Abstract: Following a radiological or nuclear emergency, medical intervention requires rapid assessment of the exposure of people usually through determination of internal dose. For the plutonium urine bioassay, besides thermal ionization mass spectrometry (TIMS) and alpha spectrometry methods, inductively coupled plasma mass spectrometry (ICP-MS) methods have been recently developed, which can provide much higher sample throughput. In this work, three ICP-MS methods were compared with one TIMS method and two alpha spectrometry methods for the measurement of 239Pu and 240Pu in human urine samples spiked at different concentration levels. The sample throughputs for all three ICP-MS methods are similar: each instrument measures about 80 samples in the first 24 hours and 200 samples in the first 48 hours following the emergency event, if the samples arrive at the laboratory 8 hours after the event occurs. Method accuracy and precision were determined using ANSI N13.30. Method detection limits and minimum detectable amounts (MDA) were determined to evaluate method sensitivities. The sensitivities of the three ICP-MS methods were also compared with the derived urine action level (24 h urine, 500 mSv committed effective dose equivalent, inhalation exposure, maximum dose conversion factor) to evaluate their applicability to exposure situations. Abstract: Non-invasive multi-scale and multimodal 3D characterization of heterogeneous or hierarchically structured intact mesoscale samples is of paramount importance in tackling challenging scientific problems. Scanning hard X-ray tomography techniques providing simultaneous complementary 3D information are ideally suited to such studies. However, the implementation of a robust on-site workflow remains the bottleneck for the widespread application of these powerful multimodal tomography methods. In this paper, we describe the development and implementation of such a robust, holistic workflow, including semi-automatic data reconstruction. Due to its flexibility, our approach is especially well suited for on-the-fly tuning of the experiments to study features of interest progressively at different length scales. To demonstrate the performance of the method, we studied, across multiple length scales, the elemental abundances and morphology of two complex biological systems, Arabidopsis plant seeds and mouse renal papilla samples. The proposed approach opens the way towards routine multimodal 3D characterization of intact samples by providing relevant information from pertinent sample regions in a wide range of scientific fields such as biology, geology, and material sciences.

Abstract: The spatial distribution and seasonal variations of atmospheric iodine (I), selenium (Se) and caesium (Cs) depositions remain unclear and this precludes adequate inputs for biogeochemical models. We quantified total concentrations and fluxes of these elements in rainfalls from 27 monitoring sites in France with contrasted climatic conditions; monthly measurements were taken over one year (starting in 2016/09). Since speciation of I and Se can impact their behaviour in the environment, analysis of their inorganic compounds was also conducted. Our results showed that annual I concentrations in rainfall were much higher than those of Se and Cs (annual means = 1.56, 0.044 and 0.005 μg L-1, respectively). The annual iodine concentrations were highly positively correlated with those of marine elements (i.e. Na, Cl and Mg), involving higher I concentrations under oceanic climate than for transition, continental and mountainous ones. Furthermore, common patterns were found between Se concentrations and both marine and terrestrial components consistent with the various sources of Se in atmosphere. The association of Cs with two anthropogenic components (i.e. NH4+ and NO3-) used in agriculture supports the hypothesis of its terrestrial origin (i.e. from atmospheric dusts) in rainfall. We found higher rainfall concentrations of I during the warmest months for all climates. However, no specific seasonal trend occurred for Se and Cs. On annual average, rainfall contained mostly unidentified selenium compounds (inorganic Se proportions = 25-54%) and equal proportions of inorganic and unidentified I compounds. Concentrations of iodate were higher under oceanic climate consistent with an iodine marine-origin. Abstract: International audience; Current multiagent chemotherapy regimens have improved the cure rate in acute leukemia patients, but they are highly toxic and poorly efficient in relapsed patients. To improve the treatment approaches, new specific molecules are needed. The G-quadruplexes (G4s), which are noncanonical nucleic acid structures found in specific guanine-rich DNA or RNA, are involved in many cellular events, including control of gene expression. G4s are considered as targets for the development of anticancer agents. Heterocyclic molecules are well known to target and stabilize G4 structures. Thus, a new series of 2,9-bis[(substituted-aminomethyl)phenyl]-1,10-phenanthroline derivatives (1a-i) was designed, synthesized, and evaluated against five human myeloid leukemia cell lines (K562, KU812, MV4-11, HL60, and U937). Their ability to stabilize various oncogene promoter G4 structures (c-MYC, BCL-2, and K-RAS) as well as the telomeric G4 was also determined through the fluorescence resonance energy transfer melting assay and native mass spectrometry. In addition, the more bioactive ligands 1g-i were tested for telomerase activity in HuT78 and MV4-11 protein extracts.

Abstract: Sur l’échantillon total de brochets adultes analysés (n=40, dont 17 individus prélevés en 2018-2019), 25% (IC95% 14.2-40.2%) des individus proviennent de la pisciculture et 75% (IC95% 59.8-85.8%) de la reproduction naturelle. Bien qu’une incertitude demeure en raison de la taille de l’échantillon, la cohérence des récents résultats avec les analyses réalisées en 2015-2016 (Richard et al., 2018) confirme la robustesse de ces proportions et la part majoritaire de la reproduction naturelle dans le stock de brochet du lac de Serre-Ponçon. Ce résultat questionne sur les sites et les périodes de reproduction dans le lac de Serre-Ponçon au vu de l’important marnage saisonnier.

Abstract: International audience; Nano-oxides and hydroxides generate great interest as promising positive electrode materials for the development of high energy density supercapacitors. However, their usually limited ionic and electronic conductivities significantly decrease their energy storage performances when increasing the electrode's mass loading. Here, we report on a sonochemical approach to functionalize the surface of Co(OH)2 nanomaterials by EmimBF4 ionic liquid that greatly improves the stability and the electrochemical performances of high mass loading electrodes (13 mg cm−2). This surface functionalization boosts the transport properties and strongly enhances the capacity as well as the capacity retention at higher current densities compared to basic Co(OH)2 (e.g. 113.5 C g−1 vs 59.2 C g−1 at 1 A g−1). Additionally, the protective layer formed by the ionic liquid stabilizes the electrode material upon cycling in KOH aqueous electrolyte and protects the material from oxidation upon open-air storage.

