assembly of a linear pentasaccharide from monosaccharide building blocks. Chapter 3 focuses on synthetic advances towards the Glysosylphosphatidylinositol (GPI) anchor of yeast Saccharomcyes cerevisiae I and analogues of the conserved GPI core structure. The synthesis the full carbohydrate core structure of I was completed by application of this new methodology. In addition, the development of a flexible strategy towards I enabled the examination of parallel routes to GPI analogues.

8. Subcortical contributions to salience network functioning during negative emotional processing

Abstract

Core regions of the salience network (SN), including the anterior insula (aINS) and dorsal anterior cingulate cortex (dACC), coordinate rapid adaptive changes in attentional and autonomic processes in response to negative emotional events. In doing so, the SN incorporates bottom-up signals from subcortical brain regions, such as the amygdala and periaqueductal gray (PAG). However, the precise influence of these subcortical regions is not well understood. Using ultra-high field 7-Tesla functional magnetic resonance imaging, this study investigated the bottom-up interactions of the amygdala and PAG with the SN during negative emotional salience processing. Thirty-seven healthy participants completed an emotional oddball paradigm designed to elicit a salient negative emotional response via the presentation of random, task-irrelevant negative emotional images. Negative emotional processing was associated with prominent activation in the SN, spanning the amygdala, PAG, aINS, and dACC. Consistent with previous research, analysis using dynamic causal modelling revealed an excitatory influence from the amygdala to the aINS, dACC, and PAG. In contrast, the PAG showed an inhibitory influence on amygdala, aINS and dACC activity. Our findings suggest that the amygdala may amplify the processing of negative emotional stimuli in the SN to enable upstream access to attentional resources. In comparison, the inhibitory influence of the PAG possibly reflects its involvement in modulating sympathetic-parasympathetic autonomic arousal mediated by the SN. This PAG-mediated effect may be driven by amygdala input and facilitate bottom-up processing of negative emotional stimuli. Overall, our results show that the amygdala and PAG modulate divergent functions of the SN during negative emotional processing.

9. Differential engagement of the posterior cingulate cortex during cognitive restructuring of negative self- and social beliefs

Abstract

Negative self-beliefs are a core feature of psychopathology, encompassing both negative appraisals about oneself directly (i.e. self-judgment) and negative inferences of how the self is appraised by others (i.e. social judgment). Challenging maladaptive self-beliefs via cognitive restructuring is a core treatment mechanism of gold-standard psychotherapies. However, the neural mechanisms underlying the restructuring of these two kinds of negative self-beliefs are poorly understood. Eighty-six healthy participants cognitively restructured self-judgment and social-judgment negative self-belief statements during 7 Tesla functional magnetic resonance imaging scanning. Cognitive restructuring broadly elicited activation in the core default mode network (DMN), salience and frontoparietal control regions. Restructuring self-judgment relative to social-judgment beliefs was associated with comparatively higher activation in the ventral posterior cingulate cortex (PCC)/retrosplenial cortex, while challenging social-judgment statements was associated with higher activation in the dorsal PCC/precuneus. While both regions showed increased functional connectivity with the supplementary and pre-supplementary motor areas during restructuring, the dorsal PCC displayed greater task-dependent connectivity with distributed regions involved in salience, attention and social cognition. Our findings indicate distinct patterns of PCC engagement contingent upon self- and social domains, highlighting a specialized role of the dorsal PCC in supporting neural interactions between the DMN and frontoparietal/salience networks during cognitive restructuring.

Abstract

Although dementia research has been dominated by Alzheimer's disease (AD), most dementia in older people is now recognised to be due to mixed pathologies, usually combining vascular and AD brain pathology. Vascular cognitive impairment (VCI), which encompasses vascular dementia (VaD) is the second most common type of dementia. Models of VCI have been delayed by limited understanding of the underlying aetiology and pathogenesis. This review by a multidisciplinary, diverse (in terms of sex, geography and career stage), cross-institute team provides a perspective on limitations to current VCI models and recommendations for improving translation and reproducibility. We discuss reproducibility, clinical features of VCI and corresponding assessments in models, human pathology, bioinformatics approaches, and data sharing. We offer recommendations for future research, particularly focusing on small vessel disease as a main underpinning disorder.