Abstract: Microorganisms are key players in the transformation of mercury into neurotoxic methylmercury (MeHg). Nevertheless, this mechanism and the opposite MeHg demethylation remain poorly understood. Here, we explored the impact of inorganic mercury (IHg) and MeHg concentrations from 0.05 to 50 mu M on the production and degradation of MeHg in two sulfate-reducing bacteria, Pseudodesulfovibrio hydrargyri BerOc1 able to methylate and demethylate mercury and Desulfovibrio desulfuricans G200 only able to demethylate MeHg. MeHg produced by BerOc1 increased with increasing IHg concentration with a maximum attained for 5 mu M, and suggested a saturation of the process. MeHg was mainly found in the supernatant suggesting its export from the cell. Hg L-3-edge High- Energy-Resolution-Fluorescence-Detected-X-ray-Absorption-Near-Edge-Structure spectroscopy (HERFD-XANES) identified MeHg produced by BerOc1 as MeHg-cysteine(2) form. A dominant tetracoordinated beta HgS form was detected for BerOc1 exposed to the lowest IHg concentrations where methylation was detected. In contrast, at the highest exposure (50 mu M) where Hg methylation was abolished, Hg species drastically changed suggesting a role of Hg speciation in the production of MeHg. The tetracoordinated beta HgS was likely present as nano-particles as suggested by transmission electron microscopy combined to X-ray energy dispersive spectroscopy (TEM-X-EDS) and nano-X ray fluorescence (nano-XRF). When exposed to MeHg, the production of IHg, on the contrary, increased with the increase of MeHg exposure until 50 mu M for both BerOc1 and G200 strains, suggesting that demethylation did not require intact biological activity. The formed IHg species were identified as various tetracoordinated Hg-S forms. These results highlight the important role of thiol ligands and Hg coordination in Hg methylation and demethylation processes.

Those particles are prepared in the batch heterophase polymerization of styrene or n-butyl acrylate, using a water-soluble poly(sodium acrylate) alkoxyamine macroinitiator. The nitroxide-mediated controlled growth of the hydrophobic block leads to the formation of poly(sodium acrylate)-b-polystyrene or poly(sodium acrylate)-b-poly(n-butyl acrylate) amphiphilic diblock copolymers, able to self-assemble in water simultaneously to the growth step. Abstract: International audience; Here, we present a new crystallization process which, by combining microwaves and metal-induced devitrification, reduces both the time and the temperature of crystallization compared to other known methods. Titania crystallization initiates at a temperature as low as 125 °C within a few minutes of microwave radiation. Several cations induce this low-temperature crystallization, namely, Mn2+, Co2+, Ni2+, Al3+, Cu2+ and Zn2+. The crystallization mechanism is probed with electron microscopy, elemental mapping, single-particle inductively coupled plasma mass spectrometry, X-ray photoelectron spectroscopy, Auger electron spectroscopy, and scanning Auger mapping. These techniques show that the metal ion migration through the vitreous titania under microwave radiation occurs prior to crystallization. The crystalline particles are suspended in solution at the end of the treatment, avoiding particle aggregation and sintering. The crystalline suspensions are thus ready for processing into a material or employment in any other application. This combination of microwaves and metal-induced crystallization is applied here to TiO2, but we are investigating its application to other materials as an ecofriendly crystallization method.

Abstract: International audience; Herein, we report the functionalization of selective nano-domains driven by the self-assembly of a polystyrene-block-poly(vinyl benzyl azide) PS-b-PVBN3 copolymer synthesized in three steps. First, a Polystyrene macro-initiator was synthesized, and then extended with vinyl benzyl chloride by Nitroxide Mediated Polymerization to form a polystyrene-block-poly(vinyl benzyl chloride) PS-b-PVBC. Nucleophilic substitution of the chloride into an azide moiety is then performed to obtain the PS-b-PVBN which self-assembled to form cylindrical domains of polystyrene and vinyl benzyl azide. Click chemistry was then used to chemically bind functional gold nanoparticles and poly(N-isopropylacrylamide) (PNIPAM) on PVBN3 domains due to the specific anchor at the surface of the nanopatterned film. Atomic Force Microscopy (AFM) was used to observe the block copolymer self-assembly and gold nanoparticles alignment at the surface of the PVBN3 nanodomains. Thorough X-ray Photoelectron Spectroscopy (XPS) analysis of the functional film showed the evidence of the sequential grafting of nanoparticles and PNIPAM. The hybrid surface expressed thermoresponsive properties and serve as pattern to perfectly align and control the assembly of inorganic particles at the nanoscale. 2

Abstract: International audience; This work provides a detailed study on the synthesis and characterization of silica coated iron nanoparticles (NPs) by coupling Transmission Electronic Microscopy (TEM), X-ray Photoelectron Spectroscopy (XPS) and magnetic measurements. Remarkably, iron NPs (of 9 nm of mean diameter) have been embedded in silica without any alteration of the magnetization of the iron cores, thanks to an original protocol of silica coating in non alcoholic medium. Tuning the synthesis parameters (concentration of reactants and choice of solvent), different sizes of Fe@SiO2 composites can be obtained with different thicknesses of silica. The magnetization of these objects is fully preserved after 24 h of water exposure thanks to a thick (14 nm) silica layer, opening thus new perspectives for biomedical applications. Hyperthermia measurements have been compared between Fe and Fe@SiO2 NPs, evidencing the self-organization of the free Fe NPs when a large amplitude magnetic field is applied. This phenomenon induces an increase of heating power which is precluded when the Fe cores are immobilised in silica. High-frequency hysteresis loop measurements allowed us to observe for the first time the increase of the ferrofluid susceptibility and remanence which are the signature of the formation of Fe NPs chains.

Abstract: International audience; Marine mucilage has been described as worldwide phenomena occurring sporadically, or frequently in certain coastal areas. They are transitory phenomena that can remain in the photic zone for several days or weeks. Their occurrence has been more frequent and persistent, and their magnitude has increased during the last decades. Their formation typically reflects an imbalance in the marine microbial communities, and has frequently be linked to global changes. Recurrent marine mucilage events have been observed in recent years in the Southern Bay of Biscay near the French coast, mainly south of the Adour estuary. In this study, we used a fingerprinting method (T-RFLP) targeting 16S and 18S rRNA genes to investigate marine microbial community composition and dynamics over an annual cycle across the three domains of life in coastal water at three depths, in the Adour River upstream of the mouth and in the marine mucilage. In line with studies conducted in marine environments we highlighted that the dynamics of marine microbial communities in the Bay of Biscay coincided with two environmental contexts: winter and spring on one hand and summer and autumn on the other hand. More interestingly, this dynamics affects also the marine pelagic mucilage that was observed during the sampling campaigns. Thus, the composition of the marine mucilage that appears in winter/spring context diverges from the composition of the marine mucilage that occurred during the summer/autumn context. We highlighted also that marine microbial communities were partly different from marine mucilage communities with an enrichment of some microorganisms suggesting that marine mucilage behaves as a microhabitat in seawater and possesses distinct microbial assemblages from the surrounding communities.

Abstract: International audience; Two novel strictly anaerobic bacteria, strains Bs105(T) and Bs107(T), were isolated from a deep aquifer-derived hydrocarbonoclastic community. The cells were rod-shaped, not motile and had terminal spores. Phylogenetic affiliation and physiological properties revealed that these isolates belong to two novel species of the genus Desulfotomaculum. Optimal growth temperatures for strains Bs105(T) and Bs107(T) were 42 and 45 degrees C, respectively. The estimated G+C content of the genomic DNA was 42.9 and 48.7 mol%. For both strains, the major cellular fatty acid was palmitate (C-16 : 0). Specific carbon fatty acid signatures of Gram-positive bacteria (iso-C-17 : 0) and sulfate-reducing bacteria (C(17 : 0)cyc) were also detected. An insertion was revealed in one of the two 16S rRNA gene copies harboured by strain Bs107(T). Similar insertions have previously been highlighted among moderately thermophilic species of the genus Desulfotomaculum. Both strains shared the ability to oxidize aromatic acids (Bs105(T) : hydroquinone, acetophenone, paratoluic acid, 2-phenylethanol, trans-cinnamic acid, 4-hydroxybenzaldehyde, benzyl alcohol, benzoic acid 4-hydroxybutyl ester; Bs107(T) : ortho-toluic acid, benzoic acid 4-hydroxybutyl ester). The names Desulfotomaculum aquiferis sp. nov. and Desulfotomaculum profundi sp. nov. are proposed for the type strains Bs105(T) (= DSM 24088(T) = JCM 31386(T)) and Bs107(T) (= DSM 24093(T) = JCM 31387(T)).

Abstract: International audience; Au@Prussian-Blue Analogue (PBA) shell nanoheterostructures are multifunctional nano-objects combining optical properties (surface plasmon resonance) of the Au core and magnetic properties of the PBA shell. We report in this article a series of new Au core@PBA shell nano-objects with different PBA shells: Au@K/Co/[FeII(CN)6] (2) and Au@K/Ni/[CrIII(CN)6]:[FeII(CN)6] (3) single PBA shell, as well as Au@K/Ni/[FeII(CN)6]@K/Ni/[FeIII(CN)6] (4) double PBA shell and Au@K/Ni/[FeII(CN)6]@K/Ni/[FeIII(CN)6]@K/Ni/[CrIII(CN)6] (5) triple PBA shell systems. The position and intensity of the Au SPR band, as well as the magnetic behaviour of the nanoheterostructures, are strongly affected by the shell composition and its thickness.

Abstract: International audience; Herein, a novel methodology to overcome the predominance of π−π interactions on the organization of rod-coil diblock copolymer is reported. A diblock copolymer poly(3-hexylthiophene)-block-poly(ethylene glycol methyl ether methacrylate) P3HT-b-PEGMA was synthesized by a Huisgen cyclo-addition, so-called “Click chemistry”, combining the P3HT and PEGMA blocks synthesized by Atom Transfer Radical Polymerization (ATRP) and Kumada Catalyst Transfer Polymerization (KCTP), respectively. Using a dip-coating process, the original film organization of the diblock copolymer was controlled by the crystallization of the P3HT block via π−π stacking. We demonstrated changes in the self-assembly morphology of the P3HT-b-PEGMA, by incorporating gold nanoparticles GNPs coated by Poly(EthyleneGlycol) ligands. Indeed, the crystalline structuration of the P3HT sequence was counterbalanced by the addition in the film of gold nanoparticles finely localized within the copolymer PEGMA matrix. Transmission Electron Microscopy (TEM) and ToF-SIMS analysis validated the GNPs homogeneous localization into the compatible PEGMA phase. Differential Scanning Calorimetry (DSC) showed the rod block crystallization disruption. A morphological transition of the self-assembly is observed by Atomic Force Microscopy (AFM) from P3HT fibrils into out of plane cylinders-like driven by the nanophase segregation.