11. The influence of electrolyte composition and current collector morphology on the properties of anodeless lithium metal batteries

Abstract

Among all the possibilities for next-generation rechargeable batteries, lithium metal is still craved as the Holy Grail of negative electrode materials, for the high theoretical specific capacity, the lowest redox potential, and the superior energy density with respect to currently used Li-ion batteries. Such energy density can be further increased by studying the anodeless configuration: the cell setup consists of a bare copper current collector where the lithium is plated directly from the cathode. Unfortunately, lithium plating/stripping on high-area surfaces displays a series of well-known problems: unstable SEI, dendrites formation that leads to cell short-circuit, and dead-lithium formation that causes severe capacity fading. In this respect, a lot of interesting strategies have been proposed; the ones adopted in this study are the electrolyte design and the employment of engineered current collectors. For this research, the effect of three different liquid electrolytes was studied on symmetrical Li|Li coin cells: commercial LP30, a Deep Eutectic Solvent and a Dual Salt electrolyte have been employed in the first part. while using the commercial LP30 and the DES the cell overpotential escalates as the cycles increase, with the DS the cyclic potential profile remains stable between ±0.02 V even after 200 cycles, confirming the validity of such electrolyte in the formation of a stable SEI. SEM measurements of the plated lithium surfaces have confirmed this stabilizing effect and its repercussions on the lithium morphology. In the second part, the DS performance has been measured in a non-symmetrical configuration to study the effect of the current collector: Cu|Li coin cells have been used for anodeless plating and stripping. As preliminary measurements, both the smooth and the rough sides of a plain Cu foil have been tested; the coins with rough Cu have displayed the most stable potential profiles and the highest Coulombic efficiencies. 16. Sex differences in the effect of subjective sleep on fear conditioning, extinction learning, and extinction recall in individuals with a range of PTSD symptom severity

Abstract

Sleep has been found to play a key role in fear conditioning, extinction learning and extinction recall, and sleep disturbances are linked to many mental disorders including post-traumatic stress disorder (PTSD). Previous studies examining associations between sleep and fear or extinction processes primarily focused on objectively measured sleep architecture. Little research has so far focused on subjective sleep measures and particularly in clinical populations, which often experience subjectively poor sleep, including PTSD. Here we investigated whether subjective sleep disturbance, sleep onset latency, wake after sleep onset or sleep efficiency were related to fear conditioning, extinction learning or extinction recall in a large sample of individuals with a range of PTSD symptom severity (n = 248). Overall, we did not find that subjective sleep was associated with fear conditioning or extinction processes. However, exploratory analyses examining the moderating effect of sex found that shorter sleep onset latency and greater sleep efficiency were associated with improved extinction recall in women with higher PTSD symptom severity. This suggests that less time falling asleep and longer time asleep while in bed may be protective in highly symptomatic women against the commonly observed impaired extinction recall in PTSD. More studies are needed to explore sex-specific effects further.