Abstract: This work explores the possibilities for a further enhancement of the thermoelectric properties of Ca3Co4O9 by Sr-doping combined with hot-pressing. Modified hot-pressing process resulted in highly-textured and dense ceramics. Sr-doping significantly improves electrical properties, resulting in extremely large power factor (1.2 mW/K2m at 800 °C) due to simultaneous electrical resistivity decrease and Seebeck coefficient increase. The main effect on cumulative electrical performance is provided by the Seebeck coefficient, reaching 270μV/K at 800 °C. XPS revealed relatively high average cobalt oxidation state at room temperature (+3.3), compared to materials produced by conventional sintering. The results of combined XPS and Auger electron spectroscopy emphasize the importance of high densification in Ca3Co4O9-based ceramics for preventing phase decomposition and interaction with CO2 and moisture. Still, despite the exceptional electrical performance, the calculated figure-of-merit (estimated as 0.29 at 800 °C) is around the best reported in the literature due to a high thermal conductivity (4.4 W/K m at room temperature)., M. Abstract: International audience; The synthesis of iron germanide nanoparticles at the nanoscale is a challenging task. Here, we describe the preparation of nanocrystals of the hexagonal Fe1.67Ge phase via the thermolysis of single source precursors [{iPrNC(tBu)NiPr}RGe]Fe(CO)4 (where R = Cl, N(SiMe3)2) under mild conditions (200 °C). These bimetallic precursors and the corresponding germylenes [{iPrNC(tBu)NiPr}RGe] were fully characterized by spectroscopic techniques as well as single crystal X-ray diffraction. While the structural features of the molecular species were shown to be almost identical, the results of the thermolysis were highly dependent on the nature of R. When R = Cl, multimodal size distributions and non-controlled phases were obtained. Abstract: This study describes the functional characterization of two proteins, AupA and AupB, which are required for growth on alkanes in the marine hydrocarbonoclastic bacterium Marinobacter hydrocarbonoclasticus. The aupA and aupB genes form an operon whose expression was increased upon adhesion to and biofilm formation on n-hexadecane. AupA and AupB are outer and inner membrane proteins, respectively, which are able to interact physically. Mutations in aupA or/and aupB reduced growth on solid paraffin and liquid n-hexadecane, while growth on nonalkane substrates was not affected. In contrast, growth of aup mutants on n-hexadecane solubilized in Brij 58 micelles was completely abolished. Mutant cells had also lost the ability to bind to n-hexadecane solubilized in Brij 58 micelles. These results support the involvement of AupA and AupB in the uptake of micelle-solubilized alkanes and provide the first evidence for a cellular process involved in the micellar uptake pathway. The phylogenetic distribution of the aupAB operon revealed that it is widespread in marine hydrocarbonoclastic bacteria of the orders Oceanospirillales and Alteromonadales and that it is present in high copy number (up to six) in some Alcanivorax strains. These features suggest that Aup proteins probably confer a selective advantage in alkane-contaminated seawater., IMPORTANCE Bacteria are the main actors of the biological removal of hydrocarbons in seawater, and so, it is important to understand how they degrade hydrocarbons and thereby mitigate marine environmental damage. Despite a considerable amount of literature about the dynamic of microbial communities subjected to hydrocarbon exposure and the isolation of strains that degrade hydrocarbons, most of the genetic determinants and molecular mechanisms of bacterial hydrocarbon uptake remain unknown. This study identifies two genes, aupA and aupB, in the hydrocarbonoclastic bacterium Marinobacter hydrocarbonoclasticus that are present frequently in multiple copies in most of the marine hydrocarbon-degrading bacteria for which the genomic sequence is available. AupA and AupB are two novel membrane proteins interacting together that are involved in the uptake of alkanes dissolved in surfactant micelles. The function and the phylogenetic distribution of aupA and aupB suggest that they might be one attribute of the remarkable adaptation of marine hydrocarbonoclastic bacteria that allow them to take advantage of hydrocarbons.

Abstract: International audience; The interactions between a polyelectrolyte and an oppositely charged surfactant allows obtaining complexes with specific properties. Such complexes between a quaternized chitosan, obtained from the reaction between chitosan and quaternary ammonium epoxides, and sodium 1-decanesulfonate were studied. Physico-chemical characteristics were obtained from conductimetric experiments as well as surface tension studies. A strong associative electrostatic interaction between the polysaccharide and the surfactant was shown from conductimetric experiments and a degree of association equal to 0.74 was determined. The complex presents better interfacial properties than surfactant and polysaccharide alone. The critical aggregation concentration is much smaller than the critical micellar concentration of the surfactant alone in solution. Moreover the viscoelastic moduli of the interfacial layer was increased in presence of the complex by comparing with surfactant alone. All these considerations allow using the surfactant-polysaccharide complex as a better emulsion stabilizer than either surfactant or polysaccharide alone.

Abstract: The six possible VNxHy defects in diamond (a vacancy surrounded by x = 1 to 3 a vacancy surrounded by x = 1 to 3 nitrogens and y = 1 to 4-x hydrogens) are investigated at the quantum mechanical level by using the periodic supercell approach (64 atoms), an all electron Gaussian-type basis set, and hybrid functionals. It turns out that steric hindrance and short range repulsion is not such to prevent hydrogen atoms (up to 3 in VNH3) from fitting in the V cavity and saturating the dangling bonds of the carbon atoms first neighbors of the vacancy. All the investigated systems present specific IR spectra, with bending and stretching peaks above the nearly continuous band (from 400 to about 1340 cm−1) resulting from the perturbation of the perfect diamond spectrum by the vacancy and the nitrogen substitutions. These peaks, ranging from 1340 to 1700 cm−1 (bending; data refer to the harmonic approximation) and from 3100 to nearly 4000 cm−1 (stretching), might be considered fingerprints of the various defects. The C–H anharmonicity, evaluated numerically, turns out to be very sensitive to the N and H load, ranging from −202 cm−1 (redshift of the triplet state of VNH) to +57 cm−1 (blueshift of the symmetric stretching in VNH3).

Abstract: International audience; Hierarchically organized polymer films are produced with a high level of order from the combination of block copolymer nanophase segregation, “breath figure” methodology and microwave irradiation. A block copolymer based on poly(methyl methacrylate) and poly(n-butylacrylate) featuring cylindrical nanopatterns is involved in the “breath figure” process to create a microporous honeycomb structure. These films are submitted to microwave annealing to enhance the degree of ordering of the nano-segregation without the destruction of the honeycomb microstructure, which is not possible by classical thermal or solvent annealing. Ellipsometry, optical and atomic force microscopy are used to study three key parameters; the substrate nature, the film thickness and the microwave irradiation power. The silicon wafer is the substrate of choice to efficiently act as the heating transfer element and 60 seconds at 10 watts are enough to nicely order the 1 μm thick copolymer films. These conditions are eventually applied on hierarchically organized polymer films to obtain a hexagonal array of 100 nm deep holes within a matrix of perpendicularly aligned nano-cylinders.

Abstract: International audience; An analytical approach was developed to study the incorporation of selenium (Se), an important trace element involved in the protection of cells from oxidative stress, into the well-known probiotic Lactobacillus reuteri Lb2 BM-DSM 16143. The analyses revealed that about half of the internalized Se was covalently incorporated into soluble proteins. Se-enriched proteins were detected in 2D gels by laser ablation inductively coupled plasma mass spectrometry imaging (LA-ICP MSI) and identified by capillary HPLC with the parallel ICP MS (78Se) and electrospray Orbitrap MS/MS detection. On the basis of the identification of 10 richest in selenium proteins, it was demonstrated that selenium was incorporated by the strain exclusively as selenocysteine. Also, the exact location of selenocysteine within the primary sequence was determined. This finding is in a striking contrast to another common nutraceutical, Se-enriched yeast, which incorporates Se principally as selenomethionine. © 2013 by The American Society for Biochemistry and Molecular Biology, Inc.

Abstract: When half-cells are used to evaluate and understand electrochemical performance and interface properties of a given electrode, the metal electrode size is rarely mentioned. To evaluate such impact on the electrochemical performance and interphase formation, graphite/Li coin cells were used. Undersizing the Li metal electrode led to significantly lower charge/discharge capacities even at C/20 rate due to incomplete lithiation/delithiation processes of the graphite electrode edge. It also led to the formation of non-uniform Li metal deposits as well as to a graphite SEI film with a thickness and composition gradient across the electrode. Oversized Li led, however to homogeneous graphite SEI film. Overall, these results highlight the critical role of the metal electrode size in half-cells and should apply to all half-cells studies using other metal electrodes such as Na, K, Mg. Abstract: International audience; During the last few centuries oceanic island biodiversity has been drastically modified by human-mediated activities. These changes have led to the increased homogenization of island biota and to a high number of extinctions lending support to the recognition of oceanic islands as major threatspots worldwide. Here, we investigate the impact of habitat changes on the spider and ground beetle assemblages of the native forests of Madeira (Madeira archipelago) and Terceira (Azores archipelago) and evaluate its effects on the relative contribution of rare endemics and introduced species to island biodiversity patterns. We found that the native laurel forest of Madeira supported higher species richness of spiders and ground beetles compared with Terceira, including a much larger proportion of indigenous species, particularly endemics. In Terceira, introduced species are well-represented in both terrestrial arthropod taxa and seem to thrive in native forests as shown by the analysis of species abundance distributions (SAD) and occupancy frequency distributions (OFD). Low abundance range-restricted species in Terceira are mostly introduced species dispersing from neighbouring man-made habitats while in Madeira a large number of true rare endemic species can still be found in the native laurel forest. Further, our comparative analysis shows striking differences in species richness and composition that are due to the geographical and geological particularities of the two islands, but also seem to reflect the differences in the severity of human-mediated impacts between them. The high proportion of introduced species, the virtual absence of rare native species and the finding that the SADs and OFDs of introduced species match the pattern of native species in Terceira suggest the role of man as an important driver of species diversity in oceanic islands and add evidence for an extensive and severe human-induced species loss in the native forests of Terceira.

Abstract: International audience; Background: Selenoproteins (25 genes in human) co-translationally incorporate selenocysteine using a UGA codon, normally used as a stop signal. The human selenoproteome is primarily regulated by selenium bioavailability with a tissue-specific hierarchy. Methods: We investigated the hierarchy of selenoprotein expression in response to selenium concentration variation in four cell lines originating from kidney (HEK293, immortalized), prostate (LNCaP, cancer), skin (HaCaT, immortalized) and liver (HepG2, cancer), using complementary analytical methods. We performed (i) enzymatic activity, (ii) RT-qPCR, (iii) immuno-detection, (iv) selenium-specific mass spectrometric detection after non-radioactive 76 Se labeling of selenoproteins, and (v) luciferase-based reporter constructs in various cell extracts. Results: We characterized cell-line specific alterations of the selenoproteome in response to selenium variation that, in most of the cases, resulted from a translational control of gene expression. We established that UGAselenocysteine recoding efficiency, which depends on the nature of the SECIS element, dictates the response to selenium variation. Conclusions: We characterized that selenoprotein hierarchy is cell-line specific with conserved features. This analysis should be done prior to any experiments in a novel cell line. General significance: We reported a strategy based on complementary methods to evaluate selenoproteome regulation in human cells in culture.

Abstract: Cytochrome c oxidases are members of the heme-copper oxidase superfamily. These enzymes have different subunits, cofactors, and primary electron acceptors, yet they all contain identical heme-copper (CuB) binuclear centers within their catalytic subunits. The uptake and delivery pathways of the CuB atom incorporated into this active site, where oxygen is reduced to water, are not well understood. Our previous work with the facultative phototrophic bacterium Rhodobacter capsulatus indicated that the copper atom needed for the CuB site of cbb3-type cytochrome c oxidase (cbb3-Cox) is imported to the cytoplasm by a major facilitator superfamily-type transporter, CcoA. In this study, a comparative genomic analysis of CcoA orthologs in alphaproteobacterial genomes showed that CcoA is widespread among organisms and frequently co-occurs with cytochrome c oxidases. To define the specificity of CcoA activity, we investigated its function in Rhodobacter sphaeroides, a close relative of R. capsulatus that contains both cbb3- and aa3-Cox. Phenotypic, genetic, and biochemical characterization of mutants lacking CcoA showed that in its absence, or even in the presence of its bypass suppressors, only the production of cbb3-Cox and not that of aa3-Cox was affected. We therefore concluded that CcoA is dedicated solely to cbb3-Cox biogenesis, establishing that distinct copper uptake systems provide the CuB atoms to the catalytic sites of these two similar cytochrome c oxidases. These findings illustrate the large variety of strategies that organisms employ to ensure homeostasis and fine control of copper trafficking and delivery to the target cuproproteins under different physiological conditions., IMPORTANCE The cbb3- and aa3-type cytochrome c oxidases belong to the widespread heme-copper oxidase superfamily. They are membrane-integral cuproproteins that catalyze oxygen reduction to water under hypoxic and normoxic growth conditions. These enzymes diverge in terms of subunit and cofactor composition, yet they all share a conserved heme-copper binuclear site within their catalytic subunit. In this study, we show that the copper atoms of the catalytic center of two similar cytochrome c oxidases from this superfamily are provided by different copper uptake systems during their biogenesis. This finding illustrates different strategies by which organisms fine-tune the trafficking of copper, which is an essential but toxic micronutrient.