Abstract

El cáncer papilar de tiroides (CPT) tiene diferentes variantes y la mayoría de ellas presentan diferencias sutiles. La variante oncocítica (VO) es un subtipo poco frecuente de CPT, sobre el pronóstico de la cual existen resultados controvertidos en la literatura. Investigamos su agresividad y curso clínico comparándolos con la variante clásica (VC) y la variante de células altas (CA) de CPT en diferentes estadios. En este estudio de cohortes retrospectivo se incluyeron: 100 muestras simples de VO, 71 de CA y 1.219 de VC. Las muestras VO se compararon con las VC y las de CA sobre la base de parámetros de pronóstico independientes. La recurrencia de la VO también se comparó estadio por estadio con la VC y CA. La edad media fue de 46,8 años y la relación hombres/mujeres de 25/75 para la VO. Las tasas de recurrencia en nuestro estudio fueron del 16% en VO; del 13,5% en VC y del 56% en CA. Existe una diferencia estadísticamente significativa con respecto a la recurrencia entre el estadio 1 y el estadio 4 comparando la VO y la VC (p = 0,023; p = 0,03, respectivamente). También hay una diferencia estadísticamente significativa con respecto a la recurrencia entre el estadio 1 y el estadio 4 comparando la VO y la CA (p = 0,001; p = 0,024, respectivamente). Se puede suponer que la VO tiene un comportamiento entre la VC y la CA, pero muy cercana a la CA. La VO parece ser un poco más agresiva que la VC. A pesar de un tamaño de muestra inadecuado para los estadios 2 y 3, nuestros hallazgos implican un mayor riesgo de recurrencia para la VO que para la VC en los estadios avanzados (estadios 3 y 4) y la VC tiene un pronóstico más desfavorable que VO en estadios precoces (estadios 1 y 2), según el modelo de estadio pareado. Papillary thyroid cancer (PTC) has many variants and most of them are mild tumors. Oncocytic variant (OV) is a rare subtype of PTC. There are controversial results about its prognosis in the literature. We investigated its aggressivity and clinical course by comparing it with classical variant (CV) and tall cell variant (TV) of PTC over a stage-matched design. Pure 100 OV, 71 TV and 1,219 CV were included in this retrospective cohort study. OV was compared with CV and TV according to independent prognostic parameters. OV was also compared stage by stage with CV and TV for recurrence. Mean age was 46,8 years and male/female ratio 25/75 for OV. The recurrence rates in our study were 16% in OV, 13,5% in CV and 56% in TV. There is a statistically significant difference according to recurrence between stage 1 and stage 4 OV and CV (P = 0.023, P = 0.03, respectively). There is also a statistically significant difference between stage 1 and stage 4 OV and TV according to recurrence (P = 0.001, P = 0.024, respectively). OV can be supposed to behave between CV and TV, but very closer to CV. OV seems to be slightly more aggressive than CV. Despite an inadequate sample size for stage 2 and 3, our findings imply an increased recurrence risk for OV than CV at the advanced stages (stage 3 and 4) and CV has an unfavorable prognosis than OV at early stages (stage 1 and 2) according to stage-matched model. Ovarian ischemia-reperfusion (I/R) injury develops due to oxidative stress and inflammation that occurs when tissue is deprived of oxygen. Ovarian I/R damage also causes infertility. The aim of this study is to see the effect of ovarian I/R damage on reproductive functions and to determine the protection of antioxidant and anti-inflammatory cerebrolysin in possible damage. Albino Wistar-type female rats were divided into sham operation (SG), ischemia-reperfusion (IR) and cerebrolysin+ischemia-reperfusion (CIR) groups. After I/R application with cerebrolysin, the ovaries of six animals from each group were taken and biochemical and histopathological examinations were performed. Six animals from each remaining group were left to breed. Our test results showed that cerebrolysin decreased I/R-related oxidant, proinflammatory cytokine and tissue damage and increased antioxidant levels. In addition, cerebrolysis significantly inhibited the delay of the maternity period. These findings indicate that cerebrolysin may be helpful in the treatment of I/R-related ovarian damage and infertility.

20. THE IN SITU POTENTIAL OF SYNTHETIC NANO-HYDROXYAPATITE FOR TOOTH ENAMEL REPAIR

Abstract

This study was designed to evaluate whether nano-hydroxyapatite toothpastes with or without fluoride would be more advantageous than a fluoride toothpaste in the repair of eroded enamel in situ. Twenty-one subjects participated in this single-blind, randomized, cross-over design study with three 7-day treatment phases. In each phase, the subjects wearing a palatal appliance containing five sterilized enamel specimens used either one of the two test regimens (1% nano-hydroxyapatite toothpaste and 2.25% nano-hydroxyapatite/1450 parts per million (ppm) fluoride toothpaste) or one control (1400 ppm fluoride toothpaste). Enamel specimens were extraorally demineralized (4 × 5 min/day) and were intraorally treated with the toothpastes (2 × 2 min/day). The nano-hydroxyapatite toothpaste groups exhibited significantly higher surface microhardness than did the standard fluoride toothpaste group (p < 0.05). Enamel surface hardness was increased only by nano-hydroxyapatite toothpastes after in situ treatment compared with the baseline (p < 0.05). Morphological analysis demonstrated an apatite-type crystal deposition on the eroded enamel surface produced by nano-hydroxyapatite toothpastes, while fluoride toothpaste failed to show any significant surface deposition. Chemical analysis showed a higher content of calcium and phosphorus in the enamel surface treated with nano-hydroxyapatite toothpastes compared with that in the control one (p < 0.05). It is concluded that home use of nano-hydroxyapatite containing toothpastes may have a protective effect against erosion at the enamel surface.