Abstract: International audience; This work reports on the synthesis of hard carbon spheres (HCS) and the impact of the pyrolysis temperature (1500 to 1900 °C) on the properties of HC and its relationship with the electrochemical performance in potassium-ion batteries (KIBs). Comparison with commercial graphite performance is provided as well. Spherical morphology, disordered structure, and low surface area were obtained for the HCSs. Most properties (interlayer space, active surface area, and oxygen-based functional groups) were found to decrease with increasing pyrolysis temperature, except for the helium density and closed porosity, which increase. However, graphite presents a flake-like morphology with a larger particle size, a higher helium density, an ordered structure with a smaller interlayer distance, and no closed pores. Electrochemical tests in a half-cell vs K+/K showed that HCSs perform better than graphite with higher initial Coulombic efficiency (ICE) and better specific capacities. The HCSs pyrolyzed at 1500 and 1700 °C exhibit the best initial Coulombic efficiency, ICEs of 54 and 62%, and specific capacities of 254 and 247 mAh g–1 (C/20, 11.5 mA g–1), respectively. The ICE is affected by multiple surface and bulk parameters but also by electrolyte formulation (67% for 0.8 M KFSI vs 62% for 0.8 M KPF6). The capacity is governed by diffusive phenomena, and a larger interlayer graphitic spacing and defects favor a better insertion of K ions. Closed pores did not lead to an improvement in capacity. Furthermore, HCSs exhibit significantly better capacity retention (97%) than graphite (84%), especially when cycled at high current rates (up to 10C depotassiation rate).

Abstract: International audience; Understanding the processes involved in the control of arsenic (As) dynamics within soils has become a challenging issue for soil and water quality preservation. Interactions between mineralogical phases, organic ligands and bacterial communities - closely linked to the chemical conditions of the medium - were thus investigated through a geochemical and microbiological experimental study involving the reduction of As(V)-doped lepidocrocite within the soil. Reducing conditions were established as soon as the experiment started, followed by a release of dissolved organic carbon corresponding to a release of acetate. Scanning Electron Microscopy observations pointed out a large bacterial colonization occurring on the lepidocrocite leading to a 3-dimensionally shaped biodissolution of lepidocrocite. The taxonomic diversity evolved throughout the experiment, and thus it demonstrated the evolution of the metabolic activities of the bacteria. At the beginning, lepidocrocite was mainly colonized by bacteria belonging to the Geobacter genus (Deltaproteobacteria) (26%) and Bacillus and Oxalophagus fermentative related genera (Firmicutes) (72%). After two weeks, Geobacter spp. and Firmicutes represented 54% and 30% of the bacterial community, respectively. Although still dominated by Geobacter spp. (34%) at the end of the experiment, the bacterial diversity had increased. After 3 and 8 weeks of incubation, the presence of the arsB and ACR3(1) genes, encoding transporters involved in As detoxification processes, indicated that this community harbored As-resistant or As-transforming genera able to contribute to the transformation of As(V) into As(III).

Abstract: Recent studies on mercury (Hg)-stable isotopes in Alaskan seabird eggs and ringed seal livers illustrated the control of sea ice cover on marine methyl-Hg photochemistry. Here, complementary marine mammal tissues have been analyzed to document variations in Hg-, carbon (C)-, and nitrogen (N)-stable isotope compositions of Arctic marine food webs. Hg-stable isotope ratios were measured in liver samples of 55 beluga whales (Delphinapterus leucas) and 15 polar bears (Ursus maritimus) collected since 1990. Large variations in delta Hg-202 (approximate to 2.1 parts per thousand) and Delta Hg-199 (approximate to 1.7 parts per thousand) are observed between species and within species stocks covering the Gulf of Alaska-Bering Sea-Arctic Ocean regions. Polar bears, mainly feeding on ringed seal (delta N-15 shift of 4.2 parts per thousand), show identical liver Delta Hg-199 of 0.5 parts per thousand, confirming the absence of metabolic mass-independent fractionation, and 0.33 +/- 0.11 parts per thousand enrichment in heavy Hg isotopes. Beluga whale liver total Hg concentrations increase with age, reflecting lifetime bioaccumulation, while Hg speciation shifts to inorganic Hg with age as a result of hepatic methyl-Hg breakdown. Delta Hg-200 variations in biota show a small, 0.1 parts per thousand decrease from North Pacific Ocean to Arctic Ocean habitats, suggesting atmospheric Hg deposition to be important in the Pacific and terrestrial Hg inputs to dominate in the Arctic Ocean. Similar to seabird eggs, a consistent south to north gradient in Delta Hg-199 baseline is seen in mammal liver tissues, confirming sea ice cover as a control factor on marine Hg photoreduction and Delta Hg-199. Arctic Ocean beluga whales have near zero Delta Hg-199, indicating that terrestrial Hg and in-situ-produced methyl-Hg are not measurably photoreduced in the Arctic Ocean before entering the marine food web.

Abstract: International audience; The monitoring and use of data concerning the quality of effluents transiting through the sanitation systems (sewerage networks and wastewater treatment plants) are necessary to increase the level of control and optimization of their operation. The Innovation and Environment Department of Siaap (Syndicat interdepartmental pour I'assainissement de I'agglomeration parisienne) has initiated, with its scientific partners, R&D actions aimed at developing processing ways to transform raw data into useful information for sanitation system operators. In this paper, mathematical laws are proposed and discussed that make it possible to link easily available physical measurements in sewerage networks [turbidity, conductivity) to the effluent quality: particle matter concentration, organic matter concentration (Chemical Oxygen Demand [COD}), ammonium (NH∗1 and total phosphorus (Ptot) concentrations. Results show that 1) turbidity allows an accurate estimation of particle matter and COD concentrations and 2) conductivity allows a fairly accurate estimation of ammonium concentrations and a quite good estimation of the phosphorus concentrations. Even if conclusions are valid for both studied watersheds (Paris and Pau), mathematical laws are significantly different considering the area but also considering time. This is mostly the case considering the correlation between conductivity and nitrogen for the Paris catchment area.