28. A comparative study of song in European thrushes

Abstract

In this thesis several aspects of the singing behaviour of six European members of the genus Turdus have been studied. It has been-demonstrated that these six species show three distinct types of singing behaviour. They are; 1. Species with small repertoire size and stereotyped song characteristics (ring ousel and redwing). It is suggested that song in these species functions primarily as a territorial 2. Species advertisement. with medium to large repertoire size and stereotyped song characteristics (blackbird and mistle thrush). It is suggested and observations support the contention that one important function for song in these species is territorial. The large repertoire characteristic of individuals of these species indicates that there may also be an important mate attraction component. 3. Species with large to extremely large repertoire size and variable song characteristics (song thrush and, possibly, fieldfare but results are based on measurements from only one recording of one individual). Observations indicate that the song of the song thrush probably functions primarily in mate attraction and it is suggested that song may play a similar role in the fieldfare. It is suggested that in other avian species in which individuals possess large repertoire sizes and sing in a complex or unpredictable manner that song is likely to function primarily in mate attraction. A series of experiments was conducted on individuals of the mistle thrush, blackbird and song thrush. These revealed that individuals of these species use a second kind of singing behaviour in situations simulating territorial intrusion. (Jommon to all species is an increase in sound frequency of song during or following playback of conspecific song. The singing behaviour of three individual song thrushes was studied in an attempt to find rules governing sequencing of song types. The most important point to emerge is that individuals of this species seem to organise their singing behaviour in a hierarchial manner. It is possible to identify groups of song types within which transitions occur more frequently than at random. This is like the singing behaviour of the blackbird and it may be that hierarchical organisation of singing behaviour is a common phenomenon in species in which individuals possess large song repertoires. The functional significance of such behaviour is unclear. Finally, with a view to confirming or refuting Hartshorne's anti-monotony threshold principle, the singing behaviour of the six European species and that of eight others from various faunal regions was studied to see to what extent it conforms with predictions of the principle. Several measures of song continuity and versatility were i applied and it was found that for macro measures (see Chapter 4) there is a very significant relationship between the two parameters. 'Other predictions of Hartshorne were not confirmed. It is suggested that although the principle seems to apply to some aspects of Turdus song its major failing is that it does not provide an explanation for interspecific differences in repertoire size.

This study aims to optimize the heating length in terms of the sensory properties of sweet soy sauce made from coconut water. This experiment used a Complete Randomized Design consisting of 7 treatment levels, with each combination of treatments repeated 3 times so that the total was 21 treatments. It was concluded that the length of heating affects the color, taste, and viscosity of sweet soy sauce. Panelists favored the 160-minute warm-up time in terms of color, taste, and viscosity. The color favorability score was 5.64 (like), taste was 4.87 (somewhat like), viscosity was 5.61 (like), and aroma was 5.27 (somewhat like). Abstract

PubMed ID: 26184899 The aim of the present study was to investigate the possible protective effects of boron, an antioxidant agent, against arsenic-induced oxidative stress in male and female rats. In total, 42 Wistar albino male and female rats were divided into three equal groups: The animals in the control group were given normal drinking water, the second group was given drinking water with 100 mg/L arsenic, and the third group was orally administered drinking water with 100 mg/kg boron together with arsenic. At the end of the 28-day experiment, arsenic increased lipid peroxidation and damage in the tissues of rats. However, boron treatment reversed this arsenic-induced lipid peroxidation and activities of antioxidant enzymes in rats. Moreover, boron exhibited a protective action against arsenic-induced histopathological changes in the tissues of rats. In conclusion, boron was found to be effective in protecting rats against arsenic-induced lipid peroxidation by enhancing antioxidant defense mechanisms. © 2015 Wiley Periodicals, Inc.

Abstract

In this study, Salmonella Enteritidis strains isolated from dust and environmental materials from different flocks located in Turkey’s Western Black Sea region were examined by pulsed field gel electrophoresis (PFGE) and multilocus sequence typing (MLST). A total of 59 S. Enteritidis strains isolated from broiler breeder and hatchery flocks, and one S. Enteritidis strain isolated from a stool sample of a farm worker were examined. PFGE analysis revealed two major PFGE groups and nine different macro restriction profiles. It was determined that 85% (51/60) of the strains were close to each other and comprised Group I. All S. Enteritidis strains had the same sequence type (ST): ST11. Isolation of strains with a single genotype suggests that there may be a cross transmission between the flocks.