Abstract: International audience; In contrast to other trace elements that are cofactors of enzymes and removed from proteins under denaturing conditions, Se is covalently bound to proteins when incorporated into selenoproteins, since it is a component of selenocysteine aminoacid. It implies that selenoproteins can undergo several biochemical separation methods in stringent and chaotropic conditions and still maintain the presence of selenium in the primary sequence. This feature has been used to develop a method for the detection of trace levels of human selenoproteins in cell extracts without the use of radioactive isotopes. The selenoproteins are separated as a function of their isoelectric point (pI) using iso-electrofocusing (IEF) electrophoretic strips and detected by laser ablation-inductively coupled plasma mass spectrometry (LA-ICP MS). This method, therefore referred to as IEF-LA-ICP MS, allowed the detection of several selenoproteins in human cell lines, including Gpx1, Gpx4, TXNRD1, TXNRD2, and SELENOF.

Abstract: International audience; Blood and feathers are the two most targeted avian tissues for environmental biomonitoring studies, with mercury (Hg) concentration in blood and body feathers reflecting short and long-term Hg exposure, respectively. In this work, we investigated how Hg isotopic composition (e.g., δ202Hg and Δ199Hg) of blood and feathers from either seabird chicks (skuas, n = 40) or adults (penguins, n = 62) can accurately provide information on exposure to Hg in marine ecosystems. Our results indicate a strong correlation between blood and feather Hg isotopic values for skua chicks, with similar δ202Hg and Δ199Hg values in the two tissues (mean difference: −0.01 ± 0.25 ‰ and −0.05 ± 0.12 ‰, respectively). Since blood and body feathers of chicks integrate the same temporal window of Hg exposure, this suggests that δ202Hg and Δ199Hg values can be directly compared without any correction factors within and between avian groups. Conversely, penguin adults show higher δ202Hg and Δ199Hg values in feathers than in blood (mean differences: 0.28 ± 0.19‰ and 0.25 ± 0.13‰), most likely due to tissue-specific Hg temporal integration. Since feathers integrate long-term (i.e., the intermoult period) Hg accumulation, whereas blood reflects short-term (i.e., seasonal) Hg exposure in adult birds, the two tissues provide complementary information on trophic ecology at different time scales.

Abstract: Aim: Land-use change typically goes hand in hand with the introduction of exotic species, which mingle with indigenous species to form novel assemblages. Here, we compare the functional structure of indigenous and exotic elements of ground-dwelling arthropod assemblages across four land-uses of varying management intensity. Location: Terceira Island (Azores, North Atlantic). Methods: We used pitfall traps to sample arthropods in 36 sites across the four land-uses and collated traits related to dispersal ability, body size and resource use. For both indigenous and exotic species, we examined the impact of land-uses on trait diversity and tested for the existence of non-random assembly processes using null models. We analysed differences in trait composition among land-uses for both indigenous and exotic species with multivariate analyses. We used point–biserial correlations to identity traits significantly correlated with specific land-uses for each element. Results: We recorded 86 indigenous and 116 exotic arthropod species. Under high-intensity land-use, both indigenous and exotic elements showed significant trait clustering. Trait composition strongly shifted across land-uses, with indigenous and exotic species being functionally dissimilar in all land-uses. Large-bodied herbivores dominated exotic elements in low-intensity land-uses, while small-bodied spiders dominated exotic elements in high-intensity land-uses. In contrast, with increasing land-use intensity, indigenous species changed from functionally diverse to being dominated by piercing and cutting herbivores. Main conclusions: Our study revealed two main findings: first, in high-intensity land-uses, trait clustering characterized both indigenous and exotic elements; second, exotic species differed in their functional profile from indigenous species in all land-use types. Overall, our results provide new insights into the functional role of exotic species in a land-use context, suggesting that, in agricultural landscape, exotic species may contribute positively to the maintenance of some ecosystem functions.

Abstract: International audience; Otolith microchemistry can provide crucial information to address gaps of knowledge in spatio-temporal ecology of fish species. However, understanding the seasonal variability of water chemistry and its effect on otolith signatures is needed to interpret fish movements. Otolith multi-elemental signatures were used to examine the diadromous migration and small-scale movements of a tropical goby (Sicyopterus lagocephalus) within a watershed in La Réunion Island, Indian Ocean. The elemental ratios Sr:Ca, Ba:Ca and Sr:Ba recorded monthly in the water of nine sampling sites varied between localities and were correlated with otolith signatures, whereas the Mg:Ca signature was not. The incorporation of some trace elements in otoliths was influenced by the size of fish and site where they were caught, reflecting an effect of endogenous and exogenous factors. Despite these sources of variation, the elemental signatures did not overlap for three river sections distributed along the 26-km-long watershed. The diadromous migration and fish lifetime movements between these river sections were reconstructed using an original process based on random forest analysis. This approach revealed distinct migratory behaviours after fish entered in the river, with some individuals progressing slowly in the watershed whereas other rapidly reached the upper localities. As adults, S. lagocephalus showed a restricted exploratory behaviour and a strong site fidelity to a particular part of the watershed. The detection of adult movement was typically only between adjacent locations and is possibly explained by shifts in local intraspecific hierarchies and/or displacement during extreme discharge caused by cyclones.

Abstract: International audience; The concentrations of dissolved and particulate inorganic mercury (IHg(II)) and methylmercury (MeHg) from the contaminated Laranjo Bay (main freshwater discharge from the Antuã River) were measured by species-specific isotope dilution during six sampling campaigns at high and at low tide. Different effective riverine concentrations were calculated, based on salinity profiles, for specific hydrological conditions. The export fluxes of total Hg and MeHg (324 and 1.24 mol year−1, respectively) from the bay to the rest of the Aveiro Lagoon are much higher than the input fluxes from the Hg source (3.9 and 0.05 mol year−1) and from the Antuã River (10.4 and 0.10 mol year−1). Resuspension of contaminated sediments from Laranjo Bay is crucial for the transport of both IHg(II) and MeHg. Methylation and/or selective enrichment into biogenic particles is responsible for the mobilization of MeHg. Sorption of dissolved IHg(II) onto suspended particles limits its export flux. This is one of the rarest examples where both speciation fluxes and partitioning of mercury are studied in a contaminated coastal environment. Despite the lower fraction of total MeHg (relative to total Hg), the contaminated lagoon may have an impact on coastal areas, particularly if change in the lagoon geometry occurs, due to sea level rise.

When functional, these take part in for instance bacterial adhesion and biofilm formation. However, several of the peptides/proteins have been identified in various diseases. One of the current approaches to discover a cure against these relies on therapeutics which either prevent or accelerate peptide/protein aggregation and/or clear readily formed aggregates. Owing to the common interest of scientists from all disciplines to identify a cure against the diseases of public health concern, there are overwhelming numbers of publications dealing with these two approaches. Since amyloid aggregation could be regarded as a nucleated polymerization, which is an established mechanism of polymer self-assembly, we recently tackled the issue of amyloid aggregation using the theories and methods established in polymer science. Abstract: Lanthanide polymer-labelled antibodies were investigated to improve the analytical figures of merit of homogeneous immunoassays with inductively coupled plasma mass spectrometry (ICP-MS) detection for multiplexed biomarker analysis in human serum samples. Specific monoclonal antibodies against four cancer biomarkers (CEA, sErbB2, CA 15.3 and CA 125) were labelled with different polymer-based lanthanide group to increase the number of metal labels per binding site. After the immunoreaction of the biomarkers with the specific antibodies, antigen-antibody complexes were separated by size-exclusion chromatography followed by ICP-MS detection. The polymer label could be loaded with 30-times more atoms of the lanthanide that the lanthanide-DOTA complex traditionally used for this purpose elsewhere [1] which resulted in a 10-fold improvement in both sensitivity and detection limits. Analytical figures of merit obtained with the lanthanide polymer labelling strategy make the detection of the biomarkers feasible below the threshold reference values used in clinical analysis. This labelling method was successfully validated by analyzing a control human serum spiked with the four biomarkers at three different concentration levels. For all the biomarkers studied, the recovery values ranged from 95% to 110% whereas inter-assay and intra-assay precision were lower than 8%. Results obtained with this approach were equivalent to those obtained by heterogenous-based immunoassays based on the detection by electro-chemiluminescence or ELISA. However, the method developed offers better analytical figures of merit using a smaller amount of sample. Abstract: Metal halide perovskite nanocrystals (NCs) are promising for photovoltaic and light-emitting applications. Due to the softness of their crystal lattice, structural modifications have a critical impact on their optoelectronic properties. Here we investigate the size-dependent optoelectronic properties of CsPbI3 NCs ranging from 7 to 17 nm, employing temperature and pressure as thermodynamic variables to modulate the energetics of the system and selectively tune the interatomic distances. By temperature-dependent photoluminescence spectroscopy, we have found that luminescence quenching channels exhibit increased non-radiative losses and weaker exciton-phonon coupling in bigger particles, in turn affecting the luminescence efficiency. Through pressure-dependent measurements up to 2.5 GPa, supported by XRD characterization, we revealed a NC-size dependent solid-solid phase transition from the γ-phase to the δ-phase. Importantly, the optical response to these structural changes strongly depends on the size of the NC. Our findings provide an interesting guideline to correlate the size and structural and optoelectronic properties of CsPbI3 NCs, important for engineering the functionalities of this class of soft semiconductors.

Abstract: International audience; Sample preparation is a crucial point for quantitative multi-elemental analses by LA-ICP-MS of powdered geological materials. Four different methods are compared in this study with respect to signal stability and intensity as follows: the preparation of glass beads (GlassB) by alkaline fusion method and three grinding and pelletizing methods relying on the use of an organic binder (VanBind, vanillic acid), an adhesive binder (MixGlue, methyl methacrylate) and a sol–gel process for glass formation (SolGel, chemical reaction of tetraethoxysilane), respectively. Sixty elements were analyzed by means of a ns-UV (213 nm) laser ablation system coupled to a single collector sector field ICP-MS with a low or medium mass resolution. Signal stability was found to strongly depend on the sample homogeneity provided by the preparation method. These methods were applied to three geological standard materials (CRM). The following criteria were used to evaluate and compare the methods: (1) proportion of the measurement cycles which are above a given signal intensity threshold (defined here as signal average ± 3 times the standard deviation), (2) signal stability of the analyzed nuclides (internal precision estimated by the relative standard deviations on raw count rates), (3) signal stability of the internal standards added to the samples, (4) external precision estimated by the relative standard deviation over five preparations for each geological CRM. For the majority of the analyzed elements, signals measured for samples prepared with the four methods are reproducible. Specific contamination in one or several elements (Cr, Fe, Co, Ni, Cu, Mo, W, Au and Bi) was observed depending on the sample preparation method. In addition, compared to grinding made with PTFE material, grinding performed with tungsten carbide material was found to produce better homogeneity, especially for the sol–gel and mixing with glue protocols, although some metallic contamination (W and Co) was observed. Thanks to the suppression of grain effects by alkaline melting, the glass bead method systematically provided signal stability and percentage of “over the threshold” close to those of the NIST glasses. This may be explained by the preparation of more homogeneous samples by alkaline melting. Finally, the described methods were found to be reproducible for the majority of the analyzed elements.

Abstract: cited By 11; International audience; Discharge/charge process of classical lithium sulphur battery proceeds through intermediate polysulphides which are soluble in classical electrolyte systems. Due to concentration gradient soluble polysulphides easily diffuse/migrate out from cathode composite forming non-uniform distribution of the sulphur within cathode. Eventually polysulphides can be completely reduced on the metallic lithium anode. In this work we compare the sorption properties of manganese modified zeolite silicalite-1 (MnS-1) with a cathode composite containing SBA-15 additive and a cathode composite without additive. Careful analysis using XPS and FIB microscopy equipped with EDX show improved retention of polysulphide species within cathode composite in the case of MnS-1 zeolite as an additive. Interestingly, the amount of sulphur species detected by XPS on the metallic lithium is very similar regardless on cathode composite we use. Finally, similar cycling behaviour can be observed if MnS-1 zeolite is used as an interlayer between composite cathode and separator. © 2014 Elsevier B.V.