Abstract

Background: There is an increasing trend for administration of invasive radiological interventions, laparoscopic surgery, and transplantation procedures in recent years, and determining the vascular variations prior to these procedures is crucially important. Coeliacomesenteric trunk (CMT) is among these variations. This study aimed to retrospectively evaluate this rare anomaly by computed tomography (CT).Materials and Methods: A total of 1000 CT angiography images were analysed retrospectively, and the patients with mesenteric and coeliac arteries arising from the abdominal aorta with a single root were identified. The level that CMT arose, and its branching patterns were determined individually for all patients.Results: Ten patients (6 males and 4 females) with a mean age of 50.2 years (17-87 years) had CMT in CT images.Conclusions: The knowledge of variations in the CMT prior to vascular or laparoscopic interventions will contribute to early intervention in case of a complication, or to avoid from a potential damage. Abstract

Resumen Objetivo Los linfomas del sistema nervioso central (SNC) son linfomas no Hodgkin extranodales, agresivos y fatales, albergados en el SNC en el diagnóstico inicial. Su pronóstico es malo, teniendo la enfermedad un resultado fatal en comparación con el linfoma no Hodgkin sistémico. Se han indicado una serie de sistemas de estratificación del riesgo para estimar el pronóstico, basados principalmente en el nivel sérico de lactato deshidrogenasa, la edad, el índice de Karnofsky, el compromiso de las estructuras profundas del cerebro y la concentración proteica del líquido cefalorraquídeo. La PET/TC con 18 F-FDG tiene un elevado valor pronóstico en cuanto a la supervivencia global y la supervivencia libre de enfermedad en muchos cánceres y linfomas. Nuestro objetivo fue investigar los índices tumorales metabólicos mediante 18 F-FDG PET/TC para estadificación primaria, como marcadores pronósticos de los linfomas primarios del SNC. Material y métodos Se incluyó en este estudio retrospectivo a una cohorte de 14 pacientes con linfoma primario de células B grandes del SNC (estadio i ). Se realizó una PET/TC con 18 F-FDG de estadificación primaria, calculándose en todos los pacientes, y con anterioridad al tratamiento, los parámetros cuantitativos tales como el valor estandarizado de captación máximo, el valor de captación medio, el volumen tumoral metabólico y la glucólisis tumoral total (TLG). Se realizaron modelos de regresión de Cox para determinar su relación con el tiempo de supervivencia. Resultados En la evaluación de todos los factores de riesgo potencial con impacto sobre la recidiva/metástasis (edad, sexo, lactato deshidrogenasa sérico, compromiso de las estructuras profundas del cerebro, el valor estandarizado de captación máximo, el valor de captación medio, el volumen tumoral metabólico y la TLG), realizada mediante análisis univariante, el valor de TLG reflejó una significación estadística (p = 0,02). Conclusión Los parámetros tumorales metabólicos resultan de utilidad para la estimación pronóstica de los linfomas primarios del SNC. En especial, la TLG constituye el parámetro más importante, y puede desempeñar un papel en el tratamiento del paciente. Objective Primary central nervous system (CNS) lymphoma is an aggressive and fatal extranodal non-Hodgkin lymphoma jailed in CNS at initial diagnosis. Its prognosis is poor and the disease has a fatal outcome when compared with systemic non-Hodgkin lymphoma. A few baseline risk stratification scoring systems have been suggested to estimate the prognosis mainly based on serum lactate dehydrogenase level,age, Karnofsky performance score, involvement of deep brain structures and cerebrospinal fluid protein concentration. 18 F-FDG PET/CT has a high prognostic value with respect to overall survival and disease-free survival in many cancers and lymphomas. We aimed to investigate metabolic tumor indexes on primary staging 18 F-FDG PET/CT as prognostic markers in primary CNS lymphoma. Material and methods Fourteen patients with primary CNS diffuse large B-cell lymphoma (stage i ) were enrolled in this retrospective cohort study. Primary staging 18 F-FDG PET/CT was performed and quantitative parameters like maximum standardized uptake value, average standardized uptake value, metabolic tumor volume and total lesion glycolysis (TLG) were calculated for all patients before the treatment. Cox regression models were performed to determine their relation with survival time. Results In the evaluation of all potential risk factors impacting recurrence/metastases (age, sex, serum lactate dehydrogenase, involvement of deep brain structures, maximum standardized uptake value, average standardized uptake value, metabolic tumor volume, and TLG) with univariate analysis, TLG remained statistically significant ( P = .02). Conclusion Metabolic tumor parameters are useful in prognosis estimation of primary CNS lymphomas, especially TLG, which is the most important one and may play a role in patient management.
